The effect of abscisic acid (ABA) and CaCl2 in the medium on RNA synthesis in embryonic axes of seeds of chick-pea (Cicer arietinum L. cv. Castellana) during the first hours of germination has been studied. ABA decreases the incorporation of 3H-uridine in the embryonic axes and modifies the mRNA populations by preventing the disappearance of three polypeptides of 25, 22 and 21.6 kDa and inducing the appearance of two polypeptides of 35 and 27 kDa absent in the control. Calcium increases the effect of ABA on the synthesis of these mRNAs and seems to be involved in the synthesis of at least the polypeptide of 25 kDa, as this polypeptide disappears in the treatments with EGTA and Verapamil. Moreover, cytosolic calcium seems to be necessary for the accumulation of these messengers since treatment with TMB-8 (chelating agent for endogenous calcium) decreases the intensity of the bands corresponding to these 5 polypeptides. The possible synergistic action of ABA and calcium in this process is discussed.  相似文献

Changes in the activities of adenyl cyclase, cyclic AMP phosphodiesterase, protein phosphokinase, RNase, protease, DNA, RNA and protein synthesis during the initial imbibition phase of the germination cycle of Cicer arietinum (chick pea, Bengal gram) are reported. Activation of adenyl cyclase and phosphorylation of cellular proteins appears to precede RNA and protein synthesis in the imbibed seeds.  相似文献

A lectin was isolated from seed extracts of Cicer arietinum by (NH4)2SO4 precipitation and subsequent ion exchange chromatography and gel filtration. Affinity chromatography on desialylated human IgM coupled to AH-Sepharose was also performed, but the amount bound was very low. The lectin has a molecular mass of about 44000 Da, as determined by ultracentrifugation and gel filtration. Dodecyl sulphate polyacrylamide-gel electrophoresis showed one band corresponding to a molecular mass of 26000 Da. N-Terminal amino acid sequence analyses indicate only one type of chain, suggesting that the lectin is probably dimeric. The amino acid composition is given. Papainized human erythrocytes of the different ABO groups were agglutinated equally well by the Cicer lectin, whereas untreated cells reacted weakly and only in the presence of bovine serum albumin. Simple sugars did not inhibit the agglutination, but some glycoproteins did inhibit. The lectin is probably nonmitogenic against human lymphocytes. Antigenic analyses in an enzyme-linked immunosorbent assay (ELISA) showed only a weak cross-reaction between Cicer and the lectins in the Vicieae tribe. Thus, our physicochemical and antigenic studies of the Cicer lectin support the botanical reasons recently given for removing the genus Cicer from the Vicieae tribe.  相似文献

The combined action of the inhibitors antimycin A and cyanide with benzohydroxamic acid indicates the presence of a cyanide-resistant pathway of respiration in chick pea (Cicer arietinum L.) seeds. The appearance of this pathway takes place during germination. During the first 12 hours of germination, the respiration is predominantly cyanide-sensitive, showing after this time a shift to an “alternate” respiration which is sensitive to benzohydroxamic acid, reaching the maximal cyanide resistance between 72 and 96 hours of germination. The appearance of the alternate pathway is initiated by high O2 concentrations and depends on cytoplasmic protein synthesis, since its appearance is inhibited by cycloheximide but not by chloramphenicol. Actinomycin D has no effect on the appearance of the alternate pathway. Our results indicate, in agreement with other authors, that the branching point is located between the flavoproteins and cytochromes b, probably at the level of ubiquinone, but the possibility of more than one branching point of the electron flow is also considered.  相似文献

Cicer arietinum L. cv. G 62404 was seen to have a very low fruit-setpercentage at low temperatures and as temperatures increased,either in nature or in culture conditions, the fruit-set alsoincreased. This effect of temperature could also be seen inpercent pollen germination and pollen tube growth. M 450, amutant of G 62404, was seen to have a better fruit-set percentageat low temperature and better pollen germination. This may bedue to differences in malic acid concentration, which increasedpollen germination, and pollen tube growth under in vitro cultureconditions.  相似文献

Ethylene production by isolated chick-pea embryonic axes during the initial stages of germination has been studied. Maximum production of ethylene occurs when growth is entirely due to cell elongation and before mitotic activity begins. This peak increases three-fold in the presence of calcium, but it is diminished by osmotic inhibitors, polyamines and abscisic acid (ABA), with a parallel fall in growth rate. Fusicoccin stimulates ethylene production and counteracts the effects of polyethylene glycol; thiourea, which breaks thermodormancy in chick-pea seeds, reduces ethylene production but does not counteract the effects of osmotic inhibitors. Of the polyamines studied, spermine (in low concentrations, 0.1 to 1.0 m M ) is the only one to stimulate ethylene production and cell elongation. It is concluded that there is a close relationship between ethylene production and cell elongation.  相似文献

