The aqueous fraction of Fushimi sweet pepper increased the repair effect of the solvent control against UV-induced cyclobutane pyrimidine dimers in human fibroblast to 150%, but ordinary sweet pepper did not have a statistically significant effect. When Fushimi sweet pepper was boiled, the activity of the aqueous fraction was elevated to 209% of the control (p<0.05), while that of the grilled state was decreased to 125% of the control. The repair activity of a dialyzate (MW<12,000) of the aqueous fraction from Fushimi sweet pepper showed 191% of the control (p<0.05). The dialyzate was contained 1.9% in the weight of the fresh fruit body of Fushimi sweet pepper, and the activity can be stable in its boiling state, and it might be therefore considered to be the worthy source for expecting the DNA repair activity in human diet.  相似文献

Abstract

Field experiments were carried out at the Teaching and Research Farm of the Abubakar Tafawa Balewa University, Bauchi during the rainy season of 2005. The study was carried out with sole objective of evaluating the efficacy of six selected plant materials (sweetsop, red pepper, garlic, neem, mahogany and gmalina) against the major insect pests of cowpea variety, Dan Sokoto. The experiment was laid down in randomised complete block design with seven treatments. Each treatment was replicated three times. The results of the study showed that all the plant materials used were significantly (p < 0.05) better than control where no plant material was used in controlling the population of B. tabaci, E. dolichi, M. sjostedti and C. tomentosicollis at 1, 2 and 3 days after application of the treatments. Similarly the effects of these plant materials on the number of seeds/pod showed a significant (p < 0.05) difference between plots treated with plant materials and control. On the grain yield of the crop, all the plant materials showed significant effect except mahogany and the control which were statistically similar. Furthermore, the order of level of control indicated that sweet sop has (70.7%), garlic (69.3%), neem (61.0%), red pepper (54.0%), ash (30.9%), and mahogany (3.5%). The result of the present finding therefore recommends the use of sweet sop, garlic and neem as they were found to be the most promising in the control of major cowpea insect pests.  相似文献

Gastric cancer is the second most common cancer worldwide. The involvement of reactive oxygen species (ROS) in the pathogenesis of gastric malignancies is well known. Many human tumours have shown significant changes in the activity and expression of superoxide dismutase (SOD), which might be correlated with clinical–pathological parameters for the prognosis of human carcinoma. The aim of this study is the detection of MnSOD and CuZnSOD activity and their expression in gastric adenocarcinoma and healthy tissues. Gastric samples (adenocarcinoma and healthy tissues) harvested during endoscopy or resected during surgery were used to determine MnSOD and CuZnSOD activity and expression by spectrophotometric and Western blotting assays. The total SOD activity was significantly higher (p<0.05) in healthy mucosa with respect to gastric adenocarcinomas. No differences were found in MnSOD activity and, on the contrary, CuZnSOD activity was significantly lower (p<0.001) in cancer samples with respect to normal mucosa. The rate of MnSOD/CuZnSOD activity in adenocarcinoma was over ninefold higher than that registered in healthy tissues (p<0.05). Moreover, in adenocarcinoma MnSOD activity represented the 83% of total SOD with respect to healthy tissues where the ratio was 52% (p<0.001). On the contrary, in cancer tissues, CuZnSOD activity accounted for only 17% of the total SOD (p<0.001 if compared with the values recorded in normal mucosa). After immunoblotting, MnSOD was more expressed in adenocarcinoma with respect to normal mucosa (p<0.001), while CuZnSOD was similarly expressed in adenocarcinoma and healthy tissues. The SOD activity assay might provide a specific and sensitive method of analysis that allows the differentiation of healthy tissue from tumour tissue. The MnSOD to CuZnSOD activity ratio, and the ratio between these two isoforms and total SOD, presented in this preliminary study might be considered in the identification of cancerous from healthy control tissue.  相似文献

