During fruit development of oil palm (Elaeis guineensis) oil deposition in the mesocarp startedca12–13 weeks after flowering (WAF) and continued until the fruit ripened at 20 WAF. Over the next 1–2 weeks oil continued to be deposited but the fruit became loose and readily detached from the bunch. The lipids extracted at this stage contained over 50 % free fatty acids andca6%, polar lipids. The major fatty acids in the storage triacylglycerols were 16:0,18:1 and 18:2. The fatty acid composition of the neutral lipid classes and polar lipids during oil deposition were similar except that the latter also contained a high proportion of 18:3. Longer chain acids (20:3 and 22:0) were detected in certain lipid classes at 8 and 12 WAF.  相似文献

Abstract

Macaw palm fruits have a diverse biochemical constitution, and there is significant commercial interest in this species among food, pharmaceutical, cosmetics, and bioenergy industries. We evaluated changes in the reserve compounds of macaw palm mesocarp and seeds from fruits stored for 1 year under three different conditions. Protein and carbohydrate levels were highest in the embryo than in the endosperm. Fatty acid profiles were very similar over time under all storage conditions and in each structure evaluated, with the embryo composition being very similar to the mesocarp. Macaw palm oil remained well preserved under all storage conditions tested, but seed reserves and seed viability are best maintained at room temperatures. The endosperm contained higher levels of saturated fatty acids than either the embryo or mesocarp, making seeds more resistant to oxidative deterioration than the mesocarp. The results showed that the composition of the mesocarp oil promises the production of high-quality biodiesel from this structure, and changes in carbohydrate and protein levels show that laboratory conditions are the most efficient for maintaining seed quality during storage.

Protoplasts were enzymatically prepared from the mesocarp of two species of oil palm (Elaeis guineensis Jacq. and E. oleifera HBK and Cortes) 16–20 weeks after anthesis and from rapidly multiplying embryogenic cultures of E. guineensis. The protoplasts were purified by density gradient centrifugation in 20% (w/v) sucrose. Radioactive incorporation studies showed that the protoplasts metabolized [1-14C]acetate to lipids, water-soluble compounds and 14CO2. The [14C]fatty acids obtained consisted mainly of C16: 0, C18: 0 and C18: 1. C16: 1, a very minor fatty acid in palm oil, was also labelled and accounted for 8–39% of total fatty acids synthesized by the mesocarp and embryogenic culture protoplasts. The ratio of labelled C18: 0 to C18: 1 was found to vary with the age of the fruit from which the protoplasts were prepared. Thin layer chromatography (TLC) of the labelled lipids showed the presence of all neutral acylglycerol classes. However the distribution of radiolabel in the various classes differed from those previously reported for oil palm mesocarp [K.C. Oo et al. Lipids, 20 (1985) 205] and embryoid tissue slices [E. Turnham and D.H. Northcote, Phytochem., 23 (1984) 35]. Ozonolysis showed that all the labelled C18: 1 acid was vaccenic acid.  相似文献

以CTAB法提取油棕(Elaeis guineensis)中果皮5个不同发育时期(G1~G5)的小RNA。从前期研究获得的油棕小RNA测序数据库中筛选12个候选miRNA,实时荧光定量PCR法(qRT-PCR)检测其在果实发育过程中的表达量变化,并进一步对显著差异表达的miRNA进行靶基因预测。结果表明:中果皮5个不同发育时期小RNA的OD260/OD280比值在1.7~2.0之间;浓度分别是289、364、476、213、390 ng/μL;qRT-PCR检测结果显示,12个候选miRNA在5个发育时期均显著性差异表达,特别是在中果皮发育第4个时期(G4)和第5个时期(G5)表达量极显著增高,其中miR395和miR156在第4个时期表达量最高;miR395和miR528在发育第5时期表达量最高;靶基因预测结果显示差异表达的部分miRNA,其靶基因可能参与了脂肪酸代谢通路,如磷脂酸磷酸脂酶和磷脂酶D。本研究筛选的与脂肪酸代谢相关的miRNA为今后油棕脂肪酸代谢调控通路研究提供了可能的线索。  相似文献

