Two actinomycete strains, 2-19(6)(T) and 2-30-b(28)(T), which produced single, non-motile noduler to warty spore surfaces, were isolated from sandy soil in Chokoria, Cox's Bazar, Bangladesh. A polyphasic study was carried out to establish the taxonomic position of these strains. Morphological and chemotaxonomic characteristics of these strains coincided with those of the genus Micromonospora. Phylogenetic analysis using 16S rDNA sequences indicated that these strains should be classified in the genus Micromonospora. The 16S rDNA sequence of strain 2-19(6)(T )showed closest similarity to the type strains of M. mirobrigensis (98.9%) and M. carbonacea (98.8%), and the strain 2-30-b(28)(T) to the type strains of M. purpureochromogenes (99.4%), M. halophytica (99.3%) and M. aurantiaca (99.2%). Furthermore, a combination of DNA-DNA hybridization results and some differential physiological and biochemical properties indicated that these strains were distinguished from the phylogenetically closest relatives. These strains therefore represent two novel species, for which the name Micromonospora chokoriensis sp. nov. and Micromonospora coxensis sp. nov. are proposed. The type strains are 2-19(6)(T) (=JCM 13247(T) =MTCC 8535(T)) and 2-30-b(28)(T) (=JCM 13248(T)=MTCC 8093(T)).  相似文献

An actinomycete strain 232617T was isolated from a composite mangrove sediment sample collected in Haikou, China. Phylogenetic analysis of the 16S rRNA gene sequence of strain 232617T indicated the highest similarity with Micromonospora siamensis TT2-4T (99.05%), Micromonospora krabiensis A-2T (98.99%) and Micromonospora carbonacea DSM 43815T (98.91%). The gyrB gene sequence analysis also indicated that 232617T should be assigned to the genus Micromonospora. The cell wall contains meso-DAP and glycine. The major menaquinones were MK-10(H4) and MK-10(H6), with MK-9(H4) as minor components. The characteristic whole-cell sugars are xylose, arabinose and glucose. The phospholipid profile comprises phosphatidylethanolamine, diphosphatidlglycerol and phosphatidylinositol mannoside. The DNA G+C content is 71.5 mol%. Furthermore, a combination of DNA–DNA relatedness and some physiological and biochemical properties indicated that the novel strain could be readily distinguished from the closest related species. On the basis of these phenotypic and genotypic data, strain 232617T represents a novel species of the genus Micromonospora, for which the name Micromonospora haikouensis sp. nov. is proposed. A previous phylogenetic study on type strains of the genus Micromonospora and Micromonospora species bearing non-validly published names has pointed towards the species status of several of latter strains. A novel actinomycete, designated strain NEAU-st4T, was isolated from a soil sample collected from Shaanxi province, Northwest China and characterized using a polyphasic approach. 16S rRNA gene sequence analysis revealed that strain NEAU-st4T has the highest sequence similarities with Actinomycetospora rishiriensis RI109-Li102T (99.4 %), Actinomycetospora corticicola 014-5T (99.1 %), Actinomycetospora chiangmaiensis YIM 0006T (98.8 %) and Actinomycetospora iriomotensis IR73-Li102T (98.2 %). However, the low level of DNA–DNA relatedness differentiated strain NEAU-st4T from its closest phylogenetic neighbours. The main chemotaxonomic properties of strain NEAU-st4T, such as the diamino acid of the peptidoglycan, the whole-cell hydrolysates, the predominant menaquinones and the phospholipid profile, supported its classification within the genus Actinomycetospora. The distinctive morphology of this strain compared with that of other members in the genus Actinomycetospora is the formation of sporangia directly on the substrate hyphae. Phenotypic and genotypic differences also allowed the distinction of the strain from closely related species. Consequently, strain NEAU-st4T represents a new species of the genus Actinomycetospora, for which the name Actinomycetospora atypica sp. nov. is proposed. Four isolates, with an optimum temperature of about 30°C and an optimum pH for growth of 6.0-6.5, were recovered from a borehole head of a mineral water aquifer in Portugal and from the stored bottles produced on site. Strains F2-63(T) and F2-178 were yellow-pigmented and formed non-motile rod-shaped cells. Strains G6M-30(T) and G6-54 were whitish-pigmented, translucent and form rod-shaped cells with a polar flagellum. The four strains were strictly aerobic, oxidase and catalase positive. The major fatty acids of strains F2-63(T) and F2-178 were C(18:1)ω7c and C(16:0), and the major fatty acids of strains G6M-30(T) and G6-54 were C(18:1)ω7c and C(16:1)ω7c. Ubiquinone 8 was the major respiratory quinone. Based on 16S rRNA gene sequence analysis, physiological and biochemical characteristics two new species of the genus Nevskia are described; Nevskia aquatilis represented by strains F2-63(T) (=LMG 26345 =CECT 7897) and F2-178 (=LMG 26344 =CECT 7898) and Nevskia persephonica represented by strains G6M-30(T) (=DSM 24987 =CECT 7975) and G6-54 (=DSM 25048 =CECT 7976).  相似文献

