Summary The horizontal distribution of three species of tubificid worm (Tubifex tubifex, Limnodrilus hoffmeisteri and L. udekemianus in the sediment of a small stream was examined. Worm distribution was most strongly correlated with the distribution of leaf litter. This relationship was examined in the laboratory by means of choice chambers. It was concluded that; 1) the addition, to an inorganic substratum, of conditioned leaf material, enhanced its attractiveness to the worms; 2) a combination of silt-clay and leaf material was preferred by the worms to mixtures of leaf material and coarser inorganic substrata 3) some leaf species were more attractive to the worms than others; 4) there were differences between the worm species in their preferences for the various leaf species; 5) none of the three tubificid species was exclusively associated with its preferred leaf species; 6) worms would switch to less attractive leaf species if preferred alternatives were unavailable; 7) leaf material only attracted the worms once it had become conditioned; 8) this appeal was lost when the leaves were autoclaved; and 9) substratum choice was independent of worm size.We believe the worms were attracted to the leaves because the associated microfloras provided a concentration of bacterial food. Differences between the three tubificid species in their preferences for the various leaf species probably reduced trophic competition.  相似文献

Adult Schistosoma mansoni worms were transplanted from 8 nonhuman primates with chronic infections into 8 naive recipients, in an effort to test the hypothesis that worm fecundity reduction in chronic infections is the result of host immunity or some other host effect. Techniques for perfusing living donors without the added use of anti-schistosomal drugs and for reducing the likelihood of post-operative bacterial endotoxemia and septic shock are described. Fecundity values in terms of eggs per day per female worm were obtained for the worms in their original and in their new hosts and compared. In 3 experiments, perfusions were incomplete and the donors were saved, enabling direct comparisons of fecundity to be made in subpopulations of worms in both their original and new hosts, after equal life spans. In only 1 of the 8 transplantations was there a clear increase in fecundity after surgical introduction into a naive host. Therefore, these experiments fail to support the hypothesis that reduced fecundity of S. mansoni worms in permissive nonhuman primate hosts is a reversible result of host immunity or some other host-derived factor. Despite this negation, further evidence for reduced worm fecundity in older infections was obtained. In the absence of in vivo evidence for immune-mediated antifecundity, worm senescence is the most likely explanation for this finding, with irreversible immune damage to the worms being a less attractive alternative hypothesis.  相似文献

A recent study suggests that postdauer Caenorhabditis elegans hermaphrodites produce more self‐sperm and have larger brood sizes than worms that bypass diapause. Why might natural selection favor increased self‐sperm production in postdauer hermaphrodites? This question is addressed by developing an age‐structured model for an exponentially growing worm population descending from a founder postdauer hermaphrodite. It is assumed that natural selection favors those founders that have the largest number of living descendants at some fixed future time. Increased self‐sperm production in postdauer hermaphrodites can then evolve when the diapause‐bypassing descendants suffer a higher mortality rate than their parental postdauer founders.  相似文献

To validate the protective efficacy against schistosomiasis by immunization with cells from juvenile Schistosoma japonicum in a murine model and to analyze possible factors related to protection, in this study, two independent repeated vaccination trials were performed. After three subcutaneous vaccinations, in trial one, in the absence of adjuvant, primary juvenile worm cells (pJCs) from S. japonicum induced remarkable average reductions in worm burden (54.3%), liver eggs per gram (LEPG) load (59.8%) as well as egg granulomas size (66.5%) compared to PBS control group (P<0.01), which were significantly higher than those elicited by fractions of juvenile worm cells (JCFs) or fractions of juvenile worms (JWFs) (P<0.05). Non-cell components of worms (WNCs) showed no significant protection. In trial two, compared to PBS control group, significant protective effect was also observed for cultured juvenile worm cells (cJCs) from S. japonicum with 58.4% worm reduction and 68.1% LEPG reduction (P<0.01). However, cultured adult worms cells (cACs) showed significantly higher worm burden (P<0.05) and egg burden (P<0.01) when compared to cJCs. Immunological analysis of trial two revealed that cJCs engendered a Th1-biased mixed Th1/Th2 type of immune response while cACs elicited a Th2-type response. Our data indicated that immunization with both primary and cultured cells from S. japonicum juvenile worms provided high immunoprotection, for which the physical character of immunogens, stage-specific parasite and the type of immune response induced might be responsible, suggesting that vaccination with whole cells from S. japonicum larvae is a promising approach to produce protective immunity against schistosomiasis.  相似文献

Background

Breeding livestock for improved resistance to disease is an increasingly important selection goal. However, the risk of pathogens adapting to livestock bred for improved disease resistance is difficult to quantify. Here, we explore the possibility of gastrointestinal worms adapting to sheep bred for low faecal worm egg count using computer simulation. Our model assumes sheep and worm genotypes interact at a single locus, such that the effect of an A allele in sheep is dependent on worm genotype, and the B allele in worms is favourable for parasitizing the A allele sheep but may increase mortality on pasture. We describe the requirements for adaptation and test if worm adaptation (1) is slowed by non-genetic features of worm infections and (2) can occur with little observable change in faecal worm egg count.

