Diluted bull semen samples were bombarded with a 24 Mev proton beam. Dose response curves for the fraction of cells which survived the bombardment and for the average velocity of the surviving cells were measured. Target theory indicated a cross section of the sensitive volume of 2.1 × 10-10 cm2. Respiration measurements showed that the oxidative phosphorylation in the sperm remained coupled after the bombardments. The efficiency with which free energy from ATP hydrolysis was converted into mechanical work by the sperm was found to decrease after proton bombardment. The half-value dose for this effect was two and a half times higher than the half-value dose for motility damage. These respiration measurements indicate that the damage due to the bombardment is not to the metabolic system or to the contractile system in the sperm flagellum, but to a control system for the motility. The results of the target theory shows that this control system is localized in a small element of approximately 1600 A diameter. The centriole is tentatively proposed as being this control element.  相似文献

Hydrated T2 bacteriophage were irradiated with 0.75 to 90 kev electron beams. A thin foil isolated the sample chamber from the electron gun source. Survival (plaque formation) was observed. Apparent cross-sections and D37 doses were determined. The maximum cross-section of about 5 × 10-3 μ2 is roughly equal to the cross-sectional area presented by the phage core. As beam energy was increased the average D37 dose first attained a minimum value of about 23 kr for 1 kev electrons (which penetrate the relatively inert protein coat) after which the average D37 dose rose with beam energy to a maximum value of about 50 kr for fully penetrating beams. These dependencies suggest that the radiosensitive structure exists as a peripheral shell rather than a uniformly sensitive core. A tentative model for the phage structure, based on this and other evidence, is presented.  相似文献

The relation between survival and dose of ultraviolet (UV) light or X rays was, with only one exception, nonlinear for late-log-phase cultures of four species of Rhizobium. The ld90 for different bacterial strains ranged from 400 to 3,000 ergs per mm2 for UV treatment, and from 3.0 to 7.5 kr for X-ray treatment.  相似文献

Summary In the ultraviolet (UV)-mutable bacterium, Myxococcus xanthus, dose response curves for the induction of rifampicin-resistant (Rifr) mutants were compared with dose response curves for Weigle(W)-reactivation of the UV-irradiated phage Mx4 at a phage survival of 5x10–6. In most strains examined, including a uvr mutant, these curves are largely similar. Unexpectedly the UV-sensitive strain M. xanthus Bt, which is unable to perform W-reactivation, is nevertheless UV-mutable. This result may indicate that the repair pathway involved in phage reactivation is only partly responsible for UV-mutagenesis or alternatively is not able to act on phage DNA in M. xanthus Bt cells. N-methyl-N-nitro-N-nitrosoguanidine (MNNG) treatment of M. xanthus cells also results in marked W-reactivation of the UV-irradiated phage Mx4 at the same survival of 5x10–6. The MNNG-stimulated phage reactivation is of the same order of magnitude as the UV-stimulated phage reactivation. Also the dose response curves for the induction of Rifr mutants by MNNG and the MNNG-stimulated phage reactivation are quite similar. This coincidence may indicate that misrepair mutagenesis is involved in both UV and MNNG-mutagenesis. It is suggested that M. xanthus is a useful organism with which to study misrepair mutagenesis in bacteria.  相似文献

We have measured volume fraction dependence of the sedimentation curve of swine erythrocytes in a physiological saline solution at 10 degrees C, 20 degrees C, 30 degrees C and 40 degrees C. The sedimentation curves were found to consist of initial constant velocity region and final plateau region at the lower temperatures of 10 degrees C and 20 degrees C, while modified S-shaped curves were observed at the higher temperatures of 30 degrees C and 40 degrees C. The volume fraction dependence of the initial slope v of the sedimentation curve was fitted well to the following exponential type equation at all the temperatures: v = vs,exp (1 - H)exp[-(BH + CH2)] where vs,exp is the velocity in infinite dilution corresponding to the Stokes velocity and H is the volume fraction of erythrocytes. The volume fraction dependence of the relative velocity v/vs,exp was in close agreement with a semi-empirical equation derived for slurrys in the field of chemical engineering at the lower temperatures, while a small deviation between the observed and calculated curves was found at the higher temperatures. The volume fraction dependence of v at 20 degrees C was also analyzed on a theory recently developed by Oka. The explicit functional form of the medium up-flow factor phi (H) and the deformability factor f in the theory were determined using the experimental data.  相似文献