Occupational exposure to photocopiers has been indicated as being responsible for a number of health complaints, particularly effects on the respiratory, immunological, and nervous systems. In this study, we investigated oxidative and genotoxic damage in photocopier operators by assessing catalase activity (CAT), reduced vs. oxidized glutathione ratio (GSH/GSSG), level of lipid peroxidation (TBARS), damage index by Comet assay (DICA), and buccal cells with micronuclei (BCMN). Our results reveal that the TBARS levels in operators were increased (27%; p<0.05) but that no significant alterations to GSH/GSSG or CAT activity were observed. The DICA and the number of BCMN were significantly increased (134% and 100%, respectively; p<0.05) in the exposed group. There was a significant association between the time in months spent at work and DNA damage in lymphocytes (rs?=?0.720; p<0.001) and buccal cell with MN (rs?=?0.538; p<0.001). Because laser printers and photocopiers have become increasingly used, it is important to control human exposure using reliable biomarkers.  相似文献

Chilli pepper is an important vegetable in the diet of Ghanaians but there are some constraints militating against increased production notable among which is nematode infestation. A study was therefore carried out at the plant house of the Faculty of Agriculture, University for Development Studies, Nyankpala to investigate the efficacy of neem products against root-knot nematode of chilli pepper (Capsicum annum L.). The treatments which included 20 g neem leaf powder, 20 g neem cake, 20 g neem seed powder and control were arranged in a completely randomised design with four replications. Neem products significantly reduced (p < 0.05) nematode population, with 20 g of neem leaf powder recording the highest reduction. Similarly, plants treated with neem leaf powder produced a significantly lower (p < 0.05) number of root galls compared to the other treatments. Neem leaf powder could be used as a nematicide for nematodes control as a replacement for synthetic nematicides.  相似文献

Yam is an important crop which serves as a source of income to small-holder farmers as well as a foreign exchange earner. Among the constraints faced by yam farmers are pests and diseases, especially deterioration caused by microbes during storage. Since over dependence on pesticides is being discouraged, neem seed and ginger extracts were evaluated as potential control agents against rot-causing fungi. The study was conducted in the Spanish laboratory, at the Faculty of Agriculture, University for Development Studies, Nyankpala Campus. Isolations were made from rotted yam tubers sampled from the Tamale Central market with potato dextrose agar (PDA) as the growth medium. Growth inhibition of the isolates was determined by growing pure cultures on PDA plates amended with 2?ml each of ethanol and aqueous extracts of neem seed and ginger as well as carbendazim. A pathogenicity test proved that Aspergillus flavus, A. niger, A. ochraceus and Penicillium digitatum isolated from the rotted tubers were responsible for the rot. A. niger had a significantly higher (p?<?0.05) occurrence (40%) than the others. Growth inhibitions by carbendazim and ethanol extracts of neem seed and ginger were comparable but significantly higher (p?<?0.05) than the aqueous extracts of neem seed and ginger as well as the control. However, the aqueous extracts of neem seed and ginger had a significantly higher (p?<?0.05) inhibition than the control. For instance, growth inhibition of A. niger by carbendazim, ethanol neem seed and ginger extracts were 70.5, 69.6 and 65.5%, respectively, while inhibition by aqueous neem seed and ginger extracts were 33.9 and 24.8%, respectively. Since aqueous extracts of neem seed and ginger significantly inhibited (p?<?0.05) growth of the rot-causing fungi, they can be used as surface protectants of stored tubers.  相似文献

We investigated the effects of Sarcodon aspratus, Agaricus bisporus, and Lentinula edodes aqueous extracts on the tenderization of bovine longissimus dorsi muscle. Meat quality and muscle protein degradation were examined as well. Beef chunks were marinated in distilled water (control), 5% S. aspratus (SA), 5% A. bisporus (AB), or 5% L. edodes (LE) extracts. SA was shown to have a higher enzymatic activity (p < 0.001) and water-holding capacity than LE (p < 0.01). SA and AB extracts exhibited lower shear force values compared with the control (p < 0.05). SA, AB, and LE showed superior muscle proteolytic effects compared with the control. SA demonstrated the ability to degrade myosin heavy chains and actin, which was not observed after AB and LE extract treatments. This suggests that SA extract may affect tenderization. Taken together, our results show that aqueous extract of S. aspratus affects the tenderness of the bovine longissimus dorsi muscle.  相似文献

