For improvement of mass production of the rhabditid biocontrol nematodes Steinernema carpocapsae and Steinernema feltiae in monoxenic liquid culture with their bacterial symbionts Xenorhabdus nematophila and Xenorhabdus bovienii, respectively, the effect of the initial nematode inoculum density on population development and final concentration of dauer juveniles (DJs) was investigated. Symbiotic bacterial cultures are pre-incubated for 1 day prior to inoculation of DJs. DJs are developmentally arrested and recover development as a reaction to food signals provided by their symbionts. After development to adults, the nematodes produce DJ offspring. Inoculum density ranged from 1 to 10 × 103 DJ per milliliter for S. carpocapsae and 1 to 8 × 103 DJs per milliliter for S. feltiae. No significant influence of the inoculum density on the final DJ yields in both nematode species was recorded, except for S. carpocapsae cultures with a parental female density <2 × 103 DJs per milliliter, in which the yields increased with increasing inoculation density. A strong negative response of the parental female fecundity to increasing DJ inoculum densities was recorded for both species with a maximum offspring number per female of >300 for S. carpocapsae and almost 200 for S. feltiae. The compensative adaptation of fecundity to nematode population density is responsible for the lack of an inoculum (or parental female) density effect on DJ yields. At optimal inoculation density of S. carpocapsae, offspring were produced by the parental female population, whereas S. feltiae always developed a F1 female population, which contributed to the DJ yields and was the reason for a more scattered distribution of the yields. The F1 female generation was accompanied by a second peak in X. bovienii density. The optimal DJ inoculum density for S. carpocapsae is 3–6 × 103 DJs per milliliter in order to obtain >103 parental females per milliliter. Density-dependent effects were neither observed on the DJ recovery nor on the sex ratio in the parental adult generation. As recovery varied between different batches, assessment of the recovery of inoculum DJ batches is recommended. S. feltiae was less variable in DJ recovery usually reaching >90%. The recommended DJ inoculum density is >5 × 103 DJs per milliliter to reach >2 × 103 parental females per milliliter. The mean yield recorded for S. carpocapsae was 135 × 103 and 105 × 103 per mililiter for S. feltiae.  相似文献

The rhabditid nematodes Steinernema carpocapsae and Steinernema feltiae are used in biological control of insect pests. Mass production is done in liquid culture media pre-incubated with their bacterial symbionts Xenorhabdus nematophila and Xenorhabdus bovienii, respectively, before nematode dauer juveniles (DJs) are inoculated. As a response to food signals produced by the bacterial symbionts, the DJs exit from the developmentally arrested dauer stage (they recover development) and grow to adults, which produce DJ offspring. Variable DJ recovery after inoculation often causes process failure due to non-synchronous population development and low numbers of adult nematodes. This contribution investigated the influence of the bacterial cell density on DJ recovery and development to adults. At higher density of 1010 bacterial cells ml−1, a higher percentage of DJ recovery was induced, and adults occurred earlier in both Steinernema spp. than at lower density of 109 and 108 cells ml−1. Xenorhabdus symbionts produce phase variants. Recovery in bacteria-free supernatants was lower than in supernatants containing bacterial cells for both primary and secondary phase Xenorhabdus spp. and lower in secondary than in primary phase supernatants or cell suspensions. In general, recovery was lower for Steinernema feltiae and the time at which 50% of the population had recovered after exposure to the food signal was longer (RT50 = 17.1 h) than for Steinernema carpocapsae (RT50 = 6.6 h). Whereas >90% S. carpocapsae DJs recovered in hemolymph serum of the lepidopteran insect Galleria mellonella, recovery of S. feltiae only reached 31%. Penetration into a host insect prior to exposure to the insect’s food signal did not enhance DJ recovery. Consequences for liquid culture mass production of the nematodes and differences between species of the genera Steinernema and Heterorhabditis are discussed.  相似文献

