A rapid soil burial method for assessing the susceptibility of polyurethanes to biodeterioration was developed. The time of the test was reduced by prestressing the polyurethanes. The degree of deterioration was measured by following changes in the appearance of the polyurethanes and in selected physical properties. It was found that pre-stressing produced significant reductions in the tensile strength of a known susceptible polyurethane after burial in soil for 2 weeks. The reduction was greater than that found with unstressed polyurethanes buried for 26 weeks in active soil. Changes in tensile strength were less after burial for 4 weeks in sterile soil than after burial in active soil for the same period. The results suggest that deterioration of polyurethane during soil burial is a result of both chemical and microbial action.  相似文献

Representative varieties from the carrot groups, Chantenay, Nantes, Berlikum and Autumn King, together with three Australian varieties, were tested for susceptibility to attack by Cavariella aegopodii (Scop.). Tests were done at different average temperatures in cages in the insectary by inoculating plants with apterous forms of the aphid and frequently checking the extent of colonization over the next 20 or so days. Field sowings of the varieties were also made in each of 3 years, and over the immigration period and the subsequent population development and decline of the aphid, regular counts were made of the alates settling on a standard number of plants of each variety, and of the progeny produced. Some differences in susceptibility to attack were noted but, because the main development of C. aegopodii on field carrots is limited to two generations, these were too small to be of practical value in terms of plant resistance, and on all varieties large numbers of aphids could be found. Although the Australian variety Osborne Park was thought to have some resistance to C. aegopodii in New Zealand, it proved to be of average susceptibility. Autumn King appeared to be the most aphidsusceptible variety but could be tolerant to motley dwarf virus. Nantes was not as tolerant to aphid attack as the other varieties and as it is also intolerant to motley dwarf virus, might suffer much damage in years favouring aphid expansion. Berlikum seemed to be the least susceptible to aphid attack. Temperature differences affected expressions of varietal susceptibility less than they affected aphid fecundity and a few degrees increase in temperature more than halved the number of young born in a generation.  相似文献

Eleven carrot cultivars were tested to compare their susceptibility to carrot fly attack at Wellesbourne and at Mepal, Cambridgeshire in 1969. Significant differences in the percentages of unattacked roots were observed but not all the cultivars behaved consistently in this respect at the two sites. Foliage height and the percentage unattacked roots were not significantly correlated for these cultivars. Four of the cultivars representing the different levels of susceptibility were retested in fen and in mineral soil in microplots at Wellesbourne in 1970. Soil type significantly affected plant growth but not the relative susceptibilities of the cultivars to carrot fly attack. Plant size was positively correlated with insect attack and accounted for most, but not all, of the differences in susceptibility between the cultivars. In 1971 also, cv. Royal Chantenay was significantly less attacked than Speed's Norfolk Giant from August to December even after allowing for the large differences between the two cultivars in plant size. The experiments demonstrated a consistent difference in the relative susceptibilities of cvs Royal Chantenay and Speed's Norfolk Giant. The difference was correlated with plant size but allowances for foliage and root size failed to account for all of the difference. The results suggested a more fundamental basis for part of the difference in the susceptibility of the cultivars to carrot fly attack.  相似文献

To improve the hemocompatibility of polyurethanes, an amine monomer containing a long fluorine tail and phosphatidylcholine polar headgroups, 2-amino-3-oxo-3-(2-(2,2,3,3,4,4,5,5,6,6,7,7,8,8,8-pentadecafluorooctan amido) ethyl amino) propyl phosphorylcholine (FASPC) was firstly synthesized and characterized. Then four kinds of fluorinated phosphatidylcholine end-capped polyurethanes with different chemical structures were prepared. The surface properties of these prepared polyurethanes were characterized using X-ray photoelectron spectroscopic analysis (XPS) and water contact angle measurements. The results indicated that the phosphatidylcholine (PC) polar headgroups along with the fluorine tail could be easily enriched on the top surfaces, and the PC groups could be highly oriented on the outmost surface when the polymer film was in contact with water for only 30?s at room temperature. The evaluation of hemocompatibity was carried out via fibrinogen adsorption and platelet adhesion. Fibrinogen adsorption (37°C for 90 min) decreased by 98% to 87% compared to that on ordinary polyurethane surfaces, and almost no platelet adhesion and activation was observed at 37°C for 2?h.  相似文献

