Kinetics of nitrate uptake by freshwater algae   总被引:2,自引:2,他引:0  
The kinetics of nitrate (NO3 ) uptake, the maximum uptake velocity (Vm) and the half-saturation constant (Ks), were determined for 18 species of batch-cultured freshwater algae grown without nitrogen limitation. Values of Ks ranged from 0.25 to 6.94 µM l–1 Chlorella pyrenoidosa Chick, and Navicula pelliculosa (Breb.) Hilse, respectively. Values of Vm ranged from 0.51 to 5.07 µM l–1 h–1 for Anabaena A7214 and Nitzschia W-32 O'Kelley, respectively. The mean positive values of Ks for Chlorophyta, Cyanophyta and Chrysophyta were 1.89, 3.67 and 4.07 µM l–1, respectively. The mean values of Vm for the same phyla were 1.61, 1.02 and 2.97 µM l–1 h–1 105 cells–1, respectively. The ranges of these kinetic parameters encompass values of kinetic parameters for marine and freshwater species in batch culture, for freshwater algae grown in N-limited chemostats and for natural populations of freshwater phytoplankton. Thus, in spite of variability between species, uptake parameters for both marine and freshwater algae are identical.  相似文献

When an initial cell loading of about 30–40 µg chlorophyll (Chl)·g–1 gel and alginate suspension of 3% (w/v) were used for immobilization of Chlamydomonas reinhardtii, the resulting cell beads showed optimum nitrite uptake rate, at 30° C and pH 7.5, of 14 µmol NO inf2 sup– ·mg–1 Chl·h–1, the photosynthetic and respiratory activities being about 120 µmol O2 produced·mg–1 Chl·h–1, and 40 µmol O2 consumed ·mg–1 Chl·h–1, respectively. The nitrite uptake activity required CO2 in the culture and persisted after 8 days of cells immobilization, or in the presence of 0.2 mm ammonium in the medium. Our data indicate that alginate-entrapped C, reinhardtii cells may provide a stable and functional system for removing nitrogenous contaminants from waste-waters.Correspondence to: C. The uptake of nitrate, ammonium and phosphate was examined in vitro in seedlings of the seagrass Amphibolis antarctica ((Labill.) Sonder ex Aschers.). Uptake of all three nutrients was significantly correlated with external concentration up to 800 µ g l–1. The uptake of nitrate (0–200 µ g NO3-N g dry wt–1 h–1) was significantly lower than the uptake of ammonium (0–500 µ g NH4-N g dry wt–1 h–1), suggesting that the seedlings have a higher affinity for this form of nitrogen in the water column.Data were in general agreement with uptake rates recorded for other seagrasses, notably Zostera marina. In comparison to the dominant macroalgae for the same region, seedlings had either similar or higher uptake rates in relation to external concentration, lending support to the hypothesis that seedlings, which do not possess roots, behave like macroalgae in terms of nutrient acquisition from the water column.A comparison with literature data on adult seagrass suggests, however, that seagrasses show lower uptake rates than macroalgae suggesting that the macroalgae, which are totally reliant on the water column for nutrients, are more efficient at uptake than seagrasses which may potentially use the sediment for a nutrient source.  相似文献

Physical and chemical conditions, particulate matter and N-uptake were characterized at two sampling sites at the eastern German coast of the Baltic Sea (Pomeranian Bay) over the annual period of 1997 (February–November). The inshore sampling sites (5 m water depth) differed with respect to the potential influences of river run-off and salt water exchange (mean values of salinity: 7.05 and 8.72 PSU), respectively. The mean org-Cdiss/org-Cpart-ratios (4.9 and 12.6) fell in the same order of magnitude (1.0–12.6) as values of neighboring inshore waters, and increasing values reflect an enhancement of the trophic level. Beside differences of nitrogen concentrations (dissolved inorganic nitrogen: 1.8–23.8 and 0.9–9.9 mol l–1), particulate nitrogen (4.30–41.01 and 2.69–9.08 mol l–1) and absolute uptake of N-nutrients (mean sum of NH4 +, urea, NO3 uptake rates: 0.141 and 0.087 mol l–1 h–1), specific uptake of 15N-labelled nutrients (NH4 +, urea, NO3 ) as well as the relationships between the measured variables characterize distinguishable inshore systems. The high variability at the shallow sampling sites prevents, however a simple resolution of the seasonal courses. Light dose could be identified as a potential key in order to describe long-term variations of N-uptake at the station with higher organic matter concentration (station KW), but phytoplankton development is better reflected in the seasonal course of N-uptake at the other station. Specific nitrogen uptake rates (NH4 +: 0.0009–0.0353 h–1, urea: 0.0001–0.0137 h–1, NO3 : 0.000004–0.0009 h–1) and relative nitrogen preferences indicate extraordinary importance of reduced nitrogenous nutrients (NH4 +, urea) at both stations throughout the year.  相似文献