Potassium retentivity and survival of yeast were studied after exposure to various kinds and conditions of irradiation. The radiations used were: 2537 A ultraviolet, 3500 to 4900 A long-ultraviolet and short visible, and 250 kvp1 x-rays. Both potassium retentivity and survival are decreased by these radiations. The dose-response of survival is about 16 times as sensitive as is potassium retentivity after 2537 A irradiation. Potassium retentivity is about twice as sensitive as survival after irradiation of 3500 to 4900 A. Survival after x-irradiation under aerobic conditions is five times as sensitive as potassium retentivity. Survival of cells irradiated with x-rays under anaerobic conditions was about half as sensitive as under aerobic conditions. The response of potassium retentivity to x-radiation at 25°C. under anaerobic conditions is only slightly affected below 160 kr, at which dose the slope abruptly increases to that obtained under aerobic conditions; lowering the temperature to 0°C. moves this point to about 300 kr. These differential effects are indicative of interaction of radiations with the yeast cell at sites that independently control survival and the retention of potassium.  相似文献

Characterizing the biological effects of flattening filter-free (FFF) X-ray beams from linear accelerators is of importance, due to their increasing clinical availability. The purpose of this work is to determine whether in vitro cell survival is affected by the higher dose-per-pulse present in FFF beams in comparison with flattened X-ray beams. A Varian TrueBeam® linear accelerator was used to irradiate the T98G, V79-4 and U87-MG cell lines with a single fraction of 5 Gy or 10 Gy doses of X-rays. Beams with energies of 6 MegaVolt (MV), 6 MV FFF and 10 MV FFF were used, with doses-per-pulse as measured at the monitor chamber of 0.28, 0.78 and 1.31 mGy/pulse for 6 MV, 6 MV FFF and 10 MV FFF, respectively. The dose delivered to each Petri dish was verified by means of ionization chamber measurements. No statistically significant effects on survival fraction were observed for any of the cell lines considered, either as a function of dose-per-pulse, average dose rate or total dose delivered. Biological effects of higher instantaneous rates should not be excluded on the basis of in vitro experimental results such as the ones presented in this work. The next step toward an assessment of the biological impact of FFF beams will require in vivo studies.  相似文献

Abstract

The binding of copper ion to the surfaces of the marine diatom Phaeodactylum tricornutum was studied in seawater pH 8. The cell wall complexing capacity and the average surface complex formation constant were determined by voltammetric titration curves. The presence of biotic particles at a concentration of 2.7 X 106 cells mL?1 does not hinder the measurement of stripping currents and the voltammetric fraction of non-reducible copper is the same as the copper bound to the cell surfaces as measured by atomic absorption spectrometry.  相似文献

Laser light scattering has been employed to determine the swimming speed distribution and the fraction of motile cells in samples of bovine spermatozoa. As predicted from theory, average trajectory velocities determined by laser light scattering were approximately four times the average translational speed estimated using light microscopy. The proportion of motile spermatozoa decreased with time at the same rate when samples were prepared in either HEPES or phosphate buffers. However, whereas the mean swimming velocity declined slowly in HEPES buffer, it dropped rapidly when phosphate buffer was used. Dilution (in the range 40–0.4×106 spermatozoa·ml-1) in either of these two buffers reduced the fraction of motile spermatozoa in the sample, but the mean swimming velocity of the remaining active spermatozoa was unchanged. Lowering the temperature from 37° C to 15° C reduced the mean swimming speed by a factor of 2–3 and the fraction of motile cells by a factor of 4–5.  相似文献

Aim: To develop an inactivation kinetic model to describe ultraviolet (UV) dose‐response behaviour for micro‐organisms that exhibit tailing using two commonly referenced causes for tailing: physical shielding of micro‐organisms and phenotypic persistence. Methods and Results: Dose‐response data for Escherichia coli, Mycobacterium terrae and Bacillus subtilis spores exposed to UV radiation were fit to the phenotypic persistence and external shielding (PPES) model. The fraction of persistent micro‐organisms in the original population (Npersistent/Ntotal) that exhibited reduced sensitivity to UV radiation was estimated by the PPES model as approx. 10?7, 10?5 and 10?4 for E. coli, B. subtilis spores and Myco. terrae, respectively. Particle shielding effects were evaluated for Myco. terrae and resulted in additional reduction in UV sensitivity. Conclusions: Tailing occurred in laboratory experiments even when clumping and shielding were eliminated as major factors in UV resistance, suggesting that phenotypic persistence in addition to shielding may be important to consider when evaluating dose‐response curves for disinfection applications. Significance and Impact of the Study: The PPES model provides a mechanistically plausible tool for estimating the dose–response behaviour for micro‐organisms that exhibit tailing in dispersed and aggregated settings. Accurate dose‐response behaviour (including the tailing region) is critical to the analysis and validation of all UV disinfection systems.  相似文献