Striking varietal differences in susceptibility to attack by the lettuce root aphid, Pemphigus bursarius L., were first found in lettuces grown at Wellesbourne in 1955. Subsequent work has confirmed that White Favourite and Imperials E-4 and 19551-M are highly resistant; Imperial 45634-M, Continuity and Iceberg are markedly resistant. Midas, Salad Bowl and Imperial 4164 each appear to be a mixture of susceptible and resistant plants.
Immigrant winged forms of P. bursarius showed no preference for colonizing any particular variety of lettuce, and it seems that resistance to attack results from antibiosis.
It is suggested that varietal differences in the composition of root sap may account for the differences in susceptibility to lettuce root aphid, but analyses of water and alcohol extracts of root sap from resistant and susceptible lettuce varieties have not shown consistent differences.  相似文献

The effect of metabolic inhibitors and hormones on the susceptibility of P815 murine mastocytoma cells to antibody-C and cell-mediated killing, and on the ability of the cells to synthesize DNA, RNA, protein, carbohydrate, and lipid was tested. Pretreatment of the cells with adriamycin, actinomycin D, and puromycin increased the sensitivity of the cells to killing by rabbit anti-P815 antibody plus guinea pig C, but not by allogeneic P815-sensitized spleen cells. Conversely, mitomycin C treatment enhanced the cells' sensitivity to cell-mediated, but not antibody-C, killing. Insulin and hydrocortisone, but not epinephrine, were effective in decreasing the susceptibility of the cells to killing by both antibody-C and cell-mediated attack systems. The kinetics of the drug-induced increase and the hormone-induced decrease in susceptibility of the cells to antibody-C killing correlated with a decrease and increase, respectively, in the ability of the cells to incorporate fatty acid into complex cellular lipid. No such correlation was found between cellular lipid synthesis and tumor cell susceptibility to cell-mediated killing. The ability of the cells to resist either form of immune attack was not dependent on the ability of the cells to synthesize DNA, RNA, protein, or complex carbohydrate. These results suggest that the susceptibility of these tumor cells to antibody-C vs cell-mediated killing may be linked to different metabolic properties of the cells, and may reflect differences in the mechanisms of humoral vs cellular immune attack.  相似文献

Mixed-function oxidation of Escherichia coli glutamine synthetase by ascorbate, oxygen, and iron has previously been shown to cause inactivation of the enzyme and enhanced susceptibility to proteolytic attack by a variety of proteases. One of these proteases, from rat liver, is a high molecular weight cysteine proteinase which does not degrade native glutamine synthetase at neutral pH. Although inactive, the oxidized glutamine synthetase preparations used in this study were only partially degraded by this proteinase. Some of the subunits were degraded to acid soluble products with no detectable intermediates; the remaining subunits had not become susceptible to proteolytic attack during the limited exposure to the ascorbate mixed-function oxidation system. Several mammalian enzymes which are known to be inactivated by mixed-function oxidation were tested as substrates for the proteinase. Native rabbit muscle enolase and pyruvate kinase were resistant to degradation, but their oxidatively inactivated forms were degraded. Oxidized phosphoglycerate kinase and creatine kinase were also preferentially degraded. Moreover, trypsin degraded oxidized preparations of all of these enzymes faster than control preparations. Oxidative inactivation of superoxide dismutase by hydrogen peroxide caused a slight increase in susceptibility to proteolytic attack, but the enzyme was still relatively resistant to degradation both by the cysteine proteinase and by trypsin. Although oxidation conditions may not have been optimal for demonstrating enhanced proteolytic susceptibility, the results do indicate that mixed-function oxidation can render some mammalian enzymes, as well as bacterial glutamine synthetase, susceptible to degradation. Mixed-function oxidation of these proteins may be a mechanism of marking them for intracellular turnover.  相似文献

