Summary The non-tandem inverted duplication in the 2-m DNA of Saccharomyces cerevisiae has a length of 0.19 m and is located asymmetrically along the molecule. The majority of the dumb-bell structures that are formed upon denaturation and selfannealing of the 2-m monomer consists of the renatured inverted duplication sequences as double stranded stem and two single stranded loops of 0.67 m±0.06 m (S-loop) and 0.86 m±0.05 m (L-loop) length. Two additional size classes which comprised 5–10% of the measured molecules had contour lengths of around 1.7 m and 2.1 m. The smaller dumb-bells contained two S-loops and the larger dumb-bells contained two L-loops as was shown by heteroduplex mapping with an HindIII fragment from the L-loop. Two models which assume illegitimate or site specific recombination, are presented to explain the generation of double S-loop and double L-loop molecules. At least part of the 4-m and 6- circular molecules present in the yeast supercoiled DNA fraction are shown to be dimers and trimers of 2-m monomers, but often with inverted loop segments most probably due to intramolecular recombination between sequences of the inverted duplication.2-m DNA is used to indicate the supercoiled DNA fraction although in our measurements the average monomeric length is 1.9 mPart of this work has been presented at the Conference: The Genetics and Biogenesis of Chloroplasts and Mitochondria, Munich, August, 1976  相似文献

Summary The order and stoichiometry of the binding of phlorizin and sodium to the renal brush-border membraned-glucose transporter are studied. The experimental results are consistent with a random-binding sites is one-to-one. When the kinetics of phlorizin binding are measured as a function of increasing sodium concentration no significant variation is found in the apparent number of binding sites; however, the apparent binding constant for phlorizin decreases rapidly from approximately 16 m at [Na]=0 to 0.1 m at [Na]=100mm and approaches 0.05 m as [Na]. The experimental data are fit to a random carrier-type model of the coupled transport of sodium andd-glucose. A complete parameterization of the phlorizin binding properties of this model under sodium equilibrium conditions is given.  相似文献

Summary In addition to high molecular weight plastomic DNA, chloroplasts of Acetabularia cliftonii also contain small supercoiled DNA molecules (Green 1976). Restriction enzyme analysis of this 4.28±0.15 m DNA resulted in a 14.1 kbp circular restriction map. Southern blot analysis revealed that the high molecular weight plastomic DNA of A. cliftonii contains all of the 4.3 m DNA restriction fragments suggesting that the whole sequence of the 4.3 m DNA is an integral component of the plastome.Heterologous DNA hybridizations showed that 4.3 m DNA has no homology with either chloroplast ribosomal DNA from mustard or total chloroplast DNA from spinach. Cryphodera kalesari n. sp., recorded on Colebrookea oppositifolia Sm. from the Kalesar forests (Haryana), India, is described. Females: swollen, saccate, with two to three lip annules, with vulval lips protruding. Second stage juveniles: length 384±19 m, spear=26.5±1 m, tail 46.5±6 m, hyaline portion 21.5±2.5 m, en face dorsal and ventral arcs of head skeleton bifurcate. Males: not found.  相似文献

The distribution of mitochondria, their content and concentration (expressed as the ratio of the mean volume of mitochondria and the surface of the sensory axon) were determined in group-III and-IV nerve fibres innervating the knee joint capsule in the cat. Mitochondria mainly accumulated in axonal swellings (beads) and end bulbs of the terminal branches. Between single nerve fibres, marked differences in the content and the concentration of mitochondria were obtained in proximal portions (inside of the perineurium) and in distal portions (unmyelinated sensory endings). In group-III nerve fibres, the mitochondrial concentration ranged from 0.005 to 0.030 m3/m2 (proximal portion) and from 0.016 to 0.080 m3/m2 (distal portion). In unmyelinated group-IV nerve fibres, the values also showed a broad variation ranging from 0.001 to 0.011 m3/m2 (proximal portion) and from 0.003 to 0.019 m3/m2 (distal portion). The wide range of mitochondrial concentrations may reflect different energy consumption during receptive processes: nerve fibres with a low mechanical threshold and a high probability of excitatory events may be rich in mitochondria, whereas fibres with a high mechanical threshold and a low probability of excitatory events may be poor in mitochondria.  相似文献

