微卫星DNA(microsatellite DNA)广泛存在于真核生物的基因组中。由于其具有突变频率快、多态性丰富、呈共显性遗传、通用性等特点,已成为近年来被广泛应用的分子遗传标记。本研究对10个海参微卫星DNA进行了克隆与测序。结果表明:90%的微卫星DNA序列存在长度多态性,这为进一步研究海参的分子标记辅助育种奠定了基础。  相似文献

海参i型溶菌酶基因及其编码产物的结构特点   总被引:7,自引:0,他引:7  
通过RT-PCR 和 RACE PCR技术,从海参(Stichopus japonicus)体壁中克隆得到一种溶菌酶基因(GenBank:EF036468).生物信息软件分析表明,其中全长cDNA为 713 bp,5′非编码区(UTR)246 bp,3′UTR 29 bp,开放阅读框438 bp,编码145个氨基酸,包括溶菌酶成熟肽124个氨基酸和信号肽21个氨基酸.对海参溶菌酶与多种无脊椎动物的c、g和i型溶菌酶进行分析比较,发现它与i型溶菌酶有较高的同源性,并具有i型溶菌酶高度保守的2个活性位点,即Glu34和Ser50.活性位点附近具有i型溶菌酶的一段特有的氨基酸保守序列MDVGSLSCG(P/Y)(Y/F)QIK,所以推断克隆的海参溶菌酶为i型.另外,通过搜索蛋白保守结构域数据库,发现海参溶菌酶与医用水蛭失稳酶相似性最高,并且这2个酶的三级结构模型也极其相似.因此推测,海参i型溶菌酶具有双功能特性,既能作用于细菌细胞壁的糖苷键使细胞裂解,又具有失稳酶的一些生化功能,能够水解纤维蛋白,这些特点在海参自溶过程中发挥重要的作用.  相似文献

Twenty microsatellite markers were first developed from the Japanese sea cucumber Stichopus japonicus using an enrichment protocol. Of the 20 microsatellite loci, 19 loci were polymorphic in the population examined. At these polymorphic loci, the number of alleles per locus varied from 2 to 15, and the observed heterozygosities ranged from 0.03 to 0.97, which is considerably higher than those previously found for allozymes. The high variability of the microsatellite markers identified in this study will make them excellent tools for genetic analyses of S. japonicus.  相似文献

The mass production of juvenile seeds of the sea cucumber,Stichopus japonicus has recently developed by the SeaFarming Center of Saga Prefecture. Methods for the culture ofperiphicic diatoms have been improved. There are three importantsteps in propagating the diatoms. The first step is theenrichment, with the addition of the nutrient salts, undercontrolled light intensity. The second step is washing withhigh pressure seawater and reversal of the plates. The laststep is elimination of copepods, which feed on diatoms, usinga pesticide. Small periphitic diatoms such as Navicula,Amphora, Achnanthes, and Nitzschia are easily culturedat a density of more than one million cells cm–2, andthese diatoms are able to induce larval metamorphosis andserve as a food source for juvenile sea cucumbers.  相似文献

Two Ca2 +–dependent lectins were purified from the sea cucumber Stichopus japonicus by affinity chromatography on lactosyl-Sepharose 4B and ion-exchange chromatography on Q-Sepharose. Their molecular masses were estimated to be 13kDa (SJL-I) and 15kDa (SJL-II) on SDS-PAGE. SJL-I agglutinated rabbit erythrocytes as well as human A, B, and O-type erythrocytes, but SJL-II agglutinated only rabbit erythrocytes. Hemagglutination by SJL-I was competitively inhibited by N-acetyl-D- galactosamine and galactose-containing carbohydrates. On the other hand, only lactose, melibiose, and raffinose gave weak inhibition of hemagglutination by SJL-II, suggesting that SJL-II may have high specificity for particular complex carbohydrate(s) on the surface of rabbit erythrocytes. SJL-II was activated at ten times lower Ca2+ concentration than SJL-I. Both lectins lost activity in acidic pH, while SJL-I appeared more stable down to pH 4.5.  相似文献

The genetic relationship among the three color variants (Red, Green, and Black) of the Japanese sea cucumber, S. japonicus, was investigated using 11 microsatellite markers. Genetic differentiation testing among the three sympatric color types showed the strong heterogeneity of Red (p < 0.001), while no significant difference was observed between Green and Black (p = 0.301 to 0.961). UPGMA trees constructed from 10 sample lots from 5 localities showed two distinct clusters, one from the Red types and the other from the Green and Black types. In addition, the sympatric Green and Black formed one subcluster with strong bootstrap support at each locality. These results indicate the separate species status of Red and the other color types, and also support the population identity of sympatric Green and Black.  相似文献

