Water extracts of the red-tide dinoflagellate Alexandrium excavatumgrown at ‘high’ light intensity (200 µE m–2s–1) show a broad absorbance maximum in the UV regionof the spectrum (310–360 nm). Using TLC and reverse-phaseHPLC a series of mycosporine-like amino acids have been characterized:mycosporine-glycine (max = 310 nm), palythine (max = 320 nm),asterina-330 (max = 330 nm), shinorine (max = 334 nm), porphyra-334(max= 334 nm), palythenic acid (max = 337 nm) and the isomericmixture of usujirene and palythene (max = 359 nm). From theobserved spectral changes during transference from ‘low’(20 µE m–2 s–1) to ‘high’ (200µE m–2 s–1) light intensities and vice versa,the series of compounds are supposed to be biogenically relatedto one another. The presence of these compounds in A.excavatumis discussed in relation to their possible role in the photoprotectionto deleterious UV radiation.  相似文献

Primary production was measured for 7 years, using the in situ14C-method in hypertrophic Hartbeespoort Dam, South Africa,to examine the influence of light and water temperature on theupper limit of Microcystis aeruginosa production. Water temperaturesvaried from 11 to >25°C and chlorophyll concentrationsreached 6500 mg m–3. The maximum volumetric rate of production(Amax) was 12->8800 mg C m–3 h–1 with areal productions(A) of 69->3300 mg C m–2 h–1 for euphotic zonedepths of <0.5–8.4 m. The intrinsic parameters of phytoplanktonproduction (, Amax/B, Ik) indicated that the phytoplankton populationwas adapted to high light levels. Both Amax/B and Ik were correlatedwith temperature. Under optimal conditions, , the theoreticalupper limit of A, was calculated to be 2.8 g Cm–2 h–1,while the measured rate was 2.5 g Cm–2 h–1. Measuredareal rates exceeding were overestimated due to methodologicalproblems when working with Microcystis scums. Light and watertemperature interacted to yield high production rates: watertemperature through its direct effect on photosynthetic ratesand indirectly in the formation of diurnal mixed layers; lightindirectly through water temperature and directly through itsattenuation and induction of light-adapted physiology in Microcystis.  相似文献

The physical factors controlling algal primary production weredemonstrated from data collected for a hypertrophic lake. Amaxranged between 12.4 and 5916 mg C m–3 h–1. Arealrates (A) varied between 46.9 and 3381 mg C m–2 h–1.The factors permitting and controlling production were subjectivelyseparated into two categories. In category 1, nutrients (N +P), which were in overabundance, permitted large standing cropsof Microcystis aeruginosa to develop (>1000 µg chla 1–1). Wind patterns determined the dramatic spatialand temporal changes in algal standing crop which could dropto 2.7 µg chl a 1–1. In category 2 were the factorswhich affected the rate processes. The buoyancy mechanism ofMicrocystis usually kept the alga in the euphotic zone. A powerrelationship (r = 0.92, n = 54) between A and Amax/min showedthat with increasing phytoplankton vertical stratification,Amax was increasingly important in the integral. The saturationparameter IK and photosynthetic capacity were temperature dependent.Variations of A were significantly related to changes in watercolumn stability (g cm cm–2) because both axes of thephotosynthesis depth-profile were affected by stability changes.  相似文献

This work aimed to study the impacts of acquisition and assimilationof various nitrogen sources, i.e. NO3, NH4+ or NH4NO3,in combination with gaseous NH3 on plant growth and acid-basebalance in higher plants. Plants of C3 Triticum aestivum L.and C4 Zea mays L. grown with shoots in ambient air in hydroponicculture solutions with 2 mol m–3 of nitrogen source asNO3, NH4+ or NH4NO3 for 21 d and 18 d, respectively,had their shoots exposed either to 320 µg m–3 NH3or to ambient air for 7 d. Variations in plant growth (leaves,stubble and roots), and OH and H+ extrusions as wellas the relative increases in nitrogen, carbon and carboxylatewere determined. These data were computed as H+/N, H+/C, (C-A)/N,and (C-A)/C to analyse influences of different nitrogen sourceson acid-base balance in C3 Triticum aestivum