The localized surface plasmon resonances of multilayered nanostructures are studied using finite difference time domain simulations and plasmon hybridization method. Concentric metal–dielectric–metal (MDM) structure with metal core and nanoshell separated by a thin dielectric layer exhibits a strong coupling between the core and nanoshell plasmon resonance modes. The coupled resonance mode wavelengths show dependence on the dielectric layer thickness and composition of core and outer layer metal. The aluminum-based MDM structures show lower plasmon wavelength compared with Ag- and Au-based MDM nanostructures. The calculated refractive index sensitivity (RIS) factor is in the order Ag–Air–Ag>Au–Air–Au>Al–Air–Al for monometallic multilayered nanostructures. Bimetallic multilayered nanostructures support strong and tunable plasmon resonance wavelengths as well as high RIS factor of 510 nm/refractive index unit (RIU) and 470 nm/RIU for Al–Air–Au and Ag-Air-Au, respectively. The MDM structures not only exhibit higher index sensitivity but also cover a wide ultraviolet–near-infrared wavelengths, making these structures very promising for index sensing, biomolecule sensing, and surface-enhanced Raman spectroscopy.  相似文献

The tunneling of surface plasmon waves between two slabs of dielectric prisms superposed on the metal surface is studied. The prism with the incident surface plasmon wave is superposed by a stack of graphene sheets. The analytical theory is built to connect the Fermi energy of graphene with the Goos-Hänchen shift of the transmitted surface plasmon waves. The obtained results may be useful for developing integral switching devices on the basis of surface plasmon polaritons.

In the present study, we have investigated the extinction spectra of coated sphere (using dipole model) with different core–shell radius, in which the core is TiO2 and the shell is made up of silver or gold nanoparticles. Nanoparticles exhibit surface plasmon resonance peak; these plasmonic peaks are highly tunable in wavelength range of 300 to 1,100 nm; in fact, the blue and red shifting of resonance peak highly depends on the core–shell thickness. The broadness of resonance peaks are analysed in terms of full width at half maxima (FWHM), and the width of these resonance peaks is also the function of core–shell radius.  相似文献

Ignition of hydrocarbon–oxygen mixtures by means of a nanosecond surface dielectric barrier discharge (NSDBD) was studied experimentally. The propagation velocity of the flame wave and the ignition delay time in mixtures of oxygen with methane, ethane, ethylene, and dimethyl ether were measured using a high-speed camera. The experiments were carried out at room temperature and gas mixture pressures in the range of 0.75–1.25 atm. It is shown that, for all hydrocarbons under study, the flame velocity decreases with reducing pressure and stoichiometric ratio, as well as when the mixture is diluted with molecular nitrogen. Theoretical analysis of the processes in the NSDBD plasma and measurements of the flame velocity in hydrocarbon-containing mixtures without plasma agree qualitatively with the measurement results, except for the increasing dependence of the flame velocity on the pressure, which is decreasing in experiments without a discharge plasma.  相似文献

We numerically investigate the buried effects of surface plasmon resonance (SPR) modes for the periodic silver-shell nanopearl dimer (PSSND) array and their solid counterparts with different buried depths in a silica substrate by means of finite element method with three-dimensional calculations. The investigated PSSND array is an important novel geometry for plasmonic metal nanoparticles (MNPs), combining the highly attractive nanoscale optical properties of both metallic nanoshell and cylindrical pore filled with a dielectric. Numerical results for SPR modes corresponding to the effects of different illumination wavelengths, absorption spectra, pore–dielectric, electric field components and total field distribution, charge density distribution, and the model of the induced local field or an applied field of the PSSND array are reported as well. It can be found that the buried MNPs with cylindrical pore filled with a dielectric in a substrate exhibit tunable SPR modes corresponding to the bonding and antibonding modes that are not observed for their solid counterparts.  相似文献

Continuous monitoring of air quality and rapid detection of pollutants are highly desirable in urban planning and development of smart cities. One of the primary greenhouse gases responsible for environmental degradation and respiratory diseases is nitrogen dioxide (NO2). Existing gas sensors for measuring NO2 concentration suffer from drawbacks such as cross-sensitivity, limited range, and short life span. On the other hand, optical sensors, in particular, surface plasmon resonance (SPR) sensors, have emerged as a preferred alternative owing to advantages like high selectivity, immunity to electromagnetic interference, and low response time. In this work, we design and simulate a NO2 sensor based on a glass waveguide coated with a gold film. Surface plasmons are excited at the interface by a 400–500-nm light source, incident at an angle of 43.16°. To enhance the sensitivity, we further coat the waveguide with three layers of carbon-silver (C–Ag) nanodots, which increases the surface plasmon field amplitude by nearly 7 times, in the absence of NO2. When NO2 concentration is varied in the range of 0–200 ppm, a corresponding change is observed in the reflected amplitude. In the absence of the C–Ag nanodots layer, the sensitivity is only 0.00042%/ppm, and on addition of C–Ag nanodots, the sensitivity increases significantly to 0.14235%/ppm which is nearly 343 times higher. These results demonstrate the efficiency of implementing nanodots in SPR sensor to detect and trace concentrations of contaminants in the gas phase.

