A rapid method for predicting the buckwheat flour ratio of dried buckwheat noodles was developed by using the fluorescence fingerprint and partial least squares regression. Fitting the calibration model to validation datasets showed R(2)=0.78 and SEP=12.4%. The model was refined for a better fit by deleting several samples containing additional ingredients. The best fit was finally obtained (R(2)=0.84 and SEP=10.4%) by deleting the samples containing vinegar, green tea, seaweed, polysaccharide thickener, and yam. This result demonstrates that a calibration model with high accuracy could be constructed based on samples similar in material composition. The developed methodology requires no complex preprocessing, enables rapid measurement with a small sample amount, and would thus be suitable for practical application to the food industry.  相似文献

In situ Raman spectroscopy was employed for real‐time monitoring of simultaneous saccharification and fermentation (SSF) of corn mash by an industrial strain of Saccharomyces cerevisiae. An accurate univariate calibration model for ethanol was developed based on the very strong 883 cm?1 C–C stretching band. Multivariate partial least squares (PLS) calibration models for total starch, dextrins, maltotriose, maltose, glucose, and ethanol were developed using data from eight batch fermentations and validated using predictions for a separate batch. The starch, ethanol, and dextrins models showed significant prediction improvement when the calibration data were divided into separate high‐ and low‐concentration sets. Collinearity between the ethanol and starch models was avoided by excluding regions containing strong ethanol peaks from the starch model and, conversely, excluding regions containing strong saccharide peaks from the ethanol model. The two‐set calibration models for starch (R2 = 0.998, percent error = 2.5%) and ethanol (R2 = 0.999, percent error = 2.1%) provide more accurate predictions than any previously published spectroscopic models. Glucose, maltose, and maltotriose are modeled to accuracy comparable to previous work on less complex fermentation processes. Our results demonstrate that Raman spectroscopy is capable of real time in situ monitoring of a complex industrial biomass fermentation. To our knowledge, this is the first PLS‐based chemometric modeling of corn mash fermentation under typical industrial conditions, and the first Raman‐based monitoring of a fermentation process with glucose, oligosaccharides and polysaccharides present. 苦荞(Fagopyrum tataricum)芽菜是一种新兴健康食品,该研究通过不同光周期(0、4、8、12、16和20 h·d~(-1))处理苦荞芽菜,测定其生物量、叶绿素和主要营养成分的含量以及黄酮合成相关基因的表达水平,以明确不同光周期对苦荞芽菜品质的影响机理。结果表明:(1)随着苦荞芽菜生长时间的增加,芽菜生物量总体呈上升趋势,16 h·d~(-1)光周期时芽菜生长状态最好,鲜重最大。(2)不同光周期对苦荞芽菜各营养成分含量的影响有所差异,可溶性糖含量在芽菜萌发第2天开始逐渐下降,10 d后含量仅为第2天的13.5%~14.5%;花青素含量在芽菜萌发第2天光周期为12 h·d~(-1)时最高(2.16 mg/g),不同光周期处理4~10 d后均显示出降低趋势;芦丁是苦荞芽菜中主要的黄酮类化合物,芦丁含量在不同光周期处理后有所变化且适当的光照有利于其合成和积累,并在处理第4天光周期为16 h·d~(-1)时含量最高(59.60 mg/g)。(3)qRT-PCR分析表明,不同光周期处理的苦荞芽菜中各黄酮合成相关基因的表达量在第2天最高,随生长周期其表达量均不同程度降低,但黄酮醇合成支路关键酶基因FtFLS1和花青素支路基因FtDFR2表达量上升、且显示出强烈的光诱导特征。研究发现,光周期对苦荞芽菜的生物量影响较大,黑暗有利于胚轴伸长,长光周期(16 h·d~(-1))有利于鲜重的增加;在苦荞萌发过程中,苦荞芽菜可溶性糖、花青素和芦丁含量整体呈减少趋势,可溶性糖和花青素含量均在光周期16 h·d~(-1)处理第2天时最高,芦丁含量在光周期为16 h·d~(-1)处理第4天时最高,建议苦荞芽菜在光周期为16 h·d~(-1)生长4 d时采食。  相似文献

Present investigation deals with a search for plant sources which have high α-glucosidase activities. As a result of the screening test on 8 kinds of plant source (spring wheat, maize, buckwheat, malted barley, oat, soybean, azuki bean and kidney bean), buckwheat seed was found to have the highest α-glucosidase (maltase) activity. Changes in α-glucosidase and amylase activities during the germination stages of buckwheat were also examined. It was found that α-glucosidase was rich in seeds and seedlings at initial stages of germination. In addition, the probable existence of glucamylase activity was suggested in the former, and a rapid appearance of α-amylase was recognized in the latter.

