Summary This paper provides the results of studies which characterized conductive36Cl flux in basolaterally enriched membrane vesicles prepared from rabbit renal outer medulla. Conductive36Cl uptake was studied under two different experimental conditions. In the first,36Cl flux was driven by an inside positive voltage created with oppositely directed Cl and gluconate gradients. In the second, an inwardly direct K+ gradient was used to drive36Cl uptake. By these two methods, voltage-sensitive36Cl uptake was shown to comprise about 45 and 65%, respectively, of the initial rates of total36Cl flux. Separate paired studies demonstrated that the conductive36Cl uptake was inhibited by the Cl channel blocker diphenylamine-2-carboxylate (DPC) with an IC50 for DPC of 154 m. The voltagedependent36Cl uptake had an activation energy of 6.4 kcal/mole. This36Cl conductance had an anion selectivity sequence of I>ClNO 3 gluconate.  相似文献

Summary Active HCO 3 t- secretion in the anterior rectal salt gland of the mosquito larva,Aedes dorsalis, is mediated by a 11 Cl/HCO 3 exchanger. The cellular mechanisms of HCO 3 and Cl transport are examined using ion- and voltage-sensitive microelectrodes in conjunction with a microperfused preparation which allowed rapid saline changes. Addition of DIDS or acetazolamide to, or removal of CO2 and HCO 3 from, the serosal bath caused large (20 to 50 mV) hyperpolarizations of apical membrane potential (V a) and had little effect on basolateral potential (V bl). Changes in luminal Cl concentration alteredV a in a repid, linear manner with a slope of 42.2 mV/decaloga Cl l –. Intracellular Cl activity was 23.5mm and was approximately 10mm lower than that predicted for a passive distribution across the apical membrane. Changes in serosal Cl concentration had no effect onV bl, indicating an electrically silent basolateral Cl exit step. Intracellular pH in anterior rectal cells was 7.67 and the calculated was 14.4mm. These results show that under control conditions HCO3 enters the anterior rectal cell by an active mechanism against an electrochemical gradient of 77.1 mV and exits the cell at the apical membrane down a favorable electrochemical gradient of 27.6 mV. A tentative cellular model is proposed in which Cl enters the apical membrane of the anterior rectal cells by passive, electrodiffusive movement through a Cl-selective channel, and HCO 3 exits the cell by an active or passive electrogenic transport mechanism. The electrically silent nature of basolateral Cl exit and HCO3 entry, and the effects of serosal addition of the Cl/HCO3 exchange inhibitor, DIDS, on and transepithelial potential (V ic) suggest strongly that the basolateral membrane is the site of a direct coupling between Cl and HCO 3 movements.  相似文献

Summary The present studies examined some of the properties of Cl channels in renal outer medullary membrane vesicles incorporated into planar lipid bilayers. The predominant channel was anion selective having aP Cl/P K ratio of 10 and a unit conductance of 93 pS in symmetric 320mm KCl. In asymmetric KCl solutions, theI-V relations conformed to the Goldman-Hodgkin-Katz equation. Channel activity was voltage-dependent with a gating charge of unity. This voltage dependence of channel activity may account, at least in part, for the striking voltage dependence of the basolateral membrane Cl conductance of isolated medullary thick ascending limb segments. The Cl channels incorporated into the planar bilayers were asymmetrical: thetrans surface was sensitive to changes in ionized Ca2+ concentrations and insensitive to reducing KCl concentrations to 10mm, while thecis side was insensitive to changes in ionized Ca2+ concentrations, but was inactivated by reducing KCl concentrations to 50mm.  相似文献

Summary The rate of Cl influx in intactChara was inhibited whenever the ATP concentration was reduced by application of metabolic inhibitors. In perfused cells, however, a net influx of Cl against its electrochemical gradient could be observed in the absence of ATP. Addition of ATP to the perfusion medium slightly stimulated Cl influx in one experiment but had no effect in another. Addition of ADP, NADH or metabolic inhibitors did not alter the influx rate. Consideration of the potential energy gradients across theChara plasmalemma in the perfused state leads to the conclusion that Cl influx occurs by cotransport with H+ or OH.  相似文献

