The purpose of this study was to investigate the efficiency of superdisintegrants in promoting tablet disintegration and drug dissolution under varied media pH. Significant reductions in the rate and extent of water uptake and swelling were observed for both sodium starch glycolate (Primojel) and croscarmellose sodium (Ac-Di-Sol) in an acidic medium (0.1 N HCl) but not for crospovidone NF (Polyplasdone XL10), a nonionic polymer. When Primojel and Ac-Di-Sol were incorporated in model formulations, a significant increase in tablet disintegration time was observed for slowly disintegrating tablets (lactose-based tablets) but not for the rapidly disintegrating tablets (dicalcium phosphate-based tablets). The dissolution rate of the model drug, hydrochlorothiazide, was found highly dependent on both tablet disintegration efficiency and the solubility of base material(s) in the testing medium. A laser diffraction particle size analyzer proved to be an effective tool for determining the intrinsic swelling of disintegrant particles in different media. Water uptake and swelling were confirmed as 2 important functions of superdisintegrants. The reduced water uptake and swelling capacity of disintegrants containing ionizable substituents in an acidic medium can potentially jeopardize their efficiency in promoting tablet disintegration and the drug dissolution rate. 全球气候变化将增加未来高温与干旱的发生频率和强度,然而高温与干旱的交互作用对农作物生长、养分含量及其利用效率的影响还不甚清楚。因此,研究高温与干旱交互作用对农作物生理生态的影响将为准确评价农作物对未来极端气候条件的响应提供科学依据。选取全球第四大经济作物——西红柿为研究对象,在人工智能气候箱中模拟高温和干旱环境。共设置两个水分处理(正常浇水;干旱)与两个温度处理(常温-26℃/19℃(白天/夜间);高温-42℃/35℃(白天/夜间)(7d))。主要测定指标包括生物量以及生物量分配、比叶面积、养分含量(全氮、全磷)、光合元素利用效率(光合氮素利用效率、光合磷素利用效率)。研究表明,高温、干旱单独作用以及交互作用均显著降低了根、茎、叶生物量以及总生物量,并且高温干旱交互作用使总生物量降低最多。在生物量分配方面,高温单独作用显著降低了根质量分数以及根冠比,而干旱单独作用增加了根质量分数、茎质量分数以及根冠比,但降低了叶质量分数。在养分含量方面,高温单独作用导致叶片全氮、全磷含量显著降低、茎全磷含量显著增加、根全磷含量显著降低。干旱单独作用导致叶片、茎全磷含量显著降低、根全氮含量显著升高。高温与干旱交互作用对生物量分配及养分含量的影响与干旱胁迫单独作用类似。在光合元素利用效率方面,高温、干旱单独作用均降低了幼苗光合氮素利用效率、光合磷素利用效率,并且高温加剧了干旱对光合磷素利用效率的影响。因此,在未来气候变化情况下,高温与干旱交互作用可能会对农作物产生更大威胁。  相似文献

Variousoptions exist for re-engineering of traditionalserum-free formulations (containing insulin,transferrin and other protein factors) withnon-protein substitutes. Caution must also beexercised with sourcing of non-protein additives,particularly amino acids and lipids, to avoidintroducing adventitious contaminants. Simpleguidelines facilitate adaptation, cryopreservation andrecovery of many cell types within a protein-freeculture environment. Scrupulous maintenance offacility and equipment and monitoring of processwater, air handling systems and technical personnelare required to ensure that approved raw materials arecorrectly formulated and dispensed. Simple method of Arabidopsis thaliana w.t. cv. Columbia (L.) Heynh. cultivation in liquid nutrient medium is presented. After 5 weeks of growth in soil, the plants were transferred to modified Hoagland nutrient medium. This allowed us to cultivate Arabidopsis in conditions comparable to all other hydroponically grown higher plants used in plant physiology and plant stress physiology experiments. Absence of agar in growth medium and free access to whole root system makes this method useful also in experiments concerning root physiology.  相似文献

Bacillus flexus was isolated from local soil sample and identified by molecular methods. In inorganic nutrient medium (IM) containing sucrose as carbon source, yield of biomass and polyhydroxyalkanoate (PHA) were 2 g/l and 1 g/l (50% of biomass), respectively. Substitution of inorganic nitrogen by peptone, yeast extract or beef extract resulted in biomass yields of 4.1, 3.9 and 1.6 g/l, respectively. Corresponding yields of PHA in biomass was 30%, 40% and 44%. Cells subjected to change in nutrient condition from organic to inorganic, lacked diaminopimelic acid in the cell wall and the concentration of amino acids also decreased. Under these conditions the extractability of the polymer from the cells by hot chloroform or mild alkali hydrolysis was 86–100% compared to those grown in yeast extract or peptone (32–56%). The results demonstrated that growth, PHA production and the composition of cell wall of B. flexus are influenced by the organic or inorganic nutrients present in the growth medium. Cells grown in inorganic medium lysed easily and this can be further exploited for easier recovery of the intracellular PHA.  相似文献

