ABSTRACT. Amber‐preserved shells of testate amoebae often provide as many diagnostic features as the tests of modern taxa. Most of these well‐preserved microfossils are morphologically assignable to modern species indicating either evolutionary stasis or convergent evolution. Here we describe two Lower Cretaceous testate amoebae that are clearly distinguishable from modern species. Centropyxis perforata n. sp. and Leptochlamys galippei n. sp. possessed perforate shells that were previously unknown in these genera. They are preserved in highly fossiliferous amber pieces from the Upper Albian (ca. 100 million years old) of Archingeay/Les Nouillers (Charente‐Maritime, southwestern France). Syninclusions of soil and litter dwelling arthropods and microorganisms indicate a limnetic‐terrestrial microhabitat at the floor of a coastal conifer forest.  相似文献

In annual grasslands that experience a mediterranean-type climate, the synchrony between plant senescence and peak solar radiation over summer results in high litter sun exposure. We examined the decomposition of both shaded and sun-exposed litter over summer and inferred the effects of photodegradation from changes in mass loss and litter chemistry. The carry-over effects of summer litter exposure on wet season decomposition were also assessed, and the attenuation of photodegradation with litter layer thickness was used to estimate the proportion of grass litter lignin susceptible to photodegradation under different treatments of a factorial global change experiment. Over summer, mass loss from grass and forb litter exposed to ambient sunlight ranged from 8% to 10%, whereas lignin decreased in grass litter by approximately 20%. After one year of decomposition, mass losses from grass leaves exposed to sunlight over summer were more than double the mass losses from summer-shaded leaves. When shade litter layer thickness was varied, mass losses over summer for all treatments were also approximately 8%; however, lignin decreased significantly only in the low shade treatments (0–64 g m−2 of shade litter). Aboveground production of annual grasses nearly quadrupled in response to the combined effects of N addition, elevated atmospheric CO2, increased precipitation and warming. The estimated proportion of grass litter lignin experiencing full photodegradation ranged from 100% under ambient conditions to 31–62% in plots receiving the combined global change treatments. These results reveal an important role of sun exposure over summer in accelerating litter decomposition in these grasslands and provide evidence that future changes in the quantity of litter deposition may modulate the influence of photodegradation integrated across the litter layer.  相似文献

Using Scots pine seedlings as experimental hosts, the general reactions and recognition patterns of living conifer tissues challenged in vitro with either parasitic, saprotrophic, epiphytic or mutualistic microbial species were investigated. Three distinct reaction and recognition patterns were observed. In one, a necrosis type of browning reaction occurred that restricted vascular penetration of the seedlings when inoculated with a saprotroph (Marasmius androsaceus) or a mycorrhizal-associated bacterium (Pseudomonas fluorescens). The second pattern, a kind of necrosis in which the host defence system was ultimately ineffective, was typically observed in seedlings challenged with the S or P types of the root rot fungus (Heterobasidion annosum), the fine root parasite (Fusarium avenaceum) or the saprotrophic wood decay fungus (Phlebia gigantea). The third type, described as non-recognition, was observed on seedlings showing no necrosis and was particularly common with seedlings inoculated with either mycorrhizal fungi (Suillus granulatus, Piloderma croceum) or saprotrophs (Trichoderma aureoviride, Coriolus versicolor), or an unidentified rod-shaped bacterial isolate. Since necrosis is often linked to rapid accumulation of H2O2, increases in peroxidase activity were determined. Only seedlings challenged with the S-type of H. annosum had a systemic increase in peroxidase levels of needles, shoot and roots. Similarly, only H. annosum, F. avenaceum, P. gigantea and the Pseudomonas bacterium caused a two- to four-fold increase in the peroxidase level of inoculated roots, results suggesting that a relationship exists between the degree of necrosis and the ratio of peroxidase induction. But the higher levels of peroxidase activity was not correlated with increased resistance to disease development.  相似文献