 从链霉菌 C- 3662发酵上清液中 ,通过硫酸铵沉淀 ,CM- Sepharose Fast Flow和 Phenyl-Sepharose Fast Flow等层析色谱 ,分离纯化得到了具有纤溶活性的蛋白酶 CGW- 3,反向 HPLC鉴定纯度为 90 % ;每立升发酵上清液可得到 8mg纯品 ,活性回收率 46% ,CGW- 3为一单肽链蛋白 ,分子量 2 2 72 1 ,对丝氨酸蛋白酶抑制剂 PMSF敏感 ,对 EDTA不敏感 ;其 N端 1 5个氨基酸的顺序为 VVGGTRAAQGEFPFM,与微生物来源的胰蛋白酶类丝氨酸蛋白酶有较高的同源性 . CGW- 3的等电点 p I9.0 ,纤溶活性的最适 p H为 7.5~ 8.0 ,对温度比较敏感 .CGW- 3不仅具有直接降解纤维蛋白作用 ,而且能够激活纤溶酶原  相似文献

Eleven polymorphic microsatellite markers were identified in expressed sequence tags generated from Stichopus japonicus cDNA libraries. The numbers of alleles ranged from three to 10, and the expected and observed heterozygosities ranged from 0.378 to 0.870 and from 0.077 to 0.690, respectively. Significant deviations from Hardy–Weinberg expectations were observed at eight loci due to homozygote excess, suggesting the widespread occurrence of null alleles. The microsatellite markers will be useful for examining genetic population structure, parentage analysis and mapping studies of S. japonicus.  相似文献

Bacillus sp. JER02 is a bacterial strain that can be grown in a medium containing organic solvents and produce a protease enzyme. JER02 protease was purified with a yield of 31.9% of total protein and 328.83-fold purification. K m and Vmax of this protease were established as 0.826 µM and 7.18 µmol/min, respectively. JER02 protease stability was stimulated about 80% by cyclohexane. It exhibited optimum temperature activity at 70°C. Furthermore, this enzyme was active in a wide range of pH (4-12) and showed maximum activity at pH 9.0. The nonionic detergents Tween-20 and Triton X-100 improved the protease activity by 30 and 20%, respectively. In addition, this enzyme was shown to be very stable in the presence of strong anionic surfactants and oxidizing agents, since it retained 77%, 93%, and 98% of its initial activity, after 1 hr of incubation at room temperature with sodium dodecyl sulfate (SDS), sodium perborate (1%, v/v) and H2O2 (1%, v/v), respectively. Overall, the unique properties of the Bacillus sp. JER02 protease suggested that this thermo- and detergent-stable, solvent-tolerant protease has great potential for industrial applications.  相似文献

使用DEAE-Sephadex A-50、POROS 50HS及POROS 20PI等色谱分离技术,从蕲蛇粗毒中分离提纯得到1个具有凝血活力的组分,经SDS-PAGE和PAGE检测均为一条带,非还原条件下相对分子质量24.3kDa,还原条件下相对分子质量33.0kDa;其凝血活力为91.0NIH u/mg。该组分不具有激活因子ⅩⅢ的活性,肝素不影响该组分的凝血活性,EDTA部分抑制其活性,苯甲基磺酰氟(PMSF)则会产生不可逆抑制作用,该酶N-末端氨基酸序列为VIGGNGXDINEHRFLVAFF,经分析表明该酶是一种新的类凝血酶。  相似文献

短裙竹荪子实体经生理盐水抽提、硫酸铵沉淀、DEAE Sepharose和SephadexG 10 0柱层析纯化得到短裙竹荪凝集素 (Dityophoraduplicata(Bosc)Fischerlectin) ,简称DDFL .DDFL经PAGE显示单一条带 ,SDS PAGE测得其亚基分子量为 2 2 3kD ,SephadexG 10 0凝胶过滤测得分子量为 4 5 3kD ,DDFL不含中性糖 ,IEF测得其等电点为 3 92 .该凝集素对供试的 4种血型人血和兔、小牛、鸭、鸡、鲫鱼以及青蛙血红细胞具有凝集作用 ,但不凝集鳖红细胞 .它还可以凝集小鼠脾脏淋巴细胞和小鼠S180 肉瘤细胞 ,对兔红细胞的凝集作用可被乳糖、棉子糖、半乳糖、α 甲基半乳糖、β 甲基半乳糖和N 乙酰半乳糖胺所抑制 .氨基酸组成分析表明 ,DDFL含有 17种氨基酸 ,其中天冬氨酸、丝氨酸、苯丙氨酸和丙氨酸含量较高 .经测定 ,其N末端为甘氨酸 .DDFL对热、酸和碱具有一定的稳定性 ,经 6 0℃处理 10min ,可保持较高的活性 ,在pH 4 0~ 9 0范围内较稳定 ,其凝血活性依赖于Mg2 + 和Ca2 + 二价阳离子 ,Mn2 + 和Zn2 + 则无影响 .DDFL对小鼠腹腔注射的半致死量为 70 6 3mg kg .  相似文献

