We carried out a field evaluation of the MM3-SERO ELISA for the diagnosis of Fasciola hepatica infection, by analysing serum and milk samples from individual cows and samples from bulk milk tanks. The diagnostic performance of the assay was assessed with serum samples from all 257 cows in eight fluke-free herds, and 240 cows with natural fasciolosis (diagnosed in vivo and/or post-mortem). Assay performance for individual milk samples was determined by analysis of paired serum and milk samples from 947 lactating cows from 33 F. hepatica-infected farms. The diagnostic usefulness of the assay for bulk tank milk was evaluated by analysis of bulk milk from infected (33) and non-infected (35) farms. For serum samples, the sensitivity, specificity and diagnostic accuracy of the assay were respectively 99.2% (95% CI: 97.0%–99.9%), 100% (95% CI: 98.6%–100%) and 0.997 (95% CI: 0.987–1.000). The only two infected animals in which serum antibodies were not detected had very low parasitic burdens (with only 2 and 3 flukes observed). The performance of the MM3 SERO ELISA for individual milk samples was similar to that for serum samples, and the stepwise linear regression revealed a strong correlation between the results for the milk samples and the serum samples (R2 = 0.84; p < 0.001). The agreement between results obtained with pairs of serum and milk samples was very high: there was matching classification in 96% (910/947) of paired samples (kappa = 0.92; p < 0.001). Individual milk samples may therefore be used, instead of serum samples, in the MM3-SERO ELISA, for reliable detection of seropositive cows. Testing bulk tank milk samples enabled detection of infected herds, even when the within-herd prevalence of infection was as low as 12%. We conclude that the MM3-SERO ELISA is a sensitive and highly specific test for serodiagnosis of bovine fasciolosis, and can be used with individual samples of either serum or milk. Animal welfare in livestock production is of great interest to consumers. The organic farming approach strives to ensure animal welfare based on preventive measures, but there are very few scientific studies that compare the actual differences in animal welfare between organic and conventional farms. Those studies that have been carried out frequently focus on specific aspects of animal welfare, mostly health issues. The aim of the present study, therefore, was to investigate the effects of the farming system on the welfare of dairy cows in a more holistic way. Although this study was carried out in just two federal states of Germany, the results could serve as a suitable model for the whole country. We used the Welfare Quality assessment protocol to measure welfare for dairy cattle (Welfare Quality®, 2009) and the results showed significant differences (P < 0.05) between organic and conventional farms, but there was also considerable variance between individual farms of the same farming system. Organic farms scored higher in all four Welfare Quality® principles: “Good Feeding”, “Good Housing”, “Good Health” and “Appropriate Behavior” compared to conventional farms. In particular, organic farms obtained higher scores with respect to Welfare Quality® measures of resting comfort, which contributes to a lower percentage of lameness; organic farms also implemented less painful methods for disbudding, or indeed carried out no disbudding, and provided access to pasture and outdoor exercise. However, organic farms still have room for improvement, especially with respect to animal health. Therefore, outcome‐based specifications should be included in the current (purely action-oriented) European regulation of organic production (EC, 2008; EU, 2018) to safeguard the health‐related aspects of animal welfare. 相似文献

