Studies of the relationship between sexual behavior and reproductive endocrinology in females offer critical insight into sexual selection, mate choice, and female reproductive strategies in primates. We examine the relationship between sexual solicitations and urinary estradiol in female Sichuan golden monkeys (Rhinopithecus roxellana) living under semiwild conditions at the Shanghai Wild Animal Park, China. We collected data on the frequency of sexual solicitations and urinary samples on 4 adult females during 2 mating seasons. We determined urinary estradiol levels via radioimmunoassay. Our results indicate that during the ovarian cycle, solicitation focused on the periovulatory period around the estradiol peak. The frequency of female solicitations rose to a peak after the peak of estradiol level, suggesting that ovarian steroids modulate sexual behavior during the fertile phase of the ovarian cycle. We also found that females frequently solicited matings during pregnancy, most of which the α male accepted and that resulted in copulations. During pregnancy, however, there was no obvious relationship between sexual solicitation and urinary estradiol. We conclude that in Rhinopithecus roxallana, sexual behavior and estradiol production do not strictly correlate during different parts of the reproductive cycle. We discuss additional relationships between social interactions, dominance, sexual behavior, and endocrine function.  相似文献

Administration of tamoxifen orally (3 mg/kg/day) during the post-ovulatory period from Days 16 to 20 or from Days 18 to 30 of female bonnet monkeys mated between Days 9 and 14 of the cycle resulted in inhibition of pregnancy establishment in 90-100% of monkeys tested. The pregnancy establishment in control female monkeys after exposure to the male during one ovulatory cycle was 66%. The effect of tamoxifen was not due to interference with luteal function because there was no reduction in serum progesterone concentrations after drug treatment. Exogenously administered progesterone could not reverse the inhibitory effect of tamoxifen on pregnancy establishment. The effect of tamoxifen was dose-dependent. We suggest that tamoxifen could be developed as an effective post-ovulatory contraceptive for regulation of female fertility.  相似文献

The effects of a progesterone antagonist, lilopristone (ZK 98.734), on induction of menstruation, inhibition of implantation or pregnancy, and termination of early and mid-pregnancy were studied in bonnet monkeys. In the regularly menstruating animals, administration of lilopristone (25 mg/day, s.c.) during the mid-luteal phase (Days 20-22 of the menstrual cycle) induced menstruation within 2-4 days after the initiation of treatment. A premature drop in circulating progesterone levels was also observed. The luteolytic effect of lilopristone was prevented by exogenous treatment with hCG; however, the animals showed premature menstruation, in spite of high progesterone levels (above 4 ng/ml). Treatment around the time of implantation (between Days 8 and 12 after the mid-cycle peak in estradiol levels) in mated animals provided 100% pregnancy protection. Treatment of pregnant animals on Days 30-32 of the menstrual cycle, i.e. about Day 20 after the estradiol peak, induced abortion in 8 of 10 animals. A significant (p less than 0.05) decrease in serum progesterone levels was observed on Day 3 after the initiation of treatment. However, the decrease was slower (slope: -0.36, r: 0.96) compared to that observed in nonpregnant animals (slope: -0.72, r: 0.95). In the other two animals, pregnancy was not affected. However, when the treatment was delayed until about Day 50 after the estradiol peak, all four animals aborted. This study suggests that lilopristone is a progesterone antagonist with a potential to induce menstruation, inhibit nidation, and terminate pregnancy. The antifertility effects are mediated through blocking progesterone action at the endometrium as well as decreasing progesterone bioavailability, which appears to be due to its effects on gonadotropin release.  相似文献

An attempt has been made in this paper to review our present understanding of luteal function during the periimplantation period and in particular hormonal requirement for implantation and maintenance of early pregnancy in the non-human primate.In a fertile cycle thecorpus luteum is apparently rescued from luteolysis by chorionic gonadotropin secreted by the implanted blastocyst, In the bonnet monkey the serum progesterone titers during the luteal phase of a fertile cycle seems higher compared to that of nonmated cycling monkeys. This suggested that thecorpus luteum is receiving some stimulatory signal from the blastocyst even prior to implantation. The recent demonstration that human blastocyst in culture secretes into the medium human chorionic gonadotropin essentially support the above assumption. However, attempts to extend the luteal phase of cycling unmated monkeys with exogenous human chorionic gonadotropin injection has hitherto not met with complete success suggesting that there could be other than chorionic gonadotropin, additional luteal stimulatory factors the unimplanted blastocyst is secreting.Corpus luteum is the principle source of both progesterone and estrogen produced during the periimplantation period and dysruption of luteal function, brought about by either lutectomy or ovariectomy or luteinizing hormone antiserum treatment, followed by progesterone supplementation leads to maintenance of pregnancy. This has lead to questioning the need for estrogen in the maintenance of early pregnancy. Recent work using Zuclomiphene, an antiestrogen during days 5–11 of cycle in rhesus monkeys mated between day 9–14, has however, suggested that estrogen may be required for implantation. Further work is needed to arrive at an unequivocal decision regarding the need for estrogen in maintenance of early pregnancy in the primate.  相似文献

