简要介绍金纳米棒的光学性质和合成方法,重点阐述其在生物医学领域研究的最新进展,并对其今后的研究方向进行展望.金纳米棒是一种胶囊状的金纳米颗粒,具有一个横向等离子共振吸收峰和一个纵向等离子共振吸收峰,分别对应其横轴和纵轴两个特征尺寸.通过调节金纳米棒的长径比,纵向等离子共振吸收峰可由可见光区跨越至近红外光区.金纳米棒这一独特的光学性质在生物和化学传感方面有着广泛而重要的应用前景.  相似文献

Gold nanorods, with their unique morphology and optical properties have offered new prospects for biomedical and biosensing applications. Herein, the interaction between gold nanorods and a model protein has been monitored using spectroscopic techniques. The enzyme retains high fraction of its native structure with a slight increase in the helical content at the expense of β-turns. Kinetic investigations revealed notable increase of enzyme affinity for substrate without significant decrease in the Vmax. Emission spectra of tryptophan residues in the presence of chaotropic agent highlighted the maintenance of internal quenching due to the induced compactness. Comparison of the gold nanorod treated lysozyme with free enzyme revealed higher thermodynamic stability under denaturing condition. Results from this study encourage the possibility of utilizing gold nanorods as promising nanocarrier candidates for a new generation of drug delivery applications.  相似文献

The usage of gold nanoparticles (Au NPs) in biological applications has risen significantly over the last 10 years. With the wide variety of chemical and biological functionalization available and their distinctive optical properties, Au NPs are currently used in a range of biological applications including sensing, labeling, drug delivery, and imaging applications. Among the available particles, gold nanorods (Au NRs) are particularly useful because their optical absorption can be tuned across the visible to near infrared region. Here, we present a novel application of Au NRs associated with low power laser exposure of NG108‐15 neuronal cells. When cells were irradiated with a 780 nm laser, the average number of neurons with neurites increased. A similar stimulatory effect was observed for cells that were cultured with poly‐(4‐styrenesulfonic acid)‐coated and silica‐coated Au NRs. Furthermore, when the NG108‐15 cells were cultured with both bare and coated Au NRs and then irradiated with 1.2–7.5 W/cm2 at 780 nm, they showed a neurite length increase of up to 25 µm versus control. To the best of our knowledge, this effect has never been reported before. While the pathways of the stimulation is not yet clear, the data presented here demonstrates that it is linked to the absorption of light by the Au NRs. These initial results open up new opportunities for peripheral nerve regeneration treatments and for novel approaches to addressing central nervous system axons following spinal cord injury. The novel cellular-like gold nanofeet (CGNF) with movable gold core, which are derived from gold/silver core shell nanorods, have been generated by galvanic reaction protocol at room temperature. The optical property based on localized surface plasmon resonance (LSPR) has been evaluated in comparison with solid gold nanofeet, suggesting that obviously high LSPR sensitivity of CGNF contributes to enhancing optical effect for detection of analytes. In contrast with superquenching properties of nanogold for fluorescence detection of pollutants, highly sensitive detection of heavy metal contaminations, e.g., mercury ions, have been implemented via DNA functionalized silica-coated CGNF on the basis of surface enhanced fluorescence (SEF) approach.  相似文献

Gold nanorods have strong absorption bands in the near-infrared region, in which light penetrates deeply into tissues. The absorbed light energy is converted into heat by gold nanorods, the so-called 'photothermal effect'. Hence, gold nanorods are expected to act not only as on-demand thermal converters for photothermal therapy but also as controllers of a drug-release system responding to irradiation by near-infrared light. To achieve a controlled-release system that can be triggered by light irradiation, double-stranded DNA (dsDNA) was modified on gold nanorods. When the dsDNA-modified gold nanorods were irradiated by near-infrared light, the single-stranded DNA (ssDNA) was released from gold nanorods due to the photothermal effect. The amount of released ssDNA was dependent upon the power and exposure time of light irradiation. Release of ssDNA was also observed in tumors grown on mice after light irradiation. Such a controlled-release system of oligonucleotide triggered by the photothermal effect could expand the applications of gold nanorods that have unique optical characteristics in medicinal fields.  相似文献

