Vitellogenin is a phospholipoglycoprotein precursor of egg yolk. In mature female fish, vitellogenin is synthesized and secreted by the liver in response to circulating estrogens. Vitellogenin is normally undetectable in the blood of male fish, but can be induced by exposure to compounds possessing estrogenic activity. Thus, the presence of vitellogenin in blood of male fish can serve as a useful biomarker for assessing previous exposure to estrogenic compounds. In the present study, we report identification and purification of vitellogenin in the mosquitofish (Gambusia affinis). Anti-vitellogenin immune serum was generated and used to develop an immunoblot assay for detection of vitellogenin. A combination of immunoblotting and densitometric scanning was used to assess the time- and dose-dependent effects of 17alpha-ethynylestradiol on vitellogenesis in male G. affinis. The results indicate that changes in the level of vitellogenin in mosquitofish blood can be reliably detected by the immunoblot assay, and that the mosquitofish may be a useful bioindicator organism for detecting estrogenic contamination of the aquatic environment.  相似文献

There is increasing concern about man-made chemicals in the aquatic environment that mimic oestrogens because they may disrupt reproductive function. Vitellogenin, a precursor of egg-yolk in fish and other oviparous animals, may be used as a biomarker for “oestrogen” exposure. This study investigated the use of a radioimmunoassay developed to carp (Cyprinus carpio) vitellogenin to measure vitellogenin in other species of fish, especially cyprinids that would be of value for field and laboratory studies on oestrogenic xenobiotics. Of the nine families of fish studied, only vitellogenin from cyprinids (to which the carp belongs) showed good cross-reactivity in the carp vitellogenin radioimmunoassay. Vitellogenin from cyprinids native to Europe that cross reacted in the carp vitellogenin radioimmunoassay included: bream (Abramis brama), roach (Rutilus rutilus), rudd (Scardinius erythropthalmus), gudgeon (Gobio gobio) and minnow (Phoxinus phoxinus). Vitellogenin from cyprinids used widely in ecotoxicology that cross reacted in the carp vitellogenin radioimmunoassay included: fathead minnow (Pimephales promelas), zebrafish (Brachydanio rerio) and goldfish (Carassius auratus). In the cyprinids studies, the concentrations of vitellogenin in mature females were between a few hundred and a thousand microgram per millilitre. Concentrations of plasma vitellogenin in immature females were always greater than 200 ng·m-1, whereas in males (with the exception of the fathead minnow) plasma vitellogenin concentrations were less than 20 ng·ml-1 (and generally, much lower). The results suggest that the structure of vitellogenin is highly conserved within the cyprinid family and that the carp vitellogenin radioimmunoassay may be used to measure the concentrations of vitellogenin in plasma from a wide variety of cyprinids.  相似文献

A monoclonal antibody to vitellogenin of channel catfish (Ictalurus punctatus) was made, and its specificity was demonstrated using Western blots of serum from female fish, estradiol-treated male fish, untreated male fish, vitellogenin purified by three different methods and egg extracts. 2. An enzyme-linked immunosorbent assay (ELISA), using this monoclonal antibody, detected vitellogenin in the plasma of 59 out of 60 untreated 17-24-month-old male channel catfish with a mean concentration of 338 micrograms/ml and a maximum concentration of 4240 micrograms/ml. 3. The effect of the juvenile hormone analogue hydroprene on the appearance in the haemolymph of the yolk-protein precursor vitellogenin in male Diploptera punctata has been assessed using rocket immuno-electrophoresis. Vitellogenin was induced in a dose-dependent fashion, with a minimum dose of 10 μg hydroprene required for its appearance. Implantation of male corpora allata into male and female hosts also resulted in the appearance of vitellogenin in the haemolymph of the hosts, with females showing apparent greater sensitivity to the implantation.The identity of vitellogenin in male haemolymph was further confirmed using Ouchterlony double immunodiffusion and SDS polyacrylamide gel electrophoresis of immunoprecipitates. The electrophoretic pattern of immunoprecipitated haemolymph from females and hydroprene-treated males was similar, further confirming that the immunoprecipitable product was in fact vitellogenin.  相似文献

