Regeneration competence of aerial roots of Cleisostoma raeimeferum (Orchidaceae) from in vivo and in vitro sources was tested. The protocorm-Iike bodies and shoot buds were obtained from 2 w old in vivo grown aerial roots and 20 wold in vitro grown roots on Murashige and Skoog medium containing sucrose (3%) (w/v), casein-hydrolysate (2 g l?1), coconut water (15%) (v/v), citric acid (200 mg l?1) and different plant growth regulators. The morphogenetic response from in vivo grown roots was poor and only 20% of the cultures yielded protocorm-like bodies and shoot buds on medium containing IAA (2 µM) and kinetin (2 µM) in combination after 75 d of culture. While 100% morphogenetic response was exhibited by in vitro grown roots on MS medium enriched with IAA (1 µM) and kinetin (1 µM) in combination only after 25 d of culture initiation. The response initiated at the cut ends of the roots and subsequently the entire root length was taken over. Both IAA and kinetin singly stimulated mostly callusing of the explants. The rooted plantlets and multiple shoot buds were obtained after 30 d of culture from protocorm-like bodies and shoot buds on basal medium enriched with IAA (2 µM) and kinetin (6 µM) in combination. The well developed rooted plants could be obtained for transferring to potting mix after ~24 w of culture initiation.  相似文献

A methodology to regenerate whole plants of Pinus radiata from apical meristems of 3- and 7-year-old trees was developed. Meristematic domes with two or three leaf primordia were excised from surface-sterilized branch tips of field-grown plants and cultured in LP medium with half strength macronutrients (1/2 LP) and full strength micronutrients. The early growth of meristems required approximately 12 weeks, including a recovery stage during the first 2 weeks. After 8 weeks, some meristems developed abnormal phenotypes and died during the subsequent stages of development. However, healthy meristems elongated and formed shoots when they were transferred to LP medium supplemented with MS vitamins, 30 mg l–1 casein hydrolysate, and 0.4 g l–1 agar plus 2.85 g l–1 Gelrite. Meristems that developed vigorous shoots were used for rooting experiments when they were 2 cm in length. Whole plants were obtained after 5 days of root induction in water-agar medium containing 8.2 M IBA and 5.4 M NAA and 1 month culture in LP medium with 10 g l–1 sucrose. Plants regenerated from meristems were further propagated by rooting of cuttings. Of the rooted cuttings, 10% were morphologically juvenile.  相似文献

The influence of several endogenous factors on micropropagation of mature elite trees of beech (Fagus sylvatica L.) has been investigated. February was the most beneficial month for explantation of dormant buds, since infection with endogenous bacteria was still low and in vitro growth of the plant material was the highest. The genotypes tested gave different results concerning their growth on various tissue culture media. Out of 51 mature grafted genotypes only seven could be established in vitro. Grafting of branches of mature stock plants on juvenile rootstocks resulted in a high increase of multiplication rate compared with corresponding mature material, which could not be subcultured in vitro. Apical buds, larger than 20 mm, from 1-year-old shoots proved to be the most suitable explant source. Nodal explants, excised from young shoots from mature trees of Syzygium cuminii, were cultured on MS medium supplemented with different concentrations of BA or kinetin. Among these, BA (0.5 or 1.0 mg l–1) induced greening and opening of the incipient shoot buds, which however did not elongate. Elongation of the shoot buds was facilitated on MS medium with 1.0 mg l–1 BA supplemented with casein hydrolysate (1.5 g l–1) or glutamine (200 mg l–1). Nodal explants (microcuttings), taken from shoots developed in vitro, also developed multiple shoots when cultured on MS with1 mg l–1 BA. These explants did not require an additional supply of reduced nitrogen, for further normal development. Shoots developed from explants from mature trees and microcuttings were rooted by sub-culturing them on Knop's medium supplemented with 2% sucrose and 1 mg l–1 IAA. The plants that developed in vitro were planted in soil and were transferred to the field after an acclimatization period of 7–8 months. These plants have been thriving well for more than three years and have no apparent phenotypic aberrations.  相似文献