The presence of 10-6 M bovine insulin in chemically defined nutrient medium prevented cell death and improved cell proliferation of the ciliate Tetrahymena pyriformis when inoculated at low initial cell density. The action of insulin was found to be restricted to the 22-30 fragment of the B-chain. These results suggest that small peptides are involved in regulation of cell survival and cell proliferation in Tetrahymena pyriformis.  相似文献

猎物匮乏是影响东北虎(Panthera tigris altaica)种群恢复的关键因素之一。容纳量研究是开展东北虎猎物恢复工作的必要前提。通过Maxent模型、聚类分析和训练随机树分类等方法,结合调查数据,预测了吉林省张广才岭南部黄泥河林业局东北虎主要猎物的适宜栖息地空间分布,解译了植被类型,在此基础上基于不同植被类型动物可采食部分代谢能、不同生境等级食物可利用率、马鹿(Cervus elaphus)和狍(Capreolus capreolus)生境等级重叠情况以及动物冬季能量需求,分析了东北虎猎物的冬季营养容纳量。结果表明:黄泥河林业局狍、野猪(Sus scrofa)和马鹿的适宜栖息地分别占研究区域总面积的52.8%、40.7%和25.4%;从猎物获取能量来看,以山杨(Populus davidiana)、桦树(Betula)、核桃楸(Juglans mandshurica)为主的植被类型是马鹿、狍可获得能量较多的生境,以蒙古栎(Quercus mongolica)、核桃楸为主的植被类型是野猪可获得能量较多的生境。东北虎猎物种群的综合冬季营养容纳量为574只马鹿(0.29只/km2),7016只狍(3.54只/km2),4785只野猪(2.38只/km2)。  相似文献

萌生更新是森林更新的重要方式, 是硬叶栎林受到干扰后植被恢复的主要机制。以位于青藏高原东南缘的川西折多山东坡川滇高山栎(Quercus aquifoliodes)灌丛为研究对象, 调查分析了砍伐后灌丛萌生过程中基株根系和萌株生物量动态、营养元素含量, 以及基株根系和土壤对萌株生长过程中的营养元素供应动态。结果表明, 川滇高山栎灌丛平均地上和地下生物量分别为(11.25 ± 0.92) t·hm-2和(34.85 ± 2.02) t·hm-2, 具有较大的根冠比(3.10:1); 萌生过程中, 萌株生物量呈线性增加趋势, 以灌丛活细根生物量变化为最大, 其次是活中根和活粗根, 树桩和根蔸生物量变化最小; 萌生过程中, 灌丛细根和中根N、P含量表现为先增加、后降低的变化趋势, 萌生初期树桩、粗根和根蔸中N和K的含量明显下降, 根蔸中Ca含量略有下降, 而P没有明显下降, 根系Mg含量变化幅度较大, 灌丛地下根系储存了较多的营养元素; 土壤、树桩、粗根和根蔸是川滇高山栎灌丛砍伐后0-120天萌生生长的主要营养来源, 砍伐后60天, 萌株生长所需的营养除K元素主要来源于根系外, 其余营养元素主要来源于土壤; 在砍伐后60-120天, 基株根系对萌株生长所需的N、K和Ca贡献较大, 而对P和Mg的贡献较小; 在砍伐后120-180天, 根系除K元素对萌生生长还保持较大的贡献外, 对其余营养元素的贡献均较小。高山栎林管理要注重加强地下根系的保护。  相似文献

成都平原属典型的水田农业区,复种指数高,农作物一年两熟或三熟。为探究高土地利用率地区农田土壤养分特征及其空间分布格局,基于成都市郫都区2015年测土配方施肥数据,运用经典统计学和地统计学方法揭示该区域农田土壤养分的描述性统计特征及其空间变异。结果显示:郫都区农田土壤有机质(SOM)、总氮(TN)、碱解氮(AN)、速效磷(AP)、速效钾(AK)含量均值分别为22.35 g·kg^-1、1.15 g·kg^-1、95.38 mg·kg^-1、21.01 mg·kg^-1和104.58 mg·kg^-1,不同土壤类型及不同种植模式农田土壤养分差异总体较小,表明高强度的人类活动使郫都区土壤养分含量变得均一;半方差分析显示,SOM和AK的块金系数分别为0.39和0.62,其空间变异由土壤母质、地形、气候条件等结构性因素和耕作制度、施肥条件等随机性因素共同作用引起;TN、AN和AP的块金系数分别为0.87、0.94和0.97,其空间变异性主要受耕作制度、施肥条件等随机性因素影响;结合已有研究来看,郫都区农田SOM块金系数呈下降趋势而TN、AN和AP块金系数呈增加趋势,表明随着郫都区作物产量与作物复种指数的不断提高,以蔬菜为主的旱作模式代替传统的水旱轮作模式、复合肥代替农家肥等生产方式的改变正在影响着郫都区农田土壤养分的空间分布格局。  相似文献

