Mercury (Hg) and selenium (Se) determinations were carried out to evaluate human exposure to those elements through fish consumption in Spain and Portugal. Atomic fluorescence spectroscopy (AFS) was applied in a cold vapor mode for total mercury quantification and was also hyphenated to gas chromatography (GC) to achieve the speciation of organomercurial species in fish samples. The results obtained show the highest concentration of Hg in swordfish and tuna (0.47+/-0.02 and 0.31+/-0.01 microg g-1, respectively), and a much lower concentration in sardine, mackerel shad, and octopus (0.048+/-0.002, 0.033+/-0.001, and 0.024+/-0.001 microg g-1, respectively). The determination of alkyl mercury compounds revealed that 93-98% of mercury in the fish samples was in the organic form. Methylmercury (MeHg) was the only species found in the three fish species with higher mercury content.Total selenium concentration was high in sardine, swordfish, and tuna (0.43+/-0.02, 0.47+/-0.02, and 0.92+/-0.01 microg g-1, respectively), but low in mackerel shad and octopus (0.26+/-0.01 and 0.13+/-0.01 microg g-1, respectively). Speciation of selenium compounds was done by high-performance liquid chromatography in conjunction with inductively coupled plasma mass spectrometry (LC-ICP-MS). Selenomethionine (SeMet) was the only selenium compound identified in the fish samples with higher selenium content.Among the fish species studied, sardine had the most favourable Se:Hg and SeMet:MeHg molar ratios; therefore, its consumption seems to be preferable.  相似文献

The influence of dietary selenium (Se) on mercury (Hg) toxicity was studied in weanling male Long Evans rats. Rats were fed AIN-93G-based low-Se torula yeast diets or diets augmented with sodium selenite to attain adequate- or rich-Se levels (0.1, 1.0 or 15 μmol/kg, respectively) These diets were prepared with no added methylmercury (MeHg) or with moderate- or high-MeHg (0.2, 10 or 60 μmol/kg, respectively). Health and weights were monitored weekly. By the end of the 9-week study, MeHg toxicity had impaired growth of rats fed high-MeHg, low-Se diets by approximately 24% (p < 0.05) compared to the controls. Growth of rats fed high-MeHg, adequate-Se diets was impaired by approximately 8% (p < 0.05) relative to their control group, but rats fed high-MeHg, rich-Se diets did not show any growth impairment. Low-MeHg exposure did not affect rat growth at any dietary Se level. Concentrations of Hg in hair and blood reflected dietary MeHg exposure, but Hg toxicity was more directly related to the Hg to Se ratios. Results support the hypothesis that Hg-dependent sequestration of Se is a primary mechanism of Hg toxicity. Therefore, Hg to Se molar ratios provide a more reliable and comprehensive criteria for evaluating risks associated with MeHg exposure.  相似文献

BackgroundThe main exposure route to methylmercury (MeHg) is from eating fish and shellfish containing this compound. Since 2004, women of childbearing age in Spain have been urged not to eat some species (eg, tuna, shark, and swordfish), instead choosing low-MeHg seafood as part of a healthy diet.ObjectiveTo describe maternal total blood mercury (THg) and serum selenium (Se) in a cohort of pregnant women living in Spain as it relates to fish intake during the three trimesters and to assess whether or not Spanish women of childbearing age follow the recommendations listed in fish advisories and choose fish species with lower mercury levels.MethodsWe studied 141 female volunteers of childbearing age (16–45 years), interviewing all participants about their overall eating habits and seafood intake. Hg and Se levels were tested using cold-vapor atomic absorption spectrometry (CVAAS) and electrothermal atomic absorption spectrometry (ETAAS), respectively.ResultsAverage THg levels in pregnant women were 2.89 μg/L (standard deviation [SD], 2.75 μg/L, geometric mean [GM], 2.19 μg/L), and THg GM was positively associated with fish intake. Mean Se levels in pregnant women were 73.06 μg/L (SD, 13.38 μg/L), and Se levels were found to increase with tuna intake. In 16 (12%) pregnant women, THg was higher than the level recommended by the U.S. Environmental Protection Agency (EPA) (6.4 μg/L). A positive association was also found between THg and serum Se.ConclusionWomen of childbearing age in Spain had higher THg levels than women in other Western studies. Our study observed that 12% of women had THg levels above the safety limit set by the EPA (6.4 μg/L), and 31% had levels above the relevant benchmark level of 3.5 μg/L suggested by various researchers.  相似文献

