Mass‐reared sterile tephritid flies released in sterile insect technique (SIT) programmes exhibit behaviour, physiology and longevity that often differ from their wild counterparts. In the present study, video recordings of flies in laboratory cages are used to determine whether the sequential processes of mass‐rearing and sterilization (using gamma radiation) that are integral to SIT affect general activity patterns of male and female Queensland fruit flies Bactrocera tryoni (Froggatt) (Diptera: Tephritidae) (‘Q‐flies'). Compared with wild flies, mass‐reared flies exhibit a marked reduction in overall activity, and further reduction is found after sterilization. In terms of the frequency of activities, both fertile and sterile mass‐reared Q‐flies fly less often and exhibit more bouts of inactivity and grooming than wild Q‐flies. In addition, in terms of the duration of activities, fertile and sterile mass‐reared Q‐flies spend less time flying and more time walking, grooming and being inactive than wild Q‐flies. Although fertile and sterile mass‐reared flies are similar in other regards, sterile mass‐reared flies spend more time being inactive than fertile mass‐reared flies. These findings raise new questions about how changes in behaviour and activity levels may influence the performance of mass‐reared sterile Q‐flies in the field, as well as the physiological and metabolic processes that are involved. The frequency and duration of inactivity could provide a simple but powerful and biologically relevant test for quality in mass‐rearing and SIT programs.  相似文献

Flight, walking and resting behaviours of male Aedes albopictus (Skuse) were observed in a flight cage. These behaviours were also associated with repetitive acts of grooming. The effects of sugar meal and period of the day on the total duration, the frequency and the duration of each individual grooming behaviour were studied. Fed or unfed males were individually released in a transparent plastic flight cage and direct visual observations using Observer software were carried out for 2?h during the morning and 2?h in the afternoon. There was no significant difference in the grooming behaviour between morning and afternoon observations. Fed male mosquitoes groomed longer than unfed males. The sugar meal seemed an important influencing factor of the grooming behaviour by giving more energy and making different parts of the mosquito sticky. The functions of grooming behaviour are discussed in regard to other behaviours such as mating, copulation, sensory detection and flight.  相似文献

Oögenesis in normal and small house flies is compared. In small female house flies the onset of vitellogenesis may be delayed by 2–4 days and more. Normal flies fed with sugar only may complete previtellogenic ovarian growth within the first 4–5 days after emergence and may start vitellogenesis immediately when protein is fed after this time. In contrast to this, small flies are not ready to start vitellogenesis when fed proteins after a sugar diet of 5 days. It is suggested that small flies need a protein food for the completion of the previtellogenic period of ovarian growth. Once vitellogenesis has started further ovarian development is normal in small flies.  相似文献

The effects of spinosad bait and various insecticides, the presence of sugar in insecticides, and diet on feeding responses and mortality in western cherry fruit fly, Rhagoletis indifferens Curran (Dipt., Tephritidae), were determined. Numbers of feeding events on insecticides with sugar were greater than on insecticides alone, but there was only a small effect of diet on feeding responses to insecticides with sugar. Feeding durations on imidacloprid, thiamethoxam and acetamiprid with sugar were shorter than on sugar water and spinosad bait, as the neonicotinoids paralysed flies quickly. Flies that fed on sugar only (nitrogen‐starved) suffered higher mortalities when exposed to spinosad, thiamethoxam and azinphos‐methyl than to imidacloprid, acetamiprid and indoxacarb, and mortality in between these two groups of treatments when exposed to spinosad bait. Mortalities were greater when sugar was added to insecticides, and were higher in nitrogen‐starved than fully‐fed (yeast extract + sugar fed) flies. Flies that fed once on thiamethoxam were killed more quickly than those that fed once on spinosad bait and spinosad. Results suggest that thiamethoxam is comparable to spinosad in its effects on mortality, and that using it with sugar in bait may also have similar results as using spinosad bait or spinosad. One benefit of using thiamethoxam with sugar may be that it kills flies more quickly, before they can oviposit, than spinosad bait, although whether a fly will feed on it may depend on how much sugar or nitrogenous food it has eaten.  相似文献