1 Wireworms, the soil dwelling larvae of click beetles, Agriotes spp., have recently become a more prevalent pest of potatoes. The present study investigated whether potato varieties showed variable susceptibility to wireworm herbivory, and also tested whether increased susceptibility was associated with lower concentrations of glycoalkaloids. Twelve varieties were originally screened across a range of experimental scales, including laboratory and tunnel experiments and a large‐scale field trial involving over 2000 tubers. 2 In laboratory no‐choice tests, Maris Peer, Marfona and Rooster varieties were significantly more susceptible to wireworm attack, with 63% of tubers showing damage, compared with just 15% of the less susceptible varieties of King Edward, Nadine and Maris Piper. There was also greater tissue consumption and weight gain when wireworms were reared on the most susceptible varieties. 3 In choice tests, wireworms showed a significant preference for those varieties previously identified as being the most susceptible to wireworm herbivory (4.2 holes per tuber) compared with the least susceptible (1.2 holes per tuber). Similar patterns of susceptibility were seen in the field trial, although there was generally more variation in susceptibility. 4 In a tunnel experiment, Marfona and Maris Peer were significantly more susceptibile to wireworm attack (47% of tubers showing damage) compared with Nadine, King Edward and Maris Piper (27% of tubers showing damage). Although Nadine, in particular, had the highest glycoalkaloid concentrations (309.33 mg/kg) and lowest amounts of wireworm herbivory, the relationship between susceptibility and glycoalkaloid concentrations was weak, suggesting that this is unlikely to be the sole mechanism underpinning varietal susceptibility.  相似文献

1 Attack by shoot borers (Hypsipyla grandella Zeller) is the main factor limiting the cultivation of Cedrela odorata and Swietenia macrophylla, two economically important members of the mahogany family. No viable methods of pest control are currently available. To assess for genetic variation in susceptibility to pest attack, a combined progeny/provenance test of C. odorata and a provenance test of S. macrophylla were established separately at CATIE, Turrialba, Costa Rica, and assessed intensively over an 84-week period. 2 Variation in height growth between provenances was highly significant in both species, provenance means varying by a factor of 2.7 and 1.2 in C. odorata and S. macrophylla, respectively. Cedrela odorata also displayed provenance variation in foliar phenology, as 94.7% of the trees from a ‘wet zone’ provenance remained foliated, whereas > 35% of trees from three ‘dry zone’ provenances abscised their leaves during the dry season. 3 Both species displayed significant genetic variation in susceptibility to shoot borer attack. At the peak of attack during the second year of growth, the effect of provenance was highly significant in both species, the mean number of attacks per tree varying between 0.8–2.4 and 0.6–1.3 in different provenances of C. odorata and S. macrophylla, respectively. A provenance of C. odorata from San Carlos, Costa Rica, displayed consistently lower susceptibility to pest attack, being subjected to fewer attacks during the first year and demonstrating a higher mean height to first point of damage. 4 The nitrogen, total tannin and proanthocyanidin concentration of foliage varied significantly between C. odorata provenances: nitrogen concentration was significantly lower and tannin and proanthocyanidin contents were significantly higher in trees from the San Carlos provenance, Costa Rica. This study therefore suggests that proanthocyanidins may reduce susceptibility of C. odorata to H. grandella, at least during the early period of growth. 5 These results provide support for the development of plant resistance as a strategy for managing shoot borers. In particular, there may be scope for selecting for high foliar proanthocyanidin content and the ability to tolerate attack by vigorous apical growth.  相似文献

Since the discovery of the soybean aphid, Aphis glycines Matsumura (Hemiptera: Aphididae), in North America in 2000, chemical control has been the most effective method to manage aphid outbreaks. Increased insecticide use in soybean raises the possibility of developing insecticide resistance in soybean aphid, and monitoring insecticide susceptibility is essential to maintain pesticide tools. We developed a simple and reliable aphid-dip bioassay by using a tea strainer that resulted in -90% survival in controls. Using this technique, we tested susceptibility of a greenhouse strain of soybean aphid that has never been exposed to insecticides, and field-collected aphid strains from two counties in Michigan. Aphid susceptibility was tested for five insecticides by dipping groups of five aphids in each insecticide dose for 10 s. After 48 h, aphids were classified as dead or alive, and counted. Aphids from all strains were highly susceptible to chlorpyrifos, lambda-cyhalothrin, esfenvalerate, and dimethoate, with LC50 and LC90 values well below the recommended application rates. However, aphids showed less susceptibility after 48 h to neonicotinoid imidacloprid, with higher LC90s and wider fiducial limits. This illustrated the potential limitation of using a 48-h assay to evaluate insecticides with longer-term, sublethal impacts. Nevertheless, this study made use of a simple aphid-dip method to test and compare insecticide susceptibility of soybean aphid. In the event of a field failure, the aphid populations involved can be tested in comparison to a susceptible greenhouse strain to determine the extent of resistance development.  相似文献