The details of plant lipid metabolism are relatively well known but the regulation of fatty acid production at the protein level is still not understood. Hence this study explores the importance of phosphorylation as a mechanism to control the activity of fatty acid biosynthetic enzymes using low and high oleic acid mesocarps of oil palm fruit (Elaeis guineensis variety of Tenera). Adaptation of neutral loss-triggered tandem mass spectrometry and selected reaction monitoring to detect the neutral loss of phosphoric acid successfully found several phosphoamino acid-containing peptides. These peptides corresponded to the peptides from acetyl-CoA carboxylase and 3-enoyl-acyl carrier protein reductase as identified by their precursor ion masses. These findings suggest that these enzymes were phosphorylated at 20th week after anthesis. Phosphorylation could have reduce their activities towards the end of fatty acid biosynthesis at ripening stage. Implication of phosphorylation in the regulation of fatty acid biosynthesis at protein level has never been reported.  相似文献

We applied metabolic control analysis to the Kennedy pathway for triacylglycerol formation in tissue cultures from the important oil crops, olive (Olea europaea L.) and oil palm (Elaeis guineensis Jacq.). When microsomal fractions were incubated at 30 degrees C rather than 20 degrees C, there was an increase in triacylglycerol labelling. This increase was accompanied by a build up of diacylglycerol (DAG) radioactivity in olive but not in oil palm, suggesting that the activity of DAG acyltransferase (DAGAT) was becoming limiting in olive. We used 2-bromooctanoate as a specific inhibitor of DAGAT and showed that the enzyme had a flux control coefficient under the experimental conditions of 0.74 in olive but only 0.12 in oil palm. These data revealed important differences in the regulation of lipid biosynthesis in cultures from different plants and suggest that changes in the endogenous activity of DAGAT is unlikely to affect oil accumulation in oil palm crops.  相似文献

The lipid and fatty acid composition of the haustorium of thedeveloping seedling of the West African oil palm, Elaeis guineensisJacq. has been studied using the combined techniques of thin-layerand gas-liquid chromatography. In addition to triglycerides,which represented over 75 per cent of the total lipids, therewere present small quantities of free fatty acids, diglyceridesand polar lipids. The two glycolipids, monogalactosyl and digalactosyldiglycerides, formed the bulk of the polar lipids with smallamounts of phosphatidyl choline and phosphatidyl inositol. In general, the fatty acid pattern of the haustorium was intermediatebetween that of the palm kernel oil and the palm fruit mesocarp,and resembled to a great extent the fatty acids of the kerneltesta. It is suggested, from the presence of the biological membranelipids and lipolytic enzymes, that the main function of thehaustorium is that of food mobilization and transport for theyoung plant.  相似文献

Oil palm fruits exposed to temperatures of 15 °C and belowshowed a significant increase in free fatty acid (FFA) contentin the mesocarp. This effect was most pronounced in fruits exposedto 5 °C when FFA levels exceeding 70% of the total oil wereobserved. The increase in FFA was accompanied by an increasein lipid-soluble phosphorus levels and a decrease in carotenecontent. Palm oil is currently the leading edible oil consumed worldwide. Triacylglycerol (TAG) and diacylglycerol (DAG) are the dominant lipid classes in palm oil. Other lipid classes present in crude palm oil, such as phospholipids and galactolipids, are very low in abundance. These low-abundance lipids constitute key intermediates in lipid biosynthesis. In this study, we applied multiple lipidomic approaches, including high-sensitivity and high-specificity multiple reaction monitoring, to comprehensively quantify individual lipid species in crude palm oil. We also established a new liquid chromatography-coupled mass spectrometry method that allows direct quantification of low-abundance galactolipids in palm oil without the need for sample pretreatrnent. As crude palm oil contains large amounts of neutral lipids, our direct-detection method circumvents many of the challenges encountered with conventional lipid quantification methods. This approach allows direct measurement of lipids with no hassle during sample preparation and is more accurate and precise compared with other methods.  相似文献

