The binding in vitro of tritiated aldosterone to domestic duck (Anas platyrhynchos) kidney tissue has been investigated. Using tissue from animals on a normal diet, tritiated aldosterone was specifically bound to kidney cytosol with an apparent equilibrium dissociation constant of about 9 nM and number of binding sites in the 20 fmol/mg protein range. These values did not show statistically significant changes when the cytosol originated from animals with salt activated nasal glands. Kidney cytosols labeled with tritiated aldosterone sedimented with a single peak at 8S in a linear sucrose gradient (10–30%) and this peak was quenched by excess, radioinert aldosterone. Following incubation of labeled cytosols with crude nuclei, the cytosols became depleted of the label and aldosterone was translocated to the Tris-soluble and Tris-insoluble, 0,4 M KCl soluble nuclear fractions. Kidney cytosols metabolized aldosterone extensively to a compound presumed to be 3α,5β-tetrahydroaldosterone. However, only unchanged aldosterone became receptor-bound. It was concluded that the duck kidney possesses aldosterone receptors, though competition studies indicated that the specificity of these receptors might be different from those described in the mammalian kidney.  相似文献

An in vitro system has been designed to study the interaction between intestinal cytoplasmic 1, 25-dihydroxyvitamin D receptors and highly purified homologous nuclei. Cytosolic receptors, labeled with 1, 25-dihydroxyvitamin D, specifically associate with intestinal nuclei as ternary chromatin complexes, an event which proceeds in the absence of a demonstrable change in sedimentation coefficient. This interaction is a saturable process with a dissociation constant of 2.2 × 10?10 M at 0° C, and is specific for the 3.3S cytoplasmic receptor since neither the 4.1S nor the 6S vitamin D binding proteins associate in a similar fashion. In addition, the interaction is hormone-dependent, since free receptors also do not bind. The development and characterization of an in vitro system such as this is the beginning in an attempt to assess the molecular action of these receptors on intestinal nuclei.  相似文献

Constant levels of cellular unsaturated fatty acids were obtained by growing a fatty acid desaturase mutant of Saccharomyces cerevisiae in glucose limited chemostat cultures supplemented with various concentrations of Tween 80. An increase in the frequency of cytoplasmic respiratory incompetent mutants was observed in cultures growing at low cellular levels of unsaturated fatty acids. This effect has been shown to result from an increase in the rate of mutation as the cellular unsaturated fatty acid level is decreased. The majority of induced petite mutants are ?° (contain no mitochondrial DNA).  相似文献

R5020(17,21-dimethyl-19-nor-4,9-pregnadiene-3,20-dlone)-binding components with sedimentation coefficient of 8S were detected in sexual skin cytosols from estrogen-primed ovarlectomized Japanese monkey (Macaca fuscata fuscata). In contrast, little 8S binding was found In similar preparations from the abdominal skin. The dissociation constants and the number of binding sites of the components were l.6×10?10M and 36 fmoles/mg cytosol protein, respectively. The 8S binding components were specific for progestational compounds. Incubation with pronase abolished the 8S binding. Thermal experiments revealed the thermolabile nature of the components. Moreover, the concentration of the R5020-binding components was markedly increased by estradiol-17β 3-benzoate injections. We conclude from these results that the cytosols from the sexual skin of estrogen-primed female monkeys contain progesterone receptors.  相似文献

The control of cortisol secretion by ACTH and of aldosterone secretion by angiotensin is exerted upon separate cell populations in the adrenal cortex. Cells of the zona faciculata and the zona glomerulosa, while sharing common steroidogenic pathways, are affected differently by hormones and drugs. Fasciculata cells demonstrate increased cAMP formation and cortisol output primarily in response to ACTH. ACTH receptors, when occupied by hormone, transmit an activating signal to membrane-bound adenylate cyclase by a mechanism that may require the translocation of Ca2+. Although the precise way in which increased intracellular cAMP leads to increased steroidogenesis is unknown, protein phosphorylation and new protein synthesis are probably involved. Glomerulosa cells also respond to ACTH, but are uniquely responsive to physiological concentrations of angiotensin II and K+. The responsiveness of these cells to angiotensin may be governed by alterations in receptor number. Whether occupied angiotensin receptors activate steroidogenesis via cAMP is uncertain, but alterations in Ca2+ distribution within the cell may again be involved. Dopamine probably exerts a tonic inhibitory effect on glomerulosa cell function. Competitive inhibitory analogs for both ACTH and angiotensin II are available, but thus far all inhibitors have retained weak agonist properties. Because the regulatory processes for both cortisol and aldosterone are complex, a wide variety of drugs can affect rates of steroidogenesis invivo.  相似文献

