The growth of the primary seminal root of maize (Zea mays L.) is characterized by an initial negative gravitropic reaction and a later positive one that attains a plagiotropic liminal angle. The effects of temperature and water potential of the surrounding soil on these gravitropic reactions were studied. Temperatures of 32, 25, and 18C and soil water potentials of −5,−38, and −67 kPa were imposed and the direction of growth was measured for every 1 cm length of the root. The initial negative gravitropic reaction extended to a distance of about 10cm from the graln. Higher temperatures reduced the initial negative gravitropic reaction. Lower soil water potential induced a downward growth at root emergence. A mathematical model, in which it was assumed that the rate of the directional change of root growth was a sum of a time-dependent negative gravitropic reaction and an establishment of the liminal angle, adequately fitted the distance-angle relations. It was suggested that higher temperatures and/or a lower water potential accelerated the diminution of the intitial negative gravitropic reaction.  相似文献

Five nitrate:ammonium ratios at two N-levels were tested with and without nitrapyrin [2 chloro-6-(trichloromethyl) pyridine] for grain production on a sandy soil. Treatments were applied to field maize as nutrient solutions, in one application, six weeks after planting. Nytrapyrin resulted in an increase in grain yield at a nitrate:ammonium ratio of 1:3 but in a decrease at a 0:1 ratio. The optimum nitrate:ammonium ratio was close to 1:3 with nitrapyrin and close to 3:1 without nitrapyrin. Nitrapyrin had an effect on NH4 +-N in the topsoil and NO3 -N in the subsoil at 70 days after application. Interactions of nitrate:ammonium ratios and N-levels were shown for leaf N concentration, soil mineral N and soil pH.  相似文献

The objective of this work was to study elongation curves of maize axile roots throughout their elongation period under field conditions. Relationships between their elongation rate and the extension rate of their branched region were also studied. Maize, early-maturing cultivar Dea, was grown on a deep, barrier-free clay loam (depth 1.80m). Trenches were dug during four periods until after silking and axile roots were excavated. Parameters measured were total length and the lengths of basal and apical unbranched zones. The rank of the bearing phytomer and general data about the carrying plant were also recorded. Results showed that axile roots from lower phytomers had similar elongation rates irrespective of the rank of the carrying phytomer. This elongation rate declined with root age. A monomolecular elongation model was fitted to the experimental data. Elongation was much slower in roots from upper phytomers. A rough linear relationship was found between the elongation rate of axile roots and the length of the apical unbranched zone. This result suggests that laterals appeared on a root segment a constant time after it was formed. Possible mechanisms with may account for the declining elongation rate with root age (increasing distance from aerial parts or adverse environmental conditions in deep soil layers) and variability between individual roots are also discussed.  相似文献

Summary The auxin-binding protein ABP-1 was localised immunocytochemically in coleoptiles and immature embryos ofZea mays. Two primary polyclonal antibodies raised against ABP-1 and secondary antibodies were either labelled with FITC or 10 nm gold particles for light microscopy, and with 10 nm gold particles for transmission electron microscopy. Light microscopy revealed that ABP-1 was localised in the epidermal cells of etiolated maize coleoptiles, in subepidermal parenchymatic mesophyll cells of the coleoptile and in the companion cells of the vascular bundles. Most labelling was found in the cytoplasm, less in nuclei and vacuoles and cell walls appeared negative. The region of the plasma membrane exhibited prominent labelling. Embryos showed low labelling throughout their tissues just after excision, but after culture for 7 days intensive labelling was found in the epidermis of the scutellum. Quantitative electron microscopy confirmed that ABP-1 was present in the cytoplasm of epidermal, mesophyll, and companion cells of coleoptiles. Gold particles were neither found in cell walls nor in the cuticle. Areas with ER and dictyosomes within epidermal and mesophyll cells of coleoptiles had a denser labelling with gold particles than elsewhere. Labelling at the plasma membrane, being the site where the auxin binds to the ABP, was observed at low levels in all cells examined, which is due to the method applied. Epidermal cells of embryos cultured for 5 days exhibited high levels of gold particles in ER and nuclei, and lower levels in the cytoplasm. The effects of the divalent metal ions Zn, Cd, Hg, Cu and Pb on the ATPase activity of a plasma membrane fraction isolated from roots ofZea mays have been investigated. When Mg-ions (3 mM), with or without K-ions (50mM) are included in the reaction medium, inhibition of ATPase activity was found in all cases, the relative order of the inhibitors over the concentration range 10 to 100M, being Hg>>CuCd>ZnPb. Below 1.0M only Hg caused substantial inhibition. In the absence of Mg ions, Zn and to a lesser extent Cd, activated the enzyme up to a concentration of 1 mM, activity being further stimulated in the presence of K-ions (50mM). No activation of ATPase activity was found for Hg, Cu or Pb. It was concluded that Zn-ATP and Cd-ATP are both alternative substrates for the enzyme. Further experiments showed that both Km and Vmax for the substrates Zn-ATP and Cd-ATP are very much lower than for the usual substrate Mg-ATP.These present results are discussed in relation to the known actions of these divalent cations on the trans-root potential and H-ion efflux in excised maize roots (Kennedy and Gonsalves, 1987).  相似文献

