It is well established that male rats have an advantage in acquiring place-learning strategies, allowing them to learn spatial tasks more readily than female rats. However many of these differences have been examined solely during acquisition or in 24h memory retention. Here, we investigated whether sex differences exist in remote long-term memory, lasting 30d after training, and whether there are differences in the expression pattern of molecular markers associated with long-term memory maintenance. Specifically, we analyzed the expression of protein kinase M zeta (PKMζ) and the α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA) receptor subunit GluA2. To adequately evaluate memory retention, we used a robust training protocol to attenuate sex differences in acquisition and found differential effects in memory retention 1d and 30d after training. Female cohorts tested for memory retention 1d after 60 training trials outperformed males by making significantly fewer reference memory errors at test. In contrast, male cohorts tested 30d after 60 training trials outperformed females of the same condition, making fewer reference memory errors and achieving significantly higher retention test scores. Furthermore, given 60 training trials, females tested 30d later showed significantly worse memory compared to females tested 1d later, while males tested 30d later did not differ from males tested 1d later. Together these data suggest that with robust training males do no retain spatial information as well as females do 24h post-training but maintain this spatial information for longer. Males also showed a significant increase in synaptic PKMζ expression and a positive correlation with retention test scores, while females did not. Interestingly, both sexes showed a positive correlation between retention test scores and synaptic GluA2 expression. Furthermore, the increased expression of synaptic PKMζ, associated with male memory but not with female memory, identifies another potential sex-mediated difference in memory processing.  相似文献

Up-regulation of GluN2B-containing N-methyl-d-aspartate receptors (NMDARs) expression and trafficking is the key mechanism for remifentanil-induced hyperalgesia (RIH), nevertheless, the signaling pathway and pivotal proteins involved in RIH remain equivocal. PKMζ, an isoform of protein kinase C (PKC), maintains pain memory storage in neuropathic pain and inflammatory pain, which plays a parallel role regulated by NMDARs in long-term memory trace. In the present study, Zeta Inhibitory Peptide (ZIP), a PKMζ inhibitor, and a selective GluN2B antagonist Ro-256981 are injected intrathecally before remifentanil infusion (1 μg kg?1 min?1 for 1 h, iv) in order to detect whether GluN2B contributes to RIH through affecting synthesis and activity of PKMζ in spinal dorsal horn. Nociceptive tests are measured by Paw withdrawal mechanical threshold (PWT) and paw withdrawal thermal latency (PWL). The L4–L6 segments of dorsal horn taken from rats with RIH are for determining expression of PKMζ and pPKMζ by Western blot and immunohistochemistry. Our data suggest that remifentanil infusion causes an increase of PKMζ in expression and phosphorylation in rats with nociceptive sensitization, beginning at 2 h, peaked at 2 days, and returned to basal level at 7 days. ZIP (10 ng) could block behavioral sensitization induced by remifentanil. Ro25-6981 dosage-dependently attenuated mechanical and thermal hyperalgesia and reversed expression of PKMζ and pPKMζ, indicating that GluN2B-containing NMDA receptor facilitates development of RIH through mediating expression and activity of spinal PKMζ in rats. Although detailed mechanisms require further comprehensive study, the preventive role of Ro25-6981 and ZIP provide novel options for the effective precaution of RIH in clinics.  相似文献

The persistent activity of protein kinase Mzeta (PKMζ), a brain-specific, constitutively active protein kinase C isoform, maintains synaptic long-term potentiation (LTP). Structural remodeling of the postsynaptic density is believed to contribute to the expression of LTP. We therefore examined the role of PKMζ in reconfiguring PSD-95, the major postsynaptic scaffolding protein at excitatory synapses. In primary cultures of hippocampal neurons, PKMζ activity was critical for increasing the size of PSD-95 clusters during chemical LTP (cLTP). Increasing PKMζ activity by overexpressing the kinase in hippocampal neurons was sufficient to increase PSD-95 cluster size, spine size, and postsynaptic AMPAR subunit GluA2. Overexpression of an inactive mutant of PKMζ did not increase PSD-95 clustering, and applications of the ζ-pseudosubstrate inhibitor ZIP reversed the PKMζ-mediated increases in PSD-95 clustering, indicating that the activity of PKMζ is necessary to induce and maintain the increased size of PSD-95 clusters. Thus the persistent activity of PKMζ is both necessary and sufficient for maintaining increases of PSD-95 clusters, providing a unified mechanism for long-term functional and structural modifications of synapses.  相似文献

