Vascular elasticity is important in physiological and clinical problems. The mechanical properties of the great saphenous vein (GSV) deserve attention. This research aims to measure the radial elasticity of ex vivo GSV using the optical coherence elasticity (OCE). The finite element model of the phantom is established, the displacement field is calculated, the radial mechanical characteristics of the simulation body are obtained. Furthermore, we performed OCE on seven isolated GSVs. The strain field is obtained by combining the relationship between strain and displacement to obtain the radial elastic modulus of GSVs. In the phantom experiment, the strain of the experimental region of interest is mainly between 0.1 and 0.4, while the simulation result is between 0.06 and 0.40. The radial elastic modulus of GSVs ranged from 3.83 kPa to 7.74 kPa. This study verifies the feasibility of the OCE method for measuring the radial elastic modulus of blood vessels.  相似文献

旨在探讨如何高效获取优质的人大隐静脉(Great saphenous vein)原代干细胞。大隐静脉剪碎后分别采用组织块贴壁法和Ⅱ型胶原酶消化法获取血管壁细胞。倒置显微镜下观察不同时间段两组血管壁细胞形态学变化,台盼蓝染色测定血管壁细胞存活率,流式分选CD34和CD117双阳性干细胞,免疫荧光进一步证实。细胞培养至第三代(P3)时组织块贴壁法提取的细胞出现纤维化老化,而酶消化法提取的细胞仍有集落样生长,细胞存活率分别为(91.7±1.2)%和(97.2±0.7)%(P=0.005)。流式分选结果显示:组织块法和酶消化法获得CD34和CD117双阳性细胞所占比例分别为(0.16±0.05)%和(0.44±0.07)%,差异有统计学意义(P=0.005)。免疫荧光染色显示,分选出的干细胞培养1周后组织块法获得双阳性干细胞的阳性率(89.41±2.06)%,酶消化法为(94.03±1.83)%,P0.05。流式细胞仪检测分选出的干细胞中CD31、VEGF2和SMA阳性率分别为(0.12±0.01)%、(0.19±0.02)%和(0.45±0.01)%,与阴性对照组差异无统计学意义(P0.05),排除成熟内皮细胞和平滑肌细胞存在的可能性。分选出的干细胞进行管腔形成实验进一步证实其具有向内皮分化的潜能。上述结果显示,采用酶消化法可以获得形态更好、数量更多、存活率相对更高的干细胞,可广泛用于临床和基础研究。  相似文献

Summary Neurofilament immunoreactive nerve fibers were demonstrated in human skin using indirect immunohistochemical technique with antibodies to neurofilament polypeptides. Neurofilament-positive fibers were seen as free nerve endings in the epidermis and in dermal papilla, in Meissner's corpuscles and as fibers crousing in the dermis. Strongly fluorescent nerve fibers were also seen around hair follicles, sweat gland ducts and sometimes in relation to blood vessels. From the distribution pattern it was concluded that predominantly sensory nerve fibers were labelled and that this technique may be used to study reinnervation of cutaneous sensory nerved following tramatic injuries and surgical procedures.  相似文献

 The distribution of serotonin-immunoreactive (5HT-IR) nerve cells and fibers was thoroughly investigated immunohistochemically in the rat stomach, duodenum, jejunum, ileum, and colon. The immunoreactivity of the 5HT neurons was compared between non-treated controls and animals treated with colchicine, colchicine plus 5-hydroxytryptophan (5HTP), colchicine plus pargyline, and reserpine. The intensity of immunoreactivity in nerve fibers as well as nerve cell bodies was enhanced mostly in colchicine plus pargyline treated animals, therefore these animals were used for an observation of precise localization of 5HT in the rat gastrointestinal (GI) tract. Immunoreactivity in the nerve cell bodies and fibers was completely abolished in the GI tract of reserpine treated animals. The pattern of localization and projection of 5HT-IR neurons was similar in all segments of the rat GI tract. 5HT-IR nerve cell bodies were located in the myenteric plexus and showed the distinctive features of Dogiel type I neurons. Prominent bundles of varicose fibers traversed the myenteric ganglia and some of them surrounded the cell bodies of immunopositive and immunonegative neurons. 5HT-IR nerve fibers were located in the submucous plexus, densely entwined about the submucosal blood vessels. Most characteristically, 5HT-IR nerve fibers invaded the lamina propria of mucosa where they underlay the crypt epithelium. In conclusion, the present study showed that 5HT-IR neurons located in the myenteric plexus projected fibers widely in the rat GI tract. The localization of fibers in the lamina propria of mucosa implies that this neuron may exert an important role in the epithelial function of the GI tract. Accepted: 8 October 1996  相似文献