The fathead minnow (Pimephales promelas) has been proposed as a model species for assessing the adverse effects of endocrine-disrupting chemicals (EDCs) on reproduction and development. The purpose of these studies was to develop baseline reproductive biology and endocrinology data for this species to support interpretation of tests with potential EDCs. Pairs of reproductively-active fathead minnows (n=70) were evaluated with respect to reproductive cyclicity in terms of spawning interval and fecundity. The mode and mean (+/-SE) spawning intervals for the fish in this study were 3.0 and 3.7+/-0.1 days, respectively. The mean number of eggs produced per spawn was 85+/-2.8. Animals were sacrificed at periodic intervals during the established spawning cycle and measurements made of gonadal condition (gonadosomatic index [GSI], histopathology) and plasma concentrations of vitellogenin and sex steroids (beta-estradiol, testosterone, 11-ketotestosterone). The GSI in females varied significantly as a function of spawning interval, with the largest values occurring day 2 post-spawn, just prior to the interval of maximum spawning activity. Plasma beta-estradiol concentrations in females also varied significantly relative to peak values in the GSI and spawning activity. Vitellogenin concentrations in the female, and male GSI and steroid concentrations did not vary significantly relative to position in the spawning cycle. Concentrations of beta-estradiol in females and 11-ketotestosterone in males were positively correlated with testosterone concentrations.  相似文献

Vitellogenin synthesis is induced in juvenile rainbow trout (Salmo gairdneri) and juvenile sea trout (Salmo trutta) by estradiol-17 beta. A purification procedure for vitellogenin from trout plasma by precipitation with MgCl2-EDTA and subsequent anion exchange chromatography on DEAE-Sephacel is described. The total lipid contents of purified rainbow trout and sea trout vitellogenins are 18 and 19%, respectively. Approximately 2/3 of the lipids are phospholipids, while the remainder consists of triglycerides and cholesterol. Phosphorus determinations on delipidated vitellogenin yield a phosphorus content of 0.63% in rainbow trout and 0.58% in sea trout vitellogenin. Native (dimeric) vitellogenins from rainbow trout and sea trout both have an apparent molecular weight of 440,000, when estimated by gel filtration on Sepharose 6B.  相似文献

Anaesthesia with MS-222 followed by intra-peritoneal (ip) injection (with a 10% ethanol in corn oil carrier) of fathead minnow either as one or three (weekly) treatments did not affect survival, behaviour or secondary sexual characteristics of the fish. Fecundity of the fish, as indicated by fertility and hatching success, was also unaffected. Gonadal condition (relative gonad mass, histopathology) was not altered in either sex. Male and female plasma sex steroids (β;-oestradiol, testosterone, 11-ketotestosterone) and male vitellogenin concentrations were not significantly affected by the treatments. Females subjected to either ip treatment regime had significantly higher plasma vitellogenin concentrations than control females. However, based on previous data, this difference did not appear to be treatment-related. Overall, exposure of fathead minnows to chemicals via the ip route should not confound the interpretation of toxicity tests with potential endocrine disrupting chemicals.  相似文献

The detection of vitellogenin, a yolk precursor protein, may serve as a biomarker for exposure to environmental oestrogens as its induction by xenobiotic oestrogens in the immature and male fish has been reported repeatedly. In the present work, juvenile brown trout were injected with oestradiol (5 g g–1 body weight oestradiol benzoate) in order to assess the induction and organ distribution of vitellogenin by means of immunohistochemistry. In addition, brown trout collected from Swiss rivers were analysed. Vitellogenin was detected in the oestradiol-injected juvenile trout but not in uninjected controls. The presence of vitellogenin was also demonstrated in a male and an immature feral brown trout from one of two locations downstream of three sewage treatment plants. In contrast, no positive staining was found in livers of trout upstream of the respective plants. The results demonstrate the suitability of immunohistochemistry for monitoring feral fish fo r the presence of vitellogenin production.  相似文献

