To test the effect of seed treatment with fungicides on the development of mycorrhizal fungi, bean seeds were treated with fungicide dry or vehicled in the organic solvents, ethanol or dichloromethane and then planted in soil inoculated with the mycorrhizal fungus Glomus macrocarpum and/or the plant pathogenic fungus Fusarium solani. Measurements were made at 4 day intervals, to evaluate the location and extent of colonization of either Glomus macrocarpum or Fusarium solani in the root system. Most combinations of fungicide-solvent had little effect on the extent of colonization by each fungus individually. However, when both fungi were inoculated together, symptoms of F. solani were seen only in the tips of roots which indicate that the mycorrhizal fungus was able to limit the occurrence of the pathogenic fungus.  相似文献

We studied the diversity of oomycetes in a declining reed belt (Phragmites australis) of Lake Constance, Germany, using conventional baiting with specific reed and standard oak baits, accompanied by molecular techniques. Apart from an Aphanomyces sp. and a Phytophthora sp., baiting from reed rhizosphere samples from flooded, as well as drier, littoral sites revealed only Pythium spp. A total of 67 oomycete isolates was classified according to PCR-RFLP banding patterns and ITS sequencing, and 18 different sequence types could be separated. The majority of these seemed previously unknown species, as indicated by the degree of similarity to those deposited in nucleotide databases. Species communities in both flooded and drier habitats or both reed and oak baits clearly differed from one another, and only few species occurred in both dry and flooded sites, or in both oak and reed baits. A frequently occurring group of related Pythium species appeared to be specifically associated with reed, and these were the only species that proved pathogenic towards this host in vitro. Our study proved that unexplored natural ecosystems harbour diverse communities of oomycete species with specific habitat and host preferences within close-by, but ecologically contrasting, sites. Among the species isolated, those associated with the predominating plant might accumulate and thus may be reed pathogens of considerable importance.  相似文献

Growth response of hot pepper (Capsicum annuum L.) inoculated with the arbuscular mycorrhizal (AM) fungus, Glomus intraradices Schenck and Smith was evaluated in a greenhouse study. Three treatments in a soil-based medium amended with rock phosphate were: (1) control (CON), (2) inoculation of G. intraradices as a freshly prepared soil mixture of spores, hyphae and colonized roots of Sorghum vulgare (FM), and (3) inoculation of the fungus as cold-stored mixed inoculum (CM). Colonization at 14 weeks after inoculation with CM was 42.5%, but was significantly lower with FM (14.5%). Inoculation with G. intraradices as FM and CM increased growth of pepper, and total phosphorus and nitrogen uptake in shoots and roots compared with the CON treatment. Inoculation with CM resulted in significant increases in plant dry weight and chlorophyll concentration compared to the FM and CON treatments. Acid phosphatase activity in the rhizosphere was generally increased by AM fungal treatments. Highest acid phosphatase activity occurred at 14 weeks after inoculation with CM. Alkaline phosphatase activity in the CM treatment was significantly higher compared to that in CON and FM treatments throughout the growth period. Thus, cold storage of mixed inoculum enhanced colonization and growth-promoting activity of G. intraradices compared to freshly prepared inoculum.  相似文献

The effects of the soil yeasts Rhodotorula mucilaginosa, Cryptococcus laurentii and Saccharomyces kunashirensis on the arbuscular mycorrhizal (AM) fungus Glomus mosseae (BEG 12) was studied in vitro and in greenhouse trials. The presence of yeasts or their soluble and volatile exudates stimulated the percentage spore germination and hyphal growth of G. mosseae. Percentage root length colonized by G. mosseae and plant dry matter of soybean (Glycine max L. Merill) were increased only when the soil yeasts were inoculated prior to the AM fungus. Higher beneficial effects on AM colonization and plant dry matter were found when the soil yeasts were inoculated as an aqueous solution rather than as a thin agar slice. Although soluble and volatile exudates of yeasts benefited the AM symbiosis, their modes of action were different.This revised version was published online in May 2004 with corrections to the section of the article.  相似文献