This work was conducted to study the effect of sucrose and 2,4-D combinations on induction of haploid plants of a summer squash cultivar through anther culture; therefore, sucrose was used at 30, 60, 90, 120 and 150 g l−1and 2, 4-D was used at 0.1, 1.0, 2.5 and 5.0 mg l−1on solid MS anther culture medium. Anthers at the mid or late uninucleate microspore stage without filament were excised from sterilized buds and plated on 20 different induction media. The most plantlets resulted from the induction medium supplemented with 150 g l−1sucrose and 5 mg l−12, 4-D. Root tips from 20 plantlets were cytologically examined under a light microscope. The results revealed ten diploid (2n>= 2x= 40) and ten haploid (2n= x= 20) plants. This revised version was published online in June 2006 with corrections to the Cover Date.  相似文献

An efficient system for the in vitro plant and shootregeneration of Lilium longiflorum was developed andaccomplished using transverse thin cell layers (tTCL) of young stems.tTCLs were cut transversely along young stems from which the shoot-tipshad been removed. Sections were measured accurately using a graded gridand were cut in 4 mm × 4 mm × 1 mm cubes, eliminatingepidermal tissue, and were cultured on one-half MS medium containing 8 gl–1 agar, different sucrose concentrations (10, 20, 30 or 40g l–1), and with or without 1 mg l–1 activatedcharcoal (AC). Plants formed on the surface of tTCLs within 60 days onone-half MS medium containing 8 g l–1 agar and 20 gl–1 sucrose. Sections of 1 mm taken just below the apicalarea developed buds within 15 days, whereas the sections closer to thebase required about 45 days. Shoot regeneration was enhanced whensucrose concentration was used at 30 or 40 g l–1 after 60days of culture. No root formation occurred. Both shooting and rootingoccurred when sucrose was used at 20 g l–1. The plantletswere transferred to soil and grew well under greenhouseconditions.  相似文献

Plantlet regeneration via organogenesis was achieved in callus cultures derived form mature leaves, stems and leaves, petioles and roots of young seedling of Psoralea corylifolia on Murashige and Skoog medium supplemented with 2.5–3.0 mg L-1 BA, 1.0 mg L-1 NAA and 3% (w/v) sucrose. The rate of shoot bud regeneration was positively correlated with the concentration of hormones in the nutrient media. Shoot buds regenerated more readily from juvenile explants (seedling source) as compared to the mature explants. Addition of adenine sulphate (5 mg L-1) to the culture medium increased the growth of shoot buds. Optimum responses were obtained in hypocotyl and leaf explants using NAA in combination with BA, the highest rate of shoot bud regeneration being in hypocotyl explants. Rooting was readily achieved on the differentiated shoots on MS basal media without growth regulators. Regenerated plantlets were successfully established in the greenhouse.  相似文献

Mature trees of European grey alder (Alnus incana) were micropropagated on a modified MS medium containing 2.5 M BA, 6.2 mM (500 mg l-1) NH4NO3 and 1.5% glucose. Prior to in vitro culture, mature scions were multiplied through grafting and cutting techniques. Shoot tips from cuttings were established in vitro. After six months of culture, shoots were rooted either in vitro or in vivo and plantlets were transferred to greenhouse conditions.  相似文献

