一组鸡源乳酸菌产乳酸及其耐受特性研究*   总被引:3,自引:0,他引:3  
研究了12株(K9、D17、C1、C12、D11、D14、C2、D9、K6、C21、D1和D7)分离自肉鸡肠道的乳酸菌的产乳酸能力及其中3株产酸能力强的菌株的耐受特性。12株乳酸菌产乳酸结果表明:12h内,K6产乳酸速度最快,其次为K9和C1,24h时,D17乳酸浓度最高,48h时C1终乳酸浓度最高。K9、D17和C1的耐受试验结果表明:C1菌株耐酸能力最强,pH2时,C1菌株培养3h后还能检测到活菌,D17和K9菌株培养1h后就已经检测不到活菌。在胆盐浓度0.08%-0.40%范围内,C1、D17和K9均有一定的耐受能力,随着胆盐浓度的升高,C1、D17和K9的存活数呈现缓慢的下降趋势。3株菌中D17耐热能力最强,经80%处理后仍有10^4.9/mL存活数,而K9和C1已检测不到活菌;C1对热最敏感,65℃处理后存活数由10^8/mL降为10^3/mL。  相似文献

研究了5株(L1、12、L3、L5和L7)分离自仔猪肠道的乳酸菌的产乳酸能力及抑菌特性。结果表明:L5菌株产乳酸的速度最快,培养液中乳酸含量最高,L5菌株培养液pH值的下降速度最快,终末pH值最低,而L1菌株产乳酸的速度最慢,培养液乳酸含量最低。5株乳酸菌对大肠杆菌K88、K99、987P、O141和大肠杆菌E1及金黄色葡萄球菌均有不同程度的抑制作用;排除酸的影响后仍有22%~53%抑菌效果;经热处理后保持有92%以上的抑菌效果;蛋白酶处理后保持85%以上的抑菌效果。  相似文献

Recent research in the area of importance of microbes has revealed the immense industrial potential of exopolysaccharides and their derivative oligosaccharides from lactic acid bacteria. However, due to lack of adequate technological knowledge, the exopolysaccharides have remained largely under exploited. In the present review, the enormous potentials of different types of exopolysaccharides from lactic acid bacteria are described. This also summarizes the recent advances in the applications of exopolysaccharides, certain problems associated with their commercial production and the remedies.  相似文献

Four class IIa bacteriocins (pediocin PA-1, enterocin A, sakacin P, and curvacin A) were purified to homogeneity and tested for activity toward a variety of indicator strains. Pediocin PA-1 and enterocin A inhibited more strains and had generally lower MICs than sakacin P and curvacin A. The antagonistic activity of pediocin-PA1 and enterocin A was much more sensitive to reduction of disulfide bonds than the antagonistic activity of sakacin P and curvacin A, suggesting that an extra disulfide bond that is present in the former two may contribute to their high levels of activity. The food pathogen Listeria monocytogenes was among the most sensitive indicator strains for all four bacteriocins. Enterocin A was most effective in inhibiting Listeria, having MICs in the range of 0.1 to 1 ng/ml. Sakacin P had the interesting property of being very active toward Listeria but not having concomitant high levels of activity toward lactic acid bacteria. Strains producing class IIa bacteriocins displayed various degrees of resistance toward noncognate class IIa bacteriocins; for the sakacin P producer, it was shown that this resistance is correlated with the expression of immunity genes. It is hypothesized that variation in the presence and/or expression of such immunity genes accounts in part for the remarkably large variation in bacteriocin sensitivity displayed by lactic acid bacteria.Many lactic acid bacteria (LAB), including members of the genera Lactococcus, Lactobacillus, Carnobacterium, Enterococcus, and Pediococcus, are known to secrete small, ribosomally synthesized antimicrobial peptides called bacteriocins (26, 29, 34). Some of these peptides undergo posttranslational modifications (class I bacteriocins), whereas others are not modified (class II bacteriocins) (29, 34). Class II bacteriocins contain between 30 and 60 residues and are usually positively charged at a neutral pH. Studies of a large number of class II bacteriocins have led to subgrouping of these compounds (29, 34). One of the subgroups, class IIa, contains bacteriocins that are characterized by the presence of YGNG and CXXXXCXV sequence motifs in their N-terminal halves as well as by their strong inhibitory effect on Listeria (e.g., 3, 4, 22, 23, 27, 28, 31, 38, 45) (Fig. (Fig.1).1). Carnobacteriocin BM1 most probably is identical to piscicocin V1b (4). Sakacin P most probably is identical to bavaricin A (30). Curvacin A is identical to sakacin A (2). The consensus sequence includes residues conserved in at least 8 of the 12 sequences shown; 100% conserved residues are underlined.Class IIa bacteriocins act by permeabilizing the membrane of their target cells (1, 5, 6, 9, 10, 26, 28). The most recent studies on the mode of action of these bacteriocins indicate that antimicrobial activity does not require a specific receptor and is enhanced by (but not fully dependent on) a membrane potential (9, 28). Little is known about bacteriocin structure, and unravelling the relationships between structure and function is one of the great challenges in current bacteriocin research. A logical starting point for structure-function studies is a thorough study of the differences in activity and target cell specificity between naturally occurring homologous bacteriocins. A few such studies have been described, but these suffer from either a very limited number of tested indicator strains or the use of culture supernatants instead of purified bacteriocins (3, 4, 17, 45). The use of purified bacteriocins for comparative analyses is absolutely essential, since it is becoming increasingly evident that bacteriocin producers produce more than one bacteriocin (4, 8, 38, 48; this study).In the present study, the activities of four pure class IIa bacteriocins (pediocin PA-1, enterocin A, curvacin A, and sakacin P) (Fig. (Fig.1)1) were tested against a large number of LAB as well as several strains of the food pathogen Listeria monocytogenes. The bacteriocins were purified from their respective producer strains by use of an optimized purification protocol yielding highly pure samples. The contribution of disulfide formation was assessed and found to be important for activity. The effects of the purified bacteriocins on (noncognate) class IIa bacteriocin-producing strains are described, and the implications of our findings for immunity and resistance are discussed.  相似文献