A UV reactor with an annular design, a total liquid volume of 460[emsp4 ]ml, and outfitted with a single lamp with 1690[emsp4 ]mW of germicidal power was tested. Coliphage MS2 was used as a bioactinometer to measure the UV dose at a flow rate of 56.7[emsp4 ]ml/sec in water with a very low absorbance. The Beers Law coefficient was A100.003. The measured dose (MS2 bioactinometry) was 35.2±1.1[emsp4 ]mW-sec/cm2.A retention time distribution was generated with a dye tracer study. The reactor was modeled as if flow was confined to ten equal volume paths existing as concentric rings around the lamp. The UV intensity along each path (ith intensity) was calculated to generate a simulated distribution of UV intensity in the reactor. The retention time distribution was subdivided to estimate the retention time associated with each decile jth time) of the total flow.Seven methods of associating the ith intensity with the jth retention time were used to produce simulated dose distributions for the reactor. The average UV dose for each distribution was calculated as the average of the products of I and t (AP protocol) and by the apparent survival (AS protocol), in which the predicted survival along each path was averaged to back-calculate dose from the reference batch inactivation curve. The average dose predicted assuming that time and intensity were independent was 51.5[emsp4 ]mW-sec/cm2 based on the arithmetic average (AP protocol). Using the apparent survival method, the predicted dose for the independent distribution (I independent of t) was 36.4[emsp4 ]mW-sec/cm2. Three methods of developing dependent structure between time and intensity were tested. In the best possible case for stratified flow (I negatively correlated with t) the calculated (AS) intensity was 46.3[emsp4 ]mW-sec/cm2. In the worst case for stratified flow (I positively correlated with t) the AS intensity was 32.0[emsp4 ]mW-sec/cm2. In a rational case where flows were assumed to be distributed parabolically (low flow at the wall and at the lamp) produced an AS intensity of 37.7[emsp4 ]mW-sec/cm2. When either time or intensity was averaged, while the other variable was allowed to keep its distribution, the (AS) dose (time averaged 43.3[emsp4 ]mW-sec/cm2, intensity averaged 41.0[emsp4 ]mW-sec/cm2), yielded a poor prediction compared to the measured value.The errors associated with averaging time, intensity, or both, far outweigh the errors associated with choosing a rational distribution or an independent distribution of time and intensity in the prediction. This observation is generally true whenever an organism is exposed to UV light in a flow through reactor such that the range of doses is within the portion of the inactivation curve exhibiting strong exponential decay.  相似文献

In this study, we measured environmental outdoor gamma dose rates (terrestrial and cosmic) in 204 stations across the province of Artvin. The average outdoor gamma dose rate for the province of Artvin was found to be 174.81 nGy/h. The radiological map of Artvin was drawn with the help of measured outdoor gamma dose rates. Annual effective dose equivalent and lifetime cancer risk values were figured up for the adults in Artvin province by using measured outdoor gamma dose rates. The average annual dose equivalent and excess lifetime cancer risk were 214.5 μSv/y and 7.5 × 10?4, respectively. The values acquired were compared with the similar studies done around the world. The average annual effective dose equivalent and excess lifetime cancer risk were found to be approximately three times higher than the world average.  相似文献