A simulation rumen model has been developed to function under non-steady state conditions in order to allow prediction of nutrient availability in dairy cows managed under discontinuous feeding systems. The model simulates availability of glycogenic, aminogenic and lipogenic nutrients to lactating dairy cows fed discontinuously. The model structure considers input of up to three different feeds fed independently at any time during the day. Feeds are described by their nitrogen (N), carbohydrate and fatty acid fractions. The N containing feed fractions include ruminally undegraded crude protein (CP), ruminally insoluble but potentially degradable CP, ruminally soluble CP and ammonia N. The feed carbohydrate fractions include ruminally undegradable neutral detergent fibre (NDF), ruminally degradable NDF, ruminally insoluble starch, ruminally soluble starch and sugars. The fatty acids in the feeds are divided between long chain fatty acids and volatile fatty acids (VFA). Additionally four pools were defined representing absorption of amino acids, glucose, long chain fatty acids and volatile fatty acids. The rumen microbial population is represented as a single pool. Besides a flexible structure, new features to the extant model include adoption of the concept of chewing efficiency (or chewing effectiveness) during eating, variable fractional ruminal absorption rates of VFA and variable fractional ruminal degradation rates of NDF as a function of rumen liquid pH, as well as a variable rumen volume which directly affects rumen concentrations of metabolites. The model continuously (i.e., by minute) predicts release of soluble components from the feeds in the rumen, concentration and absorption of fermentation end products in the rumen, rumen pools of nutrients and microbial biomass dynamics, as well as passage of microbial biomass and non-fermented nutrients from the rumen, in response to various feeding strategies. Model evaluation covered a wide range of feeding strategies that included pasture and housed feeding systems. Overall, the mean square prediction error (MSPE) as a percentage of the observed mean was relatively low (<10%) with a high amount of the total variation explained by random variation (>65%). Deviation from unity varied between 23% (rumen dry matter content) and 25% (NDF), indicating some consistent over and/or under prediction. A more detailed evaluation was done based on studies available that reported diurnal behaviour of key model outputs such as rumen pools, rumen pH, and rumen VFA. The predictions broadly simulated the observed values quantitatively, relative to general diurnal patterns, and relative to differences between treatments in the predicted diurnal patterns. Results show that the model provides a tool to assess potential outcomes of changing feeding strategies which may be particularly valuable in assessing selection of feeds, amounts and times of the day to offer the feeds. The continuous nature of the simulated output also allows determination of the time(s) of the day that ruminal (and/or post-ruminal) delivery of nutrients may limit ruminal output of nutrients (and/or availability of nutrients) to support milk nutrient synthesis. 相似文献

Two tree shades (TS1 and TS2) and an artificial shade structure (AS) were evaluated using black globe temperatures (BGTs) to assess their effectiveness in reducing heat load. The artificial structure consisted of a black woven polypropylene cloth providing 80% shade, mounted on 2.5-m-high eucalyptus posts. The work was carried out at Rafaela Experimental Station, Argentina, during the summer (January and February) 1994. BGTs and floor temperatures were measured in concrete floor holding pens with and without artifical shade. The results showed no difference between TS1, TS2 and AS, their average BGTs being 30.2 (SD 0.58), 29.0 (SD 0.70) and 30.2 (SD 0.74)°C, respectively. BGTs under all three shades were significantly lower (P<0.01) than the average BGT recorded outside: 35.5 (SD 1.12)°C. Average BGTs in holding pens were 32.4 (SD 1.38) and 39.9 (SD 1.91)°C for shaded and non-shaded areas (P<0.01). The corresponding average floor temperatures were 27.8 (SD 0.68)°C and 47.7 (SD 2.13)°C (P<0.01). To assess the effects of shade on animal well-being, afternoon rectal temperatures (RT) and respiratory rate (respirations per minute, RR) of lactating cows were recorded twice a week. Rectal temperatures were significantly higher for non-shaded cows (P<0.01), mean RT being 40.1 (SD 0.65)°C vs 39.3 (SD 0.42)°C for the shaded animals. Corresponding RRs were 78.9 (SD 18.0) and 60.7 (SD 10.6) (P<0.05). It was concluded that: (1) tree and artificial shades produced similar effects, (2) shading the holding pen with an 80% shading cloth was effective in reducing heat load and floor temperatures, and (3) access to shade in our pasture-based system improved animal well-being. In France, numerous Protected Designation of Origin (PDO) cheese value chains have revised their specifications to address environmental protection, particularly at the farm level. In this stage, it is nevertheless impossible to state whether the proposed modifications may ensure the sustainability of environmental protection as no environmental sustainability diagnostic has been conducted. The aim of this study is to construct an assessment method for the Cantal PDO farms.A multi-criteria assessment approach was implemented in the following 5 steps: definition of the framework of the study, determination of the principles and criteria, definition of the indicators, aggregation of all elements of the assessment, and finally, test of the assessment method on farms. Focus groups meetings were organised for the first four steps. The panel discussions were based on data obtained from both the literature and surveys of stakeholders operating in the value chain and within the territory.The obtained assessment method comprises 4 principles characterised by 33 indicators. The most important principle (45%), management of grassland resources, is a key economic and environmental point of the studied farms. It reflects the necessary intensive use of grass by the animals in the PDO farms. Few existing methods addressed this principle that required the design of several new indicators. The second principle, impact of agricultural practices (25%), is often included in environmental sustainability assessment methods The adopted indicators are, thus, drawn from existing methods and adapted to the context of the farms studied. The management of the farm buildings and the landscape principle (18%) is often considered in social sustainability. Its role in environmental sustainability can be explained by its strong link with the image of PDO products. Primarily drawn from the literature, the used indicators have been extended to make the assessment method as objective as possible. Finally, the management of local, energy and water resources principle has the lowest weighting (12%). This can be explained by the limited number of indicators.This assessment method was constructed for the Cantal PDO farms by considering their environment and particularities. In consequence, the stakeholders have taken ownership of it. It could be used by other ruminant systems based on grassland. The development of numerous indicators enhanced the pool of existing indicators. This assessment method was sensitive and allowed for the discrimination between farms. 相似文献