A leading candidate in the process of memory formation is hippocampal long-term potentiation (LTP), a persistent enhancement in synaptic strength evoked by the repetitive activation of excitatory synapses, either by experimental high-frequency stimulation (HFS) or, as recently shown, during actual learning. But are the molecular mechanisms for maintaining synaptic potentiation induced by HFS and by experience the same? Protein kinase Mzeta (PKMζ), an autonomously active atypical protein kinase C isoform, plays a key role in the maintenance of LTP induced by tetanic stimulation and the storage of long-term memory. To test whether the persistent action of PKMζ is necessary for the maintenance of synaptic potentiation induced after learning, the effects of ZIP (zeta inhibitory peptide), a PKMζ inhibitor, on eyeblink-conditioned mice were studied. PKMζ inhibition in the hippocampus disrupted both the correct retrieval of conditioned responses (CRs) and the experience-dependent persistent increase in synaptic strength observed at CA3-CA1 synapses. In addition, the effects of ZIP on the same associative test were examined when tetanic LTP was induced at the hippocampal CA3-CA1 synapse before conditioning. In this case, PKMζ inhibition both reversed tetanic LTP and prevented the expected LTP-mediated deleterious effects on eyeblink conditioning. Thus, PKMζ inhibition in the CA1 area is able to reverse both the expression of trace eyeblink conditioned memories and the underlying changes in CA3-CA1 synaptic strength, as well as the anterograde effects of LTP on associative learning.  相似文献

Background

This study assessed the long-term safety and efficacy of tiotropium Respimat, a long-acting inhaled anticholinergic bronchodilator, in asthma, added on to inhaled corticosteroids (ICS) with or without long-acting β2-agonist (LABA).

Methods

285 patients with symptomatic asthma, despite treatment with ICS±LABA, were randomised 2:2:1 to once-daily tiotropium 5 μg, tiotropium 2.5 μg or placebo for 52 weeks (via the Respimat SoftMist inhaler) added on to ICS±LABA, in a double-blind, placebo-controlled, parallel-group study (NCT01340209). Primary objective: to describe the long-term safety profile of tiotropium. Secondary end points included: trough forced expiratory volume in 1 second (FEV1) response; peak expiratory flow rate (PEFR) response; seven-question Asthma Control Questionnaire (ACQ-7) score.

Results

At Week 52, adverse-event (AE) rates with tiotropium 5 μg, 2.5 μg and placebo were 88.6%, 86.8% and 89.5%, respectively. Commonly reported AEs with tiotropium 5 μg, 2.5 μg and placebo were nasopharyngitis (48.2%, 44.7%, 42.1%), asthma (28.9%, 29.8%, 38.6%), decreased PEFR (15.8%, 7.9%, 21.1%), bronchitis (9.6%, 13.2%, 7.0%), pharyngitis (7.9%, 13.2%, 3.5%) and gastroenteritis (10.5%, 3.5%, 5.3%). In the tiotropium 5 μg, 2.5 μg and placebo groups, 8.8%, 5.3% and 5.3% of patients reported drug-related AEs; 3.5%, 3.5% and 15.8% reported serious AEs. Asthma worsening was the only serious AE reported in more than one patient. At Week 52, adjusted mean trough FEV1 and trough PEFR responses were significantly higher with tiotropium 5 μg (but not 2.5 μg) versus placebo. ACQ-7 responder rates were higher with tiotropium 5 μg and 2.5 μg versus placebo at Week 24.

