研究寡核苷酸芯片的重复性与间隔臂(spacer)和探针长度之间的相关性。设计12条不同长度的带有不同spacer的探针,与749bp荧光标记靶序列杂交。扫描分析三次杂交结果,用Quantrray定量分析软件进行分析,随探针长度的延长,杂交信号的变异系数逐步降低,15mer的探针杂交的信号较弱,杂交不够稳定,重复性也相对较差,20mer、25mer、30mer的探针的变异系数逐渐降低。spacer为15时变异系数最小。说明选择spacer为poly(dT)15的25mer和30mer的探针可以获得较好的重复性。  相似文献

The distribution of the Pacific herring of various sizes from the Okhotsk and Gizhigin-Kamchatka populations is discussed. The dependence of herring food composition on the condition of the plankton community is demonstrated. In the spring season the feeding rate of juveniles and fattening adult herring depended on the degree of its fatness and on the maturity of the gonads in the prespawning period.  相似文献

目的测定近交系大鼠MIJ、HFJ和封闭群Wistar大鼠的脏器系数、体温、呼吸频率、肠管长度,观察两个近交系大鼠的生理表型。方法 采用常规方法对大鼠脏器系数、体温、呼吸频率及肠管长度进行测定,并对HFJ、MIJ、Wistar大鼠三者间进行比较。结果①体重:HFJ、MIJ、Wistar大鼠同性别间比较差异无显著性。②脏器系数:HFJ与Wistar大鼠雄性间心脏、肺,雌性间子宫、肝;MIJ与Wistar大鼠雄性间肺、脾,雌性间脑、脾,差异均有显著性。HFJ与MIJ大鼠雄性间脑,雌性间脑和脾差异有显著性。HFJ大鼠不同性别间肝、脑、眼球差异有显著性。MIJ大鼠不同性别间,脑、心脏、肾、肺、脾和眼球差异均有显著性。③体温:HFJ雌性大鼠明显高于Wistar雌性,雄性间差异无显著性。HFJ大鼠雌性高于雄性,差异有显著性。④呼吸频率:HFJ雌性大鼠低于同性Wistar大鼠,MIJ大鼠雌性和雄性均高于同性别Wistar大鼠,差异有显著性。⑤肠管长度:HFJ与Wistar同性别间比较,肠管总长和小肠差异有显著性。MIJ与Wistar大鼠雄性间肠管总长、大肠、小肠、盲肠差异均有显著性,雌性间肠管总长、小肠、盲肠差异有显著性。HFJ与MIJ大鼠同性别间差异均无显著性,不同性别的HFJ和MIJ的肠管总长、大肠和小肠差异有显著性。结论 MIJ和HFJ近交系大鼠的脏器系数、体温、呼吸频率、肠管长度都各有其独特的生理概貌。  相似文献

For many years, the expression "cutoff effect of anesthesia," has been used to denote the failure of the higher alcohols or paraffins to produce anesthesia. As such, it is used to assess the plausibility of specific models, proposed for anesthesia. However, the uses were shown, in many respects, to be problematic. This article augments the notion of the cutoff to fit for all cases in which only some of the molecules in a homologous series are anesthetics. We find that the location of the cutoff points is affected by three free energy quantities: that of the adsorption of the agent to the anesthetic "site" (f(sl,site)), that of the perturbation of the site (f(ll,site)), and that of the evaporation of the agent from its pure condensed phase (Deltamu degrees (evaporation)). This outcome indicates that the cutoff cannot be attributed to a single parameter. In addition, the analyses that attribute the cutoff to the failure of compounds to obey the much-used Meyer-Overton correlation will have to be amended. This article shows that cutoff results can be used to elucidate the structure of a site.  相似文献

以贵州4个茶叶产地(贵定、都匀、凤冈、湄潭)生产的新鲜茶叶(贵定云雾茶、都匀毛尖、凤冈锌硒茶、湄潭翠芽)及种植土壤为材料,研究了不同茶树叶片及种植土壤的锌、硒含量,研究不同土壤类型及不同深度土壤锌、硒含量的差异;茶树当年生新叶(一芽二叶)和生长1年以上老叶的锌、硒含量变化;土壤锌、硒含量与新老叶片锌、硒含量之间的关系。结果显示,随着茶园土壤深度(0~20 cm,20~40 cm)的增加,锌含量增加了2.0%~18.0%,而土壤硒含量则没有明显变化;不同样地土壤和同一样地土壤不同深度的锌、硒含量差异均达显著水平(Sig.0.01),但差异未见明显规律;茶树新叶锌、硒含量比老叶分别高2.3~4.0倍和1.2~3.0倍;新叶的锌、硒含量与土壤锌、硒含量呈正相关关系,且新叶中的锌、硒含量之间也呈正相关关系。表明土壤中的锌、硒含量来源于成土母岩并直接影响茶叶的品质,因此合理选择土壤类型对种植优质茶和规划发展茶产业具有重要意义。  相似文献