Heterorhabditis bacteriophora is used in biological control of soil-borne insect pests in horticulture and turf. Mass production is carried out in monoxenic liquid cultures pre-incubated with the symbiont of the nematodes, the bacterium Photorhabdus luminescens, before nematode dauer juveniles (DJ) are inoculated. As a response to bacterial food signals, the DJ recover from the developmentally arrested dauer stage, grow to adults and produce DJ offspring. Variable DJ recovery after inoculation into cultures of P. luminescens often causes process failure due to low numbers of adult nematodes in the medium. In order to enhance DJ recovery, improve nematode population management and increase yields, the optimal timing for DJ inoculation was sought. The process parameter pH and respiration quotient (RQ) were recorded in order to test whether changes can be used to identify the best moment for DJ inoculation. When DJ were inoculated during the lag and early logarithmic growth phases of P. luminescens cultures, DJ recovery was low and almost no nematode reproduction was obtained. High populations of P. luminescens phase variants were recorded. Recovery and yields increased when DJ were inoculated during the latter log phase during which the RQ dropped to values <0.8 and the pH reached a maximum. The highest DJ recovery and yields were observed in cultures that were inoculated during the late stationary growth phase. This period started with the increase of the pH after its distinct minimum at pH <8.0. Thus optimal timing for DJ inoculation can be defined through monitoring of the pH in the P. luminescens culture.  相似文献

For commercial use of the entomopathogenic nematodes Steinernema carpocapsae and Steinernema feltiae in biological control of insect pests, they are produced in liquid culture on artificial media pre-incubated with their symbiotic bacteria Xenorhabdus nematophila and Xenorhabdus bovienii, respectively. After 1 day of the bacterial culture, nematode dauer juveniles (DJs) are inoculated, which recover development. The adult nematodes produce DJ offspring, which are harvested and can be sprayed. This study determined optimal temperatures to obtain high DJ progeny within a short process time. Temperatures assessed were 23°C, 25°C, 27°C, and 29°C for S. carpocapsae and 20°C, 23°C, 25°C, and 27°C for S. feltiae. The recovery of inoculated DJs was hardly affected and was reduced only in S. carpocapsae at 29°C. The fecundity (eggs in uterus) in S. carpocapsae reached a maximum at 27°C; whereas, maximum yields were recorded at 25°C. For both Steinernema spp., highest DJ densities were obtained after 15 days incubation at 25°C. Optimal culture temperature for both nematode species is 25°C. S. carpocapsae was more sensible to suboptimal temperature than S. feltiae. Results on total DJ density and DJ proportion of the total nematode population were more variable at non-optimal temperature condition for S. carpocapsae than for S. feltiae. Suboptimal culture temperature also reduced DJ infectivity.  相似文献

Phase I cells of Photorhabdus luminescens produce two types of intracellular crystalline inclusion proteins designated CipA and CipB. The genes encoding CipA and CipB proteins from P. luminescens H06 were expressed respectively in Escherichia coli and these cells were used to feed the axenic first juveniles (J1) of three Steinernema nematode isolates in liquid cultures and on agar plates. In liquid cultures, the axenic J1 juveniles of all three test Steinernema nematode isolates were able to produce next dauer juveniles (DJs) in the E. coli cultures with at least one of the expressed Cip proteins, but unable to develop beyond the next J1 stage without expressed Cip proteins. For each target nematode isolate, addition of the supernatant of the bacterial culture of its Xenorhabdus symbiont to the tested liquid cultures did not induce the formation of DJs. However, on LB agar plates with different test E. coli cultures, all J1 juveniles of the three Steinernema strains finally developed into next DJs. It seemed that the metabolite pathway of the test bacteria in both culture systems was different. The presence of the Cip proteins has a significant influence on the DJ formation of the Steinernema nematodes in liquid culture system.  相似文献

Heterorhabditis indica is a potential agent for the biological control of grubs in sugarcane fields in India. The type strain LN 2 was transferred to monoxenic cultures on its symbiont Photorhabdus luminescens and successfully produced on solid media. In liquid cultures, a mean dauer juvenile yield of 457 000 was obtained with a maximum of 648 000 per ml. Comparatively high yields have not been reported before. Therefore, costs related to the liquid culture production of H. indica will be lower than for other entomopathogenic nematodes currently used in biocontrol. Different bacterial clones had no significant influence on the dauer juvenile yields in liquid media. The exit from the dauer juvenile stage (recovery) after inoculation and the number of hermaphrodites significantly decreased when culture temperature was increased from 25-30 ° C; the dauer juvenile yields were not affected. The cell density of P. luminescens in batch cultures was higher at 25 and 30 ° C than at growth temperatures of 35 and 37 ° C. In continuous culture, the bacterial growth was inhibited when the growth temperature reached 38 ° C. After approximately 60 h, the bacteria adapted to higher temperature and the growth rate increased again. When the temperature was further increased to 40 ° C, the bacterial growth was inhibited.  相似文献