Conductive cooling, which is based on direct contact between a cow lying down and a cooled surface (water mattress, or any other heat exchanger embedded under the bedding), allows heat transfer from the cow to the cooled surface, and thus alleviate heat stress of the cow. Conductive cooling is a novel technology that has the potential to reduce the consumption of energy and water in cooling dairy cows compared to some current practices. A three-dimensional conduction model that simulates cooling thermally-stressed dairy cows was developed. The model used a computational fluid dynamics (CFD) method to characterize the air-flow field surrounding the animal model. The flow field was obtained by solving the continuity and the momentum equations. The heat exchange between the animal and the cooled water mattress as well as between the animal and ambient air was determined by solving the energy equation. The relative humidity was characterized using the species transport equation. The conduction 3-D model was validated against experimental temperature data and the agreement was very good (average error is 4.4% and the range is 1.9–8.3%) for a mesh size of 1117202. Sensitivity analyses were conducted between heat losses (sensible and latent) with respect to air temperature, relative humidity, air velocity, and level of wetness of skin surface to determine which of the parameters affect heat flux more than others. Heat flux was more sensitive to air temperature and level of wetness of the skin surface and less sensitive to relative humidity. 相似文献

The aim of this study was to evaluate the effects of dietary Quebracho tannin extract (QTE) on feed intake, apparent total tract digestibility (ATTD), excretion of urinary purine derivatives (PD) and milk composition and yield in dairy cows. Fifty Holstein cows were divided into two groups. To reach a similar performance of both groups, cows were divided according to their milk yield, body weight, days in milk and number of lactations at the start of the experiment averaging 33.2 ± 8.2 kg/d, 637 ± 58 kg, 114 ± 73 d and 2.3 ± 1.6 lactations, respectively. The cows were fed a basal diet as total mixed ration containing on dry matter (DM) basis 34% grass silage, 32% maize silage and 34% concentrate feeds. Three dietary treatments were tested, the control (CON, basal diet without QTE), QTE15 (basal diet with QTE at 15 g/kg DM) and QTE30 (basal diet with QTE at 30 g/kg DM). Two treatments were arranged along six periods each 21 d (13 d adaptation phase and 8 d sampling phase). The ATTD of DM and organic matter were reduced only in Diet QTE30, whereas both QTE treatments reduced ATTD of fibre and nitrogen (N), indicating that QTE impaired rumen fermentation. Nevertheless, feed intake was unaffected by QTE. In Diet CON, urinary N excretion accounted for 29.8% of N intake and decreased in treatments QTE15 and QTE30 to 27.5% and 17.9%, respectively. Daily faecal N excretion increased in treatments CON, QTE15 and QTE30 from 211 to 237 and 273 g/d, respectively, which amounted to 39.0%, 42.4% and 51.7% of the N intake, respectively. Hence, QTE shifted N excretion from urine to faeces, whereas the proportion of ingested N appearing in milk was not affected by QTE (average 30.7% of N intake). Daily PD excretion as indicator for microbial crude protein (CP) flow at the duodenum decreased in treatment QTE30 compared with Diet CON from 413 to 280 mmol/d. The ratios of total PD to creatinine suggest that urinary PD excretion was already lower when feeding Diet QTE15. While there was no effect of Diet QTE15, treatment QTE30 reduced milk yield, milk fat and protein. Both QTE treatments reduced milk urea concentration, which suggest that ruminal degradation of dietary CP was reduced. In summary, adding QTE at dosages of 15 and 30 g/kg DM to diets of lactating dairy cows to improve feed and protein use efficiency is not recommended. 相似文献