The levels of mercury (Hg) and selenium (Se) and their correlation in human scalp hair of 29 pairs of pregnant women and their newborns living at a high-Hg and low-Se area, the Second Song-Hua-Jiang River System, have been studied by instrumental neutron activation analysis. Our results indicate that the Hg level in infant hair samples are roughly close to that in their mothers, whereas the Se level in infants is much higher. Furthermore, the molar ratios of Se/Hg in newborns are also 40% higher than those in mothers. It demonstrates that infants are able to absorb more selenium from their mothers’ bodies to protect against or alleviate the intoxication of Hg. This “autoprotective mechanism” likely plays a critical role during fetal development. The possible chemical species of Hg and Se in hair is also discussed.  相似文献

Samples of 7 species of piscivorous, omnivorous, and herbivorous fish caught at 12 different sites on the Madeira River, Amazon Basin, were analyzed for selenium and mercury. Selenium was determined by anodic stripping voltammetry and mercury by cold vapor atomic absorption spectrophotometry. The means for selenium concentrations ranged from 0.49 to 3.11 nmol/g and for mercury from 0.41 to 6.66 nmol/g depending on the fish species. The molar ratios of Hg:Se increased according to the fish trophic level. Piscivorous species had the highest mean ratio (4.0) and herbivorous species the lowest (0.9). There was a positive and statistically significant correlation between selenium and mercury concentrations for the herbivorous species (r = 0.716;p = 0.0088) not seen for omnivororus and piscivorous species (r = -0.2032;p = 0.3407). These findings are significant for the fish-eating population of the Madeira River because the ingestion of mercury would always be in excess of selenium.  相似文献

Seafood is an important source of nutrients for fetal neurodevelopment. Most individuals are exposed to the toxic element mercury through seafood. Due to the neurotoxic effects of mercury, United States government agencies recommend no more than 340 g (12 oz) per week of seafood consumption during pregnancy. However, recent studies have shown that selenium, also abundant in seafood, can have protective effects against mercury toxicity. In this study, we analyzed mercury and selenium levels and selenoprotein mRNA, protein, and activity in placenta of a cohort of women in Hawaii in relation to maternal seafood consumption assessed with dietary surveys. Fish consumption resulted in differences in mercury levels in placenta and cord blood. When taken as a group, those who consumed no fish exhibited the lowest mercury levels in placenta and cord blood. However, there were numerous individuals who either had higher mercury with no fish consumption or lower mercury with high fish consumption, indicating a lack of correlation. Placental expression of selenoprotein mRNAs, proteins and enzyme activity was not statistically different in any region among the different dietary groups. While the absence of seafood consumption correlates with lower average placental and cord blood mercury levels, no strong correlations were seen between seafood consumption or its absence and the levels of either selenoproteins or selenoenzyme activity.  相似文献

The potential toxicity of mercury (Hg) content in fish has been widely evaluated by the scientific community, with Methylmercury (MeHg) being the only legislated species (1 mg kg−1, maximum concentration allowed in predatory fish). On the other hand, selenium (Se) is recognized to decrease its toxicity when both elements are simultaneously administrated. In the present paper, the total content of Se and Hg and their species in fish of high consumption, such as tuna, swordfish, and sardine, have been evaluated. The percentage of MeHg is higher than 90% of total Hg content. The results show that, for all of them, the Se/Hg ratio is significantly higher than one, being the maximum ratio for sardine. As only studying the bioaccessible fraction the extent of a toxic effect caused by an element can be predicted, the bioaccessibility of both analytes through an in vitro digestion method has been carried out. The results show that MeHg in all fishes is very low bioaccessible in both gastric and intestinal digestion. Because the MeHg bioaccessible fraction might be correlated to the Se content, the potential toxicity cannot be only related to the total Hg content but also to Se/Hg ratio.  相似文献

