芽生孢子是许多真菌通过出芽方式产生的无性孢子,在液体发酵中主要以这种形式大量繁殖。真菌杀虫剂活性成分分生孢子的大量生产使用液体发酵得到的芽生孢子作为接种物,另外,它也是当今虫生真菌遗传转化的重要受体,因而研究虫生真菌芽生孢子的形态和发生特点具有重要意义。本研究分别对液体发酵中的球孢白僵菌、粉棒束孢、蝉棒束孢、环链棒束孢、玫烟色棒束孢、细脚棒束孢、斜链棒束孢、金龟子绿僵菌、蝗绿僵菌和蜡蚧霉等10种常见虫生真菌芽生孢子的形成过程进行显微观察,了解其分生孢子产生过程的异同。结果表明,芽生孢子的产生方式有两种类型:1)蝉棒束孢在其整个生活史中以菌丝生长为主,芽生孢子产生的数量很少。2)其他各种真菌芽生孢子产生方式相似,在菌丝体形成后就开始大量以菌丝出芽或缢缩产生芽生孢子,接着还可通过芽生孢子的出芽或缢缩断裂产生新的芽生孢子。芽生孢子的产生分为3个时期:初期先由菌丝形成芽生孢子;指数期芽生孢子大量增殖,菌丝和芽生孢子都可产生芽生孢子;后期以芽生孢子产新芽生孢子为主要方式。  相似文献

南瓜白粉病病原菌鉴定及寄主范围测定   总被引:2,自引:0,他引:2  
通过对南瓜白粉病病原菌的闭囊壳、分生孢子及萌发方式的观察和鉴别寄主感病反应鉴定,结果表明引起甘肃省武威地区南瓜白粉病的病原菌为苍耳单囊白粉菌Podosphaera xanthii。该白粉病菌闭囊壳形成的最适条件为温度20℃、相对湿度70%和光照强度4,400lx。寄主范围测定结果表明,该病菌不侵染丝瓜、小麦、辣椒、番茄、苜蓿、红三叶草、架豆和菜豆,可侵染除丝瓜之外的其余9种瓜类植物和绿豆、红小豆、向日葵,其中对南瓜、绿豆、西葫芦、红小豆、向日葵、甜瓜和黄瓜的致病性最强,发病率均达到100%,病情指数分别为15.56、14.51、13.33、13.33、13.07、12.22和12.22。侵染过程观察发现,感病南瓜人工接种白粉菌12h后,分生孢子芽管从侧面萌发,于24h芽管伸长,于36h形成菌丝,于72h形成稠密的网状菌丝,于96h形成分生孢子梗及串生分生孢子。  相似文献

几种植物油对莱氏野村菌分生孢子萌发的影响   总被引:1,自引:0,他引:1  
【目的】研究从银纹夜蛾(鳞翅目:夜蛾科)[Argyrogramma agnata Staudinger(Lepidoptera:Noctuidae)]幼虫上分离得到的莱氏野村菌菌株Nr0815分生孢子,于不同温度下在8种植物油中萌发率和萌发速率的差异。【方法】采用紫外分光光度计测定了Nr0815分生孢子在8种植物油中的吸光度,并用载玻片萌发法测定了72 h内不同温度下分生孢子在不同植物油中的萌发率以及不同观察时间段的萌发速率。【结果】莱氏野村菌分生孢子在供试植物油中分散良好。在10–30°C内,莱氏野村菌分生孢子在各植物油中均能正常萌发,其中在大豆油中累积萌发率均最高。从孢子萌发速率来看,23°C时除大豆油和菜籽油外,其他植物油中孢子的萌发速率在72 h内均随着时间的延长而上升,在48–72 h时的萌发速率最大。10°C时的萌发速率最小,48 h后才开始萌发。从孢子萌发后芽管生长来看,23°C下孢子萌发后72 h时大豆油中的芽管长度最大,为(24.89±1.04)μm,葵花籽油中芽管最短,仅为(9.10±1.00)μm。【结论】明确了温度和植物油对莱氏野村菌孢子萌发的影响,也为该菌油剂开发利用提供理论基础。  相似文献