This study was designed to determine the estrogenic effect of the phytoestrogen genistein on several measures of endocrine function in adult Japanese medaka (Oryzias latipes) relative to 17-beta-estradiol. Adult animals of both sexes were exposed to 75, 750 and 30,000 ng/fish (average fish weight equals 0.26 g) of genistein by i.p. injection, with a positive control group treated with 300 ng/fish of 17-beta-estradiol, while a negative control group received a vehicle-only (corn oil) injection. Content of vitellogenin, the yolk glycoprotein made in the liver in response to estradiol stimulation, was measured using Western blots. Circulating estradiol and testosterone levels were measured using a steroid-enzyme immunosorbant assay. The ability of ovaries and testes to synthesize and release estradiol and testosterone was determined by ex vivo incubation of gonads with 25-hydroxycholesterol. Vitellogenin, while induced by 17-beta-estradiol, was not increased in the liver of individuals treated with genistein. In females, genistein treatment at 750 and 30,000 ng increased the estradiol production of ovaries more than the 17-beta-estradiol treatment. In males, genistein treatment resulted in decreased testosterone production from ex vivo testis and a comparable reduction in circulating testosterone level. The changes in vitellogenin, circulating steroids and ex vivo steroidogenesis in medaka in response to genistein are similar to that of 17-beta-estradiol. However, some endpoints are more sensitive to estradiol treatment (vitellogenin), while others are more sensitive to genistein (male testosterone and ovarian estrogenesis).  相似文献

Sexual maturation in sea trout ( Salmo trutto L.) running up the River Calonne (Normandy, France), at the finnock stage, is characterized. Autopsy of 118 finnock in summer showed that 87 ± 7% aged 2+ and 33 ± 15% aged 1 + were undergoing gametogenesis. In autumn all the fish were maturing. The analysis of plasma levels of 11-ketotestosterone, 17β-oestradiol, maturational gonadotropin and vitellogenin showed that only females in vitellogenesis can be well identified in summer (plasma vitellogenin greater than 0.30 mg ml-1). The morphological recognition of females among 134 autumn finnock and the ELISA assay of plasma vitellogenin of 814 summer finnock, demonstrated that vitellogenic females represented 12 ± 2%0 of the 1 + individuals and 46 ± 2% of 2+ fish captured during the 1985 run. The question of a possible relationship between the salinity tolerance ofjuvenile trout migrating towards the sea in the spring and their age on first migration in the river is discussed.  相似文献

Vitellogenin (Vg) of the barfin plaice Liopsetta pinnifasciata was isolated and purified. Using SDS-PAGE, barfin plaice Vg was dissociated into polypeptides with molecular masses of 180, 98, 70, 52, 41, and 37 kDa. Rabbit polyclonal antibodies to this protein were obtained and a specific highly sensitive diagnostic enzyme-linked immunosorbent assay was developed for Vg detection in the blood plasma of the barfin plaice, which was tested on fish from natural habitats. The plasma Vg concentration in L. pinnifasciata from a contaminated location of Amursky Bay (Sea of Japan) in November varied from 5.295 to 28.367 mg/ml in females and from an undetectable level to 0.957 mg/ml in males. The presence of Vg in the blood plasma of male L. pinnifasciata from Amursky Bay, which was detected for the first time, is indicative of ecosystem pollution by chemicals with estrogenic activity.  相似文献

This study was performed to improve the purification of Xenopus vitellogenin and establish the radioimmunoassay. The procedure of purification consisted of ammonium precipitation, DEAE-Sephadex chromatography and Sephadex G-200 gel chromatography. Using this procedure, 934 mg vitellogenin was purified from 49 ml of estradiol treated female Xenopus plasma (about 19 mg/ml). Vitellogenins purified from male and female plasma after a single injection of estradiol showed good correspondence in electrophoretic patterns and amino acid compositions, indicating that vitellogenin synthesis in the male occurs in four different genes as in the female. The radioimmunoassay for vitellogenin was established using an antibody in the plasma obtained from rabbits injected with purified Xenopus female vitellogenin. The titer was 20,000 times dilution of the plasma, and the minimum detectable amount of vitellogenin was 0.1 microgram. The cross-reactivity of this antibody with newt vitellogenin was about 65% and that of chick 6%. The cross-reaction was also observed in female bullfrog plasma. Vitellogenin content was increased gradually during the first 6 days after injection of estradiol in female and the elevated level of vitellogenin dropped afterward.  相似文献