The membrane Ca2+Mg2+ ATPase of density (age) separated human erythrocytes was examined for its stimulation by the cytosols of these cell groups. On the assumption that the stimulatory activity in the cytosol is only calmodulin, it was consistently observed that the young cytosol had a significantly higher activity towards the membrane Ca2+Mg2+ ATPase activity (from any age group) than did the old cell cytosol. The data clearly demonstrates decided differences in the expression of calmodulin activity in cytosols from young and old erythrocytes and would support the conclusion that calmodulin activity is altered during in vivo aging of these cells. Possible mechanisms for these alterations are discussed.  相似文献

While 2-aminoanthracene and 2-aminofluorene are converted to frameshift mutagens by microsomal preparations from rat livers, the microsomes from the intestinal mucosa of the same animals, under the experimental conditions used herein, either have little such activity or lack it altogether. Cell-free extracts of the colon anaerobe Bacteroidesfragilis may exhibit such activity to varying degrees depending upon the conditions of incubation. However mixtures consisting of cell-free extracts from B. fragilis and microsomes from intestinal mucosa demonstrate significant- more than additive- activity in converting these chemicals to mutagens.  相似文献

DNA-free minicells of Escherichia coli will not allow growth of phage T-7, nor is RNA synthesis stimulated by phage infection. Thus, these miniature cells seem not to contain in vivo RNA polymerase activity. However, DNA-dependent RNA polymerase activity can be unmasked in extracts with poly(dA-T) and Mn2+. A low protein diet (5% as compared to a control 21% protein diet, ad libitum) caused a significant decrease in the concentration of liver cytoplasmic glucocorticoid receptors; the equilibrium dissociation constant (Kd) did not change. The maximum decrease occurred in two weeks and was reversible upon substitution of the low protein by a control diet. This influence of protein deficiency could not be attributed to elevated plasma corticosterone levels since a comparable increase in plasma corticosterone of calorie-deficient rats (21% protein diet in restricted quantity) did not decrease glucocorticoid receptors and the difference in receptor levels of control and protein deficient animals persisted following adrenalectomy. These results suggest that glucocorticoids might not exert their usual biologic effects in the presence of protein malnutrition.  相似文献

Mouse L cell interferon (IFN αβ) inhibited the differentiation of mouse 3T3-Li fibroblasts into adipocytes. IFN αβ also inhibited hexose monophosphate shunt (HMP) activity in these cells. HMP activity is required for the reducing power necessary for the conversion of 3T3-Li fibroblasts to adipocytes. Both IFN blockage of differentiation and HMP activity were reversed by the reducing agent 2-mercaptoethanol, probably through interaction with membrane receptors and not through direct inactivation of IFN. Several non-antiviral effects of IFN αβ on cellular function, including differentiation and immunoregulation, may be mediated at the biochemical level through blockage of HMP activity.  相似文献