ABSTRACT. The effect of diet on the activity of male Calliphora vicina R.D. (Diptera, Calliphoridae) was studied. Protein–fed males (PF) were significantly more active than males fed only on sugar and water (NPF); as a consequence they ingested more sugar. The phenomenon was probably due to PF insects spending more time in flight than NPF, as no significant differences were found between the wingbeat frequencies and flight speeds of tethered flies of the two treatments. Protein ingestion also had a physiological effect, as PF flies had heavier thoraces than NPF flies with increased titres of protein.  相似文献

Feeding on yeast hydrolysate (a source of nitrogen) has a strong influence on the physiology and behaviour of the Queensland fruit fly (Q‐fly), Bactrocera tryoni (Froggatt) (Diptera: Tephritidae), affecting longevity, sexual maturation, oogenesis, and mating performance. In this study, we demonstrate that access to yeast hydrolysate also influences the development of attraction to cue‐lure in Q‐flies. We provided virgin Q‐flies various periods of access to yeast hydrolysate (continuous, 48 h, 24 h, or deprived). Attraction of males to cue‐lure was increased and occurred at an earlier age when they were fed yeast hydrolysate. Males given continuous access were strongly attracted to cue‐lure at a younger age (8 days after emergence), but by 12 days after emergence attraction of males given access to yeast hydrolysate for 48 h did not differ from males given continuous access. Attraction by males deprived or given just 24 h access to yeast hydrolysate was always significantly lower than those of males with continuous access. Male attraction to cue‐lure was highest in the early morning. While cue‐lure is most often thought of as a male attractant, virgin female Q‐flies were caught in cue‐lure traps at dusk at ages when they are known to be sexually mature. We suggest that cue‐lure or similar natural chemicals play a role in the Q‐fly mating system. γ‐Irradiation used to induce sterility had no significant effect on attraction to cue‐lure by Q‐flies.  相似文献

Queensland fruit fly, Bactrocera tryoni (“Q‐fly”), is Australia’s most economically important insect pest of horticultural and commercial crops especially in the eastern regions. The sterile insect technique (SIT) has been adopted as an environmentally benign and sustainable approach for management of Q‐fly outbreaks. High‐performance larval diets are required to produce the millions of flies needed each week for SIT. Yeast products contribute amino acids (protein) to fruit fly larval diets, as well as carbohydrate, fat and micronutrients, but there can be substantial variation in the nutritional composition and suitability of yeast products for use in larval diets. Gel larval diets have recently been developed for large‐scale rearing of Q‐fly for SIT, and composition of these diets requires optimization for both performance and cost, including choice of yeast products. We assessed performance of Q‐flies reared on gel larval diets that contained debittered brewer’s yeast (Lallemand LBI2240), hydrolysed yeast (Lallemand FNILS65), inactivated brewer’s yeast (Lallemand LBI2250) and inactivated torula yeast (Lallemand 2160‐50), including blends. Q‐flies performed poorly when reared on diets containing only or mostly hydrolysed yeast in terms of pupal number, pupal weight and percentage of fliers. Performance was also poor on diets containing high proportions of torula yeast. Overall, debittered brewer’s yeast is recommended as the best option for Q‐fly gel larval diet, as it is cheap, readily available, and produces flies with good performance in quality control assays. Inactivated brewer’s yeast produced flies of comparable quality with only a modest increase in cost and would also serve as an effective alternative.  相似文献

The aim of this research was to investigate whether or not sugar dusting can stimulate the grooming behaviour in Apis mellifera L. (Hymenoptera: Apidae), an important defensive mechanism against Varroa destructor Anderson & Trueman (Acari: Varroidae), and to assess the most effective dose and frequency of treatment. The criterion for evaluation of grooming potential was the percentage of damaged mites (PDM) among the total number collected on the bottom boards of the hives. In each sugar‐treated group PDM was significantly higher in comparison both with the negative control (no treatment) and with the values preceding the treatment. The results point to a stimulating effect of sugar on the grooming behaviour at all doses and frequencies tested. Treatment frequency influenced the stimulating effect of sugar: treatments at 3‐ and 7‐day intervals with 30 and 40 g resulted in significantly higher PDMs than the least frequent treatment (every 14 days); dusting with 20 g influenced PDM only when repeated at 3‐day intervals. Because treatments at 3‐day intervals are time‐consuming, those with 40 or 30 g repeated every 7 days may be recommended. In the positive control (hives treated with amitraz), average PDM was significantly lower than in the negative control and all sugar‐treated groups. Possible causes of the stimulating effect of sugar dusting on bee grooming behaviour are discussed.  相似文献

