Using extraction with 0.75% aqueous ammonium oxalate, the following polysaccharide fractions were isolated: tanacetans TVF, TVS, and TVR from floscules, sprouts, and roots, respectively, of Tanacetum vulgare L., spread throughout the European North of Russia. The sugar chain of tanacetan TVF consists of D-galacturonic acid (61.4%), arabinose (14.7%), galactose (10.2%), and rhamnose (3.7%) as the main constituents as well as xylose, glucose, mannose, apiose, and 2-O-methylxylose in trace amounts. Tanacetans TVS and TVR were shown to differ in the sugar quantitative composition. They contain 67 and 28% galacturonic acid, respectively. A partial acid hydrolysis of the tanacetan TVF gave a polysaccharide fragment TVF1, -1,4-D-galacturonan (GalA 98.2%). Digestion with pectinase (-1,4-D-polygalacturonase) resulted in fragment TVF3, containing residues of arabinose (27.1%) and galactose (17.3%). NMR spectroscopy allowed detection of the terminal residues of -Araf and -Galp as well as of the residues of -Arap substituted in 3,5- and 5-positions. Thus, tanacetan TVF was proved to be a pectic polysaccharide.  相似文献

Protoplasts isolated from cultured rice cells of an A-58 cytoplasmic male sterile line (A-58 MS)(Oryza sativa L.) were used to investigate the regeneration of rice plants. A cultured cell line (T3) of A-58 MS with a high growth rate and dense cytoplasm was selected. About 10% of the protoplasts prepared from this established cell line plated in RY-2 (a new medium) formed colonies. The calli formed shoots and roots in the regeneration medium and developed into whole plants.Protoplasts also were prepared from suspension cultures of 25 other varieties of rice using the same methods. The protoplasts isolated from two of the 25 varieties, Fujiminori and Toyotama, had high rates of cell division in RY-2 medium. Red clover (Trifolium pratense L.) seedlings were screened for the ability to regenerate plantlets from hypocotyl-derived callus tissue. Media sequences described by Beach and Smith (1979) and Collins and Phillips (1982) and a variation using media from both sequences were tested. Plantlets were regenerated from three out of 642 genotypes. In all three cases, callus was initiated on B5C medium and regeneration was accomplished on SPL medium. Attempts to regenerate plants from petiole-derived callus tissue have so far been successful only with regenerants of clone F49. Petiole callus from epicotyl-derived F49 plants proved to be non-regenerative. Pollen viability varied significantly among individuals regenerated from callus cultures of clone F49. Root tip squashes from F49 regenerants revealed the normal diploid chromosome number (2n=14). Forty samples of inflorescences and leaves of wild Tanacetum vulgare L. var. vulgare were collected in 20 habitats from Lithuania. The essential oils were analyzed by GC and GC/MS. The 57 identified compounds in the oils made up 80.7–99.6%. According to the cluster analysis the volatile oils were divided into four groups with 1,8-cineole (23.6–46.3%, 11 oils), trans-thujone (35.7–78.4%, 6 samples), camphor (19.8–61.8%, 17 oils) and myrtenol (13.1–24.9%, 6 samples) as main constituents. The inflorescences and leaves of tansy plants formed the oils with the same dominating constituent in 15 of the 20 habitats investigated. The leaves in five localities produced oils of the 1,8-cineole chemotype, while the inflorescences biosynthesized oils of the camphor type in three habitats and of the myrtenol type in two habitats. Amounts of the 1,8-cineole in all leaf oils were greater than that in inflorescence oils of the plants from the same locality. An opposite correlation was determined for camphor, myrtenol, cis- and trans-thujone. The myrtenol chemotype was not noticed earlier in the essential oils of T. vulgare.  相似文献