It was concluded that ungerminated buckwheat seed was excellent as the material for the preparation of plant α-glucosidase.  相似文献

An AUDPC model was developed to describe the relationship between Alternaria porri infection and yield loss in garlic. Percentage yield loss was regressed against AUDPC and gave an acceptable fit to a linear model with an intercept of -0.35, a slope of 0.09 and R2= 0.85 during 1990–91 and an intercept of 1.91, a slope of 0.08 and R2= 0.87 during 1991–92. The effect of various levels of leaf damage on garlic bulb yield was also studied to simulate damage caused by A. porri. Significant yield reduction by 25% defoliation at 5 wk, 50% defoliation at 4 wk and 75% defoliation at 3 wk before bulb maturity were recorded. No significant yield reduction from 2 wk before bulb maturity could, however, be noticed even at the highest levels of leaf damage. Linear regression models were fitted for predicting yield at various levels of defoliation at different weeks before bulb maturity. These models can be used to describe the consequences of disease epidemics, projecting possible losses during the season and justifying the need for chemical disease control.  相似文献

土壤有机碳是影响土壤肥力的最重要因素之一。生物炭由于具有高度芳香化碳结构和发达孔隙结构等特性,可以作为一种土壤改良剂,提高土壤有机碳含量,改善土壤物理结构,近些年成为农业环境领域研究的热点。分别采用传统方法和可见光近红外光谱(VIS-NIRS,400-2500 nm)技术对施加不同用量生物炭的土壤有机碳含量进行检测和对比分析,以期为含生物炭土壤的有机碳分析建立有效预测模型。通过比较不同样本选择方法(Kennard-Stone(KS),Random selection(RS)和Sample set partitioning based on joint x-y distances(SPXY))和光谱预处理方法(Savitzky-Golay平滑(SG)、倒数的对数log(1/R)、标准正态变量变换(SNV)、一阶导数(Der1)、二阶导数(Der2)和多元散射校正(MSC)),以3种模型(组合间隔偏最小二乘模型(Synergy Interval Partial Least Squares,siPLS),遗传算法-支持向量机模型(Genetic Algorithm-Support vector machine,GA-SVM)和随机森林模型(Random Forest,RF))来建立生物炭土壤有机碳预测模型。结果表明:(1)施加生物炭增加了土壤有机碳含量,增加幅度随生物炭添加量的提高呈增加趋势;(2)土壤反射率随土壤有机碳含量的增加而降低,在1410、1920和2200 nm光谱附近存在明显的吸收谷;(3)对比3种样本选择方法,KS方法所划分的样本集相对于RS方法和SPXY方法更适用于生物炭土壤有机碳模型的建立;(4)以SG+MSC预处理结合GA-SVM方法建立的模型精度最高,校正集的Rcal2和RMSECV值分别为0.9526和0.4839,验证集的R2val和RMSEP值分别为0.8598和0.9987,RPD值为2.6017。该模型因具有精度高且模拟效果较好等优点,可用于含生物炭土壤的有机碳含量的科学预测。  相似文献

Based on the fluorescence quenching of Terbium (III)‐sodium hexametaphosphate (Tb/SHMP) chelates in the presence chromate (III), a sensitive fluorimetric method was developed for the determination of trace amounts of chromium (III) in aqueous solutions. Under the optimum conditions, the linear calibration graph was obtained (R = 0.996). The linear range and detection limit of Cr (III) were 7.69 × 10?7 to 1.15 × 10?4 mol L?1 and 4.50 × 10?7 mol L?1, respectively. The proposed method had a wider linear range and was proved to be very sensitive, rapid and simple. The method was applied successfully to the determination of chromium (III) in the synthetic samples and real water samples. Moreover, the reaction mechanism was discussed through the fluorescence lifetime and proved to be dynamic quenching behavior. Copyright © 2010 John Wiley & Sons, Ltd.  相似文献