The precise mechanisms of mercury accumulation and retention are still unclear. Generally, the association of mercury with selenium is used to explain these phenomena. It seems that the presence of coaccumulated endogenous Se can protect cells from the harmful effects of Hg. However, as speculated by some authors, this binding of Se to Hg can also result in a relative deficiency of biologically available Se needed for selenoenzyme syntheses. Deriving from the assumption that Hg deposited in tissues is bound to Se in a 1:1 ratio, the quantity of non-Hg bound Se could be calculated by the difference between the molar contents of the two elements (Semol–Hgmol). In this study we applied such an approach to the data from our previous investigation, where Hg and Se concentrations were determined in autopsy samples of mercury exposed retired Idrija mercury mine workers, Idrija residents living in a Hg contaminated environment and a control group with no known Hg exposure from the environment. Based on these data we tried to estimate the influence of Hg exposure on the physiologically available selenium content in selected tissues, particularly endocrine glands and brain tissues. Comparing the calculated values of (Semol– Hgmol) it was found that for Idrija residents the values were similar to those of the control group and as expected, diminished values were found in some mercury-loaded organs of retired Idrija miners. It could be speculated that in Idrija residents Hg sequestration of selenium is sufficiently compensated by increased Se levels, but that particularly in active miners and in some organs of retired miners, the activity and/or synthesis of selenoenzymes could be disturbed. We report here some results of a long-term (19 month) study with cats fed methylmercury (MeHg) in nutritionally balanced diets based on fish. By using either freshwater pike (low in Se) or canned tuna (high in Se) as the major protein source, basal diets with low levels of MeHg were prepared having different Se content, all Se being of natural origin. The basal diets produced no signs of toxicity or pathological changes over the l9-month period. In cats fed basal diets spiked with medium or high levels of MeHg, evidence for delayed onset of toxic effects from the added MeHg was observed with the tuna diets compared to pike diets. In brain, muscle, and blood, the activity of GSH peroxidase, a selenoenzyme, was decreased by Hg. In liver, substantial accumulation of Hg with Se occured (molar Hg/Se ratio approximately 1.4 to 1.8) but GSH peroxidase activity was unaffected. We suggest that the coaccumulation of Hg and Se in liver measures the extent to which MeHg has been metabolically transformed by metabolism to Hg++, and inactivated by deposition as a Hg/Se complex of low bioavailability. The accumulation of Hg and Se in liver was much greater in cats fed tuna compared to pike, out of proportion to the relatively small differences in Hg and Se content of the tuna and pike basal diets. Some mechanisms are described by which selenium, vitamin E, and other factors might facilitate MeHg breakdown to inorganic Hg during long term low level exposure to MeHg.  相似文献

Selenium–mercury interactions were most extensively studied in relation to alleviation of Hg toxicity by added selenium. This presentation considers the influence of mercury on endogenous selenium, on its tissue and cellular “status” after lifelong or acute exposure to mercury vapor (Hgo). Discussed are data obtained from (1) humans living near or working in a mercury mine, and (2) rats experimentally exposed in the mine. Mercury vapor is unique—or similar to methylmercury—because of its ability to penetrate cell membranes and so invade all cells, where it is oxidized in the biologically active form (Hg++) by catalase. Such in situ-generated ions can react with endogenously generated highly reactive Se metabolites, like HSe−, and render a part of the selenium unavailable for selenoprotein synthesis. Data on human populations indicate that in moderate Hg exposure combined with an adequate selenium supply through diet, Se bioavailability can be preserved. On the other hand, the results of an acute exposure study emphasize the dual role of selenium in mercury detoxification. Besides the well-known Se coaccumulation through formation of nontoxic Hg–Se complexes, we observed noticeable Se (co)excretion, at least at the beginning of exposure. The higher Hg accumulation rate in the group of animals with lower basal selenium levels can also point to selenium involvement in mercury excretion. In such conditions there is a higher probability for decreased selenoprotein levels (synthesis) in some tissues or organs, depending on the synthesis hierarchy.  相似文献

The aim of this study was to determine the biomagnification of mercury through the principal prey of the blue shark, Prionace glauca, off the western coast of Baja California Sur, Mexico, as well as the relationship between mercury and selenium in blue sharks. High levels of mercury were found in shark muscle tissues (1.39?±?1.58?μg/g wet weight); these values are above the allowed 1.0?μg/g for human consumption. The mercury to selenium molar ratio was 1:0.2. We found a low correlation between mercury bioaccumulation and shark size. Juveniles have lower concentrations of mercury than adults. Regarding the analyzed prey, the main prey of the blue shark, pelagic red crab, Pleuroncodes planipes, bioaccumulated 0.04?±?0.01?μg/g Hg wet weight, but the prey with higher bioaccumulation was the bullet fish Auxis spp. (0.20?±?0.02?μg/g wet weight). In terms of volume, the red crab P. planipes can be the prey that provides high levels of mercury to the blue shark.  相似文献