The surviving fraction of synchronous V79 Chinese hamster cells was measured in two post-irradiation generations after a 300-rad X-ray dose in G1 by comparing the colony multiplicity in irradiated and control cultures. In addition, the ability of the irradiated population to form colonies was measured immediately after G1 irradiation or at 6, 32, 75 or 96 h after X-irradiation. Formulae were used in conjunctionwith previously observed transmission and survival parameters of chromosome aberrations2 to predict the amount of cell death at any given time after irradiation. The results indicate that the survival pattern of these cells can indeed be predicted on the basis of cell loss from chromosome aberrations.It is likely that an asymmetrical chromosome exchange (dicentric, centric ring, or tricentric) and a chromosome deletion are equally capable of causing cell death, whereas translocations or inversions apparently do not lead to inviability. Furthermore, cell death is rapid: 45% of the total observed death occurs in the first two post-irradiation generations. The initial decrease in viability is caused predominantly by the formation of anaphase bridges, while cell death from fragment loss becomes increasingly important in later generations. In fact, it is probable that, on the average, a cell that loses a single acentric fragment will survive one generation.  相似文献

The effect of different doses of synthetic antioxidant β-(4-hydroxy-3,5-ditertbutylphenyl)propionic acid (phenosan) on the development of spontaneous leukemia in AKR mice was studied. The drug efficiency was determined from the survival curves, animal life spans, and the incidence of leukemia. Phenosan exhibited a pronounced antitumor activity at therapeutic (10?4 mol/kg, 4 administrations) and ultra-low (10?14 mol/kg, 4 administrations) doses. The dose of 10?4 mol/kg proved most efficient to increase the life span of the short-lived subpopulation, while the dose of 10?14 mol/kg increased the life span of the long-lived subpopulation. The ultra-low dose of the drug seems promising as a prophylactic agent.  相似文献

Pulse gel electrophoresis was used to measure the reduction of mobilities of λ-DNA-Hind III fragments ranging from 23.130 to 2.027 kilobase pairs in Tris borate buffer solutions mixed with either hexammine cobalt(III), or spermidine3+ trivalent counterions that competed with Tris+ and Na+ for binding onto polyion DNA. The normalized titration curves of mobility were well fit by the two-variable counterion condensation theory. The agreement between measured charge fraction neutralized and counterion condensation prediction was good over a relatively wide range of trivalent cation concentrations at several solution conditions (pH, ionic strength). The effect of ionic strength, trivalent cation concentration, counterion structure, and DNA length on the binding were discussed based on the experimental measurements and the counterion condensation theory. © 1996 John Wiley & Sons, Inc.  相似文献

Summary BCG, C. parvum, and reovirus were used as immunostimulants in treating murine mammary adenocarcinoma (A-10) after tumor burden had been minimized with BCNU. Immunostimulants were administered at different times with respect to chemotherapy. Different doses were used to determine the optimal response as measured by survival. BCG induced the best response when 6.67×105 organisms were given 2 days after chemotherapy. The optimal response with C. parvum was observed after a dose of 0.35 mg was given 1 or 2 days after chemotherapy. Similarly, reovirus produced the best response when 1010 plaque-forming units were given 2 days after chemotherapy. Summary EL4 lymphoma was grown as an ascitic tumor in the peritoneal cavity of C57Bl/6 mice. Animals with different tumor burdens (either 107 or 109 cells) were treated with a single intraperitoneal injection of BCNU using doses from 20–40 mg/kg. Response as measured by mean survival time and percent survival was dependent on tumor burden and dose of drug. The objective of chemotherapy was to increase the mean survival time, but not the percent survival, in order to evaluate the therapeutic effect of reovirus. Mice were given 108, 109, or 1010 Pfu of reovirus at various times with respect to chemotherapy. The number of mice cured after treatment with both BCNU and reovirus was significantly greater compared to mice treated with BCNU only. Mice cured with combination therapy developed tumor-specific immunity as measured by cytotoxic lymphocytes and serum, and resistance to a lethal tumor challenge. PurposePoint detectors are frequently used to measure patient's maximum skin dose (MSD) in fluoroscopically-guided interventional procedures (IP). However, their performance and ability to detect the actual MSD are rarely evaluated. The present study investigates the sampling uncertainty associated with the use of grids of point detectors to measure MSD in IP.MethodChemoembolisation of the liver (CE), percutaneous coronary intervention (PCI) and neuroembolisation (NE) procedures were studied. Spatial dose distributions were measured with XR-RV3 Gafchromic® films for 176 procedures. These distributions were used to simulate measurements performed using grids of detectors such as thermoluminescence detectors, with detector spacing from 1.4 up to 10 cm.ResultsThe sampling uncertainty was the highest in PCI and NE procedures. With 40 detectors covering the film area (36 cm × 44 cm), the maximum dose would be on average 86% and 63% of the MSD measured with Gafchromic® films in CE and PCI procedures, respectively. In NE procedures, with 27 detectors covering the film area (14 cm × 35 cm), the maximum dose measured would be on average 82% of the MSD obtained with the Gafchromic® films.ConclusionThermoluminescence detectors show good energy and dose response in clinical beam qualities. However the poor spatial resolution of such point-like dosimeters may far outweigh their good dosimetric properties. The uncertainty from the sampling procedure should be estimated when point detectors are used in IP because it may lead to strong underestimation of the MSD.  相似文献

