1 Deterrent and toxic substances known to affect Hypsipyla grandella larvae were bioassayed to determine their possible systemic activity under laboratory conditions in Turrialba, Costa Rica. 2 Leaflets from Spanish cedar Cedrela odorata seedlings grown in vitro on culture media treated with different plant extracts were excised from the seedlings and exposed to H. grandella larvae. 3 Both Azatin EC (derived from neem, Azadirachta indica) and plant extracts (from Quassia amara wood, Ruta chalepensis leaves and Sechium pittieri fruits) were able to translocate within Spanish cedar seedlings and express their biological activity against H. grandella larvae. 4 Rates of leaflet consumption were nil for seedlings treated with the commercial standard (carbofuran) and Azatin EC, both causing larval death. 5 Intermediate levels of consumption were reached with the three plant extracts, all of them causing deterrence, because they precluded feeding activity by larvae when exposed to treated leaflets.  相似文献

Biological activity of a plant extract (common rue, Ruta chalepensis) and a semi purified fraction (from "tacaco cimarrón", Sechium pittieri) on mahogany shootborer larvae (Hypsipyla grandella) was studied. A randomized complete block design, with four replications, was used. H. grandella third instar larvae were exposed for 24 h to Cedrela odorata leaf discs dipped in several treatment dissolutions of each extract (0.1, 0.32, 1.0, 3.20, and 10%); afterwards, each larva was transferred to a flask containing an artificial diet and was allowed to complete its development. Variables measured included food consumption (foliar area eaten in 24 h), mortality, and developmental effects (developmental time for each larval instar and the pupa, and pupal weight). The common rue extract showed a clear antifeedant activity at a concentration as low as 0.32%, whereas the "tacaco cimarrón" fraction caused toxicity, especially at the two highest concentrations (3.20 and 10%).  相似文献

A methanol extract of Cassia tora seeds was successively partitioned with diethyl ether, chloroform, ethyl acetate, and water, and the antitumor-promoting activity of the solvent fractions was determined by inhibition of Epstein- Barr virus early antigen (EBV-EA) activation induced by teleocidin B-4 in Raji cells. The diethyl ether (68.7%) and chloroform (91.2%) fractions and the hydrolysate (94.3%) of the ethyl acetate fraction had strong inhibitory activities. The chloroform and ethyl acetate fractions were chromatographed on silica gel and further purified by HPLC. Three active compounds, obtusifolin-2-glucoside (75.0%), chryso-obtusin-6-glucoside (56.8%), and norrubrofusarin- 6-glucoside (39.4%), were obtained from the ethyl acetate fraction, and two active compounds, questin (97.9%) and chryso-obtusin (53.8%), were isolated from the chloroform fraction.  相似文献

Resistance in oak ( Quercus spp.) to defoliation by caterpillars of Tortrix viridana L. was studied in an oak wood and a mixed wood in Roudsea Wood National Nature Reserve. When defoliation is severe in the oak wood a few trees there and most of the oaks in the mixed wood retain relatively undamaged canopies. Their resistance was found to be phenological. The resistant trees in the oak wood flush either later or earlier than those that become defoliated; those in the mixed wood flush later. On late-flushing trees the buds open too late for newly hatched larvae to penetrate them, the resultant mortality reducing the larval population below the level at which damage is severe. The early-flushing trees carry as many larvae per leaf cluster as non-resistant trees but do not show severe defoliation because they produce more foliage and 'grow away' from the attack.  相似文献

The methanol extract of the rhizomes of Gloriosa superba Linn (Colchicaceae) and its subsequent fractions in different solvent systems were screened for antibacterial and antifungal activities. Excellent antifungal sensitivity was expressed by the n-butanol fraction against Candida albicans and Candida glaberata (up to 90%) and against Trichophyton longifusus (78%) followed by the chloroform fraction against Microsporum canis (80%). In the antibacterial bioassay, the crude extract and subsequent fractions showed mild to moderate antibacterial activities. Chloroform fraction displayed highest antibacterial sensitivity against Staphylococcus aureous (88%) followed by the crude extract (59%). The total phenol content of the crude extract and fractions of the plant expressed no significant correlation with the antimicrobial activities.  相似文献

The aqueous extract of dried bonito (Katsuobushi) was distilled under reduced pressure. The resulting distillate with diethyl ether and the extract was separated into acidic, phenolic, basic and neutral fractions. The neutral fraction was further fractionated into ten sub-fractions by silica gel column chromatography. All these sub-fractions were analyzed by gas chromatography and gas chromatography-mass spectrometry.

