Direct evidence obtained by means of the technique of pulse radiolysis-kinetic spectrometry, with measurements in the time range 10−6 to 1 s, is presented that, consequent upon reaction of a single H-atom with a single molecule of ferricytochrome c, a reducing equivalent is transmitted via the protein structure to the ferriheme moiety. Such transmission accounts for at least 70% of the total reduction of the ferri to the ferro state of cytochrome c. The remainder of the total reduction takes place without stages resolvable on the time scale of these experiments. Reduction brought about by H atoms appears to follow a different course than reduction by hydrated electrons. In the latter case, intramolecular transmission of reducing equivalents could not be demonstrated (Lichtin, N. N., Shafferman, A. and Stein, G. (1973) Biochim. Biophys. Acta 314, 117–135).

Not every H-atom reacts with ferricytochrome c at a site which results in conversion of the Fe(III) state to the Fe(II) state. Approximately half of reacting H-atoms do not produce reduction.

The following second order rate constants have been determined in solutions of low ionic strength at 20±2 °C: k[H+ferricytochrome c] = (1.0±0.2) · 1010 M−1 · s−1 at pH 3.0 and 6.7; k[H+ferrocytochrome c] = (1.3±0.2) · 1010 M−1 · s−1 at pH 3.0; k[eaq + ferrocytochrome c] = (1.9±0.4) · 1010 M−1 · s−1 at pH 6.7.  相似文献

The rate of electron transfer between reduced cytochrome ƒ and plastocyanin (both purified from parsley) has been measured as k = 3.6 · 107 M−1 · s−1, at 298 °K and pH 7.0, with activation parameters ΔH = 44 kJ · mole−1 and ΔS = +46 J · mole−1 · °K−1. Replacement of cytochrome ƒ with red algal cytochrome c-553, Pseudomonas cytochrome c-551 and mammalian cytochrome c gave rates at least 30 times slower: k = 5 · 105, 7.5 · 105 and 1.0 · 106 M−1 · s−1, respectively.

Similar measurements made with azurin instead of plastocyanin gave k = 6 · 106 and approx. 2 · 107 M−1 · s−1 for reaction of reduced azurin with cytochrome ƒ and algal cytochrome respectively.

Rate constants of 115 and 80 M−1 · s−1 were found for reduction of plastocyanin by ascorbate and hydroquinone at 298 °K and pH 7.0. The rate constants for the oxidation of plastocyanin, cytochrome ƒ, Pseudomonas cytochrome c-551 and red algal cytochrome c-553 by ferricyanide were found to be between 3 · 104 and 8 · 104 M−1 · s−1.

The results are discussed in relation to photosynthetic electron transport.  相似文献

The reaction between a cytochrome oxidase from Pseudomonas aeruginosa and oxygen has been studied by a rapid mixing technique. The data indicate that the heme d1 moiety of the ascorbate-reduced enzyme is oxidized faster than the heme c component. The oxidation of heme d1 is accurately second order with respect to oxygen and has a rate constant of 5.7 · 104 M−1 · s−1 at 20 °C. The oxidation of the heme c has a first-order rate constant of about 8 s−1 at infinite concentration of O2. The results indicate that the rate-limiting step is the internal transfer of electrons from heme c to heme d1. These more rapid reactions are followed by more complicated but smaller absorbance changes whose origin is still not clear.

The reaction of ascorbate-reduced oxidase with CO has also been studied and is second order with a rate constant of 1.8 · 104 M−1 · s−1. The initial reaction with CO is followed by a slower reaction of significantly less magnitude. The equilibrium constant for the reaction with CO, calculated as a dissociation constant from titrimetric experiments with dithionite-reduced oxidase, is about 2.3 · 10−6 M. From these data a rate constant of 0.041 s−1 can be calculated for the dissociation of CO from the enzyme.  相似文献

1. 1. The superoxide anion radical (O2) reacts with ferricytochrome c to form ferrocytochrome c. No intermediate complexes are observable. No reaction could be detected between O2 and ferrocytochrome c.

