【目的】探究不同生境巨菌草内生固氮菌群落组成多样性及其分异规律。【方法】采用高通量测序固氮酶nif H标靶基因方法,研究了我国6个典型地区的巨菌草内生固氮菌群,包括福建闽侯县、新疆墨玉县、内蒙古阿拉善左旗、青海贵德县、甘肃安定区、海南那大镇,结合地理气候因子统计,分析了固氮菌多样性的环境驱动机制。【结果】共获得64122条nif H基因的有效序列,640个OTUs,归属于6个门、10个纲、17个目、24个科、33个属和39个种。不同地区巨菌草中优势内生固氮菌群的种类和丰度存在较大的差异。在门水平上,福州闽侯县、甘肃安定区、新疆墨玉县、内蒙古阿拉善左旗和青海贵德县5个地区的优势菌门均为变形菌门,海南那大镇的优势菌门为变形菌门和蓝藻菌门;属水平上,不同地区巨菌草最优势内生固氮菌类群分别为:福州闽侯县(变形菌门中未定属,80.56%);新疆墨玉县(变形菌门中未定属,33.14%);内蒙古阿拉善左旗(变形菌门中未定属,76.23%);甘肃安定区(α-变形菌纲中的未定属,53.78%);海南那大镇(变形菌门中未定属,38.37%);青海贵德县(变形菌门中未定属,46.12%)。Alpha多样性和Beta多样性分析表明,不同地区巨菌草内生固氮菌群落的多样性存在较大的差异,海南那大镇样本中巨菌草各类内生固氮菌群的多样性及丰富度最高,福建闽侯县样本中巨菌草各类内生固氮菌群的多样性及丰富度最低。典范对应分析(CCA)结果表明,年均降雨量和年均气温是影响巨菌草内生固氮菌群变化的主要因素,其次是土壤有机质、土壤全氮和土壤p H。【结论】不同地区巨菌草内生固氮菌群落的组成及丰度存在着较大的差异,海南那大镇巨菌草内生固氮菌群的种类及相对丰度较高,本研究可为巨菌草内生固氮菌群的资源开发及其固氮微生物肥料的菌种选育和生产应用提供理论支持。  相似文献

During a survey of nitrogen-fixing Burkholderia associated with sugarcane in Tamil Nadu, some endophytes were isolated on PCAT medium. Isolation was based on the use of the selective PCAT medium. Four isolates were studied, all belonging to the genus Burkholderia. One of them, MG43 was consistently more active in reducing acetylene and was identified as Burkholderia vietnamiensis. This isolate was used to inoculate micro-propagated sugarcane plantlets in a comparison with two other diazoptrophs, viz. Gluconacetobacter diazotrophicusT and Herbaspirillum seropedicaeT. Inoculated plants and uninoculated controls were used in a pot experiment followed by two field experiments under different rates of nitrogen fertilisers. MG43 and G. diazotrophicus performed best in sugarcane, their natural host. Biomass increase due to MG43 inoculation reached 20% in the field. Inoculated plants were heavily colonised by the inoculated bacterium (up to 115,000 CFU g−1 root fresh weight). Inoculation by a combinaison of the three strains performed less well than inoculation by a single MG43 suspension. Ecological implications are discussed, as well as the potential of these bacteria to provide a feasible alternative to higher N fertilisers rates in a low input and long term sustainable rural economy.  相似文献