One hundred and sixty-five compounds were identified and 12 compounds were tentatively identified from the neutral fraction. Among them, 111 compounds were newly identified as flavor components of Katsuobushi.  相似文献

Methanol extract was prepared from the fronds of Pteris biaurita and partial purification was done by solvent partitioning with diethyl ether and ethyl acetate, followed by hydrolysis and further partitioning with ethyl acetate. The three fractions, thus obtained were bioassayed separately against five test fungi--Curvularia lunata, Fomes lamaoensis, Poria hypobrumea, Fuasrium oxysporum and a bacterium--Bacillus pumilus, by spore germination, radial growth and agar cup techniques. Results revealed that ethyl acetate fraction (III) contained the active principle. TLC plate bioassay of the active fraction revealed inhibition zone at an Rf of 0.5-0.65. Silica gel from this region was scraped, eluted in methanol and subjected to UV-spectrophotometric analysis. An absorption maxima of 278 nm was recorded. HPLC analysis of TLC-eluate revealed a single peak with retention time of 8.1 min. GC-MS analysis revealed six major peaks in the retention time range of 7.2-10.9 min. Comparison with GC-MS libraries revealed that the extracts may contain a mixture of eicosenes and heptadecanes.  相似文献

In vitro antioxidant activities of three selected Indian red seaweeds - viz., Euchema kappaphycus, Gracilaria edulis and Acanthophora spicifera were evaluated. Total phenolic content and reducing power of crude methanol extract were determined. The antioxidant activities of total methanol extract and five different solvent fractions (viz., petroleum ether (PE), ethyl acetate (EA), dichloromethane (DCM), butanol (BuOH) and aqueous) were also evaluated. EA fraction of A. spicifera exhibited higher total antioxidant activity (32.01 mg ascorbic acid equivalent/g extract) among all the fractions. Higher phenolic content (16.26 mg gallic acid equivalent/g extract) was noticed in PE fraction of G. edulis. Reducing power of crude methanol extract increased with increasing concentration of the extract. Reducing power and hydroxyl radical scavenging activity of E. kappaphycus were higher compared to standard antioxidant (alpha-tocopherol). The total phenol content of all the seaweeds was significantly different (P<0.05). In vitro antioxidant activities of methanol extracts of all the three seaweeds exhibited dose dependency; and increased with increasing concentration of the extract.  相似文献

Zizyphus spina-christi (Rhamnaceae family) is an edible plant used in folk medicine. Therefore, it is of interest to report the cytotoxic effects of Z. spina-christi bark crude extract on human cell lines. Crude ethanol extract of Z. spina-christi bark was fractionated with increasing polarity (diethyl ether, chloroform, ethyl acetate and butanol fractions). The fractions were examined for their cytotoxicity against human colon cancer (HCT-116 and CACO-2), cervical cancer (HeLa and HEp-2), lung carcinoma (A-549), hepatocellular carcinoma (HepG-2), breast cancer (MCF-7) and prostate cancer (PC-3) cell lines using viability assay. Diethyl ether fraction of Z. spina-christi showed the highest cytotoxic effects among the four extracts of Z. spina-christi. The IC50 of diethyl ether fraction was 7.14, 11.2, 11.6, 15.4, 39.8, 42.2, 84.2 and 153.8 µg/ml on HepG-2, A-549, CACO-2, HCT-116, MCF-7, PC-3, HeLa, and HEp-2 cell lines, respectively. Data shows that the diethyl ether fraction of Z. spina-christi showed effective cytotoxic effects in colon, lung and hepatocellular cancer cell lines.  相似文献

