Abscisic acid (ABA), auxins, and cytokinins (CKs) are known to be closely linked to nitrogen signaling. In particular, CKs control the effects of nitrate availability on plant growth. Our group has shown that treatment with high nitrate concentrations limits root growth and leaf development in maize, and conditions the development of younger roots and leaves. CKs also affect source-sink relationships in plants. Based on these results, we hypothesized that CKs regulate the source-sink relationship in maize via a mechanism involving complex crosstalk with the main auxin indole-3-acetic acid (IAA) and ABA. To evaluate this hypothesis, various CK metabolites, IAA, and ABA were quantified in the roots and in source and sink leaves of maize plants treated with high and normal nitrate concentrations. The data obtained suggest that the cis and trans isomers of zeatin play completely distinct roles in maize growth regulation by a complex crosstalk with IAA and ABA. We demonstrate that while trans-zeatin (tZ) and isopentenyladenine (iP) regulate nitrate uptake and thus control final leaf sizes, cis-zeatin (cZ) regulates source and sink strength, and thus controls leaf development. The implications of these findings relating to the roles of ABA and IAA in plants’ responses to varying nitrate concentrations are also discussed.  相似文献

Background and aims

The root surface of a plant usually exceeds the leaf area and is constantly exposed to a variety of soil-borne microorganisms. Root pathogens and pests, as well as belowground interactions with beneficial microbes, can significantly influence a plants' performance. Unfortunately, the analysis of these interactions is often limited because of the arduous task of accessing roots growing in soil. Here, we present a soil-free root observation system (SF-ROBS) designed to grow maize (Zea mays) plants and to study root interactions with either beneficial or pathogenic microbes.

Methods

The SF-ROBS consists of pouches lined with wet filter paper supplying nutrient solution.

Results

The aspect of maize grown in the SF-ROBS was similar to soil-grown maize; the plant growth was similar for the shoot but different for the roots (biomass and length increased in the SF-ROBS). SF-ROBS-grown roots were successfully inoculated with the hemi-biotrophic maize fungal pathogen Colletotrichum graminicola and the beneficial rhizobacteria Pseudomonas putida KT2440. Thus, the SF-ROBS is a system suitable to study two major belowground phenomena, namely root fungal defense reactions and interactions of roots with beneficial soil-borne bacteria.

Conclusions

This system contributes to a better understanding of belowground plant microbe interactions in maize and most likely also in other crops.  相似文献

Aims

This study explores soil nutrient cycling processes and microbial properties for two contrasting vegetation types along an elevational gradient in subarctic tundra to improve our understanding of how temperature influences nutrient availability in an ecosystem predicted to be sensitive to global warming.

Methods

We measured total amino acid (Amino-N), mineral nitrogen (N) and phosphorus (P) concentrations, in situ net N and P mineralization, net Amino-N consumption, and microbial biomass C, N and P in both heath and meadow soils across an elevational gradient near Abisko, Sweden.

Results

For the meadow, NH4 + concentrations and net N mineralization were highest at high elevations and microbial properties showed variable responses; these variables were largely unresponsive to elevation for the heath. Amino-N concentrations sometimes showed a tendency to increase with elevation and net Amino-N consumption was often unresponsive to elevation. Overall, PO4-P concentrations decreased with elevation and net P immobilization mostly occurred at lower elevations; these effects were strongest for the heath.

Conclusions

Our results reveal that elevation-associated changes in temperature can have contrasting effects on the cycling of N and P in subarctic soils, and that the strength and direction of these effects depend strongly on dominant vegetation type.  相似文献

In plants of wheat (Triticum aestivum L.) grown in the media with nitrate (NO 3 ? plants), ammonium (NH 4 + plants), and without nitrogen (N-deficient plants), the response to oxidative stress induced by the addition of 300 mM NaCl to the nutrient solution was investigated. Three-day-long salinization induced chlorophyll degradation and accumulation of malondialdehyde (MDA) in the leaves. These signs of oxidative stress were clearly expressed in NO 3 ? and N-deficient plants and weakly manifested in NH 4 + plants. In none of the treatments, salinization induced the accumulation of MDA in the roots. Depending on the conditions of N nutrition, salt stress was accompanied by diverse changes in the activity of antioxidant enzymes in the leaves and roots. Resistance of leaves of NH 4 + plants to oxidative stress correlated with a considerable increase in the activities of ascorbate peroxidase and glutathione reductase. Thus, wheat plants grown on the NH 4 + -containing medium were more resistant to the development of oxidative stress in the leaves than those supplied with nitrate.  相似文献