Zusammenfassung Die schwachelektrischen Mormyridae haben eine dreischichtige Epidermis, deren innere Schicht aus nur etwa 0,22 m dicken sechseckigen Zellen von ca. 60 m Durchmesser besteht. Die etwa 2 m dicken, linsenförmigen Kerne von 7,6 m Durchmesser liegen am Zellrand. Die Zellen sind zu Säulen aufgeschichtet. Ihr Rand ist ausgezackt und dort, wo er die Säulengrenze erreicht, auf etwa 0,34 m verdickt. In der Nähe der Säulengrenzen sind die Zellen über Desmosomen mit den Nachbarn in der eigenen und in der angrenzenden Säule verbunden. Diese Epidermisschicht ist auf die Körperpartien beschränkt, in denen auch Elektrorezeptoren ausgebildet sind.Die beiden anderen Epidermisschichten haben den üblichen Aufbau einer Fischepidermis, abgesehen vom Fehlen der Becherzellen.
Ultrastructure of the electroceptor epidermis of the Mormyridae (Teleostei, Pisces)
Summary The weakly electric fish of the family Mormyridae have a three layered epidermis, with a medium layer consisting of hexagonal cells of only 0.22 m in thickness and about 60 m in diameter. The lens-shaped nuclei are about 2 m thick and 7.6 m in diameter and are situated near the border of the cells. The cells are piled up to hexagonal columns. Their margin is serrate and where it reaches the boundary of the column, it has a thickness of about 0.34 m. Close to the boundaries of the columns, the cells are linked to their neighbours within the column and of the adjoining column by desmosomes. This layer of the epidermis is confined to those regions of the body surface which also contain electroreceptors.The other layers of the epidermis have a structure as usual in fish, except for the lack of goblet cells.

Résumé Capillaria dessetae n. sp. est décrit à partir de spécimens parasites du Lagomorphe Ochotona roylei récoltés au Népal. Le mâle est caractérisé par la présence de deux ailes latéro-caudales longues de 65–70 m, une bourse caudale soutenue par deux expansions bifurquées, un spicule long de 190–220 m, un cirre inerme et un canal éjaculateur long de 1200–1300 m. La femelle est caractérisée par des ornementations cuticulaires péri-vulvaires et un oeuf de grande taille (65–69 × 32–38 m) à surface finement réticulée. Les espèces C. legerae et C. murissylvatici (toutes deux parasites de Rongeurs) sont très proches morphologiquement, avec en particulier des dimensions du spicule et une morphologie des bourses caudales très comparables. Ces trois espèces sont néanmoins bien différenciées par les caractéristiques des oeufs. Une représentation linéaire schématique de l'appareil reproducteur mâle interne est proposée, pour ces trois espèces. Ceci met en relief des différences de longueur du canal éjaculateur et du sphincter, qui sont rarement utilisées en systématique, et permet une diagnose facile des espèces. Une telle représentation schématique linéaire serait utile dans toutes les études taxonomiques de Capillariinae.
Capillaria dessetae n. sp. is described from specimens which are parasites of Ochotona roylei collected in Nepal. Males are characterized by the presence of two laterocaudal alae 65–70 m long, a caudal bursa supported by two forked protrusions, a spicule 190–220 m in length, a non-spiny cirrus and an ejaculatory canal 1200–1300 m in length. Females are characterized by cuticular ornamentations associated with the vulva and large eggs (65–69 × 32–38 m) which have a finely striated surface. The species C. legerae and C. murissylvatici (both parasitic in rodents) are morphologically very close, particularly in the spicule dimensions and in the caudal bursa. These three species, however, are clearly differentiated by characteristics of the eggs. Linear schematic representations of the internal male reproductive tracts are given for the three species. This emphasises the differences in the lengths of the ejaculatory canal and sphincter, which are generally not used in systematics, but which allow clear differential diagnoses of the species. Such linear diagrams would be useful in all taxonomic studies of the Capillariinae.