The common thread of evolution runs through all science disciplines, and the concept of evolution enables students to better understand the nature of the universe and our origins. “Science and the Concept of Evolution” is one of two interdisciplinary science Core courses taken by Dowling College undergraduates as part of their General Education requirements. The course examines basic principles and methods of science by following the concept of evolution from the big bang to the origin and evolution of life. Case studies of leading scientists illustrate how their ideas developed and contributed to the evolution of our understanding of the world. Evidences for physical, chemical, and biological evolution are explored, and students learn to view the evolution of matter and of ideas as a natural process of change over space and time.  相似文献

This article explores the relationship between Vietnam's national interests and the international law of the sea. Vietnam's national interests in the marine sphere include defense (the maintenance of a maritime buffer); international relations (enhancing its regional position through joint development and favorable resolution of boundary and fisheries disputes); and economics (stimulating development of its maritime resources). Although ratification of the 1982 Law of the Sea Convention would be somewhat disadvantageous to Vietnam's maritime defense strategy, many provisions would promote Vietnam's interests. Moreover, ratification and implementation of the Convention would signal Vietnam's support for international law and its desire to “rejoin”; the community of Southeast Asian nations. It might also encourage resolution of its boundary disputes, thus opening new areas to resource exploration and development by foreign companies.  相似文献

在果树休眠期,通过主干注射果树营养液替代果园土壤施肥,对苹果外部形态、果实品质及其生理作用影响进行了研究。结果表明,萌芽期提前,花期早而且盛花期花朵开放整剂稳定;萌芽率、成花率、成枝率提高;果树根系鲜重和数量显著增加,其中20-40cm土层中增长最为显著;叶片的光合速率和叶绿素(a b)含量也大幅度上升;果实品质明显改善,产量增加。从总体看,果树营养注射液有广阔的发展前景和推广价值。  相似文献

Two types of selection operate (and complement each other) in plants under domestication: (a) conscious or intentional selection applied by the growers for traits of interest to them; (b) unconscious or automatic selection brought about by the fact that the plants concerned were taken from their original wild habitats and placed in new (and usually very different) human-made or human-managed environments. The shift in the ecology led automatically to drastic changes in selection pressures. Numerous adaptations vital for survival in the wild environments lost their fitness under the new sets of conditions. New traits were automatically selected, resulting in the build-up of characteristic “domestication syndromes,” each fitting the specific agricultural environment provided by the farmer. The present paper assesses the evolutionary consequences of the introduction of the wild plants into several sets of contrasting farming situations. These include: (a) the type of maintenance applied, whether seed planting or vegetative propagation; (b) the plant organs for which the crop has been grown, whether they are reproductive parts or vegetative parts; (c) the impact of the system of tilling, sowing, and reaping on the evolution of grain crops; (d) the impact of the horticultural environment on fruit crops.  相似文献

Viruses and cells co-evolve due to the parasitic nature of viruses. Yet there are no models suggesting how the unicellular organisms and their viruses might co-evolve structurally. Here, in this study, we plunge into this unexplored field from a wide perspective and try to describe some of the intriguing ways in which viruses may have shaped the cellular life forms on the ancient Earth. At first we propose a scenario where viruses act as a driving force in the emergence of bacterial cell walls by providing favorable intermediates for the otherwise improbable steps in the cell wall generation. We also discuss the role of viruses in the evolution of cell surface components such as receptors and second membranes. Finally we focus on hypothetical proto-viruses, the selfish abusers of the RNA-world, in explaining some of the very early stages in the origin and evolution of life. Proto-viruses may be responsible for creating the first true cells in order to support their selfish needs. In this model we also suggest a logical pathway to explaining the emergence of modern viruses.  相似文献

1. By using dibromothymoquinone as the electron acceptor, it is possible to isolate functionally that segment of the chloroplast electron transport chain which includes only Photosystem II and only one of the two energy conservation sites coupled to the complete chain (Coupling Site II, observed P/e2 = 0.3–0.4). A light-dependent, reversible proton translocation reaction is associated with the electron transport pathway: H2O → Photosystem II → dibromothymoquinone. We have studied the characteristics of this proton uptake reaction and its relationship to the electron transport and ATP formation associated with Coupling Site II.