Conclusions

The long-term tiotropium Respimat safety profile was comparable with that of placebo Respimat, and associated with mild to moderate, non-serious AEs in patients with symptomatic asthma despite ICS±LABA therapy. Compared with placebo, tiotropium 5 μg, but not 2.5 μg, significantly improved lung function and symptoms, supporting the long-term efficacy of the 5 μg dose.

A model is presented that shows that reduced fertility in humans can be explained as part of an evolved strategy to maximize long-term fitness in the face of periodic calamities that result in demographic crashes. Three conditions must be met for this model to be plausible: (1) human population history has been characterized by local periods of growth punctuated by recurrent crashes caused by calamities such as climatically induced resource shortfalls; (2) a strategy is available to individuals that increases the probability of survival through a crash, but that, to implement, requires diverting resources away from producing more offspring; and (3) long-term fitness benefits to increased survivorship through a crisis must outweigh or equal the fitness benefits that would accrue to putting the same resources into higher fertility. We present a model that shows that increases in survivorship can outweigh the benefits of higher fertility even if crises are neither very frequent nor particularly severe.  相似文献

β1 integrin regulates multiple epithelial cell functions by connecting cells with the extracellular matrix (ECM). While β1 integrin-mediated signaling in murine epithelial stem cells is well-studied, its role in human adult epithelial progenitor cells (ePCs) in situ remains to be defined. Using microdissected, organ-cultured human scalp hair follicles (HFs) as a clinically relevant model for studying human ePCs within their natural topobiological habitat, β1 integrin-mediated signaling in ePC biology was explored by β1 integrin siRNA silencing, specific β1 integrin-binding antibodies and pharmacological inhibition of integrin-linked kinase (ILK), a key component of the integrin-induced signaling cascade. β1 integrin knock down reduced keratin 15 (K15) expression as well as the proliferation of outer root sheath keratinocytes (ORSKs). Embedding of HF epithelium into an ECM rich in β1 integrin ligands that mimic the HF mesenchyme significantly enhanced proliferation and migration of ORSKs, while K15 and CD200 gene and protein expression were inhibited. Employing ECM-embedded β1 integrin-activating or -inhibiting antibodies allowed to identify functionally distinct human ePC subpopulations in different compartments of the HF epithelium. The β1 integrin-inhibitory antibody reduced β1 integrin expression in situ and selectively enhanced proliferation of bulge ePCs, while the β1 integrin-stimulating antibody decreased hair matrix keratinocyte apoptosis and enhanced transferrin receptor (CD71) immunoreactivity, a marker of transit amplifying cells, but did not affect bulge ePC proliferation. That the putative ILK inhibitor QLT0267 significantly reduced ORSK migration and proliferation and induced massive ORSK apoptosis suggests a key role for ILK in mediating the ß1 integrin effects. Taken together, these findings demonstrate that ePCs in human HFs require β1 integrin-mediated signaling for survival, adhesion, and migration, and that different human HF ePC subpopulations differ in their response to β1 integrin signaling. These insights may be exploited for cell-based regenerative medicine strategies that employ human HF-derived ePCs.  相似文献

Background

Previous analyses from a randomized trial in women aged 24–45 have shown the quadrivalent HPV vaccine to be efficacious in the prevention of infection, cervical intraepithelial neoplasia (CIN) and external genital lesions (EGL) related to HPV 6/11/16/18 through 4 years. In this report we present long term follow-up data on the efficacy, safety and immunogenicity of the quadrivalent HPV vaccine in adult women.

Methods

Follow-up data are from a study being conducted in 5 sites in Colombia designed to evaluate the long-term immunogenicity, effectiveness, and safety of the qHPV vaccine in women who were vaccinated at 24 to 45 years of age (in the original vaccine group during the base study [n = 684]) or 29 to 50 years of age (in the original placebo group during the base study [n = 651]). This analysis summarizes data collected as of the year 6 post-vaccination visit relative to day 1 of the base study (median follow-up of 6.26 years) from both the original base study and the Colombian follow-up.

Results

There were no cases of HPV 6/11/16/18-related CIN or EGL during the extended follow-up phase in the per-protocol population. Immunogenicity persists against vaccine-related HPV types, and no evidence of HPV type replacement has been observed. No new serious adverse experiences have been reported.

