Laboratory experiments were conducted using pure cultures ofAcinetobacter under anaerobic/aerobic cyclic conditions to explain the release and uptake of soluble phosphate in an activated sludge process showing enhanced biological phosphate removal (EBPR). Under anaerobic/aerobic cyclic conditions in a Sequencing Batch Reactor (SBR), COD uptake concurrent with soluble phosphate release byAcinetobacter was not significant during the anaerobic periods, indicating that EBPR would not be established in pure cultures. However,Acinetobacter cells accumulated higher phosphate content (5.2%) in SBR than that obtained (4.3%) from batch experiments. These results suggest thatAcinetobacter sp. may not follow the proposed pattern of behavior of poly-P bacteria in EBPR activated sludge plants.  相似文献

Polyphosphate accumulation by Paracoccus denitrificans was examined under aerobic, anoxic, and anaerobic conditions. Polyphosphate synthesis by this denitrifier took place with either oxygen or nitrate as the electron acceptor and in the presence of an external carbon source. Cells were capable of poly-β-hydroxybutyrate (PHB) synthesis, but no polyphosphate was produced when PHB-rich cells were incubated under anoxic conditions in the absence of an external carbon source. By comparison of these findings to those with polyphosphate-accumulating organisms thought to be responsible for phosphate removal in activated sludge systems, it is concluded that P. denitrificans is capable of combined phosphate and nitrate removal without the need for alternating anaerobic/aerobic or anaerobic/anoxic switches. Studies on additional denitrifying isolates from a denitrifying fluidized bed reactor suggested that polyphosphate accumulation is widespread among denitrifiers.  相似文献

Aerobic production of rhamnolipid by Pseudomonas aeruginosa was extensively studied. But effect of medium composition on anaerobic production of rhamnolipid by P. aeruginosa was unknown. A simplifying medium facilitating anaerobic production of rhamnolipid is urgently needed for in situ microbial enhanced oil recovery (MEOR). Medium factors affecting anaerobic production of rhamnolipid were investigated using P. aeruginosa SG (Genbank accession number KJ995745). Medium composition for anaerobic production of rhamnolipid by P. aeruginosa is different from that for aerobic production of rhamnolipid. Both hydrophobic substrate and organic nitrogen inhibited rhamnolipid production under anaerobic conditions. Glycerol and nitrate were the best carbon and nitrogen source. The commonly used N limitation under aerobic conditions was not conducive to rhamnolipid production under anaerobic conditions because the initial cell growth demanded enough nitrate for anaerobic respiration. But rhamnolipid was also fast accumulated under nitrogen starvation conditions. Sufficient phosphate was needed for anaerobic production of rhamnolipid. SO4 2? and Mg2+ are required for anaerobic production of rhamnolipid. Results will contribute to isolation bacteria strains which can anaerobically produce rhamnolipid and medium optimization for anaerobic production of rhamnolipid. Based on medium optimization by response surface methodology and ions composition of reservoir formation water, a simplifying medium containing 70.3 g/l glycerol, 5.25 g/l NaNO3, 5.49 g/l KH2PO4, 6.9 g/l K2HPO4·3H2O and 0.40 g/l MgSO4 was designed. Using the simplifying medium, 630 mg/l of rhamnolipid was produced by SG, and the anaerobic culture emulsified crude oil to EI24 = 82.5 %. The simplifying medium was promising for in situ MEOR applications.  相似文献

In this study, high-throughput pyrosequencing was applied on the analysis of the microbial community of activated sludge and biofilm in a lab-scale UV/O3- anaerobic/aerobic (A/O) integrated process for the treatment of petrochemical nanofiltration concentrate (NFC) wastewater. NFC is a type of saline wastewater with low biodegradability. From the anaerobic activated sludge (Sample A) and aerobic biofilm (Sample O), 59,748 and 51,231 valid sequence reads were obtained, respectively. The dominant phylotypes related to the metabolism of organic compounds, polycyclic aromatic hydrocarbon (PAH) biodegradation, assimilation of carbon from benzene, and the biodegradation of nitrogenous organic compounds were detected as genus Clostridium, genera Pseudomonas and Stenotrophomonas, class Betaproteobacteria, and genus Hyphomicrobium. Furthermore, the nitrite-oxidising bacteria Nitrospira, nitrite-reducing and sulphate-oxidising bacteria (NR-SRB) Thioalkalivibrio were also detected. In the last twenty operational days, the total Chemical Oxygen Demand (COD) and Total Organic Carbon (TOC) removal efficiencies on average were 64.93% and 62.06%, respectively. The removal efficiencies of ammonia nitrogen and Total Nitrogen (TN) on average were 90.51% and 75.11% during the entire treatment process.  相似文献