Organogenesis was induced in callus derived from mature zygotic embryos of six families of loblolly pine (Pinus taeda L.) within 24 weeks of culture. Elongation of adventitious buds was achieved on TE medium supplemented with 0.5 mg l−1 indole-3-butyric acid (IBA) and 2 mg l−1 6-benzyladenine (BA). The most suitable medium for root formation proved to be TE medium supplemented with 0.1 mg l−1 IBA, 1 mg l−1 BA, and 0.5 mg l−1 gibberellic acid (GA3). One hundred and sixty-nine regenerated plantlets were transferred to a perlite:peatmoss:vermiculite (1:1:1) soil mixture, and 98 plantlets survived in the field. Shoot apical meristems of seedling and mature eastern white pine trees were excised and grownin vitro. Placing the meristems on filters instead of directly on agarose-solidified nutrient medium enhanced survival of both juvenile and mature meristems. Applying forcing treatments to mature branches improved survival and growth of dissected meristems compared with meristems from non-forced branches in experiments conducted over two years. No consistent differences were observed among 2-, 4-, and 6-week forcing treatments. Including 5.37 nM (0.001 mg l-1) l-naphthaleneacetic acid in the culture medium did not affect meristem survival or growth. Some meristems from seedlings grew rapidly, produced primary leaves, underwent internode elongation, and in three cases, produced adventitious roots. Meristems from mature trees did not grow as rapidly as seedling meristems. The leaves produced by mature meristems appeared to be scale leaves and a few of these had brachyblast primordia in the axils. The shoots derived from mature meristems did not produce adventitious roots.  相似文献

Summary Explants of axillary buds excised from mature adult stems of Swainsona formosa (G. Don) J. Thompson (syn. Clianthus formosus) were cultured on Murashige and Skoog medium supplemented with a range of auxins, cytokinins, and sucrose concentrations. Auxins did not increase shoot or bud numbers above controls, and 2,4-dichlorophenoxyacetic acid was the only auxin to significantly increase callus production. Benzyladenine or thidiazuron incorporated into the medium at 0.1 μM stimulated shoot and bud production, and shoot growth occurred following removal of cytokinins from the medium after 4 wk. Shoot number increased linearly with sucrose concentration up to 40 g l−1, but shoot height and the number of cytokinin-induced buds were optimal at sucrose levels of 20–30 g l−1. Roots were initiated in vitro following treatment of cuttings with 0.1% indole-3-butyric acid and 0.1% α-naphthaleneactic acid. Plantlets were successfully established in soil but were plagiotropic and exhibited distichous phyllotaxy.  相似文献

Shoot tips and leafy bud fragments removed from offshoots of adult date palms (Phoenix dactylifera L.) were cultured on a nutrient medium containing the Murashige and Skoog inorganic salts, 453 M 2,4-dichlorophenoxyacetic acid, 14.8 M N6-(2-isopentenyl)adenine and 3 g l-1 activated charcoal to develop nodular callus after 8 months of culture. Callus was cultured in agar-solidified and stationary or shaken liquid media containing half-strength MS inorganic salts, 3 g l-1 activated charcoal and different sucrose concentrations to study the influence of these factors on somatic embryogenesis. Nodal segements were taken from juvenile shoots of mature 100 year-old trees of saucer magnolia (Magnolia x soulangiana Soul.-Bod.) and cultured on Standardi and Catalano medium supplemented with 1.33 μmol·dm−3 BA, 0.54 μmol·dm−3 NAA, 58 μmol·dm−3 sucrose and 6.0 g·l−1 agar-agar. After 8 weeks, separated shoots were transferred to rooting medium with half-strength macronutrients (basal medium) supplemented with 0.3% activated charcoal and one of carbohydrates: arabinose, cellulose, fructose, galactose, glucose, lactose, mannose, rhamnose, ribose, sorbose, sucrose or xylose at 20 g·dm−3 and 7.0 g·dm−3 agar-agar. After 13 weeks of culture, shoot number, fresh and dry weight of shoots and roots, total root length and number of roots/per shoot were recorded. Percentages of rooted shoots were calculated. Fructose, mannose and xylose were the most effective carbon source on shoot proliferation followed by sucrose. The rooting response was induced by cellulose and xylose. Arabinose, rhamnose and sorbose inhibited root formation. The number of adventitious roots produced per shoot was stimulated by cellulose and xylose. Total biomass (shoot plus roots) of the plantlets was the highest at fructose and cellulose.  相似文献