Conclusions

Vaccination with qHPV vaccine provides generally safe and effective protection from HPV 6-, 11-, 16-, and 18-related genital warts and cervical dysplasia through 6 years following administration to 24–45 year-old women.

We test a strategy for analyzing species richness in a landscape. This strategy is based on the joint analysis of (local), (turnover) and (landscape) diversities. We assessed the spatial and temporal relations among , and diversity of bats (Phyllostomidae and Mormoopidae) in a tropical landscape. In a spatial dimension, diversity depends on the diversity of the most species-rich community. The value of spatial diversity between habitats was very low. A high diversity was found in a cornfield, which may be attributed to the reduced extent of the field (compared with a more extensive field) that allows the arrival of individuals from nearby rich communities. In a temporal dimension, within habitat cumulative richness over sampling period may be considerably different from the average richness. These differences are attributed to temporal turnover during short time intervals. Therefore, cumulative richness may be viewed as the temporal equivalent of within-habitat diversity, which results of both average and temporal turnover. We discuss, which value must be taken as an estimate of habitat species richness, the average or the cumulative , and the implications that this decision can have in the evaluation of biodiversity.  相似文献

Understanding of population dynamics in a fragmented habitat is an issue of considerable importance. A natural modelling framework for these systems is spatially discrete. In this paper, we consider a predator–prey system that is discrete both in space and time, and is described by a Coupled Map Lattice (CML). The prey growth is assumed to be affected by a weak Allee effect and the predator dynamics includes intra-specific competition. We first reveal the bifurcation structure of the corresponding non-spatial system. We then obtain the conditions of diffusive instability on the lattice. In order to reveal the properties of the emerging patterns, we perform extensive numerical simulations. We pay a special attention to the system properties in a vicinity of the Turing–Hopf bifurcation, which is widely regarded as a mechanism of pattern formation and spatiotemporal chaos in space-continuous systems. Counter-intuitively, we obtain that the spatial patterns arising in the CML are more typically stationary, even when the local dynamics is oscillatory. We also obtain that, for some parameter values, the system’s dynamics is dominated by long-term transients, so that the asymptotical stationary pattern arises as a sudden transition between two different patterns. Finally, we argue that our findings may have important ecological implications.  相似文献

Summary Simulation models have recently been used to suggest that spatial heterogeneity, acting on small spatial scales within local populations, may allow parasitoids and other natural enemies to regulate host or prey populations in ways that would not be detected by conventional (k-factor) analyses of life table data. However, additional study of these models suggests that local extinction may be a frequent event in the simulated interactions. The spreading of risk concept appears more applicable to the simulated populations than a classical view emphasizing tight regulation around stable equilibrium points. The spreading of risk viewpoint also appears to shed additional light on questions raised in the recent debate between Dempster (1983); Hassel (1985); Dempster and Pollard (1986), concerning the modeling of spatial heterogeneity and regulation in temperate-zone insect populations.  相似文献

Annual influenza B virus epidemics and outbreaks cause severe influenza diseases in humans and pose a threat to public health. China is an important epidemic area of influenza B viruses. However, the spatial, temporal transmission pathways and the demography history of influenza B viruses in China remain unknown. We collected the haemagglutinin gene sequences sampled of influenza B virus in China between 1973 and 2018. A Bayesian Markov chain Monte Carlo phylogeographic discrete approach was used to infer the spatial and temporal phylodynamics of influenza B virus. The Bayesian phylogeographic analysis of influenza B viruses showed that the North subtropical and South subtropical zones are the origins of the Victoria and Yamagata lineage viruses, respectively. Furthermore, the South temperate and North subtropical zones acted as transition nodes in the Victoria lineage virus dispersion network and that the North subtropical and Mid subtropical zones acted as transition nodes in the Yamagata lineage virus dispersion network. Our findings contribute to the knowledge regarding the spatial and temporal patterns of influenza B virus outbreaks in China.  相似文献

Aims

Transforming growth factor-β (TGF-β), fascin, nuclear factor-kappa B (NF-κB) p105, protein-kinase C-zeta (PKC-ζ), partioning-defective protein-6 (Par-6), E-cadherin and vimentin are tumor promoting molecules through mechanisms involved in cell dedifferentiation. In soft tissue sarcomas, their expression profile is poorly defined and their significance is uncertain. We aimed to investigate the prognostic impact of TGF-β1, NF-κB p105, PKC-ζ, Par-6α, E-cadherin and vimentin in non-gastrointestinal stromal tumor soft tissue sarcomas (non-GIST STSs).