The arginine kinase gene of sea cucumber Stichopus japonicus was cloned and inserted into the prokaryotic expression plasmid pET-21b. The protein was expressed in a soluble and functional form in Escherichia coli and purified by Blue Sepharose CL-6B, DEAE-32, and Sephadex G-100 chromotography with a final yield of 83 mgL(-1) of LB medium. The specific activity, electrophoretic mobility, and isoelectric focusing were all identical with those of arginine kinase that was purified from sea cucumber muscle. The fluorescence emission spectrum of arginine kinase had a maximum fluorescence at a wavelength of 330 nm upon excitation at 295 nm. These results are the first report of this purified protein.  相似文献

Urea titration was used to study the inactivation and unfolding equilibrium of arginine kinase (AK) from the sea cucumber Stichopus japonicus. Both fluorescence spectral and circular dichroism spectral data indicated that an unfolding intermediate of AK existed in the presence of 1.0 to 2.0 M urea. This was further supported by the results of size exclusion chromatography. The spectral data suggested that this unfolding intermediate shared many structural characteristics with the native form of AK including its secondary structure, tertiary structure, as well as its quaternary structure. Furthermore, according to the residual activity curve, this unfolding intermediate form still retained its catalytic function although its activity was lower than that of native AK. Taken together, the results of our study give direct evidence that an intermediate with partial activity exists in unfolding equilibrium states of AK during titration with urea.  相似文献

冬瓜的果肉中发现了丰富的蛋白水解酶.用硫酸铵将冬瓜果肉汁分级盐析,得到粗酶液.再经DEAE-Sepharose FF离子交换层析和Superdex-75柱层析等步骤得到一种电泳纯的冬瓜蛋白酶.SDS-PAGE测得其分子量为64 kDa.以酪蛋白做底物时,该酶的最适反应温度为70℃,最适作用pH为6.5,在pH 4.5~10.5,40~70℃范围内较稳定.PMSF强烈抑制该蛋白酶的活性.另外,Hg~(2+)对该酶有强烈的抑制作用,Mn~(2+)离子对其有保护作用,Zn~(2+)、Ca~(2+)和Cu~(2+)等离子对其活性没有影响.  相似文献

酰基辅酶A结合蛋白(acyl-CoA-binding protein,ACBP)对长链脂酰基辅酶A(long-chainfatty acyl-CoA esters,LCACoA)有很高的亲和力,因而对LCACoA在细胞内的运输和利用过程起重要的作用。本文采用RACE技术从鲈鱼肝脏中克隆了Acbp基因的全长cDNA序列,该基因全长cDNA 679 bp,5'端和3'端的非翻译区分别为83 bp和326 bp,开放阅读框为270 bp。推测编码89个氨基酸,理论等电点为5.44,分子量为10.14 kDa。鲈鱼Acbp与青鳉鱼、银鳕鱼、大西洋鲑和人的同源性分别为87%、84%、78%和68%。用RT-PCR和实时定量PCR检测鲈鱼肌肉、心脏、眼、大脑、消化道、肾脏、脂肪组织、脾脏、鳃和肝脏等10种组织的Acbp基因的表达情况,结果表明,在肾脏和肝脏的表达量高,肌肉、眼睛和大脑中表达低。定量PCR检测表明鲈鱼肝脏Acbp的表达在饥饿时明显下降,胰岛素上调其表达,葡萄糖对其表达则没有影响。  相似文献

对中国北方海域江蓠属养殖龙须菜(Gracilaria lemaneiformis)进行了溴过氧化物酶分离纯化及性质的研究。粗提液中酶催化检测反应不稳定, 活力单位较低或无; 经DEAE cellulose 52离子交换层析, 去除了结构多糖及藻胆蛋白, 酶催化反应稳定, 得到比活力为2.8的电泳纯溴过氧化物酶。对纯化溴过氧化物酶性质研究表明: 该溴过氧化物酶为单体酶, 分子量约66 kD, 溴化单氯双甲酮时的最适pH值为6.0, 在40°C以下和pH 3.0~9.0之间有很好的稳定性。钒酸盐可提高该溴过氧化物酶的催化活性, 而Fe2+、Fe3+、Cu2+、Zn2+和EDTA等化合物对其有较显著的抑制作用。反应动力学实验表明, 该酶对Br-、H2O2的Km分别为53.5 mmol/L和38 mmol/L。  相似文献