Peatlands, with organic soils, are usually dominated by one or a few species. Above and belowground ecological processes are regulated by the characteristics of the dominant species in the peat. Understanding how these species relate to climatic or water chemistry gradients will help to predict the fate of those ecosystems under current climate change. The patterns of abundance and occurrence of 12 major peat-forming species from different structural plant groups were quantified along gradients of elevation, precipitation, and water chemistry. The species were distributed in four major structural groups: cushion plants, true mosses, sedges, and Sphagnum mosses. A response curve for each species was built with Generalized Additive Models. Niche breadths, species tolerances, and species optima were estimated using bootstrap resampling. Our results showed that species were limited in their elevational ranges; Sphagnum species were biased toward low to intermediate elevations, sedges, and true mosses to intermediate elevations and cushion plants to very high elevations. Sphagnum species were more likely to occur in sites with low precipitation, while sedges preferred wet sites. Sphagnum species preferred habitats with acidic pH. The species tolerances and optimum distributions are an indication to the vulnerability of the species to changes in their environment. Species with limited tolerances are more vulnerable, such as the narrow elevational distribution of cushion plants or the low pH and narrow tolerances observed for Sphagnum species. Climate and hydrological changes will very possibly affect the distribution of those species forcing changes on ecosystem functioning.  相似文献

Background and aims

Production of indole-3-acetic acid (IAA) by Azospirillum brasilense is one of the most important mechanisms underlying the beneficial effects observed in plants after inoculation with this bacterium. This study determined the contribution of the hisC1 gene, which encodes aromatic amino acid aminotransferase-1 (AAT1), to IAA production, and analyzed its expression in the free-living state and in association with the roots of wheat.

Methods

We determined production of IAA and AAT activity in the mutant hisC::gusA-sm R . To study the expression of hisC1, a chromosomal gene fusion was analyzed by following β-glucuronidase (GUS) activity in vitro, in the presence of root exudates, and in association with roots.

Results

IAA production in the hisC mutant was not reduced significantly compared to the activity of the wild-type strain. AAT1 activity was reduced by 50% when tyrosine was used as the amino acid donor, whereas there was a 30% reduction when tryptophan was used, compared to the activity of the wild-type strain. Expression of the fusion protein was up-regulated in both logarithmic and stationary phases by several compounds, including IAA, tryptophan, tyrosine, and phenyl acetic acid. We observed the expression of hisC1 in bacteria associated with wheat roots. Root exudates of wheat and maize were able to stimulate hisC1 expression.

Conclusions

The expression data indicate that hisC1 is under a positive feedback control in the presence of root exudates and on plants, suggesting that AAT1 activity plays a role in Azospirillum–plant interactions.  相似文献

Many studies have tried to explain the physiological mechanisms of the alpine treeline phenomenon, but the debate on the alpine treeline formation remains controversial due to opposite results from different studies. The present study explored the carbon-physiology of an alpine shrub species (Quercus aquifolioides) grown at its upper elevational limit compared to lower elevations, to test whether the elevational limit of alpine shrubs (<3 m in height) are determined by carbon limitation or growth limitation. We studied the seasonal variations in non-structural carbohydrate (NSC) and its pool size in Q. aquifolioides grown at 3000 m, 3500 m, and at its elevational limit of 3950 m above sea level (a.s.l.) on Zheduo Mt., SW China. The tissue NSC concentrations along the elevational gradient varied significantly with season, reflecting the season-dependent carbon balance. The NSC levels in tissues were lowest at the beginning of the growing season, indicating that plants used the winter reserve storage for re-growth in the early spring. During the growing season, plants grown at the elevational limit did not show lower NSC concentrations compared to plants at lower elevations, but during the winter season, storage tissues, especially roots, had significantly lower NSC concentrations in plants at the elevational limit compared to lower elevations. The present results suggest the significance of winter reserve in storage tissues, which may determine the winter survival and early-spring re-growth of Q. aquifolioides shrubs at high elevation, leading to the formation of the uppermost distribution limit. This result is consistent with a recent hypothesis for the alpine treeline formation.  相似文献