Patients and Methods

Tumor samples and clinical data from 249 patients with non-GIST STS were obtained, and tissue microarrays (TMAs) were constructed for each specimen. Immunohistochemistry (IHC) was used to evaluate marker expression in tumor cells.

Results

In univariate analysis, the expression levels of TGF-β1 (P = 0.016), fascin (P = 0.006), NF-κB p105 (P = 0.022) and PKC-ζ, (P = 0.042) were significant indicators for disease specific survival (DSS). In the multivariate analysis, high TGF-β1 expression was an independent negative prognostic factor for DSS (HR = 1.6, 95% CI = 1.1–2.4, P = 0.019) in addition to tumor depth, malignancy grade, metastasis at diagnosis, surgery and positive resection margins.

Conclusion

Expression of TGF-β1 was significantly associated with aggressive behavior and shorter DSS in non-GIST STSs.  相似文献

Spatial and temporal variability in the concentration of saprophytic bacteria was investigated in surface waters of Paranaguá and Antonina bays.Seven points along a profile from the entrance of Paranaguá Bay to the innermost part of Antonina Bay, were sampled monthly from November 1985 to November 1986. Temperature, dissolved oxygen, pH and precipitation data were also measured.ZoBell 2216E culture medium was utilized with freshwater and 32% salinity water.Principal component analysis of biotic and abiotic data showed a gradient from the innermost bay to the baymouth bar with an increase in bacteria concentration at Antonina Bay. Significant variation was not observed among sampling stations at Paranaguá Bay, except for February 1986 when there was a homogeneous increase of bacteria throughout both bays in association with an increase in temperature and precipitation. There was also no strong variation of saprophyte numbers at sampling locations near Paranaguá city, despite domestic and harbour waste input.We conclude that the outer region of the estuary is highly influenced by the adjacent ocean and that the inner part has typical estuarine characteristics. The fish biodiversity of Lake Nokoué, a lagoon in Bénin, was monitored for one year from August 2000 to July 2001 by means of samples from the artisanal fishery collected once a month at three stations. A total of 5 933 fish weighing 95 322g (mean individual weight 16.0g) belonging to 51 species, 47 genera and 34 families were sampled. An abundance of juveniles was observed, confirming the importance of the lake as nursery grounds, rich in exploitable habitats like other tropical estuaries and lagoons. The fish fauna comprised three main components: i) an estuarine component (strict, freshwater and marine origins) which is the basis of the lagoon fauna community and which is stable in space and time; ii) a marine component containing fishes which appeared in the lagoon during the dry season where the salinity remains high (>20‰) and iii) a freshwater component with fishes coming into the lagoon from the river during the flood period. The magnitude of the spatial and seasonal fluctuations of the last two components varies depending on the area: marine (Zogbo), freshwater (Vêki) or intermediate (Ganvié). The global species richness and the relative abundances of fish vary between stations and from month to month. November, with 20 species on average, and January, with 6 species on average, are significantly different as regards species richness.  相似文献

