The purpose of this study was to develop a model to predict (1) air and product temperatures, (2) product moisture, and (3) air humidity during an aqueous coating process using a Bohle Lab-Coater. Because of the geometrical properties and the airflow, the drum of the Bohle Lab-Coater can in principle be divided into 2 zones of equal size—the drying and the spraying zones. For each zone, 4 balance equations could be set up describing the change of the air humidity, the product moisture, the enthalpy of the air, and the enthalpy of the product in each zone. For this purpose, knowledge regarding heat and mass transfer and also the motion of the tablets in drums was used. Based on the considerations of the heat and mass transfer, a set of first-order coupled ordinary differential equations (ODEs) was developed. This set of ODEs can be solved numerically. In this part, the development of the model is described in detail, whereas the application of the model can be found in part 2.

The goal of this study was to utilize risk assessment techniques and statistical design of experiments (DoE) to gain process understanding and to identify critical process parameters for the manufacture of controlled release multiparticulate beads using a novel disk-jet fluid bed technology. The material attributes and process parameters were systematically assessed using the Ishikawa fish bone diagram and failure mode and effect analysis (FMEA) risk assessment methods. The high risk attributes identified by the FMEA analysis were further explored using resolution V fractional factorial design. To gain an understanding of the processing parameters, a resolution V fractional factorial study was conducted. Using knowledge gained from the resolution V study, a resolution IV fractional factorial study was conducted; the purpose of this IV study was to identify the critical process parameters (CPP) that impact the critical quality attributes and understand the influence of these parameters on film formation. For both studies, the microclimate, atomization pressure, inlet air volume, product temperature (during spraying and curing), curing time, and percent solids in the coating solutions were studied. The responses evaluated were percent agglomeration, percent fines, percent yield, bead aspect ratio, median particle size diameter (d50), assay, and drug release rate. Pyrobuttons® were used to record real-time temperature and humidity changes in the fluid bed. The risk assessment methods and process analytical tools helped to understand the novel disk-jet technology and to systematically develop models of the coating process parameters like process efficiency and the extent of curing during the coating process.

Recent studies have shown the importance of monitoring microenvironmental conditions (temperature, relative humidity) experienced by the tablet bed during a pan coating process, thereby necessitating the need to understand how various process parameters influence these microenvironmental conditions. The process parameters studied in this work include exhaust air temperature, spray rate, inlet airflow rate, gun-to-bed distance, coating suspension percent solids, and atomization and pattern air pressure. Each of these process parameters was found to have an impact on the tablet bed relative humidity (RH), as measured using PyroButton data logging devices. A higher tablet bed RH was obtained with an increase in spray rate and atomization air pressure and with a decrease in exhaust air temperature, inlet airflow rate, gun-to-bed distance, suspension percent solids, and pattern air pressure. Based on this work, it can be concluded that the tablet bed thermodynamic conditions are a cumulative effect of the various process conditions. A strong correlation between the tablet bed RH and the frequency of tablet coating defect (logo bridging) was established, with increasing RH resulting in a higher percent of logo bridging events.

Measurements of expiration temperatures were carried out under different climatic conditions. In one series of experiments the ambient air temperature was varied, in another the relative humidity of the ambient air. The temperatures of the ambient air ranged between –5°C and 30°C and the relative humidity between 10% and 90%. The results reveal a high variability of the expiration temperatures, when the ambient air temperature is changed, and almost constant expiration temperatures, when the relative humidity is altered but the ambient air temperature is kept constant. Nasal expiration temperatures are more sensitive to changes of the meteorological parameters than oral expiration temperatures.

Summary Leaf gas exchange of Vigna unguiculata was influenced by short-term (day-to-day) changes in soil temperature and the response depended upon the aerial environment. When aerial conditions were constant at 30° C leaf temperature, high air humidity and moderate quantum flux, CO2 assimilation rate and leaf conductance increased with increases in soil temperature from 20 to 35° C, and this response was reversible. Decreases in CO2 assimilation rate and leaf conductance were observed at root temperatures above 30° C when root temperatures were increased from 20° C to 40° C and when air humidity was decreased in steps during the day. In contrast, varying soil temperatures between 20 to 35° C had no influence on gas exchange when shoots were subjected to a wide range of temperatures during each day.The gain ratio A/E remained constant at different air humidities when root temperature was less than or equal to 30° C indicating optimal gas exchange regulation, but changed with humidity at higher root temperatures. Leaf conductance responded independently from leaf water potential which remained relatively constant during individual experiments. The results indicate that plant responses to high root temperatures may have relevance to plant performance in semi-arid environments. They also illustrate the importance of controlling soil temperatures when studying the responses of potted plants in controlled aerial environments.Dedicated to K.F.

