Chromosome number determinations from 118 populations of eight species of the aster genus Virgulus are reported for the first time. Based on these counts and literature reports the geographic distributions of diploids and tetraploids in V. concolor, V. novae-angliae, V. oblongifolius, and V. patens have been determined. In the first three species, diploids were widely distributed, while tetraploids were more restricted. In V. patens, diploids were restricted to the southwestern portion of the species’ range, while tetraploids were found throughout the range. Both V. adnatus and V. walteri were found only at the tetraploid level throughout their ranges. Two other species with restricted distributions were consistently found to be high level polyploids: V. georgianus with 2n = 50, V. grandiflorus with 2n = 60. The following new combinations are listed: Virgulus georgianus (Alexander ex Small) Semple; Virgulus subsect. Brachyphylli (Torr. & Gray) Semple; and Virgulus sect. Riassunto Lo studio comprende una revisione critico-sperimentale della specie Actinomyces albus, della quale vengono considerati come sinonimi circa 30 nomi speeifici, fra i quale A. chromogenus, A. odorifer, A. thermophylus p.p.; della specie é data una diagnosi ed una particolareggiata descrizione.Sono inoltre studiate le specie A. viridis, (= A. viridochromogenus) e A. innominatus, n. nomen. Quest'ultima é preceduta da una breve discussione sulla specie A. homini.
Summary Twenty-six strains of Actinomyces albus are studied redescribed from morphological, cultural and biochemical standpoints. Many biological activities of A. chromogenus, A. odorifer and A. thermophilus are in common with other species of the same genus, so that they may be considered for sub-specific, (not specific) differentiation. A discussion on A. farcinicus, A. albidoflavus and A. aureus has been originated from mislabeling as A. albus; the group including the two last named species (flavus group) must be revised. A few strains classified A. farcinicus are in no doubt true A. albus, but this real specific entity remains to be revised from Nocard's strain. A. viridis, for the first time described by Lombardo-Pellegrino, has been redescribed three times as a new species under the same binomial, and the fourth as A. viridochromogenes. A. hominis Bostroem is an uncorrect determination for the species originally described by Waksmann and Curtiss, and it is renamed A. innominatus, the binomial A. (Streptothrix) hominis Auct. being a nomen ambiguum. In conclusion, 30 bionmial are appended in sinonimy to A. albus, including Cladothrix dichotoma Macé (1888) non Cohn, G. invulnerabilis Acosta et G. Rossi, C. odorifera Rullm. Actinomyces chromogenus Gasp., A. thermophilus Auct., p.p., A. (Streptothrix) Sanninii (Cif.) Westerd., A. Almquisti Duché, A. Gougeroti Duché, and so on.