Leaf discs from four strains and petioles from six strains of Gossypium hirsutum were cultured on a variety of media. Callus formed from explants on all media, though embryogenesis was highly specific. Embryos formed from only three strain x media combinations. A small percentage of these embryos developed into plantlets. Callus tissue was induced on barley mesocotyl explants of germinated seven-day-old seedlings on MS medium supplemented with 2,4-D or 2,4,5-T in high concentrations. Two morphologically different tissue cultures were maintained in vitro for a long time: a callus tissue without organogenesis and a culture with high rhizogenic capacity. Shoots and plantlets were generated when the auxin-media induced callus was transferred to medium supplemented with 3 M TIBA. In 62% of cultures, during the first five subcultures, four to twentyeight plants per single mesocotyl were obtained. Some cultures produced shoots even in the 9th subculture, being in culture for nearly 14 months. The largest number of plants obtained per one mesocotyl was forty. Summary The cell ultrastructure in three types of callus obtained from leaf explants ofAesculus hippocastanum L. has been studied. Remarkable differences have been shown between the cells of the forerunner E1 callus and those of the callus arising from it, according to the culture conditions.The peculiar characteristics of E1 are the scarcity of intercellular spaces and the occurrence of autophagic vacuoles in the cells.An embryogenic friable callus (E2) is formed in time when E1 is maintained on solid culture medium. The E2 cells show cytological features typical of a higher metabolic level and contain starch. Diffused middle lamella digestion leads to the detachment of small embryogenic cell aggregates consisting of vacuolated parenchymatous-like cells and small meristematic cells which may be regarded as embryoids initials.Shaking E1 in the same liquid medium and subsequent culture on solid medium lead to the differentiation of a non-embryogenic callus (NE), whose cells are very large and highly vacuolated, devoid of starch and with organelle-rich cytoplasm.  Morphological and genetical differences of twenty Finnish tansy (Tanacetum vulgare) genotypes were studied. The genotypes were distinguishable by morphology, and number of flower heads, which correlated positively with height of the plant and height of the corymb. The mean nuclear DNA content in the leaves of tissue-cultured tansy plantlets was 8.86 pg, and variation between genotypes was 27%. The genotypes were also distinguishable on the basis of their RAPD patterns. The RAPD and morphological data were subjected to analysis of principal components, according to which the genotypes were separated into two main groups. Group I included 7 genotypes from southern Finland and 2 genotypes from the eastern lake district. They started to flower later, had more flower heads and nodes per stem, and the corymb was longer than in the other genotypes that originated in the western and central part of Finland and formed group II. Our data are supported by previous studies and suggest that group I may represent native tansy populations in Finland, whereas group II may represent tansy genotypes that have been transported to other parts of Finland through agriculture and attempts of domestication. Protoplasts isolated enzymatically from seedling roots, hypocotyls and cotyledons of Lotus corniculatus L. produced callus which underwent prolific shoot regeneration. Summary Regeneration of adventitious shoots of pothos (Epipremnum aureum Linden and Andre) ‘Jade’ was obtained using leaf and petiole explants preprated from shoot tips of 3-yr-old greenhouse-grown plants. Explants were cultured on Murashige and Skoog (MS) basal medium supplemented with thidiazuron (TDZ), 6-(4-hydroxy-3-methy-trans-2-butenyl-amino)purine (zeatin) or N-isopentenylaminopurine (2iP) individually with α-naphthaleneacetic acid (NAA) in 18 combinations. Callus was initiated from cut surface and along the midrib or major vein of leaf sections. Shoot regeneration from leaf and petoole explants occurred in 30d on medium containing 1, 5 or 10μM TDZ with 0.5 or 1.0μM NAA except petioles on medium with 10 μM TDZ and 1.0 μM NAA where regeneration failed. More time (50d) was needed for shoot regeneration when explants were cultured on medium containing either 2iP or zeatin with NAA. Regeneration frequencies were up to 20% and 50% for leaf and petiole explants, respectively. Shoot numbers per responding explant attained 30 for leaf and petiole explants on medium containing TDZ but only one to four on medium containing either 2iP or zeatin. These results indicate that TDZ is a more effective cytokinin for in vitro regeneration of pothos than either zeatin or 2iP.. Shoots elongated readily and rooted well on MS basal medium, without plant growth regulators. Plantlets acclimatized rapidly and grew vigorously in the greenhouse after transfer to pots containing a commerecial potting medium.  相似文献