A new automated procedure for nanomolar nitrate analysis was applied to the study of uptake of low nitrate concentrations (< 100 ngat l–1) by phytoplankton. The precision of this analytical method (± 3 ngat l–1) made it possible to monitor the absorption of very low quantities of nitrate over short term periods by a low cell-density culture of the marine diatom Phaeodactylum tricornutum, where the levels of particulate nitrogen and chlorophyll were equivalent to those found in oligotrophic areas (0.5 µgat N l–1 and 0.4 µg l–1 respectively). By continuous monitoring of nitrate disappearance from the culture medium, we are able to describe accurately the transient uptake responses of the diatom after a spike addition of trace quantities of nitrate and thus to provide new information on the still largely unknown small-scale phenomenon of pulsed nitrate supply in the upper layer of stratified oceans and rapid uptake of these nitrate patches by phytoplankton.The results show that a N-limited culture of Phaeodactylum tricornutum is immediately capable of taking up trace quantities of nitrate (< 100 ngat l–1) at high rates (0.10–0.14 h–1) . These initial rates are one order of magnitude higher than the theoretical rates calculated from the Michaelis-Menten equation and are close to the level of V max (0.15 h–1) obtained when cells are exposed to saturating nitrate concentrations. This rapid initial uptake would be a considerable advantage in oligotrophic areas where nanomolar nitrate supply is thought to be episodic. The present results suggest that phytoplankton evolve adaptations to utilize the available nitrate at the spatial and temporal scales at which it occurs. On the other hand, we can consider this physiological adaptation as evidence of nitrate pulses in the field which would invalidate the steady-state approach to the oligotrophic ecosystems.  相似文献

The turnover times of glucose, averaged for 0–10 m in the upper waters of Lake Kinneret and measured by the addition of single or multiple concentrations of substrate, ranged from 23 to 188 hours and 1 to 87 hours respectively. Potential uptake rates (estimated as Vmax) ranged from 0.095 to 1.94 µg glucose l–1h–1, while measured uptake rates varied from 0.09 to 1.1 µg glucose l–1h–1. Concentrations of dissolved carbohydrates and glucose averaged 0.71 mg glucose equivalents l–1 and 39 µg glucose l–1 respectively. No evident relationships between glucose cycling and any fractions of dissolved organic matter, phytoplankton biomass or primary productivity were found. Turnover times were generally most rapid immediately after the decline of the spring Peridinium bloom. The respiration percentage of incorporated glucose ranged from 25% to 61% with highest values during the summer months. Respiration may be influenced by the nature of the indigenous bacterial population as well as by temperature. Daily heterotrophic glucose carbon uptake was about 9% of the photosynthetic incorporation and could provide a bacterial yield of about 7 × 104 ml–1d–1.  相似文献

The uptake of ammonium, nitrate and phosphate by laboratory-grown young sporophytes of Laminaria abyssalis was measured in a perturbed system (batch mode) at 18 °C and 35 ± 5 µE m–2 s–1 photon flux density. Uptake of all appeared to follow saturation-type nutrient uptake kinetics. The NO inf3 sup– (K s = 14.0 µM, V max = 5.0 µmol h–1 g–1 dry wt) and NH inf4 sup+ (K s = 4.6 µM, V max= 2.0 µmol h–1 g–1 dry wt) were taken up simultaneously, although NH inf4 sup+ was taken up more rapidly. Values of K 3 and V max for phosphate were, respectively, 2.21 µM and 0.83 µmol h–1 g–1 dry wt. Nitrate and phosphate were both consumed in similar rates (V max /Ks 0.37) at low concentrations. NH inf4 sup+ , thus, might be a more efficient form of N fertilizer if artificial enrichment of seawater is used.  相似文献