Some physical, catalytic, and regulatory properties of ketopantoate hydroxymethyltransferase (5,10-methylenetetrahydrofolate: alpha-ketoisovalerate hydroxymethyltranferase) from Escherichia coli are described. This enzyme catalyzes the reversible synthesis of ketopantoate (Reaction 1), an essential precursor of pantothenic acid. (1) HC(CH3)2COCOO- + 5,10-methylene tetrahydrofolate f in equilibrium r HOCH2C(CH3)2COCOO- + tetrahydrofolate It has a molecular weight by sedimentation equilibrium of 255,000, a sedimentation coefficient (S20,w) of 11 S, a partial specific volume of 0.74 ml/g, an isoelectric point of 4.4, and an absorbance, (see article), of 0.85. Polyacrylamide gel electrophoresis in sodium dodecyl sulfate and amino acid analyses give a subunit molecular weight of 27,000 and 25,700, respectively; both procedures indicate the presence of 10 identical subunits. The NH2-terminal sequence is Met-Tyr---. The enzyme is stable and active over a broad pH range, with an optimum from 7.0 to 7.6. It requires Mg2+ for activity; Mn2+, Co2+, Zn2+ are progressively less active. The enzyme is not inactivated by borohydride reduction in the presence of excess substrates, i.e. it is a Class II aldolase. Reaction 1f is partially inhibited by concentrations of formaldehyde (0.8 mM) and tetrahydrofolate (0.38 mM) below or near the Km values, apparent Km values are 0.18, 1.1 and 5.9 mM for tetrahydrofolate, alpha-ketoisovalerate, and formaldehyde, respectively. For Reaction 1r, apparent Km values are 0.16 and 0.18 mM, respectively, for ketopantoate and tetrahydrofolate, and the saturation curves for both substrates show positive cooperativity. Forward and reverse reactions occur at similar maximum velocities (Vmax approximately equal to 8 mumol of ketopantoate formed or decomposed per min per mg of enzyme at 37 degrees). Only 1-tetrahydrofolate is active in Reaction 1; d-tetrahydrofolate, folate, and methotrexate were neither active nor inhibitory. However, 1-tetrahydrofolate was effectively replaced with conjugates containing 1 to 6 additional glutamate residues; of these, tetrahydropterolpenta-, tetra-, and triglutamate were effective at lower concentrations than tetrahydrofolate itself; they were also the predominant conjugates of tetrahydrofolate present in E. coli. Alpha-Ketobutyrate, alpha-ketovalerate, and alpha-keto-beta-methylvalerate replaced alpha-ketoisovalerate as substrates; pyruvate was inactive as a substrate, but like isovalerate, 3-methyl-2-butanone and D- or L-valine, inhibited Reaction 1. the transferase has regulatory properties expected of an enzyme catalyzing the first committed step in a biosynthetic pathway. Pantoate (greater than or equal to 500 muM) and coenzyme A (above 1 mM) all inhibit; the Vmax is decreased, Km is increased, and the cooperativity for substrate (ketopantoate) is enhanced. Catalytic activity of the transferase is thus regulated by the products of the reaction path of which it is one component; transferase synthesis is not repressed by growth in the presence of pantothenate.  相似文献

Fine hair-like appendages on the cell walls of the another smut Ustilago violacea are described. These hairs are termed fimbriae because of their close similarity to the fimbriae (pili) found on certain Gram-negative bacteria. Cells of U. violacea may carry more than 200 fimbriae varying in length from about 0.5 mum to over 10 mum, and having a diameter of about 60-70 A. Some fimbriae produce knobs similar to those found on bacterial sex fimbriae. Log-phase cells are the most densely fimbriated, while stationary phase cells are devoid of fimbriae. The cells can be defimbriated by sonication, high-speed agitation, or centrifugation through a 40% sucrose solution. The fimbriae can regenerate in these defimbriated cells in about 1 h. This regeneration is inhibited by both cycloheximide and rifampin, but not by chloramphenicol and therefore appears to depend on de novo protein synthesis on cytoplasmic ribosomes. Similar long fimbriae are found on U. maydis and Leucosporidium (Candida) scottii. Short fimbriae, about 0.5 mum long, were found on all the other species of yeast-like fungi examined (Rhodotorula, Saccharomyces, Schizosaccharomyces, Hansenula, Lipomyces, Nadsonia, and Torulopsis spp.).  相似文献

A comprehensive mechanism is proposed for the photolysis of transition metal complexes in equilibrium solutions. This involves the photo-oxidation, a possibility of energy transfer and several secondary thermal reactions, e.g. forward and back reactions with photochemically-formed H· radicals. The scheme includes the individual quantum yield (IQY) of each species. The photo-oxidations of FeSO4, FeClO4, FeCl2 in aqueous acidic solutions were studied by continuous irradiation. The IQY of Feaq2+ (0.2), FeSo4 (0.3), FeHSO4+ (0.67), FeClO4+ (0.06–0.2), FeCl+ (0.8), FeCl2 (0.3) were determined. The IQY was found to be dependent on H+ concentration, due to competition with geminal recombination within the solvent cage. This dependence, and the efficiency of energy transfer between excited iron(III) species and iron(II) in ground state, are discussed.  相似文献