In vitro zinc uptake by human erythrocytes was studied under a range of zinc concentrations representing three different plasma zinc levels i.e., zinc deficient [0.35–0.61 ppm], zinc normal [0.74–1.59 ppm], and zinc excess [1.65–2.3 ppm]. Further, interactions of physiological levels of riboflavin, flavin adenine dinucleotide (FAD), nicotinic acid, nicotinamide adenine dinucleotide (NAD), thiamine, thiamine pyrophosphate (TPP), folic acid, and ascorbic acid with zinc uptakes were studied in independent experiments. In control experiments, as compared to the normal zinc state, the rate of change of zinc uptake over change in zinc levels was 1.6 times in the excess state and 0.12 times in the deficient state, indicating three distinct patterns. Under the zinc-deficient state, thiamine significantly enhanced the zinc uptakes (p<0.05), whereas ascorbic acid and riboflavin inhibited zinc uptakes (p<0.05). The percent hemolysis of the cells was also significantly lower in the presence of thiamine (p<0.05). Under normal and excess zinc states, the vitamin-zinc interactions were not significant. The results suggest that with erythrocytes as the vehicles, thiamine might be playing an enhancer role in uptake of zinc, whereas the action of ascorbic acid might be inhibitory for zinc uptakes under deficient zinc states.  相似文献

The predatory mite Iphiseius degenerans (Berlese) is commercially available as a biological control agent of thrips and spider mites in greenhouse crops. Developmental duration and immature survival of I. degeneransreared on nine types of food (almond pollen, apple pollen, castor bean pollen, plum pollen, sweet pepper pollen, Tetranychus urticaeKoch, Frankliniella occidentalis(Pergande), Ephestia kuehniella Zeller eggs and Artemia franciscana Kellogg cysts) and on three substrates (Multicel, sweet pepper leaf, and bean leaf) were determined in the laboratory. All experiments were carried out at 25 °C. On Multicel, mean developmental times on pollen ranged from 6.0 to 7.1 days, with the lowest value recorded on almond pollen and the highest on sweet pepper pollen. When reared on castor bean pollen significantly longer developmental times were obtained on a sweet pepper leaf compared to a bean leaf or Multicel. Developmental duration when offered T. urticaeon Multicel ranged between 6.1 and 6.9 days, on a bean leaf development was completed in 8.0 days. The longest developmental times on Multicel were recorded on Ephestia eggs (7.0 days) and on decapsulated Artemia cysts (7.5 days). No development beyond the protonymphal stage occurred in the absence of food or when encapsulated Artemia cysts or thrips larvae were offered on Multicel. On a sweet pepper leaf and a bean leaf, respectively 6.7 and 10.0% of the eggs reached adulthood when thrips larvae were provided as food; developmental times recorded here, were 9.0 and 8.3 days. Overall, immature mortality occurred mainly in the protonymphal stage and ranged from 0.0 to 36.7%. In conclusion, I. degenerans is able to feed on a variety of natural and unnatural foods, but thrips larvae and sweet pepper pollen are unfavourable food for immature development. This could compromise the establishment of this biological control agent when used against thrips in sweet pepper crops.  相似文献