In most populations of Solidago altissima, a small proportion of plants have a growth pattern in which the stem bends early in the season so that the apex is pointing downward (i.e., a “candy-cane” stem) then straightens up again later in the season. The majority of plants, however, do not show this pattern. Instead their stems remain erect from emergence through flowering and senescence. It has recently been shown that the candy-cane stems are a result of a ducking strategy that reduces the risk of attack by apex-attacking herbivores. With such a resistance advantage, it is unclear why the candy-cane morphology is always in the minority, and why the erect morphology persists at all in S. altissima populations. In this study, we tested the hypothesis that the advantage of ducking is inversely frequency dependent and thus will not be an effective resistance strategy when ducking plants are in the majority. In a series of trials, we introduced gall flies (Eurosta solidaginis) into enclosures with relatively low (15%) and relatively high (85%) frequencies of candy-cane versus erect stems. Candy-cane stems were more resistant than erect stems regardless of relative frequency; thus, the hypothesis was rejected. Overall, attack rates were lower for both stem types in the high candy-cane frequency groups than in the low candy-cane frequency groups. This frequency-dependent attack rate suggests that erect-stemmed plants gain “associational resistance” by having a majority of candy-cane neighbors, while candy-cane plants suffer from “associational susceptibility” when surrounded by erect-stemmed neighbors.  相似文献

Eggs from reciprocal hybrids between the C57BL/6By and BALB/cBy strains were tested for their susceptibility to attack by hyaluronidase and pronase. There were significant reciprocal differences between the F1 females in the responses of their unfertilized eggs to both enzymes. The F1 hybrids from BALB mothers showed the increased susceptibility characteristic of C57BL whilst the F1 hybrids with C57BL mothers were more resistant to both enzymes, like BALB mice. Eggs from the four kinds of reciprocal F2 hybrid females also showed patroclinous patterns of susceptibility. A patroclinous difference was found between reciprocal crosses of the CXBD and CXBE recombinant inbred strains but not in crosses between recombinant inbred strains with similar phenotypes. Cross fostering did not alter the phenotypes of the C57BL and BALB females or those of their reciprocal F1 hybrids. The findings are interpreted in terms of differential genomic imprinting of paternally inherited information. The possible general usefulness of patroclinous differences between reciprocal F1 females in revealing differences in imprinting is noted.  相似文献

Sixty-eight halobacteria, including both culture collection strains and fresh isolates from widely differing geographical areas, were tested for susceptibility to arsenate, cadmium, chromium, cobalt, copper, lead, mercury, nickel, silver, and zinc ions by an agar dilution technique. The culture collection strains showed different susceptibilities, clustering into five groups. Halobacterium mediterranei and Halobacterium volcanii were the most metal tolerant, whereas Haloarcula californiae and Haloarcula sinaiiensis had the highest susceptibilities of the culture collection strains. Different patterns of metal susceptibility were found for all the halobacteria tested, and there was a uniform susceptibility to mercury and silver. All strains tested were multiply metal tolerant.  相似文献

Ozonolysis was used to obtain polyols with terminal primary hydroxyl groups and different functionalities from trilinolein (or triolein), low-saturation canola oil, and soybean oil. The functionality of the model polyol from triolein (trilinolein) was 3.0 and that of soy polyol was 2.5, due to the presence of unreactive saturated fatty acids, while canola gave a polyol with a functionality of 2.8. All polyols exhibited a high tendency to crystallize at room temperature. The resulting waxes had melting points comparable to that of paraffin and very low viscosities in the liquid state. The polyols were cross-linked using 4,4'-methylenebis(phenyl isocyanate) to give polyurethanes. Glass transitions (T(g)) for the model-, canola-, and soy-based polyurethanes were 53, 36, and 22 degrees C, respectively. The about 30 degrees C lower T(g) of the soy-based polyurethane than that of the model polyurethane was the result not only of lower functionality but also of the presence of saturated fatty acids in the former. Polyurethane from the canola polyol had intermediate cross-linking density and properties. These polyurethanes displayed excellent mechanical properties and higher glass transition temperatures compared to polyurethanes from epoxidized and hydroformylated polyols of the same functionality, presumably due to the absence or lower content of dangling chains in the former.  相似文献