2. 2. At 20 °C the rate constant for the reaction at pH 4.7 to 6.7 is 1.4 · 106 M−1 · s−1 and as the pH increases above 6.7 the rate constant steadily decreases. The dependence on pH is the same for tuna heart and horse heart cytochrome c. No reaction could be demonstrated between O2 and the form of cytochrome c which exists above pH ≈ 9.2. The dependence of the rate constant on pH can be explained if cytochrome c has pKs of 7.45 and 9.2, and O2 reacts with the form present below pH 7.45 with k = 1.4 · 106 M−1 · s−1, the form above pH 7.45 with k = 3.0 · 105 M−1 · s−1, and the form present above pH 9.2 with k = 0.

3. 3. The reaction has an activation energy of 20 kJ mol−1 and an enthalpy of activation at 25 °C of 18 kJ mol−1 both above and below pH 7.45. It is suggested that O2 may reduce cytochrome c through a track composed of aromatic amino acids, and that little protein rearrangement is required for the formation of the activated complex.

4. 4. No reduction of ferricytochrome c by HO2 radicals could be demonstrated at pH 1.2–6.2 but at pH 5.3, HO2 radicals oxidize ferrocytochrome c with a rate constant of about 5 · 105–5 · 106 M−1 · s−1

The reduction of acetylated, fully succinylated and dicarboxymethyl horse cytochromes c by the radicals CH3CH(OH), CO2, O2, and eaq′ and the oxidation of the reduced cytochrome c derivatives by Fe(CN)3−6 were studied using the pulse radiolysis technique. Many of the reactions were also examined as a function of ionic strength. By obtaining rate constants for the reactions of differently charged small molecules redox agents with the differently charged cytochrome c derivatives at both zero ionic strength and infinite ionic strength, electrostatic and conformational contributions to the electron transfer mechanism were effectively partitioned from each other in some cases. In regard to cytochrome c electron transfer mechanism, the results, especially those for which conformational influences predominate, are supportive of the electron being transferred in the heme edge region.  相似文献

Horse-heart ferrocytochrome c has been labeled with N-(2,2,5,5-tetramethyl-3-pyrrolidinyl-1-oxyl) iodoacetamide at methionine-65. The paramagnetic resonance spectrum of labeled ferricytochrome c indicates a weak immobilization of the radical (τc = 9.3·10−10 sec) which becomes stronger upon binding of labeled cytochrome c to cytochrome c-depleted mitochondrial membranes (τc = 3.3·10−9 sec). The hyperfine coupling constant remains, however, unchanged (16.7 ± 0.1 gauss) indicating that the cytochrome c binding site is highly polar. The region where cytochrome c is bound to the membrane is insensitive to large variations of medium viscosity.  相似文献

1. The reduction of cytochrome c oxidase by hydrated electrons was studied in the absence and presence of cytochrome c.

2. Hydrated electrons do not readily reduce the heme of cytochrome c oxidase. This observation supports our previous conclusion that heme a is not directly exposed to the solvent.

3. In a mixture of cytochrome c and cytochrome c oxidase, cytochrome c is first reduced by hydrated electrons (k = 4 · 1010 M−1 · s−1 at 22 °C and pH 7.2) after which it transfers electrons to cytochrome c oxidase with a rate constant of 6 · 107 M−1 · s−1 at 22 °C and pH 7.2.

4. It was found that two equivalents of cytochrome c are oxidized initially per equivalent of heme a reduced, showing that one electron is accepted by a second electron acceptor, probably one of the copper atoms of cytochrome c oxidase.

5. After the initial reduction, redistribution of electrons takes place until an equilibrium is reached similar to that found in redox experiments of Tiesjema, R. H., Muijsers, A. O. and Van Gelder, B. F. (1973) Biochim. Biophys. Acta 305, 19–28.  相似文献

The interaction of horse ferricytochrome c with the reagents [Fe(EDTA)(H2O)] and [Cr(CN)6]3− were studied at pH 7 and 25°C by 1H-NMR spectroscopy. Two binding regions near to the heme crevice of cytochrome c were identified. Both regions bound both reagents but they exhibited different selectivities.