Cord blood is widely used as surrogate tissue in epigenome-wide association studies of prenatal conditions. Cell type composition variation across samples can be an important confounder of epigenome-wide association studies in blood that constitute a mixture of cells. We evaluated a newly developed cord blood reference panel to impute cell type composition from DNA methylation levels, including nucleated red blood cells (nRBCs). We estimated cell type composition from 154 unique cord blood samples with available DNA methylation data as well as direct measurements of nucleated cell types. We observed high correlations between the estimated and measured composition for nRBCs (r = 0.92, R2 = 0.85), lymphocytes (r = 0.77, R2 = 0.58), and granulocytes (r = 0.72, R2 = 0.52), and a moderate correlation for monocytes (r = 0.51, R2 = 0.25) as well as relatively low root mean square errors from the residuals ranging from 1.4 to 5.4%. These results validate the use of the cord blood reference panel and highlight its utility and limitations for epidemiological studies.  相似文献

Fermented buckwheat sprouts (FBS) are used as multifunctional foods. Their production process includes fermentation with lactic acid bacteria. The major strains were found to include Lactobacillus plantarum, Lactobacillus brevis, Lactobacillus pentosus, Lactococcus lactis subsp. lactis, and Pediococcus pentosaceus in an investigation of the lactic acid bacteria. We searched for the functional components, and nicotianamine (NA) and 2″-hydroxynicotianamine (HNA) were identified as angiotensin I-converting enzyme (ACE) inhibitors. NA and HNA increased during fermentation. Indole-3-ethanol was identified as an antioxidant (a SOD active substance), and may have been generated from tryptophan during fermentation because it was not contained in green buckwheat juice. A safety test demonstrated that FBS contained were safe functional food components, showing negative results in buckwheat allergy tests. Any buckwheat allergy substances might have been degraded during the fermentation process.  相似文献

ABSTRACT The metal bioadsorption potential of shell dust of the freshwater snail Melanoides tuberculata (MTSD) was evaluated under laboratory conditions using cadmium as a model metal. As bioadsorbent, MTSD exhibited a biosorption capacity of 27.03 mg g?1 at pH 6, indicating potential to remove cadmium from aqueous solution. The adsorption data fit more to the Langmuir (R2 = 0.998) equation than the Freundlich (R2 = 0.761) equation at equilibrium condition. The kinetics of biosorption followed the pseudo-second-order model (R2 = 0.999) better than the Lagergren model (R2 = 0.676), as was evident from the regression analysis. The presence of calcium ions appears to have facilitated ion exchange with cadmium along with the binding of different functional groups, as revealed through Fourier transform infrared (FT-IR) analysis. It is apparent from these observations that MTSD can act as low-cost and efficient bioadsorbent for cadmium bioremediation from aquatic habitats. Use of the shells of M. tuberculata for metal biosorption will promote the utility of a waste material of biological origin for bioremediation of heavy metals such as cadmium.  相似文献

Comparative morphological and physiological studies of the ancestral forms (Fagopyrum homotropicumO. and F. esculentumssp. ancestraleO.) and the various morphogenotypes of common buckwheat (F. esculentumMoench.) were carried out in glasshouse experiments in a soil culture. A considerable reduction of plant morphogenesis, a restriction of the growth of the vegetative organs, and the ecological and agricultural specialization (accompanied by the change in the primary adaptive life strategy from competitive (c) to ruderal (r) or to combined c/rtype) occurred in the evolution of common buckwheat. The reduction of morphogenesis was accompanied by changes in some of the morphological and anatomical structures and their functions, primarily, the source–sink relationships among plant organs. Compensatory mechanisms were developed, and the nutritional compounds became reutilized as an additional source for fruit formation and ripening.  相似文献