A feasibility study of lactic bacteria as potential probiotics in larval cultures of marine fish was performed by investigating the survival of five strains of lactic bacteria in seawater by readily standardized procedures at different temperatures and salinities. These conditions were chosen in such a way that their combinations define a complete first-order factorial design. Depending on the strain and the ambient conditions, the survival adhered to first-order kinetics in some cases, and to the Gompertz equation in others. The half lives (t 0.5) calculated from these models were subsequently introduced as responses to the factorial designs, estimating the coefficients of empirical equations that describe the group effect of temperature and salinity on t 0.5. Simply additive effects were found in two cases, a negative first-order interaction in another case, while another two required second-order models.  相似文献

Antisera prepared against two isologous malic enzymes from Lactobacillus casei strains 64H and M40 were used to survey and categorize the various malic enzymes found within this diverse species. In addition to detecting three major antigenic variants of malic enzyme within this group, both antisera readily reacted with Streptococcus faecalis malic enzyme. The cross-reactions between the L. casei malic enzyme antisera and the S. faecalis malic enzyme indicated that these iso-functional enzymes found in two apparently diverse groups of organisms were immunologically homologous. A scheme proposing a common ancestry for the two species S. faecalis and L. casei based on the results of quantitative immunological studies is presented.  相似文献

The envelope proteins of 5 strains of the genus Desulfotomaculum and 12 strains of the genus Desulfovibrio were studied by sodium dodecyl sulfate-polyacrylamide gel electrophoresis and immunoblotting. The Desulfovibrio strains exhibited a typical gram-negative cell envelope, whereas the cell envelope of Desulfotomaculum strains appeared to be gram-positive. A close relationship between strains of Desulfotomaculum nigrificans was observed. A comparison between different species of Desulfotomaculum revealed some degree of similarity between Desulfotomaculum nigrificans and Desulfotomaculum ruminis, whereas Desulfotomaculum orientis seemed unique. The strains of Desulfovibrio salexigens were quite different from the strains of the other species of Desulfovibrio. In two of the strains of Desulfovibrio desulfuricans, a species-specific antigen was observed. The strains of Desulfovibrio vulgaris, Desulfovibrio africanus, and Desulfovibrio gigas and one strain of Desulfovibrio desulfuricans exhibited a similar outer membrane protein profile and also showed very similar antigenic reactions.  相似文献

将前人报道的乳酸菌质粒提取方法与大肠杆菌质粒提取方法相结合,对常用试剂盒质粒提取方法进行改进,建立了一种快速、安全、高效的乳酸菌质粒提取方法。提取过程中,溶菌酶最佳浓度为20mg/ml,最佳处理时间为30min,同时避免了毒性物质溴化乙锭的使用。多次实验结果表明,采用改进后的方法可明显提高乳酸菌天然质粒的提取效果。且重复性好,为下一步乳酸菌的分子改良奠定了基础。  相似文献

The influence of four bacteriocins from lactic acid bacteria on the proton motive force (PMF) of sensitive organisms was investigated. Pediocin PA-1 (20 μg/ml) and leuconocin S (48.5 μg/ml) mediated total or major PMF dissipation of energized Listeria monocytogenes Scott A cells in a concentration-dependent manner, as has been shown for nisin. Lactacin F (13.5 μg/ml) caused 87% PMF depletion of energized Lactobacillus delbrueckii ATCC 4797 cells, also in a concentration-dependent fashion. The energy requirements for the activity of these four bacteriocins were determined by using the ionophores nigericin and valinomycin to carry out partial and specific deenergization of the target organisms. Pediocin PA-1, leuconocin S, and lactacin F acted in an energy-independent manner, whereas the activity of nisin was confirmed to be energy dependent. These results together with published reports on other bacteriocins suggest that the bacteriocins of lactic acid bacteria share a common mechanism, the depletion of PMF.  相似文献

利用指示菌生长曲线法和琼脂再培养法研究了乳酸杆菌AT-1产生的乳酸菌素对指示菌(Escherichiacoli.sp.4447)的作用方式。与大多数抑菌剂的作用规律相似,乳酸菌素在极低浓度(<24WU:106个E.colisp.4447)时,能刺激指示菌的生长;达到一定浓度之后,开始表现出抑菌作用,并且随其浓度逐渐增高(40~80WU:106个E.colisp.4447),抑菌作用更加显著直至完全抑制指示菌的生长。取检测平板上透明圈区域的琼脂块于肉汤液体培养基中37℃再培养24h,指示菌仍能生长,该结果表明:由乳酸杆菌AT-1产生的乳酸菌素对指示菌的生长呈现抑制作用。  相似文献

The distribution of mucosa-associated bacteria, bifidobacteria and lactobacilli and closely related lactic acid bacteria, in biopsy samples from the ascending, transverse, and descending parts of the colon from four individuals was investigated by denaturing gradient gel electrophoresis (DGGE). Bifidobacterial genus-specific, Lactobacillus group-specific, and universal bacterial primers were used in a nested PCR approach to amplify a fragment of the 16S rRNA gene. DGGE profiles of the bifidobacterial community were relatively simple, with one or two amplicons detected at most sampling sites in the colon. DGGE profiles obtained with Lactobacillus group-specific primers were complex and varied with host and sampling site in the colon. The overall bacterial community varied with host but not sampling site.  相似文献