A Gram-negative, strictly aerobic, non-motile, rod-shaped and psychrophilic bacterial strain, PAMC 27137T, was isolated from the marine sediment of the Ross Sea, Antarctica. Strain PAMC 27137T was observed to grow at 4–10 °C, at pH 6.5–7.5 and in the presence of 2.5–4.0 % (w/v) sea salts. Phylogenetic analysis based on the 16S rRNA gene sequence indicated that strain PAMC 27137T belongs to the genus Lacinutrix showing the high similarities with Lacinutrix mariniflava JCM 13824T (97.6 %) and Lacinutrix algicola JCM 13825T (97.1 %). Genomic relatedness analyses based on the average nucleotide identity and the genome-to-genome distance showed that strain PAMC 27137T is clearly distinguished from the most closely related Lacinutrix species. The major fatty acids (>5 %) were identified as iso-C15:1 G (19.9 %), iso-C15:0 (19.3 %), iso-C17:0 3-OH (11.3 %), summed feature 9 (C16:0 10-methyl and/or iso-C17:1 ω9c as defined by MIDI, 9.1 %), iso-C15:0 3-OH (7.5 %), and anteiso-C15:1 A (5.8 %). The polar lipids were found to consist of phosphatidylethanolamine, an unidentified aminolipid, an unidentified aminophospholipid, and five unidentified phospholipids. The major respiratory quinone was identified as MK-6. The genomic DNA G+C content was determined to be 32.1 mol%. Based on the data from this polyphasic taxonomic study, strain PAMC 27137T is considered to represent a novel species of the genus Lacinutrix, for which the name Lacinutrix jangbogonensis sp. nov. is proposed. A white-coloured bacterium, SGM1-15T, was isolated from a paddy soil sample from Suwon, Republic of Korea. The cells were strictly aerobic, Gram-negative and curved rod-shaped. A phylogenetic analysis based on 16S rRNA gene sequences revealed that strain SGM1-15T was closely related to Curvibacter delicatus LMG 4328T (97.6% similarity) and Caenimonas koreensis EMB320T (97.5% similarity). The major respiratory quinone system was Q-8 and the predominant cellular fatty acids were C16:0 (39.9%), summed feature 3 (C16:1 ω7c and/or iso-C15:0 2-OH; 24.3%) and C17:0 cyclo (22.7%). The major polar lipids were diphosphatidylglycerol, phosphatidylglycerol and phosphatidylethanolamine. The major polyamines were 2-hydroxypurescine, purescine and spermidine. The DNA G+C content was 68.7 mol%. On the basis of the phylogenetic, physiologicl and chemotaxonomic data, stain SGM1-15T represents a novel species of the genus Caenimonas, for which the name Caenimonas terrae sp. nov. is proposed. A Gram-positive, moderately halotolerant, rod-shaped bacterium, designated YIM 94025T, was isolated from a soil sample from a salt lake in Xinjiang province, north-west China. Strain YIM 94025T was observed to grow at 25–45 °C (optimum 37 °C), 0–22 % NaCl (optimum 2–10 %) and pH 6.0–9.0 (optimum pH 8.0). Phylogenetic analyses based on 16S rRNA gene sequences revealed that the organism belongs to the genus Tenuibacillus and exhibited sequence similarity of 98.0 % to the closest type strain, Tenuibacillus multivorans AS 1.3442T. The predominant menaquinone was found to be MK-7; the cell-wall peptidoglycan diamino acid was meso-diaminopimelic acid; the polar lipids consisted of diphosphatidylglycerol, phosphatidylglycerol, phosphatidylinositol, an unidentified phospholipid and an unknown lipid; and the major fatty acids were found to contain iso-C15:0, anteiso-C15:0 and iso-C16:0. The chemotaxonomic characteristics of strain YIM 94025T are consistent with those of the genus Tenuibacillus. The level of DNA–DNA relatedness value between YIM 94025T and T. multivorans AS 1.3442T was 36.6 ± 4.5 %. The G+C content of the strain YIM 94025T was determined to be 38.5 %. Based on the comparative analysis of physiological, biochemical and chemotaxonomic data, as well as DNA–DNA hybridization results, the isolate is considered to represent a novel species of the genus Tenuibacillus, for which the name Tenuibacillus halotolerans sp. nov., is proposed, with the type strain of YIM 94025T (=CCTCC AB 2012860T = KCTC 33046T).  相似文献