Abstract

Rod-shaped gold nanoparticles (‘nanorods’) have recently attracted widespread attention due to their unique optical properties and facile synthesis. In particular, they can support a longitudinal surface plasmon, which results in suspensions of them having a strong extinction peak in the upper visible or near-infrared parts of the spectrum. The position of this peak can be readily tuned by controlling the shape of the rods. In addition, the surface of the nanorods can be functionalized by a very wide variety of molecules. This has led to interest in their use as selective biomarkers in biodiagnostics or for selective targeting in photothermal thearapeutics. Here, we review the recent advances in the use of gold nanorods in these applications. Additionally, the information available regarding their biocompatibility is discussed.  相似文献

By absorption spectroscopy method optical properties of gold nanorods (10x38 nm) and their interaction with globular protein bovine hemoglobin and bovine serum albumin were investigated. Nanorods behavior was studied in water solution and in solution of 97 mM NaCl under ultrasound action during 90 min and results were then compared. In water solutions nanorods coagulation (aggregation) was observed with reduced optical density of the longitudinal plasmon band widening at lamda>800 nm. In NaCI solution absorption spectra evolution had complex character and was in some degree analogous to the result that was obtained for two-dimensional grids of gold nanoparticles when changing the distance between them. By interacting with serum albumin stabilization of colloid solution and dissociation of nanorods aggregates were observed.  相似文献

Therapeutic and diagnostic methods based on photomechanical effects are attracting much current attention in contexts as oncology, cardiology and vascular surgery, for such applications as photoacoustic imaging or microsurgery. Their underlying mechanism is the generation of ultrasound or cavitation from the interaction of short optical pulses with endogenous dyes or targeted contrast agents. Among the latter, gold nanorods are outstanding candidates, but their use has mainly been reported for photoacoustic imaging and photothermal treatments. Conversely, much less is still known about their value as a precision tool for photomechanical manipulations, such as to impart local damage with high spatial resolution through the expansion and collapse of microbubbles. Here, we address the feasibility of gold nanorods exhibiting a distribution of surface plasmon resonances between about 900 to above 1100 nm as a contrast agent for photoacoustic theranostics. After testing their cytotoxicity and cellular uptake, we discuss their photostability and use to mediate cavitation and the photomechanical destruction of targeted cells. We find that the choice of a plasmonic band peaking around 1064 nm is key to enhance the translational potential of this approach. With respect to the standard alternative of 800 nm, at 1064 nm, relevant regulations on optical exposure are less restrictive and the photonic technology is more mature.   相似文献

Nondegenerate pump probe differential transmission experiments on gold nanorods with varying longitudinal surface plasmon resonance have revealed a new phenomenon where the polarity of the transient transmission signal can be reversibly switched between photo bleaching and photo-induced absorption by controlling probe fluence. Under the usual case where probe fluences are nominal, photo bleaching effect is observed for the nanorods with longitudinal surface plasmon resonance energy smaller than the probe photon energy. The laser-induced melting of the nanorods or change in their shape is ruled out for the observed optical switching effect. A quantitative understanding of the results is attempted by invoking a cascaded two-photon absorption dominant beyond a threshold probe fluence of ~75 μJ/cm2.  相似文献

Electron beam induced deposition (EBID) is used to grow on a transparent substrate plasmonic antennas formed by gold nanorods. We first discuss the influence of the growth parameters on the geometrical homogeneity of the structures. The optical response of optimized rods with different aspect ratios are measured using scattering spectroscopy. The optical data show antenna resonances in good agreement with 3D numerical simulations for pure gold antennas, validating EBID as a novel relevant technique for the fabrication of plasmonic nanostructures.  相似文献