【背景】菊科(Asteraceae)外来入侵植物欧洲千里光(Senecio vulgaris L.)来源于欧洲,广泛分布于我国西南和东北地区,在湖北高海拔山区也有分布。在入侵过程中,内生细菌可能在其获取氮磷营养方面起到了一些关键性作用。【目的】探究欧洲千里光内生固氮菌和溶磷菌的多样性和功能,为理解其入侵机制及防治提供参考。【方法】选择来自6个不同种群的种子,萌发后转移到花盆生长6-8周,并从每个种群中各挑选9株生长情况良好的植株,对其叶片和根组织表面进行消毒处理。使用基于nifH基因(固氮功能基因)的高通量测序方法对植物的固氮微生物群落结构和多样性进行研究。通过涂布平板法和平板划线法,在固体无氮培养基(Ashby)和无机磷培养基(inorganic phosphate, NBRIP)上对植物内生菌进行分离、纯化,对纯化的固氮菌株和溶磷菌株进行16S rRNA基因测序。采用钼锑抗比色法分析纯化溶磷菌株的溶磷能力。【结果】基于nifH基因的内生菌高通量测序结果表明,欧洲千里光叶样本中固氮菌多样性显著高于根样本;固氮菌群落中丰度最高的属是慢生根瘤菌属(Bradyrhizobium,30.9%...  相似文献

We isolated and characterized novel insoluble phosphate (P)-solubilizing bacteria tolerant to environmental factors like high salt, low and high pHs, and low temperature. A bacterium M6 was isolated from a ginseng rhizospheric soil and confirmed to belong to Burkholderia vietnamiensis by BIOLOG system and 16S rRNA gene analysis. The optimal cultural conditions for the solubilization of P were 2.5% (w/v) glucose, 0.015% (w/v) urea, and 0.4% (w/v) MgCl2·6H2O along with initial pH 7.0 at 35°C. High-performance liquid chromatography analysis showed that B. vietnamiensis M6 produced gluconic and 2-ketogluconic acids. During the culture, the pH was reduced with increase in gluconic acid concentration and was inversely correlated with P solubilization. Insoluble P solubilization in the optimal medium was about 902 mg l−1, which was approximately 1.6-fold higher than the yield in NBRIP medium (580 mg l−1). B. vietnamiensis M6 showed resistance against different environmental stresses like 10–45°C, 1–5% (w/v) salt, and 2–11 pH range. The maximal concentration of soluble P produced by B. vietnamiensis M6 from Ca3(PO4)2, CaHPO4, and hydroxyapatite was 1,039, 2,132, and 1,754 mg l−1, respectively. However, the strain M6 produced soluble P with 20 mg l−1 from FePO4 after 2 days and 100 mg l−1 from AlPO4 after 6 days, respectively. Our results indicate that B. vietnamiensis M6 could be a potential candidate for the development of biofertilizer applicable to environmentally stressed soil.  相似文献

【背景】植物内生菌长期与宿主共生,对宿主生长发育产生影响。葛根作为重要的药食两用作物,葛根内生菌的研究具有重要实践意义。【目的】对广西葛根根部内生细菌进行分离、鉴定及促植物生长特性分析,旨在了解该药食同源植物内生细菌种群结构及其促生特性,为分析内生菌群体在药食同源植物产量和品质形成的作用及其内生细菌资源的开发利用提供参考。【方法】采用6种不同的培养基从广西葛根的根瘤、根系和根愈伤组织分离内生细菌,16S rRNA基因测序和系统发育分析内生细菌的分布特征和遗传多样性,采用生理生化方法测定分离菌株的固氮活性、溶磷特性、产生嗜铁素、分泌吲哚乙酸(indole-3-aceticacid,IAA)等促生特性。【结果】从葛根根瘤、根系和根部愈伤组织中共分离得到223个菌株,16S rRNA基因测序鉴定这些菌株隶属于2门4纲10科19属,其中芽孢杆菌属、假单胞菌属、土壤杆菌属、肠杆菌属为葛根优势菌群;内生细菌数量和群落组成存在明显的组织特异性,其数量表现为根瘤>根系>根愈伤组织,但其种群多样性表现为根愈伤组织>根系>根瘤。不同培养基分离出的细菌种群丰富度有差异。从供试菌株中筛...  相似文献