The relevance of this finding to the electron-transfer function of cytochrome c is discussed.  相似文献

The reduction of ferricytochrome c by O2 and CO2 was studied in the pH range 6.6–9.2 and Arrhenius as well as Eyring parameters were derived from the rate constants and their temperature dependence. Ionic effects on the rate indicate that the redox process proceeds through a multiply-positively charged interaction site on cytochrome c. It is shown that the reaction with O2 and correspondingly with O2 of ferrocytochrome c) is by a factor of approx. 103 slower than warranted by factors such as redox potential. Evidence is adduced to support the view that this slowness is connected with the role of water in the interaction between O2/O2 and ferri-ferrocytochrome c in the positively charged interaction site on cytochrome c in which water molecules are specifically involved in maintaining the local structure of cytochrome c and participate in the process of electron equivalent transfer.  相似文献

1. Fluoride is a mixed-type inhibitor of the cytochrome c oxidase activity with a Ki for the free enzyme of 10 mM and a Ki for the cytochrome c-complexed enzyme of 35 mM.

2. Fluoride shifts the γ-band of the enzyme from 423 to 421 nm and the -band from 597 to 598 nm. The difference spectrum (oxidized enzyme in the presence of fluoride minus oxidized enzyme) has peaks at 400, 453, 482, 605 and 638 nm and troughs at 430, 520, 552 and 674 nm. The changes in absorbance are small (about 3% at absorbance maxima) with respect to those of other hemoproteins.

3. On addition of fluoride to isolated cytochrome c oxidase 3 reactions can be distinguished: (I) a bimolecular binding reaction (Kon = 4 M−1 · s−1 and koff = 2.9 · 10−2s−1 at 25 °C, pH 7.4) contributing at 638 nm and 430 nm; (II) a first-order reaction (k = 2.4 · 10−2) s−1 at 22 °C, pH 7.2) visible mainly at 430 nm and (III) a very slow reaction with a half-time in the order of 10 min.

4. The spectroscopic dissociation constants for the fluoride binding, determined from Hill plots using the absorbance changes at 638 and 430 nm, are similar (7 and 10 mM, respectively, at 22 °C, pH 7.2).

5. A mechanism for the reaction is discussed in which the bimolecular binding reaction is followed by a conformational change of the enzyme-fluoride complex.  相似文献

1. Rate constants for reduction of paraquat ion (1,1′-dimethyl-4,4′-bipyridy-lium, PQ2+) to paraquat radical (PQ+·) by eaq and CO2· have been measured by pulse radiolysis. Reduction by eaq is diffusion controlled (k = 8.4·1010 M−1·s−1) and reduction by CO2· is also very fast k = 1.5·1010 M−1·s−1).

2. The reaction of paraquat radical with oxygen has been analysed to give rate constants of 7.7·108 M−1·s−1 and 6.5·108 M−1·s−1 for the reactions of paraquat radical with O2 and O2·, respectively. The similarity in these rate constants is in marked contrast to the difference in redox potentials of O2 and O2· (− 0.59 V and + 1.12 V, respectively).

3. These rate constants, together with that for the self-reaction of O2·, have been used to calculate the steady-state concentration of O2· under conditions thought to apply at the site of reduction of paraquat in the plant cell. On the basis of these calculations the decay of O2· appears to be governed almost entirely by its self-reaction, and the concentration 5 μm away from the thylakoid is still 90% of that at the thylakoid itself. Thus, O2· persists long enough to diffuse as far as the chloroplast envelope and tonoplast, which are the first structures to be damaged by paraquat treatment. O2· is therefore sufficiently long-lived to be a candidate for the phytotoxic product formed by paraquat in plants.  相似文献

Submitochondrial particles isolated from Tetrahymena pyriformis contain essentially the same redox carriers as those present in parental mitochondria: at pH 7.2 and 22 °C there are two b-type pigments with half-reduction potentials of −0.04 and −0.17 V, a c-type cytochrome with a half reduction potential of 0.215 V, and a two-component cytochrome a2 with Em7.2 of 0.245 and 0.345 V.