As part of an ongoing investigation of human mast cell heterogeneity, we have isolated, partially purified, and characterized the uterine mast cell and compared it with mast cells isolated from other organs. The average histamine content of myometrium and leiomyofibroma obtained from hysterectomies was 2.1 +/- 0.3 (mean +/- SEM) microgram/g of tissue (n = 10), and the histamine content of the two tissues did not differ significantly. A mild collagenase, hyaluronidase, and DNase digestion was used to disperse the uterine mast cells, with an average yield of 9.5% (range, 0 to 21%). The average histamine/uterine mast cell was 2.1 +/- 0.2 pg (n = 3), and 61 +/- 7% (n= 3) of the uterine mast cells survived overnight culture. Early purification efforts with Percoll gradients have yielded up to 80% pure uterine mast cells, with an average of 27 +/- 10% (n = 5). Uterine mast cells released histamine in response to the secretogogues anti-IgE and A23187 but did not respond to substance P or to the basophil secretogogues FMLP, C5a, and 12-O-tetradecanoylphorbol-13-acetate. After 1 microgram/ml anti-IgE stimulation, the uterine mast cell appeared to make significant quantities of PGD2 (89 +/- 26 ng/10(6) cells, n = 6) (p less than 0.05), as assayed by RIA. Simultaneously, leukotriene C4 release was 45 +/- 15 ng/10(6) cells, (n = 6) (p less than 0.05), as assayed by RIA. Combined gas-chromatography mass spectroscopy analysis of anti-IgE-stimulated cell supernatants confirmed the production of PGD2. In pharmacologic studies, isobutyl-methylxanthine and isoproterenol blocked anti-IgE-induced histamine release. The uterine mast cell is similar to the lung mast cell in terms of response to secretogogues and release of arachidonic acid metabolites. Ultrastructurally, the uterine mast cell contains scroll granules, crystal granules, combined granules, homogeneously dense granules, and large lipid bodies, many with focal lucencies within them. Particle granules, most frequently present in gut mast cells of mucosal origin, were absent from uterine mast cells. Although certain features are analogous to the ultrastructure of skin or lung mast cells, the combination of structures is distinctive for uterine mast cells.  相似文献

Multiple strains of Campylobacter coli, C. jejuni, C. helveticus, C. lari, C. sputorum, and C. upsaliensis isolated from animal, clinical, or food samples have been analyzed by matrix-assisted laser desorption ionization-time of flight mass spectrometry (MALDI-TOF MS). Whole bacterial cells were harvested from colonies or confluent growth on agar and transferred directly into solvent and then to a spot of dried 3-methoxy-4-hydroxycinnamic acid (matrix). Multiple ions in the 5,000- to 15,000-Da mass range were evident in spectra for each strain; one or two ions in the 9,500- to 11,000-Da range were consistently high intensity. "Species-identifying" biomarker ions (SIBIs) were evident from analyses of multiple reference strains for each of the six species, including the genome strains C. jejuni NCTC 11168 and C. jejuni RM1221. Strains grown on nine different combinations of media and atmospheres yielded SIBI masses within +/-5 Da with external instrument calibration. The highest-intensity C. jejuni SIBIs were cytosolic proteins, including GroES, HU/HCj, and RplL. Multiple intraspecies SIBIs, corresponding probably to nonsynonymous nucleotide polymorphisms, also provided some intraspecies strain differentiation. MALDI-TOF MS analysis of 75 additional Campylobacter strains isolated from humans, poultry, swine, dogs, and cats revealed (i) associations of SIBI type with source, (ii) strains previously speciated incorrectly, and (iii) "strains" composed of more than one species. MALDI-TOF MS provides an accurate, sensitive, and rapid method for identification of multiple Campylobacter species relevant to public health and food safety.  相似文献