A group of endophytic diazotrophs were isolated from surface-sterilized roots and stems of different sugarcane varieties in the Tamilnadu region of India. From these, four isolates were selected, based on the highest acetylene reduction activity. Gene-specific PCR amplification confirmed the presence of nif-D genes in those isolates. The 16S rRNA sequence of isolates GR4 and GR7 had a 99.5% sequence similarity to the Pseudomonas sp. pDL01 (AF125317) and 16S rDNA sequence of isolate GR3 had a 100% similarity to that of Burkholderia vietnamiensis (AY973820). The 16S rDNA sequence of isolate GR9 was 99.79% similar to that of the Klebsiella pneumoniae type strain (KPY17657). Colonization by the isolates was confirmed using micropropagated sugarcane and sterile rice seedlings. Isolate GR9, identified as Klebsiella pneumoniae, was consistently more active in reducing acetylene as compared with the other isolates. The effects of GR9 and the sugarcane diazotroph Gluconacetobacter diazotrophicus were compared in inoculated micropropagated sugarcane plantlets. The effects of K. pneumoniae GR9, and four other diazotrophs, G. diazotrophicus, Herbaspirillum seropedicae, Azospirillum lipoferum 4B, and Burkholderia vietnamiensis in inoculated rice seedlings were compared. GR9 alone or in combination with the other diazotrophs performed best under pot conditions. The combined effects of nitrogen fixation and endophytic colonization of this diazotroph may be useful for the development of bio-inoculants.  相似文献

This paper informs the characterization by 16SrDNA partial sequence analysis of an endophytic diazotrophic bacterium isolated from roots of the halophyte shrub Prosopis strombulifera. The bacterium produced ABA, IAA, GA1, GA3 and jasmonic acid in chemically-defined culture medium as assessed by GC-EIMS. The results emphasize the role of phytohormones produced by endophytic bacteria in the association host-beneficial microorganisms, especially under conditions of adverse environments.  相似文献

A total of 250 endophytic fungal isolates, representing 30 morphotaxa, were isolated and characterised, they were collected from the different living symptomless parts of date palm trees of orchards of six Egyptian governorates. Colonisation was greater in samples from the midrib than in those from laminar tissue and slightly greater at the tip of the lamina compared with the base of the leaf. Acremonium spp. were frequently isolated as date palm root endophytes. Acremonium isolates were screened in Petri dishes to select the highest antagonistic one against an Algerian isolate of Fusarium oxysporum f.sp. albedinis. Two-week-old axenically reared date palm seedlings grown in Petri dishes were directly injected with spore suspension (1.5?×?107 spores/ml) of a pure culture of the virulent antagonistic isolate of Acremonium sp. One week after endophytic colonisation, date palm seedlings were then challenged with the pathogen, Fusarium albedinis. The challenged seedlings exhibited a significant reduction in wilt symptom percentage (by 87.0%), while the seedlings exposed to Fusarial toxin without pathogen exhibited the wilt disease symptoms. This indicates that the endophyte ably depresses any toxic action of F. albedinis. The endophytic fungus was recovered from sites distant from the point of inoculation after six?months from the application, indicating that the Acremonium sp. has the potential to move throughout the tissue plant, even the end time of trial. The Acremonium mode of action, as a biocontrol agent, was discussed.  相似文献

用微生物学传统分离培养方法从珙桐茎、叶及叶柄中分离到内生细菌56株,选取17株内生菌,提取其基因组DNA,并以此为模板,PCR扩增其16S rDNA,扩增出约1 500 bp大小的DNA条带,对15株内生菌16S rDNA测序,用BLAST软件对测序结果进行相似性比对,发现11株为Bacillus属,相似性为95%-98%;2株为Lysinibacillus属,相似性为97%和99%;1株为Bordetella属,相似性为95%;1株为非培养细菌的同源菌,相似性96%。芽孢杆菌为珙桐内生细菌优势菌属。通过构建系统发育树发现这15株内生菌明显聚为2大支。  相似文献