Results

Adaptation in worms was found to be primarily influenced by overall worm fitness, viz. the balance between the advantage of the B allele during the parasitic stage in sheep and its disadvantage on pasture. Genetic variation at the interacting locus in worms could be from de novo or segregating mutations, but de novo mutations are rare and segregating mutations are likely constrained to have (near) neutral effects on worm fitness. Most other aspects of the worm infection we modelled did not affect the outcomes. However, the host-controlled mechanism to reduce faecal worm egg count by lowering worm fecundity reduced the selection pressure on worms to adapt compared to other mechanisms, such as increasing worm mortality. Temporal changes in worm egg count were unreliable for detecting adaptation, despite the steady environment assumed in the simulations.

Conclusions

Adaptation of worms to sheep selected for low faecal worm egg count requires an allele segregating in worms that is favourable in animals with improved resistance but less favourable in other animals. Obtaining alleles with this specific property seems unlikely. With support from experimental data, we conclude that selection for low faecal worm egg count should be stable over a short time frame (e.g. 20 years). We are further exploring model outcomes with multiple loci and comparing outcomes to other control strategies.  相似文献

The purple globe crab, Randallia ornata (Randall, 1839) (Decapoda: Leucosiidae) is a common crustacean found on sandy beaches from Northern California to Baja California, Mexico. An undescribed species of Carcinonemertes, which shares biological features with a partially described worm from Tasmania was recovered from R. ornata. The worm reaches 10 mm in length and secretes a sturdy tube with a distinctive spiral shape. The total prevalence of the worm on R. ornatawas 70% (85% for ovigerous female crabs, 67% for post-ovigerous female crabs, and 45% for male crabs). Intensities reached 32 worms per crab. Adult worms were found in crab egg masses, regressed adults were recovered from under the abdomen of post-ovigerous females, and encysted juveniles were found on the gill lamellae of non-ovigerous female crabs and occasionally from the gill lamellae of male crabs.  相似文献

Concomitant immunity and its consequence against infection play roles in regulating worm burdens in helminthiasis. Under natural conditions, this immunity is generated by exposure to repeated low dose or trickle infection. In this study, concomitant immunity was induced in mice exposed repeatedly to infection with Echinostoma malayanum and its protective effect on a challenge infection evaluated. A profile of worm burden from exposure to 10 metacercariae/mouse/week rose rapidly during the first 2 weeks reaching a plateau from week 3 to 8 post infection. Based on a cumulative dose of infection, worm recoveries were around 75% in the first 2 weeks, dropped to 50% at week 3 and 19% at week 8. After week 2, adult worm burden was constant and no juvenile worms were found after week 3 of the experiment. To examine the effect of resistance against reinfection, mice in the experimental group were primarily infected with 10 metacercariae/week for 5 weeks, treated with praziquantel and were challenged with 75 metacercariae/animal. The number of worms recovered from the experimental groups was significantly lower than that from naïve control groups beginning from 24 h to 28 days post challenge. The worms in the experimental group showed growth retardation and the proportion of adult worms was lower than that in the control animals especially during the first 3 weeks of the experiment. Parasite fecundity was also suppressed compared with that in the control group. The selective effects of protective immunity on establishment, growth, and fecundity of challenged worms affected the population dynamics of E. malayanum which is a similar phenomenon to concomitant immunity in schistosomiasis.  相似文献