Abstract.  1. Neonate evergreen bagworms, Thyridopteryx ephemeraeformis (Haworth) (Lepidoptera: Psychidae), disperse by dropping on a strand of silk, termed silking , and ballooning on the wind. Larvae construct silken bags with fragments of plant foliage. This species is highly polyphagous, feeding on more than 125 species of woody plants of 45 families. The larvae commonly infests juniper ( Juniperus spp.) and arborvitae ( Thuja spp.), but rarely feed on deciduous hosts such as maples. The hypothesis is proposed that polyphagy in T. ephemeraeformis is maintained by variation among larvae in dispersal behaviour, and time constraints on the opportunity to disperse, but patterns of host species preference result from a predisposition for larvae to settle on arborvitae and juniper but disperse from other hosts.
2. Consistent with that hypothesis, laboratory experiments revealed: (a) starved larvae varied in their tendency to disperse from paper leaf models; (b) starved larvae readily silked only during their first day; (c) larvae became increasingly sedentary the longer they were exposed to plant foliage; (d) when provided with several opportunities to silk, larvae became sedentary after exposure to arborvitae foliage, but repeatedly silked after exposure to maple ( Acer species) foliage or paper; and (e) larvae were less inclined to silk from foliage of arborvitae than from maple.
3. Field experiments supported the hypothesis by demonstrating that: (a) neonates tended to disperse from maple leaves while larvae older than 1 day tended to settle and remain; and (b) neonates were less likely to disperse from arborvitae and juniper trees than from maples.  相似文献

The susceptible species Cedrela odorata and Swietenia macrophylla to attack by Hypsipyla grandella (Zeller) larvae were grafted onto the resistant species Khaya senegalensis and Toona ciliata. Six-month-old grafted plants were then compared to their reciprocal grafts and to both intact (non-grafted) and autografted plants for damage due to H. grandella larvae and for their effects on larval performance. Two experiments were conducted: one in which the apical bud of the main plant shoot was inoculated with H. grandella eggs, and the other in which the bud was inoculated with third instars. Damage in each experiment was assessed by the number of frass piles, number and length of tunnels, number of damaged leaves, and damage to the apical bud. Larval performance was evaluated in terms of time to reach pupation and pupal weight and length. In both experiments, plant damage differed significantly among treatments (P < 0.03). Resistant rootstocks conferred resistance to susceptible scions. In both experiments, grafting by itself, regardless of the rootstock and scion combination, also reduced damage caused by H. grandella larvae. Scions of autografted susceptible species had similar resistance to susceptible scions grafted on resistant rootstocks. Few larvae reached pupation, and their pupal weight and length were similar.  相似文献

Bioefficacy of hexane, chloroform and ethyl acetate extracts of the seeds of Ceasalpinea bonduc (L.) Roxb. was studied against third instar larvae of Spodoptera litura at 0.25, 0.5, 1, 2.5 and 5.0% concentrations. Significant antifeedant, larvicidal and pupicidal activities and least LC50 values were observed in chloroform extract. The chloroform extract was subjected to fractionation using silica gel column chromatography. Six fractions were obtained; among these, the third fraction showed high antifeedant, larvicidal and pupicidal activities at 1000 ppm concentration. Abnormalities in adults were also observed. All the activities were concentration dependent. C. bonduc could be useful in integrated pest management programme.  相似文献

Abstract  Hexane, petroleum ether, chloroform, ethyl acetate, methanol and water extracts of Clausena anisata [(Willd.) Hook F. Ex Benth] leaves and roots were evaluated against Helicoverpa armigera (Hübner) for antifeedant activities. Antifeedant activity was confirmed, and was found to be higher in root extracts than those of the leaf. Chloroform and petroleum ether extracts of the root showed strongest antifeedant activities (DC50s [concentration (C) causing 50% deterrence compared with the control] 0.014% and 0.016% respectively), and root extracts were fractionated using silica gel column chromatography. One fraction of the chloroform and one of the petroleum ether root extracts was active; and on the basis of mass spectroscopy and 1H and 13C nuclear magnetic resonance spectral data, the active compounds in the two fractions were confirmed to be identical, and identified as osthol [2H-1-Benzopyran-2-one, 7-methoxy-8-(3-methyl-2-butenyl)]. The highest concentration of osthol was found in the chloroform root extract. Antifeedant activities of the root extracts, as measured by DC50 values, were highly correlated with their osthol contents. Approximately 99% of the variation in bioactivity of the root extracts could be accounted for by variation in osthol content; osthol therefore, appeared to be an antifeedant component of C . anisata to H. armigera . This may provide a new approach to managing this pest.  相似文献

