Since its publication, it has been brought to our attention that an error was introduced during typesetting. In the text several instances of concentrations are erroneously presented as mg−1: these should be kg mg−1.

In the July 2000 issue of New Phytologist , we published the research paper entitled 'Dynamics of root systems in native grasslands: effects of elevated atmospheric CO2' by J. A. Arnone III, J. G. Zaller, E. M. Spehn, P. A. Niklaus, C. E. Wells and C. Körner. Since its publication, it has come to our attention that there are two important errors. In Table 1, for alpine grassland (Switzerland, 2480 m) in year 2 (cool), the observed soil depth should be given as 0–10 cm (as originally printed there is no soil depth given). In Table 2, the period covered should be 28 April 1994 to 6 April 1995, not 28 April 1994 to 6 April 1996, as erroneously stated.

In the January 1999 issue of New Phytologist , we published the research paper entitled 'The effect of light on the growth and reproduction of Floerkea proserpinacoides ' by Margaret F. McKenna and Gilles Houle ( New Phytol . (1999) 141 , 99–108). Since its publication, the authors have identified two important errors in the text. First, values given for leaf area ratio (LAR) and specific leaf area (SLA) have been given erroneously in units of cm2 g−1; the correct units are cm2 mg−1. Second, the values for unit leaf rate (ULR) have been given incorrectly as tenfold lower than they should be; these should be multiplied by ten to give the correct values in mg m−2 d−1.

Since its publication, an error in the published title has been brought to our attention: this refers to the ampelomyces , but the name should be Ampelomyces , meaning the genus.

New Phytologist has, throughout its history, been a forum for open debate – indeed, in his opening editorial (1902), Tansley referred to 'easy communication and discussion' as being the main ideas for the journal launch. The Internet now offers exciting new ways for sharing information, and this has challenged all information providers to reassess what they offer. It is our intention that the new Forum section in the journal should formalize our standing commitment, and encourage serious dialogue – through scientific commentary, correspondence and opinion.
As the journal approaches its hundredth year, so the need for dialogue is stronger than ever. More and more science is published, making it harder, year on year, to keep abreast of important developments. Science is also global, making discussion confined to those within one country or economic bloc to be too restrictive. And as the pressure to publish is as strong as it has ever been, so the free flow of original and unorthodox ideas can too easily become stifled.
New Phytologist , as a high-impact, high-quality journal, with its international scope and fierce independence – the Trust is not answerable to any society or commercial publisher – is a respected vehicle for such material. New Phytologist welcomes this month, and introduces, its new Managing Editor, Jonathan Ingram. He comes to New Phytologist from Elsevier Science's Trends in Plant Science , where he has worked as Assistant Editor since the highly successful launch of that magazine in January 1996.
Jonathan's introduction to plant science, at Oxford over 10 years ago, was through the excellent tuition of Vernon Butt in a traditional botany degree. He then took a DPhil with Andrew Smith and Chris Leaver, learning biochemical and molecular research methods while studying malate decarboxylation associated with Crassulacean acid metabolism. He subsequently moved to the Max Planck Institute for Breeding Research in Cologne, Germany, where he did research with Dorothea Bartels into the molecular mechanism of drought tolerance in the resurrection plant Craterostigma plantagineum , specifically the role of sucrose-phosphate synthase.
These are challenging times in science publishing, particularly as technological advances force rapid change, but there are also many exciting new opportunities. Jonathan's experience and background will help New Phytologist remain in the vanguard of plant journals, while maintaining its traditions of scientific excellence and friendly service to authors and readers, traditions carefully nurtured by his predecessor David Stribley.

Conventional cognitive testing of monkeys is time‐consuming and involves single‐caging and food or water deprivation. Here we report a novel test of global cognitive performance that can be completed in a short time period without food/water or social restrictions. Nine mazes of increasing difficulty were developed using a standard puzzle feeder, and the maze‐solving performance of ten young and five aged female cynomolgus monkeys (Macaca fascicularis) was tested. The young monkeys solved maze configurations at higher levels of difficulty and solved the first level of difficulty more quickly than aged monkeys. This task discriminated performance by age in nonhuman primates as do more conventional forms of cognitive testing and indicates that this task may be a quick and easy assessment of global cognitive function. The sex and stage of gonadal development of longfinned river eels Anguilla reinhardtii , captured from nine river catchments in New South Wales, Australia, between 1999 and 2001, were determined macroscopically. Sex was verified by histology. Histology was also necessary, however, to accurately define stages of gonadal development, particularly in individuals <600 mm in total body length. Anguilla reinhardtii displayed asynchronous gamete development. The most advanced cells present in migrating male and female A. reinhardtii were spermatocytes and pre-vitellogenic oocytes, respectively. Gonadal development stages were positively correlated with body size in both sexes. Females, however, were significantly larger than males and their gonads matured over a broader size range. Size at sexual differentiation (42–60 cm for males and 50–76 cm for females) was much larger than for most other anguillids that have been studied, with the exception of the New Zealand longfinned eel Anguilla dieffenbachii . Corresponding with its large range in size at sexual differentiation was a relatively large range in size at migration for both males (44–62 cm) and females (74–142 cm).  相似文献