Thylakoids were isolated from the leaves of three different plants (Pisum sativium L., Lactuca sativa L., and Raphanus sativus L.). The addition of HCO 3 ? to a suspension of salt-and HCO 3 ? -epleted thylakoids (suspended in salt-free medium) raised the rate of O2 evolution up to fourfold. This stimulation could be partially replaced by the addition of chloride or nitrate ions. However, the addition of HCO 3 ? in the presence of Cl? or NO 3 ? resulted in a higher stimulation of O2 evolution (sixfold in the presence of nitrate and sevenfold in the presence of chloride). On the other hand, the addition of HCO 3 ? to the thylakoids depleted from salt only raised the rate of O2 evolution by 10–15%, whereas 40–70% was obtained by the addition of nitrate or chloride ions. The fluorescence induction studies indicated a significant decrease in the yield of the variable fluorescence of the salt- and HCO 3 ? -depleted thylakoids. A partial increase in the fluorescence yield was obtained by the addition of HCO 3 ? alone. A typical fluorescence induction curves were obtained by the addition of HCO 3 ? in the presence of Cl? or NO 3 ? ions. The data obtained suggest a similar role for chloride and nitrate ions in O2 evolution in the Hill-reaction, which is restricted at the donor side of photosystem II, whereas bicarbonate plays its role at both sides (acceptor and donor sides). The presented data are those obtained for the thylakoids of P. sativium, which were more or less similar to those obtained for L. sativa and R. sativus.  相似文献

The Cl? transport properties of the luminal border of bovine tracheal epithelium have been investigated using a highly purified preparation of apical plasma membrane vesicles. Transport of Cl? into an intravesicular space was demonstrated by (1) a linear inverse correlation between Cl? uptake and medium osmolarity and (2) complete release of accumulated Cl? by treatment with detergent. The rate of Cl? uptake was highly temperature-sensitive and was enhanced by exchange diffusion, providing evidence for a carrier-mediated transport mechanism. Transport of Cl? was not affected by the ‘loop’ diuretic bumetanide or by the stilbene-derivative anion-exchange inhibitors SITS (4-acetamido-4′-isothiocyanostilbene-2,2′-disulfonic acid) and DIDS (4,4′-diisothiocyanostilbene-2,2′-disulfonic acid). In the presence of the impermeant cation, tetramethylammonium (TMA+), uptake of Cl? was minimal; transport was stimulated equally by the substitution of either K+ or Na+ for TMA+. Valinomycin in the presence of K+ enhanced further Cl? uptake, while amiloride reduced Na+-stimulated Cl? uptake towards the minimal level observed with TMA+. These results suggest the following conclusions: (1) the tracheal vesicle membrane has a finite permeability to both Na+ and K+; (2) the membrane permeability to the medium counterion determines the rate of Cl? uptake; (3) Cl? transport is not specifically coupled with either Na+ or K+; and, finally (4) Cl? crosses the tracheal luminal membrane via an electrogenic transport mechanism.  相似文献

Summary The net loss of KCl observed in Ehrlich ascites cells during regulatory volume decrease (RVD) following hypotonic exposure involves activation of separate conductive K+ and Cl transport pathways. RVD is accelerated when a parallel K+ transport pathway is provided by addition of gramicidin, indicating that the K+ conductance is rate limiting. Addition of ionophore A23187 plus Ca2+ also activates separate K+ and Cl transport pathways, resulting in a hyperpolarization of the cell membrane. A calculation shows that the K+ and Cl conductance is increased 14-and 10-fold, respectively. Gramicidin fails to accelerate the A23187-induced cell shrinkage, indicating that the Cl conductance is rate limiting. An A23187-induced activation of42K and36Cl tracer fluxes is directly demonstrated. RVD and the A23187-induced cell shrinkage both are: (i) inhibited by quinine which blocks the Ca2+-activated K+ channel. (ii) unaffected by substitution of NO 3 or SCN for Cl, and (iii) inhibited by the anti-calmodulin drug pimozide. When the K+ channel is blocked by quinine but bypassed by addition of gramicidin, the rate of cell shrinkage can be used to monitor the Cl conductance. The Cl conductance is increased about 60-fold during RVD. The volume-induced activation of the Cl transport pathway is transient, with inactivation within about 10 min. The activation induced by ionophore A23187 in Ca2+-free media (probably by release of Ca2+ from internal stores) is also transient, whereas the activation is persistent in Ca2+-containing media. In the latter case, addition of excess EGTA is followed by inactivation of the Cl transport pathway. These findings suggest that a transient increase in free cytosolic Ca2+ may account for the transient activation of the Cl transport pathway. The activated anion transport pathway is unselective, carrying both Cl, Br, NO 3 , and SCN. The anti-calmodulin drug pimozide blocks the volume- or A23187-induced Cl transport pathway and also blocks the activation of the K+ transport pathway. This is demonstrated directly by42K flux experiments and indirectly in media where the dominating anion (SCN) has a high ground permeability. A comparison of the A23187-induced K+ conductance estimated from42K flux measurements at high external K+, and from net K flux measurements suggests single-file behavior of the Ca2+-activated K+ channel. The number of Ca2+-activated K+ channels is estimated at about 100 per cell.  相似文献