Abstract The distribution of soil microorganisms is generally believed to be patchy and to reflect habitat heterogeneity. Despite this general rule, the amount of existing data on species distribution patterns is scarce. Testate amoebae (Protozoa; Rhizopoda) are an important component of soil microbial communities and are increasingly used in ecological and paleoecological studies of Sphagnum-dominated peatlands, but data on the spatial structure of communities are completely lacking. This is an important aspect since quantitative models used for paleoecological reconstruction and monitoring are based on species assemblages. We explored the distribution patterns of testate amoebae distribution in a macroscopically homogeneous Sphagnum carpet, down to a scale of several centimeters. Distributions maps of the species and spatially constrained sample groups were produced. Multivariate and individual spatial autocorrelations were calculated. The importance of spatial structure was quantified by canonical correspondence analysis. Our ultimate goal is to find the finest resolution of environmental monitoring using testate amoebae. The distribution patterns differed among species, resulting in a complex spatial structure of the species assemblage in a whole. Spatial structure accounted for 36% of the total variation of species abundance in a canonical correspondence analysis constrained by spatial variables. This structure was partly correlated to altitude (microtopography) at a very fine scale. These results confirmed the existence of significant broad- and fine-scale spatial structures within testate amoebae communities that could in part be interpreted as effects of ecological gradients. This shows that, on a surface area of 0.25 m2, ecological conditions which look uniform from a macroscopic point of view are not perceived as such by Sphagnum-inhabiting organisms. Therefore, testate amoebae could prove very useful to monitor fine-scale ecological processes or disturbances. Studies of the species' spatial distribution patterns in combination with autoecological studies are needed and should be included in the toolbox of biomonitoring itself.  相似文献

In this study, spatial concordance among microcrustaceans, rotifers and testate amoebae was examined in a small pond in Central Brazil. The main goal was to test how consistently different taxonomic groups ordinate 9 sites distributed along the pond's main axis. Microcrustaceans were more abundant during the dry season, characterized by waters with high transparency and dissolved oxygen concentration. An increase in species richness and densities of rotifers and testate amoebae was detected during rainy season, with low values of Secchi disk and concentrations of dissolved oxygen. However, the ordination patterns generated by these groups were different. Concordant patterns across sites were found only between cladocerans and copepods. These results suggest that great care should be taken when ordination patterns based on some taxonomic groups are extrapolated to other groups for detecting environmental changes. The ecology of peatland testate amoebae is well studied along broad gradient from very wet (pool) to dry (hummock) micro-sites where testate amoebae are often found to respond primarily to the depth to water table (DWT). Much less is known on their responses to finer-scale gradients, and nothing is known of their possible response to phenolic compounds, which play a key role in carbon storage in peatlands. We studied the vertical (0–3, 3–6, and 6–9 cm sampling depths) micro-distribution patterns of testate amoebae in the same microhabitat (Sphagnum fallax lawn) along a narrow ecological gradient between a poor fen with an almost flat and homogeneous Sphagnum carpet (fen) and a “young bog” (bog) with more marked micro-topography and mosaic of poor fen and bog vegetation. We analyzed the relationships between the testate amoeba data and three sets of variables (1) “chemical” (pH, Eh potential, and conductivity), (2) “physical” (water temperature, altitude, i.e., Sphagnum mat micro-topography, and DWT), and (3) phenolic compounds in/from Sphagnum (water-soluble and primarily bound phenolics) as well as the habitat (fen/bog) and the sampling depth. Testate amoeba Shannon H′ diversity, equitability J of communities, and total density peaked in lower parts of Sphagnum, but the patterns differed between the fen and bog micro-sites. Redundancy analyses revealed that testate amoeba communities differed significantly in relation to Eh, conductivity, water temperature, altitude, water-soluble phenolics, habitat, and sampling depth, but not to DWT, pH, or primarily bound phenolics. The sensitivity of testate amoebae to weak environmental gradients makes them particularly good integrators of micro-environmental variations and has implications for their use in paleoecology and environmental monitoring. The correlation between testate amoeba communities and the concentration of water-soluble phenolic suggests direct (e.g., physiological) and/or indirect (e.g., through impact on prey organisms) effects on testate amoebae, which requires further research.  相似文献