The biosynthesis of nucleic acids and proteins was studied in rat uterus by following the incorporation of [3H]-thymidine, [3H]-uridineand[14C]-leucinein control and pregnant rats in the presence and absence of two anti-implantation drugs. One of the drugs, 78/224 caused a significant increase in incorporation whereas the other drug, Centchroman, caused an inhibition in incorporation of all the three precursors. The implications of these changes in the light of estrogenicity, agonist and antagonist actions of anti-estrogens have been analysed. The importance of homeostatic mechanisms involved in nucleic acids and proteins for the maintenance of constant internal milieu for blastocyst attachment has been discussed.  相似文献

A novel lectin (CAA-II) was isolated and purified from the seeds of Cicer arietinum by ammonium sulphate fractionation and affinity chromatography on an N-acetyl-D-galactosamine-linked agarose column. The lectin is composed of four identical subunits of 30 kDa and the molecular mass of the native lectin was estimated to be 120 kDa by gel filtration chromatography and confirmed by mass spectrometry. The lectin showed agglutination activity against rabbit erythrocytes (trypsin-treated and untreated) as well as against human erythrocytes. Haemagglutination inhibition assays showed that the lectin is a galactose-specific protein having a high affinity for N-acetyl-D-galactosamine. The molecular weight, haemagglutination pattern, carbohydrate specificity and N-terminal amino acid sequence indicated that the lectin is clearly distinct from the previously reported chickpea lectin CAA-I.  相似文献

A cDNA library was constructed in lambda TriplEx2 vector using poly (A(+)) RNA from immature seeds of Cicer arietinum. The lectin gene was isolated from seeds of chickpea through library screening and RACE-PCR. The full-length cDNA of Chichpea seed lectin(CpGL)is 972 bp and contains a 807 bp open reading frame encoding a 268 amino acid protein. Analysis shows that CpSL gene has strong homology with other legume lectin genes. Phylogenetic analysis showed the existence of two main clusters and clearly indicated that CpSL belonged to mannose-specific family of lectins. RT-PCR revealed that CAA gene expressed constitutively in various plant tissues including flower, leaf, root and stem. When chickpea lectin mRNA level was checked in developing seeds, it was higher in 10 DAF seeds and decreased throughout seed development.  相似文献

Chickpea is a staple protein source in many Asian and Middle Eastern countries. The seeds contain carotenoids such as beta-carotene, cryptoxanthin, lutein and zeaxanthin in amounts above the engineered beta-carotene-containing golden rice level. Thus, breeding for high carotenoid concentration in seeds is of nutritional, socio-economic, and economic importance. To study the genetics governing seed carotenoids in chickpea, we studied the relationship between seed weight and concentrations of beta-carotene and lutein by means of high-performance liquid chromatography in segregating progeny from a cross between an Israeli cultivar and wild Cicer reticulatum Ladiz. Seeds of the cross progeny varied with respect to their carotenoid concentration (heritability estimates ranged from 0.5 to 0.9), and a negative genetic correlation was found between mean seed weight and carotenoid concentration in the F3. To determine the loci responsible for the genetic variation observed, the population was genotyped using 91 sequence tagged microsatellite site markers and two CytP450 markers to generate a genetic map consisting of nine linkage groups and a total length of 344.6 cM. Using quantitative data collected for beta-carotene and lutein concentration and seed weight of the seeds of the F2 population, we were able to identify quantitative trait loci (QTLs) by interval mapping. At a LOD score of 2, four QTLs for beta-carotene concentration, a single QTL for lutein concentration and three QTLs for seed weight were detected. The results of this investigation may assist in improving the nutritional quality of chickpea.  相似文献

An acid phosphatase (EC 3.1.3.2.) from the embryonic axes of chickpea seeds ( Cicer arietinum L. cv. Castellana) was purified by ammonium sulphate precipitation, chromatography on Sephacryl S-200 and polyacrylamide gel electrophoresis. The preparation has an apparent molecular weight of 39 kDa, pH optimum for p -nitrophenylphosphate hydrolysis of 5.25, and K m of 0.57 m M . The enzyme hydrolyzed all the mono- and di-phosphorylated sugars tested, but had no effect on ATP, ADP, AMP and phosphoenolpyruvate. Phosphate was a competitive inhibitor. Mg2+. Ca2+, Hg2+, Fe3+, arsenate, K+ and Zn2+ were inhibitory. Mn2+, dithiothreitol and EDTA had no effect, and polyamines were activators.  相似文献