Chemostat culture of Acinetobacter calcoaceticus KB-2 was done under palm oil-limiting conditions for cell production, and variation of cell compositions and yield coefficients were investigated in connection with the specific growth rates. At the concentration of 0.6% palm oil, the productivity of cells and yield coefficient were 4.76 g cells/l/h and 1.18 g cells/g palm oil, respectively, at a practical dilution rate of 0.85 h−1. About 80% of the palm oil was assimilated by the strain, and the maintenance coefficient was 0.035 g palm oil/g cells/h. Although the carbohydrate content remained essentially constant when the growth rate was varied, the lipid, protein, and nucleic acid contents were increased slightly at higher growth rates. Although the protein content increased only 3%, the protein yield coefficient (Yp) increased about 1.5 times over the range of specific growth rates between 0.1 and 0.7 h−1. The increase in Yp was due to the higher protein content of the biomass and to higher values of the cell yield coefficient.  相似文献

Food grade fisheries have reached their sustainable limits while aquaculture production has increased to meet consumer demands. However, for growth in aquaculture to continue and utilise sustainable, feeding ingredients, alternatives to fish oil (FO), the predominant lipid component of fish diets, must be developed. Therefore, there is currently considerable interest in the regulation of fatty acid metabolism in fish in order to determine strategies for the best use of plant oils in diets for commercially important cultured fish species. Plant oils are characteristically rich in C18 polyunsaturated fatty acids (PUFA) but devoid of C20 and C22 highly unsaturated fatty acids (HUFA) found in FO. The fatty acyl desaturase enzyme activities involved in the biosynthesis of HUFA from PUFA are known to be under nutritional regulation and can be increased in fish fed diets rich in plant oils. However, fatty acid desaturase activity is also known to be modulated by water temperature in fish. The present study aimed to investigate the interaction between water temperature and diet in the regulation of fatty acid metabolism in rainbow trout. Trout, acclimatized to 7, 11 or 15 degrees C, were fed for 4 weeks on diets in which the FO was replaced in a graded manner by palm oil. At the end of the trial, fatty acyl desaturation/elongation and beta-oxidation activities were determined in isolated hepatocytes and intestinal enterocytes using [1-14C]18:3n-3 as substrate, and samples of liver were collected for analysis of lipid and fatty acid composition. The most obvious effect of temperature was that fatty acid desaturation/elongation and beta-oxidation were reduced in both hepatocytes and intestinal enterocytes from fish maintained at the highest water temperature (15 degrees C). There were differences between the two tissues with the highest desaturation/elongation and beta-oxidation activities tending to be in fish held at 11 degrees C in the case of hepatocytes, but 7 degrees C in enterocytes. Correlations between fatty acid metabolism and dietary palm oil were most clearly observed in desaturation/elongation activities in both hepatocytes and enterocytes at 11 degrees C. The highest beta-oxidation activities were generally observed in fish fed FO alone in both hepatocytes and enterocytes with palm oil having differential effects in the two cell types.  相似文献

This work was designed to study the effect of different lipid sources on the activities of lipoprotein lipase and lipogenic enzymes in adipose tissue from rats fedad libitum or energy-controlled diets. Male Wistar rats were fed diets containing 40% of energy as fat (olive oil, sunflower oil, palm oil or beef tallow), for 4 wk. Underad libitum feeding no differences were found among dietary fat groups in final body weight, adipose tissue weights and total body fat. Under energy-controlled feeding, despite isoenergetic intake, rats fed the beef tallow diet gained significantly less weight than rats fed the other three diets. Beef tallow fed rats showed the lowest values for adipose tissue weights and total body fat. When rats had free access to food no effect of dietary lipid source on lipogenic enzyme activities was found. In contrast, under energy-controlled feeding rats fed the beef tallow diet showed significantly higher activities of glucose-6-phosphate dehydrogenase and fatty acid synthase than rats fed the other three diets. Heparin-releasable lipoprotein lipase activity in perirenal and subcutaneous adipose tissues was not different among rats fed olive oil, safflower oil, palm oil or beef tallow. When comparing both adipose tissue anatomical locations, significantly higher activities were found in subcutaneous than in perirenal fat pad independently of dietary fat. In conclusion, under our experimental protocol, lipogenesis in rat adipose tissue does not seem to be affected by dietary fat type.  相似文献