Tropical peatlands represent globally important carbon sinks with a unique biodiversity and are currently threatened by climate change and human activities. It is now imperative that proxy methods are developed to understand the ecohydrological dynamics of these systems and for testing peatland development models. Testate amoebae have been used as environmental indicators in ecological and palaeoecological studies of peatlands, primarily in ombrotrophic Sphagnum-dominated peatlands in the mid- and high-latitudes. We present the first ecological analysis of testate amoebae in a tropical peatland, a nutrient-poor domed bog in western (Peruvian) Amazonia. Litter samples were collected from different hydrological microforms (hummock to pool) along a transect from the edge to the interior of the peatland. We recorded 47 taxa from 21 genera. The most common taxa are Cryptodifflugia oviformis, Euglypha rotunda type, Phryganella acropodia, Pseudodifflugia fulva type and Trinema lineare. One species found only in the southern hemisphere, Argynnia spicata, is present. Arcella spp., Centropyxis aculeata and Lesqueresia spiralis are indicators of pools containing standing water. Canonical correspondence analysis and non-metric multidimensional scaling illustrate that water table depth is a significant control on the distribution of testate amoebae, similar to the results from mid- and high-latitude peatlands. A transfer function model for water table based on weighted averaging partial least-squares (WAPLS) regression is presented and performs well under cross-validation (r \(^{2}_{apparent} \,=\, 0.76, \text {RMSE} \,=\, 4.29; \mathrm {r}^{2}_{jack} \,=\, 0.68, \text {RMSEP} \,=\, 5.18\) ). The transfer function was applied to a 1-m peat core, and sample-specific reconstruction errors were generated using bootstrapping. The reconstruction generally suggests near-surface water tables over the last 3,000 years, with a shift to drier conditions at c. cal. 1218-1273 AD.  相似文献

Winter deicing operations occur extensively in mid- to high-latitude metropolitan regions around the world and result in a significant reduction in road accidents. Deicing salts can, however, pose a major threat to water quality and aquatic organisms. In this paper, we examine the utility of Arcellacea (testate amoebae) for monitoring lakes that have become contaminated by winter deicing salts, particularly sodium chloride. We analysed 50 sediment samples and salt-related water property variables (chloride concentrations; conductivity) from 15 lakes in the Greater Toronto Area and adjacent areas of southern Ontario, Canada. The sampled lakes included lakes in proximity to major highways and suburban roads and control lakes in forested settings away from road influences. Samples from the most contaminated lakes, with chloride concentrations in excess of 400 mg/l and conductivities of >800 μS/cm, were dominated by species typically found in brackish and/or inhospitable lake environments and by lower faunal diversities (lowest Shannon diversity index values) than samples with lower readings. Q-R-mode cluster analysis and detrended correspondence analysis (DCA) resulted in the recognition of four assemblage groupings. These reflect varying levels of salt contamination in the study lakes, along with other local influences, including nutrient loading. The response to nutrients can, however, be isolated if the planktic eutrophic indicator species Cucurbitella tricuspis is removed from the counts. The findings show that the group has considerable potential for biomonitoring in salt-contaminated lakes, and their presence in lake sediment cores may provide significant insights into long-term benthic community health, which is integral for remedial efforts.  相似文献

In Asia, especially in China, our knowledge of the distribution of testate amoebae is still limited. In this paper, the geographical distribution of testate amoebae in Tibetan Plateau and northwestern Yunnan Plateau, southwest China and their relationships with the climatic factors have been studied. We found testate amoebae shifted in the most dominant species and increased in species (or genus) richness from northwest to southeast. Further, the linear regression analyses revealed that both species richness and genus richness have higher positive correlations with the mean temperature of the warmest month and annual mean precipitation as contrasted with the mean altitude, which showed weak negative correlation. This indicates that the temperature and precipitation are more significant influences on the richness than the altitude. The cluster analysis based on the community structure, defined by Sørenson’s coefficient matrix, suggested four groups from the 10 physiographical regions. This geographical distribution pattern was also closely related with the climatic regionalization. The present climatic regionalization pattern of the study area originated from the uplift of Tibetan Plateau and mainly occurred in or after the late Pleistocene. Therefore, the geographical distribution of testate amoebae in our study area may have experienced complicated and drastic changes corresponding to the variation of the climate caused by the geological events.  相似文献

