整合分析(meta-analysis)是对同一主题下多个独立实验结果进行综合的统计学方法,被认为是到目前为止最好的数量综合方法,其统计量为效应值,反应比(InR)和Hedges'd值是生态学应用中最常用的两个效应值。以综合植物生理生态学指标对大气CO2浓度倍增响应为实例,比较这两种效应值的不同之处。采用两种效应值指标会对同一个生理生态指标产生不同的总效应值大小,有时甚至会改变效应值的方向;InR相对Hedges'd更易产生正效应值;Hedges'd较InR可拉大不同生理生态指标之间总效应值的差异;Hedges'd具有正负效应对称性,而InR却具有正负效应的不对称性。  相似文献

Quantitative trait locus (QTL) mapping is often done in a single segregating population, such as a backcross or an intercross. Both QTL location and effect size are then estimated from the same dataset. This approach results in an over-estimate of effect size for two reasons: (1) LOD scores, which are maximized over numerous point-wise tests, are correlated with estimated effect size and (2) small effect QTLs are often undetected in underpowered experiments, yielding inflated effect sizes for detected QTLs (the Beavis effect). When it is impractical to maintain or generate large population sizes, an alternative is to use two populations, one for initial detection and localization and a second for a locus-matched estimate of effect size, not conditioned on significance. Recombinant inbred (RI) panels are eminently suitable for this approach, as each strain genotype can be sampled repeatedly. We present mapping results from the LXS RI panel for two behavioral phenotypes relating to ethanol response: low-dose ethanol activation and loss of righting following high-dose injection. Both the phenotypes were measured in two or three independent cohorts, which were then used to re-estimate effect size. Many small-effect QTLs replicated using this approach, but in all cases, effect size, in the replicate cohorts, was reduced from the initial estimate, often substantially. Such a reduction will have important consequences for power analyses in which sample sizes are determined for subsequent confirmation studies.  相似文献

Summary General expressions are established for means, variances and covariances, taking into account the effect of interactions between individuals. For simplicity, only groups of size two are considered so that interactions are restricted to pairs of individuals; genetic effects are reduced to additivity and to additive × additive interaction between the direct effect of an allele and the associate effect of another. The effect of family structure is studied, and some consequences of interest to plant breeding are discussed by an application to general combining ability selection. From a population genetic point of view, this approach reveals a new application of coefficients of kinship.  相似文献

The search for the alleles that matter, the quantitative trait nucleotides (QTNs) that underlie heritable variation within populations and divergence among them, is a popular pursuit. But what is the question to which QTNs are the answer? Although their pursuit is often invoked as a means of addressing the molecular basis of phenotypic evolution or of estimating the roles of evolutionary forces, the QTNs that are accessible to experimentalists, QTNs of relatively large effect, may be uninformative about these issues if large‐effect variants are unrepresentative of the alleles that matter. Although 20th century evolutionary biology generally viewed large‐effect variants as atypical, the field has recently undergone a quiet realignment toward a view of readily discoverable large‐effect alleles as the primary molecular substrates for evolution. I argue that neither theory nor data justify this realignment. Models and experimental findings covering broad swaths of evolutionary phenomena suggest that evolution often acts via large numbers of small‐effect polygenes, individually undetectable. Moreover, these small‐effect variants are different in kind, at the molecular level, from the large‐effect alleles accessible to experimentalists. Although discoverable QTNs address some fundamental evolutionary questions, they are essentially misleading about many others.  相似文献

应用复方克痛宁注射液,治疗各种痛证82例,显效50%,有效43.9%,总有效率93.9%。其中大多数病例都是经用其它药或疗法无效或效差而用本药获效的。对关节痛、神经血管性头痛等顽固性疼痛显著独特。用药次数最少1次,最多35例,仅极个别有轻微短暂口干、头晕副作用,各项常规化验检查均无异常,效果显著高于克痛宁或盐酸平痛新单用。临床应用的结果与动物试验观察到的协同增效结果一致。  相似文献

The kinematics of flight in the little brown bat are described, both with and without the influence of ground effect. These data are used to estimate aerodynamic power costs and the reduction in power due to ground effect. An effect of low temperature (0-2 degrees C) on meiosis was studied in different varieties from various ecological zones. It is shown that the Mexican varieties are most sensitive to the effect of low temperature. The percentage of sterile pollen and disturbances in anaphase I in them are the highest. The local varieties with respect to these characters are more resistant to the effect of low temperature.  相似文献

A method is presented for estimating the treatment effect in a meta-analysis when some of the studies are comparative studies, and the rest are noncomparative, historical control studies. A random-effects model is used in which the baseline effect in each study is random, but the treatment effect is constant. With this model the appropriate contribution of the historical studies can be determined. Extensions of the model are developed to accommodate preliminary tests for bias, and for the possibility that the treatment effect is heterogeneous.  相似文献