Generalist parasites may disproportionately use certain hosts because of different benefits associated with each host species. I measured the growth rate of the marine snail Trichotropis cancellata, a facultative kleptoparasite that can suspension feed and steal food, on different hosts to determine the relative nutritional benefits of each host. The variation in tentacle (feeding structure) area among the hosts studied had the potential to provide parasitic snails with different amounts of nutrition for growth. In field experiments, suspension-feeding snails isolated from potential hosts grew at a similar rate to snails on brachiopods and significantly more slowly than snails on the following polychaete worms: Serpula columbiana (Serpulidae), Pseudopotamilla ocellata (Sabellidae), Schizobranchia insignis (Sabellidae), and Eudistylia vancouveri (Sabellidae). However, choice among worm hosts affected snail growth rates only in the fall, when phytoplankton levels are low. At this time, snails parasitizing the sabellids Schizobranchia and Eudistylia grew more quickly than snails on Serpula. In the spring and summer, with high levels of phytoplankton, Trichotropis grew at similar rates on all worm species tested. Trichotropis spent approximately the same time stealing food from each worm host species, >50% of the time the worms had their tentacles extended (the difference among hosts was not significant). This finding demonstrates that the similarity of snail growth rates on different worm species is not due to the snails compensating for poor hosts (worms that provide food at a slower rate) by spending more time stealing food. Snails in choice experiments preferred live Serpula to empty Serpula tubes, indicating that at least some of the cue(s) snails use to identify hosts are derived from living host tissues. In choice racks containing live Serpula and live Schizobranchia, snails did not choose one host worm significantly more often than the other. Because Trichotropis grows faster on sabellids than serpulids in the fall, I predicted that snails in nature would infect sabellids more often than other species. However, snails were usually distributed randomly among host species. In the few cases where the snails showed a significant preference among host species, proportionally more snails were found on serpulids than on sabellids or sabellarids. This study is the first to quantify under natural conditions the growth benefits of a kleptoparasite across the range of possible hosts, and implies that factors other than growth rate influence host choice specificity in the marine kleptoparasite T. cancellata.  相似文献

The oligochaete worm Nais elinguis was counted during a year and a half in a full-scale, completely mixed, municipal activated sludge plant consisting of four aeration tanks connected in parallel. Simultaneously the operating variables, i.e. effluent quality, energy costs in kWh for oxygen supply in the aeration tanks, and sludge-disposal were measured. The number of worms varied both seasonally and among the aeration tanks. A major worm bloom resulted in a low sludge volume-index, lower energy consumption for oxygen supply expressed in kWh and, depending on the temperature, less sludge-disposal. The worms had no influence on the effluent quality.  相似文献

Mice were infected orally with 1,6, or 30 cysticercoids of Hymenolepis diminuta. These were allowed to develop for different periods of time before elimination with anthelminthic, thus exposing the hosts to antigens from the prestrobilate, early strobilate, or fully strobilate worms. Other groups of mice were immunized by intraperitoneal (ip) implantation of a live strobilate worm or by ip implantation of live worms from cysticercoids excysted in vitro. Strong protection against challenge with a surgically transplanted strobilate worm was achieved by prior infection with 6 or 30 worms eliminated as early as Day 3 of infection. By this time these worms would not have strobilated. Conversely, a single worm, strobilating extensively over 16 days, stimulated only weak protection. Parenteral implantation of excysted worms protected mice but parenteral implantation of a strobilate worm had no effect. It is suggested that (i) the tapeworm protective antigens are primarily related to the scolex and/or the germinative region; (ii) the number of worms and the duration of antigenic stimulation in an immunizing infection determine the magnitude of a protective secondary response.  相似文献

Inequality in body sizes is a common feature in populations of helminth parasites, with potential consequences for egg production and population genetics. Inequalities in body lengths and the effects of intraspecific competition on worm length were studied in a species of mermithid nematode parasitic in the crustacean Talorchestia quoyana (Amphipoda: Talitridae). The majority of the 753 worms recovered were relatively small, and an analysis using a Lorenz curve and Gini coefficient suggested that there were no marked inequalities in body lengths among the worms. Total worm length in the 356 infected amphipods (i.e. the sum of the lengths of all the worms in a host) increased steadily as a function of the number of worms per amphipod, whereas the length of the longest worm per amphipod peaked in amphipods harbouring intermediate numbers of worms. This last result was not significantly accounted for by the observed increase in host size with increasing intensity of infection, but resulted from a correlation between worm length and host size. As the number of worms per amphipod increased, the relative sizes of the second-, third-, and fourth-longest worms per host increased markedly. This means that relative inequalities in sizes become less pronounced, i.e. subordinate worms get closer in size to the longest worm, as the number of worms per host increases. The main consequence of this phenomenon is that worm sizes in the mermithid population are more homogeneous than they would be if intraspecific competition had stronger effects on worm growth.  相似文献