The effect of temperature on pine wilt development in Scots pine (Pinus sylvestris) was examined in three experiments. Container-grown pines (4-6 years old) inoculated with 1,500 Bursaphelenchus xylophilus were incubated at constant temperatures in growth chamber for 8 weeks, then at a temperature range of 15-30 C in a greenhouse for 10-12 weeks. Nematode infection was greater, tree mortality was higher, and disease incubation was shorter at 32 and 30 C than at 25, 23, 18, and 11 C. Foliar symptoms developed more rapidly and uniformly at higher temperatures. Ninety-five percent of tree deaths at 32 and 30 C and 88% at 25 and 23 C occurred within the 8-week exposure to constant temperatures. Mortality at 18, 16, and 11 C occurred only after transfer to the greenhouse. Results indicate that pine wilt incidence is directly related and disease incubation period is inversely related to temperature and that high-temperature stress predisposes Scots pine to lethal infection by B. xylophilus.  相似文献

Peatland testate amoebae (TA) are well-established bioindicators for depth to water table (DWT), but effects of hydrological changes on TA communities have never been tested experimentally. We tested this in a field experiment by placing Sphagnum carpets (15 cm diameter) collected in hummock, lawn and pool microsites (origin) at three local conditions (dry, moist and wet) using trenches dug in a peatland. One series of samples was seeded with microorganism extract from all microsites. TA community were analysed at T0: 8–2008, T1: 5–2009 and T2: 8–2009. We analysed the data using conditional inference trees, principal response curves (PRC) and DWT inferred from TA communities using a transfer function used for paleoecological reconstruction. Density declined from T0 to T1 and then increased sharply by T2. Species richness, Simpson diversity and Simpson evenness were lower at T2 than at T0 and T1. Seeded communities had higher species richness in pool samples at T0. Pool samples tended to have higher density, lower species richness, Simpson diversity and Simpson Evenness than hummock and/or lawn samples until T1. In the PRC, the effect of origin was significant at T0 and T1, but the effect faded away by T2. Seeding effect was strongest at T1 and lowest vanished by T2. Local condition effect was strong but not in line with the wetness gradient at T1 but started to reflect it by T2. Likewise, TA-inferred DWT started to match the experimental conditions by T2, but more so in hummock and lawn samples than in pool samples. This study confirmed that TA responds to hydrological changes over a 1-year period. However, sensitivity of TA to hydrological fluctuations, and thus the accuracy of inferred DWT changes, was habitat specific, pool TA communities being least responsive to environmental changes. Lawns and hummocks may be thus better suited than pools for paleoecological reconstructions. This, however, contrasts with the higher prediction error and species’ tolerance for DWT with increasing dryness observed in transfer function models.  相似文献

Seasonality of Glutathione Dynamics in Scots Pine and Bilberry   总被引:2,自引:0,他引:2
Abstract: Seasonal changes in the glutathione status of Scots pine ( Pinus sylvestris L .) needles and bilberry ( Vaccinium myrtillus L.) stems and leaves were investigated during 1995 and 1997–1998, respectively. The glutathione concentration, the proportion of reduced glutathione (GSH%) and glutathione reductase (GR) activity were measured at monthly intervals throughout one year. The glutathione concentration and GSH% in current year's Scots pine needles were highest during late summer, but decreased during autumn. It is suggested that exchange reactions between free and protein-bound glutathione may be involved in this reduction. Both metabolites decreased towards summer in the previous year's needles. GR activity exhibited the lowest values in summer and the highest values in winter in both needle generations. In bilberry stems, glutathione peaked during midwinter, while GSH% remained high and relatively constant throughout the year. GR activity was highest in August, after which it gradually declined. In bilberry leaves, the total glutathione concentration was high in early summer. This peak was followed by a sharp decrease during midsummer. GR activity in the leaves was very low, but showed a clear increase towards autumn. Obviously, glutathione metabolism showed considerable seasonal variation in Scots pine needles and in bilberry leaves and stems, but the pattern of variation differed between the studied species and organs.  相似文献