Among the genus Populus, the sections Populus (white poplar), Aigeiros Duby (black poplar) and Tacamahaca Spach contain many tree species of economical and ecological important properties. Two parental maps for the inter-specific hybrid population of Populus adenopoda × P. alba (two species of Populus section) were constructed based on SSR and SRAP markers by means of a two-way pseudo-test cross mapping strategy. The same set of SSR markers developed from the P. trichocarpa (belonging to Tacamahaca section) genome which were used to construct the maps of P. deltoides and P. euramericana (two species of Aigeiros section) was chosen to analyze the genotype of the experimental population of P. adenopoda × P. alba. Using the mapped SSR markers as allelic bridges, the alignment of the white and black poplar maps to each other and to the P. trichocarpa physical map was conducted. The alignment showed high degree of marker synteny and colinearity and the closer relationship between Aigeiros and Tacamahaca sections than that of Populus and Tacamahaca. Moreover, there was evidence for the chromosomal duplication and inter-chromosomal reorganization involving some poplar linkage groups, suggesting a complicated course of fission or fusion in one of the lineages. A poplar consensus map based on the comparisons could be constructed will be useful in practical applications including marker assisted selection.  相似文献

Concern about colonization of marshesby plant species such as Phragmites australisand Lythrum salicariahas highlighted the needfor management strategies. However, there is a lack ofinformation in the literature on which to base thesedecisions. This study compares the alpha diversity ofmarshes to assess the impact of invasion by Phragmitesand Lythrum. Species occurrence andstem density were measured in marshes dominated by Phragmites, Lythrum, Typhaspp., or otherherbaceous perennials in the Charles River watershedin eastern Massachusetts, USA, and species richness,Shannon's H, Simpson's reciprocal (1/D), and Pielou'sJ were compared among six community types. The threediversity indices had significantly higher values forTypha-Lythrummarshes than for any of the othermarsh types (Tukey test, p< 0.05), with mean values(± s.d.) of H = 2.00 ± 0.74, 1/D = 3.51± 1.68 and J = 0.69 ± 0.1. Marshes dominatedby Phragmiteshad the lowest diversity, with H= 0 and D = 1, i.e. they were monospecific. Typhadominated marshes had the second lowest values,with H = 0.17 ± 0.05, 1/D = 1.05 ± 0.01, andJ = 0.11 ± 0.03. These results support the ideathat a reduction in diversity can be expected inmarshes colonized by Phragmites. However, thehigh diversity found in the Typha-Lythrummarshes contradicts the expectation of lower diversityafter invasion by Lythrum. This information mayalter marsh management decisions.  相似文献

Human alpha-thrombin, the thromboplastin activation product of prothrombin with high clotting and esterase activity, was produced from Cohn Fraction III paste. The procedure started with 0.4 to 3.2 kg of frozen paste and was completed in 2 or 3 days. Some 23 g of thrombin were recorded for 65 quantitated preparations made from 11 lots of Fraction III paste. These preparations were obtained at protein concentrations of 3.9 +/- 1.3 mg/ml with a yield of 340 +/- 110 mg/kg of paste, which represented 48 +/- 14% of the clotting potential extracted as prothrombin. They had specific clotting activities of 2.8 +/- 0.4 U.S. (NIH) units/microng of protein and titrated to 88 +/- 8% active with p-nitrophenyl-p'-guanidinobenzoate (NPGB). Those (N - 29) examined by labeling with [14C]diisopropyl phosphorofluoridate (iPr2P-F) and electrophoresing in sodium dodecyl sulfate (SDS)-polyacrylamide gels were found to contain only (N = 4) or predominantly alpha-thrombin (97 +/- 3%) and corresponding amounts of ists degradation product, beta-thrombin (2.6 +/- 3.1%). No plasmin(ogen), prothrombin complex factors (II, VII, IX, IXalpha, X, Xalpha), or prothrombin fragments were detected in representative preparations. As produced in 0.75 M NaCl, pH approximately 6, thrombin was stable for approximately 1 week at 4 degrees and for greater than 1 year at less than or equal to 50 degrees; freeze-dried thrombin stored at 4 degrees for greater than 1 year displayed stable clotting activity and no vial to vial variation, permitting its use for reference purposes. Human thrombin generated by Taipan snake venom activation was compared with that produced by rapid thromboplastin activation: after treatment with [14C]iPr2P-F, greater than 95% of the label in both thrombins migrated at the same rate during electrophoresis in SDS; identical pairs of NH2-terminal residues were released in three consecutive Edman degradation cycles.  相似文献