The effects of prostaglandin (PG)F2alpha and PGF2alpha, 1-15 lactone were compared in luteal phase, non-pregnant and in early pregnant rhesus monkeys. Animals treated with either PG after pretreatment with human chorionic gonadotropin (hCG) had peripheral plasma progesterone concentrations that were not statistically different from those in animals treated with hCG and vehicle. However, menstrual cycle lengths in monkeys treated with PGF2alpha, 1-15 lactone were significantly (P less than 0.02) shorter than those in vehicle treated animals. In the absence of hCG pretreatment, plasma progesterone concentrations were significantly (P less than 0.008) lower by the second day after the initial treatment with either PGF2alpha or PGF2alpha, 1-15 lactone than in vehicle treated monkeys. Menstrual cycle lengths in monkeys treated with either PG were significantly (P less than 0.04) shorter than those in animals treated with vehicle. There were no changes in plasma progesterone concentrations in early pregnant monkeys treated with PGF2alpha, and pregnancy was not interrupted. In contrast, plasma progesterone declined and pregnancy was terminated in 5 of 6 early pregnant monkeys treated with PGF2alpha, 1-15 lactone. These data indicate that PGF2alpha, 1-15 lactone decreases menstrual cycle lengths in non-pregnant rhesus monkeys. More importantly, PGF2alpha, 1-15 lactone terminates early pregnancy in the monkey at a dose which is less than an ineffective dose of PGF2alpha.  相似文献

This study, based on three years of mating behavior observations and 10 years of birth records, reveals that Sichuan golden monkeys in captivity displayed a marked seasonality of mating behavior and births. The peak of matings occurred around October, and births occurred in March-June. The birth peak followed the mating peak by six to seven months. This seasonal cycle of matings and births was similar to observations made in the wild, where both temperature and food resources were favorable in spring. The time delay between peaks of matings and births was the approximate length of gestation, which implies that mating behavior was concentrated during the period of conception. We suggest that the peak of births in captive Sichuan golden monkeys occurred during the time of year with the most favorable environmental conditions, and the peak of matings corresponded with the period of conception.  相似文献

The expression of endometrial beta3 integrin and insulin-like growth factor binding protein-1 (IGFBP-1) was studied in cycling and pregnant vervet monkeys. There were clear changes of beta3 integrin expression during the menstrual cycle, with the strongest immunostaining observed on day 26. Moderate to strong expression was observed during pregnancy. The expression of IGFBP-1 during the menstrual cycle was weak but upregulated during pregnancy with moderate to strong staining. The administration of a single dose of onapristone at 10 mg/kg on days 17, 21 and 22 of the menstrual cycle, followed by a biopsy on days 22, 22 and 26, respectively, and during pregnancy (34-44 days menstrual age) 24 h before the biopsy, disrupted and desynchronized the endometrium. However, no effect on beta3 integrin expression could be observed and staining reflected the untreated patterns. The same applied to IGFBP-1 except that during pregnancy the expression of this protein was reduced or abolished. The results suggest that beta3 integrin is associated with endometrial receptivity in vervet monkeys and that IGFBP-1 plays an important role during pregnancy in this species. The administration of onapristone appeared to only influence IGFBP-1 expression. To our knowledge, this is the first time that these endometrial proteins have been investigated in vervet monkeys. This study should therefore contribute to improving our understanding of the reproductive function of this species.  相似文献

During two consecutive mating seasons from August 2001 to December 2002, we observed the sexual behavior of Sichuan golden monkeys in the Chengdu Zoo and the Shanghai Wild Animal Park. We collected urine samples from females in the early morning. We also measured urinary estradiol and progesterone levels using radioimmunoassay. Based on the estradiol concentration during the menstrual cycle, we discovered that the length of the menstrual cycle was 286.33 1.67 days. The length of pregnancy was about 200 days. The level of estradiol after impregnation was significantly different with the level during the ,same period in the non-receptive part of the cycle. Thus we could diagnose early pregnancy in female Sichuan golden monkeys by checking their urinary estradiol level .  相似文献