When investigating insecticide resistance of pest insects, for example, the pollen beetle Meligethes aeneus, it is relevant to differentiate toxicological and molecular genetic data between male and female specimens. A molecular sex determination method would allow resistance testing to be run without prior sorting of the samples. A one-step quantitative RT-PCR method for quantification of the yolk protein vitellogenin expression in the pollen beetle was established. The expression level of vitellogenin relative to tubulin was determined. Pollen beetles were tested at different time points during their development to determine if vitellogenin is a reliable molecular marker for detection of sexually mature females. The differentiation between females and males by relative expression of vitellogenin to tubulin is conditional regarding the life cycle. Sexually mature females and males could easily be distinguished, whereas immature specimens could not be seperated. Vitellogenin expression is a successful marker for identification of sexually mature pollen beetles. Females from the spring populations showed vitellogenin expression when the temperature was above 10.2°C. Further, detailed observations of vitellogenin throughout the spring indicated a strong relationship between daily temperatures and vitellogenin expression, which is an indicator of oviposition ability.  相似文献

Vitellogenin was isolated by affinity chromatography and gel filtration from landlocked Atlantic salmon plasma. Vitellogenin was labelled with iodine-131 using iodogen and an homologous radioimmunoassay was developed. There was poor immunological cross-reactivity with vitellogenin or plasma from other teleosts. Parallelism of the vitellogenin standard to the displacement by plasma of vitellogenic salmon allowed the assay to be used to evaluate the seasonal concentration profile of vitellogenin in female adult salmon. Extracts of liver or ovary from female Atlantic salmon also yielded displacements parallel to the vitellogenin standard in the assay.  相似文献

Various adverse effects related to landfill leachate have made leachates an important issue in past decades, and it has been demonstrated that landfill leachate is an important source of environmental estrogens. In this study, we employed chemical analysis of some already evaluated estrogenic substances, in combination with a bioassay using several specific biomarkers (e.g., plasma vitellogenin and sex steroids, enzyme activity of gonad gamma-glutamyl transpeptidase, and gonadosomatic index) to evaluate the estrogenic activities in outlets from different stages of the leachate treatment process. The results indicated that 5 environmental estrogens (4-t-octylphenol, bisphenol A, di-ethyl phthalate, di-n-butyl phthalate, and diethylhexyl phthalate) were detected by a gas chromatography-mass spectrometry, and the concentrations in leachate samples were 6153 ng/L, 3642 ng/L, 2139 ng/L, 5900 ng/L, and 9422 ng/L, respectively. Leachate (1∶200 diluted) induced the synthesis of plasma vitellogenin and led to decreased enzyme activity of gonad gamma-glutamyl transpeptidase and gonadosomatic index in male goldfish (Carassius auratus) after a 28-day exposure, while increased circulating 17β-estradiol level was also observed in males exposed to treated effluent. Although the target EEs were partially removed with removal rates varying from 87.2% to 99.77% by the “membrane bioreactor+reverse osmosis+aeration zeolite biofilter” treatment process, the treated effluent is still estrogenic to fish. The method combined chemical techniques with the responses of test organisms allowing us to identify the group of estrogen-like chemicals so that we were able to evaluate the overall estrogenic effects of a complex mixture, avoiding false negative assessments.  相似文献

The effect of juvenile hormone (JH) III on the hemolymph composition of vitellogenin was examined in Perillus bioculatus. Adult females were treated topically with JH III, and the premature presence of vitellogenin in the hemolymph was then detected using electrophoresis and Western blot analyses. JH III treatment resulted in a dose-dependent early production of vitellogenin that was detectable 48 h before vitellogenin was present in non-treated insects. Vitellogenin was not observed in the hemolymph of JH III-treated adult males. The techniques reported here may be useful for the detection, isolation and characterization of compounds with JH-like activity in P. bioculatus and other species of Heteroptera (which are thought to have JH-like substances other than the JHs with known chemical identity). These same techniques may also provide a method for researchers to investigate the interactions of JH-like compounds and other substances, such as ecdysteroid, in the regulation of vitellogenesis in Heteroptera.  相似文献