We report for the first time that the sensitivity of the luminol–hypochlorite chemiluminescence (CL) reaction was enhanced approximately 10 times by the addition of phloxine B. The maximum wavelength of CL emission shifted from 431 to 595 nm in the absence and presence, respectively, of phloxine B, suggesting that an efficient chemiluminescence resonance energy transfer occurred between a luminol donor and a phloxine B acceptor in the luminol–hypochlorite–phloxine B system. Based on this observation, a simple, rapid and sensitive microflow injection CL method, using a microchip with spiral channel configurations, was developed for the determination of hypochlorite. Under optimized conditions, a linear calibration curve (R2 = 0.9944) over the range 0.1–10.0 µmol/L was obtained, with a detection limit of 0.025 µmol/L (S:N = 3). The relative standard deviation (RSD) was found to be 4.2% (n = 10) for 2.5 µmol/L hypochlorite. The sample consumption was only 2 μL, with a sample throughput of 90/h. 1. A growth model, originally developed for brown trout (Salmo trutta), has now been fitted to data for Atlantic salmon (S. salar) and stone‐loach (Barbatula barbatula) from English populations, and Arctic charr (Salvelinus alpinus) from Sweden. The model relates growth rate to temperature for a fish of standard size and the functional relationship has a triangular shape with a sharp peak at the optimal temperature for growth and zero growth at the base of the triangle. It was unsuitable for growth data for Norwegian salmon, and a curvilinear Ratkowsky model provided a better fit, though the experimental protocol was different in the Norwegian and English experiments. 2. The Norwegian salmon were kept in groups in each tank, had to compete for food, and had to be divided into slow, moderate and fast growers before the Ratkowsky model could be fitted. Each English salmon was kept in its own tank and fed individually. For replicate experiments, fish of similar size were selected. Variation among fish kept under similar conditions was therefore small, and the triangular model was essentially for individual fish, not groups of fish. 3. The present simulation study tests the hypothesis that individual differences in the growth response could account for the curvilinear growth‐temperature relationship for the Norwegian salmon. The triangular model was used to generate the growth response to temperature for a group of salmon, each fish having a slightly different temperature preference and growth rate. The result was a curvilinear response, well approximated by the Ratkowsky model (adjusted R2 = 0.96). When the variability in individual temperature preference was increased, the Ratkowsky model was an even better fit (adjusted R2 = 0.98). Therefore, the apparent discrepancy between the two models was reconciled by allowing for individual differences in temperature preference and growth rate within groups of fish.  相似文献

The objective of this study was to use diffuse reflectance near infrared spectroscopy (NIRS) to determine racemic compound content in physical mixtures composed primarily of the enantiomorph and to assess the error, instrument reproducibility and limits of detection (LOD) and quantification (LOQ) of the method. Physical mixtures ranging from 0 to 25% (±)-tartaric acid in (+)-tartaric acid were prepared and spectra of the powder samples contained in glass vials were obtained using a Foss NIRSystems Model 5000 monochrometer equipped with a Rapid Content Analyzer scanning from 1100 to 2500 nm. A calibration curve was constructed by plotting (±)-tartaric acid weight percent against the 2nd derivative values of log (1/R) vs λ at a single wavelength, normalized with a denominator wavelength (1480 nm/1280 nm). Excellent linearity was observed (R2=0.9999). The standard error of calibration (SEC) was 0.07 and the standard error of prediction (SEP) for the validation set was 0.11. Instrument and method errors for samples in the 2% composition range ((±)-tartaric acid in (+)-tartaric acid) were less than 1% RSD and 3% RSD, respectively. The practical LOD and LOQ were 0.1% and 0.5%, respectively, and comparable to the calculated LOD and LOQ. These studies show that NIRS can be used as a rapid and sensitive quantitative method for determining racemate content in the presence of the enatiomerically pure crystal in the solid-state. The biological rhythms of microalgal growth within a hydraulically integrated serial turbidostat algal reactor (HISTAR) were examined after comparison of a simple mechanistic productivity model with actual data yielded a standard error of prediction (SEP) of 62%. The data used for this study were taken on cultures of Selenastrum capricornutum grown under continuous 400-watt metal halide lighting. Fourier series analysis (up to five harmonics) was used to model the biorhythms and differentiate them from stochastic processes. Regression analyses revealed that the best Fourier series fit for the data was a three harmonic summation. Regression analyses on additional harmonic summations did not increase r 2 by more than 1%. The three harmonics were summed and incorporated into the growth term of the simple model, and the resultant full model was calibrated. The mechanistic HISTAR productivity model was greatly enhanced by the addition of the biological rhythm component, resulting in a SEP of <24.8 %. The period of the first harmonic was 13.4 days, which is very close to a circasemilunar rhythm (14.8 days). In summary, the predictive power of productivity models for continuous microalgal cultures can be dramatically improved with the inclusion of a biorhythm analysis.  相似文献