Simple, reproducible, high frequency, improved plant regeneration protocol in Eastern Cottonwood (Populus deltoides) clones, WIMCO199 and L34, has been reported. Initially, aseptic cultures established from axillary buds of nodal segments from mature plus trees on MS liquid medium supplemented with 0.25 mg l−1 KIN and 0.25 mg l−1 IAA. Nodal and internodal segments were found to be extra-prolific over shoot apices during course of aseptic culture establishment, while 0.25 mg l−1 KIN concentration played a stimulatory role in high frequency plant regeneration. Diverse explants, such as various leaf segments, internodes, and roots from in vitro raised cultures, were employed. Direct plant regeneration was at high frequency of 92% in internodes, 88% in leaf segments, and 43% in root segments. This led to the formation of multiple shoot clusters on established culture media with rapid proliferation rates. Many-fold enhanced shoot elongation and growth of the clusters could be achieved on liquid MS medium supplemented with borosilicate glass beads, which offer physical support for proliferating shoots leading to faster growth in comparison to semi-solid agar or direct liquid medium. SEM examination of initial cultures confirmed direct plant regeneration events without intervening calli. In vitro regenerated plants induced roots on half-strength MS medium with 0.15 mg l−1 IAA. Rooted 5- to 6-week-old in vitro regenerated plants were transferred into a transgenic greenhouse in pots containing 1:1 mixture of vermicompost and soil at 27 ± 2°C for hardening and acclimatization. 14- to 15-week-old well-established hardened plants were transplanted to the field and grown to maturity. The mature in vitro raised poplar trees exhibited a high survival rate of 85%; 4-year-old healthy trees attained an average height of 8 m and an average trunk diameter of 25 cm and have performed well under field conditions. The regeneration protocol presented here will be very useful for undertaking genetic manipulation, providing a value addition to Eastern Cottonwood propagation in future.  相似文献

Rapid micropropagation of Trichopus zeylanicus Gaertn. subsp. travancoricus Burkil ex Narayanan, a rare ethnomedicinal herb endemic to the Western Ghats of southern India, was achieved by culturing shoot tips (0.3–0.5 cm) of 2-month-old axenic seedlings on Woody Plant Medium. Among the cytokinins tested, only BAP induced callus-free multiple shoot bud formation, with a maximum of 8.5±0.4 buds per explant being obtained with 2.0 mg.l–1 BAP after 8 weeks of culture. Shoot tips containing proliferated buds were divided and subcultured on medium containing 0.2 mg.l–1 BAP to produce 12.0±1.0 shoots per explant in 6 weeks. Excision of buds after culture initiation, with subculture of the debudded basal tissue in 2 successive passages yielded 20.0±1.0 and 13.5±0.5 buds per explant respectively. Each bud cultured in turn for 4 weeks on WPM with 1.0 mg.l–1 BAP formed 3.8±0.4 secondary buds which were repeatedly recultured to increase bud production. Altogether this method enabled an estimated harvest of 7848 buds from a single shoot tip in 28 months. Shoots (3–5 cm) developed from bud cultures were rooted in half-strength WPM medium with 0.5 mg.l–1 each of NAA and IBA, and 90–100% of the rooted plants were established in the field after hardening. Clonal propagation in vitro of raisin tree (Hovenia dulcis Thunb.) was achieved using axillary buds from mature trees and young plants. Explants cultured on Murashige-Skoog’s medium with 1/3 of the original salt concentration, supplemented with 0.5 mg l-1 BAP and 0.5 mg l-1 IAA, showed proliferation of new shoots in 4-5 weeks. Adventitious shoot proliferation was also stimulated in subsequent subcultures in the presence of BAP. The shoots rooted when transferred to 1/3 Murashlge and Skoog’s medium with 0.1 mg l-1 of IBA. Plantlets thus formed were successfully transplanted to the field after a short acclimatization period.  相似文献