Effects of Pb2+, Ni2+, Hg2+ and Se4+ on cultured human glioma U-343MG cells were investigated considering uptake, toxicity and, in combination with radiation, clonogenic cell survival. The cells were exposed to 0-100 m of the metals for a week before the evaluation. The tests showed a tendency to toxicity with 10 m nickel although not significant (P > 0.05). Selenium, lead and mercury exerted a significant toxicity (P < 0.05) at 2.5 m, 10 m and 1 m, respectively. To challenge the clonogenic cell survival capacity, the cells were irradiated with60Co photons after being exposed to the highest nontoxic concentration of the different metals. The clonogenic cell survival tests, after irradiation, showed no significant change if the cells were exposed to 5 m nickel, 0.5 m selenium or 5 m lead compared with those not exposed. Mercury, 0.1 m, gave a relative reduction in survival compared with only irradiated cells of 58 ± 17%. Thus, only mercury affected the radiation-induced damage and/or repair. When exposed to the highest nontoxic concentrations of the different metals, the cultures did not display a significant uptake ratio (metal concentration ratio of exposed cells to control cells) of nickel (3.1 ± 3.3), only a small uptake ratio of selenium (4.0 ± 0.4), while there was a large uptake ratio of both lead (2.6 ± 1.7) x 102 and mercury (1.5 ± 0.2) x 101. The results indicated that nickel was neither especially toxic nor influenced the clonogenic cell survival after irradiation. Mercury was more toxic and also influenced the radiation sensitivity. Lead was taken up strongly but did not influence the radiation sensitivity. Selenium accumulated but gave no detectable effect on the radiation sensitivity.  相似文献

Factors associated with the production of extracellular lipase and proteinase by Pseudomonas fluorescens B52 during the late-log, early-stationary phase of grown were examined. Active lipase production by resting cell suspensions was observed when cells were harvested during the log phase (A600 of 0.3–0.9) Resting suspensions of younger cells (A600<0.1) synthesized lipase after a significant lag. Addition of cells of the proteinase-and lipasedeficient mutant P. fluorescens RM14 to B52 cells at low density resulted in stimulation of lipase and proteinase production. Similar results were found using cell-free culture fluid of RM14. Gel filtration on Biogel P2 revealed that the stimulatory factor co-chromatographed with the iron(III) siderophore, pyoverdine. Partially purified pyoverdine stimulated enzyme synthesis at a concentration of 6 M while having no effect on activity of preformed enzyme. Production of pyoverdine and extracellular enzymes was also stimulated by transferrin, a strong iron(III) binding protein. Growth of B52 in deferrated media was limited to 27% of that found with untreated media. Maximum pyoverdine, proteinase and lipase synthesis was obtained at a final iron(III) concentration of 5.75 M. Growth was maximal in 8.75 M iron(III) while synthesis of pyoverdine, proteinase and lipase was reduced to 3.6, 6.6 and 30% respectively in 23.75 M iron(III). Lipase activity in cell-free culture fluid was slightly inhibited by the addition of up to 400 M iron(III) while proteinase activity was unaffected. In dilute cell suspensions, lipase synthesis was more sensitive to iron(III) than was proteinase (50% inhibition at 1.6 M and a maximum of 40% inhibition at 5.0 M, respectively). In the case of lipase, added pyoverdine was able to partially protect enzyme production from the effects of iron(III). Summary The influence of cis-diamminedichloroplatinum(II) (cis-DDP) on human blood monocyte functions in vitro was studied. With cis-DDP added to the cell suspensions in concentrations between 102 and 10–6 M just before assay of monocyte functional capacity no significant changes in chemotactic, chemokinetic, phagocytic, or fungicidal activities were observed. However, with preincubation at 1 M for 60 min cis-DDP selectively inhibited chemotaxis, but not chemokinesis or phagocytosis. This inhibition was more pronounced at 37° C than at 4° C, and was not reversed by washing of the cells. The implications of these findings for clinical oncology are discussed.  相似文献

Summary When grown in a chemically defined medium, Streptomyces clavuligerus excreted cephamycin C, in addition to other components, throughout most of the growth phase. Ferrous iron and oxygen are required for the biosynthesis of this antibiotic and the concentration of these cofactors was manipulated to maximize cephamycin C production. The iron content of the chemically defined medium was shown to be sub-optimal for antibiotic production and the addition of 130 g/ml ferrous iron almost doubled the cephamycin C levels to 200 g/ml. When dissolved oxygen was maintained at saturation levels, only 60–80 g/ml cephamycin C was produced, and the intermediate penicillin N accumulated to high levels (50 g/ml). This suggests that the high concentration of dissolved oxygen had a greater effect on the enzymes catalysing the conversion of penicillin N to cephamycin C, than on those involved in the earlier steps of the pathway leading to the formation of penicillin N.  相似文献