Both conventional and genetic engineering techniques can significantly improve the performance of animal cell cultures for the large-scale production of pharmaceutical products. In this paper, the effect of such techniques on cell yield and antibody production of two NS0 cell lines is presented. On the one hand, the effect of fed-batch cultivation using dialysis is compared to cultivation without dialysis. Maximum cell density could be increased by a factor of ~5–7 by dialysis fed-batch cultivation. On the other hand, suppression of apoptosis in the NS0 cell line 6A1 bcl-2 resulted in a prolonged growth phase and a higher viability and maximum cell density in fed-batch cultivation in contrast to the control cell line 6A1 (100)3. These factors resulted in more product formation (by a factor ~2). Hydrological and chemical structures off the Rhône River estuary resulting from the introduction of the river flow into the Mediterranean Sea are described. The effect of the fresh-water/sea-water interface on the distribution of inorganic and organic matter off the Rhône river is investigated. Strong vertical gradients of inorganic and dissolved organic matter such as lipids characterized the first few meters in this area (from 83.7 to 0.6 N-NO3 µgat l–1, from 6.39 to 0.92 N-NH4 µgat l–1 and from 299 to 73 µg l–1 of total dissolved lipids). At the interface, substantial increases of particulate organic (PON: from 45 µg l–1 at surface to 118 µg l–1 at the interface, POC: from 462 to 876 µg l–1, total particulate lipids: from 33 to 648 µg l–1) and suspended matter in general (from 18 to 22.2 mg l–1) were observed. High phytoplanktonic production may account for some of this enrichment, although passive accumulation might also be involved.  相似文献

Exchange of phosphate between components of the reef bottom and the water column were studied on reefs around Heron Island (Great Barrier Reef), both in aquaria and in in situ enclosures, using radioactive phosphorus (32P) as a tracer. Living corals, dead corals, coral rubble overgrown with periphyton, and soft sediments of coral sand were used in experiments. In all of these components of bottom reef biotopes, two opposite flows of inorganic phosphate were recorded and measured, i.e. the rate of PO4-P uptake from water (Ac), and its release (Ae). At ambient PO4-P concentrations in water of 0.1– 0.3 µmoll–1, both flows varied in living corals and coral rubble between 10 and 70 µg P kg–1 h–1, 3–10 mg P m–2 day–1, and in coral sand between 10 and 30 µg P kg–1 h–1, or 2–7 mg P m–2 day–1. Under the latter concentration range (which is typical for coral reef areas), the reciprocal PO4-P flows almost balanced each other, so that net uptake (At) was very low. Often it approached zero or was positive, showing that a net PO4-P release had taken place. The uptake flow (Ac) in living coral was much more dependent on the PO4-P content in overlying water than was the release flow (Ae). The influence of conditions of illumination upon the values of Ac and Ae was comparatively low. The data obtained are used to discuss problems of phosphorus balance and dynamics in coral reef ecosystems.  相似文献

Phosphorus (PO4-P) and nitrogen (NH4-N) excretion rates of Euchlanis dilatata lucksiana, a rotifer, isolated from Lake Loosdrecht (The Netherlands) and cultured in the lake water at 18–19 °C, were measured in the laboratory.In a series of experiments, the effects of experiment duration on the P and N excretion rates were examined. The rates measured in the first half-hour were about 2 times higher for P and 2–4 times for N than the rates in the subsequent three successive hours which were quite comparable.Eight experiments were carried out in triplicate, 4 each for P and N excretion measurements, using animals of two size ranges: 60–125 µm and > 125 µm. The specific excretion rates varied from 0.06 to 0.18 µg P.mg–1 DW.h–1 and 0.21 to 0.76 µg N.mg–1 DW.h–1. Generally an inverse relationship was observed between the specific excretion rates and the mean individual weight. The excretion rates of Euchlanis measured by us are lower than those reported for several other rotifer species, most of which are much smaller than Euchlanis.Extrapolating the excretion rates of Euchlanis to the other rotifer species in Lake Loosdrecht, and accounting for their density, size and temperature, rotifer excretion appears to be a significant, potential nutrient (N,P) source for phytoplankton growth in the lake. The excretion rates for the rotifers appear to be about two thirds of the total zooplankton excretion, even though the computed rotifer mean biomass is about one-third of the total zooplankton biomass.  相似文献