Summary This study clarifies the relationships between the root mucilage and the epidermal cell-wall in axenically grown maize roots. Mucilage appears to overlay the epidermal cell-wall which has two distinct layers. This mucilage is well preserved after cationized ferritin fixation and PATAg staining. Summary Dehydrogenase and urease activities, bacterial and fungal populations and physicochemical characteristics of maize (Zea mays L.) field soils have been studied for one crop cycle. A comparison has been made among soils of three different agricultural systemsviz permanent agriculture on plain lands in valleys, recently introduced terrace land agriculture and age old ‘slash and burn’ type of shifting agriculture on slopes. Results demonstrate that the enzyme activities, microbial population as well as most of the physico-chemical characteristics of soils followed the trend permanent agriculture on plain lands>terrace land agriculture>‘slash and burn’ type of shifting agriculture. Moisture and nutrient levels and topography of the lands were found to be major factors responsible for the trend.  相似文献

The crude lysosomal fraction of corn seedling root tips contains an arylsulphatase (E.C. 3.1.6.1) which hydrolysed p-nitrophenyl sulphate at pH 8.0 but had no activity towards p-nitrocatechol sulphate. The Km value for p-nitrophenyl sulphate was 1.24 mM. The hydrolysis of p-nitrophenyl sulphate was linear up to 2 h and the rate was proportional to the amount of enzyme added. The enzyme was strongly inhibited by cyanide, fluoride and phosphate ions and did not resemble the arylsulphatases of bacterial and animal origin.  相似文献

Trajectories of maize nodal roots were studied to test the hypothesis that roots which appear on a common internode have similar geometrical characteristics, and to assess the effect of soil temperature on root trajectory. Treatments consisted of three sowing dates, a comparison between mulched and non-mulched soil, a replication of one sowing date for two years in two locations, and a comparison between two cultivars at one sowing date. All these sources of variation, except the cultivar, had an appreciable effect on the trajectories of roots which appeared on the first four internodes. The horizontal component of the trajectory differed significantly between treatments, ranging from 93 to 700 mm in roots which appeared on the second internode, and from 71 to 569 for those on the third internode. The original hypothesis had, therefore, to be rejected. Mean soil temperature during the 100°C.days after root appearance accounted for the differences in trajectory between location, year, sowing date and mulch treatments, and for the differences between internodes within each location. The critical period during which temperature affected root trajectory probably began at root appearance, and ended between 50 and 100°C days after root appearance, i.e. when the root was less than 100 mm long.  相似文献

The direction of root growth can be studied by analyzing the trajectories of roots growing in soil. Both the primary seminal root and nodal roots of maize attain a preferred, or liminal, angle of growth that deviates from the vertical. These roots are said to be plagiogravitropic. Experiments using plants grown in soil-filled boxes revealed that the primary seminal root is truly plagiogravitropic. It shows both positive and negative gravitropism in response to gravity stimuli and tends to maintain its direction even after growing around obstacles. These are experimental results suggesting that plagiogravitropic growth is controlled by internal factors. The orientation of the grain affects the establishment of the liminal angle of the primary seminal root, and both the position of their node of origin and the root diameter are closely related to the plagiogravitropic behaviour of nodal roots. Several external factors are also known to influence plagiogravitropism. Low soil water content causes a decrease in the angle of growth and soil mechanical resistance suppresses the gravitropic curvature. Plagiogravitropic behaviour of both seminal and nodal roots plays a significant role in shaping the root system.  相似文献