Coffee is the main source of chlorogenic acid in the human diet, and it contains several chlorogenic acid isomers, of which the 5‐caffeoylquinic acid (5‐CQA) is the predominant isomer. Because there are no available data about the action of chlorogenic acids from instant coffee on hepatic glucose‐6‐phosphatase (G‐6‐Pase) activity and blood glucose levels, these effects were investigated in rats. The changes on G‐6‐Pase activity and liver glucose output induced by 5‐CQA were also investigated. Instant coffee extract with high chlorogenic acids content (37.8%) inhibited (p < 0.05) the G‐6‐Pase activity of the hepatocyte microsomal fraction in a dose‐dependent way (up to 53), but IV administration of this extract did not change the glycaemia (p > 0.05). Similarly, 5‐CQA (1 mM) reduced (p < 0.05) the activity of microsomal G‐6‐Pase by about 40%, but had no effect (p > 0.05) on glucose output arising from glycogenolysis in liver perfusion. It was concluded that instant coffee extract with high content of chlorogenic acids inhibited hepatic G‐6‐Pase in vitro, but failed to reduce the glycaemia probably because the coffee chlorogenic acids did not reach enough levels within the hepatocytes to inhibit the G‐6‐Pase and reduce the liver glucose output. Lack of reliable assays for DNA repair has largely prevented measurements of DNA repair from being included in human biomonitoring studies. Using newly developed modifications of the comet assay we tested whether a fruit‐ and antioxidant‐rich plant‐based intervention could affect base excision repair (BER) and nucleotide excision repair (NER) in a group of 102 male volunteers. BER and NER repair capacities were measured in lymphocytes before and after a dietary intervention lasting 8 weeks. The study had one control group, one group consuming three kiwifruits per day and one group consuming a variety of antioxidant‐rich fruits and plant products in addition to their normal diet. DNA strand breaks were reduced following consumption of both kiwifruits (13%, p = 0.05) and antioxidant‐rich plant products (20%, p = 0.02). Increased BER (55%, p = 0.01) and reduced NER (?39%, p < 0.01) were observed in the group consuming a wide variety of plant products. Reduced NER was also observed in the kiwifruit group (?38%, p = 0.05), but BER was not affected in this group. Here we have demonstrated that DNA repair is affected by diet and that modified versions of the comet assay can be used to assess activity of different DNA repair pathways in human biomonitoring studies. The effect of bacterial antagonism against the causal agent, Colletotrichum truncatum was assessed as a potential alternative in managing anthracnose in chilli pepper. Out of 104 contrasting bacterial colonies isolated from rhizosphere soil of a forest floor, five isolates caused the radial growth inhibition greater than 90% (Significant at p?<?0.05 level) of C. truncatum in dual cultures. Based on 16S rRNA analysis, these antagonistic bacterial isolates were identified as Pseudomonas aeruginosa, Burkholderia arboris, Burkholderia gladioli and Burkholderia rinojensis. The selected promising antagonists showed nearly 100% inhibition of the spore germination of C. truncatum in vitro. These antagonists produced antifungal compounds which are diffusible in nature. Microscopic studies of blackened fungal hyphae, which were subjected to antagonism showed many deformations such as thickening, swelling and malformation. In vivo study revealed that C. truncatum inoculated chilli pepper seeds treated with the five antagonists significantly inhibited the incidence of seed colonisation (p?<?0.05) by the pathogen. At the post emergence stage, the survival percentages and vigour indices of all the antagonists’ treatments, except B. rinojensis strain 1, were significantly higher compared to the untreated control. The efficacy of the selected antagonists in managing anthracnose fruit rot was 100% at the colour breaking stage of chilli pepper fruits. These bacterial antagonists had a negative effect on C. truncatum spore attachment and subsequent colonisation on chilli pepper leaves except in the treatment of B. arboris. The results of in-vitro and in-vivo studies, suggest that the screened antagonistic bacterial isolates are potential biocontrol agents and need to be further studied for the biochemical basis of their activity against C. truncatum.  相似文献