Measurement of gas exchange rates in plant tissue culture vessels
The aerial microenvironment in culture vessels has a significant effect on the growth and development of plantlets in vitro. Since the gas exchange between outside air and inner air can influence the microenvironment of culture vessel, it is necessary to measure the air exchange rate for various vessels. In this study, water vapor was used as the tracer gas, and the change of absolute humidity inside the vessel was calculated continuously by the measured values of a relative humidity sensing element. The outside environment was maintained at constant humidity level by a saturated salt solution. The RH data were transformed into absolute humidity and the specific humidity ratio. The air exchange rates of several tissue culture vessels were then calculated. The exchange rate was between 0.0145 h–1 to 0.0376 h–1. This technique provides an inexpensive, rapid and simple way to determine the air exchange rate of a culture vessel within a short period. The effects of the air current velocities on the exchange rates of vessels were also determined.

Abstract. Seasonal data on leaf conductance (gl) for three different apple cultivars grown in four separate orchards with different aged trees was studied between 1979 and 1985. A number of empirical models for predicting leaf conductance from environmental measurements were compared using this data and a general method for adapting such models for the prediction of different data sets is proposed. Although stepwise multiple regression identified relative humidity or vapour pressure as important variables, it frequently did not identify the optimal set of independent variables, which often did not include either of these. There was no advantage in regressing gl against principal components of the environment, rather than against the raw environmental variables. A simple model involving air vapour pressure deficit, air temperature and a hyperbolic function of irradiance was found to explain between 32 and 62% of the variance in gl for the different data sets. Parameters fitted for one data set led to the effective prediction of gl in other years or plots. The model fit could generally be improved significantly by including soil moisture deficit among the independent variables.

Peanut hull (peanut pods and waste materials from processing of peanuts) was pelleted through a 3/16-in. (4.76 mm) diameter die. The effect of change in moisture content (4.2-21.2%, wet basis) of the pellets (due to exposure of the pellets to low or high humidity storage environments) on physical properties were measured. Pelleting increased the bulk density of the peanut hull from 151 kg/m(3) to more than 600 kg/m(3). Bulk density, particle density and durability of the pelleted peanut hull of pellets were significantly affected by moisture content attained by the pellets during exposure to the low or high humidity environment. Air relative humidity significantly influenced the rate at which the pellets exchange moisture with the environment while air temperature did not. The values of the constants of the GAB model were obtained from moisture sorption isotherm at 25 degrees C.

There is growing concern about the adverse effects of fungal bioaerosols on the occupants of damp dwellings. Based on an extensive analysis of previously published data and on experiments carried out within this study, critical limits for the growth of the indoor fungi Eurotium herbariorum, Aspergillus versicolor, and Stachybotrys chartarum were mathematically described in terms of growth limit curves (isopleths) which define the minimum combination of temperature (T) and relative humidity (RH) at which growth will occur. Each growth limit curve was generated from a series of data points on a T-RH plot and mathematically fitted by using a third-order polynomial equation of the form RH = a(3)T(3) + a(2)T(2) + a(1)T + a(0). This fungal growth prediction model was incorporated within the ESP-r (Environmental Systems Performance [r stands for "research"]) computer-based program for transient simulation of the energy and environmental performance of buildings. For any specified location, the ESP-r system is able to predict the time series evolution of local surface temperature and relative humidity, taking explicit account of constructional moisture flow, moisture generation sources, and air movement. This allows the predicted local conditions to be superimposed directly onto fungal growth curves. The concentration of plotted points relative to the curves allows an assessment of the risk of fungal growth. The system's predictive capability was tested via laboratory experiments and by comparison with monitored data from a fungus-contaminated house.