The radial growth response to climate of white spruce in open grown conditions was examined at two measurement heights [breast height (1.3 m aboveground) versus stump height (0.3 m aboveground)] in order to examine the role of sampling height in dendroclimatic analysis. Twenty-four white spruce [Picea glauca (Moench) Voss] islands were sampled within Epinette prairie in the Spruce Woods Provincial Park (SWPP) located in south-western Manitoba. The structure of breast height and stump height residual chronologies were essentially the same after standardization and autoregressive (AR) modeling to stabilize growth variability, and AR modeling to remove temporal autocorrelation and thereby reduce low-frequency variation and enhance high-frequency variation. Correlation and response function analysis indicated that white spruce at the two sampling heights responded similarly to total precipitation, moisture index (precipitation subtract potential evapotranspiration), and minimum, mean, and maximum temperature. Both sampling heights also responded strongly to the cumulative effects of precipitation during the period of August of the previous year (t−1) to July of the current year (t). Regardless of the sampling heights considered, the results indicate that white spruce at its southern distribution limit responded strongly to moisture stress intensified by high temperatures. The results support the recommendation, at least for samples of white spruce at its southern distribution limit, that samples taken at stump height can be safely incorporated into tree-ring chronologies without the risk of diminishing the strength of chronologies in detecting climatic signals.  相似文献

Pterocarpus officinalis (Jacq.) seedlings inoculated with the arbuscular mycorrhizal fungus, Glomus intraradices, and the strain of Bradyrhizobium sp. (UAG 11A) were grown under stem-flooded or nonflooded conditions for 13 weeks after 4 weeks of nonflooded pretreatment under greenhouse conditions. Flooding of P. officinalis seedlings induced several morphological and physiological adaptive mechanisms, including formation of hypertrophied lenticels and aerenchyma tissue and production of adventitious roots on submerged portions of the stem. Flooding also resulted in an increase in collar diameter and leaf, stem, root, and total dry weights, regardless of inoculation. Under flooding, arbuscular mycorrhizas were well developed on root systems and adventitious roots compared with inoculated root systems under nonflooding condition. Arbuscular mycorrhizas made noteworthy contributions to the flood tolerance of P. officinalis seedlings by improving plant growth and P acquisition in leaves. We report in this study the novel occurrence of nodules connected vascularly to the stem and nodule and arbuscular mycorrhizas on adventitious roots of P. officinalis seedlings. Root nodules appeared more efficient fixing N2 than stem nodules were. Beneficial effect of nodulation in terms of total dry weight and N acquisition in leaves was particularly noted in seedlings growing under flooding conditions. There was no additive effect of arbuscular mycorrhizas and nodulation on plant growth and nutrition in either flooding treatment. The results suggest that the development of adventitious roots, aerenchyma tissue, and hypertrophied lenticels may play a major role in flooded tolerance of P. officinalis symbiosis by increasing oxygen diffusion to the submerged part of the stem and root zone, and therefore contribute to plant growth and nutrition.  相似文献

We developed a new technique for monoconidial culture of the most aggressive isolate in a given population of Bipolaris sorokiniana, to facilitate the evaluation of spot blotch resistance in wheat and barley. Blotched portions of infected barley leaves were placed on a glass slide in a moist chamber for production of conidia by associated fungal hyphae. Conidia were collected separately and grown on water agar discs. Individual water agar discs having conidium growth were inoculated on barley leaves. The conidium producing the earliest symptom with the largest lesion was considered most aggressive. This lesion was incubated in a moist chamber and the conidial offspring were tested for pathogenicity. When a uniform infection was observed, a small piece of the lesion was cut using a sterilized scalpel, surface sterilized with NaOCl, and inoculated in the centre of Petri dishes containing potato dextrose agar medium. The inoculated Petri dishes were incubated at 25 ± 1 °C to yield monoconidial cultures of the most aggressive isolate. Variability in symptom expression caused by the most aggressive isolate of a given population was much less than variability in symptom expression caused by all isolates collectively. The techniques will be useful for plant pathologists and breeders in screening for spot blotch resistance in wheat and barley.  相似文献

It was the aim of this study to determine the way in which low temperature modifies the effect of a competing grass on nitrogen fixation of a forage legume. White clover (Trifolium repens L.) was grown in monoculture or in different planting ratios with timothy (Phleum pratense L.) or perennial ryegress (Lolium perenne L.) in growth chambers at either 7.5/5°C (LoT) or 15/10°C (HiT) average day/night temperatures, and with 2.5 or 7.5 mM 15N-labelled nitrate in the nutrient solution.Competition with grass led to a marked increase in the proportion of clover nitrogen derived from symbiosis (% Nsym). This increase was slower at LoT where % Nsym was reduced considerably; it was closely related to the reduction in the amount of available nitrate as a result of its being utilized by the grass.Nitrogen concentration in white clover herbage and dry matter yield per clover plant were reduced, for the most part, when a competing grass was present. The amount of nitrogen fixed per plant of white clover decreased markedly with temperature. Low temperature consequently accentuated competition for nitrate. The capacity of white clover to compete successfully was limited by its slower growth and nitrogen accumulation.  相似文献