During a survey of endophytic diazotrophic bacteria associated with different rice varieties in Tamilnadu, some “endophytes” were obtained. Thirteen bacterial isolates from surface-sterilized roots and shoots were obtained in pure culture, which produced indole acetic acid (IAA) and reduced acetylene to ethylene. Polymerase chain reaction (PCR) amplification confirmed the presence of nif-H gene in all the isolates. Morphological, biochemical, and molecular characteristics indicated that all of them belonged to the genus Burkholderia One of them, MGK3, was consistently more active in reducing acetylene, and 16S rDNA sequences of isolate MGK3 confirmed its identification as Burkholderia vietnamiensis. Colonization of rice root was confirmed by strain MGK3 marked with gusA gene. The inoculated roots showed a blue color, which was most intense at the points of lateral root emergence and at the root tip. Transverse sections of roots, 15 days after inoculation, revealed beta-glucuronidase (GUS) activity within many of the cortical intercellular spaces next to the stele and within the aerenchyma. Nitrogen fixation was quantified by using 15N isotope dilution method with two different cultivars grown in pot and field experiments. Higher nitrogen fixation was observed in variety Ponni than in ADT-43, where nearly 42% (field) and 40% (pot) of the nitrogen was derived from the atmosphere (% Ndfa). Isolate MGK3 was used to inoculate rice seedlings in a comparison with four other diazotrophs, viz., Gluconacetobacter diazotrophicus LMG7603, Herbaspirillum seropedicae LMG6513, Azospirillum lipoferum 4B LMG4348, and B. vietnamiensis LMG10929. They were used to conduct two pot and four field inoculation experiments. MGK3 alone, and combined with other diazotrophs, performed best under both pot and field conditions: combined inoculation produced yield increases between 9.5 and 23.6%, while MGK3 alone increased yield by 5.6 to 12.16% over the uninoculated control treatment.  相似文献

Plant seeds are carriers of both beneficial bacteria and pathogens. Using the 16S rRNA gene clone library technique, we conducted a preliminary study on the community diversity and population succession dynamics of endophytic bacteria in seeds of reciprocal cross hybrid maize at different seed developmental stages. In both hybrid lines (108A and 108B), more types of endophytic bacteria were found at the proembryo-forming stage than in the other two stages, including 29 and 23 bacterial operational taxonomic units (OTUs), respectively. Undibacterium (39.20 and 30.00 % in 108A and 108B, respectively) was the first dominant bacterium to appear. At the milky stage, fewer types of endophytic bacteria in 108A and 108B appeared, including 18 and 16 OTUs, respectively, and the abundance of the dominant genus Burkholderia in the two seed samples reached 73.38 and 80.43 %, respectively. Limnobacter appeared as the second and third endophytic dominant bacterium in 108A (4.55 %) and 108B (5.07 %), respectively, in both seed samples. At the dough stage, the abundance of the first dominant bacterium, Burkholderia, in 108A and 108B was 78.26 and 84.80 %, respectively. Pantoea appeared as the second endophytic dominant bacterium in the both seeds (9.42 and 4.80 % in 108A and 108B, respectively). This is the first study on endophytic bacteria present during several crucial stages of the dynamic grain growth process of plant seeds conducted using culture-independent methods.  相似文献

The effect of endophytic and rhizospheric bacteria was studied on salt stress in a local paddy rice (Oryza sativa L.) variety GJ-17. Plants inoculated with endophytic bacterium Pseudomonas pseudoalcaligenes showed significantly higher concentration of glycine betaine-like quaternary compounds and higher shoot biomass at lower salinity levels. While at higher salinity levels, mixture of both P. pseudoalcaligenes and Bacillus pumilus showed better response against the adverse effects of salinity. However, accumulation of proline showed an opposite trend against plant growth promoting rhizobacteria (PGPR) treatment in salinity stress. Proline concentration increased with salinity but decreased in plants inoculated with either of the PGPRs or mixture of both P. pseudoalcaligenes and B. pumilus. The present study shows that inoculation of paddy rice (Oryza sativa L.) with a mixture of endophytic and rhizospheric bacteria could serve as a useful tool for alleviating salinity stress.  相似文献