Gold nanorods exhibit strong absorbance of light in the near infrared region, which penetrates deeply into tissues. Since the absorbed light energy is converted into heat, gold nanorods are expected to act as a contrast agent for in vivo bioimaging and as a thermal converter for photothermal therapy. To construct a gold nanorod targeted delivery system for tumor a peptide substrate for urokinase-type plasminogen activator (uPA), expressed specifically on malignant tumors, was inserted between the PEG chain and the surface of the gold nanorods. In other words, we constructed PEG–peptide-modified gold nanorods. After mixing the gold nanorods with uPA, the PEG chain was released from the surface of the gold and subsequently nanorod aggregation took place. The formation of the aggregation was monitored as a decrease in light absorption at 900 nm. Tumor homogenate induced a significant decrease in this absorption. Larger amount of the PEG–peptide-modified gold nanorods bound to cells expressing uPA in vitro compared with control gold nanorods, which had scrambled sequence of the peptide. The PEG–peptide-modified gold nanorods showed higher accumulation in tumor than the control after they were injected intravenously into tumor-bearing mice, however, the density of the peptide on the surface of the gold nanorods was a key factor of their biodistributions. This targeted delivery system, which responds to uPA activity, is expected to be a powerful tool for tumor bioimaging and photothermal tumor therapy.  相似文献

A localized surface plasmon resonance (LSPR) sensor surface was fabricated by the deposition of gold nanorods on a glass substrate and subsequent immobilization of the DNA aptamer, which specifically bind to thrombin. This LSPR aptamer sensor showed a response of 6‐nm λmax shift for protein binding with the detection limit of at least 10 pM, indicating one of the highest sensitivities achieved for thrombin detection by optical extinction LSPR. We also tested the LSPR sensor fabricated using gold bipyramid, which showed higher refractive index sensitivity than the gold nanorods, but the overall response of gold bipyramid sensor appears to be 25% less than that of the gold nanorod substrate, despite the approximately twofold higher refractive index sensitivity. XPS analysis showed that this is due to the low surface density of aptamers on the gold bipyramid compared with gold nanorods. The low surface density of the aptamers on the gold bipyramid surface may be due to the effect of shape of the nanostructure on the kinetics of aptamer monolayer formation. The small size of aptamers relative to other bioreceptors is the key to achieving high sensitivity by biosensors on the basis of LSPR, demonstrated here for protein binding. The generality of aptamer sensors for protein detection using gold nanorod and gold nanobipyramid substrates is anticipated to have a large impact in the important development of sensors toward biomarkers, environmental toxins, and warfare agents. Active application of gold nanoparticles for various diagnostic and therapeutic purposes started in recent decades due to the emergence of new data on their unique optical and physicochemical properties. In addition to colloidal gold conjugates, growth in the number of publications devoted to the synthesis and application of multifunctional nanocomposites has occurred in recent years. This review considers the application in biomedicine of multifunctional nanoparticles that can be produced in three different ways. The first method involves design of composite nanostructures with various components intended for either diagnostic or therapeutic functions. The second approach uses new bioconjugation techniques that allow functionalization of gold nanoparticles with various molecules, thus combining diagnostic and therapeutic functions in one medical procedure. Finally, the third method for production of multifunctional nanoparticles combines the first two approaches, in which a composite nanoparticle is additionally functionalized by molecules having different properties.  相似文献

In this work, we present the experimental study of the nonlinear absorption of gold nanospheres and nanorods in aqueous suspension, using picosecond white-light supercontinuum open-aperture Z-scan. We demonstrate a saturable absorption effect in all particle suspensions at low-pulse energy. In the high-pulse energy regime, the apparent reverse-saturable absorption, observed in gold nanorods, was determined to be induced by photodegradation. Using the Lorentzian deconvolution method for the absorption spectra, we explain the variations on nonlinear optical effects and prove that saturable absorption only occurs within the plasmonic bands.  相似文献

Noble metal nanoparticles hold great potential as optical contrast agents due to a unique feature, known as the plasmon resonance, which produces enhanced scattering and absorption at specific frequencies. The plasmon resonance also provides a spectral tunability that is not often found in organic fluorophores or other labeling methods. The ability to functionalize these nanoparticles with antibodies has led to their development as contrast agents for molecular optical imaging. In this review article, we present methods for optimizing the spectral agility of these labels. We discuss synthesis of gold nanorods, a plasmonic nanoparticle in which the plasmonic resonance can be tuned during synthesis to provide imaging within the spectral window commonly utilized in biomedical applications. We describe recent advances in our group to functionalize gold and silver nanoparticles using distinct antibodies, including EGFR, HER-2 and IGF-1, selected for their relevance to tumor imaging. Finally, we present characterization of these nanoparticle labels to verify their spectral properties and molecular specificity.  相似文献