The leaf skeletonizer Uraba lugens Walker (Lepidoptera: Nolidae), an Australian species, locally known as "gumleaf skeletonizer", is well established in New Zealand. This insect has the potential to become a serious pest of forestry and amenity eucalypts (Eucalyptus spp.) and is the focus of a long-term management program. The use of synthetic chemical or biological insecticides is one possible control method within an integrated control program. A series of dose-response trials were conducted using laboratory bioassays to test the efficacy of several insecticides against U. lugens: pyrethroids, spinosad, Bacillus thuringiensis kurstaki Berliner (Btk) and an insect growth regulator, Mimic. Pyrethroids and spinosad proved highly effective against U. lugens larvae, achieving 100% mortality after 3-6-d exposure. The performance of Btk was lower against gregarious skeletonizing larvae compared with solitary chewing larvae. When good coverage of the target foliage is achieved, >90% mortality is possible with Btk. Mimic performed poorly against U. lugens compared with other insecticides tested (<60% mortality). The Eucalyptus species on which larvae were feeding significantly altered insecticide efficacy. Treatments applied to Eucalyptus nitens (Deane & Maiden) Maiden had reduced efficacy compared with E. cinerea F. Muell. ex Benth. or E. fastigata Deane & Maiden. Cooler temperatures also reduced insecticide efficacy, presumably by decreasing movement and food consumption by U. lugens. Recommendations on spray applications to control U. lugens in New Zealand are given.  相似文献

Background

Tribolium castaneum (Herbst) is a harmful pest of stored grain and flour-based products in tropical and subtropical region. In the present study, rhizome of Drynaria quercifolia (J. Smith) was evaluated for pesticidal and pest repellency activities against T. castaneum, using surface film method and filter paper disc method, respectively. In addition, activity of the isolated compound 3,4-dihydroxybenzoic acid was evaluated against the pest.

Results

Chloroform soluble fraction of ethanol extract of rhizome of D. quercifolia showed significant pesticidal activity at doses 0.88 to 1.77 mg/cm2 and significant pest repellency activity at doses 0.94 to 0.23 mg/cm2. No pesticidal and pest repellency activity was found for petroleum ether, ethyl acetate and methanol soluble fractions of ethanol extract as well as for 3,4-dihydroxybenzoic acid.

Conclusion

Considering our findings it can be concluded that chloroform soluble fraction of rhizome of D. quercifolia is useful in controlling T. castaneum of stored grain and flour-based products.

The seaweed Sargassum (Sargassaceae) as tropical alternative for goats' feeding. The nutritive value of seaweed (Sargassum spp.) was studied in Baja California Sur, Mexico. Twenty female Nubian goats (43-weeks old) were randomly distributed into two groups of 10 goats each and were housed in individual pens. One group was fed with a control diet and the other with a diet supplemented with 25% of Sargassum spp. Feed and water intake were recorded daily and individually for 60 days. The weight of each goat was recorded every 15 days. The nutritional content of Sargassum spp. was 89% dry mater, 8% crude protein, 31% ash, 2% ether extract, and 39% carbohydrates. Fiber fractions, minerals, vitamins, fatty acids, and antinutritional factors were also determined. There were no significant differences in body weight (8.6 kg control and 9 kg experimental), feed intake (1.3 kg control and 1.6 kg experimental), and feed conversion rate (11.1 control and 12.6 experimental). Water consumption was greater in the goats that ate the Sargassum diet (5.3 1). From these results, Sargassum spp. can be considered as an alternative feedstuff for goats.  相似文献

