探索亮菌甘蔗渣固体发酵产纤维素和木质素降解酶的能力,对甘蔗渣的充分利用,以及开发新的、高效的产纤维素降解酶和木质素降解酶的微生物资源极为重要。本研究用专一的底物分别对亮菌分泌的外切葡聚糖酶、内切葡聚糖酶、β-葡萄糖苷酶、锰过氧化酶、漆酶和木质素过氧化物酶的酶活力进行测定。结果显示亮菌固态发酵可以分泌外切葡聚糖酶、内切葡聚糖酶、木质素过氧化物酶和漆酶。其酶活力随着时间的延长而增长,达到最高峰值后,开始下降。其中,漆酶酶活力最高,第55天达到最大值为(2 336±183)U/g。各种酶的最高酶活力峰值出现时间不尽相同,体现出多种酶之间很好的协同作用。因此,亮菌甘蔗渣固体发酵具有分泌纤维素和木质素降解酶的能力,尤其产具有高酶活力的漆酶,具有潜在的工业化应用前景。  相似文献

目前真菌漆酶酶活的测定方法多样,没有统一的标准,致使不同研究之间的漆酶酶活无法进行比较分析,也造成漆酶产品在酶活质量意义上的混乱。因此,对测定真菌漆酶酶活的各种不同的分光光度法进行了综述和比较分析,认为采用ABTS法作为测定漆酶酶活的方法较具合理性和科学性,建议作为漆酶酶活测定的统一方法。  相似文献

酶促反应速度是酶促反应动力学中的一个基本概念。一定条件下 ,酶催化反应的活力大小要由该反应条件下酶促反应速度来反映和确定 :酶促反应速度越小 ,酶活力越小 ;反之 ,酶促反应速度越大 ,酶活力也越大。在研究底物浓度、酶浓度、温度、pH、激活剂、抑制剂等因素对酶活力的影响时 ,都必须测定酶促反应的速度。为了排除实验中存在的一些干扰 ,一个酶的酶促反应的速度要在酶促反应进行的初期测定 ,即测定所谓的酶促反应的初速度。在教学过程中 ,学生不易弄清楚酶促反应速度与酶促反应的初速度之间的关系。我们发现 :当一般地问起酶反应速度…  相似文献

透明质酸酶是以降解透明质酸为主的糖苷酶。近年来国内外关于透明质酸酶各方面的研究日益增多,透明质酸酶的构效关系及生物学应用越来越引起人们的关注。对透明质酸酶的相关研究进行了综述,主要阐述了各类型透明质酸酶的研究概况包括人透明质酸酶、牛睾丸透明质酸酶、毒液透明质酸酶、微生物透明质酸酶。简要介绍了透明质酸酶的活性测定、酶活抑制剂及透明质酸酶制剂的应用情况,最后对透明质酸酶的研究前景进行展望。  相似文献

限制性内切酶催化的酶解反应是分子生物学实验中的常用技术.鉴于酶切反应时间长,有人提出采用微波释放的热效应催化限制性内切酶解反应,以期缩短酶解时间.但微波是否能代替传统酶切反应还需验证.本研究设计了一系列的酶切 试验,验证微波酶切是否有效.结果表明:短时间(2 min)微波炉高火处理未导致DNA降解及内切酶失活.对于质粒的单酶切和部分双酶切,短时间(2 min)微波炉处理可替代水浴加热完成酶解反应.对于部分双酶切体系,微波炉处理酶切不完全,不能替代温水浴.使用微波酶解需严格控制时间,切勿时间过长.  相似文献

一株产琼胶酶细菌的分离、鉴定及其琼胶酶基本性质   总被引:1,自引:0,他引:1  
【目的】分离海洋来源的琼胶酶产生菌,对其进行分类鉴定,并研究其所产琼胶酶的基本酶学性质,为琼胶酶的应用研究及开发利用奠定基础。【方法】通过以琼脂为唯一碳源的选择培养基分离产琼胶酶的菌株;利用16S rRNA基因序列分析、表型和生理生化特征对菌株进行鉴定;通过DNS-还原糖法测定琼胶酶活性;利用显色底物法测定琼胶酶的类型;对菌株所产琼胶酶粗酶的酶学性质进行初步研究。【结果】分离到一株产琼胶酶的菌株NTa,16S rRNA基因序列分析显示该菌株属于寡养单胞菌属(Stenotrophomonas sp.);该菌株主要产胞外琼胶酶,可分泌α-琼胶酶和β-琼胶酶;琼胶酶粗酶的最适反应温度和pH分别为40℃和7.0,并且琼胶酶在温度低于30℃,pH为7.0-9.0时稳定;Ca2+对琼胶酶粗酶具有促进作用,Ag+、Fe2+、Ba2+、Mn2+、Cu2+、Zn2+和Fe3+均可不同程度地抑制酶的活性;EDTA对琼胶酶粗酶活性具有抑制作用;琼胶酶粗酶对检测的抑制剂、去垢剂及变性剂有较好的抗性。【结论】海洋细菌Stenotrophomonas sp.NTa是一种新型的产琼胶酶菌株,可同时分泌α-琼胶酶和β-琼胶酶,具有潜在开发利用价值。  相似文献

Juncus Linn. is a cosmopolitan genus with about 240 species, widely distributed in both hemispheres, but most abundant in the frigid and temperate zones. In the present paper, a revision of this genus in China is made, and a key to the species is provided. The utility of 36 presumptive determinative tests for 32 pathovars of Pseudomonas syringae was investigated. A total of 395 strains was examined. Most strains of 12 of these pathovars ( Ps. syringae pv. cannabina, Ps. syr. delphinii, Ps. syr. glycinea, Ps. syr. helianthi, Ps. syr. lachrymans, Ps. syr. mori, Ps. syr. morsprunorum, Ps. syr. phaseolicola, Ps. syr. 'porri', Ps. syr. papulans, Ps. syr. savastanoi and Ps. syr. tabaci ) formed clusters when test data were compared by centroid analysis. Pseudomonas syr. syringae, Ps. syr. aptata, Ps. syr. atrofaciens, Ps. syr. dysoxyli and Ps. syr. japonica formed a single cluster, indicating their possible synonymy. Strains of Ps. syr. antirrhini and Ps. syr. tomato were indistinguishable, as were those of Ps. syr. garcae and Ps. syr. oryzae. Strains of Ps. syr. berberidis, Ps. syr. coronafaciens, Ps. syr. eriobotryae, Ps. syr. maculicola, Ps. syr. passiflorae, Ps. syr. pisi and Ps. syr. striafaciens and Ps. syr. tagetis did not form distinguishable clusters.
The tests which reliably differentiated pathovars are recorded in a determinative scheme.  相似文献