The fluoride (F), calcium (Ca), magnesium (Mg), zinc (Zn), copper (Cu), and phosphorus (P) content in potable water and food samples (cereals, pulses, and vegetables) from endemic and nonendemic villages for fluorosis were analyzed. It was found that the F content in water was significantly higher (p<0.01) in endemic villages (4.20±1.6 ppm) than control villages (0.63±0.15 ppm), whereas the Ca, Cu, and Mg contents were found to be significantly lower (p<0.05) in endemic villages compared to control villages. However, there was no significant difference in Zn and P contents between the villages. Foods (cereals, pulses, and vegetables) grown in endemic villages contained significantly higher (p<0.01) fluoride content as compared to control villages. There was no significant difference in Ca, Mg, P, and Zn contents in food grown in endemic and control villages. Copper content in cereals (p<0.05), pulses (p<0.01), and vegetables (p<0.01) in endemic villages was found significantly higher as compared to control villages. The overall prevalence of dental fluorosis in six endemic villages was 97.4% in boys and 96% in girls, which was significantly higher (p<0.01) than that of control villages, where it was 10.5% in boys and 8.3% in girls. The prevalence of dental fluorosis was positively correlated (r=0.125, p<0.01) to fluoride and negatively correlated to Ca and Cu content in drinking water in endemic villages.  相似文献

We hypothesized that dexanabinol can prevent neuronal death by protecting neuronal lysosomes from nitric oxide (NO)-mediated toxicity, and in turn, by suppressing the release of cathepsins during cerebral ischemia. Focal cerebral ischemia was induced in two sets of animals by permanent middle cerebral artery occlusion. The first set was used to monitor NO concentration and cathepsin activity, while the second was used for histological examination with hematoxylin and eosin, and TUNEL staining. In post-ischemic brain tissue, NO content and cathepsin B and L activity increased (p < 0.05). Dexanabinol treatment reduced NO concentration and cathepsin activity to the control level (p > 0.05). The number of eosinophilic and apoptotic neurons increased in the post-ischemic cerebral cortex (p < 0.05). However, dexanabinol treatment lowered both of these (p < 0.05). We conclude that dexanabinol might be a useful agent for the treatment of stroke patients.  相似文献

This article deals with the features of the evaluation of changes in the functional state during the professional activity of operators. We estimated the dynamics of the functional state and efficiency of operators under the effect of aircraft noise and individual psychophysiological characteristics. We also investigated changes in the functional state of operators depending on their psychological characteristics. It was found that changes in the functional state of the organism in operators with high levels of anxiety, low degrees of extraversion, and high emotional lability were more pronounced in the modeling of operator activity with exposure to aircraft noise, as evidenced by significant changes in psychophysiological parameters. After 60 minutes of noise stress, the latent period of a simple and a complex sensorimotor reaction in introverts increased by 9.1% (p < 0.05) and 8.1% (p < 0.05), respectively. In the group of subjects with high levels of anxiety, there was a decrease in the general state, activity, and mood by 18.9% (p < 0.01), 20.9%, and 14.9% (p < 0.05), respectively, and a decrease in the accuracy of reaction to a moving object by three times (p < 0.05). The latent period of simple sensorimotor reaction in emotionally labile subjects after the exercise was increased by 15% (p < 0.05), and the reaction time on a moving object was increased by 8.9% (p < 0.05) as compared with the group of emotionally stable subjects. Thus, the data suggest that changes in the functional state of operators depends on the level of extraversion, personal anxiety, and emotional lability, and does not depend on the level of behavioral regulation, which must be taken into consideration during the assessment of functional state.  相似文献