Aspalathus linearis (burm f.), commonly known as rooibos, grows in nutrient and organic matter poor sandy soils that limit its growth. In this study, samples of nodules from both declined and healthy rooibos plants were collected to determine the frequency of nodule nitrogen-fixing and endophytic bacteria. Standard microbiological procedures as well as sequence analysis of the 16S rRNA revealed that more than 75% of the bacterial isolates from the healthy plants contained microsymbionts belonging to the Rhizobium group and the remaining 25% were characterized as Pseudomonas and Burkholderia spp. The nodule from the declined plants lacks a sufficient number of rhizobia and was mostly white in color, small and contains the free-living endospore-forming Bacillus and other endophytic Burkholderia and Pseudomonas spp. The results provide a baseline data on the microsymbionts of rooibos nodules in Citrusdal and highlighted the need for further investigation using additional techniques.  相似文献

重金属胁迫对植物内生细菌群落结构的影响在很大程度上是未知的,目前也很少有研究超积累植物内生细菌的群落结构与多样性对根际土壤中重金属的响应。【目的】探索在不同镉污染水平下,超积累(HE)和非超积累生态型(NHE)东南景天的根系、茎和叶片中内生细菌的群落结构与多样性的变化及其差异性,试图从植物-内生菌之间的相互关系的角度补充解释2种生态型东南景天对有效态镉忍耐和积累能力的差异。【方法】采用Illumina新一代测序方法分析了在不同Cd~(2+)浓度土壤上生长的2种生态型东南景天根、茎和叶中的内生细菌群落结构。【结果】高浓度Cd~(2+)抑制NHE东南景天的生长,内生细菌的丰富度和多样性也降低;然而,高浓度Cd~(2+)促进HE东南景天的生长,茎和根系内生细菌的丰富度增加。在3种土壤上,2种生态型东南景天叶片、茎和根系内生细菌均以变形菌门(Proteobacteria)、厚壁菌门(Firmicutes)、拟杆菌门(Bacteroidetes)和放线菌门(Actinobacteria)占优势。随着土壤中Cd~(2+)浓度的增加,HE东南景天叶片中Gammaproteobacteria纲、Negativicutes纲和Clostridia纲的相对丰度显著增加,茎中Alphaproteobacteria纲的相对丰度显著增加,Clostridia纲的相对丰度显著减少;NHE东南景天叶片中Alphaproteobacteria纲、Gammaproteobacteria纲和Clostridia纲的相对丰度没有显著变化,茎中Negativicutes纲的相对丰度显著减少,根系中Betaproteobacteria纲和Clostridia纲的相对丰度显著减少,Negativicutes纲却显著增加。在高Cd~(2+)污染土壤(50mg/kg)上,HE东南景天叶片中Sphingomonas属和茎中Veillonella属的相对丰度均大于NHE,且HE东南景天根系内生细菌的第一、第二、第三优势菌Veillonella、Sphingomonas、Prevotella属细菌均没有出现在NHE东南景天根系。【结论】土壤Cd~(2+)污染水平对2种生态型东南景天叶、茎、根中的内生菌群落结构有显著影响。  相似文献

A range of different species of diazotrophic bacteria has been found in tissues and the rhizosphere of oil palm plants, suggesting a potential to benefit from biological nitrogen fixation (BNF). A few studies have confirmed that plantlets at nursery stage can benefit significantly from BNF after inoculation with Azospirillum spp. but no data are available regarding the benefit from naturally-occurring diazotrophic bacteria in oil palm. The results described here were derived from two pot trials laid out under controlled conditions with plantlets from two important regions for palm oil production in Brazil, as well as from different field sites of mature oil palm plantations. The 15N natural abundance technique was employed to estimate plant dependence on BNF (%Ndfa) by the different ecotypes grown in soil and previously characterized as hosting diazotrophic bacteria. From both pot trials it was possible to identify some ecotypes of high potential for N2-fixation that reached in some cases approximately 50%Ndfa. However, the accuracy of measurement still needs to be improved using more suitable reference plants for pot experiments. Values of δ 15N signals from oil palm and reference plants in the field were inconclusive concerning any benefit from BNF to oil palm, owing to apparently high temporal and spatial variability of δ 15N of the plant-available N in the heterogeneous soil matrix for the different palm and reference plant tested.  相似文献