The number of laboratories using the free living nematode C. elegans is rapidly growing. The popularity of this biological model is attributed to a rapid generation time and short life span, easy and inexpensive maintenance, fully sequenced genome, and array of RNAi resources and mutant animals. Additionally, analysis of the C. elegans genome revealed a great similarity between worms and higher vertebrates, which suggests that research in worms could be an important adjunct to studies performed in whole mice or cultured cells. A powerful and important part of worm research is the ability to use transgenic animals to study gene localization and function. Transgenic animals can be created either via microinjection of the worm germline or through the use of biolistic bombardment. Bombardment is a newer technique and is less familiar to a number of labs. Here we describe a simple protocol to generate transgenic worms by biolistic bombardment with gold particles using the Bio-Rad PDS-1000 system. Compared with DNA microinjection into hermaphrodite germline, this protocol has the advantage of not requiring special skills from the operator with regards to identifying worm anatomy or performing microinjection. Further multiple transgenic lines are usually obtained from a single bombardment. Also in contrast to microinjection, biolistic bombardment produces transgenic animals with both extrachromosomal arrays and integrated transgenes. The ability to obtain integrated transgenic lines can avoid the use of mutagenic protocols to integrate foreign DNA. In conclusion, biolistic bombardment can be an attractive method for the generation of transgenic animals, especially for investigators not interested in investing the time and effort needed to become skilled at microinjection.  相似文献

Fourteen tubificid species and one lumbriculid make up the oligochaete population of the northern half of Lake Constance, a meso- to mesoeutrophic lake. Tubifex tubifex makes up 72% of the total abundance, the clean water species Stylodrilus heringianus only 0.1%. No correlation exists between total worm abundance and organic carbon content of the sediment, only 4% of the organic carbon being used by the worms. A highly significant correlation between particulate organic matter loading and its organic carbon content from 10 tributaries and total oligochaete abundance is demonstrated. The ecological index IPA is defined and used to assess the biologically relevant trophic status of the lake bed. This integrates information on the percent representation of three groups of worm species classified on atrophic basis and total abundance of the worms. Despite control of the increase in phosphorus loadings progressive eutrophication of sediments was observed after an 8 year interval.  相似文献

An outbreak of the nemertean, Carcinonemertes epialti, was observed on Hemigrapsus oregonensis during October, 1982 at Campbell Cove, Bodega Harbor, California. Mean worm intensity (296 worms/crab) was the highest recorded for this nemertean egg predator on H. oregonensis. During the outbreak, male crabs were found to harbor more worms than both non-ovigerous and ovigerous females. Crab egg mortality was substantial; 83% of the ovigerous females experienced 75–100% brood mortality. The seasonal peak in worm abundance coincided with the seasonal low in crab reproduction at this locality. A method for estimating the impact of C. epialti on H. oregonensis natality was developed using crab size and fecundity, and worm prevalence and intensity. For a non-outbreak sampling period, a mean of 5.6% egg mortality was experienced by infested crabs for the period selected. Thus, brood mortality during the outbreak was much greater than that experienced at non-outbreak periods. Heavy fishing pressure on some commercially important crab species has been suggested as a possible factor inducing worm outbreaks and facilitating their continued persistence. These observations suggest that fisheries are not necessarily responsible for the outbreaks of nemerteans on commercially important crab species. However, fishing pressure may still be a sufficient condition to promote nemertean outbreaks.  相似文献

BackgroundGuinea worm (Dracunculus medinensis) was detected in Chad in 2010 after a supposed ten-year absence, posing a challenge to the global eradication effort. Initiation of a village-based surveillance system in 2012 revealed a substantial number of dogs infected with Guinea worm, raising questions about paratenic hosts and cross-species transmission.Methodology/principal findingsWe coupled genomic and surveillance case data from 2012-2018 to investigate the modes of transmission between dog and human hosts and the geographic connectivity of worms. Eighty-six variants across four genes in the mitochondrial genome identified 41 genetically distinct worm genotypes. Spatiotemporal modeling revealed worms with the same genotype (‘genetically identical’) were within a median range of 18.6 kilometers of each other, but largely within approximately 50 kilometers. Genetically identical worms varied in their degree of spatial clustering, suggesting there may be different factors that favor or constrain transmission. Each worm was surrounded by five to ten genetically distinct worms within a 50 kilometer radius. As expected, we observed a change in the genetic similarity distribution between pairs of worms using variants across the complete mitochondrial genome in an independent population.Conclusions/significanceIn the largest study linking genetic and surveillance data to date of Guinea worm cases in Chad, we show genetic identity and modeling can facilitate the understanding of local transmission. The co-occurrence of genetically non-identical worms in quantitatively identified transmission ranges highlights the necessity for genomic tools to link cases. The improved discrimination between pairs of worms from variants identified across the complete mitochondrial genome suggests that expanding the number of genomic markers could link cases at a finer scale. These results suggest that scaling up genomic surveillance for Guinea worm may provide additional value for programmatic decision-making critical for monitoring cases and intervention efficacy to achieve elimination.  相似文献