The distribution of myelinated and nonmyelinated nerve fibers of the saphenous nerve of cats in the ventral and dorsal roots of the spinal cord was investigated by methods improving the signal—noise ratio in records of evoked responses from the nerve. The fibers of this nerve enter the spinal cord through roots of segments L4–6. Nerve fibers with conduction velocities of between 80 and 0.38 m/sec were distributed in the dorsal roots of these segments. Four groups of nerve fibers with conduction velocities of 80–60, 40–30, 12.0–3.0, and 1.1–0.51 m/sec, possibly afferent in nature, were found in the ventral roots. The conditions of origin and detection of low-amplitude potentials in the roots of the spinal cord and the probable functional role of the nerve fibers in the ventral roots are discussed.Research Institute of Applied Mathematics and Cybernetics, N. Endovenous laser treatment (ELT) is a minimally invasive technique used to treat varicose veins. In this technique, an optical fiber is inserted into the vein to be treated and the laser lights while the optical is slowly withdrawn. The average linear endovenous energy density (LEED) delivered during the procedure is determinant in the success of this therapy. Recanalization and side effects are energy-dependent. Thus, the success of the therapy is mainly related to the pullback speed of the fiber. Ideally this speed has to be 1 mm/s. The purpose of this investigation was to evaluate in real time the fiber pullback speed. In order to measure in real time the motion of the fiber inside the saphenous vein, magnetic tracking was used. The system was evaluated during the treatment procedures for 18 patients. The mean pullback speed was 1.02 ± 0.15 mm/s similar to the speed the physician wanted to obtain. Based on quality control, safety standards, and regulations official documents for medical lasers, where a 20% accuracy is admitted; only one patient was treated with an average pullback speed inferior to 0.8 mm/s (0.77 cm/s) and two patients were treated with an average pullback speed superior to 1.2 mm/s (1.26 mm/s and 1.32 mm/s). Consequently, even for a well-trained physician, variations can be observed. These variations could easily explain why complications (too low speed and consequently too high LEED) or recurrences (too fast speed and consequently too low LEED) are sometimes reported in clinical studies. A standardized or an automatic withdrawal of the fiber could possibly improve this technique but remains to be carefully evaluated.  相似文献

Summary The distribution and density of the noradrenergic and acetylcholinesterase-positive nerve fibres were histochemically studied in different uterine regions of prepubertal and cycling rats in dioestrus and oestrus. Besides the rich and double innervation of blood vessels, both types of nerve fibre were found in the myometrium and cervical musculature. The non-vascular noradrenergic network looked denser at the tubal end of the horns and at the cervix, whereas the acetylcholinesterase-positive innervation was poor at the tubal end, increasing toward the cervix. Contrasting with the middle third of the uterine horn, at the tubal end, the myometrial longitudinal layer was much more innervated than the circular one, especially by the noradrenergic nerve fibres. The prepubertal rats presented an adult pattern of uterine autonomic innervation. In the cycling animals, this innervation was nearly the same during oestrus and dioestrus regarding both the density of nerve fibres and intensity of the histochemical reactions.  相似文献