Two experiments were carried out in a growth chamber to investigate the phosphorus (P)-uptake efficiency of Fagopyrum esculentum Moench (buckwheat) and Triticum aestivum (spring wheat) from a Ca-bound form. The first experiment was based on a sand-culture system with either rock phosphate (RP) or CaHPO4 (CaHP) as the P source and nitrate or ammonium nitrate as nitrogen source. A highly calcareous soil was used in the second experiment. Buckwheat was shown to be highly efficient in taking up Ca-bound P compared to spring wheat. When plants were supplied with nitrate, the total P uptake by buckwheat from RP was nearly 10-fold higher than that of spring wheat (20.1 compared with 2.1 mg P pot–1). Changing nitrogen source from nitrate only to ammonium nitrate increased P uptake by spring wheat substantially, but not buckwheat. High P-uptake efficiency of buckwheat was also demonstrated using the field soil, but to a lesser extent, which may be related to the difference in Zn supply between sand culture and field soil. It is suggested that buckwheat may be included in intercropping or crop rotation systems to activate P sources in calcareous soils. The principal mechanism of P uptake efficiency of buckwheat may be its ability to acidify the rhizosphere; however, further study is needed to unravel the regulation of root excretion of H+ and its molecular basis in order to exploit buckwheat's genetic capability to utilise sparingly soluble P from soil.  相似文献

Isolated pistils of dimorphic buckwheat (Fagopyrum esculentum Moench.) flowers were treated with phosphatase inhibitors (ocadaic acid and cantharidin) and with calcium antagonists (verapamil, La3+, and A23187). They were subsequently cross- or self-pollinated, and the growth of pollen tubes was observed under the fluorescence microscope. All treatments suppressed inhibition of pollen tubes growth suggesting that protein phosphatases and calcium signaling may be involved in self-incompatibility signal transduction in buckwheat. Rapid development in the glutamate fermentation industry has dictated the need for effective fermentation monitoring by rapid and precise methods that provide real-time information for quality control of the end-product. In recent years, near-infrared (NIR) spectroscopy and multivariate calibration have been developed as fast, inexpensive, non-destructive and environmentally safe techniques for industrial applications. The purpose of this study was to develop models for monitoring glutamate, glucose, lactate and alanine concentrations in the temperature-triggered process of glutamate fermentation. NIR measurements of eight batches of samples were analyzed by partial least-squares regression with several spectral pre-processing methods. The coefficient of determination (R 2), model root-mean square error of calibration (RMSEC), root-mean square error of prediction (RMSEP) and residual predictive deviation (RPD) of the test calibration for the glutamate concentration were 0.997, 3.11 g/L, 2.56 g/L and 19.81, respectively. For the glucose concentration, R 2, RMSEC, RMSEP and RPD were 0.989, 1.37 g/L, 1.29 g/L and 9.72, respectively. For the lactate concentration, R 2, RMSEC, RMSEP and RPD were 0.975, 0.078 g/L, 0.062 g/L and 6.29, respectively. For the alanine concentration, R 2, RMSEC, RMSEP and RPD were 0.964, 0.213 g/L, 0.243 g/L and 5.29, respectively. New batch fermentation as an external validation was used to check the models, and the results suggested that the predictive capacity of the models for the glutamate fermentation process was good.  相似文献

A very low-angle light-scattering photometer is described with respect to optical features, scattering cell, correction factors, and absolute calibration in the angular range 2°–35°. An improved microfiltration apparatus was used to obtain essentially dust-free aqueous solutions for very low-angle light scattering. The instrument was calibrated with silicotungstic acid, an absolute molecular-weight standard, and the calibration was confirmed with the use of several secondary standards. Very low-angle light-scattering measurements were made to determine the weight-average molecular weight M?r and z-average radius of gyration Rg,z of a commerical preparation of calf-thymus DNA. Microfiltration of the solutions allowed measurements down to 6°. The value M?r = 20.0 × 106 obtained by extrapolating 6°–9° data to 0° is more than three times that from 30°–75° data (6.38 × 106) but ~20% smaller than that from 10–35° data (23.7 × 106). The experimental errors in M?r and Rg,z are estimated to be ±8% and ±14%, respectively. Combined 6°–75° data from two photometers fit well a theoretical scattering curve for a model wormlike coil of the same M?r as the DNA sample.  相似文献

Background: Professional oral health care can reduce the number of oral bacteria and days of fever and inhibit the development of pneumonia. A novel detection apparatus was developed by applying the dielectrophoretic impedance measurement method. Methods: First, to determine the accuracy of this apparatus, employing standard samples of Escherichia coli. Next, to evaluate the oral environment, samples were taken from the tongue in elderly (mean age: 86.6 years) in nursing home. Results: In the first study, a good correlation was observed between the two methods (R = 0.999). In the second study, there were significant correlations between measurement values obtained using this apparatus and those obtained by the culture method (R = 0.852), as well as those obtained by the FM method (R = 0.885). Conclusion: Our data showed that this rapid oral bacterial detection apparatus is effective in evaluating the oral hygiene to prevent pneumonia in the elderly.  相似文献