Haplodontium zangii X.R.Wang &; J.C.Zhao, a new moss species from Xizang, China, is described and illustrated. Previously, specimens of H. zangii have been identified as Mielichhoferia himalayana Mitt. However, H. zangii is distinctly different from M. himalayana in having excurrent costae with short awns (vs long denticulate awns), short-pyriform capsules, 0.8–1?mm (vs long-pyriform capsules, 2.5–3?mm), and densely papillose exostome teeth (vs smooth or vertically striped exostome teeth). Haplodontium zangii is similar to H. macrocarpum (Hook.) J.R.Spence, which was traditionally placed in Mielichhoferia Nees &; Hornsch. as M. macrocarpa (Hook.) Bruch &; Schimp. The main differences between H. zangii and H. macrocarpum are in the morphology of the leaves, capsules, guide cells, and stomata. Mielichhoferia himalayana and another Chinese species of Mielichhoferia, M. sinensis Dix., are also transferred to Haplodontium Hampe, a new genus in the bryoflora of China, as H. himalayanum (Mitt.) X.R.Wang &; J.C.Zhao and H. sinensis (Dix.) X.R.Wang &; J.C.Zhao. A morphological comparison and a key to the three species of Haplodontium in China as well as to H. macrocarpum, a species that is likely to be found in China, are provided.  相似文献

An increase in solar ultraviolet-B (UVB) radiation reaching the earth's surface is an important consequence of stratospheric ozone depletion. UVB has important effects on human health, both beneficial and harmful. Recent research has found that solar UVB reduces the risk of over 20 types of cancer, respiratory diseases caused by viruses, autoimmune diseases, and, likely, several other diseases, in addition to the well-known effects on bone diseases. On the other hand, solar UVB is an important risk factor for non-melanoma skin cancer and cataracts. Human epidemiological studies have provided evidence that solar UVA may be a more important risk factor for melanoma than UVB. If this result is correct, melanoma risk is not related to ozone depletion. We consider the net effect of solar UVB on human health to be beneficial at or near current levels.  相似文献

To investigate whether similar physiognomic areas support similar faunas, the richness, similarity, and community structure of the ant fauna attracted to baits in an area of the Brazilian Cerrado sensu stricto at the core of the domain and in two Cerrado 'islands' (one of sensu stricto and another of 'cerradão') were compared. In each locality, four collecting events were performed (two during the day and two at night) capturing ants attracted to 400 sardine baits (200 in soil, 200 in vegetation). Ants of six subfamilies, 33 genera, and 121 species were identified visiting baits in the three areas (180 species estimated). The most speciose genera were Camponotus (27 species), Pheidole (13), Solenopsis (11), and Crematogaster (8). The similarity values between pair sites can be considered medium in relation to other studies. The highest value was recorded for the core area and the 'island' of Cerrado sensu stricto (Morisita-Horn = 0.57). The two Cerrado 'islands' were found to be 0.44 similar and the core area and the 'cerradão' 0.41 similar. The ordination analysis (non-metric multidimensional scaling) revealed discontinuities in species distributions between areas of Cerrado sensu stricto and 'cerradão,' and distinctive ant faunas relative to the temporal (nocturnal and diurnal) and foraging niches (ground and vegetation). The ant fauna at ground level in the 'cerradão' and Cerrado sensu stricto showed the greatest dissimilarity. The data suggest that areas of Cerrado are characterized by relatively high local ant fauna richness, and that community structure is influenced by the distance between sites and the diversity of vegetation.  相似文献