Vitellogenin derived from the blood of estrogen-treated Pleurodeles waltl was identified by immunochemical and electrophoretic analyses, using an antiserum against plasma vitellogenin isolated by dimethylformamide precipitation. Pleurodeles vitellogenin migrates as four bands on native PAGE, designated alpha-, beta-, gamma- and delta- VTG, with apparent mol. wts of 250,000, 270,000, 280,000 and 520,000 respectively. In the plasma, from estrogen-treated males like from ovariectomized estrogen-treated females, an additional band (mu-VTG) was found by native PAGE, never observed in estrogen-treated female plasma. It has a mol. wt of about 380,000 and shows complete immunological cross-reactivity with the vitellogenin antiserum. At least two polypeptides, termed VTG-I and VTG-II (mol. wt = 180,000 and 210,000) were identified by SDS-PAGE. Rocket immunoelectrophoresis displays three distinct precipitate lines indicating major immunological differences between the plasma vitellogenins.  相似文献

Estrogenic chemicals such as alkylphenols (APs) have been shown to disrupt the reproductive system of male fish. In the present study, the effects of the estrogenic chemical octylphenol (100 microg g(-1)) and 17 beta-estradiol on the semen production and the biochemical composition of the seminal fluid of the viviparous eelpout (Zoarces viviparus) were investigated at the time of spawning. After 10 days of octylphenol or estrogen treatment, vitellogenin (Vtg) synthesis was induced as indicated by increased plasma vitellogenin concentration. In accordance with the increased vitellogenin concentration, hepatosomatic index (HSI), total protein concentration, and total calcium concentration were also increased, and free amino acids concentration was decreased in blood plasma. Octylphenol treatment caused a decrease in the gonadosomatic index (GSI) and the milt volume and an increase in the spermatocrit. The histological examination revealed that octylphenol affected the normal lobular structure, including the Sertoli cells. In the majority of the octylphenol-treated fish, trapped sperm cells were observed in parts of the seminiferous lobules and the sperm ducts. The biochemical composition of the seminal fluid was also affected by the octylphenol or estrogen. The seminal plasma concentrations of magnesium, calcium, and total protein were elevated, and the concentration of free amino acids was reduced in the treated fish. This study indicates that octylphenol inhibits the seminal fluid production and changes its biochemical composition in eelpouts.  相似文献

Administration of estradiol-17beta to male Xenopus laevis induces synthesis and secretion by the liver of the egg yolk precursor protein vitellogenin. RNA extracted from livers of estradiol-17beta-treated Xenopus laevis directs the synthesis of the entire 200,000-dalton vitellogenin monomer in a cell-free protein synthesizing system derived from rabbit reticulocytes. Vitellogenin synthesized in vitro was isolated and quantitated by indirect immunoprecipitation and identified by comparison to authentic [14C]vitellogenin. The in vitro product and [14C]vitellogenin co-migrate on electrophoresis in sodium dodecyl sulfate-polyacrylamide gels and they exhibit identical immunoprecipitation curves. Xenopus laevis vitellogenin messenger RNA has a sedimentation coefficient of approximately 30 S in sucrose density gradients. It can be purified approximately 60-fold from cell RNA by poly(U)-Sepharose chromatography and therefore appears to contain a polyadenylate sequence. Vitellogenin mRNA and vitellogenin synthesis in vivo could not be detected in unstimulated male Xenopus laevis. The relative rate of vitellogenin synthesis and the level of vitellogenin mRNA were determined at various times following the administration of estradiol-17beta. Vitellogenin synthesis is maximal 12 days after estradiol-17beta administration when it comprises approximately 70% of cell protein synthesis. The level of vitellogenin mRNA and the intracellular rate of vitellogenin synthesis exhibit a close correspondence from 4 to 16 days after administration of estradiol-17beta.  相似文献