Background and aims

Trees allocate a high proportion of assimilated carbon belowground, but the partitioning of that C among ecosystem components is poorly understood thereby limiting our ability to predict responses of forest C dynamics to global change drivers.

Methods

We labeled sugar maple saplings in natural forest with a pulse of photosynthetic 13C in late summer and traced the pulse over the following 3 years. We quantified the fate of belowground carbon by measuring 13C enrichment of roots, rhizosphere soil, soil respiration, soil aggregates and microbial biomass.

Results

The pulse of 13C contributed strongly to root and rhizosphere respiration for over a year, and respiration comprised about 75 % of total belowground C allocation (TBCA) in the first year. We estimate that rhizosphere carbon flux (RCF) during the dormant season comprises at least 6 % of TBCA. After 3 years, 3.8 % of the C allocated belowground was recovered in soil organic matter, mostly in water-stable aggregates.

Conclusions

A pulse of carbon allocated belowground in temperate forest supplies root respiration, root growth and RCF throughout the following year and a small proportion becomes stabilized in soil aggregates.  相似文献

Aims

Phenanthrene is one of the ubiquitous, persistent organic pollutants commonly found in soil and sediments. The study will provide insight regarding the feasibility of nitrogen-assisted phytoremediation.

Methods

To study the effects of various nitrogen forms on cucumber seedling phenanthrene tolerance, hydroponic experiments were conducted in a greenhouse.

Results

Under phenanthrene stress, decreases in plant growth and biomass were more pronounced with a nitrate supply than with ammonium. In addition, phenanthrene concentrations in plants fed with ammonium were higher than those fed with nitrate. The reduction in plant protein and sugar, increases in nitrogen and phosphate concentrations, and increased activity of antioxidative enzymes may contribute to the phenanthrene stress response and adaptation. Higher peroxidase, superoxide dismutase, and catalase activities were found in ammonium-fed plants as compared to nitrate-fed plants under phenanthrene stress. Moreover, the reduction in soluble protein content and increases in phenanthrene transport and accumulation in non-photosynthetic organs may enable ammonium-fed plants to adapt more effectively to adverse conditions.

Conclusions

Overall, these results suggest that ammonium nutrition could provide a useful tool to improve the growth and adaption of plants under phenanthrene stress.  相似文献

Aims

Some rhizobia can convert insoluble P into available forms for plant growth but the underlying mechanisms for this are not understood. In this study, the function of rhizobia in P acquisition from P sources for soybean was studied.

Methods

Four rhizobial strains were employed to evaluate their phosphate-solubilizing (PS) activity, their ability to mediate pH changes in growth medium for different P sources, and IAA production. A sand culture experiment using different P sources was carried out to characterize P acquisition changes of soybean plants with or without rhizobium inoculation. Rhizospheric acidification in soybean was further analyzed in hydroponics.

Results

Our results showed that all the tested rhizobial strains exhibited significant PS activity for different P sources in the order of Ca-P>Al-P>Phy-P??Fe-P as indicated by the halo/colony ratio technique and increased Pi percentage in the solid and liquid phases, respectively. Furthermore, all of the rhizobial strains could acidify the growth medium for all P sources except Phy-P, but only three of them produced IAA. Compared to non-nodulated plants, the nodulated plants had greater plant biomass and P content in sand culture for all the tested P sources, especially for Ca-P. Moreover, H+ and total acid exudation was more significantly enhanced in the nodulated plants in hydroponics.