BACKGROUND : The covalent modification of interferon (IFN) α2b with monomethyoxy polyethylene glycol (PEG) reduces its clearance rate and increases its half‐life. High doses of interferon (IFN) α2b have previously been shown to affect maintenance of pregnancy in rhesus monkeys. Given the role of ovarian hormones in reproductive function and pregnancy, this study was conducted to assess the effects of PEG‐IFNα2b or IFNα2b (comparative control) on ovarian hormones and menstrual cyclicity in cynomolgus monkeys. In addition, the potential for reversibility of PEG‐IFNα2b or IFNα2b‐related observations was assessed. METHODS : Monkeys were administered 3,105 µg/m2 human recombinant (hr) IFNα2b or 52, 262, or 4,239 µg/m2 PEG‐hr‐IFNα2b every other day for one menstrual cycle, followed by a post‐dose period of up to two menstrual cycles. RESULTS : Monkeys administered 3,105 µg/m2 hr‐IFNα2b or 52, 262, or 4,239 µg/m2 PEG‐hr‐IFNα2b exhibited transient decreases in food consumption, leukocyte and erythrocyte parameters. Monkeys administered 3,105 µg/m2 hr‐IFNα2b exhibited lengthened menstrual cycles that were associated with a delay in reaching peak ovarian hormone levels and lower respective peak concentrations. Similarly, monkeys administered 4,239 µg/m2 PEG‐hr‐IFNα2b exhibited lengthened menstrual cycles and a delay in reaching peak ovarian hormone levels and slightly lower respective peak concentrations. Post‐dosing menstrual cycle length, estradiol and progesterone profiles exhibited evidence of recovery in both the hr‐IFNα2b and the high‐dose PEG‐hr‐IFNα2b groups. CONCLUSIONS : Administration of hr‐IFNα2b or PEG‐hr‐IFNα2b at high doses to cynomolgus monkeys resulted in similar effects on menstrual cycles, estradiol and progesterone profiles, and exhibited evidence of reversibility upon cessation of dosing. These results suggest that the previously observed high‐dose IFNα‐related effects on the maintenance of pregnancy in monkeys are likely the result of altered ovarian function. A major limitation to progress in primate embryology is the lack of an adequate supply of preimplantation embryos. We describe a method for recovering preimplantation-embryos in bonnet monkeys (Macaca radiata ) using a nonsurgical uterine flushing technique similar to the one previously employed in rhesus monkeys. Forty cyclic females were screened for cervical cannulation, and 10% of these had an impassable cervix. Eleven females suitable for cannulation were selected, and 27 menstrual cycles were monitored over a 5-mo period. Seventy-one percent of the cycles showed estrogen peaks, which were observed between Days 9 and 14 of the cycle. Following natural mating, uterine flushings were performed on Days 5 to 8 of pregnancy (Day 0 = the day following the estrogen peak). Of the 27 recovery attempts, 9 (33.3%) resulted in the recovery of ovulation products, including those of an unfertilized oocyte and empty zona (2 cases), retarded cleavage-stage (4 to 8-cell) embryos (4 cases), morula (1 case) and blastocysts (2 cases). These results show, for the first time, that the nonsurgical uterine flushing technique can be successfully performed to recover uterine-stage preimplantation embryos from bonnet monkeys.  相似文献

The Yunnan snub-nosed monkey (Rhinopithecus bieti) is one of the most endangered species in the world, and it is endemic to China. According to our knowledge, there was no information on reproduction for this species. The present study was designed to understand the characteristics of reproductive hormone secretion during the menstrual cycle and pregnancy of this species by monitoring urinary estrone conjugate (E1C), pregnanediol-3-glucuronide (PdG), bioactive follicle-stimulating hormone (FSH), and luteinizing hormone (LH). The urine samples were collected each day from four adult females for eight menstrual cycles, and once in 3 days during pregnancy (three full-term pregnancies, one mid-term abortion). The steroid conjugate was tested by radioimmunoassays (RIAs), and bioactive FSH and LH levels were measured in vitro by the sensitive bioassays granulosa cell aromatize bioassay (GAB) and rat interstitial cell testosterone (RICT), respectively. The results showed that: 1) E1C presented a preovulatory peak (183.9 +/- 8.6 ng/mgCr) followed by a definite elevation of PdG; 2) PdG in the luteal phase (754.4 +/- 30.6 ng/mgCr) was three- to fivefold higher than during the corresponding follicular phase (198.3 +/- 11.4 ng/mgCr); 3) the peaks of bio-LH and bio-FSH were on the same day, while the E1C peak was 1 or 2 days before the peaks for these two hormones; 4) bio-FSH levels were higher in the follicular phase than in the luteal phase, and bio-LH levels elevated slightly in the luteal phase; 5) the mean cycle length was 23.6 +/- 3.5 days (n = 3) based upon successive urinary LH peaks; 6) based on the interval from the day of E1C peak to the day of parturition, the gestation was 203.7 +/- 2.5 days (n = 3); and 7) both E1C and PdG increased and remained high after pregnancy, with a sharp decrease in basal levels following parturition or mid-term abortion. The results suggested that the pattern of reproductive hormones for R. bieti is similar to that of other Old World monkeys, but the concentration of the hormones is different from that of other species. This species has a longer progestation period, which may be related to its classification status.  相似文献