The selective enzymatic hydrolysis of 3′,5′-di-O-acetylthyidine (1) was studied. The lipases from porcine pancreas and Aspergillus niger, and pig liver esterase, all catalysed selective hydrolysis of the 5′O-acetyl group, but the lipase from Candida cylindracea catalysed selective hydrolysis of the 3′-O-acetyl group. Highest selectivity, leading to essentially pure 3′-O-acetylthymidine, was achieved using porcine pancreatic lipase in dilute solution at pH 7.5. Provision of an artificial interface in the form of polystyrene beads led to a significant increase in the rate of hydrolysis, accompanied by a marked fall in selectivity. Other changes in the hydrolysis conditions, such as raising the concentration of substrate or adding cosolvent, also led to a fall in selectivity.  相似文献

For the development of efficient anti-cancer therapeutics against the HER receptor family it is indispensable to understand the mechanistic model of the HER receptor activation upon ligand binding. Due to its high complexity the binding mode of Heregulin 1 beta (HRG1β) with its receptor HER3 is so far not understood. Analysis of the interaction of HRG1β with surface immobilized HER3 extracellular domain by time-resolved Surface Plasmon Resonance (SPR) was so far not interpretable using any regular analysis method as the interaction was highly complex. Here, we show that Interaction Map (IM) made it possible to shed light on this interaction. IM allowed deciphering the rate limiting kinetic contributions from complex SPR sensorgrams and thereby enabling the extraction of discrete kinetic rate components from the apparently heterogeneous interactions. We could resolve details from the complex avidity-driven binding mode of HRG1β with HER3 by using a combination of SPR and IM data. Our findings contribute to the general understanding that a major conformational change of HER3 during its activation is induced by a complex sequential HRG1β docking mode.  相似文献

Soluble oligomers of the amyloid-β (Aβ) peptide play a key role in the pathogenesis of Alzheimer's disease, but their elusive nature makes their detection challenging. Here we describe a novel immunoassay based on surface plasmon resonance (SPR) that specifically recognizes biologically active Aβ oligomers. As a capturing agent, we immobilized on the sensor chip the monoclonal antibody 4G8, which targets a central hydrophobic region of Aβ. This SPR assay allows specific recognition of oligomeric intermediates that rapidly appear and disappear during the incubation of synthetic Aβ(1-42), discriminating them from monomers and higher order aggregates. The species recognized by SPR generate ionic currents in artificial lipid bilayers and inhibit the physiological pharyngeal contractions in Caenorhabditis elegans, a new method for testing the toxic potential of Aβ oligomers. With these assays we found that the formation of biologically relevant Aβ oligomers is inhibited by epigallocatechin gallate and increased by the A2V mutation, previously reported to induce early onset dementia. The SPR-based immunoassay provides new opportunities for detection of toxic Aβ oligomers in biological samples and could be adapted to study misfolding proteins in other neurodegenerative disorders.  相似文献

Two previously obtained, full-size, fully human antibodies that reversibly bind the active form of an enzyme belonging to the subtype EC 3.2.1, which is used for substitutive enzymatic therapy in lysosomal storage diseases, have been characterized by surface plasmon resonance and biolayer interferometry methods. It was shown under normal physiological conditions that the designed antibodies specifically bound with the antigen (KD ~ 10–8 M) and rapidly dissociated at neutral pH in 60% ethylene glycol while leaving the enzymatic activity unchanged. Dan ue to their properties, the developed antibodies can be used in industry as affinity ligand in the isolation of therapeutic substances of the enzyme.  相似文献

The mechanisms governing telomere replication in humans are still poorly understood. To fill this gap, we investigated the timing of replication of single telomeres in human cells. Using in situ hybridization techniques, we have found that specific telomeres have preferential time windows for replication during the S-phase and that these intervals do not depend upon telomere length and are largely conserved between homologous chromosomes and between individuals, even in the presence of large subtelomeric segmental polymorphisms. Importantly, we show that one copy of the 3.3 kb macrosatellite repeat D4Z4, present in the subtelomeric region of the late replicating 4q35 telomere, is sufficient to confer both a more peripheral localization and a later-replicating property to a de novo formed telomere. Also, the presence of β-satellite repeats next to a newly created telomere is sufficient to delay its replication timing. Remarkably, several native, non-D4Z4–associated, late-replicating telomeres show a preferential localization toward the nuclear periphery, while several early-replicating telomeres are associated with the inner nuclear volume. We propose that, in humans, chromosome arm–specific subtelomeric sequences may influence both the spatial distribution of telomeres in the nucleus and their replication timing.  相似文献