The filtering rates of14C labelledChlamydomonas sp. by 3 dominant species of rotifers were studied in eutrophic Lake Aydat. They varied from 4 to 53 µl ind–1 h–1 forKeratella cochlearis, from 2 to 56 µl.–1h–1 forKeratella quadrata and from 3 to 52 µl ind–1 h–1 forKellicottia longispina. Their maximum assimilation efficiency was 32%. At the measured grazing rates, these populations could clear the water in less than two days during July. In Lake Aydat, the rotifers community could play an important role in the regulation of seasonal succession of phytoplankton and bacteria.  相似文献

Feeding experiments with two species of carnivorous copepod, Mesocyclops longisetus (Thiebaud) and Mesocyclops kieferi Van de Velde from Barra Bonita, a eutrophic reservoir in São Paulo, Brasil, were performed using two common types of prey: Ceriodaphnia cornuta, a cladoceran, with a mean body length of 464 µm (including spines) or 393 µm (without spines), and Brachionus calyciflorus, a rotifer with a mean body length of 350 µm (including spines) or 279 µm (without spines).Both species showed higher consumption rates on Brachionus than on Ceriodaphnia. For Mesocyclops longisetus, the average rates were: 2.19 prey ind–1 h–1 (Brachionus), and 1.30 prey ind–1 h–1 (Ceriodaphnia). For Mesocyclops kieferi, the rates were 1.85 prey ind–1 h–1 (Brachionus) and 0.60 prey ind–1 h–1 (Ceriodaphnia). In situ uptake of inorganic carbon and nitrogen by the aquatic liverworts Jungermannia vulcanicola Steph. and Scapania undulata (L.) Dum. was measured in an acid stream, Kashiranashigawa, Japan. The uptake activities were similar in the both species. The activities were highest at the tip of shoots, and decreased gradually towards the base. Carbon uptake at the tip in the light was 10.4 × 10–4 for J. vulcanicola and 8.1 × 10–4 g C g dry wt–1 h–1 1 for S. undulata. Ammonium was effectively incorporated into the shoots, and the uptake activity at the tip was between 1.9 × 10–5 and 5.8 × 10–5 g N g dry wt–1 h–1. Nitrate uptake was smaller than ammonium uptake. The ratio of dark to light uptake in ammonium uptake experiments was larger than that in carbon uptake experiments. These results suggest that the liverworts use ammonium as a major nitrogen source, and that ammonium uptake was less dependent on light than carbon uptake.  相似文献

The clearance rates (CRs) of bacteria by Filinia longiseta were measured at 19°C, both in situ in Lake Loosdrecht and in the laboratory during summer. The tracer particles used in the field were: (1) 0.51 µm fluorescent microspheres, and (2) fluorescently labelled bacteria (FLB). A third type of tracer particle, natural [methyl-3H]-thymidine-labelled bacteria (< 1.2 µm), were used as a radiotracer in a laboratory experiment. The uptake of the first two tracer-particle types was measured by microscopic examination of the rotifer guts. In the third case, the uptake of radioactivity was determined by liquid scintillation counting. The rate of uptake of the microspheres decreased 10 min after the start of the experiment, probably because the gut passage time was exceeded. Using a 5 min feeding time, the rate of uptake of microspheres was higher than that of the FLB, though the variation in the uptake in both cases was high. The ingestion rates and CRs of bacteria by F. longiseta based on the fluorescent tracers were: microspheres, 5115 bact.ind–1 h–1 and 0.368 µl ind–1 h–1; FLB, 2252 bact.ind–1 h–1 and 0.162 µl ind–1 h–1. The mean CR using the thymidine-labelled natural bacteria and a 10 min feeding time was 0.179 µl ind–1 h–1. Thus, the CR based on the microsphere method was twice as high as for the other two methods.  相似文献