Because it is likely that antigen sensitization is not restricted to airway smooth muscle but probably involves all tissues in the animal, we decided to test the hypothesis that saphenous vein from pollen extract-sensitized dogs is sensitized and is, in addition, mechanically altered. To this end, we studied responses to specific antigen challenge and length-tension and force-velocity relationships in sensitized (SSV) and control saphenous veins (CSV). The antigen challenge revealed that the venous smooth muscle was strongly sensitized and developed a Schultz-Dale response, the two main mediators of which were histamine and norepinephrine. Length-tension relationship studies showed that whereas there is no difference in maximum isometric tension development between SSV and CSV [93.95 +/- 7.34 and 87.86 +/- 4.00 (SE) mN/mm2, respectively], SSV exhibited a significantly greater maximum isotonic shortening capacity of 0.613 +/- 0.009 optional length (lo) vs. 0.578 +/- 0.012 lo for CSV. Unloaded shortening velocity (Vo), which reflects the cross-bridge cycling rate, was determined at different times after the onset of electrical stimulation. Maximum Vo was attained early (5 s) in the contraction; a 15% decline in Vo was observed at the plateau of the contraction (15 s). At 5 s, Vo of SSV (0.316 +/- 0.019 lo/s) was significantly higher than that of CSV (0.269 +/- 0.018 lo/s), although Vos were same at 15 (0.249 +/- 0.021 lo/s for SSV and 0.237 +/- 0.019 lo/s for CSV). Immunoreactivity of galanin (GAL) was detected in the nerve fibers distributed within the intervascular stroma of the bullfrog carotid labyrinth. GAL-immunoreactive fibers are numerous, and some are close to the sinusoidal plexus. Most GAL fibers appear as thin processes with some varicosities. A combination of indirect double immunofluorescence labelling and image processing clearly demonstrated that the distribution patern of GAL fibers is different from that of SP fibers. This indicates that GAL and SP do not coexist in the same nerve fibers. The role of GAL fibers may be different from that of previously reported neuropeptides (substance P, calcitonin gene-related peptide, vasoactive intestinal polypeptide, neuropeptide Y, and others) as a neuromodulator in controlling vascular tone of the labyrinth.  相似文献

Summary The distribution of nerve fibers in molars, periodontal ligament and gingiva of the rat shows a complex pattern. Decalcified material including the alveolar bone was sectioned in three different planes and stained by means of immunohistochemistry for detection of the neurofilament protein (NFP); the immunoreactive neural elements were clearly visualized in three-dimensional analyses. NFP-positive nerve fibers formed a subodontoblastic plexus in the roof area of the dental pulp; some of them entered the predentin and dentin directly through the dentinal tubules. This penetration was found mainly in the pulp horn, and was limited to a distance of about 100 m from the pulpo-dentinal junction. In the periodontal ligament, NFP-positive nerve fibers were found densely distributed in the lower half of the alveolar socket. Two types of nerve terminals were recognized in the periodontal ligament: free nerve endings with tree-like ramifications, and expanded nerve terminals showing button- or glove-like shapes. The former tapered among the periodontal fibers, some even reaching the cementoblastic layer. The latter were located, frequently in groups, within the ligament restricted to the lower third of the alveolar socket. A well-developed plexus of NFP-positive nerves was revealed in the lamina propria of the free gingiva, the innervation being denser toward the epithelium of the gingival crevice. The characteristic distribution of NFP-immunoreactive nerve fibers revealed in this study is discussed in relation to region-specific sensations in the teeth and surrounding tissues.  相似文献

ABSTRACT: BACKGROUND: As a main cause of mortality in developed countries, Coronary Artery Disease (CAD) is known as silent killer with a considerable cost to be dedicated for its treatment. Coronary Artery Bypass Graft (CABG) is a common remedy for CAD for which different blood vessels are used as a detour. There is a lack of knowledge about mechanical properties of human blood vessels used for CABG, and while these properties have a great impact on long-term patency of a CABG. Thus, studying these properties, especially those of human umbilical veins which have not been considered yet, looks utterly necessary. METHODS: Umbilical vein, as well as human Saphenous vein, are respectively obtained after cesarean and CABG. First, histological tests were performed to investigate different fiber contents of the samples. Having prepared samples carefully, force-displacement results of samples were rendered to real stress--strain measurements and then a fourth-order polynomial was used to prove the non-linear behavior of these two vessels. RESULTS: Results were analyzed in two directions, i.e. circumferentially and longitudinally, which then were compared with each other. The comparison between stiffness and elasticity of these veins showed that Saphenous vein's stiffness is much higher than that of umbilical vein and also, it is less stretchable. Furthermore, for both vessels, longitudinal stiffness was higher than that of circumferential and in stark contrast, stretch ratio in circumferential direction came much higher than longitudinal orientation. CONCLUSION: Blood pressure is very high in the region of aorta, so there should be a stiff blood vessel in this area and previous investigations showed that stiffer vessels would have a better influence on the flow of bypass. To this end, the current study has made an attempt to compare these two blood vessels' stiffness, finding that Saphenous vein is stiffer than umbilical vein which is somehow as stiff as rat aortic vessels. As blood vessel's stiffness is directly related to elastin and mainly collagen content, results showed the lower amount of these two contents in umbilical vein regarding Saphenous vein.  相似文献