Summary Four inbred lines ofZea mays (33.16, B 68, N 7B, B 77) were grown in nutrient solution to which K2Cr2O7 was added to give final concentration of 5 mg/l Cr (VI). The most evident differences in metal tolerance were observed between the B 68 and 33.16 line: in fact, even though the level of Cr (VI) was almost the same in the root tissues of both lines after 6 d of treatment, in the B 68 line, Cr induced marked alterations of nuclear structure and a progressive arrest of the cell cycle in G 1. The transport of14C-IAA and14C-ABA applied exogenously to root cap toward the elongation zone was investigated in gravi- and light-stimulated primary roots ofZea mays L. cv. Golden Cross Bantam 70. No significant difference of either IAA or ABA in radioactivities was observed between upper and lower halves of elongation zones during the latent period (0–60 min after the stimulation) of gravitropic response. When quantitative analysis of endogenous IAA and ABA by an internal standard method was carried out 60 min after gravi- and/or light-stimulation, no asymmetric redistribution of either IAA or ABA was observed between upper and lower halves of elongation zones. Light irradiation increased by 20% the contents of ABA in elongation zones. These results suggest that although both IAA and ABA are basipetally transportable and can transmit their information to the elongation zone during a latent period we cannot explain the gravitropic curvature by their redistributions between the two (upper and lower) halves of primary roots ofZea. On the basis of results from the present work and previous papers, the distribution of IAA and ABA in gravistimulatedZea roots is discussed. Summary Treatment ofZea mays seedlings with a 5 mM caffeine solution inhibits cytokinesis in guard cell mother cells (GMCs), producing unicellular, binucleate aberrant stomata (a-stomata). Ventral wall (VW) strips of limited length, which usually meet the wall portions of GMCs adjoining the cortical zone of the preprophase microtubule band (PMB), are laid down in many a-stomata.In a-stomata with or without VW-strips, the periclinal walls are lined by numerous microtubules (Mts) converging on their mid-region, where local wall thickenings are deposited. When the VW-strips reach the mid-region of the periclinal walls, thickenings lined by numerous Mts rise at their free margins. In certain a-stomata an anticlinal wall column, surrounded by a dense Mt bundle, grows centripetally from either or both of the periclinal wall thickenings. In wall thickenings, the cellulose microfibrils are co-aligned with the adjacent Mts. Pore formation is initiated in all a-stomata. Deposition of an electron dense intra-wall material followed by lysis precedes pore opening. This process is closely related to the a-stornata morphogenesis. These observations show that the primary morphogenetic phenomenon in a-stomata is the establishment of an intense and stable polarity in the cytoplasm abutting on the mid-region of the periclinal walls and/or the adjacent plasmalemma area. Prime morphogenetic factor(s), including microtubule organizing centres (MTOCs), seem to function in these sites. Summary Cytokinesis in the subsidiary cell mother cells (SMCs) ofZea mays leaves grown in the presence of 5 mM of caffeine solution is usually partially inhibited. A continuous wall strip, resembling a portion of the subsidiary cell (SC) wall, is laid down in the preprophase microtubule band (PMB) cortical zone. Sometimes, the incomplete SC (SC) wall grows centripetally in the absence of a phragmoplast and the gap becomes smaller or closes. The SC nucleus escapes through the SC wall gap into the larger SMC compartment and may fuse with the other nucleus.The aberrant SMCs (a-SMCs) pass through another division cycle, reattempting to produce a SC. A typical PMB is found in the SC space, in the site of the previous PMB. Moreover, in some preprophase SMCs, the cytoplasm adjacent to the SC wall is traversed by a small number of microtubules. The preprophase nuclei are partly or totally separated from the PMB by the perforated SC wall and may lie far from the latter.Usually, one mitotic spindle is assembled. The cycling paired polarized nuclei appear to synchronize and their chromosomes line up together on a single metaphase plate. Although the mitotic spindle axis is diversely oriented, one of its poles tends to be stabilized in the proximity of the SC wall gap. These divisions separate abnormal cells. Most or all the cell plate edges fuse with wall regions far from the PMB cortical zone. However, when some of them approach the SC wall strips, they are attracted and intersect their rims. In rare occasions the cell plate, invading the SC space is guided by the PMB cortical zone to create a SC-like curved wall portion, in absence of a daughter nucleus.Observations show that the cell plate arrangement in redividing aberrant SMCs is not subjected to a strict spatial control. The disorder of polarization sequence generated by the SC wall ring and especially the perturbation of the spatial (and functional?) relationship between PMB-PMB cortical zone and the nucleus—mitotic spindle is a causal factor of the variable cell plate arrangements.  相似文献