Photorhabdus luminescens are bacterial symbionts of entomopathogenic nematodes of the genus Heterorhabditis. The bacto-helminthic complexes are used in biocontrol of insect pests in cryptic environments. For in vitro production, liquid media are incubated with P. luminescens for 24 h prior to the inoculation of nematode dauer juveniles. The nematodes develop to self-fertilizing hermaphrodites and produce offspring. The exit from the developmentally arrested dauer stage (recovery) is a response to a yet undescribed food signal. Major process instability is caused by low and unsynchronized recovery of the dauers. In living insects, dauer recovery is approximately 95% within 1 day. In liquid cultures of P. luminescens the recovery is spread over several days and varies between 0 and 81%. In complex culture media no food signal was detected. A food signal is produced by P. luminescens and excreted into the culture medium. The maximum food signal production was recorded during the late exponential growth phase. Compared to the food signal found in insects, the efficacy of the bacterial signal is much lower. The reasons for the variable activity of the bacterial food signal and its function during the nematode life cycle are discussed. The effect of intraspecific competition on the development and reproduction of the slug parasitic nematode Phasmarhabditis hermaphrodita was studied in a series of laboratory experiments. Different inocula concentrations (1, 10, 25, 50, 100, 250, 500 and 1000) of P. hermaphrodita dauer juveniles (DJ) were applied to 0.02 g of homogenised slug Deroceras reticulatum on agar in multi-well plates and the development time, yield, lipid content, and body length of females and DJs were recorded. There was an inverse relationship between dose and recorded female length or lipid content. In newly emerged DJs the body length and lipid content were relatively invariable up to a dose of 250 DJs (ranging between 850 and 925 µm, 6.8 and 7.4, respectively), and decreased at higher doses (500 and 1000 DJs). The yield increased with increasing dosage to its optimum at a dose of 100 (200,000 DJs per 1 g of substrate) DJs and thereafter decreased. In the treatment with the highest dose (1000 DJs), only a minor portion of the inoculum developed to adult stage and completed their reproduction cycle while the rest remained at the larval stage. It might, therefore, be concluded that P. hermaphrodita tolerates intraspecific competition to a certain level at which crowding becomes too severe and a part of the inoculum do not develop and probably leave the site to avoid the fatal overcrowding. In conclusion, we show that intraspecific competition negatively affects the yield and quality of DJs of P. hermaphrodita, but it seems that this nematode can partly prevent overcrowding by avoiding occupied sites.  相似文献

The aim of this study was to investigate the relationship between reactive strength in a vertical and a horizontal drop jump (DJ). Subjects (n = 28) with previous jump training experience, performed 6 vertical DJs and 6 horizontal DJs from a 0.4-m box. Contact time, height jumped, distance jumped, and reactive strength index (RSI) were calculated and analyzed. Typical error measurements (TEMCV%) and intraclass correlations (ICCs) were used to assess the intrasubject reliability. Relationships between jumps and within jumps of the aforementioned variables were assessed using ICCs. The ICC (r > 0.789) and the TEMCV% (<10%) indicated good reliability for both vertical and horizontal DJs across each variable. Contact time showed no relationship between jumps (r = 0.222) and had no effect on the vertical DJ height (r = 0.152) or horizontal DJ distance (r = 0.261). The RSI correlation (r = 0.533) indicated a large relationship between reactive ability in the horizontal DJ and the vertical DJ. Contact times were significantly lower in vertical DJs compared with horizontal DJs (p < 0.0001). This study indicated that horizontal DJs are reliable and may be better used to train reactive movements that do not require brief contact times.  相似文献