EPR spectra of the aerobic submitochondrial particles in the absence of substrate show the presence of low spin ferric hemes with g values at 3.4 and 3.0, a high spin ferric heme with g = 6, and a g = 2.0 signal characteristic of oxidized copper. In the reduced submitochondrial particles signals of various iron-sulfur centers are observed.

Cytochrome c553 is lost from mitochondria during preparation of the submitochondrial particles. The partially purified cytochrome c553 is a negatively charged protein at neutral pH with an Em7.2 of 0.25 V which binds to the cytochrome c-depleted Tetrahymena mitochondria in the amount of 0.5 nmol/mg protein with a KD of 0.8 · 10−6 M. Reduced cytochrome c553 serves as an efficient substrate in the reaction with its own oxidase. The EPR spectrum of the partially purified cytochrome c553 shows the presence of a low spin ferric heme with the dominant resonance signal at g = 3.28.

A pigment with an absorption maximum at 560 nm can be solubilized from the Tetrahymena cells with butanol. This pigments has a molecular weight of approx. 18 000, and Em7.2 of −0.17 V and exhibits a high spin ferric heme signal at g = 6.  相似文献

1. Cyanide causes a slow disappearance of the oxidized band (648 nm) of cytochrome d in particles of Azotobacter vinelandii and inhibits the appearance of the reduced band (631 nm). No effect of cyanide is found on the reduced band of cytochrome d.

2. The kinetics of the disappearance of the 648-nm band of cytochrome d with excess cyanide deviates from first-order kinetics at lower temperatures (22 °C) indicating that at least two conformations of the enzyme are involved. At higher temperatures (32 °C) the observed kinetics of the cyanide reaction are first order with a kon = 0.7 M−1·s−1 and with an estimated koff of approximately 5·10−5 s−1.

3. The value of the koff (7·10−4−14·10−4 s−1 at 32 °C) determined from the rate of reduction of cyanocytochrome d by Na2S2O4 or NADH is one order of magnitude larger than the koff value found when the enzyme is in its oxidized state.

4. No effect of cyanide is found on the spectrum of cytochrome a1.  相似文献

The reaction between a cytochrome oxidase and a copper protein from Pseudomonas aeruginosa has been studied by a rapid mixing technique. The data support the view that a complex is formed rapidly between the two proteins and is followed by a transfer of electrons in either direction. Reduced copper protein donates an electron to the heme c moiety of the oxidase with an apparent first-order rate constant of about 30 s−1 while the transfer in the reverse direction proceeds with a constant of about 120 s−1. The reaction between the copper protein and the heme c of the oxidase is followed by a much slower internal reaction involving electron transfer between the heme c and heme d.

The kinetic data have been analyzed in terms of the thermodynamics of the interactions. This analysis indicates that the copper protein has a ΔE of about 0.038 V more positive than the heme c component, a value that compares favorably with that of 0.040 V obtained by equilibrium methods. The value of ΔE obtained by the kinetic method for the internal reaction is less precise but is reasonably close to that of 0.070 V determined by an equilibrium technique.  相似文献

In this paper a number of experiments with the purple bacteria Rhodospirillum rubrum and Rhodopseudomonas capsulata is described in which the total fluorescence yield and/or the total fraction of reaction centers closed after a picosecond laser pulse were measured as a function of the pulse intensity. The conditions were such that the reaction centers were either all in the open or all in the closed state before the pulse arrived. These experiments are analysed using the theoretical formalism discussed in the preceding paper (Den Hollander, W.T.F., Bakker J.G.C., and Van Grondelle, R., Biochim. Biophys. Acta 725, 492–507). From the experimental results the number of connected photosynthetic units, λ, the rate of energy transfer between neighboring antenna molecules, kh, and the rate of trapping by an open reaction center, kot, can be estimated. For R. rubrum it is found that λ = 14−17, kh = (1−2)·1012 s−1 and kot = (4−6)·1011 s−1, for Rps. capsulata λ ≈ 30, kh ≈ 4·1011 s−1 and kot ≈ 3·1011 s−1. The findings are discussed in terms of current models for the structure of the antenna and the kinetic properties of the decay processes occurring in these purple bacteria.  相似文献