Nutrient resorption from senescing tissues increases plant nutrient-use efficiency, and may be an adaptation to nutrient limitation. In some tree species, retranslocation of nutrients from sapwood during heartwood formation is a comparable process. We measured Ca, Mg and K concentrations in Atlantic white cedar (Chamaecyparis thyoides) stemwood samples taken from two swamps in the northeastern United States and compared them to soil mineral nutrient availability at each site. We found that Ca, Mg and K concentrations were 60–700% higher in sapwood than in the immediately adjacent heartwood, indicating retranslocation of these nutrients from senescing sapwood. Sapwood nutrient concentrations were similar between the two sites. However, nutrient concentrations in the heartwood differed significantly between the sites, as did the relative degree of Ca and Mg retranslocation from senescing sapwood. We found these differences between sites to be inversely related to significant differences in exchangeable Ca, Mg and K as well as Al concentrations in the soil. These findings suggest that the degree of nutrient retranslocation from senescing sapwood may be influenced by soil nutrient availability. The changes in nutrient content of grain tissues and seedling parts of two wheat genotypes (Triticum aestivum L., Excalibur and Janz) with low or high seed Zn content were followed from imbibition to early seedling development (12 days). The grains were separated into seed coat, endosperm and embryo, while the seedlings were separated into roots and shoots. The dry weight of the seed coat did not change throughout the experimental period, whereas the endosperm weight declined rapidly from day 4 onward. The weight of embryo did not show any difference between and within cultivars. About a half of seed Zn was remobilised into shoot and roots during 12 days of growth, regardless of the initial seed Zn content in both genotypes. The seed coat contained 55–77% of the total seed nutrients in the two wheat genotypes, except in the case of S (around 40%). Manganese, Fe, Ca, K, and P were remobilised effectively from the seed coat as well as from the endosperm, while remobilisation of Zn and Cu was relatively less from the seed coat than from the endosperm. After 10 days of growth, all nutrients monitored were completely remobilised from the endosperm. Remobilised K was directed primarily into shoots; an increase in K content in shoots was relatively higher than the accumulation of dry matter, with a consequent increase in K concentration in shoot tissue. The remobilisation of some nutrients (eg. Fe, Ca and Zn) from various grain tissues during inbibition, germination and early growth is different from the remobilisation in more mature plants.  相似文献

Neisseria gonorrhoeae is difficult to cultivate in liquid medium. Currently there are no liquid media, defined or undefined, that reliably permit growth of this bacterium from low inocula. Standard clinical laboratory broths may allow multiplication of some strains of gonococci from large inocula, but such media incorporate infusates, extracts or digests and are therefore undefined. In this study, 20 gonococci of ten auxotypes were tested in various experimental media in the development of an easily prepared chemically defined, clear and protein-free liquid medium. The final medium - GW medium - allowed the growth of three clinical isolates of gonococci from inocula of <10(3) CFU mL(-1) to >10(8) CFU mL(-1) by 24 h. None of four commercially-available broths (nutrient broth, brain heart infusion, tryptone soya broth, and Mueller-Hinton broth) tested in parallel reliably supported growth of these isolates to the same extent. GW medium should be useful for studies of the growth of gonococci under different conditions and, as the medium is clear and colorless, this can be monitored turbidometrically. GW medium may be suitable as a basal medium for biochemical identification tests, antimicrobial susceptibility determinations and antimicrobial synergy studies.  相似文献

Taxol is the most effective antitumor agent developed in the past three decades. It has been used for effective treatment of a variety of cancers. A taxol-producing endophytic fungus Pestalotiopsis pauciseta (strain CHP-11) was isolated from the leaves of Cardiospermum helicacabum and screened for taxol production. The fungus was identified based on the morphology of the fungal culture and the characteristics of the spores and screened for taxol production. The amount of taxol produced by this endophytic fungus was quantified by HPLC and it produced 113.3 mg/L, thus the fungus can serve as a potential material for fungus engineering to improve taxol production. This fungal taxol also had strong anticancer activity against some cancer cells viz., BT 220, H116, Int 407, HL 251 and HLK 210 tested by Apoptotic assay and it is indicated that with the increase of taxol concentration from 0.005–0.05 mmol/L, taxol induced increased cell death through apoptosis.  相似文献