A effective protocol for complete plant regeneration via somatic embryogenesis has been developed for Ocimum basilicum L. Callus was initiated from leaf explant of young plant on supplemented with 2,4-dichlorophenoxyacetic acid (2,4-D) 1.0 mg l(-1), 3% sucrose and 0.9% agar. The calli showed differentiation of globular structure embryos when transferred to MS medium containing 2,4-D 0.5 mg l(-1) and BAP 1.0 mg l(-1). The maximum globular structure embryos were further enlarged and produced somatic embryos in MS basal medium supplemented with BAP 1.0 mg l(-1)+NAA 1.0 mg l(-1) + KN 0.5 mg l(-1). Continued formation of globular embryo and germination of embryos occurred in this medium. Complete plantlets were transferred onto specially made plastic cup containing soilrite followed by their transfer to the garden soil. Survival rate of the plantlets under ex vitro condition was 80%.  相似文献

Summary The petiole expiants of coriander when cultured on a solid or liquid Murashige and Skoog's medium supplemented with 0.5 mg/l 2,4-D developed a large number of meristemoids. These meristemoids could then be induced to form adventive embryos when transferred into a liquid MS-medium without 2,4-D. The development of the expiants leading to the formation of meristemoids was followed anatomically and by using SDS-polyacrylamide gel electrophoresis of soluble proteins at various days of culture.  相似文献

Effects of three antibiotics (cefotaxime, rifampicin and gentamicin) were tested on in vitro shoots cultures of tansy (Tanacetum vulgare L.)- These antibiotics were selected because endophytic bacteria isolated from the in vitro shoot cultures of tansy showed that all bacteria were Gram-negative and their growth was reduced by these three antibiotics. Five isolates were Enterobacteriaceae, three were fluorescent Pseudomonas, and two were aerobic bacteria. Increased concentrations of antibiotics caused usually linear or quadratic changes on the initiation of shoot growth, shoot number, growth rate, and shoot height. These changes and changes in pH of the culture media were tansy genotype-dependent following treatments with gentamicin. Also, the treatment with rifampicin or cefotaxime showed a genotype-dependent effect, because they resulted in significantly higher percentage of rooted plants in one of the three tansy genotypes tested. The growth rate and length of shoots were reduced in the media containing both gentamicin and rifampicin, but less so than in media containing both gentamicin and cefotaxime.  相似文献

A protocol for plant regeneration in Indian cultivars of barley (Hordeum vulgare L.) has been developed using mature embryo culture. The influence of various auxins 2,4-D (2,4-dichlorophenoxyacetic acid), Dicamba (3,6-dichloro-o-anisic acid) and Picloram (4-amino-3,5,6-trichloropicolinic acid) on the callus induction and subsequent plant regeneration revealed highest percent of callus induction form cultivar (cv) BL 2 on MSB5 medium (MS salts + B5 vitamins) supplemented with 6 mg l?1 Picloram, but maximum number of shoot buds (6–13) were regenerated on MSB5 medium containing 0.5 mg l?1 Picloram. Regenerated shoots were rooted on half-strength MSB5 medium. Plantlets were successfully transferred to soil and grown to maturity in greenhouse. The effect of copper sulphate revealed significant improvement in callus induction and plant regeneration when the concentration of CuSO4 was increased to 3 μM (30 times higher than normal MS medium) for cv BL 2. Regeneration potential differed for different cultivars of barley used, with highest for cv BL 2 and lowest for cv BH 924. We conclude that the Indian barley genotypes exhibit plant regeneration from mature embryo cultures. The protocol has potential application in barley improvement through genetic engineering.  相似文献