Post‐teneral diets containing yeast hydrolysate are reported to increase longevity, reproductive development and sexual performance of Queensland fruit fly (‘Q‐fly’) Bactrocera tryoni Froggatt (Diptera: Tephritidae). Consequently, diets including yeast hydrolysate are recommended for sterile Q‐flies before release in sterile insect technique (SIT) programmes. However, in some tephritids, diets including yeast hydrolysate are associated with an increased vulnerability to starvation. In the present study, the effects of yeast hydrolysate supplementation before release are considered with respect to the longevity of released Q‐fly when food becomes scarce. Experiments are carried out in three settings of varying resemblance to field conditions: 5‐L laboratory cages, 107‐L outdoor cages and 14 140‐L field cages containing potted citrus trees. In all experimental settings, compared with flies that received only sucrose, male and female Q‐flies that are provided with yeast hydrolysate during the first 2 days of adult life have a significantly shorter survival when subsequently deprived of food. Yeast supplementation appears to commit Q‐flies to a developmental trajectory that renders them more vulnerable to starvation. The practical significance of these findings for SIT depends on how often the releases are carried out under conditions in which Q‐flies experience extreme food shortages in the field.  相似文献

Wild strains of fruit flies (Diptera: Tephritidae) placed into laboratory rearing conditions are subjected to selection pressures caused by the diet, cages, density of flies, and other factors. Selection that changes mating behavior of the strain may result in less effective males released in sterile insect programs. Tests were performed to examine the effects of protein in diet and adult interactions on egg production and mating during sexual maturation of the Mexican fruit fly (Anastrepha ludens Loew) in laboratory cages. Flies were offspring of wild flies collected from Chiapas or Nuevo Leon, Mexico, and reared on Valencia oranges. Experiments demonstrated effects of yeast hydrolysate protein in adult diet and pairing with males on production of mature and immature eggs, numbers of females producing eggs, and mating with females aged 15 d. Addition of protein to 4% fructose in the adult diet approximately tripled mature egg production in females maintained for the total maturation period with an equal number of males. Females that matured without males produced approximately 33% more-mature eggs when fed protein than those fed no protein. Total egg production of females matured without males and fed sugar only or sugar with protein was more than twice that of females matured with males. Tests to examine the effects of male and female diet separately on female egg production showed slightly higher egg production in females fed protein, or females paired with males fed protein, but these differences were not significant. The most definitive effects were that combining wild strain females and males in cages during maturation reduced egg production. This effect was greatest when flies were not fed protein.  相似文献

Abstract. Adults of the dung fly Neomyia cornicina (Fabricius) were fed continuously on either dung containing no ivermectin (control dung) or dung containing 0.125 μg g-1or 0.25 μ g-1ivermectin (wet weight).Comparisons were made between the behaviour of flies during the first 24 h of dung feeding and that observed after 96 h of feeding.Subsequent experiments investigated the effects of ivermectin ingestion on three measures of locomotory ability: escape time, time to re-right, and capture time.
Analysis of behavioural data showed a significant reduction in the activity of ivermectin-fed flies compared to that of the controls.After 96 h of feeding on dung containing ivermectin, there was a significant increase in the duration of time spent standing and a reduction in duration and frequency of walking and grooming behaviours compared to controls.
Seventy-two hours after the onset of dung feeding, flies fed dung containing ivermectin took significantly longer to escape from a glass tube and to re-right themselves after overturning than flies fed control dung.The time taken to capture flies that had fed on dung containing ivermectin at 0.25 μg g-1was significantly shorter than that required to catch control flies when flies from the different treatment groups were presented blind and randomly.  相似文献