Abstract The objective of the study was to investigate the influence of two roughage-to-concentrate ratios, with or without linseed oil supplementation, on the flow of fatty acids in the intestinal chyme and the secretion in milk fat in late lactating cows. Seven late lactating cows fitted with cannulae in the dorsal rumen and simple T-shaped cannulae in the proximal duodenum were randomly assigned to four experimental periods applying an incomplete replicated 2×2 Latin square design. The rations consisted of meadow hay and a concentrate mixture given in a ratio of 70 : 30 or 30 : 70 on dry matter basis. The basal rations were fed without or with 200 g linseed oil daily. After three weeks of adaptation, samples from the duodenal chyme were taken to study the flow of fatty acids. Additionally, milk samples were analysed for their milk fat composition. Decreasing roughage/concentrate ratio and linseed oil supplementation significantly increased the flow of monounsaturated fatty acids (MUFA), trans-fatty acids (tFA) and conjugated linoleic acids (CLA) in the duodenum. Furthermore, linseed oil increased the flow of saturated fatty acids (SFA) in the duodenum. Higher concentrate portion (H 30) and linseed oil supplementation significantly decreased the milk fat content. SFA were lower (p < 0.05) and MUFA were higher (p < 0.05) in milk fat after linseed oil supplementation; H 30 resulted in more polyunsaturated fatty acids (PUFA, p < 0.05) in the milk. Linseed oil supplementation significantly increased tFA and CLA in milk fat. The higher CLA content in milk fat as compared to that in the digesta suggests that a substantial endogenous synthesis of CLA in the mammary gland tissue through Δ9-desaturase took place. Between 21% and 48% of duodenal t11-C18:1 were converted into c9, t11-CLA in milk fat. 相似文献

Heat stress is one of the greatest issues of the dairy industry in regions with hot climate. Since coat color appears to be related to heat stress adaptiveness, we compared rectal temperatures and surface temperatures of Red-and-white (RW, n = 14) and Black-and-white (BW, n = 16) Holstein cows using infrared thermography in both cold (July; mean temperature: 15.5 °C) and hot (March; mean temperature: 30.5 °C) seasons in Southern Brazil. Thermographic images were taken from the left side of the animal at a distance of 4 m. The images obtained were then analyzed using the software Testo IRSoft. The variables obtained by thermography of the body surface include the temperature of non-pigmented patches, obtained using the average of five spots on white patches in a rectangle drawn on the body of the cow from the scapula to the ilium of the cow until the middle of the ribs; the temperature of pigmented patches, obtained using an average of 5 pigmented spots on the same rectangle; the temperature at the hottest spot and the temperature at the coldest spot, within the same rectangle. Rectal temperature measures were taken by a mercury thermometer during milkings. In our findings, during the cold season, RW cows had lower temperatures on the surface of pigmented spots (p = 0.01) but did not differ from BW animals when comparing rectal temperatures (p = 0.70). During the hot season, however, RW cows had lower temperatures on white spots (p = 0.049) as well as lower rectal temperatures (p = 0.029). These results suggest that the red coat phenotype presents less absorption of solar radiation, retaining less heat. 相似文献