【目的】评估环链虫草Cordyceps cateniannulata对植物促生和植物抗氧化酶活性的影响。【方法】本研究利用浸种法将环链虫草接种于番茄植物体,在接种后的第30天和60天,通过番茄株高、根长、地上和地下部分的干鲜重指标评价其对番茄生长的影响;在接种后第10、20、30、60和90天,通过选择性培养基分析其在番茄不同组织中的生存情况,使用形态学及DNA序列比对的方法检验所分离菌株与原有菌株的一致性。在处理后的第30天,检测番茄叶片中的过氧化物酶(POD)、过氧化氢酶(CAT)、超氧化物歧化酶(SOD)及丙二醛(MDA)含量,观察环链虫草对番茄的抗氧化酶活性影响。【结果】环链虫草可定殖于番茄幼苗且对番茄生长有显著促进作用,菌株对植株的定殖偏好性分别为根部>茎部>叶部。酶活检测结果表明,处理组番茄叶片防御酶活性均呈显著升高的趋势,其中POD、CAT、SOD活性分别比对照增加了52.21%、75.31%和158.59%,MDA含量下降了35.15%。【结论】环链虫草可以通过浸种的方法感染并定殖番茄幼苗的根、茎、叶,促进番茄幼苗的生长并提高番茄抗氧化酶活性,具有较好的田间...  相似文献

The change in dried rhizome samples that were left to decompose was investigated to elucidate the effects of rhizome age on the decomposition rate of Phragmites australis. Rhizomes were classified into five age categories and placed 30 cm below the soil surface of a reed stand. After 369 days of decay, new (i.e., aged less than one year) rhizomes had lost 84% of their original dry mass, compared with a loss of 41–62% for that of older rhizomes. The exponential decay rates of older rhizomes were nearly identical to that of aboveground biomass. The nitrogen (N) concentration increased to two times its original values, but the phosphorus (P) concentration remained constant after an initial loss by leaching. The carbon to nitrogen (C:N) and carbon to phosphorus ratios (C:P) leveled out at 22:1 to 38:1 and 828:1 to 1431:1, respectively, regardless of rhizome age. The results are important to understand the nutrient cycles of reed-dominant marsh ecosystems.  相似文献

The putative plant defensin SPI1 cDNA from the conifer Norway spruce (Picea abies) is the only known plant defensin-like sequence from a gymnosperm. The predicted translational product SPI1 was not detected in the embryo or other parts of the seed by means of antibodies, but it accumulated in the root cortex after germination. In roots of seedlings infected with the root pathogenic oomycete Pythium dimorphum and the blue stain fungus Ceratocystis polonica, variable levels of SPI1 was detected during the first day as a response to the infection, however a significant increase was seen as an initial response to the root-rot fungus Heterobasidion annosum. After the first day of infection, the amount of SPI1 polypeptide was dramatically reduced in response to either of the pathogens, but not in response to the ectomycorrhizal fungus Laccaria bicolor. During the same time of infection, extensive damage to cortical root cells resulted from the infecting pathogens, but not from the mycorrhiza. These results indicate that pathogens may reduce the level of SPI1 by suppressing its expression, but may also reduce the SPI1 level by invading and disrupting the root cortical cells or by a combination of these mechanisms.  相似文献

Monotypic stands of reed canary grass, Phalaris arundinacea, replace native wetland vegetation where stormwater runoff alters hydrologic conditions, nutrient inflows, and sedimentation rates. We asked if different hydrologic conditions could explain the dominance of Phalaris and/or loss of the native grass, Spartina pectinata, and we compared the growth of each species alone and together under four hydroperiods (varying inundation frequency and duration) each at two water depths (surface saturation and flooding to 15 cm). When grown alone, aboveground biomass was similar for the two species, but Phalaris produced twice the stem length of Spartina via its low tissue density. Per unit biomass, Phalaris distributed its leaves over a larger canopy volume. Flooding reduced belowground biomass and increased total shoot length and shoot:root biomass of each species. Phalaris produced the most biomass, shoots, and total shoot length when wetter and drier conditions alternated weekly, while Spartina grew best with prolonged (4-week) inundation. When grown with Spartina, Phalaris changed its morphology by increasing its total shoot length:biomass ratio by 50%. However, ratios of Spartina:Phalaris aboveground biomass, shoot number, and total shoot length in two-species pots were not significantly affected by water depth or hydroperiod. We conclude that two plant attributes facilitate Phalaris' dominance of wetlands: its high ratio of total shoot length:biomass and its adaptable morphology (characterized herein as increased total shoot length:biomass when grown with Spartina).  相似文献