Transforming growth factor β1 (TGF-β1) is not only elevated in airways of cystic fibrosis (CF) patients, whose airways are characterized by abnormal ion transport and mucociliary clearance, but TGF-β1 is also associated with worse clinical outcomes. Effective mucociliary clearance depends on adequate airway hydration, governed by ion transport. Apically expressed, large-conductance, Ca2+- and voltage-dependent K+ (BK) channels play an important role in this process. In this study, TGF-β1 decreased airway surface liquid volume, ciliary beat frequency, and BK activity in fully differentiated CF bronchial epithelial cells by reducing mRNA expression of the BK γ subunit leucine-rich repeat-containing protein 26 (LRRC26) and its function. Although LRRC26 knockdown itself reduced BK activity, LRRC26 overexpression partially reversed TGF-β1-induced BK dysfunction. TGF-β1-induced airway surface liquid volume hyper-absorption was reversed by the BK opener mallotoxin and the clinically useful TGF-β signaling inhibitor pirfenidone. The latter increased BK activity via rescue of LRRC26. Therefore, we propose that TGF-β1-induced mucociliary dysfunction in CF airways is associated with BK inactivation related to a LRRC26 decrease and is amenable to treatment with clinically useful TGF-β1 inhibitors.  相似文献

Umbilical cord blood (UCB) is an alternative source of allogeneic hematopoietic stem cells (HSCs) for transplantation to treat various hematological disorders. The major limitation to the use of UCB-derived HSCs (UCB–HSCs) in transplantation, however, is the low numbers of HSCs in a unit of cord blood. To overcome this limitation, various cytokines or small molecules have been used to expand UCB-HSCs ex vivo. In this study, we investigated a synergistic effect of the combination of HIL-6, SR1, and UM171 on UCB-HSC culture and found that this combination resulted in the highest number of CD34+ cells. These results suggest that the combination of SR1, UM171 and HIL-6 exerts a synergistic effect in the proliferation of HSCs from UCB and thus, SR1, UM171 and HIL-6 is the most suitable combination for obtaining HSCs from UCB for clinical transplantation.  相似文献

The conjugation of polysaccharides to peptides is essential for antigen delivery and vaccine development. Herein, we show that tricine SDS-PAGE in combination with Coomassie Blue staining was adequate to determine the conjugation efficacy of a peptide (epitope 35–55 of myelin oligodendrocyte glycoprotein) to mannan. In addition, tricine SDS-PAGE and periodic acid–Schiff stains were able to monitor the redox state of mannan. Using the described protocol, more than 99.9% of a peptide containing five lysines at its N-terminus was confirmed conjugated to mannan.  相似文献

A simple, rapid and accurate liquid chromatography–tandem mass spectrometry (LC–MS/MS) method has been developed and validated for the determination of mildronate in human plasma. Following a simple protein precipitation with methanol, the analyte was separated on a C18 column by isocratic elution with methanol and 10 mM ammonium acetate (55:45; v/v), and then analyzed by mass spectrometry in the positive ion MRM mode. Good linearity was achieved over a wide range of 0.01–20 μg/mL. The intra- and inter-batch precisions (as RSD, %) were less than 7.1%. The average extraction recovery was 87.5%. The method described above has been used, for the first time, to reveal the pharmacokinetics of mildronate injection in healthy subjects. After single intravenously administration of 250, 500 and 1000 mg mildronate, the elimination half-life (t1/2) were (5.56 ± 1.55), (6.46 ± 1.07) and (6.55 ± 1.17) h, respectively. The Student–Newman–Keuls test results showed that peak plasma concentration (Cmax) and the area under the plasma concentration versus time curve from time 0 to 24 h (AUC0–24) were both linearly related to dose. The pharmacokinetics of mildronate fitted the linear dynamic feature over the dose range studied. The essential pharmacokinetic parameters of multidoses administration intravenously (500 mg, b.i.d) were as follows: t1/2 was (15.34 ± 3.14) h; Cmax was (25.50 ± 3.63) μg/mL; AUC0–24 was (58.56 ± 5.57) mg h/L. The t1/2 and AUC of multidoses administration intravenously were different from those of single-dose administration significantly. These findings suggested that accumulation of mildronate in plasma occurred.  相似文献

Otilonium bromide (OB) degrades rapidly in plasma and readily undergoes hydrolysis by the plasma esterase. In this paper, an LC–ESI-MS method has been developed for the determination of OB in human plasma. The rapid degradation of OB in plasma was well prevented by immediate addition of potassium fluoride (KF, an inhibitor of plasma esterase) to the freshly collected plasma before prompt treatment with acetonitrile. The method was validated over the concentration range of 0.1–20 ng/ml. The data of intra-run and inter-run precision and accuracy were within ±15%. The mean extraction recoveries for OB and the internal standard were higher than 93.0% and the matrix effects were negligible. The method has been successfully used in a pharmacokinetic study.  相似文献