A sequencing batch reactor (SBR) was used to remove phosphate in biological wastewater treatment as an alternative to the activated sludge process, in order to improve the low removal efficiency of phosphate and the operational instability. After a cycle of 2 h anaerobic and 4 h aerobic conditions, phosphate removal was optimized. The removal efficiencies of 5 and 50 mg phosphate l–1 by Staphylococcus auricularis under repeated anaerobic and aerobic conditions were above 90%. These results showed that a long adaptation time, one of the major problems in biological phosphate removal process, was overcome by SBR.  相似文献

Under alternating aerobic/anaerobic conditions and without additional carbon sources, a bacterial consortium consisting initially of 18 bacterial strains was obtained in a sequence batch reactor. The phosphorus removal capability could only be maintained using sterile filtrate of activated sludge as medium. The addition of calcium and magnesium salts, as well as vitamins and trace elements, to autoclaved sterile filtrate of activated sludge was not sufficient to achieve stable phosphorus removal. A further enrichment by subcultivation on solid, agar, freezing, and shortening of the aerobic and anaerobic phases led to a defined bacterial consortium consisting of four strains. On the basis of physiological and chemotaxonomic characterization, and partial 16S rRNA sequencing, one of the organisms was identified as Delftia acidovorans. A further isolate belonged to the Bacillus cereus group, and the third isolate was identified as Microbacterium sp.. The remaining strain seems to represent a new genus within the Flavobacteriaceae. Under continuous chemostat conditions, this consortium was able to remove up to 9.6 mg P/l phosphate in the aerobic phase and released up to 8.5 mg/l in the anaerobic phase. Up to 25 mg P-polyphosphate/g dry mass was stored under aerobic conditions.  相似文献

Acetate is thought to be an important substrate for phosphate removal in anaerobic/aerobic activated sludge (AS) processes. The acetate content in municipal wastewater is low, and the main organic compounds in such wastewater are particulate organic matters (POMs) that are converted to endogenous substrates in AS processes when municipal wastewater is introduced into AS reactors. The question which then arises is which substrate, acetate or POM, is important for phosphate removal in full‐scale AS plants. The rates of phosphate release and substrate uptake were determined using AS harvested from a full‐scale anaerobic/aerobic AS plant and also AS acclimated to peptone under alternate anaerobic and aerobic conditions for 26 months. The rate of phosphate release upon POM addition per AS concentration per unit of time was about 0.84 mg PO4‐P/(g MLSS·h) irrespective of the wastewater quality. This value was about 0.05 in the case of AS acclimated to peptone for 26 months. When the AS concentration is 2.5 g/L and the mixed liquor retention time is 2 h in the anaerobic zone, about 4.2 mg/L PO4‐P is released upon POM addition. Hence, phosphate can be removed from municipal wastewater using full‐scale AS plants running under these conditions.  相似文献

Summary This study examined the microbial degradation of fuel oil by nine highly adapted different commercially available mixed bacterial cultures (DBC-plus, Flow Laboratories, Meckenheim, F.R.G.) and a bacterial community from a domestic sewage sludge sample. All mixed cultures were cultivated under aerobic batch conditions shaking (110 rpm) at 20°C in a mineral base medium containing 1 or 5% (v/v) fuel oil as the sole carbon source. Percent degradation of fuel oil and the n-alkane fraction was recorded for the nine DBC-plus cultures and the mixed population of the activated sludge sample. The increase in colony counts, protein, and optical density was studied during a 31-day incubation period for DBC-plus culture A, DBC-plus culture A2 and the activated sludge sample. The activated sludge mixed culture was most effective in degrading fuel oil, but various isolated bacterial strains from this bacterial community were not able to grow on fuel oil as the sole carbon source. In contrast, the n-alkane degradation rates of the DBC-cultures were lower, but single strains from the commercially available mixed cultures were able to mineralize fuel oil hydrocarbons. Strains ofPseudomonas aeruginosa were isolated most frequently and these organisms were able to grow very rapidly on fuel oil as a complex sole carbon source. The results indicate that fuel oil degradation in domestic sewage sludge is performed by mixed populations of naturally occurring bacteria and does not depend on the application of highly adapted commercially available cultures.  相似文献

The addition of iron (III) hydroxide during methanogenic digestion of activated sludge by anaerobic sludge displaying an iron-reducing activity resulted in a microbial reduction of iron (III) with the formation of iron (II), capable of precipitating phosphates. The feasibility of eliminating 66.6 to 99.6% of the dissolved phosphate at initial concentrations of 1000 to 3500 mg PO3- 4/l by adding 6420 mg/l iron (III) hydroxide into a reactor for anaerobic fermentation of activated sludge was analyzed. The optimal ratio of iron (III) added to dissolved phosphate removed (mg) providing a 95% removal amounted to 2 : 1. These results may be used in new technology for anaerobic wastewater treatment with phosphate removal.  相似文献