A radioreceptorassay (RRA) for macaque luteinizing hormone (LH)/chorionic gonadotropin (CG) was adapted from the clinical RRA for human LH/CG, Biocept-G?, for the purposes of detection of pregnancy prior to day 20 of gestation and for estimation of the time of ovulation in macaques. The 90-min assay procedure was simple, accurate, and reliable. Seventy-five rhesus monkeys (Macaca mulatta) and 20 crab-eating monkeys (Macaca fascicularis) were tested for the presence of CG in the serum on estimated days 17–20 of pregnancy. Of a total of 160 tests, four false negative and 0 false positive tests were obtained, for an accuracy of 97.5%. The preovulatory LH peak was detected in 19 rhesus monkeys by semiquantitative RRA of LH/CG. Ovulation was confirmed in these 19 animals by the presence of a fresh corpus luteum at laparotomy 2–10 days after ovulation, collection of an embryo, pregnancy, or subsequent cycle history. The short, simple assay procedure and the low inter-and intraassay coefficients of variation (7.3 and 3.7%, respectively) allow use of this assay in an economical, predictive, as well as retrospective, capacity for estimation of the time of ovulation in rhesus monkeys. The sensitivity, reliability, species nonspecificity, simplicity, and rapidity of performance of this RRA for LH/CG are features which add up to a useful new management tool for breeding macaques for research purposes.  相似文献

A homologous double-antibody radioimmunoassay developed for humans was used to measure serum prolactin, progesterone, and estradiol in common marmosets. In the ovarian cycle of common marmosets, serum progesterone began to increase after an estradiol surge, attained a peak level, and then declined before the ensuing pre-ovulatory rise in estradiol. During the luteal phase, the change in serum concentrations of estradiol was synchronized with that of progesterone. During the ovarian cycle there was no consistent change in serum prolactin concentrations. During the last 75 days of pregnancy the prolactin level was higher than during the ovarian cycle and the first 70 days of pregnancy. Moreover, during lactation, mothers with suckling twin infants had a higher prolactin level than during the final stage of pregnancy.  相似文献

The endocrine characterization of the menstrual cycle of the stumptailed monkey was determined and compared with that of the rhesus monkey. Serum concentrations of luteinizing hormone (LH), estradiol-17beta, and progesterone were determined on a daily basis from 5 monkeys. Differences included: 1) basal and peak concentrations of LH 40% of those in rhesus monkeys, 2) an estraidol peak occurring on the day proceding LH peak, 3) an estradiol peak on the 3rd day after LH surge and representing the highest mean estradiol value during the luteal phase, and 4) absence of the usual periovulatory decline in serum progesterone. Further experimentation is needed to understand the importance of these differences.  相似文献

SYNOPSIS. Social groups of rhesus monkeys (Macaca mulatto) livingin outdoor environments exhibit a distinct seasonal mating patternand female rhesus are observed to be sexually receptive fordiscrete periods averaging about 9 days duration. In the laboratoryenvironment mating occurs throughout the year and, in the pairtest, female rhesus are observedto be sexually receptive throughall phases of a menstrual cycle, with a periovulatory peak incopulatorybehavior. The apparent conflict between results from field andlaboratory studies has been difficult to resolve because ofmethodological limitations inherent in each study environment.Studies conducted on social groups of rhesus monkeys housedin outdoor compounds, an environment in which the species typicalseasonal mating pattern is preserved and which allowsfor concomitantmeasurement of behavioral and hormonal variables, provided informationabout the covariance between female sexual behavior and ovariancycles. Female copulations were observed only in associationwith ovulatory cycles, and were limited to the follicular andperi-ovulatory phases of such cycles, demonstrating that copulatorybehavior in female rhesus monkeys is strongly influenced bycyclic hormonal variables. The studies also revealed that femalestend to conceive on the first ovulatory cycle of the season,that there was no synchrony of cycles among the females, andthat the best predictor of the timing of ovulation in a particularfemale is reproductive outcome in the previous year.  相似文献