Clostridium difficile Toxin B (TcdB) glucosylates low molecular weight GTP-binding proteins of the Rho subfamily and thereby causes actin re-organization (cell rounding). This "cytopathic effect" has been generally attributed to RhoA inactivation. Here we show that cells expressing non-glucosylatable Rac1-Q61L are protected from the cytopathic effect of TcdB. In contrast, cells expressing RhoA-Q63L or mock-transfected cells are fully susceptible for the cytopathic effect of TcdB. These findings are extended to the Rac1/RhoG mimic IpgB1 and the RhoA mimic IpgB2 from Shigella. Ectopic expression of IpgB1, but not IpgB2, counteracts the cytopathic effect of TcdB. These data strongly suggest that Rac1 rather than RhoA glucosylation is critical for the cytopathic effect of TcdB.  相似文献

 The main subject for models of postzygotic isolation has been how reproductive isolation genes (RI genes) which cause hybrid inviability or sterility spread within populations despite their deleterious effects. The models are divided into three categories according to the within-population effect of RI genes in their fixation process. (1) The beneficial effect model, where RI genes are assumed to spread within populations by a positive selective force via natural or sexual selection. (2) The neutral effect model, where RI genes are assumed not to affect the fitness of individuals in their fixation process and to be spread by genetic drift. (3) The deleterious effect model, where RI genes are assumed to exhibit some (slightly) deleterious effects in their fixation process and to be spread by genetic drift. Factors that affect the applicability of these models are discussed. If a selective force such as sexual conflict or natural selection facilitates the evolution of RI genes, the beneficial effect model should be applied. Many empirical studies have suggested that positive selection plays an important role in the evolution of hybrid male sterility. If the mutation rates of RI genes are low, and the specificity of epistatic interaction causing hybrid inviability or sterility is high, the neutral effect model should be applied. However, if the opposite condition applies, the deleterious effect model should be applied. In this paper, a discrete-time host–parasitoid model is investigated. Two biological phenomena, the Allee effect of the host population and the aggregation of the parasitism, are considered in our mathematical model. Through extensive numerical simulations, we gain some interesting findings related to Allee effect from this research. Firstly, the ranges of parameter, in which the population dynamics is chaos, are compressed when Allee effect is added. Secondly, the sensitivity to initial conditions of the host–parasitoid system decreased after adding Allee effect. Thirdly, without Allee effect, we observed two complicated dynamics, intermittent chaos and supertransients. However, when Allee effect is included, these two phenomena are replaced by another kind of phenomenon-period alternation, where chaos is eliminated. From above three novel findings, it can be concluded that dynamic complexities are alleviated by Allee effect. This conclusion is crucial in resolving the discrepancy between real population dynamics and theoretical predictions. Furthermore, the importance of this research is to help us understand the mechanisms inducing the irregular fluctuations of the natural populations.  相似文献

Access to mental representations of smaller vs. larger number symbols is associated with leftward vs. rightward spatial locations, as represented on a number line. The well-replicated SNARC effect (Spatial-Numerical Association of Response Codes) reveals that simple decisions about small numbers are facilitated when stimuli are presented on the left, and large numbers facilitated when on the right. We present novel evidence that the size of the SNARC effect is relatively stable within individuals over time. This enables us to take an individual differences approach to investigate how the SNARC effect is modulated by spatial and numerical cognition. Are number-space associations linked to spatial operations, such that those who have greater facility in spatial computations show the stronger SNARC effects, or are they linked to number semantics, such that those showing stronger influence of magnitude associations on number symbol decisions show stronger SNARC effects? Our results indicate a significant correlation between the SNARC effect and a 2D mental rotation task, suggesting that spatial operations are at play in the expression of this effect. We also uncover a significant correlation between the SNARC effect and the distance effect, suggesting that the SNARC is also related to access to number semantics. A multiple regression analysis reveals that the relative contributions of spatial cognition and distance effects represent significant, yet distinct, contributions in explaining variation in the size of the SNARC effect from one individual to the next. Overall, these results shed new light on how the spatial-numerical associations of response codes are influenced by both number semantics and spatial operations.  相似文献

It is well recognized that individuals of many species can benefit from the presence of conspecifics, a concept broadly referred to as the Allee effect. At the metapopulation level, there is an analogous but essentially different phenomenon called the Allee-like effect that leads to metapopulation extinction thresholds at low habitat occupancy. But so far not adequate attention has been paid to this phenomenon. In this paper, the Allee-like effect is introduced into a metapopulation model of one species and also that of a three-state two-species competitive system. Phase plane analysis is used to investigate the dynamics of these models. We demonstrate that the Allee-like effect alone could lead to multiple stable states in three-state two-species competitive systems at the metapopulation level, and the number of stable states decrease as the Allee-like effect becomes more severe. Severe Allee-like effects may make coexistence impossible and may even lead to the extinction of both species even if their initial habitat occupancies are high and suitable habitats are enough. It is especially noticeable that depending on their initial conditions one species may exclude the other one that subjects to a weaker Allee-like effect than the former, while the second species always excludes the first one when both species are assumed to be in the absence of the Allee-like effect. We also investigate the habitat destructive effect on the Allee-like system mentioned above. Research indicates that the existence of the Allee-like effect makes a metapopulation more susceptible to habitat destruction. All in all, the Allee-like effect is probably a destabilizing factor that, together with habitat destruction, would affect the continuous existence of species. These conclusions may have important implications for conservation and metacommunity organization.  相似文献