Callus was induced from hybrids between cultivated barley (Hordeum vulgare L. ssp. vulgare) and ten species of wild barley (Hordeum L.) as well as from one backcross line ((H. lechleri x H. vulgare) x H. vulgare). Successful callus induction and regeneration of plants were achieved from explants of young spikes on the barley medium J 25–8. The capacity for plant regeneration was dependent on the wild parental species. In particular, combinations with four related wild species, viz. H. jubatum, H. roshevitzii, H. lechleri, and H. procerum, regenerated high numbers of plants from calli.  相似文献

Transverse thin cell layers (tTCLs) (0.2–0.4 mm thick) excised from the pseudo-bulblets of L. longiflorum in vitro were placed on the Murashige and Skoog medium supplemented with 3 % sucrose and a concentration of N-(2-chloro-4-pyridyl)-N′-phenylurea (forchlofenuron, CPPU) varying from 0.1 to 10 μM. Buds were obtained from the tTCL pseudo-bulblet expiants cultured in the presence of CPPU in the light but not in the dark. The number of buds formed per tTCL was the highest with 1 or 3 μM CPPU (16.8 ± 1.1 and 16.2 ± 1.6 buds per tTCL, respectively). Testing a combination of sucrose (from 1 to 12 %) and CPPU (from 1 to 10 μM) confirmed that the number of buds produced per tTCL was the highest when 1 or 3 % sucrose was combined with 1 or 3 μM of CPPU. The regenerated shoots were transferred to soil after rooting with 10 μM indole-3-byturic acid and normal plants were produced.  相似文献

Summary The inheritance of the ability of barley anthers to produce microspore-derived callus in vitro was investigated. The genotypes selected were the two spring cultivars Dissa (D) and Sabarlis (S), the two F1 hybrids (DxS, SxD), the two backcross generations [Dx(DxS), Sx(DxS)], and an F2 generation derived from DxS. From a number of individuals of each generation, the first five spikes were harvested sequentially and after pre-treatment the anthers were removed and placed in culture. Cultures were scored for microspore callus production and plantlet differentiation. Although Dissa gave a significantly higher level of callus production than Sabarlis, the overall frequencies of green and albino plant production were higher from Sabarlis. There was no significant difference between reciprocal F1 hybrids. Analysis of variance revealed significant differences in response between the spikes sampled from the plants. This was the major source of variation in the experiment. Spike to spike variation also appeared to be a heritable character.  相似文献

Summary An in vitro propagation system was developed for Echinacea purpurea L. (purple coneflower), a medicinal plant commonly used in the treatment of colds, flu and related ailments. Echinacea seeds were found to be contaminated with systemic fungi and therefore an optimized minimal concentration of Plant Preservation Mixture (PPM) was incorporated in the seed germination medium to recover sterile seedlings. Regeneration was induced on petiole explants from 2-month-old sterile seedlings cultured on medium supplemented with benzylaminopurine (BAP) or thidiazuron (TDZ) in combination with indoleacetic acid (IAA). Two distinct forms of regeneration were identified in cultured petiole explants with histological and morphological observations, viz. the direct formation of somatic embryos on the epidermis and the de novo development of shoots from callus tissues formed in subepidermal cell layers. the results of this study have established a micropropagation system for E. purpurea that will provide sterile plant material for further investigations into medicinally active biochemicals and may facilitate mass production of high-quality E. purpurea plants for the commercial market.  相似文献

Summary Protoplasts isolated from mesophyll tissues of cauliflower (Brassica oleraceavar. botrytis) were induced to divide in culture, with 2% of them producing calli. Upon transfer to a regeneration medium containing a low auxin/cytokinin balance (0.02mg/l 1-naphthaleneacetic and 2mg/l 6-benzylaminopurine), they displayed an extensive production of hairy roots before the regeneration of shoots.