Entomopathogenic nematodes (EPNs) in the families Heterorhabditidae and Steinernematidae have considerable potential as biological control agents of soil-inhabiting insect pests. In the present study, the control potential of the EPNs Steinernema carpocapsae (TUR-S4), S. feltiae (Nemaplus), S. carpocapsae (Nemastar), S. feltiae (TUR-S3) and Heterorhabditis bacteriophora (Nematop) against a new longicorn pest, Dorcadion pseudopreissi Breuning, 1962 (Coleoptera: Cerambycidae), on turf was examined in laboratory studies. Pathogenicity tests were performed at the following doses: 50, 100 and 150 Dauer Juveniles (DJs)/larva at 25°C. Highest mortalities (75–92%) of the larvae were detected at the dose of 150 DJs/larva for all nematodes used. Reproduction capabilities of the used EPNs were examined at doses of 50, 75, 100 and 150 DJs/larva at 25°C. S. carpocapsae (TUR-S4) had the most invasions (32 DJs/larva) and reproduction (28042 DJs/larva) at the dose of 100 DJs, and the highest reproduction (per invaded DJ into a larva) was observed in H. bacteriophora (Nematop) (2402.85 DJs) at a dose of 50 DJs. The foraging behaviour of the nematodes in the presence of D. pseudopreissi and Galleria mellonella L. (Lepidoptera: Galleriidae) larvae was studied using a Petri dish filled with sand at 20°C. All of the used nematodes accumulated near the larvae section of both insect species (32–53% of recovered DJs) with a higher percentage of S. carpocapsae (TUR-S4) (53%) and H. bacteriophora (48%) (Nematop) moving towards larvae of D. pseudopreissi, than the S. feltiae strains.  相似文献

昆虫病原线虫的感染期幼虫(infective juvenile,IJ)是其一生中唯一具有侵染能力和可自由生活于寄主体外的虫态,一般滞育不取食,体外包裹着已经蜕去的第2龄幼虫的表皮,对外界不良环境的耐受能力强,又称为耐受态幼虫(dauer juvenile,DJ),类似于秀丽隐杆线虫Caenorhabditis elegans的耐受态幼虫。在食物信息的诱导下,感染期幼虫脱鞘,释放出共生细菌,恢复取食并继续发育,这个过程称为感染期幼虫的恢复(IJ recovery)。这个过程是发生在寄生性线虫入侵寄主时的发育过程,对于成功寄生是必要的,在线虫的产业化培养中发挥着重要作用,感染期线虫的恢复率及其发育的同步性直接影响了线虫的产量。本文概述了感染期线虫的恢复发育过程,并对诱导感染期线虫恢复发育的食物信号(food signals)、恢复的影响因素及其检测手段进行了综述,同时讨论了未来的研究方向。  相似文献

Agro - industrial residues and by - products available in southeastern Brazil were used as ingredients for low - cost culture media for liquid fermentation of Bacillus thuringiensis var. kurstaki. Highest spore yield was obtained with a medium containing cheese whey , soya bean milk and molasses (WSM) . Crystals and spores were produced in all media and potency of the final product was highest for nutrient broth + yeast extract medium (NBY) . There was no correlation between the number of spores in the fermented media and the potency of the preparations . Considering all three factors , the potencies , costs and yields of the final products , lowest relative cost was obtained with BMM medium ( Bombyx mori pupae + molasses) . NBY and WSM had intermediate relative cost approximately nine times higher than BMM . The cost analysis suggests that BMM medium should be preferred for local production of B. thuringiensis var . kurstaki in comparison to other media tested . The results also demonstrate the importance of considering yields , cost and potency of the B. thuringiensis preparations in selecting the production medium .  相似文献

The entomopathogenic nematode Heterorhabditis bacteriophora is commercially used in biological control of soil dwelling insect pests. It reproduces by autogamy (hermaphrodites) enabling the production of inbred lines, but also by amphimixis (through mating of male and female) which allows cross-breeding. When H. bacteriophora is produced in liquid culture, copulation of male and female is prevented and reproduction is solely by self-fertilisation of hermaphrodites. When reared in insects, crosses are possible resulting in heterozygous offspring. Heat and desiccation tolerance of these nematodes have been successfully improved by selective breeding. Trait deterioration was prevented by producing homozygous inbreds through consecutive reproduction in liquid culture, the method also used for commercial mass production. In this study, we investigated possible heterosis effects in desiccation and heat tolerance after cross-breeding of homozygous inbred lines of H. bacteriophora. Increased desiccation tolerance of the heterozygous progeny in comparison to homozygous inbred lines was recorded indicating that heterosis is a possible means for further improvement of this trait. In contrast, the heat tolerance of the heterozygous offspring was lower than that of the homozygous population. The results provide evidence for the tremendous potential of classical genetics to improve beneficial traits of a biological control agent and carry domestication of H. bacteriophora a significant step forward.  相似文献