To examine the mechanisms of earlier reported alleviation of fluoride injury in ectomycorrhizal plants by NaCl, jack pine (Pinus banksiana) and white spruce (Picea glauca) seedlings were subjected to 1 mM and 5 mM KF in the presence of either 60 mM NaCl or 10% polyethylene glycol 3350 (PEG) for 2 weeks. Before the treatments, seedlings had either been inoculated with the ectomycorrhizal fungus Suillus tomentosus or remained non-inoculated. The inoculation with S. tomentosus reduced Na uptake by shoots and roots of jack pine seedling and by roots of white spruce that were treated with 60 mM NaCl. Mycorrhizal associations also drastically decreased fluoride uptake by jack pine seedlings, but did not affect shoot fluoride concentrations in white spruce. When NaCl was replaced by PEG in the 5 mM KF treatment solution, shoot fluoride concentrations were reduced by more than twofold without corresponding reductions in transpiration rates in mycorrhizal and non-mycorrhizal white spruce seedlings. When fluoride was present in the treatment solution, Na concentrations were lower in shoots and roots of both jack pine and white spruce mycorrhizal and non-mycorrhizal seedlings. The results suggest that Suillus tomentosus may help alleviate the effects of soil fluoride and salinity in jack pine and that fluoride uptake in white spruce is sensitive to osmotic stress.  相似文献

Summary This report describes a low-cost method for generating large numbers of high quality mature white spruce (Picea glauca [Moench.] Voss) somatic embryos which survived desiccation and grew to plantlets more vigorously than excised zygotic embryos cultured in vitro. Somatic embryos from suspension culture were supported within a culture chamber on a flat absorbent pad above the surface of a liquid culture medium containing 20–50 M abscisic acid and 7.5 % polyethylene glycol. Throughout a 7 week culture period 3 L of fresh medium was pumped into one end of the chamber, while the spent medium exited by gravity from the opposite end. Over 6,300 cotyledonary stage white spruce somatic embryos were recovered after this time from a single culture chamber without manual manipulation. The somatic embryos were of excellent appearance with well developed cotyledons, and possessed high levels of storage lipids. They survived drying to about 8 % moisture content following treatment for 4 weeks at 63 % relative humidity, and following imbibition converted to normal plantlets at a frequency of 92 %, compared to 80 % for embryos grown in Petri dishes. Somatic embryos cultured within the bioreactor developed to plantlets that were 20 % longer than zygotic embryos excised from mature seed and grown in vitro, and were 38 % longer than somatic embryos cultured upon agar medium in Petri dishes.Plant Research Centre contribution No. 1523  相似文献

为研究双色蜡蘑(Laccaria bicolor)对黑松(Pinus thunbergii)幼苗生长及其根系形态的影响,在营养杯培育条件下,用双色蜡蘑液体菌剂对黑松幼苗进行接种处理,接种第15、30、60、90、120天时取样,比较接种和未接种黑松幼苗的生物量、根系形态及根系分形维数的差异。结果表明:双色蜡蘑在黑松幼苗地上植株、地下根系的生长及其生物量的积累方面都表现出明显的促进作用。接种双色蜡蘑也显著改善了根系总长度、分支数、表面积、体积等参数和根系分形维数,并对地下根系生长的促进作用时间早于地上部分,且效应显著高于地上部分。接种双色蜡蘑第15~30天时对地上部分基本无影响,但对根系促进作用明显,而地上部分在第60天时开始表现出显著的生长效应。研究发现,双色蜡蘑能够成功定殖于黑松根部,促进黑松幼苗生长及其生物量的积累,同时显著促进根系总长度、分支数、表面积和体积增加,并使根系分形维数增大,表现出明显的促生作用,且对根系发育的显著促进作用早于地上部分。  相似文献