2. The initial phase of H+ uptake, analyzed by a flash-yield technique, exhibits linear kinetics (0–3 s) with no sign of transient phenomena such as the very rapid initial uptake (“pH gush”) encountered in the overall Hill reaction with methylviologen. Thus the initial rate of H+ uptake obtained by the flash-yield method is in good agreement with the initial rate estimated from a pH change tracing obtained under continuous illumination.

3. Dibromothymoquinone reduction, observed as O2 evolution by a similar flash-yield technique, is also linear for at least the first 5 s, the rate of O2 evolution agreeing well with the steady-state rate observed under continuous illumination.

4. Such measurements of the initial rates of O2 evolution and H+ uptake yield an H+/e ratio close to 0.5 for the Photosystem II partial reaction regardless of pH from 6 to 8. (Parallel experiments for the methylviologen Hill reaction yield an H+/e ratio of 1.7 at pH 7.6.)

5. When dibromothymoquinone is being reduced, concurrent phosphorylation (or arsenylation) markedly lowers the extent of H+ uptake (by 40–60%). These data, unlike earlier data obtained using the overall Hill reaction, lend themselves to an unequivocal interpretation since phosphorylation does not alter the rate of electron transport in the Photosystem II partial reaction. ADP, Pi and hexokinase, when added individually, have no effect on proton uptake in this system.

6. The involvement of a proton uptake reaction with an H+/e ratio of 0.5 in the Photosystem II partial reaction H2O → Photosystem II → dibromothymoquinone strongly suggests that at least 50% of the protons produced by the oxidation of water are released to the inside of the thylakoid, thereby leading to an internal acidification. It is pointed out that the observed efficiencies for ATP formation (P/e2) and proton uptake (H+/e) associated with Coupling Site II can be most easily explained by the chemiosmotic hypothesis of energy coupling.  相似文献

对桑-蚕系统中镉的吸收,累积与迁移行为研究结果表明:(1)桑树对土壤镉污染有一定的耐性,桑树镉累积量和相对累积率与土壤镉浓度的关系可分别用回归方程Ta=a+blog(Sc)和log(Ra)=a+blog(Sc)描述。(2)随着土壤镉浓度的增加,镉在桑树根部的分布率明显增加,地上部分的分布率有所降低,运转到叶片的比率明显降低。(3)蚕体,蚕砂和蚕茧的镉含量,镉累积量,随着桑叶镉含量的增加而增加。  相似文献

The role of mechanisms underlying differentiation is considered in malignant transformation of hemoblastoses and epithelial tumors. In hemoblastoses, differentiation is intimately related to malignant transformation and they are underlain by the same mechanisms. Immunophenotyping of hemoblastoses is fully based on successive stages of their differentiation with characteristic expression of differentiation antigens. Unlike hemoblastoses, epithelial tumors gradually, in the course of progression, lose their differentiation due to the degradation of the connections with the microenvironment, which controls the direction and level of epithelial differentiation. Therefore, carcinomas are characterized by varying degrees of “antigenic simplification”, including the epithelial-mesenchymal transition.  相似文献

To enrich the Hennigian internodal conception of species, a new formalization of the definition of the species concept is proposed. This rigorous definition allows for considerable unification of the various, and sometimes conflicting, techniques of species delimitation used in practice. First, the domain of such a definition is set out, namely, the set of all organisms on Earth, past, present, and future. Next, the focus is on the genealogical relationship among organisms, which provides the key to analysing the giant or global genealogical network (GGN) connecting all these organisms. This leads to the construction of an algorithm revealing the topological structure of the GGN, from families to lineages, ending up with a definition of species as equivalence classes of organisms corresponding to branches of the 'tree of life'. Such a theoretical definition of the species concept must be accompanied by various recognition criteria to be operational. These criteria are, for example, the ill-named 'biological species concepts', 'phylogenetic species concepts', etc., usually, but wrongly, presented as definitions of the species concept. Besides clarifying this disputed point, the definition in the present study displays the huge diversity of the scales (time-scale and population size) involved in actual species, thus explaining away the classical problems raised by previous attempts at defining the species concept (uniparental reproduction, temporal depth of species, and hybridization).  © 2006 The Linnean Society of London, Biological Journal of the Linnean Society , 2006, 89 , 509–521.  相似文献