为了进一步评估斑点野生稻Oryza punctata的抗虫性,采用液 液分配萃取和硅胶柱层析的方法,从斑点野生稻甲醇提取物中分离获得石油醚、氯仿、乙酸乙酯、正丁醇和水的萃取物,测定了其对斜纹夜蛾Spodoptera litura 3龄幼虫拒食活性。结果显示,氯仿萃取物比其他4种萃取物具有更高的拒食活性,在25 mg/mL的浓度下,24 h和48 h的拒食率分别为55.62%和52.66%。氯仿萃取物经硅胶柱层析后,得到14个组分。比较14个组分的拒食活性,发现组分4和10为主要的活性组分。这两个组分对斜纹夜蛾幼虫中肠脂肪酶和α-淀粉酶的活性都具有一定的抑制活性,其中组分10对脂肪酶具有显著的抑制效果,以1 mg/mL的浓度活体处理48 h和72 h抑制率分别达19.82%和34.60%; 对α-淀粉酶的影响在48 h和72 h内都具有显著的抑制效果,随着处理时间的延长,其抑制率逐步提高,72 h的抑制率分别为25.06%和27.40%。结果提示斜纹夜蛾幼虫中肠脂肪酶和α-淀粉酶可能是斑点野生稻拒食活性成分的作用靶标。  相似文献

Caesalpinia crista seed extracts were investigated in the laboratory against Helicoverpa armigera (Hubner). The extracts exhibited powerful antifeedant and growth disruption activity. The toxic symptoms of the poisoned larvae included reduction in weight gain and mortality of larvae and pupae, larval-pupal intermediates and malformed adults. Maximum anti-feedance is caused by methanol extract (AI50=0.0186%), followed by hexane extract (0.0212%), ethyl acetate extract (0.0416%), butanol extract (0.0767%) and aqueous extract (0.2199%). The larval mortality ranged from 10.00 to 70.00% in different extracts. The 50% larval growth inhibition recorded at 3DAT was statistically at par by methanol and hexane extract. The percent 150 values for inhibiting normal adult emergence were 0.0287, 0.0325, 0.0485, 0.0977 and 0.0547% for methanol, hexane, ethyl acetate, aqueous and butanol extract. The biosafety evaluation of these extracts carried out against the predator, Coccinella septumpunctata showed no mortality of the adults till nine days after treatment. Though the observation taken at 10 DAT, showed slight mortality of adults by methanol extract at both 5.0 and 1.0% concentration.  相似文献

A greenhouse study was conducted to investigate the persistence of methoxyfenozide in pepper (Capsicum annuum L.) foliage. An aqueous suspension of methoxyfenozide was sprayed on pepper plants at concentrations of 72 and 144 mg of active ingredient (a.i.)/L. Foliage was collected at different intervals of time (0, 3, 5, 10, 20, 30, 40, and 50 days) after the treatment, and the methoxyfenozide residue was measured by high‐performance liquid chromatography. The foliage was also used in bioassays to determine the residual toxicity on and the consumption rate of the third‐instar larvae of the beet armyworm, Spodoptera exigua (Hübner) (Lepidoptera: Noctuidae). The methoxyfenozide concentrations observed after 50 days had decreased to 19 and 69 μg/g per sample, corresponding to a loss of 61% and 28% from the application concentrations of 72 and 144 mg a.i./L, respectively. When fitting a first‐order kinetics degradation model, the half‐life (DT50) of this compound was 76 days. Both application concentrations of methoxyfenozide caused a high mortality rate (≥97%) when the larvae were fed the pepper foliage collected at all of the time intervals. Lastly, at all of the time points, the consumption rate by the larvae was reduced to between 57% and 92% for both concentrations that were bioassayed. Our results indicate that, under the present greenhouse conditions, the degradation of methoxyfenozide was slower than that reported by other authors and that its residues were highly toxic to S. exigua larvae. The implications of these results for the management programs of this pest are discussed.  相似文献