Conclusions

Our results suggested that the PS ability of rhizobia is more related to acidification of the growth medium than IAA production. Rhizobium inoculation could enhance P acquisition in soybean, especially on soils where Ca-P is the primary P source, and the primary mechanism for rhizobial-mediated P solubilization appears to be via Pi remobilization of nodulated roots through rhizospheric acidification.  相似文献

Background and aims

Wetland plants have been widely used in constructed wetlands for the clean-up of metal-contaminated waters. This study investigated the relationship between rate of radial oxygen loss (ROL), root porosity, Zn uptake and tolerance, Fe plaque formation in wetland plants.

Methods

A hydroponic experiment and a pot trial with Zn-contaminated soil were conducted to apply different Zn level treatments to various emergent wetland plants.

Results

Significant differences were found between plants in their root porosities, rates of ROL, Zn uptake and Zn tolerance indices in the hydroponic experiment, and concentrations of Fe and Mn on roots and in the rhizosphere in the pot trial. There were significant positive correlations between root porosities, ROL rates, Zn tolerance, Zn, Fe and Mn concentrations on roots and in the rhizosphere. Wetland plants with higher root porosities and ROL tended to have more Fe plaque, higher Zn concentrations on roots and in their rhizospheres, and were more tolerant of Zn toxicity.

Conclusions

Our results suggest that ROL and root porosity play very important roles in Fe plaque formation, Zn uptake and tolerance, and are useful criteria for selecting wetland plants for the phytoremediation of Zn-contaminated waters and soils/sediments.  相似文献

The present work with transgenic poplar lines producing varying levels of trans -zeatin suggests the existence of a switching threshold for triggering ckx gene expression or suppressing cytokinin-induced auxin.

Abstract

Cytokinins have an important role in growth and developmental processes of plants. Transgenic plants with varying levels of cellular cytokinin are convenient tools for studying its role in morphogenetic as well as molecular responses. In this work, the transgenic lines producing either high level of cellular trans-zeatin (HX lines) or moderate level (MX lines) were compared with regard to their cytokinin oxidase activities and cellular auxin content. The HX lines showed typical cytokinin phenotypes including leafy shoots and spontaneous shoot formation on hormone free medium. In contrast, the MX lines did not show any striking phenotypes. However, in leaf disk culture on hormone free medium, they regenerated roots and subsequently formed shoots from the roots. Determination of cellular IAA content revealed a significant increase in the level in MX lines but not in HX lines. Of nine cytokinin oxidase genes (ckx) examined by qPCR, five were activated in HX lines but not in MX lines. Among them, ckx4 appeared to play a key role in maintaining cellular cytokinin level since it showed more than 1,000-fold increase in HX lines and in the leaf disks of untransformed control exposed to exogenous cytokinins. Although low level of cellular cytokinin did not induce the expression of ckx genes, it appeared to trigger cellular IAA biosynthesis.  相似文献

Aims

The mechanisms of belowground competition are not well understood. Addressing literature reports on competition-induced changes in tree fine root morphology, we conducted a growth experiment with tree saplings to investigate competition effects on important root morphological and functional traits in a root order-focused analysis.

Methods

European beech and European ash saplings were grown for 34 months in containers under greenhouse conditions in monoculture (2 conspecific plants), in mixture (1 beech and 1 ash) or as single plants. The root system was fractionated according to root orders and eight morphological and functional properties were determined.

Results

Root order was the most influential factor affecting the fine root traits (except for root diameter and δ13C); a significant species identity effect was found for root diameter, tissue density, N concentration and δ13C. Ash fine roots were thicker, but had lower tissue densities, contained more N and had systematically higher δ13C values than beech roots. The competition treatments had no significant effect on morphological root traits but altered δ13C in the 2nd root order.

Conclusion

Neither intra- nor interspecific root competition affected fine root morphology significantly suggesting that competition-induced root modification may not be a universal phenomenon in temperate trees.  相似文献

Background

Studying root biomass, root system distribution and belowground interactions is essential for understanding the composition of plant communities, the impact of global change, and terrestrial biogeochemistry. Most soil samples and minirhizotron pictures hold roots of more than one species or plant individual. The identification of taxa by their roots would allow species-specific questions to be posed; information about root affiliation to plant individuals could be used to determine intra-specific competition.