【背景】尖孢镰孢(Fusariumoxysporum)引起的烟草根腐病在世界烟区普遍发生,严重影响烟草产量和质量,化学农药无法有效防治病害,利用生防菌防治该病成为研究热点。【目的】明确贝莱斯芽孢杆菌(Bacillusvelenzensis)GDND-2对尖孢镰孢生长发育的抑制作用。【方法】制备含10%和20%GDND-2发酵滤液的PDA培养基,涂在玻片上,接种尖孢镰孢分生孢子,观察滤液对尖孢镰孢孢子萌发、菌丝生长、孢子形成和色素产生的影响,应用扫描和透射电镜观察菌体超微结构的变化。【结果】贝莱斯芽孢杆菌GDND-2发酵滤液延迟孢子萌发2h以上,造成芽管膨大畸形,促使菌丝提早分枝,抑制菌丝延伸,使菌丝产生畸形球状结构,10%和20%发酵滤液对菌丝生长抑制率达53.41%和61.58%。滤液延迟病菌产孢,显著抑制产孢量,影响孢子形态,刺激病菌产生色素。10%和20%发酵滤液延迟产孢20h和28h,对产孢量的抑制率为52.11%和78.85%,滤液促使病菌形成小型分生孢子。观察尖孢镰孢的超微结构,部分菌丝膨大、畸形,细胞壁变薄,细胞膜消失,细胞质渗出,胞内呈空腔结构;部分菌丝严重皱缩、扭曲、...  相似文献

用飞虱虫疠霉(Pandora delphacis)初级分生孢子接种桃蚜(Myzus persicae),24 h内定时取样,在扫描电镜下观察孢子的萌发及其对寄主体壁的入侵。结果表明,附着到蚜体表面的孢子在4 h内有30~40%已萌发产生芽管,其中多数为侵染性芽管,少数是呈叉状分枝的营养生长型芽管。具侵染性芽管的孢子部分陷入体壁蜡质层中,显示孢子有分泌物产生并作用于寄主体壁。接种10 h内,侵染性芽管通过顶端膨大的附着胞或直接穿透侵入寄主体壁。到24 h时,产生侵染性芽管的孢子全部侵入寄主体内,寄主体表仅留下少数未萌发的孢子或营养生长型芽管。初级分生孢子在蚜体表面很少产生次级分生孢子,说明桃蚜是适合该菌侵染的寄主。陷入寄主体壁的孢子不因若蚜蜕皮而被去掉,表明该菌对成蚜和若蚜都具有侵染力。  相似文献

对豫南茶园茶树轮斑病的发生与病原菌分生孢子萌发特性进行了调查和研究。茶树轮斑病在豫南茶区发生较普遍,一般夏秋发病较重,冬春发病较轻;轮斑病的病叶率和病情指数与温度、湿度、光照等生态因子关系密切;病原菌孢子在离体25℃条件下,4 h开始萌发。在茶树叶面上,病原菌分生孢子萌发明显比非叶面条件下好,病原菌菌丝生长较快,说明此病原菌与茶树叶片有高度的亲和力和较强的适应茶树叶面微环境的能力;茶树轮斑病病原菌分生孢子在pH5~7范围内萌发及芽管伸长较好,pH过小或过大均不利于病原菌的分生孢子萌发和菌丝生长。  相似文献

对马铃薯蔗糖人工培养基(PSA)上绿核菌(稻曲病菌)不同培养时期的产孢情况进行了系统的扫描电镜观察。研究结果表明,在培养的前期(前20d),菌落表面往往形成集结状菌丝结构,其上开始产生大量分生孢子;一些分散的菌丝上也可产生少量的分生孢子。而在培养的后期,菌落表面往往形成黄色子实体,内部集生大量厚垣孢子。说明绿核菌在人工培养基上前期以形成分生孢子为主,后期则以厚垣孢子为主,且厚垣孢子的量远远大于分生孢子。萌发试验表明,成熟的厚垣孢子会随着保存时间的延长萌发率急剧下降。因此,新鲜的成熟厚垣孢子是最为理想的接种体。  相似文献