The Cl/HCO 3 exchange mechanism usually postulated to occur in gastric mucosa cannot account for the Na+-dependent electrogenic serosal to mucosal Cl transport often observed. It was recently suggested that an additional Cl transport mechanism driven by the Na+ electrochemical potential gradient may be present on the serosal side of the tissue. To verify this, we have studied Cl transport in guinea pig gastric mucosa. Inhibiting the (Na+, K+) ATPase either by serosal addition of ouabain or by establishing K+-free mucosal and serosal conditions abolished net Cl transport. Depolarizing the cell membrane potential with triphenylmethylphosphonium (a lipid-soluble cation), and hence reducing both the Na+ and Cl electrochemical potential gradients, resulted in inhibition of net Cl flux. Reduction of short-circuit current on replacing Na+ by choline in the serosal bathing solution was shown to be due to inhibition of Cl transport. Serosal addition of diisothiocyanodisulfonic acid stilbene (an inhibitor of anion transport systems) abolished net Cl flux but not net Na+ flux. These results are compatible with the proposed model of a Cl/Na+ cotransport mechanism governing serosal Cl entry into the secreting cells. We suggest that the same mechanism may well facilitate both coupled Cl/Na+ entry and coupled HCO 3 /Na+ exit on the serosal side of the tissue.  相似文献

Summary The vacuolar H+ ATPase is inhibited by N-ethylmaleimide (NEM), a sulfhydryl compound, suggesting the involvement of a sulfhydryl group in this transport process. We have examined the effects of several sulfhydryl-containing compounds on the vacuolar H+ ATPase of rabbit renal cortical endosomes. A number of such compounds were effective inhibitors of endosomal H+ transport at 10–5–10–6 m, including NEM, mersalyl, aldrithiol, 5,5 dithiobis (2-nitrobenzoic acid),p-chloromercuribenzoic acid (PCMB) andp-chloromercuriphenyl sulfonic acid (PCMBS). NEM, mersalyl, aldrithiol and PCMBS had no effect on pH-gradient dissipation, whereas PCMB decreased the pH gradient faster than control. In the absence of ATP, PCMB (10–4 m) stimulated endosomal36Cl uptake, particularly in the presence of an inside-alkaline pH gradient (pHin=7.6/pHout=5.5.). This result was not an effect of PCMB on the Cl-conductive pathway. The less permeable PCMBS did not stimulate36Cl uptake. The effects of PCMB were concentration dependent and were prevented by dithioerithritol,. ATP-dependent36Cl uptake was decreased by addition of PCMB. Finally, PCMB had no effect on45Ca2+ uptake. These results support the presence of two functionally important sulfhydryl groups in this endosomal preparation. One such group is involved with ATP-driven H+ transport and must be located on the cytoplasmic surface of the endosomal membrane. The second sulfhydryl group must reside on the internal surface of the endosomal membrane and relates to a PCMB-activated Cl/OH exchanger that is functional both in the presence and absence of ATP. This endosomal transporter is similar to the PCMB-activated Cl/OH exchanger recently described in rabbit renal brush-border membranes.  相似文献