Rapid reactions occur between [OsVI(tpy)(Cl)2(N)]X (X = PF6, Cl, tpy = 2,2′:6′,2″-terpyridine) and aryl or alkyl phosphi nes (PPh3, PPh2Me, PPhMe2, PMe3 and PEt3) in CH2Cl2 or CH3CN to give [OsIV(tpy)(Cl)2(NPPh3)]+ and its analogs. The reaction between trans-[OsVI(tpy)(Cl)2(N)]+ and PPh3 in CH3CN occurs with a 1:1 stoichiometry and a rate law first order in both PPh3 and OsVI with k(CH3CN, 25°C) = 1.36 ± 0.08 × 104 M s−1. The products are best formulated as paramagnetic d4 phosphoraniminato complexes of OsIV based on a room temperature magnetic moment of 1.8 μB for trans-[OsIV(tpy)(Cl)2(NPPh3)](PF6), contact shifted 1H NMR spectra and UV-Vis and near-IR spectra. In the crystal structures of trans-[OsIV(tpy)(Cl)2( NPPh3)](PF6)·CH3CN (monoclinic, P21/n with a = 13.384(5) Å, b = 15.222(7) Å, c = 17.717(6) Å, β = 103.10(3)°, V = 3516(2) Å3, Z = 4, Rw = 3.40, Rw = 3.50) and cis-[OsIV(tpy)(Cl)2(NPPh2Me)]-(PF6)·CH3CN (monoclinic, P21/c, with a = 10.6348(2) Å, b = 15.146(9) ÅA, c = 20.876(6) Å, β = 97.47(1)°, V = 3334(2) Å3, Z = 4, R = 4.00, Rw = 4.90), the long Os-N(P) bond lengths (2.093(5) and 2.061(6) Å), acute Os-N-P angles (132.4(3) and 132.2(4)°), and absence of a significant structural trans effect rule out significant Os-N multiple bonding. From cyclic voltammetric measurements, chemically reversible OsV/IV and OsIV/III couples occur for trans-[OsIV(tpy)(Cl)2(NPPh3)](PF6) in CH3CN at +0.92 V (OsV/IV) and −0.27 V (OsIV/III) versus SSCE. Chemical or electrochemical reduction of trans-[OsIV(tpy)(Cl)2(NPPh3)](PF6) gives isolable trans-OsIII(tpy)(Cl)2(NPPh3). One-electron oxidation to OsV followed by intermolecular disproportionation and PPh3 group transfer gives [OsVI(tpy)Cl2(N)]+, [OSIII(tpy)(Cl)2(CH3CN)]+ and [Ph3=N=PPh3]+ (PPN+). trans-[OsIV(tpy)(Cl)2(NPPh3)](PF6) undergoes reaction with a second phosphine under reflux to give PPN+ derivatives and OsII(tpy)(Cl)2(CH3CN) in CH3CN or OsII(tpy)(Cl)2(PR3) in CH2Cl2. This demonstrates that the OsVI nitrido complex can undergo a net four-electron change by a combination of atom and group transfers.  相似文献

1. 1. Cyanide inhibits the catalytic activity of cytochrome aa3 in both polarographic and spectrophotometric assay systems with an apparent velocity constant of 4·103 M−1·s−1 and a Ki that varies from 0.1 to 1.0 μM at 22 °C, pH 7·3.

2. 2. When cyanide is added to the ascorbate-cytochrome c-cytochromeaa3−O2 system a biphasic reduction of cytochrome c occurs corresponding to an initial Ki of 0.8 μM and a final Ki of about 0.1 μM for the cytochrome aa3−cyanide reaction.