High performance liquid chromatography analysis of different parts of Sclerotium rolfsii-infected and healthy seedlings of chickpea (Cicer arietinum) was carried out to examine the status of phenolic compounds. Three major peaks that appeared consistently were identified as gallic, vanillic and ferulic acids. Gallic acid concentrations were increased in the leaves and stems of infected plants compared to healthy ones. Vanillic acid detected in stems and leaves of healthy seedlings was not detected in infected seedlings. There was a significant increase of ferulic acid in those stem portions located above the infected collar region compared to minimal amounts in the roots of healthy seedlings. In vitro studies of ferulic acid showed significant antifungal activity against S. rolfsii. Complete inhibition of mycelial growth was observed with 1000 g of ferulic acid/ml. Lower concentrations (250, 500 and 750 g/ml) were also inhibitory and colony growth was compact in comparison with the fluffy growth of normal mycelium. Higher amounts of phenolics were found in the stems and leaves of S. rolfsii-infected seedlings in comparison to the healthy ones. A role for ferulic acid in preventing infections by S. rolfsii in the stems and leaves of chickpea plants above the infection zone is therefore feasible.  相似文献

A procedure for the partial purification of a non-specific alkaline phosphatase (EC 3.1.3.1.) from the embryonic axes of chick-pea seeds is described. Ammonium sulphate precipitation, DEAE-cellulase chromatography, Sephacryl S-200 chroma-tography and polyacrylamide gel electrophoresis are the most important steps. The molecular weight of this non-specific enzyme, as determined by Sephacryl S–200 gel filtration and SDS–polyacrylamide gel electrophoresis, was estimated as being 68 and 78 kDa respectively; the optimum pH for p-nitrophenylphosphate hydrolysis was 7.5, and the Km for this artificial substrate was 0.5 mM. The enzyme catalyzes the hydrolysis of a variety of organic phosphate esters. The best substrates are: phos-phoenolpymvate (Km= 2.4 m M ), NADP+ (Km= 4.0 m M ), 5'-AMP (Km= 4.5 m M ), 5'-ADP (Km= 6.1 m M ) and ribose-5P (Km= 5.8 m M ); but it is unable to hydrolyze 5'-ATP, phosphocreatine and tripolyphosptiate. Phospate was a competitive inhibitor. Zn2+, K+, Hg2+ and Mo6+ were strong inhibitors, whereas F and Ca2+ inhibited weakly; Co2+ and Ni2+ were activators.  相似文献

K. KATSABOXAKIS AND K. MALLIDIS. 1996. The microflora of soak water was studied during natural fermentation of coarsely ground chickpea seeds at 32°, 37° and 42°C. The soak water was slightly salted with NaCl 0.5% (w/v) and its proportion to sample was 10 : 1 (v/w). A gas-producing Clostridium species isolated from Reinforced Clostridium Agar (RCA) plates incubated anaerobically, was found to dominate this fermentation system particularly at the higher temperatures. Gram-negative bacteria and yeasts were not found. Bacillus species were isolated from Plate Count Agar (PCA) plates and Lactobacillus, Corynebacterium, Micrococcus and Pediococcus spp. from RCA plates were incubated aerobically. Butyric was the main acid produced at 37° and 42°C and CO2 with hydrogen were identified as main fermentation gases.  相似文献

Arginase (EC 3.5.3.1) activity increased in the cotyledon, while it declined rapidly in the embryonic axis of Bengal gram ( Cicer arietinum L. cv. Desi type) seeds during germination. The decrease in enzyme activity in the embryonic axis was accompanied by changes in the properties of arginase in vitro such as decreased stability, increased heat lability and failure to bind to DEAE-cellulose. These alterations were due to the presence of a low-molecular-weight factor in the extract, which was purified and identified as a hexose derivative. During germination the concentration of the factor increased in the embryonic axis, while no detectable level of the factor was present in the cotyledons. We postulate that the factor may have a role in the regulation of arginase activity in vivo.  相似文献

Changes in calmodulin (CaM) levels in embryonic axes of Cicer arietinum L. cv. Castellana germinated under three different conditions were measured. Abscisic acid (ABA) and a temperature of 30°C, which delay chick-pea germination, respectively decreased and increased the concentration of CaM compared to the values obtained under normal germinating conditions (H2O-25°C). The CaM concentration was higher in those zones of the axes undergoing an active cell division.
The compartmentalization of CaM in 36-h-old embryonic axes grown under these three conditions was also measured. Cytosolic and mitochondrial CaM was higher in axes where a delayed germination occurred as well as in the cell walls of normally grown ones. On the other hand, CaM was higher in nuclear and microsomal fractions extracted from H2-O-25°C-treated axes. From these data we postulate that delayed germination could be an effect of altered CaM distribution in chick-pea embryonic axes.  相似文献