本试验结合曲利苯兰和荧光素钠两种染色方法的优点,从显微角度研究了尖角突脐孢菌(Exserohilum monoceras)两个菌株X-27和HN-14在稗草和水稻上萌发和侵染行为的差异。结果表明,在寄主稗草上,接种4h后尖角突脐孢菌孢子开始从一端或两端萌发形成初生芽管;10h后芽管顶端膨大形成附着胞,附着在寄主表面,两端萌发的孢子约90%一端败育,仅一端形成成熟附着胞;在接种后24h内成熟附着胞形成率与接种时间成正相关,24h后趋于稳定;16h后在成熟附着胞下方受侵染的细胞内指状吸器开始形成,随后发育为掌状吸器;接种36h后菌丝在组织表面扩展形成网状,同时稗草叶片上显现叶斑病症状,部分菌丝能在细胞间蔓延扩展。在非寄主植物水稻上,同样观察到孢子萌发产生芽管,但是萌发起始时间滞后大约4h,初生菌丝分枝明显减少,且未能观察到附着胞和吸器的产生。  相似文献

本文报直球孢白僵菌在摇瓶、发酵罐的不同培养条件过程中,通过分生孢子、芽生孢子、节生孢子、内生孢子和菌丝体断裂等方式进行无性繁殖。它的生活史属多孢类型,在不同的培养条件下,孢子生殖的类型也不同。在斜面上是产分生孢子,在液体发酵中是以芽生孢子为主,还有部份类似分生孢子的内生孢子。根据白僵菌在发酵过程中细胞的分化规律我们设计出球孢白僵菌对甾体化合物Reichstein’s转化发酵规律,获得转化的最佳效果。  相似文献

The kinetics of the development of the mycelial form of Sporothrix schenckii from yeast cells and conidia in a minimal basal medium with glucose at pH 4.0 and 25 °C were established. Germ tube formation was used as the index of germination for both yeast cells and conidia. Yeast cells were first observed to develop germ tubes after 3 h of incubation, reaching 92±5%, after 12 h of incubation. Germ tubes were first detected in conidia after 9 h of incubation, and 12 h after inoculation 92±6% of the conidia had germ tubes. After 24 h of incubation, fully developed, sporulating mycelia were observed from both yeast cells and conidia. A delay in germ tube formation from yeast cells was observed when But2cAMP(10 mM) and But2cGMP (10 mM) were added to the medium. Also the addition of caffeine, a cyclic nucleotide phosphodiesterase inhibitor, inhibited the yeast to mycelial transition. Conidial germination into the mycelial form was also inhibited when cAMP, But2cAMP and caffeine were added to the medium. These results suggest the possible involvement of cyclic nucleotides in the control of dimorphism in S. schenckii.  相似文献

The techniques of restriction enzyme-mediated integration (REMI) and electroporation (EP) were applied for the first time to improving the blastospore transformation of fungal biocontrol agent Beauveria bassiana for higher frequency. The blastospores from < or =24 h incubation in glucose-mineral medium after shaking conidia for 48 h in Subouraud dextrose broth were found most competent for integrating 1 microg plasmid DNA vectoring the phosphinothricin (PPT) resistance gene bar in 360 microL reaction system containing 100 U HindIII or XbaI. Such blastospores were also most suitable for EP transformation at the optimized field strength of 10 kV cm(-1). The optimized REMI and EP generated averagely 39 and 53 transformants microg(-1) plasmid DNA whereas polyethylene glycol (PEG) integration yielded only 22. All transformants grew well on Czapek's agar containing 400 microg PPT mL(-1) after three rounds of cultivation on the same agar excluding PPT but their parental strain showed no resistance. The target gene inserted into the genomes of 10 transformants randomly taken from REMI or EP transformation was consistently detected by both PCR and Southern blotting. Compared to the PEG integration, REMI and EP enhanced the frequency of the blastospore transformation by 73 and 137%, respectively.  相似文献