A continuous-flow anaerobic–anoxic (A2) activated sludge system was operated for efficient enhanced biological phosphorus removal (EBPR). Because of the system configuration with no aeration zones, phosphorus (P) uptake takes place solely under anoxic conditions with simultaneous denitrification. Basic operating conditions, namely biomass concentration, influent carbon to phosphorus ratio and anaerobic retention time were chosen as variables in order to assess their impact on the system performance. The experimental results indicated that maintenance of biomass concentration above 2,500 mg MLVSS/L resulted in the complete phosphate removal from the influent (i.e. 15 mg PO4 3−-P/L) for a mean hydraulic residence time (HRT) of 15 h. Additionally, by increasing the influent COD/P ratio from 10 to 20 g/g, the system P removal efficiency was improved although the experimental results indicated a possible enhancement of the competition between phosphorus accumulating organisms (PAOs) and other microbial populations without phosphorus uptake ability. Moreover, because of the use of acetate (i.e. easily biodegradable substrate) as the sole carbon source in the system feed, application of anaerobic retention times greater than 2 h resulted in no significant release of additional P in the anaerobic zone and no further amelioration of the system P removal efficiency. The application of anoxic P removal resulted in more than 50% reduction of the organic carbon necessitated for nitrogen and phosphorus removal when compared to a conventional EBPR system incorporating aerobic phosphorus removal.  相似文献

Microbial aggregates of an aerobic granular sludge can be used for the treatment of industrial or municipal wastewater, but their formation from a microbial activated sludge requires several weeks. Therefore, the aim of this research was the selection of microbial cultures to shorten the granule-forming period from several weeks to a few days. An enrichment culture with the ability to accelerate granulation was obtained by repeating the selection and batch cultivation of fast-settling microbial aggregates isolated from the aerobic granular sludge. Bacterial cultures of Klebsiella pneumoniae strain B and Pseudomonas veronii strain F, with self-aggregation indexes of 65 and 51%, respectively, and a coaggregation index of 58%, were isolated from the enrichment culture. A mixture of these strains with the activated sludge was used as an inoculum in an experimental sequencing batch reactor to start up an aerobic granulation process. Aerobic granules with a mean diameter of 446±76 μm were formed in an experiment after 8 days of cultivation, but microbial granules were absent in controls. The effects of potassium on excess uptake of phosphate in an aerobic-anaerobic activated sludge process were examined by the fill-and-draw procedure. The presence of sufficient potassium was necessary for excess uptake to occur. The contents of potassium and phosphate in the sludge at the end of each cycle of the process were correlated with each other by a non-linear equation, with some scattering. However, the sum of the molar ion valences of magnesium (Mg) and potassium (K), 2 Mg + K, was well correlated by a linear equation with the moles of P expressed as mmol/g-VSS, with a correlation coefficient of 0.993. When the potassium concentration was insufficient for the enhanced uptake of phosphate, its concentration in treated water was of the order of 0.1 mg/l. In the first anaerobic period phosphate was released into the liquid phase, but potassium was released after initial instantaneous removal, and in the successive aerobic period they were both taken up again.  相似文献

Polyphosphate accumulating bacteria were isolated from activated sludge cultured in a fill and draw system with alternating anaerobic and aerobic conditions. One of the isolates, strain NM-1 tentatively identified with the genus Micrococcus accumulated a large amount of phosphorus and its content reached 166 mg of phosphorus per g of cells. This strain accumulated phosphorus rapidly under aerobic conditions in the absence of any organic substrates, and when glucose was supplied to this strain under anaerobic conditions, it released phosphate and took up glucose in a similar manner to activated sludge that has good phosphorus eliminating capacity. Micro-aerophilic growth conditions were required for the induction of phosphorus accumulating activity.  相似文献

The aim of this work was to establish a relation between the mean cellular retention time and the ability of activated sludge to remove phosphate and ammonium. A sequencing batch reactor (SBR) with a total volume of 1.94 m3 was fed with municipal wastewater and was operated under four different organic loading rates to obtain sludge ages of 23, 16, 6, and 3 days. The operational strategy included fill, anaerobic, aerobic, settling and draw phases. The experimental work lasted 445 days. Biological phosphate removal was achieved with sludge ages from 6 to 23 days. The highest PO4-P removal rate observed was of 98% and corresponds to a 16-day sludge age; phosphate removal increased with the sludge age. A sludge age of 3 days resulted in a chemical oxygen demand (COD) removal rate of 81% and a sludge age of 23 days in a removal rate of 99%. Full nitrification was observed with a sludge age of 16 days. Nitrification increased with the sludge age. The 3-day sludge age did not allow nitrification. The phosphate concentrations in the biomass were inversely proportional to the sludge age.  相似文献