Following a precultivation with pedospheric nitrogen nutrition, Ricinus plants were supplied with nitrogen solely by spraying nitrate or ammonium solution onto the leaves during the experimental period. The chemical composition of tissues, xylem and phloem exudates was determined and on the basis of the previously determined nitrogen flows (Peuke et al., New Phytologist (1998), 138 , 657–687) the flows of potassium, sodium, magnesium, calcium, chloride and ABA were modelled. These data, which permit quantification of net-uptake, transport in xylem and phloem, and utilization in shoot and root, were compared with results obtained in plants with pedospherically-supplied nitrate or ammonium and data in the literature. Although the overall effects on the chemical composition of supplying ammonium to the leaves were not as pronounced as in pedospherically supplied plants, there were some typical responses of plants fed with ammonium (ammonium syndrome). In particular, in ammonium-sprayed plants uptake and transport of magnesium decreased and chloride uptake was increased compared with nitrate-sprayed plants. Furthermore, acropetal ABA transport in the xylem in ammonium-sprayed Ricinus was threefold higher than in nitrate-sprayed plants. Additionally, concentrations of anions were more or less increased in tissues, particularly in the roots, and transport fluids. The overall signal from ammonium-sprayed leaves without a direct effect of ammonium ions on uptake and transport systems in the root is discussed.  相似文献

SYNOPSIS. Monolayer cell cultures of embryonic turkey intestine (primary) and bovine kidney (cell line, 20th passage), maintained at 40.6 and 43 C for alternating intervals of approximately 12 hours in Basal Medium Eagle and fetal calf serum at pH 7.0–7.4, were observed for 144 hours after inoculation of Eimeria meleagrimitis sporozoites.
In turkey intestine cultures, which consisted of fibroblast-like cells and patches of epitheliul-like cells, there were decreases of 80 and 81% in the numbers of parasites between 5 and 48 hrs; in bovine cultures, 21–41% decreases. Decreases in the turkey cultures, however, were due to the nonsurvival of sporozoites in fibroblast-like cells; in epitheliul-like cells there was a 42% dcrease between 5 and 48 hrs and only 27% between 48 and 144 hours.
Trophozoites were present in bovine cells at 5 hrs. Small, mature schizonts containing only 12-28 merozoites were present in the bovine cultures and in the epitheliul-like cells within turkey intestine cultures from 48-144 hrs. Larger schizonts (50-115 by 20-70 μ) were present in bovine but not in turkey cultures from 72–144 hrs. Many of these schizonts contained far more merozoites than schizonts of any of the 3 generations described from the host.
In bovine cultures, there was an abundance of liberated merozoites at 50, 52, 74, and 76 hrs; many had reinvaded cells, sometimes as many as 50–60 per cell. In turkey cultures, liberated merozoites were found once at 144 hrs and none were intracellular. At 120 and 144 hrs in bovine cultures, abnormally developed and degenerate forms appeared; in turkey cultures, all were normal.  相似文献

Secretory carrier membrane protein 2 (SCAMP2) functions in late steps of membrane fusion in calcium-dependent granule exocytosis. A basic/hydrophobic peptide segment within SCAMP2 (SCAMP2 E: CWYRPIYKAFR) has been implicated in this function and shown to bind and sequester phosphatidylinositol 4,5-bisphosphate [PI(4,5)P2 or PIP2] within membranes through an electrostatic mechanism. We now show that alanine substitution of tryptophan W2 within SCAMP2 E substantially weakens peptide binding to negatively charged liposomes; other substitutions for arginine R4 and lysine K8 have only limited effects on binding. Electron paramagnetic resonance analysis of liposomes containing spin-labeled PIP2 shows that R4 but not K8 is critical for SCAMP E binding to PIP2. The interfacial locations of SCAMP E and its structural variants within lipid bicelles measured by oxygen enhancement of nuclear relaxation are all similar. Corresponding point mutations within full-length SCAMP2 (SC2-R204A, SC2-K208A, and SC2-W202A) have been analyzed for biological effects on dense core vesicle exocytosis in neuroendocrine PC12 cells. With the same level of overexpression, SC2-R204A but not SC2-K208A inhibited secretion of cotransfected human growth hormone and of noradrenalin. Inhibition by SC2-R204A was the same as or greater than previously observed for SC2-W202A. Analysis of noradrenalin secretion by amperometry showed that inhibitory mutants of SCAMP2 decrease the probability of fusion pore opening and the stability of initially opened but not yet expanded fusion pores. The strong correlation between SCAMP2 E interactions with PIP2 and inhibition of exocytosis, particularly by SC2-R204A, led us to propose that SCAMP2 interaction with PIP2 within the membrane interface regulates fusion pore formation during exocytosis.  相似文献

Abstract. The present investigation was designed to analyse the effects of acute lethal graft-versus-host disease (GVHD) in adult (DA x LEW)F1 rats on cellular proliferation within the liver. The influence of the host thymus on GVHD-induced proliferation was also assessed. From 1–28 days after initiation of GVHD [3H]thymidine ([3H]-TdR) was injected i.v. and rats were killed one hour later. Percentage labelled cells (LI) of periportal infiltrating cells (PIC), hepatocytes (H), and sinusoidal lining cells (SC) were counted. Mean values for control rats were 0.3 ± 0.1% (H), 0.4 ± 0.1% (SC) and 0.2 ± 0.1% (PIC). GVHD rats demonstrated a significant increase in LI of PIC (days 1–21), SC (days 2–17) and H (days 2–17). Most labelled cells in PIC were large lymphocytes. Peak LI values were 7.0 ± 1.0% PIC (day 17), 6.8 ± 0.9% SC (day 17), and 5.2 ± 0.9% H (day 7), with all cellular compartments returning to near normal LI values by day 28. Stimulation of cellular proliferation occurred in all three liver cell compartments in neonatally thymectomized (TXM) rats. The intensity of GVHD-induced cell proliferation was significantly decreased at day 7 in all compartments and PIC was dramatically decreased at day 21 in TXM-GVHD rats as compared to non-TXM-GVHD rats. It is hypothesized that the general stimulation of hepatocyte cell proliferation in GVHD is related to the secretion of lymphokines by primarily donor and secondarily host T cells in the periportal infiltrate.  相似文献