and C4 Zea maysplants. Root growth in dry weight gain was significantly reduced bytreatment with 320 µg m–3 NH3 in Triticum aestivumand Zea mays growing with different N-forms, whereas leaf growthwas not significantly affected by NH3. In comparison with C3Triticum aestivum, non-fumigated C4 Zea mays had low ratiosof OH/N in NO3–3-grown plants and of H+/N in NH4+- and NH 4NO3-grown plants. Utilization of NH3 from the atmospherereduced both the OHN ratios in NO3 -grown plantsand the H+/N ratio in NH4+ - and NH4NO3 -grown plants of bothspecies. Furthermore, Zea mays had higher ratios of (C-A)/Nin NH4+ - and NH4NO3-grown plants than Triticum aestivum. Thismeans that C4 Zea mays had synthesized more organic anion perunit increase in organic N than C3 Triticum aestivum plants.Within both species, different nitrogen sources altered theratios of (C-A)/N in the order: NH4NO3>NH4+>NO3.Fumigation with NH3 increased organic acid synthesis in NO3- and NH4+ - grown plants of Triticum aestivum, whereas it decreasedorganic acid synthesis in Zea mays plants under the same conditions.Furthermore, these differences in acid-base regulation betweenC3 Triticum aestivum and C4 Zea mays plants growing with differentnitrogen sources are discussed. In nature, large concentrations of the toxic bloom-forming dinoflagellate,Gonyaulax tamarensis, are frequently observed in the vicinityof the pycnocline. In the absence of a pycnocline the organismis usually recorded near the surface, where light levels aremore advantageous for photosynthesis. In this paper we examinethe swimming behaviour of G.tamarensis when exposed to varyingdegrees of stratification and investigate whether the maintenanceof a subsurface (pycnocline) population is the result of retentionof the algae by a physical barrier or active accumulation ofthe organisms at a density interface. The study indicates thatG.tamarensis cells presented with a halocline of S<{smalltilde}6–7 (occurring over a few centimeters) cross thissalinity barrier and accumulate at the highest available photonflux density ({small tilde}100 µmol m–2 s–1).Cells exposed to a gradient of S>{small tilde}7remain atthe halocline (pfd={small tilde}40 µmol m–2 s–1).However, when light above the pycnocline is attenuated by theaddition of food colour to the medium, the cells cross a haloclineof S=10 and accumulate at the highest available photon fluxdensity. In the absence of added nutrients (inorganic N andP) the organism fails to exhibit a phototactic response. Thus,the presence of a strong halocline does not represent an inpenetrablephysical barrier for G.tamarensis and the development of pycnoclinepopulations of this organism is a function of density, lightand nutrient climate.  相似文献

SYNOPSIS. Studies are described on the adenosine triphosphatase(ATPase) properties of myosin isolated from skeletal muscleof Coryphaenoides, a benthic fish captured at 2,200 meters depth.Ca2+-ATPase and EDTA-ATPase of Coryphaenoides myosin show thesame pH dependence as ATPase of mammalian myosin; however, ratesof ATP hydrolysis by Coryphaenoides myosin are only 5–10%of rates of ATP hydrolysis by rabbit skeletal myosin. Coryphaenoidesmyosin ATPase shows a decrease from Q10 of 2.0 at 25°C toQ10 of 1.4 a t 2°C, and undergoes irreversible denaturationat temperatures above 25°C. At pH 6.8 to pH 8.5, Coryphaenoidesmyosin ATPase undergoes activation by pressure at 25°C,but at 2°C shows negligible effect of pressure at valuesbelow 3,000 psi. The kinetic data on Ca2+-ATPase indicate valuesof 11 kcal/mole for H, –7.5 kcal/mole for TS, and –5.7cc/mole for V at 25°C, pH 7.6. Comparable data at 2°Cindicate values of 5 kcal/mole for H. –13 kcal/mole forTS, and negligible V. According to the results of 25°C,Ca2+-activatkm of myosin-ATP may involve disruption of fouror five hydrophobic or polar groups, presumably due to an "opening-up"of the myosin molecule at or near the site for ATP binding.It would also appear that Coryphaenoides myosin has undergonean adaptive change in the enzyme mechanism for ATPase such thatthe rate of ATP hydrolysis is relatively insensitive to pressureand temperature under conditions encountered by the living fish.  相似文献