莱氏绿僵菌对斜纹夜蛾的致病力及生理效应   总被引:1,自引:0,他引:1  
【目的】测定莱氏绿僵菌Metarhizium rileyi Nr5772菌株对斜纹夜蛾Spodoptera litura幼虫及蛹的致病能力,研究莱氏绿僵菌侵染后在寄主体内的发育及对寄主的生理效应,探讨莱氏绿僵菌的致病机制。【方法】采用浸渍法测定莱氏绿僵菌孢子对斜纹夜蛾3-6龄幼虫及蛹的致死中浓度(LC_(50))和致死中时(LT_(50))。采用微量注射法接种莱氏绿僵菌虫菌体,在不同时间后采集斜纹夜蛾幼虫血淋巴,在显微镜下检查虫菌体的数量、形态及寄主血细胞数量,并用酶标仪测定寄主血淋巴酚氧化酶(Phenoloxidase,PO)的活性。【结果】M.rileyi孢子对3龄斜纹夜蛾幼虫毒力最强,10 d后LC_(50)=3.12×10~6个孢子/mL,龄期越大,致病力越低;孢子浓度为5×10~9个/mL时,对3龄幼虫的致死速度最快,LT_(50)=4.55 d,致死速度随龄期的增大和浓度的降低逐渐减缓;M.rileyi孢子对蛹的致病力远低于对幼虫的致病力。注射接种虫菌体后,64 h内,虫菌体数量在寄主血腔中以幂函数的形式增长,寄主的血细胞数量没有明显的变化;在侵染初期(接种后44 h内),血淋巴PO活性正常;在侵染后期,虫菌体数量不再增加(55-64 h后),逐渐转化为菌丝体,并快速杀死寄主,PO活性受到抑制。【结论】莱氏绿僵菌Nr5772菌株对斜纹夜蛾幼虫有较强的致病力,应在害虫低龄期应用;莱氏绿僵菌在侵染初期对寄主血细胞和血淋巴PO无影响,后期则完全抑制PO活性。  相似文献

The pH reactions, ultraviolet spectra and phosphorus content of solutions of a variety of commercially available peptones all indicated, predictably, considerable differences in the chemical composition of the peptones. The effects of these differences on the outcome of experiments with Candida albicans grown in different peptone media was investigated. The fungus produced germ tubes equally effectively on all such media provided that the inoculum was kept to 10(6) blastospores/ml or less. However, expression of inducible enzyme activities in C. albicans varied extensively from peptone to peptone; there was, for example, an inverse relationship between the inorganic phosphorus content of peptones and the amount of acid phosphomonoesterase detectable in intact blastospores. The results indicated that use of different peptones in "Sabouraud's" media by different laboratories may account for some, but not all, published instances of irreproducibility of experiments with C. albicans.  相似文献

In shake flask and fermentor studies, various media components and culture inocula were tested to improve P. fumosoroseus spore production rates, yield and stability. To evaluate inoculum potential and inoculum scale-up for fermentor studies, conidia and liquid culture-produced spores of various strains of P. fumosoroseus were compared as inoculum. Inoculation of liquid cultures with blastospores at concentrations of at least 1×106 spores mL-1 resulted in the rapid production of high concentrations of blastospores (∼1×109 spores mL-1, 48 h fermentation time) for all strains tested. The rapid germination rate of blastospores (90% after 6 h incubation) compared to conidia (>90% after 16 h incubation) and the use of higher inoculum rates reduced the fermentation time from 96 to 48 h for maximal spore yields. A comparison of various complex nitrogen sources showed that liquid media supplemented with acid hydrolyzed casein or yeast extract supported the production of high concentrations of blastospores that were significantly more desiccation-tolerant (79-82% survival after drying) when compared to blastospores produced in media supplemented with other nitrogen sources (12-50% survival after drying). For rapid spore production, requirements for trace metals and vitamin supplementation were dependent on the type of hydrolyzed casein used in the medium. Fermentor studies with two strains of P. fumosoroseus showed that high concentrations (1.3-1.8×109 spores mL-1) of desiccation-tolerant blastospores could be produced in 48-h fermentations. These studies have demonstrated that the infective spores of various strains of the fungal bioinsecticide Paecilomyces fumosoroseus can be rapidly produced using deep-tank, liquid culture fermentation techniques.  相似文献