Edaphic factors such as soil temperature and moisture influence soil-dwelling insects, whose most vulnerable stages typically are eggs and young larvae. In this study, the survival of eggs and first-instar larvae of the cabbage maggot, Delia radicum L., was measured under laboratory conditions after exposure to a range of soil temperatures and moistures. When eggs were exposed to constant temperature (20-29°C) and humidity (5-200% [wt:wt]), temperature had no significant effect on survival, whereas humidity <25% [wt:wt] caused egg mortality. The gradual exposure of eggs to high temperatures resulted in low mortality below 33°C, but <5% of eggs survived at 40°C. When first-instar larvae were exposed to constant temperature (17-29°C) and humidity (5-100% [wt:wt]), both factors as well as their interaction had a significant effect on larval survival, which was nil at 5% (wt:wt) for all temperatures but increased from 21.9 to 42.8% at 17°C and from 34.1 to 55.0% at 29°C, for soil moisture contents of 15% and 100% (wt:wt), respectively. Eggs of D. radicum are resistant to low soil moisture and high temperature conditions. Larval survival tends to increase with an increase in soil temperature and moisture. It is suggested that soil temperature be integrated into insect development simulation models instead of air temperature, to build more effective models for cabbage maggot management.

The tensile properties of the outermost layer of skin of neonatal rats, the stratum corneum, were investigated at a constant strain rate as a function of moisture content and ambient test temperature. The results show that the mechnical behavior of this membrane, whose primary constituent is the fibrous protein keratin, can be significantly altered by variations in both the sorbed water content and ambient temperature. In particular, a brittle to ductile transition was observed at 25 degrees C once the hydration level exceeded 70% relative humidity. Similarly, an identical phenomenon was detected at temperatures beyond 40 degrees C for specimens whose equilibrium moisture concentrations were maintained at 10 g H2 O/100 g dry protein. Differential scanning calrimetry measurements showed the presence of a molecular relaxation process which migrated from 42 degrees C at 40% relative humidity to --18 degrees C at 95% relative humidity. It is postulated that this relaxation process, possibly corresponding to the glass transition of the fibrous protein component of stratum corneum, is primarily responsible for the observed behavior.

Effect of operating conditions on solid substrate fermentation
In this work the effects of environmental parameters on the performance of solid substrate fermentation (SSF) for protein production are studied. These parameters are (i) air flow rate, (ii) inlet air relative humidity, (iii) inlet air temperature, and (iv) the heat transfer coefficient between the outer wall of the fermentor and the air in the incubator. The air flow is supplied to effect cooling of the fermented mass by evaporation of water. A dynamic model is developed, which permits estimation of biomass content, total dry matter, moisture content, and temperature of the fermented matter. The model includes the effects of temperature and moisture content on both the maximum specific growth rate and the maximum attainable biomass content. The results of the simulation are compared with actual experimental data and show good agreement with them. The most important conclusions are that (i) the evaporative cooling of the biomass is very effective for temperature control and (ii) the air flow rate and the heat transfer coefficient have strong effects but they affect the biomass morphology and are not controllable easily. Also, a simple technique for the determination of the optimum temperature and moisture content profile for cell protein production is applied. The simulated biomass production increases considerably employing the optimum temperature and moisture content profiles. The ultimate goal is to implement the determined effects of the environmental parameters on the SSF biomass production and the temperature and moisture variation profiles to effectively control the SSF and optimize the biomass production. Summary Evaporative water loss was measured from black-tailed prairie dogs (Cynomys ludovicianus) at eight air temperatures between 10 and 40°C, and over a broad range of humidities at each temperature. Evaporation increased, decreased, or remained constant below thermoneutrality, depending on humidity, and increased at higher temperatures indirectly with humidity. Evaporation was negatively related to humidity at different temperatures with a series of linear regressions that provided for statistical comparisons. Significantly different intercepts and slopes at different temperatures occurred for prairie dogs, and for deer mice (Peromyscus maniculatus) and house mice (Mus musculus) using published data. A general model, based on the equation for a straight line, was developed to relate evaporation to humidity; the intercept and slope were empirically derived mathematical functions of temperature. The model provided three-dimensional graphic responses for evaporation that showed different interactions between temperature and humidity for the three species.

