During membrane filtration antibiotics belonging to different chemical groups are strictly absorbed on the filters. When the filters are put into liquid thioglycol medium, the residual amounts of the antibiotics on the filters did not prevent the growth of sensitive microflora experimentally added to the drug. When the filter was put onto solid nutrient medium, only resistant forms of the microbes grew as a rule on its surface, the amount of the grown microbes being 26--43 per cent of the added one. The sensitive microbes grew only in the amount of 0.3--1.3 per cent. Subsequently the residues of the antibiotic adsorbed on the filter inhibited the growth of the sensitive and partially resistant microflora.  相似文献

目的 检查和分析血液制品原料混合血浆的微生物限度.方法 按照《中华人民国和国药典》(2010年版)三部微生物限度检查法,采用薄膜过滤法检查10批混合血浆的微生物限度情况,分析混合血浆的微生物状况.结果 10批混合血浆的微生物总数量在170~ 330 cfu/mL之间.在检查的三种微生物中,细菌总数最多,霉菌次之,酵母菌数量最少.结论 成功采用薄膜过滤法检查了混合血浆中微生物限度状况.  相似文献

A universal diluting fluid (UDF) was developed for use in membrane filtration sterility testing. Diuting fluid is used to free the filter membrane of preservative residues which could inhibit microbial recovery. Current procedures employ one of several fluids described by the United States Pharmacopeia. UF was designed to utilize the neutralizing capabilities of a combination of inactivating agents while minimizing any inherent microbial toxicity. This formulation eliminates the need for multiple types of preservative-specific diluting fluids. Its neutralizing effectiveness was examined against several common preservative agents used in health care products. UDF provides significantly better microorganism recovery than dilution alone among a wide range of preservative classes.  相似文献

AIMS: To apply fluorescent staining method for fast assessment of microbial quality of herbal medicines. METHODS AND RESULTS: The number of total bacteria and esterase-active bacteria on powdered traditional Chinese medicines were enumerated by fluorescent staining method using 6-carboxyfluorescein diacetate (6CFDA) and 4',6-diamidino-2-phenylindole (DAPI), and they were compared with colony-forming units (CFU). The CFU was approximately 10(3) per gram in ginseng radix, and no bacterial colonies were detected from others. However, the total bacterial number (TDC) was more than 10(7) per gram, and number of bacteria possessing esterase activity ranged from 1 to 3% of TDC. CONCLUSIONS, SIGNIFICANCE AND IMPACT OF THE STUDY: Many bacteria in each Chinese medicine had enzyme activity and most of them could not be detected by conventional plate counting technique. Enumeration of bacterial cells on traditional Chinese medicines by fluorescent staining method requires less than 1 h. The double staining method with 6CFDA and DAPI could be applicable to rapid microbial monitoring of crude drugs.  相似文献

Gravity-driven membrane (GDM) filtration is a promising tool for low-cost decentralized drinking water production. The biofilms in GDM systems are able of removing harmful chemical components, particularly toxic cyanobacterial metabolites such as microcystins (MCs). This is relevant for the application of GDM filtration because anthropogenic nutrient input and climate change have led to an increase of toxic cyanobacterial blooms. However, removal of MCs in newly developing GDM biofilms is only established after a prolonged period of time. Since cyanobacterial blooms are transient phenomena, it is important to understand MC removal in mature biofilms with or without prior toxin exposure. In this study, the microbial community composition of GDM biofilms was investigated in systems fed with water from a lake with periodic blooms of MC-producing cyanobacteria. Two out of three experimental treatments were supplemented with dead biomass of a MC-containing cyanobacterial strain, or of a non-toxic mutant, respectively. Analysis of bacterial rRNA genes revealed that both biomass-amended treatments were significantly more similar to each other than to a non-supplemented control. Therefore, it was hypothesized that biofilms could potentially be ‘primed’ for rapid MC removal by prior addition of non-toxic biomass. A subsequent experiment showed that MC removal developed significantly faster in mature biofilms that were pre-fed with biomass from the mutant strain than in unamended controls, indicating that MC degradation was a facultative trait of bacterial populations in GDM biofilms. The significant enrichment of bacteria related to both aerobic and anaerobic MC degraders suggested that this process might have occurred in parallel in different microniches.  相似文献

The public health aspects of the use of wastewater in agriculture and the effects of the drip irrigation method on the contamination of vegetables were studied. The method used was to simulate enteric microorganisms' dissemination by contaminated irrigation water in the field. The vegetables were irrigated with an effluent inoculated with a high titer of traceable microorganisms: poliovirus vaccine and a drug-resistant Escherichia coli. The dissemination of the marker organisms in the field was followed, and the effects of certain manipulations of the drip irrigation method on the contamination of the crops by the effluent were examined. It was shown that drip irrigation under plastic sheet cover with the drip lines placed either on the soil surface or buried at a depth of 10 cm significantly reduced crop contamination from inoculated irrigation water even when massive doses of bacteria and viruses were used. The microbial contamination was found to persist in the irrigation pipes and in the soil for at least 8 and 18 days, respectively. The data indicate that the recovery of the marker organisms was affected by soil texture and environmental conditions.  相似文献