Abstract. The effect of sugar feeding on the survival of adult phorid fly Pseudacteon tricuspis is investigated. Flies fed 25% sucrose in aqueous solution continuously throughout their lifespan have greater longevity (mean ± SE longevity: female = 7.9 ± 0.8 days, male = 8.9 ± 0.9 days) than completely starved (provided no water and no sugar solution) flies, sugar-starved (provided water only) flies, or flies fed sugar solution only on their first day of adult life. Completely starved flies rarely lived beyond one day. Provision of water increases longevity by 2 days, and one full day of sugar feeding further increases longevity by an additional 1–2 days. Flies fed 50% sucrose have similar survivorship as those fed 25% sucrose. The temporal patterns of nutrient accumulation and utilization are also compared in P. tricuspis fed different diets: sugar-starved, sucrose-fed on the first day of adult life only, and sucrose-fed continuously. Adult P. tricuspis emerge with no gut sugars, and only minimal amounts of body sugars and glycogen. Although the levels of body sugars and glycogen decline gradually in sugar-starved flies, a single day of sugar feeding results in the accumulation of maximum amounts of gut sugars, body sugars and glycogen. High levels of these nutrients are maintained in female and male phorid flies fed on sucrose continuously over the observation period, whereas nutrient levels decline in flies fed only on the first day of life, beginning 1 day postfeeding. Female and male P. tricuspis emerge with an estimated 12.3 ± 2.3 and 7.2 ± 1 g of lipid reserves per fly, respectively. These teneral amounts represent the highest lipid levels detected in adult flies, irrespective of their diet, and are maintained over the life times of sucrose-fed female and male flies, but declined steadily in sugar-starved females. These data suggest that adult P. tricuspis are capable of converting dietary sucrose to body sugars and glycogen, but not lipids.  相似文献

Behavioural experimentation on Phormia regina shows that females previously fed only sucrose, and thus in a state of yeast preference, will ingest single-salt-sucrose mixtures in volumes similar to that of 10% yeast (Rachman et al., 1982). In particular, females will ingest NaCl 0.1 M in 0.2 M sucrose in volumes not statistically different from 10% yeast. In the experiments reported here, groups of females were given ad libitum access to both 0.2 M sucrose and NaCl 0.1 M in 0.2 M sucrose from days 0–4 after eclosion. Potassium and sodium salts were added to 0.2 M sucrose over a spectrum of concentrations and presented to the flies on day 6. The flies rejected all single-salt-sucrose mixtures offered. Even when the pre-test sodium salt was changed to a potassium salt during days 0–4, the flies still rejected the single-salt-sucrose mixtures offered subsequently. However, when sodium and potassium salts were mixed together in 0.2 M sucrose, females ingested the solutions in amounts similar to 10% yeast. Several different concentrations of the double-salt-sucrose solutions were ingested in volumes not statistically different from 10% yeast. Results support the underlying hypothesis that activity in the salt-sensitive chemosensory neurones is involved in protein preference.  相似文献

Abstract.  Mediterranean fruit flies ( Ceratitis capitata Wiedemann, Diptera: Tephritidae) harbor a community of diazotrophic bacteria in their digestive system. The present study aims to test the hypothesis that bacteria contribute to fly fitness by enhancing copulatory success and egg production in males and females, respectively. After eclosion, flies were fed a full diet containing peptides, sugar and minerals, or a sugar diet, lacking peptides. Subgroups from each diet were fed a mixture of the antibiotics ciprofloxacin and piperacillin. The presence of bacteria, food consumption, weight gain, lipid and protein levels, oviposition in females and copulatory success of males were quantified in the four groups. The antibiotic treatment effectively cleared the gut of bacteria. The relative amounts of food consumed (with or without antibiotics) are similar in all groups. The antibiotics do not inhibit feeding, and their ingestion does not affect dry weight or the amount of protein stored, yet females feeding on the full diet without antibiotics have increased lipid levels. Females fed the full diet produce significantly more eggs than females on the sugar diet, but the presence of bacteria does not affect numbers of eggs produced. However, in the absence of bacteria, the oviposition rate of nutritionally stressed females is significantly accelerated. The presence of bacteria in sugar fed males does not provide them with a mating advantage. Conversely, in males fed a full diet, the presence of bacteria is associated significantly with a shorter latency to mate. It is concluded that, because the bacterial community is present at all stages of the fly's life cycle, at least some species are effectively transmitted from parents to offspring, and removal of bacteria affects measurable physiological and behavioural parameters related to fitness, the association between bacteria and the medfly is mutualistic.  相似文献