Gold nanorods, rod-shaped gold nanoparticles, have strong absorbance in the near-infrared region, and the absorbed light energy can be converted to heat, the so-called photothermal effect. The gold nanorods were coated with thermoresponsive polymers, which have different phase transition temperatures that were controlled by adding comonomers, N,N-dimethylacrylamide (DMAA) or acrylamide (AAm) to N-isopropylacrylamide (NIPAM). The phase transition temperatures of poly(NIPAM-DMAA) and poly(NIPAM-AAm)-coated gold nanorods were 38 and 41 °C, respectively, while polyNIPAM-coated gold nanorods showed phase transition at 34 °C. Irradiation of the coated gold nanorods using the near-infrared laser induced a decrease in their sizes due to a phase transition of the polymer layers. Poly(NIPAM-AAm)-coated gold nanorods stably circulated in the blood flow without a phase transition after intravenous injection. Irradiation of near-infrared light at a tumor after the injection resulted in the gold specifically accumulating in the tumor. This novel accumulation technique which combines a thermoresponsive polymer and the photothermal effect of the gold nanorods should be a powerful tool for targeted delivery in response to light irradiation.  相似文献

Applications based on the optical excitation of the longitudinal surface plasmon resonance of gold nanorods (AuNRs) work at highest efficiency if all component AuNRs can be maximally excited simultaneously. This can be achieved in aligned AuNR structures, such as those embedded in uniaxially stretched polymer films. Since too high heating temperatures during film stretching cause reshaping and alteration of optical properties of the rods, a maximum allowable heating temperature is determined. The alignment of the rods is quantified by an orientational order parameter of 0.92 based on a statistically significant sample of assumed t distributed means and obtained by scanning electron microscopy. We show that a stretched AuNRs-PVA composite film has optical properties that approach the dichroic properties of an idealized ensemble of fully aligned, identical, and non-interacting AuNRs embedded in a PVA film. The idealized system is provided by FDTD simulations of a single AuNR, which we carried out using the size- and shape-adapted dielectric function of gold and the software RSOFT.  相似文献

In the present study, the effect of nanosized graphene oxide layer on thermal stability and biocompatibility of gold nanorods has been examined. The graphene oxide-wrapped gold nanorods were prepared by electrostatic interaction between negatively charged graphene oxide and positively charged nanorods. The resulting nanohybrids were then heated at different time intervals to 95 °C in a water bath to assess the effect of heat on the rods morphology. The structural changes in gold nanorods were monitored via UV-Vis spectroscopy measurements and transmission electron microscopy images. In similar experiments, the graphene oxide used to wrap gold nanorods was reduced by ascorbic acid in a 95 °C water bath. Our results indicate that while bare gold nanorods are highly vulnerable to elevated temperatures, graphene oxide and reduced graphene oxide-coated gold nanorods remain thermally stable with no structural changes. We also confirmed that the enhanced thermal stability is highly dependent on the concentration of deposited graphene oxide available on the surface of the gold nanorods. In addition, we performed an MTT (3-[4,5-dimethylthiazol-2yl]-2,5-diphenyltetrazoliumbromide) assay to make a comparison between the cytotoxicity of the nanohybrids and their primary building blocks on human dermal fibroblast cells as a normal cell line. We found evidence that graphene oxide can enhance the biocompatibility of the rods through covering toxic chemicals on the surface of them.

A low-cost and effective surface-enhanced Raman scattering (SERS) substrate consisting of aligned gold nanorods is obtained by stretching the poly(vinyl alcohol) nanorods composite film doped with the probe molecule. The SERS intensity of characteristic vibrational band of the probe is observed obviously dependent on the angle (θ) between incident polarization and major axis of nanorod. The relationship between them manifests a cos2 θ dependence. The result is illustrated from both the localized field enhancement and optical antenna effect of gold nanorod. The finite element method calculation is also performed to further confirm the conclusion.  相似文献