The present work describes a procedure that allows for the easy and rapid induction of caulogenesis in four cultivars of Brassica napus L. from transversal Thin Cell Layers (tTCLs). In order to investigate the regeneration ability of this crop, the effects of genotype, explant source and culture medium were examined on shoot regeneration. The tTCL explants were excised from hypocotyl and petiole of 2-week-old seedlings and cultured on a solid basal MS medium supplemented with α-naphthaleneacetic acid (NAA: 0.1–0.4 mg l−1), 6-benzylamino-purine (BAP: 1–4 mg l−1) and sucrose (20–40 g l−1). A significant genotypic effect was observed between the four cvs; Jumbo and Drakkar displayed higher capacities to produce shoots than Pactol and Cossair. Regeneration commenced earlier and the percentage of shoot-producing explants as well as the number of shoots per regenerating explant was greater. The comparison between the regeneration ability of different explants showed that the hypocotyls exhibited a high rate of shoot organogenesis when they were cultured on MS medium supplemented with 3 mg l−1 BAP, 0.3 mg l−1 NAA and 30 g l−1 sucrose. Adventitious shoot buds developed from 46% of the tTCLs, with a mean of 7.5 buds. Furthermore, the method was fast with shoot formation occurring by 7 days culture. Plantlets regenerated from all shoots and developed normally. The regenerated plants were fertile and identical to source plants.  相似文献

Embryogenic cultures were induced from immature avocado zygotic embryos representing different botanical races and complex hybrids. The optimum induction medium consisted of B5 major salts, MS minor salts, 0.4 mg l−1 thiamine HCl, 100 mg l−1 myo-inositol, 30 g l−1 sucrose, 0.41 μM picloram and 8 g l−1 TC agar. Somatic embryogenesis occurred directly from the explants on induction medium, and secondary embryos and proembryonic masses proliferated in liquid and on semisolid maintenance medium. Embryogenic culture maintainance was optimized in liquid, filter-sterilized MS medium, supplemented with 30–50 mg l−1 sucrose, 4 mg l−1 thiamine HCl and 0.41 μM picloram. Two types of embryogenic cultures were recognized: –genotypes that proliferated as proembryonic masses in the presence of auxin (PEM-type) and; –genotypes in which the heart stage and later stages of somatic embryos developed in the presence of auxin(SE-type). Embryogenic suspension cultures became increasingly disorganized over time, and this was associated with progressive loss of embryogenic potential. Shoot tips obtained from in vitro Rosa plants (three cultivars) were successfully cryopreserved by a combined droplet vitrification method and subsequently shoots regenerated. The excised shoot tips (1–4 mm long) were incubated in a liquid MS medium supplemented with 2.5 mg l−1 thiamine, 0.2 mg l−1 biotin, 0.2 mg l−1 pyridoxine, 0.25 mg l−1 6-benzylaminopurine (BAP), 0.5 mg l−1 gibberellic acid (GA3) and 0.08 M sucrose, for 24 h. Following that incubation shoot tips were pre-cultured in this MS medium containing 0.1 till 1.0 M sucrose for 24 and 48 h, respectively. Pre-cultured shoot tips were dehydrated with concentrated PVS2 cryoprotective solution for 10–30 min at room temperature, prior to a direct plunge in liquid nitrogen. After rapid rewarming in the above mentioned liquid medium shoot tips were plated on a modified MS medium (5 g l−1 agar) supplemented with vitamins and plant growth regulators as mentioned above for regrowth. Cryopreserved shoot tips resumed growth within 10 days and regenerated shoots within 3 weeks. The highest numbers of regrowing shoot tips were 64.44% for cv. Kardinal, 67.73% for cv. Embryogenic callus was induced from the hypocotyl region of seedlings germinated from immature embryos of orange jessamine (Murraya paniculata (L.) Jack) on Murashige & Tucker (1969) medium containing 50 g l-1 sucrose, 5.0 mg l-1 benzyladenine, 2.5 mg l-1 2,4-dichlorophenoxyacetic acid and 600 mg l-1 malt extract. Isolated protoplasts divided to produce callus on Murashige & Tucker (1969) medium containing 50 g l-1 sucrose, 0.01 mg l-1 gibberellin A4+7 and 600 mg l-1 malt extract. Callus developed to plantlets via somatic embryogenesis on Murashige & Tucker (1969) medium with 50 g l-1 lactose but no plant growth regulators.
