Summary The central projections of ocellar interneurones in two species of trichopterous insects Agrypnia varia F. and Limnephilus flavicornis F. were analysed by use of cobalt iontophoresis. The interneurones were classified into three groups: large-, medium- and small-caliber neurones based on the diameters of the axons. Seven large-diameter neurones project from each lateral ocellus into the central nervous system. Of these, four neurones terminate in the posterior slope (three ipsilateral and one contralateral). Three neurones possess branches in the contralateral posterior slope and proceed down the cervical connective into the thoracic ganglia. Medium-sized neurones connect the neuropiles of the three ocelli to each other. Small-diameter neurones contact the contralateral lobula and medulla of the optic lobes and connect the three ocellar neuropiles. Large-diameter neurones of the median ocellus were found to terminate bilaterally or ipsilaterally in the posterior slope. In the posterior slope four different subregions can be recognised: (1) the dorso-lateral, (2) the ventro-lateral, (3) the lateral, into which large-diameter interneurones of the lateral ocelli send branches, and (4) the medial, innervated by interneurones of the median ocellus. Interneurones of the median ocellus send branches into the lateral region as well.  相似文献

A new dipeptide was isolated from several tissues of Agkistroden blomhoffi (mamushi: a venomous snake in Japan), using ion-exchange resins and thin-layer chromatography. It was identified as O-phosphoserylethanolamine by mass spectrometry and comparison with synthetic compounds using several methods. This compound was contained in several mamushi tissues including the liver, heart, brain, bile, and muscle. The concentrations of O-phosphoserylethanolamine in the liver, brain, muscle, skin, heart, and bile were 7.17+/-3.11,16.98+/-4.25,37.37+/-7.88,37.56+/-8.97,23.93+/-6.11, and 22.21+/-5.76 micromol/g, respectively.  相似文献

Biogenic purine crystals function in vision as mirrors, multilayer reflectors and light scatterers. We investigated a light sensory organ in a primarily wingless insect, the jumping bristletail Lepismachilis rozsypali (Archaeognatha), an ancestral group. The visual system of this animal comprises two compound eyes, two lateral ocelli, and a median ocellus, which is located on the front of the head, pointing downwards to the ground surface.We determined that the median ocellus contains crystals of xanthine, and we obtained insights into their function. To date, xanthine biocrystals have only been found in the Archaeognatha. We performed a structural analysis, using reflection light microscopy, cryo-FIB-SEM, microCT and cryo-SEM. The xanthine crystals cover the bottom of a bowl-shaped volume in the median ocellus, in analogy to a tapetum, and reflect photons to light-sensitive receptors that are spread in the volume without apparent order or preferential orientation. We infer that the median ocellus operates as an irregular multifocal reflector, which is not capable of forming images. A possible function of this organ is to improve photon capture, and by so doing assess distances from the ground surface when jumping by determining changes in the intensity and contrast of the incident light.  相似文献

A new genus of Metopiinae, Ojuelos Khalaim, gen. n., with a single species, Ojuelos juachicus Khalaim & Ruíz-Cancino, sp. n., is described from Jalisco Province in central Mexico. Ojuelos belongs to the group of three genera (Cubus Townes & Townes, Colpotrochia Holmgren and Triclistus Förster) having a high lamella separating the antennal sockets and reaching the median ocellus (this lamella possesses a dorsal groove in it, just below the median ocellus), but differs from these genera primarily by 1) face and clypeus not convex in lateral view, 2) face separated from clypeus by a shallow transverse impression, 3) mandible with lower tooth very strongly reduced, 4) flagellomeres of antenna strongly transverse, and 5) dorsal carinae of propodeum reduced. A portion of the key to the genera of Metopiinae is provided to distinguish the new genus.  相似文献

A new species of Anteon Jurine, 1807 is described from Laos, Houaphanh Province: Anteon holzschuhi sp. n. Morphologically the new species is similar to Anteon semipolitum Olmi, 2008, but it is distinguished by the sculpture of the face partly reticulate rugose and partly with deep punctures similar to areolae; in Anteon semipolitum the face is completely punctate and unsculptured among punctures. In addition, in the new species the distance from the outer edge of a lateral ocellus to the compound eye (OOL) is about 3.3 times as long as the distance between the inner edges of a lateral ocellus and the median ocellus (OL); in Anteon semipolitum OOL is less than twice as long as OL. Published identification keys to the Oriental species of Anteon are modified to include the new species.  相似文献

Background

Glioblastoma is the most common and most lethal form of brain tumor in human. Unfortunately, there is still no effective therapy to this fatal disease and the median survival is generally less than one year from the time of diagnosis. Discovery of ligands that can bind specifically to this type of tumor cells will be of great significance to develop early molecular imaging, targeted delivery and guided surgery methods to battle this type of brain tumor.