Membrane biofouling was investigated during the early stages of filtration in a laboratory-scale membrane bioreactor operated on molasses wastewater. The bacterial diversity and composition of the membrane biofilm and activated sludge were analyzed using terminal restriction fragment length polymorphism coupled with 16S rRNA clone library construction and sequencing. The amount of extracellular polymeric substances produced by bacteria was investigated using spectroscopic methods. The results reveal that the bacterial community of activated sludge differs significantly from that of the membrane biofilm, especially at the initial phase. Phylogenetic analysis based on 16S rRNA gene sequences identified 25 pioneer OTUs responsible for membrane surface colonization. Also, the relationship between the identified bacterial strains and the system specifications was explored.  相似文献

A method applicable for beforehand determination of streptomycin sulfate powder stability by the colour of its solutions is described. The method involves filtration of dilutions of the mixture through a membrane filter followed by replication on ox blood agar and enumeration of organisms. This method was compared with the conventional drop count method, and a significant correlation was found (r = 0.985). When solutions containing antibiotic were used, the membrane filtration method gave a more reliable estimate of the viable count at the time of sampling.  相似文献

介绍了两种膜过滤细胞直接计数新方法,即不透明滤膜的金相显微镜细胞直接计数法和透明滤膜的生物显微镜细胞直接计数法的基本原理、仪器设备、分析和计算方法,并且将此方法与荧光显微镜细菌计数、血球计数板等方法进行了比较。实际计数结果表明,两种膜过滤细胞直接计数法配合了独特的酸碱处理法以分散聚团细胞,用于非环境样品细胞浓度的测定具有快速准确简便的特点。此外,该方法基本不受细胞大小的影响,除了适用于体型较大的藻类、酵母菌的快速计数以外,还可以清晰地分辨诸如光合细菌这样体积较小的细菌。  相似文献

Aims: To investigate the effectiveness of pulsed electric field (PEF) treatment as a new method for inactivation of micro-organisms in complex biomatrices and to assess this by quantifying the inactivation of Escherichia coli seeded in collagen gels. Methods and Results: PEF was applied to E. coli seeded collagen gels in static (nonflowing) chambers. The influence of electric field strength, pulse number and seeded cell densities were investigated. The highest level of inactivation was obtained at the maximum field strength of 45 kV cm−1. For low levels of E. coli contamination (103 CFU ml−1), PEF treatment resulted in no viable E. coli being recovered from the gels. However, PEF treatment of gels containing higher cell densities (≥104 CFU ml−1) did not achieve complete inactivation of E. coli. Conclusions: PEF treatment successfully inactivated E. coli seeded in collagen gels by 3 log10 CFU ml−1. Complete inactivation was hindered at high cell densities by the tailing effect observed. Significance and Impact of the Study: PEF shows potential as a novel, nondestructive method for decontamination of collagen-based matrices. Further investigation is required to ensure its compatibility with other proteins and therapeutic drugs for tissue engineering and drug delivery applications.  相似文献

An Antimicrobial Removal Device (ARD) is proposed as an alternative to membrane filtration in the sterility testing of antibiotics. The device consists of a glass vial containing adsorbent resins which completely removed the antibacterial activity of a range of antibiotics without affecting the recovery of contaminating bacteria. The method is simple to use and has several advantages compared to membrane filtration including easier aseptic handling and lack of antibiotic carry-over to subculture media. A problem with resin turbidity in subculture media was overcome by the use of an indicator medium or by subculture into fresh medium.  相似文献

Optimal conditions for determination of microbial contamination of drugs were studied on artificially contaminated powders and tablets of phenoxymethylpenicillin, ampicillin, oxacillin and dicloxacillin. The method of membrane filtration was the best for determination of the microbial contamination of the powders. However, it was not possible to wash out completely the antibiotic from the membrane filter. To prevent this it was necessary to add penicillinase into the nutrient medium onto which the filter was put for providing the microbial growth. For determination of microbial contamination of tablets direct plating of 3 per cent suspension of the tablet mass onto the surface of the nutrient medium with penicillinase was the best.  相似文献

A new primary model based on a thermodynamically consistent first-order kinetic approach was constructed to describe non-log-linear inactivation kinetics of pressure-treated bacteria. The model assumes a first-order process in which the specific inactivation rate changes inversely with the square root of time. The model gave reasonable fits to experimental data over six to seven orders of magnitude. It was also tested on 138 published data sets and provided good fits in about 70% of cases in which the shape of the curve followed the typical convex upward form. In the remainder of published examples, curves contained additional shoulder regions or extended tail regions. Curves with shoulders could be accommodated by including an additional time delay parameter and curves with tails shoulders could be accommodated by omitting points in the tail beyond the point at which survival levels remained more or less constant. The model parameters varied regularly with pressure, which may reflect a genuine mechanistic basis for the model. This property also allowed the calculation of (a) parameters analogous to the decimal reduction time D and z, the temperature increase needed to change the D value by a factor of 10, in thermal processing, and hence the processing conditions needed to attain a desired level of inactivation; and (b) the apparent thermodynamic volumes of activation associated with the lethal events. The hypothesis that inactivation rates changed as a function of the square root of time would be consistent with a diffusion-limited process.  相似文献