Summary The changes in the cytoplasmic Cl concentration, [Cl] c , are monitored at the time of withdrawal (starvation) and subsequent replacement of Cl in the outside medium. The measurement technique exploits the involvement of Cl inChara excitation. The transient clamp current due to Cl,I Cl, is separated from other excitation transients through Hodgkin-Huxley (HH) equations, which have been adjusted toChara. TheI Cl amplitude depends on HH parameters, [Cl] c and the maximum membrane conductance to Cl, . The results are discussed in terms of these quantities.I Cl and were found to fall after 6–10 hr of Cl starvation, thus supporting the hypothesis that [Cl c decreases in Cl-free medium. The best HH fit to starved data was obtained with [Cl c =3.5mm. The time-course forI Cl decline is considerably slower than the time-course of the rise of the starvation-stimulated influx. As cells starved for periods longer than 24 hr are re-exposed to Cl, it is revealed that while [Cl] c remains low during long starvation, increases to values greater than those of the normal cells. Such differences among cells starved for various lengths of time have not been detected previously.  相似文献

Summary The characteristics of Cl movement across luminal and basolateral membranes ofAmphiuma intestinal absorptive cells were studied using Cl-sensitive microelectrodes and tracer36Cl techniques. Intracellular Cl activity (a Cl i ) was unchanged when serosal Cl was replaced; when luminal Cl was replaced cell Cl was rapidly lost. Accordingly, the steady statea Cl i could be varied by changing the luminal [Cl]. As luminal [Cl] was raised from 1 to 86mM,a Cl i rose in a linear manner, the mucosal membrane hyperpolarized, and the transepithelial voltage became serosa negative. In contrast, the rate of Cl transport from the cell into the serosal medium, measured as the SITS-inhibitable portion of the Cl absorptive flux, attained a maximum whena Cl i reached an apparent value of 17mm, indicating the presence of a saturable, serosal transport step. The stilbeneinsensitive absorptive flux was linear with luminal [Cl], suggestive of a paracellular route of movement. Intracellulara Cl was near electrochemical equilibrium at all but the lowest values of luminal [Cl] after interference produced by other anions was taken into account.a Cl i was unaffected by Na replacement, removal of medium K, or elevation of medium HCO 3 . Mucosae labeled with36Cl lost isotope into both luminal and serosal media at the same rate and from compartments of equal capacity. Lowering luminal [Cl] or addition of theophylline enhanced luminal Cl efflux. It is concluded that a conductive Cl leak pathway is present in the luminal membrane. Serosal transfer is by a saturable, stilbene-inhibitable pathway. Luminal Cl entry appears to be passive, but an electrogenic uptake cannot be discounted.  相似文献

Pyocyanin (N-methyl-1-hydroxyphenazine), a redox-active virulence factor produced by the human pathogen Pseudomonas aeruginosa, is known to compromise mucociliary clearance. Exposure of human bronchial epithelial cells to pyocyanin increased the rate of cellular release of H2O2 threefold above the endogenous H2O2 production. Real-time measurements of the redox potential of the cytosolic compartment using the redox sensor roGFP1 showed that pyocyanin (100 μM) oxidized the cytosol from a resting value of − 318 ± 5 mV by 48.0 ± 4.6 mV within 2 h; a comparable oxidation was induced by 100 μM H2O2. Whereas resting Cl secretion was slightly activated by pyocyanin (to 10% of maximal currents), forskolin-stimulated Cl secretion was inhibited by 86%. The decline was linearly related to the cytosolic redox potential (1.8% inhibition/mV oxidation). Cystic fibrosis bronchial epithelial cells homozygous for ΔF508 CFTR failed to secrete Cl in response to pyocyanin or H2O2, indicating that these oxidants specifically target the CFTR and not other Cl conductances. Treatment with pyocyanin also decreased total cellular glutathione levels to 62% and cellular ATP levels to 46% after 24 h. We conclude that pyocyanin is a key factor that redox cycles in the cytosol, generates H2O2, depletes glutathione and ATP, and impairs CFTR function in Pseudomonas-infected lungs.  相似文献