One possible control strategy against the olive fly, Bactrocera oleae, the most serious olive crop pest, is the Sterile Insect Technique (SIT) application. However, a number of problems associated with this method remain that decrease the effectiveness of SIT, including the quality of reared insects. Taking the importance of the relationship between the olive fly and bacteria into consideration, the effects of probiotic diets enriched with Pseudomonas putida on B. oleae longevity and fecundity were evaluated. First, we found that the probiotic bacterium, P. putida, is conveyed from diets to the oesophageal bulb as well as to the fly midgut after feeding on the probiotic diet. Subsequently, B. oleae adults fed on either: (a) a standard full protein and sugar diet; (b) a sugar only diet; (c) a probiotic standard full protein and sugar diet; or (d) a probiotic sugar diet. Flies fed on probiotic diets were supplied with an inoculated gel containing P. putida; non‐inoculated gel was provided to the flies fed on non‐probiotic diets. B. oleae males and females that fed on sugar diets did not survive as long as those that fed on protein diets. A comparison of the longevity of adults fed on full diet and sugar with their respective probiotic diets revealed no significant difference. Males fed on the sugar only diet survived longer than males fed on probiotic sugar diet, and females fed on the full protein and sugar diet survived much longer than females fed on the full probiotic diet. As regarding fecundity, both full diets resulted in a higher number of eggs laid per female. Females fed on the probiotic sugar diet laid a higher number of eggs than females that fed on sugar only. The inoculated gel of the probiotic sugar diet contained a significantly higher quantity of leucine, isoleucine and proline than the non‐inoculated gel of the sugar only diet. The possible role of dietary bacteria in relation to functional aspects of olive fly physiology is discussed.  相似文献

Liquid larval diets have been developed for several tephritid fruit flies including Queensland fruit fly, Bactrocera tryoni (Frogatt) (Q‐fly). In liquid diets, wheat germ oil (WGO) is usually added to improve performance in some quality parameters of reared flies, especially flight ability. However, for some flies, other plant oils may be more readily available, cheaper or produce flies of superior performance. In the present study, four alternative types of plant oils – rice bran, canola, vegetable, and sesame – were incorporated into a fruit fly liquid larval diet to replace the currently used wheat germ oil and their efficacy on the quality parameters of reared Q‐fly was compared to diets containing wheat germ oil or no oil. The quality parameters included: total pupal yield (N), pupal recovery (%), larval duration (days), pupal weight (mg), adult emergence (%), adult fliers (%), rate of fliers (%), sex ratio (%), F1 egg/female/day and egg hatching (%). There were significant differences among treatments in performance of Q‐fly. Vegetable oil appeared better in terms of total pupal yield, percentage of pupal recovery, percentage of adult emergence, percentage of fliers, mean egg/female/day and % F1 egg hatch compared with other oil treatments, especially from that of WGO treated diet. The result suggests that WGO can be substituted with rice bran and vegetable oil to improve the liquid larval diet for rearing of B. tryoni, with vegetable oil being the best replacement.  相似文献

Survival and egg-laying trends were investigated in Mediterranean fruit flies (Ceratitis capitata) adults maintained on a sucrose-only diet, or on a full diet that consisted of a 3:1 sucrose and yeast hydrolizate mixture. In addition, we followed the total individual lipid and protein contents of aging flies in a cohort. Survival trends and life expectancy parameters at eclosion for males and females on full diet and for males on sucrose only were very similar. In contrast, the mortality of females on sucrose only was high early in life, but then slowed down. Egg-laying was ten times greater in female flies on full diet than in flies on sucrose only. Lipid contents in males and females on both types of diets were very similar, and harmonically oscillated with an amplitude of approximately 10 days. Successive crests of lipids tended to be smaller with the ageing of the cohort, and lipids contents significantly dropped at very advanced ages and close to the maximal age of the whole cohort. Protein contents of flies maintained on a full diet were high and stayed at a constant level throughout the entire life of the cohort. Protein levels in males and females on sucrose only dropped drastically during the first days of adult life, but then stayed stable at a low level until advanced ages. We propose that the synchronous rhythmic oscillation in lipid contents of male and female flies seems to be independently set by an internal clock. Protein reserves are allocated according to the access to protein food sources and these levels of protein are closely associated to egg production and mortality. Our results are discussed in view of resource allocation during reproduction and senescence.  相似文献