We have determined the trace element composition of anti-influenza virus mushrooms using atomic absorption spectrophotometer. The elements present in greater concentration in Ganoderma lucidum samples are selenium, iron, and zinc, with selenium being the element with the highest concentration of all, at 416 ± 38.5 mg/kg; in Cordyceps militaris samples are iron, selenium, and zinc, with iron being the element with the highest concentration of all, at 291 ± 20.9 mg/kg; in Kuehneromyces mutabilis samples are selenium, iron, and manganese, with selenium being the element with the highest concentration of all, at 203 ± 9.8 mg/kg; in Inonotus hispidus samples are zinc, selenium, and iron, with zinc being the element with the highest concentration of all, at 194 ± 16.9mg/kg; in the Collybia maculata samples are iron, selenium, and zinc, with iron being the element with the highest concentration of all, at 274 ± 22.2 mg/kg, respectively. The average metal concentrations in mushrooms decreases in the order: selenium > iron > zinc > chromium > manganese > copper > magnesium > lead. After the mice were administered (orally) with mushroom extracts for 8 weeks and inoculated intranasally with viral suspension, element levels in serum were also measured. Highly significantly increased values of Se, Zn, and Mg in the serum of mice supplemented with anti-influenza virus mushrooms were a characteristic finding. Se, Zn, and Mg present in mushrooms may play a direct or indirect role in their anti-influenza virus nature. They may provide prophylactic protection against influenza infection via stimulation of host innate immune response.  相似文献

Vertebrates, including humans, can experience adverse effects from mercury consumed in fish. Humans often prefer large predatory fish that bioaccumulate high mercury levels. Recent attention has focused on the role of selenium countering mercury toxicity, but there is little research on the selenium:mercury molar ratios in freshwater fish. We examine selenium:mercury molar ratios in freshwater fish from Tennessee at Poplar Creek which receives ongoing inputs of mercury from the Department of Energy's Oak Ridge Y-12 facility. Our objective was to determine variation of the ratios within species that might affect the protectiveness of selenium against mercury toxicity. Within species, the ratio was correlated significantly and positively with fish length only for two species. There was great individual variation in the selenium:mercury molar ratio within each species, except striped bass. The lack of a clear relationship between the selenium:mercury molar ratio and fish length, and the intraspecific variation, suggests that it would be difficult to use the molar ratio in predicting either the risk from mercury toxicity or in devising consumption advisories.  相似文献

The bioavailabilities of selenium (Se) from Se-rich fish species and Se-rich wheat were compared in a study involving 32 healthy volunteers. Initial serum Se values were 109±16 μg/L (mean±SD). For 6 wk, one group (n=11) included Se-rich bread in their diet, bringing daily average intake of Se up to 135±25 μg/d. Another group (n=11) consumed Se-rich fish daily (average Se intake: 115±31 μg/d), whereas the control group (n=10) ate their normal diet, providing 77±25 μg Se/d. Serum Se increased by 17% (P<0.01), and platelet Se increased by 30% (P<0.01) in the wheat group. Although platelet Se decreased by 11% in the fish group, no changes in serum and platelet Se in the fish or control group reached statistical significance. Glutathione peroxidase (EC 1.11.1.9; GSH-Px) activity in serum and platelets did not change during the study, nor did platelet mercury (Hg) content. Since the dietary intake of Hg, arsenium (As), and fatty acids could not satisfactorily explain the lack of response in the fish group, the results are indicative of low bioavailability of fish Se in humans. The bioaccumulation and biomagnification of mercury (Hg) and selenium (Se) were investigated in sub-tropical freshwater food webs from Burkina Faso, West Africa, a region where very few ecosystem studies on contaminants have been performed. During the 2010 rainy season, samples of water, sediment, fish, zooplankton, and mollusks were collected from three water reservoirs and analysed for total Hg (THg), methylmercury (MeHg), and total Se (TSe). Ratios of δ13C and δ15N were measured to determine food web structures and patterns of contaminant accumulation and transfer to fish. Food chain lengths (FCLs) were calculated using mean δ15N of all primary consumer taxa collected as the site-specific baseline. We report relatively low concentrations of THg and TSe in most fish. We also found in all studied reservoirs short food chain lengths, ranging from 3.3 to 3.7, with most fish relying on a mixture of pelagic and littoral sources for their diet. Mercury was biomagnified in fish food webs with an enrichment factor ranging from 2.9 to 6.5 for THg and from 2.9 to 6.6 for MeHg. However, there was no evidence of selenium biomagnification in these food webs. An inverse relationship was observed between adjusted δ15N and log-transformed Se:Hg ratios, indicating that Se has a lesser protective effect in top predators, which are also the most contaminated animals with respect to MeHg. Trophic position, carbon source, and fish total length were the factors best explaining Hg concentration in fish. In a broader comparison of our study sites with literature data for other African lakes, the THg biomagnification rate was positively correlated with FCL. We conclude that these reservoir systems from tropical Western Africa have low Hg biomagnification associated with short food chains. This finding may partly explain low concentrations of Hg commonly reported in fish from this area.  相似文献