Forty weaned male guinea pigs of 208.20±6.62 g mean body weight were divided into 4 groups of 10 animals in a randomized block design. All of the guinea pigs were fed a basal diet [25% ground maize hay, 30% ground maize grain, 22% ground chickpea (Cicer arietinum L.), 9.5% deoiled rice bran, 6% soybean meal, 6% fish meal, 1.45% mineral supplement (without Zn) and 0.05% ascorbic acid] and available green fodder. Group I served as the control (no Zn supplementation), whereas 20 ppm Zn was added in the diet in groups II, III, and IV either as zinc sulfate (ZnSO4), zinc amino acid complex (ZAAC), and ZnSO4 + ZAAC in equal parts, respectively. Experimental feeding lasted for 70 d, including a 3-d digestibility trial. Blood was collected through cardiac puncture from four animals in each group at d 0 and subsequently at the end of experimental feeding. After 40 d of experimental feeding, four animals from each group were injected with 0.4 mL of Brucella abortus cotton strain-19 vaccine to assess the humoral immune response of the animals. After 10 wk of study, four animals from each group were sacrificed to study the concentration of Zn, Cu, Co, Fe, and Mn in the liver, pancreas and spleen. Results revealed no significant difference in the feed intake, body weight gain, and digestibility of the nutrients, except for crude protein (CP) digestibility, which was significantly (p<0.05) lower in group IV. Although concentrations of serum glucose, Ca, and P and the albumin:globulin (A:G) ratio were similar in the different groups, the total protein, albumin, and serum alkaline phosphatase activity were higher in all of the Zn-supplemented groups on d 70. The serum Zn levels at the end of experimental feeding were significantly higher in groups II and III, whereas serum Mn levels were found to be significantly (p<0.05) higher in groups III and IV. The organ weights (as percentage of body weights) did not show any differences among the treatment groups. Although the Mn concentration was significantly (p<0.05) higher in the pancreas, the Cu concentration was significantly (p<0.05) reduced in the spleen in all of the Zn-supplemented groups. The humoral immune response (antibody titer values) on d 14 of vaccination was significantly (p<0.05) higher in all of the Zn-supplemented groups. It was concluded that the 20-ppm level of Zn in the diet might be adequate for growth and nutrient utilization in guinea pigs, but supplementation of 20-ppm zinc significantly improved the immune response and impact was more prominent with the ZAAC (organic source) compared to ZnSO4 (inorganic source).  相似文献

When cells are starved of their substrate, many nutrient transporters are induced. These undergo rapid endocytosis and redirection of their intracellular trafficking when their substrate becomes available again. The discovery that some of these transporters also act as receptors, or transceptors, suggests that at least part of the sophisticated controls governing the trafficking of these proteins has to do with their signaling function rather than with control of transport. In yeast, the general amino acid permease Gap1 mediates signaling to the protein kinase A pathway. Its endocytic internalization and intracellular trafficking are subject to amino acid control. Other nutrient transceptors controlling this signal transduction pathway appear to be subject to similar trafficking regulation. Transporters with complex regulatory control have also been suggested to function as transceptors in other organisms. Hence, precise regulation of intracellular trafficking in nutrient transporters may be related to the need for tight control of nutrient-induced signaling.  相似文献