Methodology/Principal Findings

We discovered two target-specific aptamers named GBM128 and GBM131 against cultured human glioblastoma cell line U118-MG after 30 rounds selection by a method called cell-based Systematic Evolution of Ligands by EXponential enrichment (cell-SELEX). These two aptamers have high affinity and specificity against target glioblastoma cells. They neither recognize normal astraglial cells, nor do they recognize other normal and cancer cell lines tested. Clinical tissues were also tested and the results showed that these two aptamers can bind to different clinical glioma tissues but not normal brain tissues. More importantly, binding affinity and selectivity of these two aptamers were retained in complicated biological environment.

Conclusion/Significance

The selected aptamers could be used to identify specific glioblastoma biomarkers. Methods of molecular imaging, targeted drug delivery, ligand guided surgery can be further developed based on these ligands for early detection, targeted therapy, and guided surgery of glioblastoma leading to effective treatment of glioblastoma.  相似文献

Despite advances in diagnostic imaging and drug discovery, primary malignant brain tumors remain fatal. Median survival for patients with the most severe forms is rarely past eight months. The severity of the disease and the lack of substantial improvement in patient survival demand that new approaches be explored in drug delivery to brain tumors. Recently, local delivery of chemotherapy to brain tumors has provided a way to circumvent the blood-brain barrier, allowing delivery of chemotherapy drugs directly to malignant cells in the brain. Two methods of local delivery have been developed: polymeric-controlled release and convection-enhanced delivery. Controlled release utilizes degradable or non-degradable polymers as carriers of chemotherapy; polymer implants or microparticles are implanted locally to introduce a sustained source of drug for periods of days or months. Convection-enhanced delivery employs the bulk flow of drugs dissolved in fluid, which is introduced intracranially using a catheter and pump. The convective fluid flow is capable of delivering drugs great distances within the brain, potentially treating invasive cells at a distance from the catheter infusion site. These two new delivery strategies are capable of delivering both standard chemotherapeutic drugs and new methods of anti-cancer therapy. Taken individually, or used in tandem, they represent a potential revolution in brain cancer treatment.  相似文献

Treatment of late-stage human African trypanosomiasis is complicated by the presence of trypanosomes within the central nervous system (CNS). The regimen commonly prescribed to treat CNS-stage disease involves the use of the trypanocidal drugs suramin and melarsoprol. Suramin does not cross the blood–brain barrier efficiently and therefore, at normal dosages, will not cure CNS-stage infections. An initial treatment with suramin is given to eliminate the parasites from the peripheral tissues. This is followed by a course of intravenous melarsoprol, which can enter the CNS. However, melarsoprol not only produces severe adverse reactions but also is extremely painful to administer. One possible method to help alleviate these problems is to reduce the total amount of melarsoprol in the treatment regimen. This study indicates a synergism between suramin and melarsoprol and demonstrates that experimental murine CNS-trypanosomiasis can be cured with a single intraperitoneal dose of 20 mg/kg suramin followed almost immediately by 0.05 ml (4.5 μmol) topical melarsoprol. These dosages will not cure the infection when administered as monotherapies. Moreover, the timing of the drug administration appears to be crucial to the successful outcome of the regimen. If the interval between injection of suramin and application of topical melarsoprol is extended from 15 min to 3 or 7 days, the infections are not cured. Although extended relapse times occur following these regimens when compared with monotherapy approaches. Thus, there is strong evidence that injected suramin and topical melarsoprol should be given almost simultaneously to achieve the most effective combination of the two drugs.  相似文献