It is generally assumed that plant assimilation constitutes the major sink for anthropogenic Nitrate NO 3 deposited in temperate forests because plant growth is usually limited by nitrogen (N) availability. Nevertheless, plants are known to vary widely in their capacity for NO 3 uptake and assimilation, and few studies have directly measured these parameters for overstory trees. Using a combination of field and greenhouse experiments, we studied the N nutrition of Acer saccharum Marsh. in four northern hardwood forests receiving experimental NO 3 additions equivalent to 30 kg N ha–1 year–1. We measured leaf and fine-root nitrate reductase activity (NRA) of overstory trees using an in vivo assay and used 15N to determine the kinetic parameters of NO 3 uptake by excised fine roots. In two greenhouse experiments, we measured leaf and root NRA in A. saccharum seedlings fertilized with 0–3.5 g NO 3 –N m–2 and determined the kinetic parameters of NO 3 and NH 4 + uptake in excised roots of seedlings. In both overstory trees and seedlings, rates of leaf and fine root NRA were substantially lower than previously reported rates for most woody plants and showed no response to NO 3 fertilization (range = non-detectable to 33 nmol NO 2 g–1 h–1). Maximal rates of NO 3 uptake in overstory trees also were low, ranging from 0.2 to 1.0 mol g–1 h–1. In seedlings, the mean V max for NO 3 uptake in fine roots (1 mol g–1 h–1) was approximately 30 times lower than the V max for NH 4 + uptake (33 mol g–1 h–1). Our results suggest that A. saccharum satisfies its N demand through rapid NH 4 + uptake and may have a limited capacity to serve as a direct sink for atmospheric additions of NO 3 .  相似文献

Sulphate uptake by Amphidinium carterae, Amphidinium klebsii and Gymnodinium microadriaticum grown on artificial seawater medium with sulphate, cysteine, methionine or taurine as sulphur source occurred via an active transport system which conformed to Michaelis-Menten type saturation kinetics. Values for K m ranged from 0.18–2.13 mM and V max ranged from 0.2–24.2 nmol · 105 cells–1 · h–1. K m for symbiotic G. microadriaticum was 0.48 mM and V max was 0.2 nmol · 105 cells–1 · h–1. Sulphate uptake was slightly inhibited by chromate and selenate, but not by tungstate, molybdate, sulphite or thiosulphate. Cysteine and methionine (0.1 mM), but not taurine, inhibited sulphate uptake by symbiotic G. microadriaticum, but not by the two species of Amphidinium. Uptake was inhibited 45–97% under both light and dark conditions by carbonylcyanide 3-chlorophenylhydrazone (CCCP); under dark conditions sulphate uptake was 40–60% of that observed under light conditions and was little affected by 3-(3,4-dichlorophenyl) 1,1-dimethylurea (DCMU).The uptake of taurine, cysteine and methionine by A. carterae, A. klebsii, cultured and symbiotic G. microadriaticum conformed to Michaelis-Menten type saturation kinetics. K m values of taurine uptake ranged from 1.9–10 mM; for cysteine uptake from 0.6–3.2 mM and methionine from 0.001–0.021 mM. Cysteine induced a taurine uptake system with a K m of 0.3–0.7 mM. Cysteine and methionine uptake by all organisms was largely unaffected by darkness or by DCMU in light or darkness. CCCP significantly inhibited uptake of these amino acids. Thus energy for cysteine and methionine uptake was supplied mainly by respiration. Taurine uptake by A. carterae was independent of light but was inhibited by CCCP, whereas uptake by A. klebsii and symbiotic G. microadriaticum was partially dependent on photosynthetic energy. Taurine uptake by cultured G. microadriaticum was more dependent on photosynthetic energy and was more sensitive to CCCP. Cysteine inhibited uptake of methionine and taurine by cultured and symbiotic G. microadriaticum to a greater extent than in the Amphidinium species. Methionine did not greatly affect taurine uptake, but did inhibit cysteine uptake. Taurine did not affect the uptake of cysteine or methionine.  相似文献