In Trifolium repens L. there were immediate transient depolarizationsof the membrane electropotential (Evo) when KH2PO4 was addedto phosphate-free media, but these were of the same magnitudeas the controls (K2SO4 and KCI). Furthermore, the extents ofdepolarization were the same as the expected effect of the addedK+ calculated using the Goldman equation. There was no significantdepolarization on adding H3PO4 to buffered media. Consequently,there was no evidence for a depolarization caused by phosphate.This result provides evidence that the H+–H2PO4 symportin roots of T. repens operates with a stoichiometry of 1: 1. In a group of control plants ( + P plants) and a group whichwere stressed by reducing the supply of phosphate (– Pplants), the – P plants had lower values for Evo than+P plants (– 118 mV and – 130 mV, respectively).The absence of phosphate from the measurement media also reducedEvo (mean effect = 9 mV). A significant difference in Evo between– P and + P plants persisted when phosphate was addedto – P plants. Stable carbon isotopes (13C) were determined for phytoplanktonand dissolved inorganic carbon (DIC) from Lake Apopka, a shallow,polymictic and hypereutrophic lake in Florida, USA. Bulk planktondominated by pico- and nanqanobacteria were enriched in 13(–13.1± 1.1%) as a result of assimilation of extremely 13C-richDIC (13C = 9.6 ± 3.0%). Diatoms (Aulacoseira spp.) hada 13C of –14.3 ± 0.6% that was slightly more negativethan that of small cyanobacteria. Meroplanktonic diatoms hada 13C (–13.6 ± 1.8%), similar to their planktoniccounterparts. The 13C of a colonial cyanobacterium (Microcystisincerta) was exceptionally heavy (–3.0 ± 1.0%)and attributed to localized carbon limitation. Seasonal variationin 13C of bulk plankton was small (4%) relative to reports forother lacustrine systems No difference in the 13C of bulk planktonhorn surface water between stratified and non-stratified periodswas found. No measurable changes in 13C of bulk plankton wereindicated in light and dark incubation experiments Frequentwind mixing of the water column, high DIC concentration, andconsistently high lake productivity were used to explain thetemporal and spatial isotope consistency of phytoplankton inthis lake.  相似文献

Light-limited photosynthetic carbon incorportion is expectedto be directly proportional to the scalar quantum irradiance.The proportionality constant is , where mis the maximum quantum yield (mol C Einstein–1 absorbed)and $$\stackrel{\&macr;}{{\hbox{ k }}_{\hbox{ c }}}$$ isthe mean spectral absorption coefficient (m2 mg–1 chla). Recent efforts to evaluate of in situphytoplankton photosynthesis are variously flawed. Lack of evidenceof proportionality and lack of correction of cosine to scalarirradiance are common deficiencies. Most data, as we interpretthem, indicate values in the range 0.0003 – 0.0006 mol C m2 Einstein1 abs mg1 chl a. New determinationsin lrondequoit Bay, New York, lie in this range. Most estimatesof at depth have been about 0.010 m2 mg–1chl a. Similar values are being obtained for total particulatesfrom lrondequoit Bay; whether detritus contributes significantlyis not yet known. Published data, in our view, all point tovalues of m in situ in the range 0.03–0.07 mol C Einstein–1abs. Uptake of the toxic heavy-metal, thallium, was studied in thecyanobacterium Synechococcus R-2 (PCC 7942) using clinicallyavailable 201Tl +. Thallium was found to distribute across theplasmalemma passively, and so the accumulation ratio of theion ([Tl+]i/[Tl+]o) could be used to calculate the apparentmembrane potential (­i,o) of the cells (ETI+i,o = ­i,o).The permeability of the plasmalemma to TI+ (PTI+ 1 to 5 nms–1)is higher than that of K+. Valinomycin does not increase thepermeability of TI+. Transient changes in the ­i,o of cells,because of electrogenic transport of ions, could be detectedfrom its effects upon the uptake rate of TI+. HCO3 hyperpolarizedSynechococcus cells, whereas NH+4, CH3NH+, and K+ led to depolarization.The use of TI+ as a reporter of ­i,o has some inherent limitations.Tl+ is toxic at very low concentrations (inhibitory effectsare apparent after about 6 h at concentrations