The effect of thyroidectomy (Tx) and subsequent treatment with triiodothy-ronine (T3) on rat kidney mitochondrial oxidative phosphorylation was examined. Thyroidectomy resulted in lowering of state 3 respiration rates and cytochrome contents. Thyroidectomized animals administered with T3 (20 Μg/100 g body wt) resulted in the nonsynchronous stimulation of state 3 respiration rates in kidney mitochondria with glutamate, Β-hydroxybutyrate, succinate and ascorbate+TMPD as substrates. Cytoch-rome contents were also elevated differentially. Increase in the state 4 respiration rates was transient and reversible. However, primary dehydrogenases were not generally altered in the Tx and T3-treated Tx animals. The results thus indicate that the T3treatment to-Tx animals brings about differential and nonsynchronous increase in the respiratory parameters and respiratory chain components of kidney mitochondria.  相似文献

Oxidative stress is observed during aging and in numerous age-related diseases. Dietary restriction (DR) is a regimen that protects against disease and extends life span in multiple species. However, it is unknown how DR mediates its protective effects. One prominent and consistent effect of DR in a number of systems is the ability to reduce oxidative stress and damage. The purpose of this review is to comprehensively examine the hypothesis that dietary restriction reduces oxidative stress in rodents by decreasing reactive oxygen species (ROS) production and increasing antioxidant enzyme activity, leading to an overall reduction of oxidative damage to macromolecules. The literature reveals that the effects of DR on oxidative stress are complex and likely influenced by a variety of factors, including sex, species, tissue examined, types of ROS and antioxidant enzymes examined, and duration of DR. Here we present a comprehensive review of the existing literature on the effect of DR on mitochondrial ROS generation, antioxidant enzymes, and oxidative damage. In a majority of studies, dietary restriction had little effect on mitochondrial ROS production or antioxidant activity. On the other hand, DR decreased oxidative damage in the majority of cases. Although the effects of DR on endogenous antioxidants are mixed, we find that glutathione levels are the most likely antioxidant to be increased by dietary restriction, which supports the emerging redox-stress hypothesis of aging.  相似文献

2004年4月4日~5月22日,在(23.0±0.5)℃条件下研究了牙鲆(平均体重,(8.80±0.18)g)继蛋白质限制后恢复投喂对其生长的影响。整个实验期间对照组(C组)连续48d饱食投喂含能17.29kJ/g,含粗蛋白50.00%的饵料。蛋白质限制阶段(1~18d)处理组T30和T40的饵料蛋白含量分别为27.95%和40.47%,但饵料含能与对照组相同,在恢复投喂阶段(19~48d)各处理组均投喂与对照组相同的饵料。实验结果表明,在蛋白质限制阶段,饵料蛋白质水平下降显著降低了处理组鱼的体重、特定生长率、饵料湿重、干重以及能量转化率(p<0.05),而摄食率、蛋白质效率和饵料蛋白质转化率随饵料蛋白水平降低而显著升高(p<0.05)。恢复投喂结束后,处理组上述各指标均恢复至对照组水平(p>0.05)。此外,除实验结束时T30组鱼体脂肪含量显著低于对照组(p<0.05)外,整个实验期间各处理组的表观消化率及鱼体成分与对照组相比均无显著差异(p>0.05)。实验结果表明,牙鲆幼鱼继蛋白质限制后的恢复投喂阶段出现了完全补偿生长效应。  相似文献

The aim of this study is to determine the effects of early and mid-gestation nutrient restriction on maternal metabolites and foetal growth. Primiparous Angus cows were synchronized and inseminated with semen from one sire. Dietary treatments were: control to gain 1 kg/week (CON) or 0.55% maintenance energy and CP requirements (nutrient restricted; NR). A subset of dams was fed NR (n=8) or CON (n=8) from days 30 to 110 of gestation. Another group was fed CON (n=8), days 30 to 190; NR (n=7), days 30 to 110 followed by CON days 110 to 190; or CON, (n=7) days 30 to 110 followed by NR days 110 to 190. Cows were harvested at days 110 or 190 of gestation, when foetal measurements and samples were collected. Cows that were NR during days 30 to 110 or 110 to 190 of gestation lost significant BW and body condition score (P<0.001), this was associated with reduced plasma glucose during NR (P<0.002). Foetal weights, empty foetal weights, abdominal and thoracic circumferences were all reduced (P<0.03) in day 110 NR animals. Foetal perirenal adipose as a percentage of empty foetal weight was increased (P=0.01) in NR day 110 female foetuses compared with CON foetus. Maternal serum triglycerides at day 110 of gestation were decreased (P<0.05) in NR dams, whereas foetal serum triglycerides were increased (P<0.05) in response to maternal NR. Foetal weights tended to be reduced (P=0.08) in NR/CON and CON/NR v. CON/CON cattle at day 190 of gestation. Empty foetal weights, abdominal and thoracic circumferences were reduced (P⩽0.03) in NR/CON and CON/NR v. CON/CON cattle. Brain weight as a percentage of empty foetal weight was increased (P<0.001) in NR/CON and CON/NR v. CON/CON cattle. Foetal perirenal adipose as a percentage of empty foetal weight was increased (P=0.003) in NR/CON and CON/NR v. CON/CON cattle. Maternal serum triglycerides at day 190 of gestation were decreased (P<0.05) in association with maternal NR. Foetal serum triglycerides at day 190 of gestation were increased (P<0.05) in response to maternal NR during early gestation but decreased by NR in mid gestation compared with CON foetuses. The data show that maternal nutrient restriction during early or mid-gestation cause’s asymmetrical foetal growth restriction, regardless if the restriction is preceded or followed by a period of non-restriction.  相似文献