Co-trimoxazole (trimethoprim—sulphamethoxazole) was compared with furazolidone in the treatment of shigellosis in two groups of 33 and 30 patients respectively. Those treated with co-trimoxazole recovered more quickly; none had shigellae in the faeces four days after the start of treatment, whereas in the group given furazolidone eight still had positive stool cultures seven days after treatment.The susceptibility of 104 shigella strains to seven antimicrobial agents was studied by plate dilution technique. All agents but tetracycline and chloramphenicol were found highly effective against most of the strains tested. All shigella isolates were resistant to sulphamethoxazole, and 63% were sensitive to trimethoprim. Potentiation of trimethoprim by sulphamethoxazole was shown in that all strains tested became sensitive to the combination of trimethoprim and sulphamethoxazole in a ratio of 1:20.  相似文献

NK cell activation is negatively regulated by the expression of target cell MHC class I molecules. We show that this relationship is nonlinear due to an NK cell activation/inhibition threshold. Ewing's sarcoma family tumor cell monolayers, which were highly susceptible to NK cells in vitro, developed a highly resistant phenotype when cultured as three-dimensional multicellular tumor spheroid structures. This suggested that tumor architecture is likely to influence the susceptibility to NK cells in vivo. Resistance of the multicellular tumor spheroid was associated with the increased expression of MHC class I molecules and greatly reduced NK cell activation, implying that a threshold of NK cell activation/inhibition had been crossed. Reducing MHC class I expression on Ewing's sarcoma family tumor monolayers did not alter their susceptibility to NK cells, whereas increased expression of MHC class I rendered them resistant and allowed the threshold point to be identified. This threshold, as defined by MHC class I expression, was predictive of the number of NK-resistant target cells within a population. A threshold permits modest changes in the target cell surface phenotype to profoundly alter the susceptibility to NK cells. Whereas this allows for the efficient detection of target cells, it also provides a route for pathogens and tumors to evade NK cell attack.  相似文献

Highly lethal terrorist attacks, which we define as those killing 21 or more people, account for 50% of the total number of people killed in all terrorist attacks combined, yet comprise only 3.5% of terrorist attacks. Given the disproportionate influence of these incidents, uncovering systematic patterns in attacks that precede and anticipate these highly lethal attacks may be of value for understanding attacks that exact a heavy toll on life. Here we examined whether the activity of terrorist groups escalates–both in the number of people killed per attack and in the frequency of attacks–leading up to highly lethal attacks. Analyses of terrorist attacks drawn from a state-of-the-art international terrorism database (The Global Terrorism Database) showed evidence for both types of escalation leading up to highly lethal attacks, though complexities to the patterns emerged as well. These patterns of escalation do not emerge among terrorist groups that never commit a highly lethal attack.  相似文献

Novel biobased aromatic triols (1,3,5-(9-hydroxynonyl)benzene and 1,3,5-(8-hydroxyoctyl)-2,4,6-octylbenzene) were synthesized through the transition-metal-catalyzed cyclotrimerization of two alkyne fatty acid methyl esters (methyl 10-undecynoate and methyl 9-octadecynoate) followed by the reduction of the ester groups to give terminal primary hydroxyl groups. A series of biobased segmented polyurethanes based on these triols, 1,4-butanediol as a chain extender and 4,4'-methylenebis(phenyl isocyanate) as a coupling agent, were synthesized. Samples were prepared with hard-segment contents up to 50%. The morphologies and thermal properties of these polyurethanes were studied by Fourier transform infrared spectroscopy, wide-angle X-ray diffraction, differential scanning calorimetry, thermogravimetric analysis, and dynamic mechanical thermal analysis. Partial crystallinity and phase separation were detected in samples with hard-segment content of 50%.  相似文献