The electron spin relaxation of iron-sulphur centres and ubisemiquinones of plant mitochondria was studied by microwave power saturation of the respective EPR signals. In the microwave power saturation technique, the experimental saturation data were fitted by a least-squares procedure to a saturation function which is characterized by the power for half-saturation (P12) and the inhomogeneity parameter (b). Since the theoretical saturation curves were based on a one-electron spin system, it became possible to differentiate between EPR signals of iron-sulphur centres which have similar g values but different P12 values. If the difference in the P12 values of the overlapped components was small, no significant deviation from these theoretical saturation curves was observed, as shown for the overlapped signals of centre S-3 and the Ruzicka centre of mung bean mitochondria. By contrast, the microwave power saturation data for the g = 1.93 signal (17–26 K) of Arum maculatum submitochondrial particles reduced by succinate could not be fitted using one-electron saturation curves. Reduction by NADH resulted in a stronger deviation. Since the iron-sulphur centres of Complex I were present only in an unusually low concentration in A. maculatum mitochondria, it was proposed that an iron-sulphur centre of the external NADH dehydrogenase contributes to the spectrum of centre S-1. For mung bean mitochondria, the g = 1.93 signal below 20 K could be attributed mainly to centre N-2. The microwave power saturation technique was also suitable for detecting magnetic interactions between paramagnetic centres. From the saturation data of the complex spectrum attributable to centre S-3 and an interacting ubisemiquinone pair in mung bean mitochondria (oxidized state) followed that centre S-3 has a faster electron spin relaxation than the ubisemiquinone molecules. It is noteworthy that the differences in the relaxation rates were maintained despite the interaction between centre S-3 and the ubisemiquinones. Furthermore, a relaxation enhancement was observed for centre S-1 of A. maculatum submitochondrial particles upon reduction of centre S-2 by dithionite. This indicated a magnetic interaction between centres S-1 and S-2.  相似文献

The relationship between nuclear DNA and cytoplasmic membrane-associated DNA, extracted from a human lymphocyte cell line, was examined by DNA-DNA reannealing and by dissociation of renatured molecules. Up to 2% of the total cellular DNA is found in the cytoplasm as cytoplasmic membrane-associated DNA and of this 2%, approximately 70% is comprised of repeated sequences. These sequences are homologous to only about 4% of the repeated sequences of nuclear DNA. The repeat fraction of cytoplasmic membrane-associated DNA consists of sequences which are only moderately repeated. The number of copies in the average “family” could range from about 1500 copies to as few as 25 copies. A small rapidly reannealing portion of cytoplasmic membrane-associated DNA (C0t < 4 × 10?3) appears to consist of sequences derived from a single “family”.About 30% of cytoplasmic membrane-associated DNA reassociates slowly with a C0t12 value of 223 (unique cytoplasmic membrane-associated DNA). This fraction has homology with about 11% of the unique sequences of nuclear DNA. However, unique cytoplasmic membrane-associated DNA comprises only about 0·6% of the total cellular DNA. If it is assumed that each cell has the same amount of cytoplasmic membrane-associated DNA, homology with 11% of the unique sequences of nuclear DNA suggests that different cells may have different unique nucleotide sequences in the cytoplasm.  相似文献

Sucrose gradient studies of rat pineal cytosol incubated with 3H-estradiol (female pineals) or 3H-5 α -dihydrotestosterone (male pineals) revealed a radioactivity peak in the 8 S region which disappeared after superior cervical ganglionectomy or incubation with excess unlabeled hormone. Ganglionectomy decreased significantly estradiol and testosterone uptake by the pineal gland in vitro as well as high affinity binding to pineal cytoplasmic and nuclear components. Norepinephrine treatment counteracted all the effects of ganglionectomy but was unable to modify hormone uptake and binding by the pineal gland of sham-operated controls. Pre-treatment with actinomycin D or propranolol but not with phentolamine impaired norepinephrine effects; propranolol blockage however was only partial. Administration of isoproterenol, L-dopa or phentolamine increased hormone uptake by denervated pineals. The effects of isoproterenol were also observed in vitro and were blocked by propranolol. These results indicate that sex steroid receptors in the pinealocytes are controlled by norepinephrine via beta-adrenergic receptors and that depletion of neural norepinephrine enhanced responsiveness of pineal hormone receptors to exogenous catecholamines.  相似文献