Intrauterine growth retardation (IUGR) has been linked to the development of type 2 diabetes in adulthood. We have developed an IUGR model in the rat whereby the animals develop diabetes between 3 and 6 mo of age that is associated with insulin resistance. Alterations in hepatic glucose metabolism are known to contribute to the hyperglycemia of diabetes; however, the mechanisms underlying this phenomenon have not been fully explained. To address this issue, intact liver mitochondria were isolated from IUGR and control offspring at different ages to examine the nature and time course of possible defects in oxidative metabolism. Phosphoenolpyruvate carboxykinase (PEPCK) expression was also measured in livers of IUGR and control offspring. Rates of ADP-stimulated (state 3) oxygen consumption were increased for succinate in the fetus and for alpha-ketoglutarate and glutamate at day 1, reflecting possible compensatory metabolic adaptations to acute hypoxia and acidosis in IUGR rats. By day 14, oxidation of glutamate and alpha-ketoglutarate had returned to normal, and by day 28, oxidation rates of pyruvate, glutamate, succinate, and alpha-ketoglutarate were significantly lower than those of controls. Rotenone-sensitive NADH-O2 oxidoreductase activity was similar in control and IUGR mitochondria at all ages, showing that the defect responsible for decreased pyruvate, glutamate, and alpha-ketoglutarate oxidation in IUGR liver precedes the electron transport chain and involves pyruvate and alpha-ketoglutarate dehydrogenases. Increased levels of manganese superoxide dismutase suggest that an antioxidant response has been mounted, and hydroxynonenal (HNE) modification of pyruvate dehydrogenase E2-(catalytic) and E3-binding protein subunits suggests that HNE-induced inactivation of this key enzyme may play a role in the mechanism of injury. The level of PEPCK mRNA was increased 250% in day 28 IUGR liver, indicating altered gene expression of the gluconeogenic enzyme that precedes overt hyperglycemia. These results indicate that uteroplacental insufficiency impairs mitochondrial oxidative phosphorylation in the liver and that this derangement predisposes the IUGR rat to increased hepatic glucose production by suppressing pyruvate oxidation and increasing gluconeogenesis.  相似文献

The sesquiterpene lactone, 'parthenin' the toxic principle of the allergenic weed Parthenium hysterophorus, inhibited 'state 3' respiration and stimulated 'state 4' respiration in rat liver and kidney mitochondria as well as ATPase activity in the presence of Mg2+ ions. Drosophila melanogaster is ideal for studying lifespan modulated by dietary restriction (DR) and oxidative stress, and also for screening prolongevity compounds. It is critical to measure food intake in the aforementioned studies. Current methods, however, overlook the amount of the food excreted out of the flies as feces or deposited in eggs. Here we describe a feeding method using a radioactive tracer to measure gender-specific food intake, retention and excretion in response to DR and oxidative stress to account for all the ingested food. Flies were fed a full, restricted or paraquat-containing diet. The radioactivity values of the food in fly bodies, feces and eggs were measured separately after a 24-hr feeding. Food intake was calculated as the sum of these measurements. We found that most of the tracer in the ingested food was retained in the fly bodies and < 8% of the tracer was excreted out of the flies as feces and eggs in the case of females during a 24-hr feeding. Under a DR condition, flies increased food intake in volume to compensate for the reduction of calorie content in the diet and also slightly increased excretion. Under an oxidative stress condition, flies reduced both food intake and excretion. Under all the tested dietary conditions, males ingested and excreted 3-5 fold less food than females. This study describes an accurate method to measure food intake and provides a basis to further investigate prandial response to DR and prolongevity interventions in invertebrates.  相似文献

The specific binding of [3H] dexamethasone to its receptor, activation of the hormone-receptor complexes and DNase I digestion of nuclear bound hormone-receptor complexes were studied in the liver of mice during dietary restriction (alternate days of feeding for 3 months) compared to animals fed ad libitum. Results indicate an increase of receptor level (fmol/mg protein) in the diet-restricted (DR) animals as compared to those fed ad libitum (AL). Scatchard analyses confirm the increase in the level of receptors in DR animals, while the affinity (Kd) remained same in both groups of mice. Protein slot-blot analysis also depicts the increase of the receptor level in DR fed compared to the AL fed animals. The extent of temperature- and salt-dependent activation of receptors showed no marked difference in AL- and DR-fed mice. DNase I extraction of bound hormone-receptor complexes from nuclei revealed similar pattern of digestion in both groups of animals.  相似文献

1. Kidneys were kept anoxic at 4 degrees , 20 degrees and 38 degrees . Mitochondria were then isolated and their oxidative phosphorylation and respiration were determined. 2. Under all conditions the rate of phosphate esterification was affected to a greater extent, or earlier, than oxygen consumption. 3. Glutamate and succinate were used as substrates. The depression of P/O ratio was greater for glutamate at 4 degrees , and for succinate at 20 degrees . 4. Anoxia abolished the inhibiting effect of fluoride on respiration. 5. Phosphate esterification, after anoxia, was higher in the presence of fluoride than its absence, whereas in control preparations they were the same. 6. The decrease in P/O ratio did not appear to be due to activation of adenosine triphosphatase, as activities of both Mg(2+)-and dinitrophenol-activated adenosine triphosphatases were decreased after anoxia.  相似文献