The relationships between xylem tension, velocity of water ascending and transpiration in tobacco plants were measured by means of the “xylem pressure probe technique” (Balling, A. and Zimmermann, U., Planta 182, 325–338, 1990). The flow velocity was determined by suction or injection of fluorescein (or FITC-labelled dextrans of various molecular weights) from the microcapillary of the pressure probe into the punctured xylem vessel, followed by serial-sectioning of the stem after a given propagation time. The distance travelled was defined as the distance from the injection point to the uppermost xylem section in which the dye could be detected. For a transpiration rate of 0.52 ± 0.12 ml . h?1, a linear dependence between the flow velocity and the tension gradients was found as expected from the Hagen-Poiseuille law. The slope of the straight lines decreased with increasing molecular weight of the fluorescent labelled compound, presumably because of (partial) plugging of the pit membranes. The average value of the flow velocity (2.5 . 10?4 ± 0.9 . 10?4 m . s?1) was one magnitude smaller than the value estimated from the geometric dimensions of the xylem vessels, but agreed well with the literature value of 2.8 . 10?4 m . s?1 for herbs (determined by the heat pulse technique; Huber, B. Ber. deutsch. bot. Ges. 50, 89–109, 1932). The average pressure gradient was determined to be 0.39 ± 0.23 MPa . m?1, in agreement with the literature (Begg, J. E. and Turner, N. C. Plant Physiol. 46, 343–346, 1970). The first response of xylem pressure (or tension) and of flow velocity to a reduction of the transpiration rate (0.14 ± 0.06 ml . h?1) occurred after about 24 h, when an increase of the xylem pressure towards higher values associated with a decrease in flow velocity was observed. In contrast, re-establishment of the normal transpiration rate brought the pressure (or tension) and the flow velocity back to normal values within half an hour. Similary, introduction of a transverse cross-sectional cut into the stem did not lead during the first 10 h to a change in xylem tension (or velocity). However, during the following day the pressure fell to relatively low values (about ?0.13 MPa). The velocity increased 10-fold. In the next two days the xylem pressure increased again to normal values (average +0.03 MPa), whereas the flow velocity assumed higher values than normal. The data are discussed in terms of the water status and storage of the adjacent tissue cells.  相似文献

PurposeBoron Neutron Capture Therapy (BNCT) is a form of hadrontherapy based on the selective damage caused by the products of neutron capture in 10B to tumour cells. BNCT dosimetry strongly depends on the parameters of the dose calculation models derived from radiobiological experiments. This works aims at determining an adequate dosimetry for in-vitro experiments involving irradiation of monolayer-cultured cells with photons and BNCT and assessing its impact on clinical settings.M&MDose calculations for rat osteosarcoma UMR-106 and human metastatic melanoma Mel-J cell survival experiments were performed using MCNP, transporting uncharged particles for KERMA determinations, and secondary particles (electrons, protons, 14C, 4He and 7Li) to compute absorbed dose in cultures. Dose-survival curves were modified according to the dose correction factors determined from computational studies. New radiobiological parameters of the photon isoeffective dose models for osteosarcoma and metastatic melanoma tumours were obtained. Dosimetry implications considering cutaneous melanoma patients treated in Argentina with BNCT were assessed and discussed.ResultsKERMA values for the monolayer-cultured cells overestimate absorbed doses of radiation components of interest in BNCT. Detailed dose calculations for the osteosarcoma irradiation increased the relative biological effectiveness factor RBE1% of the neutron component in more than 30%. The analysis based on melanoma cases reveals that the use of survival curves based on KERMA leads to an underestimation of the tumour doses delivered to patients.ConclusionsConsidering detailed dose calculation for in-vitro experiments significantly impact on the prediction of the tumor control in patients. Therefore, proposed methods are clinically relevant.  相似文献