as low as 1 mmolm–3). The rate of equilibration is slow (t1/25 to 20 min).Equilibration of TI+ takes about 2 h, which limits its valueas a membrane potential probe. Large amounts of TI+ bind tothe surface of the cells making the method impracticable formeasuring accumulation ratios of less than about 10 (­i,o)values smaller than about –60 mV). Cultures continuouslyexposed to Tl+ (10 mmol m–3) eventually become TI+ resistantby actively extruding TI+ (µTI+i,o= –3±0.2kJ mol–1) and so thallium cannot be used as a ­i,oprobe in such cells. Colonies of Trichodesmium spp. are conspicuous, macroscopiccomponents of the life in tropical and subtropical oceans. Thelarge size and the morphology of the colony raise questionsregarding the mechanism of carbon supply for photosynthesis.Constraints on these mechanisms may be indicated by the stablecarbon isotopic composition (13C) that reflects the balancebetween carbon supply and speciation, as well as the growthrate and colony size. The 13C of Trichodesmium off Bermuda measuredhere revealed a strong correlation between size of individualcolonies and season. The smallest colonies, 2–7 µgC colony–1, showed the lightest 13C composition (–19),increasing to asymptotic values of –12 above 7 µgC colony–1. The average 13C of the colonies was lightestimmediately after the onset of stratification in the SargassoSea, gradually increasing by 4 to heavier values during thesummer. We propose that the mass effect is due to increaseduse of HCO3 by the larger colonies, whereas the seasonalinfluence may be related to changes in irradiance and pCO2 affectingthe internal carbon cycling.  相似文献

The euryhaline charophyte Lamprothamnium papulosum (Wallr.)J. Gr. was adapted to media with decreasing salinities rangingfrom 550 to 0 mosmol kg–1. Vegetative plants grown inmedia with osmotic pressures (0) in the range of 550 to 130mosmol kg–1 maintained a constant turgor pressure () at309 + 7 mosmol kg–1. The ions K+, Na+ and Cl–, werethe predominant solutes in the vacuole. Changes in their concentrationsaccount for the variation in internal osmotic pressure (1) with,0. The divalent ions Mg2+, Ca2+ and were also present in significant amounts, but their concentrationsdid not alter with changes in, 0. In cells subjected to hypo-osmotic shock the regulation of was incomplete. The turgor pressure increased from 302 to 383mosmol kg–1. The first rapid response to the sudden decreasein 0 was a loss of K+ and Cl. In contrast to the decreasein ionic concentrations an accumulation of sucrose occurredwhich could account for the increase of . The increase in sucroseconcentration started 24 to 48 h after the downshock and reachedits highest value after 3 to 4 weeks. The sucrose concentrationin the vacuole was up to 320 mol m–3. During this timethe ionic content continued to decrease but did not counterbalancethe sucrose concentration sufficiently to regain the original. High sucrose levels accompanied by an enhanced were also observedduring the period of fructification (sexual reproduction: formationof antheridia and oogonia) in Lamprothamnium kept under conditionsof constant salinity. It is concluded that high sucrose content and elevated arecharacteristic of sexual reproduction in this charophyte. The luciferin-luciferase method was used to determine ATP extractedfrom darkmaintained and light-exposed samples of the green algaChlorella pyrenoidosa and of the blue-green alga Anacystis nidulans.A few measurements on Synechococcus lividus (a bluegreen thermophile,clone 65?C) are also reported.
  1. The light-minus-dark ATP levels (ATP) from aerobic cells ofChlorella and Anacystis were negative; however, ATP from Synechococcuswas positive. Large positive ATP was obtained in regularly grown(RG: moderate light) Chlorella treated with oligomycin; darklevels were reduced, light levels remained essentially unaffected.In high-light exposed (HLE) Chlorella, oligomycin reduced bothlight and dark ATP levels, but positive ATP was still obtained.However, in Anacystis, which has a different organization ofthylakoid membrane, oligomycin severely reduced both the lightand the dark ATP levels and the ATP remained negative.