The pollen tube is a cellular protuberance formed by the pollen grain, or male gametophyte, in flowering plants. Its principal metabolic activity is the synthesis and assembly of cell wall material, which must be precisely coordinated to sustain the characteristic rapid growth rate and to ensure geometrically correct and efficient cellular morphogenesis. Unlike other model species, the cell wall of the Arabidopsis (Arabidopsis thaliana) pollen tube has not been described in detail. We used immunohistochemistry and quantitative image analysis to provide a detailed profile of the spatial distribution of the major cell wall polymers composing the Arabidopsis pollen tube cell wall. Comparison with predictions made by a mechanical model for pollen tube growth revealed the importance of pectin deesterification in determining the cell diameter. Scanning electron microscopy demonstrated that cellulose microfibrils are oriented in near longitudinal orientation in the Arabidopsis pollen tube cell wall, consistent with a linear arrangement of cellulose synthase CESA6 in the plasma membrane. The cellulose label was also found inside cytoplasmic vesicles and might originate from an early activation of cellulose synthases prior to their insertion into the plasma membrane or from recycling of short cellulose polymers by endocytosis. A series of strategic enzymatic treatments also suggests that pectins, cellulose, and callose are highly cross linked to each other.Upon contact with the stigma, the pollen grain swells through water uptake and develops a cellular protrusion, the pollen tube. During its growth in planta, the pollen tube invades the transmitting tissue of the pistil and finds its way to the ovary to deliver the male gametes for double fertilization to happen (Heslop-Harrison, 1987). Depending on the species, pollen tubes can grow extremely rapidly both in planta and in in vitro conditions. To fulfill its biological function, the pollen tube has to (1) adhere to and invade transmitting tissues (Hill and Lord, 1987; Lennon et al., 1998), (2) provide physical protection to the sperm cells, and (3) control its own shape and invasive behavior (Parre and Geitmann, 2005b; Geitmann and Steer, 2006). For all of these functions, the pollen tube cell wall plays an important regulatory and structural role. Although the pollen tube does not form a conventional secondary cell wall layer, its wall is assembled in two phases. The “primary layer” is mainly formed of pectins and other matrix components secreted at the apical end of the cell. The “secondary layer” is assembled by the deposition of callose in more distal regions of the cell (Heslop-Harrison, 1987). Depending on the species, cellulose microfibrils have been found to be associated either with the outer pectic or with the inner callosic layer. Unlike most other plant cells, cellulose is not very abundant representing only 10% of total neutral polysaccharides in Nicotiana alata pollen tubes, whereas callose accounts for more than 80% in this species (Schlüpmann et al., 1994).The biochemical composition of the pollen tube cell wall has been well characterized in many species such as Lilium longiflorum (Lancelle and Hepler, 1992; Jauh and Lord, 1996), tobacco (Nicotiana tabacum; Kroh and Knuiman, 1982; Geitmann et al., 1995; Ferguson et al., 1998; Derksen et al., 2011), Petunia hybrida (Derksen et al., 1999), Pinus sylvestris (Derksen et al., 1999), and Solanum chacoense (Parre and Geitmann, 2005a). But for Arabidopsis (Arabidopsis thaliana), the model for plant molecular biology studies (Arabidopsis Genome Initiative, 2000), there is a striking lack of quantitative information concerning the composition of the pollen tube cell wall as well as the spatial distribution of its components. This is all the more surprising because numerous mutants defective in enzymes involved in cell wall synthesis exhibit a pollen tube phenotype (for example, Jiang et al., 2005; Nishikawa et al., 2005; Wang et al., 2011). Two studies have characterized the Arabidopsis pollen germinating in vitro (Derksen et al., 2002) and in vivo (Lennon and Lord, 2000), but both are qualitative rather than quantitative. A biochemical study by Dardelle and coworkers investigated the cell wall sugar composition in a more quantitative way but does not provide any detailed spatial information (Dardelle et al., 2010; Lehner et al., 2010). This lack of information is not surprising given that until recently Arabidopsis pollen was known to be rather challenging to germinate reproducibly in vitro and more difficult to manipulate than the pollen of many other plant species (Bou Daher et al., 2009). With the publication of optimized methods for in vitro germination (Boavida and McCormick, 2007; Bou Daher et al., 2009), it has become much more feasible to germinate healthy-looking Arabidopsis pollen tubes in vitro in a highly reproducible way.The precisely controlled spatial distribution of biochemical components in the pollen tube cell wall is crucial for shape generation and maintenance of this perfectly cylindrical cell (Geitmann and Parre, 2004; Aouar et al., 2010; Fayant et al., 2010; Geitmann, 2010). The pollen tube, therefore, represents an ideal model system to study the link between intracellular signaling, biochemistry, cell mechanical properties, and morphogenesis in plant cells. Because of its typically fast growth rates, it responds quickly to any environmental triggers such as pharmacological, hormone, or enzymatic treatments. Adding Arabidopsis to the group of commonly studied pollen tube species is particularly timely, because one-third of the approximately 800 cell wall synthesis genes identified in this species are expressed in or are specific to its pollen (Pina et al., 2005). Therefore, the Arabidopsis pollen tube has become a valuable system for cell wall studies, especially with the increasing availability of cell wall mutant lines (Liepman et al., 2010).Here we describe the biochemical composition of the Arabidopsis pollen tube cell wall grown in in vitro conditions using immunocytochemical labeling coupled with epifluorescence and electron microscopic techniques. Rather than relying on imaging alone, we developed a quantitative strategy to assess the precise spatial distribution of cell wall components. This quantitative approach will provide an important tool and baseline dataset for the investigation of mutant phenotypes and for the interpretation of pharmacological studies. Furthermore, we used selective and strategically combined enzymatic digestions to determine the degree of connectivity between the individual types of cell wall polysaccharide networks.  相似文献