Autopsied liver tissue samples collected from 42 males and 31 females were analyzed for copper, manganese and zinc using atomic absorption spectrometry (AAS). With the exception of two liver samples for which the copper levels were determined to be 74.8 and 104.0 g/g (dry weight), hepatic copper concentrations were found to range from 1.7 to 32.4 g/g with a mean concentration of 14.2 g/g and standard deviation of 7.0 g/g. Manganese concentrations (with the exception of one sample having 12.9 g/g) ranged from 0.22 to 4.6 g/g with a mean of 2.26 ± 1.00 g/g. Hepatic zinc levels averaged 118.3 ± 44.4 g/g and ranged from 38.5 to 231.3 g/g. There were no apparent trends for the levels of any metals versus age nor were there any differences in average hepatic metal concentrations for males and females. © Rapid Science 1998.  相似文献

Callus was initiated from in vitro grown immature leaf and ex vitro grown mature leaf and rhizome explants of Agave sisalana Perr. ex. Engelm, on MS medium containing 2,4-D (9.05 M) and kinetin (4.6 M) or 2,4-D (9.05 M), kinetin (4.6 M) and CH (1000 mg l–1) or mod. MS (NH4NO3, 1500 mg l–1) containing 2,4-D (9.05 M) and kinetin (4.6 M). Light was essential for callus formation which, however, was different in three types of explants on three different media compositions. Increasing NH4 +had a negative impact while addition of CH had a positive impact on callus formation. Shoot regeneration from callus from CH-supplemented medium only was achieved for rhizome and immature leaf tissues. The highest rate of regeneration was obtained with BA (26.6 M) as the sole hormone. Shoot buds g–1 callus varied according to BA concentrations. Shoot proliferation rate increased on half-strength MS medium containing BA (8.9 M). Microshoots developed on MS medium containing BA (2.22 M) and GA3 (1.44 M) and finally rooted on MS medium containing IAA (11.42 M). Acclimatized rooted plantlets are growing satisfactorily in ex vitro. This is the first report on plant regeneration via organogenesis of A. sisalana.  相似文献

The effects of N-(2,4-dimethyl-5-(((trifluoromethyl)sulfonyl)amino)phenyl)acetamide (mefluidide) and 3,6-dichloro-o-anisic acid (dicamba) on in vitro growth and somatic embryogenesis ofDactylis glomerata L. (orchard grass) were studied using suspension cultures and explanted leaf bases. All experiments employed modified Schenk and Hildebrandt medium amended with concentrations of dicamba ranging from 15 to 120 M (SH-15 to SH-120) and of mefluidide ranging from 1 to 100 M. SH medium without either growth regulator was used for embryo germination. Embyro production in suspension cultures with SH-30 medium plus 3 g/L casein hydrolysate was significantly reduced by 1 M mefluidide. Only 15% of these embryos germinated and produced plants compared to 84% from controls. Growth, as measured by dry weight, was significantly reduced by 50 or 100 M mefluidide. The number of embryos formed on leaf sections was significantly reduced by 20 or 25 M mefluidide. Embryos that formed with 10 M or more mefluidide were callused on both SH-15 and SH-30 media. Shoot formation was inhibited from individual embryos and embryo/callus masses that developed on either SH-15 or SH-30 medium containing 5 M or more mefluidide. Radicle emergence was significantly reduced with 10 M mefluidide regardless of 15 or 30 M dicamba. Histological examination revealed that mefluidide inhibited both shoot and root meristem development with shoot development being the more sensitive. Inhibition of both was independent of dicamba concentrations. Shoot formation was also reduced from embryos that had developed on SH-30 medium without mefluidide when transferred to medium containing mefluidide without dicamba.  相似文献

The distribution of cysteine oxidase (CO) and cysteine sulfinate decarboxylase (CSD) was examined in 12 regions of the rat central nervous system (CNS). The distribution of CO activity, expressed as mol of cysteine sulfinate formed per h per g, was the following: hypothalamus, superior and inferior colliculi, 94–99 mol/h/g; olfactory bulbs, cerebral cortex, striatum, and hippocampus, 44–51 mol/h/g; cerebellum, 71 mol/h/g; pons-medula and spinal cord, 94 and 60 mol/h/g, respectively. The distribution of CSD activity expressed as mol of cysteine sulfinate decarboxylated per h per g was the following: hypothalamus and colliculi, 14–21 mol/h/g; olfactory bulbs, cerebral cortex, striatum, hippocampus, and cerebellum, 8–13 mol/h/g; pons-medulla, 7.3; and spinal cord, 3.6 mol/h/g. No CSD activity was detected in sciatic nerve. The subcellular distribution of CO and CSD activities was studied in hypothalamus, colliculi, and cerebral cortex. CO activity was localized in synaptosomes, mitochondria, and microsomes. CSD was primarily confined to the crude mitochondrial fraction and after subfraction, recovered mainly in the synaptosomal fraction.  相似文献