Summary Intact plants can reduce external oxidants by an appearingly trans-membrane electron transport. In vivo an increase in net medium acidification accompanies the reduction of the apoplastic substrate. Up to now, several NAD(P)H dehydrogenases,b-type cytochromes, and a phylloquinone have been identified and partially purified from plant plasma membranes. The occurrence of a quinone in the plasma membrane of maize roots supports the hypothetical model of a proton-transferring redox system, i.e., an electron transport chain with a quinone as mobile electron and proton carrier. In the present study the trans-membrane electron transport system of intact maize (Zea mays L.) roots was investigated. Flow-through and ionostat systems have been used to estimate the electron and proton transport activity of this material. Application of 4,4,4-trifluoro-1-(2-thienyl)-butane-1,3-dione (thenoyltrifluoroacetone) inhibited the reduction of ferricyanide in the incubation solution of intact maize roots up to 70%. This inhibition could not be washed off by rinsing the roots with fresh incubation medium. The acidification of the medium induced after ferricyanide application was inhibited to about 62%. The effects of thenoyltrifluoroacetone on proton fluxes in the absence of ferricyanide have been characterized in a pH-stat system. The net medium acidification by maize roots was inhibited up to 75% by thenoyltrifluoroacetone in the absence of ferricyanide, while dicumarol inhibited net acidification completely. The inhibition of H+-ATPase activity was estimated with plasma membrane vesicles isolated by phase partitioning and treated with 0.05% (w/v) Brij 58. ATP-dependent proton gradients and Pi release were measured after preincubation with the effectors. Summary A method was devised for the biochemical selection of immature, haploid Zea mays embryos using Adh1 and either the Stock 6 or indeterminate gametophyte (ig in W23) high haploid-inducing systems. Haploid (Adh1 ) embryos survived exposure to levels of allyl alcohol which killed diploid (Adh1 +/Adh1 ) embryos. Of the total surviving embryos which were examined cytologically 15% (using ig) and 22% (using Stock 6) were haploid. In two experiments with Stock 6, 100% of the surviving embryos were haploid. To obtain maximum effectiveness of Stock 6 and ig, Adh1 was transferred to stock 6 and W23 backgrounds. Immature, haploid embryos are being used to develop haploid, morphogenic tissue cultures of Zea mays.  相似文献

Summary Positive relationships were observed between Zn and P in the studied calcareous soils and in the corn plants grown thereon. The high content of carbonates in these soils affected greatly the levels of extractable Zn and the Zn–P relationships. Hydrolysis of organic phosphates by corn and soybean roots   总被引:1,自引:0,他引:1  
Because of the importance of organic phosphates as sources of P for plants, this work was performed to study the hydrolysis of nine organic phosphates by sterile, intact corn (Zea mays L.) and soybean (Glycine max L.) roots. Results showed that the rates of hydrolysis ofp-nitrophenyl phosphate (PNP) in buffered solutions by roots of three varieties of corn and three varieties of soybean ranged from 13 to 22 μmol PO4−P g−1 root h−1 and from 2.1 to 2.2 μmol PO4−P 0.1 g−1 root h−1, respectively. The average rate of hydrolysis of PNP in nonbuffered solutions was 2- to 3-fold lower for corn roots and 6- to 10-fold lower for soybean roots as compared with those obtained with buffered solutions. The orthophosphate released from hydrolysis of organic P compounds in buffered solutions during a 48-h incubation of corn roots showed that the maximum rate of hydrolysis of PNP was 4 to 6 times greater than the commonly used substrates: α- and β-glycerophosphates, phenolphthalein diphosphate, and glucose-6-phosphate. The rates of hydrolysis of glucose-6-phosphate and glucose-1-phosphate were similar and about 6- to 12-fold lower than that of PNP. Phosphoethanolamine and phosphocholine were hydrolyzed slightly, ando-carboxyphenyl phosphate was not hydrolyzed. The rates of hydrolysis of organic P compounds in nonbuffered solutions by corn and soybean roots were 1 to 3 and 1 to 10 times lower than those in buffered solutions, respectively. The trends in rates of hydrolysis by soybean roots of buffered organic P substrates were similar to those observed with corn roots, with the exception of glucose-1-phosphate and phosphoethanolamine.  相似文献

The influence of different Al concentrations, (0, 60 and 120 M Al) on growth and internal concentrations of Al, Si and selected organic acids was analysed in plants of teosinte (Zea mays L. ssp. mexicana), a wild form of maize from acid soils from Mexico. The plants were grown in nutrient solutions (pH 4.0) with or without 4 M silicon. Analysis with the GEOCHEM speciation program did not reveal differences between free activities of Al3+ in solutions with and without 4 M Si, but solutions with Si yielded lower concentrations of monomeric Al species, [Al]mono, when analysed by a modified aluminon method. Plants grown on solutions with similar [Al]mono, but differing in silicon, showed highly significant differences in growth and tissue concentrations of Al and organic acids. Silicon prevented growth inhibition at [Al]mono concentrations as high as 35 M, while plants grown without Si suffered severe growth reductions with 33 M [Al]mono. In solutions with similar [Al]mono concentrations plants with Si had lower tissue Al concentrations and higher concentrations of malic acid than plants without Si. In view of both the significant influence of Si on the response of plants to Al toxicity and the fact that some soluble Si is always present in soil solutions, the addition of low Si concentrations to nutrient solutions used for Al-tolerance screening is recommended.  相似文献