Samples of cured hay have been stored in closed containers at relative humidities of 0–95% at temperatures varying from 5 to 25° C. A curve relating equilibrium moisture content to relative humidity of the store has been constructed and the incidence of first mould growth noted. An explanation has been suggested to account for the apparent discrepancy between earlier published work concerning the onset of moulding and conditions normally prevailing on the farm. In laboratory storage experiments temperature conditions are relatively constant, whereas with hay stacked in the open air the high atmospheric humidity in winter is offset by a lowered temperature, and the higher summer temperature is accompanied by a lower moisture content in the hay.

The paper describes the effect of soil moisture content andair humidity on CO2 exchange (PN), CO2 diffusion resistance(Cr) and transpiration (E) in four varieties of japonica rice(Oryza sativa L.). A decrease in soil moisture content reducedthe rate of photosynthesis to a varying degree in the varieties.Reduction in photosynthesis was attributed to increase in Cr.The effect of low soil moisture on photosynthesis and CO2 diffusionwas further intensified by decrease in air humidity. By maintaininga high humidity in the air around the leaves however, the effectof soil moisture deficiency was reduced considerably, exceptin Rikuto Norin 21 which was very sensitive to soil-moisturedeficiency alone. Dryness of the air enhanced the transpirationrate, although the increase was relatively less in the plantsfacing a simultaneous water crisis at the root surface. In plantsgrowing under flooded conditions, a decrease in air humiditycaused a slight depression in PN despite the simultaneous decreasein Cr. The secondary drying phase in freeze drying is mostly developed on a trial-and-error basis due to the lack of appropriate noninvasive process analyzers. This study describes for the first time the application of Tunable Diode Laser Absorption Spectroscopy, a spectroscopic and noninvasive sensor for monitoring secondary drying in laboratory-scale freeze drying with the overall purpose of targeting intermediate moisture contents in the product. Bovine serum albumin/sucrose mixtures were used as a model system to imitate high concentrated antibody formulations. First, the rate of water desorption during secondary drying at constant product temperatures (−22°C, −10°C, and 0°C) was investigated for three different shelf temperatures. Residual moisture contents of sampled vials were determined by Karl Fischer titration. An equilibration step was implemented to ensure homogeneous distribution of moisture (within 1%) in all vials. The residual moisture revealed a linear relationship to the water desorption rate for different temperatures, allowing the evaluation of an anchor point from noninvasive flow rate measurements without removal of samples from the freeze dryer. The accuracy of mass flow integration from this anchor point was found to be about 0.5%. In a second step, the concept was successfully tested in a confirmation experiment. Here, good agreement was found for the initial moisture content (anchor point) and the subsequent monitoring and targeting of intermediate moisture contents. The present approach for monitoring secondary drying indicated great potential to find wider application in sterile operations on production scale in pharmaceutical freeze drying.

A study was conducted on the drying of Penicillium bilaiae, a fungal micro-organism used to promote soil-bound phosphorous uptake in several crop species, such as wheat, canola and pulse crops. A wet pellet formed from a mixture of the inoculant and a starch-based carrier was air-dried to the appropriate water activity to extend the shelf-life of the viable fungal conidia. Convective air-drying was examined as a low-energy alternative to the more expensive freeze-drying technology that is currently in use. Experiments were conducted to measure the loss of conidia viability during drying in a fixed-bed, thin-layer convective dryer. The dryer air inlet temperature and relative humidity were controlled in experiments to determine the effect of thermal and dessicative stresses on conidial viability. The measured survivor fraction was determined to be dependent on solids temperature, moisture content and drying rate. Thermal stresses became significant for process temperatures above 30°C, while the survivor fraction fell sharply below a dry basis moisture ratio of 30%. Slower drying kinetics associated with high inlet air relative humidity were found to significantly improve the recovery of viable conidia. By minimising environmental stresses, survivor fractions of up to 75% could be achieved, but this result fell dramatically with the introduction of more severe conditions. A general linear statistical model is used to quantify experimental error and the significance level of each factor.