Dolichos biflorus agglutinin (DBA), which is specific for terminal α-N-acetylgalactosamine, bound to a spontaneous leukemia cell of GRSA mice, but not to lymphoid cells of the host. The DBA receptors were isolated from the leukemia cell labeled with [3H]-galactose after detergent solubilization and affinity chromatography on DBA-agarose. The major component of the receptors migrated as a glycoprotein of apparent molecular weight 100,000 upon SDS gel electrophoresis. Alkaline treatment degraded the glycoproteins, releasing oligosaccharides of molecular weight around 1,000.  相似文献

O6-Methyl[8-3H]deoxyguanosine in a synthetic DNA polymer, poly(dC, dG, m6dG), is demethylated by cell-free extracts of EscherichiacoliBr adapted by exposure to N-methyl-N′-nitro-N-nitrosoguanidine, as shown by the appearance of 3H-labeled deoxyguanosine in hydrolysates of the recovered DNA. The demethylating activity could not be detected in extracts of nonadapted E. coli. These results provide direct evidence that a previously described inducible repair activity in E. coli acts by demethylating O6-methylguanine at the DNA level.  相似文献

When bakers' yeast cells which had been grown anaerobically in galactose were aerated in the presence of 10% glucose, they showed a 40% decrease in invivo [14C]-leucine incorporation into a washed mitochondrial membrane fraction compared with cells which had been aerated in a low glucose medium. The observed catabolite repression of membrane protein synthesis was primarily due to a decrease in cytoplasmic translational activity, but this repression was entirely dependent upon concomitant mitochondrial translation. The inductions of reduced coenzyme Q cytochrome c reductase (complex III) and of cytochrome c oxidase (complex IV) activities were repressed 30 and 60%, respectively, by aeration of the cells for 8 hours in 10% glucose. The catabolite repression of the formation of these two inner membrane complexes was again shown to be dependent upon concomitant mitochondrial translation. Both the amino acid incorporation and enzyme induction data suggest that catabolite repression of both cytoplasmically and mitochondrially translated mitochondrial membrane proteins is mediated through a mitochondrially translated repressor.  相似文献

The changes in the energy substrate utilized by the remnant liver were studied in relation to the changes in the cellular energy status of 25 and 70% hepatectomized rabbits. In 25% hepatectomized rabbits, the energy charge ((ATP+0.5ADP)(ATP+ADP+AMP)) level of the remnant liver remained unchanged, the energy substrate of which was predominantly glucose, rather than fatty acid. In contrast, in 70% hepatectomized rabbits, the energy production by the mitochondria was mainly dependent upon fatty acid oxidation at the early period after hepatectomy when the energy charge level decreased remarkably, and then upon glucose oxidation, concomitant with the restoration of the energy charge. It is suggested that the changes in the energy substrate utilized are closely related to those in the energy charge level and the mitochondrial phosphorylative activity of the remnant liver following hepatectomy.  相似文献

Steroid hormones induce responses in target tissues by a mechanism involving the specific initial interaction of hormone with cytoplasmic receptor molecules. These receptors, usually localized in target tissues have high binding affinities and limited binding specificities for biologically active steroids. Examination of human leukemic lymphoblasts has revealed these receptors in some tumor samples. Their presence is well correlated with hormone responsiveness of the tumor in vitro. Similar studies on human breast cancer tumor homogenates has indicated that about 23 of primary tumors contain estrogen receptor. The absence of receptor predicts a lack of response to hormone therapy almost invariably, while the presence of receptor increases but does not assure that the tumor will be hormone responsive. Recently in vitro tissue culture systems which mimic the hormone responses observed in vivo have been developed which should significantly aid in the clarification of the mechanisms whereby steroid hormones stimulate and inhibit growth in target tissues.  相似文献

A compound was isolated from female Phthorimaea operculella (Lepidoptera: Gelechiidae) abdominal tip extracts and identified as trans-4, cis-7-tridecadienyl acetate. The synthetic compound elicits male P. operculella wing fanning and upwind orientation in the laboratory, and is attractive to males in the field. The corresponding alcohol also appeared to be present in the female extracts, but this compound was not found to increase male responses in the laboratory or the field.