A major challenge for the treatment of many central nervous system (CNS) disorders is the lack of convenient and effective methods for delivering biological agents to the brain. Mucopolysaccharidosis II (Hunter syndrome) is a rare inherited lysosomal storage disorder resulting from a deficiency of iduronate-2-sulfatase (I2S). I2S is a large, highly glycosylated enzyme. Intravenous administration is not likely to be an effective therapy for disease-related neurological outcomes that require enzyme access to the brain cells, in particular neurons and oligodendrocytes. We demonstrate that intracerebroventricular and lumbar intrathecal administration of recombinant I2S in dogs and nonhuman primates resulted in widespread enzyme distribution in the brain parenchyma, including remarkable deposition in the lysosomes of both neurons and oligodendrocytes. Lumbar intrathecal administration also resulted in enzyme delivery to the spinal cord, whereas little enzyme was detected there after intraventricular administration. Mucopolysaccharidosis II model is available in mice. Lumbar administration of recombinant I2S to enzyme deficient animals reduced the storage of glycosaminoglycans in both superficial and deep brain tissues, with concurrent morphological improvements. The observed patterns of enzyme transport from cerebrospinal fluid to the CNS tissues and the resultant biological activity (a) warrant further investigation of intrathecal delivery of I2S via lumbar catheter as an experimental treatment for the neurological symptoms of Hunter syndrome and (b) may have broader implications for CNS treatment with biopharmaceuticals.  相似文献

The present investigation describes the effect of the spasmolytic benzylbenzoates 1-9 from Brickellia veronicifolia on CaM using a functional in vitro enzymatic assay. Bovine brain PDE1 was used as a monitoring enzyme. The most active natural inhibitors of the system CaM-PDE1 were benzyl benzoates 3-5, which inhibited the activity of PDE1 in a concentration-dependent manner. In addition, three series of analogs of compound 4, compounds 10a-32a, were prepared and assayed. The benzyl benzoates from the first series, namely 10a-24a, possess no substituents on ring B but different number and position of hydroxyl or methoxy groups in ring A. The second group (25-32a), on the other hand, possesses an A ring identical to that on compound 4, but different substituents in Ring B. The most active compounds were 14a, 15a and 30a. These compounds were two to six times more potent than chlorpromazine, a well known CaM inhibitor. Benzyl benzoates 14a and 15a have methoxyl groups at C-2/C-4 and C-3/C-4 in ring A, respectively; while 30a, in addition to the methoxyl groups at C-2/C-6 of ring A, hold a benzoyloxy moiety at C-3' of ring B. Kinetic studies revealed that compounds 3, 4, 14a, 15a and 30a behave as competitive CaM antagonists.  相似文献

From the acetone extract of Teucrium tomentosum, a new antifeedant neo-clerodane diterpenoid teuctosin (1) was isolated along with teuflin (2), teucrin-H(2) (3), 6beta-hydroxyteuscordin (4), 6beta-acetylteuscordin (5) and montanin-D (6). The structure of the new compound was elucidated comprehensively using 1D and 2D NMR methods and confirmed by X-ray crystallography. All the compounds showed effective antifeedancy against Plutella xylostella and Spodoptera litura at 10 mug/cm(2) of leaf area.  相似文献

The nauplius eye consists of one median and two lateral ocelli, each within a pigment cup. The three pigment cups are made up from two multi-nucleate pigment cells: each cell forming one lateral cup and half of the median cup. The three cups are lined on the insides by tapetal cells which contain layers of reflectile crystals. Each of the ocelli contains six sensory cells which protrude from the rims of the pigment cups and the protruding parts are sheathed by the conjunctiva cells. The whole eye is enveloped by a thin membrane which also sheaths the proximal parts of the five nerve bundles that leave the eye. All the sensory cells of the lateral ocelli are similar and have rhabdomeric microvilli on the terminal end, and contain phaosomes and a multitude of other organelles and cytoplasmic inclusions. The complex median ocellus contains a superior group of three retinular cells, linked by interdigitating processes, and an inferior group consisting of a large central cell enclosed in two cup-shaped peripheral retinular cells. A two-tiered rhabdome arrangement exists, with a rather complex inferior rhabdome set made up of a central rhabdomere and two hemi-annulate rhabdomeres. The cytoplasm of the retinular cells of the median ocellus lack phaosomes but instead contain double-walled tubular elements, possibly formed by the inpushings of microvilli into adjacent cells. The possible functional significance of the unique arrangement seen in the median ocellus is discussed. The retinular cells are of the inverse type. There are no efferent nerve fibres from the brain nor any nervous connection between the lateral and the median ocelli.  相似文献

本研究通过形态解剖和BrdU免疫组织化学方法对东方蜜蜂Apis cerana Fabricius背单眼的胚后发育过程进行了比较研究,结果表明:东方蜜蜂的每一个背单眼都包括角膜晶体、角膜生成细胞、小网膜细胞以及后部的单眼神经。蜜蜂的背单眼起源自头壳上皮;其胚后发育的高峰期集中在蛹发育的前3d;其新细胞主要来源于上皮细胞和圆锥形单眼囊周围细胞的有丝分裂;单眼同脑的联系在P1期前后就已经建立;角膜晶体的形成在P5以后。说明单眼的结构和发育同其功能密切相关。  相似文献