Several methods are available for estimating heritability in disomic species, including parent-offspring regression, realized heritability, intraclass correlations of recombinant inbred lines, and diallel-cross analysis. Estimates were obtained by these various methods for a set of eight bread wheat (Triticum aestivum) lines adapted to the East African highlands, which had been intercrossed and selfed in a half-diallel arrangement to give F1, F2 and F3 generations, and F6 recombinant inbred lines. Significant genetic variation existed among parents and crosses for both grain yield and yellow rust resistance in all generations. Based on the heritability calculated from the analysis of F6 recombinant inbred lines, analysis of the F2 diallel crosses was recommended for determining the heritability of both characters in early segregating generations. The results also suggest that a form of tandem selection may be effective in developing locally adapted germplasm which combines high grain yield with yellow rust resistance. Entomopathogenic nematode production in liquid fermentation still requires improvements to maximize efficiency, yield, and nematode quality. Therefore, this study was aimed at developing a more suitable liquid medium for mass production of Steinernema feltiae, by assessing the effects of nutrient concentration, thickeners (primarily agar), and agitation speed on infective juvenile (IJ) yield. Base medium (BM) contained yeast extract (2.3%), egg yolk (1.25%), NaCl (0.5%), and corn oil (4%). All media were inoculated with Xenorhabdus bovienii, and 2 d later, with 2-d-old S. feltiae juveniles. For the nutrient concentration experiment, we evaluated the base medium versus a modified base medium containing all the components, but with 3× concentrations of yeast extract (6.9%), egg yolk (3.75%), and corn oil (12%). The nematodes and bacteria were cultured in 150-ml Erlenmeyer flasks containing 50 ml of liquid medium at (25°C) and 180 rpm on a rotary shaker incubator. To assess the effect of thickeners, IJs were inoculated in BM with agar (0.2%), carrageen (0.2%), and carboxymethyl cellulose (0.2% and 0.5%). The addition of 3× more nutrients relative to the BM resulted in a significantly lower yield of nematodes. For agar and agitation speed experiments, five levels of agar in the BM (0%, 0.2%, 0.4%, 0.6%, and 0.8% agar) and two agitation speeds (180 and 280 rpm) were evaluated for production. Increasing agitation speed from 180 to 280 rpm and higher levels of agar in the medium (> 0.2%) significantly increased the yield of bacteria. At the lower agitation speed, media amended with 0.4% and 0.6% agar produced higher nematode yields compared to media without agar. Media with 0.2% and 0.8% agar resulted in intermediate levels of nematode production. At the higher agitation speed, media supplemented with 0.8% agar resulted in the lowest yield of nematodes when compared to the other media tested. Results indicated that increasing nutrient concentration levels was detrimental to nematode production. Also, media containing agar (0.4% and 0.6%) increased nematode yields when cultures were grown at low agitation speed. When IJs were used as the inoculum, 0.2% agar also enhanced recovery and nematode yield at the higher agitation speed.  相似文献

This study compared the magnitude of muscle damage induced when consecutive drop jumps (DJs) were performed on sand vs. firm (wood) surfaces from a height of 0.6 m. Eight subjects performed DJs on a sand surface at a depth of 0.2 m (S condition), and 8 other subjects performed DJs on a wood surface (F condition). Each set consisted of 20 DJs with an interval of 10 seconds between jumps. Subjects performed 5 sets of DJs with 2 minutes between sets. Maximal isometric force, muscle soreness, and plasma creatine kinase (CK) activity were measured immediately before and immediately after the DJ exercise as well as 1, 24, 48, 72, and 96 hours after the DJ exercise. All measures changed significantly (p < 0.05) after exercise for both conditions; however, significantly (p < 0.05) smaller changes in these measures were evident for the S condition than for the F condition. These results show that DJs on a sand surface induce less muscle damage than on a firm surface. Training on sand may improve aerobic capacity or strength with a low risk of muscle damage.  相似文献