Scope

Researchers need to be able to discern plant taxa by roots as well as to quantify abundances in mixed root samples. However, roots show less distinctive features that permit identification than aboveground organs. This review discusses the primary use of available methods, outlining applications, shortcomings and future developments.

Conclusion

Methods are either non-destructive, e.g. visual examination of root morphological criteria in situ, or require excavated and excised root samples. Among the destructive methods are anatomical keys, chemotaxonomic approaches and molecular markers. While some methods allow for discerning the root systems of individual plants, others can distinguish roots on the functional group or plant taxa level; methods such as IR spectroscopy and qPCR allow for quantifying the root biomass proportion of species without manual sorting.  相似文献

Aims

Plants with precise root foraging patterns can proliferate roots preferentially in nutrient-rich soil patches. When nutrients are distributed heterogeneously, this trait is often competitively advantageous in pot experiments but not field experiments. We hypothesized that this difference is due to belowground herbivory under field conditions.

Methods

We performed pot experiments using seedlings of Lolium perenne (a more precise root foraging species) and Plantago lanceolata (a less precise root foraging species). The experiment had a two-way factorial randomized block design, with nutrient distribution pattern (homogeneous or heterogeneous) and belowground herbivore (present or absent) as the two factors. Each pot contained one seedling of each species.

Results

With no herbivore present, plant biomass was smaller in the heterogeneous nutrient treatment than in the homogeneous treatment in P. lanceolata, but not in L. perenne. Under homogeneous nutrient distribution, plant biomass was lower in both species with a herbivore present than with no herbivore. Under heterogeneous nutrient distribution, biomass reduction due to herbivory occurred only in L. perenne.

Conclusions

Roots of the precise root foraging species were grazed more under the heterogeneous nutrient distribution, suggesting that the herbivore more efficiently foraged for roots in nutrient-rich soil patches.  相似文献

Background and aims

Characterisation of genetic variation in nitrate accumulation by lettuce will inform strategies for selecting low-nitrate varieties more capable of meeting EU legislation on harvested produce. This study uses a population of recombinant inbred lines (RILs) of lettuce to determine how genotypic differences influence N uptake, N assimilation and iso-osmotic regulation, and to identify key related traits prior to future genetic analysis.

Methods

Measurements were made on plants grown to maturity in soil fertilised with ammonium nitrate, and in a complete nutrient solution containing only nitrate-N. A simple osmotic balance model was developed to estimate variations in shoot osmotic concentration between RILs.

Results

There were significant genotypic variations in nitrate accumulation when plants were grown either with nitrate alone or in combination with ammonium. Ammonium-N significantly reduced nitrate in the shoot but had no effect on its relative variability, or on the ranking of genotypes. Shoot nitrate-N was correlated positively with total-N and tissue water, and negatively with assimilated-C in both experiments. Corresponding relationships with assimilated-N and shoot weight were weaker. Estimated concentrations of total osmotica in shoot sap were statistically identical in all RILs, despite variations in nitrate concentration across the population.

Conclusions

Approximately 73% of the genotypic variability in nitrate accumulation within the population of RILs arose from differences in nitrate uptake and only 27% from differences in nitrate assimilated, irrespective of whether or not part of the N was recovered as ammonium, or whether the plants were grown in soil or solution culture. Genotypic variability in nitrate accumulation was associated with changes in concentrations of other endogenous solutes (especially carboxylates and soluble carbohydrates) and of tissue water, which minimised differences in osmotic potential of shoot sap between RILs. This offers the opportunity of using the regulation of these solutes as additional traits to manipulate nitrate accumulation.  相似文献

An efficient protocol of transformation and selection of transgenic lines of Micro-tom, a widespread model cultivar for tomato, is reported. RNA interference silencing efficiency and stability have been investigated and correlated with the number of insertions.