Pupae of the Queensland fruit fly or Q‐fly Bactrocera tryoni (Froggatt) are irradiated routinely to induce reproductive sterility in adults for use in sterile insect technique programmes. Previous studies suggest that adult sexual performance and survival under nutritional and crowding stress are compromised by the current target dose of radiation for sterilization (70–75 Gy), and that improved mating propensity and survival under stress by irradiated males may be achieved by reducing the target sterilization dose without reducing the level of induced sterility. This raises the question of the amount by which the irradiation dose can be reduced before residual fertility becomes unacceptable. The present study measures the levels of residual fertility in male and female irradiated Q‐flies at different irradiation doses (20, 30, 40, 50, 60 and 70 Gy), and investigates the possibility that fecundity and fertility increase between 10–15 and 30–35 days post emergence. Male flies require a higher dose than females to induce sterility, with no residual fertility found in females irradiated at doses of 50 Gy or above, and no residual fertility found in males irradiated at doses of 60 Gy or above. Irradiated females are more fecund at 30–35 days post emergence than at 10–15 days. However, fertility does not increase between 10 and 15 days post emergence and 30–35 days, even at doses below 50 Gy. The present study shows that there is scope to reduce the target sterilization dose for Q‐flies below that of the current dose range (70–75 Gy) at the same time as retaining an adequate safety margin above radiation doses at which residual fertility can be expected.  相似文献

Recent laboratory studies of mass‐reared flies in small cages have found that periods of just 24‐ or 48‐h access to yeast hydrolysate can substantially enhance mating performance of mass‐reared male Queensland fruit flies, Bactrocera tryoni (Froggatt) (Diptera: Tephritidae) (‘Q‐flies’). Using field cage tests that provide a better approximation of nature, we here investigated whether access to yeast hydrolysate for 48 h after adult emergence improves the ability of male and female mass‐reared, sterile Q‐flies to compete sexually with wild‐type flies that had been provided continuous access to yeast hydrolysate. Mating probability of sterile males was significantly increased by 48‐h access to yeast hydrolysate; sterile males provided 48‐h access to yeast hydrolysate had mating probability similar to that of wild males provided continuous access to yeast hydrolysate, whereas sterile males deprived of access to yeast hydrolysate had much lower mating probability. Unlike males, access to yeast hydrolysate for 48 h did not increase mating probability of sterile female Q‐flies. We instead found that wild females provided continuous access to yeast hydrolysate had higher mating probability than sterile females that did or did not have 48‐h access to yeast hydrolysate. This result raises the possibility that a bisexual Q‐fly strain might operate essentially as a male‐only release when the flies are given access to yeast hydrolysate during a 48‐h pre‐release holding period. Sterile males given access to yeast hydrolysate for 48 h mated significantly earlier in the evening than wild males and, as in other recent studies, this tendency was associated with an increased tendency to mate on the trees rather than the cage walls. There was no evidence of sexual isolation in this study, as wild and sterile mass‐reared flies showed no evidence of preferential mating with their own kind. Further studies are now needed to assess the potential for pre‐release access to yeast hydrolysate to improve sexual performance and longevity of sterile, mass‐reared, Q‐flies in the field.  相似文献

Raspberry ketone (RK) dietary supplements accelerate the emergence of sexual behaviour in developing Queensland fruit fly (Q‐fly) males and show promise as a pre‐release supplement for use in sterile insect technique (SIT) programs. However, the value of RK supplements in SIT programs would be greatly reduced if RK‐treated males are ineffective at inducing sexual inhibition in mated females. To test the effectiveness of matings by RK‐treated males, we here investigate the remating propensity of females mated by RK‐treated (1.25% or 5% RK in food) and RK‐untreated (control) males. Tested males received RK supplements mixed in sugar and yeast hydrolysate for 2 days after emerging and then received only sugar. To test for male age‐dependent effects, virgin females were mated to treated and untreated males that were 6, 8, 10, 20 or 30 days old. To test for persistence of sexual inhibition, mated females were tested for remating propensity at 1, 7 or 15 days after their first mating. RK‐treated males did not differ from control males in copula duration, and females mated by RK‐treated males did not differ from those mated by control males in remating propensity, second copula latency or second copula duration. RK‐treated Q‐fly males not only mate at younger ages but also their matings are as effective as those of untreated controls at inducing sexual inhibition in mates.  相似文献