Acetic acid is thought to be an important substrate for the removal of phosphate in anaerobic/aerobic activated sludge (AS) processes. However, the acetic acid content in municipal sewage is low, and the main organic compounds in such sewage are particulate organic matters (POM) that are converted to endogenous substrates (E(ntrapped) POM, i.e., EPOM) in AS processes. Thus, the question arises whether it is really acetate or POM, which is important for the removal of phosphate in full‐scale AS plants. AS was harvested from a full‐scale anaerobic/aerobic AS plant. The amount of phosphate released after the addition of acetic acid depends on the AS conditions, particularly the influent sewage quality. However, the amount of phosphate released by EPOM was not affected by the AS conditions, and the amount of phosphate released per AS concentration and per unit of time was calculated to be about 0.86 mg PO4‐P/g MLSS/hour. When the AS concentration is 2.5 g/L and the mixed‐liquor retention time is 2 hours in the anaerobic zone, about 4 mg/L PO4‐P is released from EPOM. Under these conditions, phosphate in such sewage is removed by full‐scale AS plants without using acetic acid. In the case of carbon deficiency, the introduction of primary sludge to the anaerobic zone promoted the release of phosphate.  相似文献

The effects of magnesium on excess uptake of phosphate in an aerobic-anaerobic activated sludge process were examined by the fill and draw procedure. The alternation of anaerobic and aerobic conditions in one cycle of fill and draw process was varied many ways.The presence of sufficient magnesium was necessary for uptake of excess phosphate. When sludge contained more phosphorus than the upper limit of phosphorus content in the usual aerobic activated sludge, 2.5% by weight, magnesium was also contained in more than an ordinary amount (0.5%). Their contents in the sludge at the end of each cycle of the process were correlated with each other by a linear equation with the correlation coefficient of 0.99. When magnesium concentration was insufficient for the uptake of excess phosphate, its concentration in the treated water was of the order of 0.1 mg/l.In the first anaerobic period both phosphate and magnesium were released, and in a successive aerobic period they were taken up again. The weight ratio of differential amounts of phosphorus and magnesium released or taken up changed with time in one cycle.Dynamic behaviors of phosphate and magnesium removal against the step change of feed magnesium concentration also showed a stoichiometric relationship supporting the correlation equation above mentioned.  相似文献

H2-producing bacteria were isolated from anaerobic granular sludge. Out of 72 colonies (36 grown under aerobic conditions and 36 under anaerobic conditions) arbitrarily chosen from the agar plate cultures of a suspended sludge, 34 colonies (15 under aerobic conditions and 19 under anaerobic conditions) produced H2 under anaerobic conditions. Based on various biochemical tests and microscopic observations, they were classified into 13 groups and tentatively identified as follows: From aerobic isolates,Aeromonas spp. (7 strains),Pseudomonas spp. (3 strains), andVibrio spp. (5 strains); from anaerobic isolates,Actinomyces spp. (11 strains),Clostridium spp. (7 strains), andPorphyromonas sp. When glucose was used as the carbon substrate, all isolates showed a similar cell density and a H2 production yield in the batch cultivations after 12h (2.24–2.74 OD at 600 nm and 1.02–1.22 mol H2/mol glucose, respectively). The major fermentation by-products were ethanol and acetate for the aerobic isolates, and ethanol, acetate and propionate for the anaerobic isolates. This study demonstrated that several H2 producers in an anaerobic granular sludge exist in large proportions and their performance in terms of H2 production is quite similar.  相似文献

A method for quantifying bacterial populations introduced into an activated-sludge microbial community is described. The method involves extraction of DNA from activated sludge, appropriate dilution of the extracted DNA with DNA extracted from nonintroduced activated sludge, PCR amplification of a gyrB gene fragment from the introduced strain with a set of strain-specific primers, and quantification of the electrophoresed PCR product by densitometry. The adequacy of the method was examined by analyzing the population dynamics of two phenol-degrading bacteria, Pseudomonas putida BH and Comamonas sp. strain E6, that had been introduced into phenol-digesting activated sludge. The density of each of the two populations determined by the PCR method immediately after the introduction was consistent with the density estimated from a plate count of the inoculum. This quantitative PCR method revealed different population dynamics for the two strains in the activated sludge under different phenol-loading conditions. The behavior of both of these strains in the activated sludge reflected the growth kinetics of the strains determined in laboratory axenic cultures.  相似文献