The purpose of this study was to examine the effect of 3 different plyometric training frequencies (e.g., 1 day per week, 2 days per week, 4 days per week) associated with 3 different plyometric training volumes on maximal strength, vertical jump performance, and sprinting ability. Forty-two students were randomly assigned to 1 of 4 groups: control (n = 10, 7 sessions of drop jump (DJ) training, 1 day per week, 420 DJs), 14 sessions of DJ training (n = 12, 2 days per week, 840 DJs), and 28 sessions of DJ training (n = 9, 4 days per week, 1680 DJs). The training protocols included DJ from 3 different heights 20, 40, and 60 cm. Maximal strength (1 repetition maximum [1RM] and maximal isometric strength), vertical height in countermovement jumps and DJs, and 20-m sprint time tests were carried out before and after 7 weeks of plyometric training. No significant differences were observed among the groups in pre-training in any of the variables tested. No significant changes were observed in the control group in any of the variables tested at any point. Short-term plyometric training using moderate training frequency and volume of jumps (2 days per week, 840 jumps) produces similar enhancements in jumping performance, but greater training efficiency (approximately 12% and 0.014% per jump) compared with high jumping (4 days per week, 1680 jumps) training frequency (approximately 18% and 0.011% per jump). In addition, similar enhancements in 20-m-sprint time, jumping contact times and maximal strength were observed in both a moderate and low number of training sessions per week compared with high training frequencies, despite the fact that the average number of jumps accomplished in 7S (420 jumps) and 14S (840 jumps) was 25 and 50% of that performed in 28S (1680 jumps). These observations may have considerable practical relevance for the optimal design of plyometric training programs for athletes, given that a moderate volume is more efficient than a higher plyometric training volume.  相似文献

Low heritability estimates in marginal or stress environments have often been used as one of the main justification for conducting selection work in environments with optimum or near‐optimum conditions for plant growth and grain yield. In this study, we have examined the relationships between grain yield and broad‐sense heritability in four groups of recombinant inbred lines (RILs) obtained from four barley crosses derived from parents differing in adaptation to stress. The RILs and the parents were grown in 13 combinations of years and locations (environments) in Syria and Jordan. Grain yields ranged from about 30 kg ha?1 to nearly 4000 kg ha?1 and genotype × environment interactions explained about half of the total variance for environmentally standardised data. Broad‐sense heritability in the individual year–location combinations varied from 0 to 0.68 and both the simple correlation and the rank correlation coefficients between grain yield and heritability were not significant. Genotype × years within individual locations, which measures the repeatability of a location in discriminating between genotypes, was also independent from the yield level, confirming that low‐yielding locations can be reliable selection environments. Also, there was no relationship between the type of cross and the magnitude of heritability in the various environments, but, as expected, the magnitude of heritability was significantly associated with the genetic distance between the parents. It is concluded that, holding all other factors affecting response to selection constant, concerns about the magnitude of heritability at low‐yielding locations are not justified and should not prevent them from being used as selection sites.  相似文献

Predicting the performance of recombinant inbred lines derived by single seed descent (SSD) following crosses between inbred lines is most reliable for traits in which epistasis and genotype environment interaction are absent. The analysis of two field trials of Brussels sprouts indicated the presence of such effects in seven quantitative characters. These effects could be minimised by transforming the scale on which each character was measured. Predictions were made of the likely performance of the recombinant inbred lines currently being produced by SSD at the Institute of Horticultural Research, Wellesbourne, compared with that of current F1 hybrid varieties. The predictions from the first trial of F2 material were encouraging for harvest date, stem length and waste weight, but low for sprout number, quality, marketable yield and total yield. Predictions from F3 material in the second trial were good for sprout number and total yield (in addition to harvest date, stem length and waste weight as before), but still low for marketable yield and quality, with less than 0 01% of lines expected to perform better than the F1. Model fitting to variances suggested the presence of linkage disequilibrium in dispersion for waste weight, but no information could be obtained about linkage disequilibrium for the two most important characters, marketable yield and quality. The broad heritability of each character except stem length was lower than 0 5 (0 3 or lower for total yield, waste weight and quality), suggesting that advanced generation selection, as used in SSD, might be more successful than selection in the early generations.  相似文献