The circulating concentrations of progesterone, FSH, and follistatin across the estrous cycle and gestation were compared in Australian merino sheep that were homozygous for the Booroola gene, FecB, or were noncarriers. The Booroola phenotype is due to a point mutation in the bone morphogenetic protein receptor 1B. Progesterone concentrations began to rise earlier and were higher in the Booroola ewes than in the noncarriers on most days of the luteal phase but not during the follicular phase of the cycle. Follistatin concentrations remained unchanged across the estrous cycle in both groups of ewes, with no differences between genotypes. FSH concentrations were higher in Booroola ewes than in noncarrier ewes on most days of the estrous cycle, with a significantly higher and broader peak of FSH around the time of estrus. Progesterone concentrations were significantly higher in early and midgestation in Booroola ewes but were lower toward the end of gestation than those in noncarriers. FSH declined in both groups across gestation, with lower concentrations of FSH in Booroola ewes during midgestation. Follistatin remained unchanged across gestation in Booroola ewes and noncarrier ewes with a twin pregnancy but declined across gestation in noncarrier ewes with a singleton pregnancy. These results suggest that follistatin concentration is not regulated by the FecB gene during the estrous cycle and pregnancy but is influenced by the number of fetuses. However, the FecB gene appears to positively affect both progesterone and FSH during the estrous cycle and across pregnancy, which suggests that bone morphogenetic proteins play an important role in the regulation of both hormones.  相似文献

The effect of pregnancy on peak O2 uptake (VO2 peak) during tethered swimming was evaluated in 10 women during their 25th and 35th wk of pregnancy, as well as 9-11 wk postpartum. The swim results were compared with cycle ergometry results obtained at similar times. The results indicated that exercise-induced maximal heart rates remained the same and were similar for the swim and cycle trials, approximately 184 +/- 4 beats/min. Cycling VO2 peak was not affected by pregnancy, averaging 1.94 +/- 0.11 l/min. Postpartum swim VO2 peak was similar to the cycle results; however, during pregnancy it was significantly lower than cycling VO2 peak (P less than 0.05; postpartum, 1.78 +/- 0.14 l/min; 25th wk, 1.64 +/- 0.12; 35th wk, 1.48 +/- 0.11). Hemoglobin concentrations and hematocrits were lower during pregnancy; however, changes in plasma volume (based on hematocrit and hemoglobin) were found to be significantly greater during cycling than during swimming and also greater during pregnancy for both modes of exercise. It was concluded that, unlike cycling, the VO2 peak of pregnant women during swimming is reduced. This reduction in VO2 peak was associated with a decreased peak ventilation (r = 0.864) but was not correlated to exercise-induced hemoconcentration (r = -0.29). Furthermore, pregnancy results in a greater-than-normal exercise-induced hemoconcentration, which may be related to pregnancy-induced changes in capillary dynamics.  相似文献

Levels of estradiol and progesterone in peripheral serum of seven cynomolgus monkeys (Macaca fascicularis) were measured from about the 30th day of pregnancy until parturition. Although the pattern of each steroid in circulation differed somewhat from the respective patterns in rhesus monkeys (Macaca mulatta), there were basic similarities. On the day of delivery, the corpus luteum was excised and in vitro incubation of dispersed luteal cells was performed. Isolated luteal cells produced progesterone under control conditions and responded to the addition of HCG with enhanced steroidogenesis. Accordingly, "rejuvenation" of the corpus luteum may occur during advanced gestation in cynomolgus monkeys. These findings, along with establishing the efficacy of the Subhuman Primate Pregnancy Test kit to diagnose pregnancy in this macaque, extend previous evidence for utility of cynomolgus monkeys as a primate model for study of steroid hormones in pregnancy.  相似文献

The present paper summarizes some of the important biological and physiological data recorded over a 30-year period on the biology of bonnet monkeys in captivity. Data on sexual maturity, menstrual cyclicity, general behaviour, endocrine profile, reproductive physiology, gestation, parturition, postpartum amenorrhoea in the female, and sexual maturity, hormone profile, and seasonal variation in sperm count of the male monkeys are presented. In addition to the biological values, weights of selected organs, vertebral and dental pattern are also presented. Menarche occurred at an age of 36±4 months and the first conception in the colony occurred at an age of 54±4 months. The average menstrual cycle length was 28±4.3 days. Majority of monkeys did not cycle regularly during March–June during which the temperature reached a peak. The pregnancy index of the colony was 80% with controlled breeding. The gestation period was 166±5 days with 6–7 months postpartum amenorrhoea. Males attained sexual maturity by the age of 6–7 years and exhibited the characteristic nocturnal surge of serum testosterone at this age and sperm concentration ranged from 116–799 millions/ejaculate.  相似文献