Nickel (Ni) is an environmental pollutant towards which human exposure can be both occupational (mainly through inhalation) and dietary (through water and food chain-induced bioaccumulation). The aim of this study was to investigate the effects of short-term Ni-administration (as NiCl2, 13 mg/kg) on the adult rat whole brain total antioxidant status (TAS) and the activities of acetylcholinesterase (AChE), Na+,K+-ATPase, and Mg2+-ATPase; in addition, the potential effect of the co-administration of the antioxidant L-cysteine (Cys, 7 mg/kg) on the above parameters was studied. Twenty-eight male Wistar rats were divided into four groups: A (saline-treated control), B (Ni), C (Cys), and D (Ni and Cys). All rats were treated once daily with intraperitoneal injections of the tested compounds, for 1-week. Rats were sacrificed by decapitation and the above-mentioned parameters were measured spectrophotometrically. Rats treated with Ni exhibited a significant reduction in brain TAS (-47%, p < 0.001, BvsA) that was efficiently limited by the co-administration of Cys (-4%, p > 0.05, DvsA; +83%, p < 0.001, DvsB), while Cys (group C) had no effect on TAS. The rat brain AChE activity was found significantly increased by both Ni (+30%, p < 0.001, BvsA) and Cys (+62%, p < 0.001, CvsA), while it tended to adjust to control levels by the co-administration of Ni and Cys (+13%, p < 0.001, DvsA; −13%, p < 0.001, DvsB). The activity of rat brain Na+,K+-ATPase was significantly decreased by Ni-administration (−49%, p < 0.001, BvsA), while Cys supplementation could not reverse this decrease (-44%, p < 0.001, DvsA). The activity of Mg2+-ATPase was not affected by Ni-administration (−3%, p > 0.05, BvsA), but was significantly reduced when combined with Cys administration (−17%, p < 0.001, DvsA). The above findings suggest that Ni short-term in vivo administration causes a statistically significant decrease in the rat brain TAS and an increase in AChE activity. Both effects can be, partially or totally, reversed to control levels by Cys co-administration; Cys could thus be considered (for future applications) as a potential neuroprotective agent against chronic exposure to Ni. The activity of Na+,K+-ATPase that was inhibited by Ni, could not be reversed by Cys co-administration. The matter requires further investigation in order to fully elucidate the spectrum of the neurotoxic effects of Ni.  相似文献

Abstract

The protecting ability of the Piper betle leaves-derived phenol, allylpyrocatechol (APC) against AAPH-induced membrane damage of human red blood cells (RBCs) was investigated. Compared to control, AAPH (50 mM) treatment resulted in significant hemolysis (55%, p < 0.01), associated with increased malondialdehyde (MDA) (2.9-fold, p < 0.001) and methemoglobin (6.1-fold, p < 0.001) levels. The structural deformation due to membrane damage was confirmed from scanning electron microscopy (SEM) images and Heinz bodies formation, while the cell permeability was evident from the K+ efflux (28.7%, p < 0.05) and increased intracellular Na+ concentration (8%, p < 0.05). The membrane damage, due to the reduction of the cholesterol/phospholipids ratio and depletion (p < 0.001) of ATP, 2,3-DPG by ?44–54% and Na+–K+ ATPase activity (43.7%), indicated loss of RBC functionality. The adverse effects of AAPH on all these biochemical parameters and the resultant oxidative hemolysis of RBCs were significantly reduced by pretreating the cells with APC (7 μM) or α-tocopherol (50 μM) for 1 h, prior to incubation with AAPH.  相似文献