A method for rapid and highly effective plant micropropagation from vegetative meristems was established for Aloe barbadensis Mill. Plant micropropagation was achieved culturing apices on medium containing 1.1 M 2,4-dichlorophenoxyacetic acid and 2.3 M kinetin for 15–30 days. High morphogenetic ability was maintained by transferring explants (after 60 days) on media containing 0.11 M 2,4-dichlorophenoxyacetic acid and 2.2 M 6-benzylaminopurine.  相似文献

This report describes the newly identified Sarcocystis inghami n. sp. from the skeletal muscles of opossums (Mammalia: Didelphidae) that were collected from south central Michigan (42° 43-42° 79N, 84° 18-84°mathtype="display">6'W), USA. The new species is distinguished from all species described from North and South American opossums by the distinctive morphology of the villar protrusions on the cyst wall. Sarcocysts of S. inghami are microscopic, up to 700 m long and 110 m wide. The sarcocyst wall is up to 7 m thick, with long, stalked protrusions which average 5.5 × 1.2 m. These are constricted at the base, expanded laterally, rounded off distally and occasionally bifid. The villar protrusions have numerous microtubules without electron–dense bodies that extend from the tips into the granular layer. Bradyzoites are 10.7 × 4.3 (8––12 × 4––5) m. This is the second species of Sarcocystis sarcocyst described from the Virginia opossum in North America.  相似文献

In liquid synthetic medium inoculated with Sclerotium rolfsii (SR), addition of 6-methylpurine (MP, 50g/ml) immediately after inoculation led to approximately 100% reduction in sclerotia production. Adenosine, and to a lesser extent guanosine, each at final concentration of 100g/ml significantly reduced inhibition of sclerotia formation by SR in presence of 50g/ml MP. Uridine and cytidine each at 100g/ml had no such effect. The inhibition of sclerotia morphogenesis could be prevented by addition of 800g/ml of adenosine together with 50g/ml MP. Reversal by adenosine of MP-induced inhibition of sclerotia development was concentration dependent.  相似文献

The light utilization efficiency and relative photon requirement of photosynthesis in pulsed and continuous light from light emitting diodes (LEDs) has been measured. First, we chacterized the photon requirement of photosynthesis from light of LEDs that differ in spectral quality. A photon requirement of 10.3±0.4 was measured using light from a 658 nm peak wavelength (22 nm half band width) LED over the range of 0–50 mol photons m–2 s–1 in 2 kPa O2 in leaves of tomato (Lycopersicon esculentum Mill., cv. VF36). Because the conversion of electrical power to photons increased with wavelength, LED lamps with peak photon output of 668 nm were most efficient for converting electricity to photosynthetically fixed carbon. The effect of pulsed irradiation on photosynthesis was then measured. When all of the light to make the equivalent of 50 mol photons m–2 s–1 was provided during 1.5 s pulses of 5000 mol photons m–2 s–1 followed by 148.5 s dark periods, photosynthesis was the same as in continuous 50 mol photons m–2 s–1. When the pulse light and dark periods were lengthened to 200 s and 19.8 ms, respectively, photosynthesis was reduced, although the averaged photon flux density was unchanged. Under these conditions, the light pulses delivered 1017 photons m–2, which we calculate to be equivalent to the capacitance of PS I or PS II. Data support the theory that photons in pulses of 100 s or shorter are absorbed and stored in the reaction centers to be used in electron transport during the dark period. When light/dark pulses were lengthened to 2 ms light and 198 ms dark, net photosynthesis was reduced to half of that measured in continuous light. Primordial initiation and development of shoot-buds has been accomplished by using shoots derived from chestnut (Castanea sativa Mill) seedlings cultured with added 6-benzylaminopurine (BAP). Germination of chestnut seeds in the presence of BAP (4 – 40 M) stimulated varying numbers of shoot-buds in those areas of the main axis that were favorably altered. When excised single shoots from these treated seeds were subcultured on a fresh medium containing BAP (4 – 40 M) continual shoot production was observed. Bud growth and shoot elongation were stimulated by transferring cultures to a reduced concentration of BAP (2 M) plus indole-3-butyric acid (IBA 0.4 M). Plant regeneration occurred in the presence of IBA (0.8 M) after a preconditioning treatment in which naphthaleneacetic acid (NAA 50 M) and kinetin (k 2 M) were applied to the tissue culture shoots for 7 days in light.  相似文献