  2. Theoligomycin (12 µM) treated Chlorella and the untreatedAnacystis and Synechococcus show the presence of cyclic photophosphorylationunder conditions in which the non-cyclic electron flow fromphotosystem II to photosystem I is blocked by 10 µM 3-(3,4-dichlorophenyl)-l,l-dimethylurea(DCMU), or not allowed to operate by the absence of CO2. Cyclicphotophosphorylation ranged from 10–30% of the maximumATP in RG, to 40–50% in HLE Chlorella. In RG Chlorella,cyclic and non-cyclic (in the absence of DCMU) photophosphorylation(ATP) saturate at about 103 ergs cm–2 sec–1 and104 ergs cm–2 sec–1 and 104 ergs cm–2 sec–1red (>640 nm) light, respectively; a lag was observed inthe light curve.
  3. In Chlorella, the addition of the photosystemI electron acceptormethyl viologen (MV; 1 mM) increased ATPby twofold. Furtheraddition of DCMU (25 µm) reduced thisto the level observedwith DCMU alone. If 1 mM reduced dichlorophenolindophenol orphenazine methosulphate (DCPIPH2 or PMSH2, respectively)wasadded along with DCMU, the ATP level was 30–40% ofthecontrol. Further addition of MV increased the JATP to be70–80%of that of the control. These and other resultsconfirm thepresence of both non-cyclic and cyclic photophosphorylationin vivo, the former predominating in Chlorella, and the latterin Anacystis and Synechococcus.

The vacuolar pH (pHv) and the cytoplasmic pH (pHc) of the marinegiant-celled green alga Chaetomorpha darwinii were measuredby pH microelectrode techniques on extracted vacuolar sap, andby the [I4C]DMO distribution method respectively. Equilibrationof DMO occurred with a half-time of about 2 h, with an apparentPDMO of 3.6 x 10–5 cm s–1, but the vacuolar concentrationof free, undissociated DMO was always less than the externalconcentration. The explanation offered for freshwater giant-celledalgae of net DMO leakage across the plasmalemma cannotapply to Chaetomorpha darwinii, since electrically-driven DMOexit from the cytoplasm should be similar across the plasmalemmaand the tonoplast in these cells with large, vacuole-positivepotential differences across the tonoplast. pHc was accordinglycomputed assuming either tonoplast or plasmalemma equilibrationof DMO, with correction for DMO metabolism within the cell.pHc was 8.0–8.3 in the light in artificial seawater (pHoabout 8.0), was some 0.5 units lower in the dark, and was slightlylower with an external pH of 7. Vacuolar pH was 6.5–6.9,without consistent effects of illumination or of external pHof 7 rather than 8. While µH+ at the tonoplast was similarto that in giant-celled freshwater algae (although with a greatercontribution from relative to pH), µH+ at the plasmalemmawas less than 8 kJ mol–1, i.e. less than one-third ofthe value in freshwater green algae. µNa+ was some 13kJ mol–1 at the plasmalemma. The possibility that theprimary active transport process at the plasmalemma of Chaetomorphadarwinii (and certain other marine algae) is Na+ efflux ratherthan H+ efflux is discussed.  相似文献

By analysing the relationship between inverse water potential(–1), and relative water content (RWC) measured on leavesof roses (Rosa hybrida cv. Sonia), grown soilless, it was foundthat a non-linear (NL) model was better suited than a linearmodel to reproduce values observed in the non-turgid region.To explain this apparent curvature, it is assumed that a reductionof the non-osmotic water fraction (Ap) takes place when decreases.Osmotic potentials () measured on fresh and frozen leaf discstend to support this hypothesis. A method for exploiting PVcurves, which takes into account the variation of Ap, is described.It delivers values for the turgor pressure (p), the relativeosmotic water content, and the mean bulk volumetric elasticitycoefficient, lower than those given by the linear model. Onthe other hand, it gives higher estimates for Ap and for . Whenapplying the traditional model to obtain estimates for waterrelations characteristics of rose leaves, and comparing resultsfrom two distinct salinity treatments (electrical conductivitiesof 1·8 mS cm–1 and 3·8 mS cm–1, respectively),one deduces a significant reduction of at turgor-loss in thehigh salinity treatment. The NL method is, in addition, ablesimultaneously to reveal a reduction of and a significant increasein p at RWC=100% this proves that soilless–grown roseplants are able to osmoregulate when subjected to a constantand relatively high degree of salinity. The effects of -hydroxy-2-pyridinemethanesulphonic acid (-HPMS)upon net photosynthesis (Pn, the CO2 compensation point (),post-lower illumination