Abstract

Given its small size and ease of cultivation, the tomato (Solanum lycopersicon) cultivar Micro-tom is of widespread use as a model tomato plant. To create and screen transgenic plants, different selectable markers are commonly used. The bar marker carrying the resistance to the herbicide glufosinate/Basta, has many advantages, but it has been little utilised and with low efficiency for identification of tomato transgenic plants. Here we describe a procedure for accurate selection of transgenic Micro-tom both in vitro and in soil. Immunoblot, Southern blot and phenotypic analyses showed that 100 % of herbicide-resistant plants were transgenic. In addition, regeneration improvement has been obtained by using 2 mg/l Gibberellic acid in the shoot elongation medium; rooting optimisation on medium containing 1 mg/l IAA allowed up to 97 % of shoots developing strong and very healthy roots after only 10 days. Stable transformation frequency by infection of leaf explants with Agrobacterium reached 12 %. Shoots have been induced by combination of 1 mg/l zeatin-trans and 0.1 mg/l IAA. Somatic embryogenesis of cotyledon on medium containing 1 mg/l zeatin + 2 mg/l IAA is described in Micro-tom. The photosynthetic psbS gene has been used as reporter gene for RNA silencing studies. The efficiency of gene silencing has been found equivalent using three different target gene fragments of 519, 398 and 328 bp. Interestingly, silencing efficiency decreased from T0 to the T3 generation in plants containing multiple copies of the inserted T-DNA, while it was stable in plants containing a single insertion.  相似文献

A plant’s growth and fitness are influenced by species interactions, including those belowground. In primary successional systems, belowground organisms are known to have particularly important control over plant growth. Exotic plant invasions in these and other habitats may in part be explained by altered associations with belowground organisms compared to native plants. We investigated the growth responses of two foundation grasses on Great Lakes sand dunes, the native grass Ammophila breviligulata and the exotic grass Leymus arenarius, to two groups of soil organisms with important roles in dune succession: arbuscular mycorrhizal fungi (AMF) and plant-parasitic nematodes (PPN). We manipulated the presence/absence of two generalist belowground species known to occur in Great Lakes dunes, Rhizophagus intraradices (AMF) and Pratylenchus penetrans (PPN) in a factorial greenhouse experiment and assessed the biomass production and root architectural traits of the plants. There were clear differences in growth and above- and belowground architecture between Ammophila and Leymus, with Leymus plants being bigger, taller, and having longer roots than Ammophila. Inoculation with Rhizophagus increased above- and belowground biomass production by ~32% for both plant species. Inoculation with Pratylenchus decreased aboveground biomass production by ~36% for both plant species. However belowground, the exotic Leymus was significantly more resistant to PPN than the native Ammophila, and gained more benefits from AMF in belowground tri-trophic interactions than Ammophila. Overall, our results indicate that differences in plant architecture coupled with altered belowground interactions with AMF and PPN have the potential to promote exotic plant invasion.  相似文献

The elevational range of the alpine cushion plant Laretia acaulis (Apiaceae) comprises a cold upper extreme and a dry lower extreme. For this species, we predict reduced growth and increased non-structural carbohydrate (NSC) concentrations (i.e. carbon sink limitation) at both elevational extremes. In a facilitative interaction, these cushions harbor other plant species (beneficiaries). Such interactions appear to reduce reproduction in other cushion species, but not in L. acaulis. However, vegetative effects may be more important in this long-lived species and may be stronger under marginal conditions. We studied growth and NSC concentrations in leaves and stems of L. acaulis collected from cushions along its full elevational range in the Andes of Central Chile. NSC concentrations were lowest and cushions were smaller and much less abundant at the highest elevation. At the lowest elevation, NSC concentrations and cushion sizes were similar to those of intermediate elevations but cushions were somewhat less abundant. NSC concentrations and growth did not change with beneficiary cover at any elevation. Lower NSC concentrations at the upper extreme contradict the sink-limitation hypothesis and may indicate that a lack of warmth is not limiting growth at high-elevation. At the lower extreme, carbon gain and growth do not appear more limiting than at intermediate elevations. The lower population density at both extremes suggests that the regeneration niche exerts important limitations to this species’ distribution. The lack of an effect of beneficiaries on reproduction and vegetative performance suggests that the interaction between L. acaulis and its beneficiaries is probably commensalistic.  相似文献