Ischemic heart disease is the leading cause of serious morbidity and mortality in Western society. One of the therapeutic approaches is based on the use of thrombolitic drugs that promote clot lysis. Even if the mechanisms leading to clot lysis are not completely understood, it is widely accepted that they depend on the complex biochemical reactions that occur among fibrin fibers and fibrinolitic agents, and by their ready diffusion into the fibers. Here we investigate the effects of specific anions on the architecture of protofibrils within fibrin fibers in fibrin gels prepared in a para-physiological solution. The results obtained through small-angle X-ray scattering (SAXS) demonstrate that the characteristic axial and longitudinal repeat distances among protofibrils are strongly affected by the action of Cl and F anions.  相似文献

Na+, Li+ and Cl− transport by brush border membranes from rabbit jejunum   总被引:1,自引:0,他引:1  
Na+, Li+, K+, Rb+, Br-, Cl- and SO4(2-) transport were studied in brush border membrane vesicles isolated from rabbit jejunum. Li+ uptakes were measured by flameless atomic absorption spectroscopy, and all others were measured using isotopic flux and liquid scintillation counting. All uptakes were performed with a rapid filtration procedure. A method is presented for separating various components of ion uptake: 1) passive diffusion, 2) mediated transport and 3) binding. It was concluded that a Na+/H+ exchange mechanism exists in the jejunal brush border. The exchanger was inhibited with 300 microM amiloride or harmaline. The kinetic parameters for sodium transport by this mechanism depend on the pH of the intravesicular solution. The application of a pH gradient (pHin = 5.5, pHout = 7.5) causes an increase in Jmax (50 to 125 pmol/mg protein . sec) with no change in Kt (congruent to 4.5 nM). Competition experiments show that other monovalent cations, e.g. Li+ and NH4+, share the Na+/H+ exchanger. This was confirmed with direct measurements of Li+ uptakes. Saturable uptake mechanisms were also observed for K+, Rb+ and SO4(2-), but not for Br-. The Jmax for K+ and Rb+ are similar to the Jmax for Na+, suggesting that they may share a transporter. The SO4(2-) system appears to be a Na+/SO4(2-) cotransport system. There does not appear to be either a Cl-/OH- transport mechanism of the type observed in ileum or a specific Na+/Cl- symporter.  相似文献