In many epidemiological studies time to event data are clustered and the physiological relationship between (time-dependent) covariates and the log hazard is often not linear as assumed in the Cox model. Introducing frailties in the Cox model can account for the clustering of the data and smoothing splines can be used to describe nonlinear relations. These two extensions of the Cox model are introduced jointly and it is shown how penalized partial likelihood techniques can be used to fit the extended model. We demonstrate the need for such a model to study the relation between the physiological covariates milk ureum and protein concentration and the log hazard of first insemination in dairy cows, with the farms as clusters. 相似文献

The herd dynamic milk (HDM) model is a dynamic model capable of simulating the performance of individual dairy animals (from birth to death), with a daily time step. Within this study, the HDM model is described and evaluated in relation to milk production, body condition score (BCS) and BCS change throughout lactation by comparing model simulations against data from published experimental studies. The model’s response to variation in genetic potential, herbage allowance and concentrate supplementation was tested in a sensitivity analysis. Data from experiments in Ireland and France over a 3-year period (2009–11) were used to complete the evaluation. The aim of the Irish experiment was to determine the impact of different stocking rates (SRs) (SR1: 3.28 cow/ha, SR2: 2.51 cow/ha) on key physical, biological and economic performance. The aim of the French experiment was to evaluate over a prolonged time period, the ability of two breeds of dairy cows (Holstein and Normande) to produce and to reproduce under two feeding strategies (high level and low level) in the context of compact calving. The model evaluation was conducted at the herd level with separate evaluations for the primiparous and multiparous cows. The evaluation included the two extreme SRs for the Irish experiment, and an evaluation at the overall herd and individual animal level for the different breeds and feeding levels for the French data. The comparison of simulation and experimental data for all scenarios resulted in a relative prediction error, which was consistently <15% across experiments for weekly milk production and BCS. In relation to BCS, the highest root mean square error was 0.27 points of BCS, which arose for Holstein cows in the low feeding group in late lactation. The objective of this study was to investigate the effects of niacin and dietary concentrate proportion on body temperature, ruminal pH and milk production of dairy cows. In a 2 × 2 factorial design, 20 primiparous Holstein cows (179 ± 12 days in milk) were assigned to four dietary treatments aimed to receive either 0 or 24 g niacin and 30% (low) or 60% (high) concentrate with the rest being a partial mixed ration (PMR) composed of 60% corn and 40% grass silage (on dry matter basis). Ambient temperature and relative humidity were determined and combined by the calculation of temperature humidity index. Respiration rates, rectal, skin and subcutaneous temperatures were measured. Milk production and composition were determined. Ruminal pH and temperature were recorded at a frequency of 5 min using wireless devices for continuous intra-ruminal measurement (boluses). pH values were corrected for pH sensor drift. The climatic conditions varied considerably but temporarily indicated mild heat stress. Niacin did not affect skin, rectal and subcutaneous temperatures but tended to increase respiration rates. High concentrate reduced skin temperatures at rump, thigh and neck by 0.1–0.3°C. Due to the technical disturbances, not all bolus data could be subjected to statistical evaluation. However, both niacin and high concentrate influenced mean ruminal pH. High concentrate increased the time spent with a pH below 5.6 and ruminal temperatures (0.2–0.3°C). Niacin and high concentrate enhanced milk, protein and lactose yield but reduced milk fat and protein content. Milk fat yield was slightly reduced by high concentrate but increased due to niacin supplementation. In conclusion, niacin did not affect body temperature but stimulated milk performance. High concentrate partially influenced body temperatures and had beneficial effects on milk production. 相似文献