Summary Chloroplast DNA (cpDNA) restriction analysis was used to classify five reforestation seedlots as to species. The material included two Sitka spruce (Picea sitchensis (Bong.) Carr.), one white spruce (P. glauca (Moench) Voss) from interior British Columbia, and two putative hybrid seedlots from the coast-interior introgression zone in British Columbia. The cpDNA patterns generated by Bam-HI and Bc1-I from individual trees of Sitka spruce, white spruce, western white spruce (P. glauca var. albertiana (S. Brown)), and Engelmann spruce (P. engelmanni (Parry)) were species-specific. They were used as reference patterns for comparisons. In addition, two controlled crosses between white and Sitka spruce were analyzed to demonstrate the paternal inheritance of cpDNA in spruces. The cpDNA restriction patterns for the five seedlots were obtained from composite samples of seedlings from each lot and compared to the typical cpDNA patterns of each species. Restriction patterns for the two Sitka spruce seedlots agreed with those from the Sitka spruce tree, while patterns for the white spruce seedlots from British Columbia agreed with those from the white spruce tree, lacking evidence of any Engelmann spruce component in the sample. On the other hand, one putative hybrid seedlot showed cpDNA patterns similar to white spruce while the other showed fragments unique to both Sitka and white spruce, indicating that this was a hybrid seedlot. The analysis of cpDNA restriction polymorphism has proven to be an effective tool for classifying seedlots in regions of introgression. To our knowledge, these results provide the first demonstration of the use of cpDNA analysis for solving practical forestry problems.  相似文献

Summary The inoculation ofAlnus rubra (red alder) withFrankia sp. can lead to a highly efficient symbiosis. Several factors contribute to the successful establishment of nitrogenfixing nodules: (1) quantity and quality ofFrankia inoculant; (2) time and method of inoculation; (3) nutritional status of the host plant.Frankia isolates were screened for their ability to nodulate and promote plant growth of container-grown red alder. Inoculations were performed on seedlings and seeds. Apparent differences in symbiotic performance could be seen when seeds or seedlings were inoculated. Plants inoculated at planting performed significantly better than those inoculated four weeks later in terms of shoot height, nodule number and shoot dry weight. If inoculation was delayed further, reduction in shoot height, nodule number and shoot dry weight resulted. The effect of fertilizer was also investigated with regard to providing optimal plant growth after inoculation. Plants receiving 1/5 Hoagland's solution minus nitrogen showed maximal plant growth with abundant nodulation. Plants receiving 1/5 Hoagland's solution with nitrogen showed excellent plant growth with significantly reduced nodulation.  相似文献

The effects of waterlogging and salinity on morphological and physiological responses in the marsh grass Sporobolus virginicus (L.) Kunth were investigated in a 4×2 factorial experiment. Plants were subjected to four salinity levels (0, 100, 200 and 400 mol m–3 NaCl) and two soil inundation conditions (drained and flooded) for 42 days. Flooding at 0 mol m–3 NaCl caused initiation of adventitious surface roots, increased internal acration and plant height, induced alcohol dehydrogenase activity (ADH), and decreased belowground biomass and the number of culms per plant. Salinity increase from 0 to 400 mol m–3 NaCl under drained conditions increased leaf and root proline concentrations and decreased photosynthesis, aboveground biomass, number of culms per plant and number of internodes per culm. Concurrent waterlogging and salinity induced ADH activity and adventitious surface roots but decreased plant height and aboveground biomass. Internal air space increased with waterlogging from 0 to 100 mol m–3 NaCl but further increases in salinity to 400 mol m–3 reduced air space. Combined waterlogging and salinity stresses, however, had no effect on photosynthesis or on the concentrations of proline in leaves or roots. These results are discussed in relation to the widespread colonization by S. virginicus of a wide range of coastal environments varying in soil salinity and in the frequency and intensity of waterlogging.  相似文献

Anin vitro selection system using microcross sections of banana and plantain cultivars belonging to AAA and AAB genomic groups were used to produce plants resistant against the Black Sigatoka disease. The fungus resistant plantlets were obtained in a double selection system. This involved in a first step the use of a fungal crude filtrate and in the second step the purified host-specific toxin 2,4,8-trihydroxytetralone extracted from the fungusMycosphaerella fijiensis (M. fijiensis), the causal agent of Black Sigatoka disease. Resistant plantlets obtained from the double selection system were inoculated with conidia ofM. fijiensis in a growth chamber to reproduce Black Sigatoka symptoms. Compared to non-treated control plantlets, which were highly susceptible to the fungus, 10.7–19.3% toxin-resistant plantlets which arose from tissues that went through the double selection system were resistant againstM. fijiensis. This technique of using micro-cross sections for selection on fungal toxins seems to be amenable to differentMusa genotypes for the production of fungus-resistant plants.F. A.