Immune mesenteric lymph node cells (IMLNC) and hyperimmune serum, alone or in combination, were transferred to recipient, syngeneic Lewis rats. On the day of cell transfer, the recipients, along with appropriate controls, were infected with 1,000Strongyloides ratti larvae. Twelve days later, the number of adult worms in the anterior small intestine was counted and the number ofS. ratti eggs in the terminal uterus of representative female worms was determined. Using these methods, we showed that worm fecundity was reduced in the IMLNC recipients, the hyperimmune serum recipients, and the recipients of both cells and serum as compared with regular controls (averages of 2.45, 2.93, and 2.98 eggs, respectively, vs. 4.73 eggs). The number of adult worms recovered from the IMLNC recipients was not reduced (474.6 worms). However, significantly fewer worms were recovered from the recipients of IMLNC-hyperimmune serum and the recipients of hyperimmune serum only as compared with regular controls (184.8 and 171.5 worms, respectively, vs. 567.2 worms). A working hypothesis, involving an interaction of serum and cellular components, is postulated as the mechanism for this protective response.  相似文献

Sludge Reduction with a Novel Combined Worm-reactor   总被引:5,自引:1,他引:4  
In China, as a result of economic, environmental and regulation factors, excess sludge treatment and disposal represents a rising challenge for small and mid-scale municipal wastewater treatment plants (WWTPs). Although the presence of worms in aerobic wastewater treatment may lead to substantial sludge reduction, the practical application is still uncontrollable because of unstable worm growth. To overcome unstable worm growth in activated sludge process, a combined worm-reactor, consisting of sections for both free-swimming and sessile worms, was developed to enhance worm growth. This plug flow type of worm-reactor has the sessile worm growing section filled with plastic carriers, and recirculation of sludge from the section for sludge settling was carried out in order to avoid worm washing out. Sludge reduction of treating the discharged excess sludge using this worm-reactor was investigated during 53 days. In the start-up phase the combined worm-reactor was inoculated with Tubificidae. Tubificidae mainly occurred in the carriers and on the bottom of the worm-reactor, however, neither Aeolosoma nor Nais was found in the worm-reactor. Results clearly showed that the average sludge reduction was 48%, and the COD removal was low, about 8%. High NH4+ -N or NO2 -N concentrations or their combination may limit the growth of Tubificidae. Few PO 43−-P release into the effluent was observed in the worm-reactor. However, results obtained from this study cannot unequivocally be attributed to the presence of worms because it did not include a control system, and thus much more work will be needed in this concept in the further research.  相似文献

One, 5, 15 and 30 worm infections of Hymenolepis diminuta were established in juvenile or adult male (Hooded Rowett or Sprague-Dawley) rats. Worm numbers and weight, and egg output were determined from day 15 to day 85 post infection. Gradual worm loss occurred only from 15 and 30 worm infections. In 5, 15 and 30 worm infections worm weight decreased from day 19 to day 50 but no weight loss occurred in single worm infections. The size range of individual worms from a multiple infection of a single rat increased markedly following infection. Adult rats showed a greater worm loss and harboured smaller worms than juvenile rats. The data will fit either a competitive or an immunological model.  相似文献

The rate of uptake of 3H2O increases in Phocanema decipiens between 8 and 13 h after it has been activated by exposure to 37°C in 0.9% Nacl containing juvenile hormone (0.4 μl/ml), while worms incubated in the absence of the hormone show no such increase. The exchange volume of an activated worm is greater than that of an unstimulated worm, but direct measurement by dry weight determination shows that both stimulated and unstimulated worms have a similar water content. The uptake of 3HO into the excretory cell is more rapid in stimulated worms, although there is no increase in uptake into the pseudoceolomic fluid. Moreover, there is an increase in water content of the excretory cell in activated worms. These observations are best explained by postulating that in an unactivated worm two compartments exist and in only one of these is water freely available for exchange. In an activated worm, the water in both compartments is available for exchange.  相似文献