Animal studies indicate that the toxic effects of methylmercury (MeHg) exposures increase when selenium (Se) status is low. Toxicity is directly proportional to Hg/Se molar ratios in critical tissues such as brain and increase dramatically as molar ratios exceed 1:1. In this study, we examined the nail as a biomonitor of Hg/Se molar ratios in kidney, liver, and brain tissues of weanling male Long-Evans rats fed controlled diets containing varying amounts of Se and MeHg. A survey of mercury (Hg) and selenium (Se) contents was performed in fish collected from lakes located in two National Parks of the northern patagonian Andean range. Two native species, catfish (Diplomystes viedmensis) and creole perch (Percichthys trucha), and three introduced species, brown trout (Salmo trutta), rainbow trout (Oncorhynchus mykiss), and brook trout (Salvelinus fontinalis), were caught from lakes Nahuel Huapi, Moreno, Traful, Espejo Chico, and Guillelmo belonging to Nahuel Huapi National Park and from lakes Futalaufquen and Rivadavia, Los Alerces National Park. In lake Moreno, fish diet items were analyzed and rainbow trout grown in a farm. Hg and Se were measured in muscle and liver tissues by instrumental neutron activation analysis. The average concentrations in muscle of Hg for all species, ages, and lakes are between 0.4 to 1.0 μg g−1 dry weight (DW) with a few fish, mainly native, exceeding the United States Environmental Protection Agency health advisory for freshwater fish limited consumption, and from 0.8 to 1.5 μg g−1 DW for Se. Average concentrations in liver of Hg in all species range from 0.4 to 0.9 μg g−1 DW. Brown trout, the top predator in these lakes, showed the lowest average Hg burden in both tissues. Se concentrations in the liver of brown and rainbow trout, up to 279 μg g−1 DW, are higher than those expected for nearly pristine lakes, exceeding 20 μg g−1 DW, the threshold concentration associated with Se toxicity. These species show lower Hg contents in muscle, suggesting a possible detoxification of Hg by a Se-rich diet. Creole perch and velvet catfish livers have lower Se concentrations, with a narrower span of values (2.3 to 8.5 μg g−1 and 3.3 to 5.5 μg g−1 DW respectively).  相似文献

The objective of the present study is to calculate linear regressions between a mother and her child with respect to their selenium concentration (ng/g) in the following traits: maternal blood and umbilical cord blood, maternal and child hair, maternal milk and child umbilical cord blood, maternal milk and meconium, maternal blood plasma, and child meconium. The data were collected at Research Hospital of the University of Yüzüncü Yıl from 30 pairs of mothers and their newborn baby. The mean maternal serum Se level in 30 mothers was 68.52 ± 3.57 ng/g and cord plasma level was 119.90 ± 18.08 ng/g. The Se concentration in maternal and neonatal hair was 330.84 ± 39.03 and 1,124.76 ± 186.84 ng/g, respectively. The Se concentration of maternal milk at day 14 after delivery was determined as 68.63 ± 7.78 ng/g (n = 13) and the concentration of Se was 418.90 ± 45.49 ng/g (n = 22) for meconium of neonatal. There was no significant difference between maternal blood and milk Se levels. However, hair Se concentration was significantly higher than milk and maternal blood Se level. For each trait comparison, the average absolute difference in log10-transformed Se concentration was calculated between a mother and her child. The observed average absolute difference was compared with a test distribution of 1,000 resampled bootstrap averages where the number of samples was maintained but the relationship between a mother and her child was randomized among samples (α = 0.05).  相似文献

The seasonal occurrence of black marlin, Istiompax indica, around Yonaguni Island, southwestern Japan, was analyzed. Black marlin was abundant from April to July, and the majority of landed fish were female (97%). None of these females was reproductively mature. The condition factor of females increased from April to July. Of 56 black marlin examined, 21 individuals contained 12 prey species; the most important prey species was skipjack tuna, Katsuwonus pelamis, and one neonatal silky shark, Carcharhinus falciformis, was also found. These results indicate that the occurrence of black marlin off Yonaguni Island was associated with feeding and not with spawning.  相似文献