对番茄内生细菌数量动态及其对青枯病的生物防治研究结果表明:番茄内生细菌可来源于种子内部。番茄不同生育期,内生细菌数量最多在成株期,其中抗病品种根、茎分别为24.3×104CFU/g鲜重和22.9×104CFU/g鲜重,感病品种根、茎分别为9.8×104CFU/g鲜重和13.4×104CFU/g鲜重。抗病品种中具有拮抗青枯菌的内生细菌菌株为17个,感病品种中7个。部分内生细菌具促进番茄种子萌发和防治番茄青枯病的作用,其中5R和3R内生菌株的防病效果分别达91.7%和81.3%。  相似文献

Aims

Endophytic bacteria are ubiquitous in plants, but little information is available on the influence of endophytic bacteria on the uptake and metabolism of PAH by plants. Thus, we seek to investigate whether the colonization of a target plant by a PAH-degrading endophytic bacterium would improve the PAH metabolism of the plant and reduce the risk of plant PAH contamination.

Methods

A pyrene-degrading endophyte was isolated from PAH-contaminated plants using enrichment culture. After root inoculation with the isolated bacterium, greenhouse container experiments were conducted. Pyrene residues in soil and plant samples were analyzed by HPLC.

Results

A pyrene-degrading endophytic bacterium, Staphylococcus sp. BJ06, was isolated from Alopecurus aequalis and could degrade 56.0 % of pyrene (50 mg?·?L?1) within 15 days. BJ06 grew and degraded pyrene efficiently under environmental conditions. The bacterium significantly promoted ryegrass growth and pyrene removal from contaminated soil in container experiments. The pyrene concentrations in ryegrass roots and shoots in endophyte-inoculated planted soil were reduced by 31.01 % and 44.22 %, respectively, compared with endophyte-free planted soil.

Conclusions

We have provided new perspectives on the regulation and control of plant uptake of organic contaminants with endophytic bacteria. The results of this study will be valuable to risk assessments of plant PAH contamination.  相似文献

【目的】通过分析树兰原球茎内生细菌群落组成、多样性特征和促生功能,探究树兰种子萌发相关的核心细菌类群及生物学功能。【方法】以树兰原球茎(树兰种子在树皮基质上共生萌发、在树叶基质上共生萌发、在MS1培养基上非共生萌发)和共生萌发基质(松树皮、腐熟树叶)共5个样本为研究材料,采用高通量测序技术分析不同萌发条件下原球茎内生细菌的16S rRNA基因多样性,比较分析细菌群落多样性和物种组成特征,通过传统的内生细菌分离方法获得共生萌发原球茎内生细菌菌株,并进行促生潜力评价。【结果】从5个研究样本中共获得2 735个可操作分类单元(operational taxonomic unit, OTU),属于41门453科876属,其中变形菌门(Proteobacteria)和放线菌门(Actinobacteria)为优势门。主坐标分析(principal coordinates analysis, PCoA)结果表明,树兰原球茎与萌发基质细菌群落结构存在差异,非共生萌发原球茎与在树皮基质上共生萌发原球茎内生细菌群落结构最为接近。功能预测表明,在树叶基质上共生萌发的原球茎内生细菌固氮功能显著高于其他萌发条件。通过分离培养,共获得内生细菌19株,分属12属16种,其中鞭毛膨胀芽孢杆菌(Tumebacillus flagellatus)、Bradyrhizobium cenepequi和人参腐殖土魏茨曼氏菌(Weizmannia ginsengihumi)为共生萌发原球茎共有种;韩国假单胞菌(Pseudomonaskoreensis)和 W. ginsengihumi兼具有溶磷、产吲哚乙酸(indole-3-acetic acid, IAA)和铁载体的潜在能力。【结论】在不同环境中萌发的树兰原球茎均有丰富的内生细菌群落定殖;从共生萌发原球茎中分离的内生细菌具有固氮、溶磷、产IAA和铁载体等促生功能。本研究为兰科植物种子萌发相关微生物资源挖掘及兰科植物与微生物互作研究提供科学依据。  相似文献