In clinical trials for the comparison of two treatments it seems reasonable to stop the study if either one treatment has worked out to be markedly superior in the main effect, or one to be severely inferior with respect to an adverse side effect. Two stage sampling plans are considered for simultaneously testing a main and side effect, assumed to follow a bivariate normal distribution with known variances, but unknown correlation. The test procedure keeps the global significance level under the null hypothesis of no differences in main and side effects. The critical values are chosen under the side condition, that the probability for ending at the first or second stage with a rejection of the elementary null hypothesis for the main effect is controlled, when a particular constellation of differences in mean holds; analogously the probability of ending with a rejection of the null hypotheses for the side effect, given certain treatment differences, is controlled too. Equations governing oxygen transport from alveolar gas to red blood cells flowing through pulmonary capillaries are written down. Some analytical predictions are made on factors affecting the rate at which this process takes place. Numerical simulations are carried out to investigate the effect of red blood cell shape, capillary dimensions, haematocrit and choice of oxygen dissociation curve on pulmonary oxygen transport. These factors all have an effect on pulmonary transport, with the effect being much more marked for simulations with low oxygen levels, typical of those seen in some subjects with respiratory disease.  相似文献

Ureases are metalloenzymes that are widespread among plants, fungi and bacteria. Urease isoforms (jack bean urease-JBU and canatoxin) from Canavalia ensiformis seeds are toxic to insects and fungi, suggesting a role in plant defense. The entomotoxic effect is due to the release of a 10-kDa peptide by cathepsin-like enzymes in the insect's midgut. Urease causes a decrease in post-feeding weight loss in Rhodnius prolixus, suggesting an effect on water balance. To investigate how this impairment occurs, we have evaluated the action of JBU and the urease-derivated peptide Jaburetox-2Ec on R. prolixus Malpighian tubules and also investigated the involvement of second messengers. JBU and Jaburetox-2Ec affect serotonin-induced secretion from Malpighian tubules. This effect is not cAMP-dependent, but the Jaburetox-2Ec effect is cGMP-dependent. Eicosanoid metabolites and calcium ions appear to be involved in JBU effect on diuresis, but are not involved in the action of Jaburetox-2Ec. Jaburetox-2Ec, but not JBU, causes a change in the transepithelial potential of the tubules. Canatoxin has a similar effect on tubules secretion, decreasing the secretion rate, but the urease from Helicobacter pylori has no significant effect. These data are helpful in our understanding of the actions of ureases and derived peptides on insects, and also reinforces the potential use of these proteins as biopesticides.  相似文献

The storage effect is defined as an increase of mutational damage after cessation of treatment. It differs from other kinds of delayed effect (replication errors, replicating instabilities) in not requiring replication of DNA. In Neurospora ad3A 38701 inos 37401 diepoxybutane (DEB) yields a storage effect for adenine reversions, but none for inositol reversions. The storage effect takes place in treated washed spores that are sedimented in a centrifuge tube, but not in spores that are agitated in water. Under the latter conditions, response to storage is gradually lost. The storage effect can be imitated by administering very small amounts of DEB to cells that had been previously treated with a moderately high dose (booster effect). During post-treatment shaking in water, responses to booster and to storage conditions disappear together; response to booster disappears at the same rate in spores that are sedimented in centrifuge tubes. No mutagenic action could be detected in eluates from heavily treated cells. We have concluded that treatment with DEB sensitizes the conidia to further small doses of DEB whether these are administered extraneously as booster or present intracellularly during storage. Sensitization is lost in the course of a few hours in shaken as well as in sedimented spores. Thus, while the storage effect is due to traces of mutagen, its gradual disappearance after treatment is not due to loss of these traces.Correlated with the ability to yield a storage effect, and probably part of the storage effect, is the response to temperature between treatment and plating. Conidia that can give a storage effect yield fewer mutations when spread on cold agar than when inplated into warm agar or heat-shocked before spreading; the excess of mutation under the latter two conditions forms part of the final storage effect. The true base line for calculation of the storage effect is therefore mutation frequency among spread spores.For DEB as mutagen, response to storage by the adenine locus and lack of response by the inositol locus are correlated with the responses of these two loci to dose of DEB and to combination treatment of DEB with UV, or DEB with nitrous acid (NA). This makes it possible to fit all observations into the picture of a general hypothesis on the cellular effects of DEB. Because of the differential response of the two loci, storage and plating procedures offer two additional means for manipulating specificity in this system.  相似文献