Milk production is responsible for emitting a range of greenhouse gases (GHGs), mainly carbon dioxide (CO2), nitrous oxide (N2O) and methane (CH4). In Life Cycle Assessments (LCA), the Global Warming Potential with a time horizon of 100 years (GWP100) is used almost universally to aggregate emissions of individual gases into so-called CO2-equivalent emissions that are used to calculate the overall carbon footprint of milk production. However, there is growing awareness that, depending on the purpose of the LCA, metrics other than GWP100 could be justified and some would give a very different weighting for the short-lived gas CH4 relative to the long-lived gases CO2 and N2O when calculating the carbon footprint. Pastoral dairy production systems at different levels of intensification differ in the balance of short- and long-lived GHGs associated with on- and off-farm emissions. Differences in the carbon footprint of different production systems could therefore be highly sensitive to the choice of GHG metric. Here we explore the extent to which alternative GHG metric choices would alter the carbon footprint of New Zealand milk production at different levels of intensification at national, regional and individual farm scales and compared to the carbon footprint of milk of selected European countries. We find that the ranking of different production systems and individual farms in terms of their carbon footprint is relatively robust against the choice of GHG metric, despite significant differences in their utilisation of pastures versus supplementary off-farm feed, fertiliser use and energy consumption at various stages of farm operations. However, there are instances where alternative GHG metric choices would fundamentally change the conclusions of LCA of different production systems, including whether a move towards higher or lower input systems would increase or decrease the average carbon footprint of milk production in New Zealand. Greater transparency about the implications of alternative GHG metrics for LCA, and the often inadvertent and implicit value judgements embedded in these metrics, would help ensure that policy decisions and consumer choices based on LCA indeed deliver the climate outcomes intended by end-users. 相似文献

For this study, we used the results of sire fertility (SF) measured on a monthly basis by the ratio between the number of pregnant cows and the number of inseminations, from a total of 905 140 inseminations carried out in Cuba between 1994 and 2003. These artificial inseminations were made using 815 sires in 3249 herds throughout the country, and were analysed using additive, non-additive linear models and a random regression model (RRM). The additive genetic (add), heterosis (het) and recombination loss (rec) coefficients were estimated according to the proportion of Zebu (Z) and Holstein (H) blood from the paternal and maternal origin of each cow. The mean level of SF was 48.8%, whereas het and rec were 9.6% and −8.4%, respectively. The heritability (h2) of a single insemination ranged from h2 = 0.011 to h2 = 0.030 for females from 0% to 100% of H genes. The additive multi-trait and RRM analyses showed the existence of heterogeneous genetic (co)variance components, as the proportion of Holstein genes in the inseminated cow increased. We found low genetic correlations for SF recorded in pure-bred and cross-bred females, with over 50% of breed differences in their additive genetic composition. The use of a RRM allows us to identify the changes in genetic (co)variance and estimated breeding values in the whole trajectory of the different proportions of Bos taurus × Bos indicus blood. 相似文献

鉴于畜牧业生态系统健康与人类、社会、经济、环境等具有密切的关系,从活力、组织结构和恢复力三方面,提出区域畜牧业生态系统健康评价指标体系及等级评价模型.其中,适宜载畜量、人均养畜量、畜群结构、草地退化沙化率、良种畜比重、舍饲、半舍饲比例是我国北方牧区草地畜牧业生态系统健康状况的主要控制因子.选取内蒙古牧区、新疆牧区、青海牧区、甘肃牧区、西藏牧区五个区域,对区域内畜牧业生态系统健康分别进行评价及诊断,评价结果为:健康状况处于\"中\"级的牧区是内蒙古(2.7730)、新疆(2.5641)以及青海牧区(2.4673);健康状况处于\"低\"级的牧区是甘肃(2.2335)和西藏牧区(1.8884).针对评价过程中暴露的活力、组织结构和恢复力问题,对5个牧区草地畜牧业生态系统健康进行了诊断,认为在中国北方牧区气候暖干化背景下,只有调整草地畜牧业发展战略,重视和确定草地资源在提供生态公共产品中的重要作用和地位,从生态效益和经济效益的要求出发,优化牧场整体生产结构,增草增畜,使草地资源得到恢复性演替,才能实现生态畜牧业的建设目标. 相似文献