Matrix metalloproteinases (MMPs) are a family of extracellular proteases capable of degrading various proteinaceous components of the extracellular matrix(ECM).They have been implicated to play important roles in a number of developmental and pathological processes,such as tumor metastasis and inflammation.Relatively few studies have been carried out to investigate the function of MMPs during postembryonic organ-development.Using Xenopus laevis development as a model system,we demonstrate here that three MMPs,stromelysin-3(ST3),collagenases-3(Col3),and Col4,have distinct spatial and temporal expression profiles during metamorphosis as the tadpole transforms into a frog.In situ hybridizations reveal a tight,but distinct,association of individual MMPs with tissue remodeling in the tail and intestine during metamorphosis.In particular,ST3 expression is strongly correlated with apoptosis in both organs as demonstrated by analyses of serial sections with in situ hybridization for ST3 mRNA and TUNEL (terminal deoxyribonucleotidyl transferase-mediated dUTP-biotin nick end labeling)for apoptosis,respectively.On the other hand,Col3 and Col4 are present in regions where extensive connective tissue remodeling take place.These results indicate that ST3 is likely to play a role in ECM-remodeling that facilitate apoptotic tissue remodeling or resorption while Col3 and Col4 appear to participate in connective tissue degradation during development.  相似文献

Changjiang Estuary, the largest estuary in China, encompasses a wide range of nutrient loading and trophic levels from the rivers to the sea, providing an ideal natural environment to explore relationships between functional diversity, physical/chemical complexity, and ecosystem function. In this study, molecular biological techniques were used to analyze the community structure and diversity of ammonia-oxidizing bacteria (AOB) in the sediments of Changjiang Estuary and its adjacent waters in East China Sea. The amoA gene (encoding ammonia monooxygenase subunit A) libraries analysis revealed extensive diversity within the β-Proteobacteria group of AOB, which were grouped into Nitrosospira-like and Nitrosomonas-like lineages. The majority of amoA gene sequences fell within Nitrosospira-like clade, and only a few sequences were clustered with the Nitrosomonas-like clade, indicating that Nitrosospira-like lineage may be more adaptable than Nitrosomonas-like lineage in this area. Multivariate statistical analysis indicated that the spatial distribution of the sedimentary β-Proteobacterial amoA genotype assemblages correlated significantly with nitrate, nitrite, and salinity. The vertical profile of amoA gene copies in gravity cores showed that intense sediment resuspension led to a deeper mixing layer. The horizontal distribution pattern of amoA gene copies was nearly correlated with the clayey mud belt in Changjiang Estuary and its adjacent area in East China Sea, where higher β-Proteobacteria phylogenetic diversity was observed. Meanwhile, those areas with high amoA copies in the surface sediments nearly matched those with low concentrations of dissolved oxygen and ammonium in the bottom water.  相似文献