Treatment of in vitro matured bovine oocytes with colcemid results in a membrane protrusion that contains maternal chromosomes, which can be easily removed by aspiration. Four experiments were designed to evaluate the overall and temporal effects of conditioned medium (CM) by bovine cumulus cells on development of nuclear transfer (NT) bovine embryos and to examine the chromosomal composition and allocation of inner cell mass (ICM) and trophectoderm (TE) of the subsequent blastocysts. The nuclear transfer embryos were cultured in various CR1aa media conditioned by preculture with bovine cumulus cells. Development to the blastocyst stage in BSA-containing CM (BCM) and serum-containing CM (SCM) were similar to co-culture group (24-30%). The 24 hr-conditioned BCM yielded higher blastocyst development than 48 and 72 hr-conditioned BCM. Temporary exposure of embryos to BCM and SCM followed by CR1aa was also studied. Morula and blastocyst development were not different among the groups cultured in BCM for 72, 96, and 168 hr, but were significantly higher (P < 0.01) than groups exposed to BCM for 24 and 48 hr, respectively. Blastocyst development in SCM for 24 hr (29%), 96 hr (25%), and 168 hr (27%) were much higher (P < 0.05) than those in SCM for 48 hr (12%) and 72 hr (10%). The analyses of chromosomal composition of the resulting blastocysts indicate approximately 80% of the blastocysts cultured in CR1aa with co-culture or groups initially exposed to BCM for 24 hr followed by culture in CR1aa were diploid. However, the incidence of diploidy were only 36-60% in SCM-cultured groups and groups cultured in BCM beyond 48 hr. Conditioned media did not affect the allocation of ICM and TE in the blastocyst. No difference was found in the ratio of inner cell mass to total cells in co-culture, BCM or SCM groups (0.424, 0.441, and 0.473, respectively). In conclusion, bovine cumulus cell-CM and CR1aa with co-culture supported comparable development and blastocyst ICM:total cell ratio of bovine NT embryos. However, CM affected the blastocyst chromosomal composition and induced higher mixploidy.  相似文献

Dose-response curves were compared for deletions [ColBR (resistant to colicin B) mutations being more than 80% deletions] and base changes (reversion of argFam to prototrophy argplus) induced in the same set of E. coli strains (wild-type for DNA repair, uvrA-, polA- and recA-) by N-methyl-N'-nitro-N-nitrosoguanidine (NTG), ethyl methanesulfonate (EMS), hydroxylamine (HA), 4-nitroquinoline I-oxide (4NQO), mitomycin C (MTC, UV and X-rays. All these agents induced deletions as well as base changes in the wild-type strain. Thus chemical mutagenesis differed in E. coli and bacteriophages in vitro, for HA, NTG, EMS and perhaps UV produced only point mutations in phage Tr. The patterns of deletion and base-change mutability in E. coli were surprisingly similar. (I) The recombination less recA- strain was mutable by only three (NTG, EMS, HA) of the seven mutagens for either deletions or base changes. (2) The uvrA- strain, unable to excise pyrimidine dimers, was very highly mutable by 4NQO and UV but immutable by MTC for both deletions and base changes. (3) The polA- strain, defective in DNA polymerase I due to a non-suppressible mutation, was very highly mutable by HA and highly mutable by MTC and 4NQO for both deletions and base changes but was highly mutable only for deletions by UV and X-rays, remaining normally mutable by the other agents for both deletions and base changes despite its high sensitivity to their inactivating action. We conclude that errors in the recA-dependent repair of induced DNA damage (after 4NQO, MTC, UV and X-rays) or errors in replication enhanced by damage to the replication system or to the template strands (after NTG, EMS, and HA) give rise to deletions as well as to base changes. From a comparative analysis of 14 dose-response curves for deletions and base changes, we conclude that the order of mutagenic efficiency relative to killing is (EMS, NTG) greater than (UV, 4NQO) greater than HA greater than (X-rays, MTC), and that X-rays, 4NQO, HA and MTC induce more ColBR deletions than Argplus base changes, whereas UV and EMS induce ColBR deletions and Argplus base changes at nearly equal rates and the specificity of NTG is intermediate between these two types.  相似文献

该研究通过野外采样和实验室测定的方法,研究了三种生境中鬼针草叶和根碳(C)、氮(N)、磷(P)化学计量特征及其与土壤养分的相关性。结果表明:(1)鬼针草的生境具有不同的资源水平。三种生境的土壤全氮(TN)和速效氮贫乏,有机碳(C)和全磷(TP)较充裕;生境Ⅰ土壤TN含量显著低于其它两个生境,生境II土壤TP含量显著低于其它两个生境,生境III土壤TN、TP含量均大于其它两个生境。(2)不同生境的鬼针草对磷(P)分配策略不同。低N生境的鬼针草叶片P含量根P含量,P较多地分配到植物体地上部分;N、P含量较高的生境中鬼针草根P含量叶P含量,P更多地分配到地下部分。(3)不同生境的鬼针草其地上部分和根的生长速率不同。低N生境的鬼针草叶片N/P和C/P值小,植物体具有较高的相对生长速率,具有地上生长竞争优势;低P生境的植物叶片N/P和C/P值大,植物体具有较慢的相对生长速率;高N、高P生境中根N/P和C/P值小,根具有较高的生长速率,保证了鬼针草的地下生长竞争优势。(4)鬼针草叶片N/P和根N/P之间呈现不显著的负相关关系,植物地上部分和地下部分为异速生长。不同生境的鬼针草具有不同的营养利用和分配策略,保证了植物强大的竞争力和入侵性。  相似文献