Summary Cl channels from basolaterally-enriched rabbit outer renal medullary membranes are activated either by increases in intracellular Cl activity or by intracellular protein kinase A (PKA). Phosphorylation by PKA, however, is not obligatory for channel activity since channels can be activated by intracellular Cl in the absence of PKA. The PKA requirement for activation of Cl channels in certain secretory epithelia is, in contrast, obligatory. In the present studies, we examined the effects of PKA and intracellular Cl concentrations on the properties of Cl channels obtained either from basolaterally-enriched vesicles derived from highly purified suspensions of mouse medullary thick ascending limb (mTALH) segments, or from apical membrane vesicles obtained from two secretory epithelia, bovine trachea and rabbit small intestine. Our results indicate that the Cl channels from mTALH suspensions were virtually identical to those previously described from rabbit outer renal medulla. In particular, an increase in intracellular (trans) Cl concentration from 2 to 50 mm increased both channel activity (P o) and channel conductance (g Cl, pS). Likewise, trans PKA increased mTALH Cl channel activity by increasing the activity of individual channels when the trans solutions were 2 mm Cl. Under the latter circumstance, PKA did not activate quiescent channels, nor did it affect g Cl. Moreover, when mTALH Cl channels were inactivated by reducing cis Cl concentrations to 50 mm, cis PKA addition did not affect P o. These results are consistent with the view that these Cl channels originated from basolateral membranes of the mTALH.Cl channels from apical vesicles from trachea and small intestine were completely insensitive to alterations in trans Cl concentrations and demonstrated markedly different responses to PKA. In the absence of PKA, tracheal Cl channels inactivated spontaneously after a mean time of 8 min; addition of PKA to trans solutions reactivated these channels. The intestinal Cl channels did not inactivate with time. Trans PKA addition activated new channels with no effect on basal channel activity. Thus the regulation of Cl channel activity by both intracellular Cl and by PKA differ in basolateral mTALH Cl channels compared to apical Cl channels from either the tracheal or small intestine.We acknowledge the able technical assistance of Steven D. Summary The potential dependence of unidirectional36Cl fluxes through toad skin revealed activation of a conductive pathway in the physiological region of transepithelial potentials. Activation of the conductance was dependent on the presence of Cl or Br in the external bathing solution, but was independent of whether the external bath was NaCl-Ringer's, NaCl-Ringer's with amiloride, KCl-Ringer's or choline Cl-Ringer's To partition the routes of the conductive Cl ion flow, we measured in the isolated epithelium with double-barrelled microelectrodes apical membrane potentialV a , and intracellular Cl activity,a Cl c , of the principal cells indentified by differential interference contrast microscopy. Under short-circuit conditionsI sc=27.0±2.0 A/cm2, with NaCl-Ringer's bathing both surfaces,V a was –67.9±3.8mV (mean ±se,n=24, six preparations) anda Cl c was 18.0±0.9mM in skins from animals adapted to distilled water. BothV a anda Cl a were found to be positively correlated withI sc (r=0.66 andr=0.70, respectively). In eight epithelia from animals adapted to dry milieu/tap waterV a anda Cl c were measured with KCl Ringer's on the outside during activation and deactivation of the transepithelial Cl conductance (G Cl) by voltage clamping the transepithelial potential (V) at 40 mV (mucosa positive) and –100 mV. AtV=40 mV; i.e. whenG Cl was deactivated,V a was –70.1±5.0 mV (n=15, eight preparations) anda Cl c was 40.0±3.8mm. The fractional apical membrane resistance (fR a) was 0.69±0.03. Clamping toV=–100 mV led to an instantaneous change ofV a to 31.3±5.6 mV (cell interior positive with respect to the mucosal bath), whereas neithera Cl c norfR a changed significantly within a 2 to 5-min period during whichG Cl increased by 1.19±0.10 mS/cm2. WhenV was stepped back to 40 mV,V a instantaneously shifted to –67.8±3.9 mV whilea Cl c andfR a remained constant during deactivation ofG Cl. Similar results were obtained in epithelia impaled from the serosal side. In 12 skins from animals adapted to either tap water or distilled water the density of mitochondria-rich (D MRC) cells was estimated and correlated with the Cl current (I Cl though the fully activated (V=–100mV) Cl conductance). A highly significant correlation was revealed (r=–0.96) with a slope of –2.6 nA/m.r. (mitochondria-rich cell and an I-axis intercept not significantly different from zero. In summary, the voltage-dependent Cl currents were not reflected infR a anda Cl a of the principal cells but showed a correlation with the m.r. cell density. We conclude that the pricipal cells do not contribute significantly to the voltage-dependent Cl conductance.  相似文献

Summary The effect of the loop diuretic furosemide (4-chloro-N-furfuryl-5-sulfamoyl-anthranilic acid) on the thiol-dependent, ouabain-insensitive K(Rb)/Cl transport in low K+ sheep red cells was studied at various concentrations of extracellular Rb+, Na+ and Cl. In Rb+-free NaCl media, 2×10–3 m furosemide inhibited only one-half of thiol-dependent K+ efflux. In the presence of 23mm RbCl, however, the concentration of furosemide to produce 50% K+ efflux inhibition (IC50) was 5×10–5 m. In Rb+ containing NaCl media, the inhibitory effect of 10–3 m furosemide was equal to that caused by NO 3 replacement of Cl in the medium. The apparent synergistic action of furosemide and external Rb+ on K+ efflux was also seen in the ouabain-insensitive Rb+ influx. A preliminary kinetic analysis suggests that furosemide binding alters both maximal K+(Rb+) transport and apparent external Rb+ affinity. In the presence of external Rb+, Na+ (as compared to choline) exerted a small but significant augmentation of the furosemide inhibition of K+(Rb+) fluxes. There was no effect of Cl on the IC50 value of furosemide. As there is no evidence for coupled Na+K+ cotransport in low K+ sheep red cells, furosemide may modify thiol-dependent K+(Rb+/Cl flux or Rb+ (and to a slight degree Na+) modulate the effect of furosemide.  相似文献