A popular sequential extraction procedure (Tessier et al. 1979) designed t o extract metals partitioned in various sediment phases, was evaluated for its selectivity. Amorphous FeOOH, FeS, and FeS2 were added separately to natural lake sediments and sequentially extracted. The selectivity of the sequential procedure for the added solid phases was evaluated by determining the difference in the mass of Fe extracted from treated and control sediments. In the experiments where sulfide minerals were added, total S was measured in the residual solids in order to confirm selectivity of the method. Concentrations of total carbon remaining in the solid phase after each extraction step were also measured to determine the selectivity of the sequential procedure for carbon. The procedure was moderately selective for Fe added as FeOOH; a mean of 77 ± 12% (p < 0.05) of the Fe added was extracted in the step designed to reduce Fe-Mn oxyhydroxides. In experiments where FeS was added, a mean of 69 ± 11% (p < 0.05) of the Fe added as FeS was extracted in the fraction designed to oxidize sulfides and organic matter. Approximately 25% of the Fe added as FeS may have been extracted prematurely. Although less precise, total S analyses confirmed that much of the FeS was extracted in the oxidation step, yielding 104 ± 87% (p < 0.05) of the S added as FeS. The procedure was highly selective for FeS2; 92 ± 14% (p < 0.05) of the Fe added as pyrite was extracted in the sulfide extraction step. Extraction of 80 ± 54% (p < 0.05) of S added as pyrite confirmed that FeS2 were selectively extracted in the sulfide extraction step. Carbon in the sediments was also selectively extracted in the oxidation step (77 ± 2.4% of total C; p < 0.05). The protective effect of black cumin (Nigella sativa=NS) on cadmium-induced oxidative stress was studied in rats. The rats were randomly divided into three experimental groups: A (conrol), B (Cd treated), and C (Cd+NS treated), each containing 10 animals. The Cd-treated and Cd+NS-treated groups were injected subcutaneously daily with CdCl2 dissolved in isotonic NaCl in the amount of 2 mL/kg for 30 d, resulting in a dosage of 0.49 mg Cd/kg/d. The control group was injected with only isotonic NaCl (2 mL/kg/d) throughout the experiment (for 30 d). Three days prior to induction of CdCl2, the Cd+NS-treated group received a daily intraperitoneal injection of 0.2 mL/kg NS until the end of the study. Cd treatment increased significantly the malondialdehyde levels in plasma and erythrocyte (p<0.01 and p<0.05, respectively) and also increased significantly the antioxidant levels (superoxide dismutase, glutathione peroxidase, and catalase) (p<0.05) compared to the control group. Cd+NS treatment decreased significantly the elevated malondialdehyde levels in plasma and erythrocyte (p<0.01 and p<0.05, respectively) and also reduced significantly the enhanced antioxidant levels (p<0.05). Cd treatment increased significantly the activity of iron levels (p<0.05) in the plasma compared to the control group. Cd+NS treatment decreased the activity of iron levels (p<0.05) in the plasma compared to the Cd-treated group. In the control group with no treatment, histology of erythrocytes was normal. In the Cd-treated group, there were remarkable membrane destruction and hemolytic changes in erythrocytes. In the Cd+NS treated group, these changes were less than in the Cd-treated group. Our results show that N. sativa exerts a protective effect against cadmium toxicity.  相似文献

Objective Hypercholesterolemia (HC) has been associated with impairment of vascular and myocardial functions. As HC could generate an alteration in the oxidative status, we studied the effects of a 1-month cholesterol diet on cardiovascular oxidative stress. Methods and Results New Zealand rabbits received cholesterol (1%) or normal chow for 1 month. At 30 days, superoxide anion levels, assessed by ESR spectroscopy, NAD(P)H oxidase (NOX) activity, and dihydroethidium (DHE) staining of aortas were higher in the cholesterol-fed (CF) group compared with control (respectively, 4.0 ± 0.6 Arbitrary Units/mg (AU/mg) vs. 2.6 ± 0.3, p < 0.05; 4231 ± 433 vs. 2931 ± 373 AU/mg, p < 0.05; 21.4 ± 1.2 vs. 12.9 ± 1.7% fluorescence/mm2, p < 0.001). NOX gp91phox and p67phox expression in the aortas were higher in the CF group vs. control (1.5 ± 0.2 vs. 0.5 ± 0.2, p < 0.001; 0.9 ± 0.2 vs. 0.3 ± 0.2, p < 0.05). The endothelium-dependent relaxation evaluated on the iliac arteries was higher in control than in the CF group (64.8 ± 10.1 vs. 13.1 ± 3.70%, p < 0.001). The cardiac diastolic pressure estimated on isolated hearts was higher in the CF group than in control (21.1 ± 4.1 vs. 10.3 ± 1.4 mmHg, p < 0.05) after 60 min of ischemia. Conclusions Hypercholesterolemia induced increased levels of superoxide in the aortas and a higher expression of NOX subunits, associated with altered vasorelaxation. The increased diastolic pressure observed in hearts, consistent with a post-ischemic contractile dysfunction might be mediated by the production of superoxide.  相似文献