3. 3. The inhibited species (a2+a33+HCN) is formed when a2+a33+ reacts with HCN, when a2+a32+HCN reacts with oxygen, or when a3+a33+HCN (cyano-cytochrome aa3) is reduced. Cyanide dissociates from a2+a33+HCN at a rate of 2·10−3 s−1 at 22 °C, pH 7.3.

4. 4. The results are interpreted in terms of a scheme in which one mole of cyanide binds more tightly and more rapidly to a2+a33+ than to a3+a33+.

Electron self-exchange in solutions of the ‘blue’ copper protein plastocyanin is catalysed by the redox-inert multivalent cations Mg2+ or Co(NH3)3+6. Measurements of specific 1H-NMR line broadening with 50% reduced solutions in the presence of these cations show that electron exchange proceeds through encounters of cation-protein complexes which dissociate at high ionic strength. In the presence of 8mM (5 equivalents/total protein) Co(NH3)3+6, with 10 mM cacodylate (pH*6.0) as background electrolyte, the bimolecular rate constant at 25°C is 7 × 104 M−1·s−1. For comparison, the ‘electrostatically screened’ rate constant measured in 0.1 M KCl in the absence of added multivalent cations is ˜ 4 × 103 M1·s−1.

The ester cleavage of R- and S-isomers N-CBZ-leucine p-nitrophenyl ester intermolecularly catalyzed by R- (a) and S-stereoisomers (b) of the Pd(II) metallacycle [Pd(C6H4C*HMeNMe2)Cl(py)] (3) follows the rate expression kobs = ko + kcat [3], where the rate constants kcat equal 25.8 ± 0.4 and 7.6 ± 0.5 dm3 mol−1 s−1 for the S- and R-ester, respectively, in the case of 3a, but are 5.7 ± 0.6 and 26.7 ± 0.5 dm3 mol−1 s−1 for the S- and R-ester, respectively, in the case of 3b (pH 6.23 and 25°C). Thus, the best catalysis occurs when the asymmetric carbons of the leucine ester and Pd(II) complex are R and S, or S and R configured, respectively. Molecular modeling suggests that the stereoselection results from the spatial interaction between the CH2CHMe2 radical of the ester and the -methyl group of 3. A hydrophobic/stacking contact between the leaving 4-nitrophenolate and the coordinated pyridine also seems to play a role. Less efficient intramolecular enantioselection was observed for the hydrolysis of N-t-BOC-S-metthionine p-nitrophenyl ester with R- and S-enantiomers of [Pd(C6H4C*HMeNMe2)Cl] coordinated to sulfur.  相似文献

Reaction center particles isolated from carotenoidless mutant Rhodopseudomonas spheroides were studied with the aim of determining the pigment composition and the molar extinction coefficients.

Two independent sets of measurements using a variety of methods show that a sample with A800 nm = 1.00 contains 20.8 ± 0.8 μM tetrapyrrole and that the ratio of bacteriochlorophyll to bacteriopheophytin is 2:1.

Measurements were made of the absorption changes attending the oxidation of cytochrome c coupled to reduction of the photooxidized primary electron donor in reaction centers, using laser flash excitation. The ratio of the absorption change at 865 nm (due to the bleaching of P870) to that at 550 nm (oxidation of cytochrome) was found to be 5.77.

These results, combined with other data, yield a pigment composition of 4 bacteriochlorophyll and 2 bacteriopheophytin molecules in a reaction center. Based on this choice, extinction coefficients are determined for the 802- and 865-nm bands: 802 nm = 288 (± 14) mM−1 · cm−1 and 865 nm = 128 (± 6) mM−1 · cm−1. For reversible bleaching of the 865-nm band, Δred - ox865nm = 112 (± 6) mM−1 · cm−1 (referred to the molarity of reaction centers). Earlier reported values of photochemical quantum efficiency are recomputed, and the revised values are shown to be compatible with those obtained from measurements of fluorescence transients.  相似文献