burst of CO2 (PLIB) and post-lower temperatureburst of CO2 (PLTB) in detached rye (Secale cereale L.) leaveswere investigated. At low concentrations ( 0.5 mol m–3),-HPMS initially stimulated Pn and decreased the magnitude ofboth PLIB and PLTB. The decreased at all concentrations of-HPMS (0.05–5.0 mol m–3. The effects of -HPMS onPn and were time-dependent and, after a few minutes, the Pnwas inhibited while values increased considerably. At a higherconcentration (5.0 mol m –3), the transient effects of-HPMS were shorter () or not observed at all (Pn. Both PLIBand PLTB, when expressed in relation to Pn, increased at higherlevels of this compound. Similar data with respect to the effectsof -HPMS on PLIB and PLTB were found for leaves of dandelion(Taraxacum officinale L.). The results suggest that -HPMS may stimulate Pn by inhibitingphotorespiration, as originally suggested by Zelitch (1966),but only at low concentrations and over a short time span. Thedecrease of PLIB and PLTB values at low -HPMS levels is consistentwith these processes being a residual activity of the glycolatepathway. The stable carbon isotope composition of particulate organicmatter expressed as 13C was measured in cultures of 13 speciesof marine microalgae in different phylogenetic groups. The effectsof salinity variations and changes in photoperiod were alsoassayed for three of them (i.e. Skeletonema costatum, Amphidiniumopercularum and Isochrysis galbana); the effect of nature ofnitrogen supply (nitrate. ammonium) was studied for one (S.costatum).These environmental parameters were chosen because of theirvariability in the ocean and their possible effects on 13C valuesof phytoplankton organic carbon. Batch culture conditions andsampling time after inoculum were strongly controlled in orderto provide cells in good physiological state which were comparablefrom one culture to the other. In the same way, sampling waslimited to the first 2 days of exponential growth, in orderto avoid a possible dissolved inorganic carbon (DIC) limitation.Carboxylase activities [of the enzyme ribulose 1,5-bisphosphatecarboxylase oxygenase (Rubisco), and the three ß carboxylases:phosphoenolpyruvate carboxylase (PEPC), phosphoenolpyruvatecarboxykinase (PEPCK) and pyruvate carboxylase (PC)] and totalchlorophyll a concentrations were assayed simultaneously. The13C values observed were between –30.2 and –12.7i.e. comparable to those observed in the world's oceans. Theisotopic composition of phytoplankton organic carbon was shownto be under the influence of the parameters tested but 13C variationsare specific to the species considered. The nature of ßcarboxylase found in each species, or systematic position, couldnot be linked to the isotopic composition of organic carbon.No linear or single correlation between 13C variations and environmentalmodifications were observed and there is no evidence for a simpleand universal relation between 13C of phytoplankters and theirenvironment. In monospecific cultures as in the field, 13C fractionationby Rubisco (and eventually by PEPCK) may be counterbalancedby other mechanisms.  相似文献

The effects of transpiration rate on the vertical gradientsof leaf and stem xylem water potential ( and ) were examinedusing hydroponic sunflower plants. Transpiration was variedby stepwise alterations of environmental conditions. The gradientsof and were relatively small (2.3 and 0.8 x 105 Pa m–1)when transpiration rates approached zero, but increased sharplyto 5.4 and 2.3 x 105 Pa m–1 as transpiration increased.However, the gradients were independent of transpiration ratesabove 0.4 g dm–2 h–1 owing to variability of theplant resistance. The gradients of I were usually less thanhalf those of I. 1 in individual leaves remained constant over a wide range oftranspiration rates (0.4—2.4 g dm–2 h–1) andeach leaf possessed a characteristic plateau value related toits elevation. I responded similarly but was approximately 2.0x 105 Pa higher than I at the same elevation. Identical resultswere obtained regardless of the procedure employed to vary transpiration. The drop in water potential between stem and leaf implies thatthe leaf resistance is appreciable. This was confirmed usingrapidly transpiring excised leaves freely supplied with water.I increased by 2.0–2.5 x 105 Pa following removal of theroot resistance but remained 2 x 105 Pa lower than similar excisedleaves in darkness. Furthermore, I in excised leaves remainedconstant over a wide range of transporting rates, demonstratingthat the leaf resistance is also variable.