The development of a filamentous, nitrogen-fixing cyanobacterial bloom was followed during July–August 1990 in a stratified basin in the central Gulf of Finland, Baltic Sea. Hydrography, dissolved inorganic, particulate and total nutrients, chlorophyll a, alkaline phosphatase activity, 32PO4-uptake and phytoplankton species were measured. The study period was characterized by wind-induced mixing events, followed by marked nutrient pulses and plankton community responses. Phosphate uptake was highest throughout the study period in the size fraction dominated by bacteria and picocyanobacteria (< 2 µm) and the proportion of uptake in the size fraction 2–10 µm remained low (2–6%). Higher phosphate turnover times were observed in a community showing signs of enhanced heterotrophic activity. The bloom of filamentous, nitrogen-fixing cyanobacteria Aphanizomenon flos-aquae was promoted by a nutrient pulse with an inorganic nutrient ratio (DIN:DIP) of 15. The results show that the quality, frequency and magnitude of the physically forced nutrient pulses have an important role in determining the relative share of the different modes of phosphorus utilization and hence in determining the cyanobacterial bloom intensity and species composition in the Baltic Sea.  相似文献

The CO2 production of individual larvae of Apis mellifera carnica, which were incubated within their cells at a natural air humidity of 60–80%, was determined by an open-flow gas analyzer in relation to larval age and ambient temperature. In larvae incubated at 34 °C the amount of CO2 produced appeared to fall only moderately from 3.89±1.57 µl mg–1 h–1 in 0.5-day-old larvae to 2.98±0.57 µl mg–1 h–1 in 3.5-day-old larvae. The decline was steeper up to an age of 5.5 days (0.95±1.15 µl mg–1 h–1). Our measurements show that the respiration and energy turnover of larvae younger than about 80 h is considerably lower (up to 35%) than expected from extrapolations of data determined in older larvae. The temperature dependency of CO2 production was determined in 3.5-day-old larvae, which were incubated at temperatures varying from 18 to 38 °C in steps of 4 °C. The larvae generated 0.48±0.03 µl mg–1 h–1 CO2 at 18 °C, and 3.97±0.50 µl mg–1 h–1 CO2 at 38 °C. The temperature-dependent respiration rate was fitted to a logistic curve. We found that the inflection point of this curve (32.5 °C) is below the normal brood nest temperature (33–36 °C). The average Q10 was 3.13, which is higher than in freshly emerged resting honeybees but similar to adult bees. This strong temperature dependency enables the bees to speed up brood development by achieving high temperatures. Finfish culture is a growing industry, and it causes a nutrient loading problem. To investigate the feasibility of an integrated culture of kelp and salmon, 15-cm long kelp (Laminaria saccharina) was grown in salmon culture effluent. The objectives were to test the effects of flow rate and kelp density on dissolved inorganic nitrogen removal (DIN), and DIN uptake and growth by the kelp. NH 4 + , NO 3 and DIN (NH 4 + + NO 3 ) loadings were in the ranges 6.2–25.4, 12.9–40.0, 19.7–52.7 mol 1–1, respectively, over the experimental period.Surplus uptake of nitrogen was not evident, because the C:N ratio (10–11) was constant in all experiments. During light periods, the kelp removed from 170–339 mol 1–1 h–1, and approximately 26–40% of the incoming DIN. The DIN uptake rate, based on daylight sampling periods, ranged between 6.1–22.5 mol g–1 dry mass h–1. The highest-flow rate, lowest-density tank had the highest DIN uptake rate. Debris from the fish effluent settling on the kelp thalli in the low-flow rate tanks affected uptake. Mean DIN uptake rate based on 3 days of growth for all flow-density combinations ranged between 5.4–8.3 mol g–1 dry mass h–1. The kelp utilized NH 4 + and NO 3 equally.The growth ranged between 6.5–9% d–1. The biomass production ranged from 1–2 g per sampling period. The highest growth rate and biomass production were achieved by kelp in the highest-flow rate, lowest-density tank. Lower DIN concentrations due to higher DIN removal rates in the other tanks and light limitation due to self-shading in the high-density tanks were probably responsible for the reduced growth rate in these tanks.Author for correspondence  相似文献