Heterozygosity and Fitness: No Association in Scots Pine   总被引:6,自引:0,他引:6       下载免费PDF全文
O. Savolainen  P. Hedrick 《Genetics》1995,140(2):755-766
The association of six quantitative traits related to fitness with heterozygosity at 12 allozyme loci has been examined in three populations of Scots pine, Pinus sylvestris. Because of several characteristics of this organism and of this extensive data set, it appeared that this study would show a positive association between heterozygosity and these traits if indeed heterozygotes had higher values for these quantitative traits. Using several different statistical techniques including analysis of variance, regression with the scaling recommended from the adaptive distance model, and multiple regression, no evidence of an association was found. For example, only between 7 and 8% of the regression tests were significant at the 5% level and half of these showed a positive association and half showed a negative association. Further, the multiple regression analysis explained on average only 5.8% of the variation observed in the six different traits and only 1.5% of this variation was explained by a positive association. Power analysis was carried out (for the first time on these type of data), both for the single locus heterozygous advantage and the association of individual multiple locus heterozygosity and the quantitative traits. For diameter and height, two traits often used in similar studies, the average power to detect a single locus heterozygous advantage of 0.10 was 0.737 and the average power to detect a mean heterozygote advantage of 0.05 per locus for multiple loci was 0.797. As a result of this study and an examination of the published results from other studies, it appears that what positive associations have been observed are probably not, in large part, due to the presence of intrinsic heterozygote advantage.  相似文献

Arcellacea (testate lobose amoebae) were examined in 24 sediment–water interface samples collected over two late August field seasons in 2010 and 2011, from James and Granite lakes, Temagami Region, Northeastern Ontario. The work was carried out to quantitatively test species–environment relationships in a lake system known to be characterized by a significant pH gradient, partially the result of contamination from the early twentieth century Northland Pyrite Mine Co., located on the shoreline in the southern basin of James Lake. Redundancy analysis confirmed that arcellacean assemblage structure was most strongly controlled by pH, explaining 14.06 % (p?<?0.002) of the total variance. Q- and R-mode cluster analysis supported by detrended correspondence analysis yielded two major faunal assemblages. The Oligotrophic Assemblage (1) had a Shannon Diversity Index (SDI) ranging up to 2.45, typical of healthy boreal lakes. This assemblage characterized samples collected from higher pH stations within James and Granite lakes away from the immediate area of the mine site, while the Low pH Assemblage 2010 (2a) and Low pH Assemblage 2011 (2b) groupings were from the very low pH environments of James Lake adjacent to the former mine site. Both low diversity assemblages (SDI ranging from 0.62 to 1.22) were characterized by Arcella vulgaris, a species known to thrive in hostile lacustrine environments. Differing depositional conditions during August 2010, a probable result of different prevailing wind patterns that summer, led to allochthonous specimens of the seasonally planktic Cucurbitella tricuspis dominating the Low pH Assemblage 2010 (2a) fauna.  相似文献

Relationships between species abundance and water table depth and soil moisture have been modeled using weighted averaging and species niches have been calculated as optima and tolerance for these parameters. Niche separation of closely related taxa has been examined in detail and it has been shown that there is often a gradient of hydrological preference within each taxon. Wet to dry gradients include those found in the Trigonopyxis arcula group (T. arcula var. major > T. arcula > T. minuta), Assulina–Valkanovia group (A. seminulum > A. muscorum > V. elegans), and Trinema lineare group (T. lineare var. truncatum > T. lineare > T. lineare var. terricola), all of which are associated with a large to small size gradient. In addition, spined forms within the Euglypha and Placocista genera have been shown to consistently occur in wetter habitats than glabrous forms of those with shorter spines. A conclusion has been drawn that palaeoecological studies should cover the lowest taxa possible within these groups to maximize the ecological indicator value of the assemblages recorded.  相似文献

Freshwater microbial diversity is subject to multiple stressors in the Anthropocene epoch. However, the effects of climate changes and human activities on freshwater protozoa remain poorly understood. In this study, the diversity and distribution of testate amoebae from the surface sediments were investigated in 51 Chinese lakes and reservoirs along two gradients, latitude and trophic status. A total of 169 taxa belonging to 24 genera were identified, and the most diverse and dominant genera were Difflugia (78 taxa), Centropyxis (26 taxa) and Arcella (12 taxa). Our analysis revealed that biomass of testate amoebae decreased significantly along the latitudinal gradient, while Shannon-Wiener indices and species richness presented an opposite trend (P?P?相似文献