利用蚊虫连续石蜡切片免疫组织化学技术,对登革Ⅱ型病毒(DEN-2)感染白纹伊蚊Aedes albopictus后的散播时间、程度及组织器官的感染顺序进行监测,以了解DEN-2在媒介白纹伊蚊体内的分布规律。结果表明:大剂量感染登革Ⅱ型病毒后,在蚊虫消化道的主要部位以及大多数组织器官包括神经及内分泌系统在内,如涎腺、脑、神经节等亦检测到病毒抗原。登革Ⅱ型病毒一旦感染并逸出中肠会迅速侵染其它组织。从各组织感染率的高低推断,病毒逸出中肠后通过血淋巴传播到其它组织的顺序通常为:前肠、涎腺、咽部神经节、脑及食管下神经节、后肠及复眼的小眼等。  相似文献

Platinum-based drugs have been used for several decades to treat various cancers successfully. Cisplatin is the original compound in this class; it cross-links DNA, resulting in cell cycle arrest and cell death via apoptosis. Cisplatin is effective against several tumor types but exhibits toxic side effects; in addition, tumors often develop resistance. An original in vitro approach is proposed to determine whether platinum-based research compounds are good candidates for further study by comparing them to marketed drugs using FTIR spectroscopy and the COMPARE analysis from the NCI. Both methods can produce fingerprints and highlight differences between the compounds, classifying the candidates and revealing promising derivatives.  相似文献

Following the rapid proliferation of organic pollutants in the surface water, the application of microfiltration technology has been extensively studied for its treatment since the 1990s. Given that the conventional treatment processes were unable to treat the excessive dissolved organic compounds, microfiltration technologies have gained momentum as effective solutions to treat the surface water. The efficacy of low-pressure membrane filtration technologies such as microfiltration and ultrafiltration has been under scrutiny ever since, and numerous research studies have aimed at enhancing their capabilities to reject the suspended solids and organic matters. This paper reviews the development trajectory of membrane technology, ranging from microfiltration to membrane bioreactors, for treating dissolved organic matters in surface water and their future potential. This is a critical review of the physicochemical and biological options such as, but not limited to, pretreatment of water using coagulation, ozonation, adsorption and/or a combination of these. On the whole, it is concluded that the membrane bioreactor system, which combines biological process and physical rejection, showed high potential in treating polluted surface water, which needs to be further investigated extensively to promote its application in water treatment plants.  相似文献

In these studies a constant ERG response was used as a measure of visual sensitivity to different wavelengths of light. The dark-adapted compound eye of Tenodera sinensis is dominated by a single class of photoreceptors. with a major peak of sensitivity at about 510–520 nm, and with a minor peak of sensitivity in the near-ultraviolet region at about 370 nm. The dark-adapted dorsal ocellus does not contain a homogeneous population of sensory receptors. The sensitivity function of the dark-adapted ocellus to longer wavelength light (yellow and red) is determined by a single receptor with a major peak of sensitivity in the green at 510–520 nm with some sensitivity in the near-ultraviolet. Sensitivity at shorter wavelengths (near-ultraviolet and blue), however, involves the stimulation of both this and a near-ultraviolet-sensitive receptor with a maximum sensitivity at about 370 nm. Anatomically, the sensory cells of the dorsal ocellus of Tenodera were determined histologically to be grouped into two distinct regions, each group making its own separate contribution to the ocellar nerve. This may represent the separation of two different photoreceptor types in the ocellus of the mantis.  相似文献

ABSTRACT. The role of ocelli in timing flight initiation of cabbage looper moths, Trichoplusia ni (Hübner) (Noctuidae), at dusk was studied under simulated sunset conditions using a multichannel actograph. The mean time of flight initiation was determined for control, sham (sham/occluded, sham ablated, unilateral ablated) and experimental (ocellus occluded, ocellus ablated moths). Ocellus occlusion delayed flight initiation on the first day following treatment but was less effective on the subsequent days. Ocellus ablation also delayed flight initiation on the first day, and also produced pronounced delays on subsequent days. In studies where the sunset was advanced 1 h, control sham ocellus occluded and unilaterally ocellus ablated moths responded to the advance, but bilaterally ocellus ablated moths did not. These results indicate that moths make use of input from the ocelli in determining the threshold light intensity for flight and in making adjustments to small light-phase changes.  相似文献