Studies on the blastospore production of Metarhizium anisopliae var. anisopliae were conducted in Adamek's medium used as a standard, enriched with lecithin, collagen, lactic acid or polyethylene glycol 200 (PEG 200) to increase spore yield and suppress mycelial pellet formation. The addition of 5% lecithin resulted in a significant 10-fold increase in spore yield up to 1.9 108 blastospores/ml compared with 1.9 107 spores/ml in the standard medium. Collagen (3%) increased the number of blastospores 3.7-fold, and lactic acid (1.5%) two-fold. A reduction of mycelial pellet formation in favour of spore production was noted with each additive. The viability of blastospores at 40IC from media with lecithin, collagen and lactic acid suspended in 25% Ringer's solution was comparable to that of spores produced in the standard medium. Striking differences were noticed in the viability of spores produced with 5% PEG 200 in standard medium. The half-life of blastospores produced in standard medium suspended in sunflower oil was 33.6 h and that of 5% PEG 200 spores only 25.2 h. In bioassays, the virulence of spores produced in standard medium to which 3% lecithin, 3% collagen, 1.5% lactic acid or 5% PEG 200 had been added was tested against third-instar nymphs of Locusta migratoria migratorioides (R. & F.). The median lethal time and the mortality of L. migratoria achieved with blastospores produced with 3% lecithin (5.7 days, 99%) was comparable to that of blastospores from standard medium (5.1 days, 98%). The virulence of blastospores from all other media with additives was significantly reduced.  相似文献

本研究通过对细脚拟青霉、蝉拟青霉、玫烟色拟青霉、金龟子绿僵菌和莱氏野村菌在疏水表面产生附着胞的荧光显微镜观察和扫描电镜观察,明确五种虫生真菌均可产生附着胞,细脚拟青霉和金龟子绿僵菌产生单附着胞和复合附着胞两种形态,均呈椭圆至长椭圆形,如遇到不适合浸染的部位,则重新产生芽管向前延伸直至找到适合入侵的部位。蝉拟青霉分生孢子多在顶端发芽成人字形,末端形成椭圆形附着胞,该拟青霉再生附着胞能力强。金龟子绿僵菌和莱氏野村菌附着胞表面有较厚的粘液层,而三种拟青霉附着胞表面均未见有粘液层。  相似文献

Nuclear behavior in the developmental process of appressoria inAlternaria alternata was investigated. In pregerminated conidia, approximately 94% of the conidial cells were uninucleate. The migration of a nucleus into an elongating germ tube from a germinating conidium was confirmed after 2h of incubation at 24±1°C in PDB. Peak frequencies of binucleate and trinucleate germ tubes were detected 1 and 2h after the peak frequency of uninucleate germ tubes, respectively. Four-and five-nucleate germ tubes did not show marked peak frrequencies. A marked peak frequency of the six-nucleate germ tubes occurred about 1 h after the peak frequency of the trinucleate germ tubes, suggesting that the nuclei in the trinucleate germ tubes each divided once within 1 h. The significance of early establishment of multinucleate appressorial cells in the colonization of host plants by pathogenicA. alternata was discussed.  相似文献

本研究旨在阐明出芽短梗霉在不同氮源培养基中形态和胞外多糖的积累及化学成分变化。采用摇瓶法培养出芽短梗霉。三种培养基的氮源分别为硝酸钠(培养基1,M1)、硫酸氨、酵母膏(培养基2,M2)和硫酸氨、蛋白胨和酵母膏(培养基3,M3)。M1培养基中,菌丝体和单细胞的生物量积累均比M2、M3低,但胞外多糖的产量则等于甚至略超过M2和M3。在指数生长的前期,白色菌丝体和酵母状细胞状态占优势。指数生长的后期,以厚垣孢子、肿大细胞和黑色菌丝体占优势。胞外多糖都能为茁霉多糖酶水解为麦芽糖和麦芽三糖,说明这些多糖的化学组成都具有(1→4,1→6)-α结构的茁霉多糖。但M1中产生的茁霉多糖结构单元为麦芽糖和麦芽三糖,且二者比例相当。M2中茁霉多糖的麦芽糖结构单元明显减少,而M3中144h后麦芽糖结构单元完全消失。这似乎表明氧化性的氮源和低溶解氧水平可能是造成茁霉多糖结构单元同时具有麦芽糖和麦芽三糖的原因。  相似文献
