Proton metabolic profiling of incisional biopsied cervical lymph node tissue specimens of 109 patients suffering from tubercular (CTBL) and non-specific (NSCLA) lymphadenitis were analyzed by high resolution magic angle spinning (HR-MAS) NMR spectroscopy. In the present study, 40 endogenous metabolites namely, myo-inositol (m-Ins), branched chain amino acids (BCAA), glutamate, serine, taurine (Tau) aromatic amino acids, choline (Cho) containing compounds and glucose were characterized. To the best of our knowledge, this is the first report on metabolic profiling of cervical tubercular lymph node tissues using HR-MAS NMR spectroscopy. The principal component analysis revealed a clear discrimination between CTBL and NSCLA tissues. Increase in the concentration of mobile poly unsaturated fatty acids, BCAA, Cho, Tau, glycine and a decrease in the concentration of lactate, phosphocholine and m-Ins was observed in CTBL cases. The partial least square discriminant analysis (PLS-DA) with R 2 = 0.95 and Q 2 = 0.92 provided >98 % of correct classification between the two groups. A PLS-DA training set model of 75 % (CTBL = 54, NSCLA = 27) of the subjects when subjected for prediction of 25 % cases (CTBL = 18, NSCLA = 10) as an unknown dataset provided more than 98 % of diagnostic accuracy in their respective histological categories. The receiver operator characteristic curve was generated from PLS-DA factor-1 projected an area under the curve of 0.962. The metabolic profile obtained from HR-MAS NMR spectroscopy may be used as surrogate markers in vivo MRS for differentiating between CTBL and NSCLA cases non-invasively.  相似文献

Seed development in Jatropha curcas L. was studied with respect to phenology, oil content, lipid profile and concentration of sterols. Seeds were collected at various stages of development starting from one week after fertilization and in an interval of five days thereafter till maturity. These were classified as stage I to stage VII. Moisture content of the seeds ranged from 8.8 to 90.3%; the lowest in mature seeds in stage VII and highest in stage I. The seed area increased as the seed grew from stage I to stage VI (0.2-10.2mm(2) per seed), however, the seed area shrunk at stage VII. Increase in seed area corresponded to increase in fresh weight of the seeds. (1)H NMR spectroscopy of hexane extracts made at different stages of seed development revealed the presence of free fatty acids (FFA), methyl esters of fatty acids (FAME) and triglycerol esters (TAG), along with small quantity of sterols. The young seeds synthesized predominantly polar lipids. Lipid synthesis was noticed nearly three weeks after fertilization. From the fourth week the seeds actively synthesized TAG. Stage III is a turning point in seed development since at this stage, the concentration of sterols decreased to negligible, there was very little FAME formation, accumulation of TAG increased substantially, and there was a sudden decrease in FFA concentration. The findings can be helpful in understanding the biosynthesis and in efforts to improve biosynthesis of TAG and reduce FFA content in the mature seeds.  相似文献

麻疯树(JatrophacurcasL.)的乳汁管能分泌含有多种药用成分的乳汁。采用石蜡切片、冰冻切片和半薄切片技术,在光学显微镜下观察麻疯树乳汁管的类型、分布和大小。结果表明,麻疯树中存在有节连接和非连接乳汁管、无节分枝和不分枝乳汁管;乳汁管普遍分布在皮层和韧皮部,茎和叶脉的木质部存在少量的乳汁管,茎、叶柄、花梗和果柄的髓部有少量分布;靠近维管组织的皮层中的乳汁管直径较大,而靠近表皮的皮层中的乳汁管直径较小,如茎中靠近维管组织的乳汁管直径为40-60μm,而靠近表皮的乳汁管直径为5-18μm。  相似文献

Microalgae are a rich source of high value compounds such as carbohydrates, lipids, proteins and bioactive compounds. In particular, microalgae have been identified as a potentially important resource for carbon-capture and as a feedstock for green biofuels. Successful cultivation of microalgae can occur under a variety of nutrient and environmental conditions with each condition producing a unique distribution of compounds. In order to steer the cultivation towards a particular distribution of compounds, rapid and accurate methods for compound identification are required. Current methods for determining the absolute quantity of each component are time consuming and arduous making cultivation optimization impractical. High-resolution magic angle spinning (HR-MAS) nuclear magnetic resonance (NMR) spectroscopy offers a robust and rapid screening method capable of ascertaining the absolute quantity of each component with minimal sample manipulation. Sample preparation consists of harvested, centrifuged and freeze-dried whole-cell Nannochloropsis granulata from large-scale photobioreactors being accurately weighed and rehydrated with deuterium oxide and placed in an HR-MAS rotor. One-dimensional HR-MAS NMR spectra were recorded under quantitative conditions to determine the lipid and carbohydrate profile of the microalgae. The total time per sample for preparation, data acquisition and analysis was approximately 1 h. Changes in resonance profiles corresponding to varying proportions of saturated and polyunsaturated fatty acids were correlated to the time of harvest. In addition, standard two dimensional experiments were used to identify the major carbohydrate components. HR-MAS NMR spectroscopy has been used to profile the lipid and carbohydrate content of N. granulata and we have begun to establish methodologies for quality analysis/quality control for cultivation of various microalgal strains.  相似文献

用透射电镜观察了麻疯树(Jatropha curcas L.)小孢子发育的超微结构。小孢子母细胞时期内质网和质体较多;减数分裂和四分体时期,细胞处于明显的代谢活跃状态,细胞器丰富,主要有内质网、线粒体、质体、高尔基体和球状体;在小孢子发育早期和晚期,线粒体和内质网仍较丰富;小孢子经过高度的不对称分裂后,形成较大的营养细胞和较小的生殖细胞,营养细胞中细胞器数量明显减少,含大量的淀粉和脂类物质,生殖细胞中脂类物质丰富;表皮、药室内壁和中层细胞在小孢子母细胞和四分体时期淀粉粒丰富,小孢子时期明显减少,绒毡层从小孢子母细胞至小孢子发育晚期的细胞器都很丰富,主要为内质网、质体和线粒体,为二胞花粉发育奠定基础。  相似文献

Magnetic resonance imaging (MRI) was used to study temporal and spatial water uptake and distribution in germinating lupine ( Lupinus luteus L.) seeds. During 24 h of imbibition, water was unevenly distributed within the seed and some anatomical parts were more hydrated than others. Water entered the seed through the hilum and micropyle. The embryonic axis was the first to show hydration followed by seed coat and later cotyledons. The changes in water status were characterized by NMR spectroscopy. Analyses of T2 relaxation times revealed a three-component water proton system (structural, intracellular and extracellular water) in germinating lupine seeds. The data on the components of transverse relaxation time studies indicated the complex exchange processes taking place between water components inside lupine seed over first 2.5 h of hydration, with a distinguished increase in structural water and decrease in other components. This speaks in favor of the high water-absorbing capacity of lupine seeds as related to high protein content. Germination was accompanied by swelling of protein bodies and changes in the organization of stored reserves with gradual disappearance of protein from the cells.  相似文献

By reviewing the existing 1H‐magnetic resonance spectroscopy literature in schizophrenia, the relationship of different sample characteristics and applied methodologies with metabolite alterations is explored. Furthermore, we emphasize common pitfalls and discrepancies in the methodological framework of the reviewed studies that introduce unwanted variation in findings and complicate the comparison of studies. A total of 92 studies were reviewed. Articles were retrieved by searching the Pubmed database. Care was taken to note down reliability and validity measures of each included study. Despite many methodological differences and shortcomings, progressive NAA reductions could be seen in several brain regions implicated in the pathogenesis of schizophrenia. In terms of treatment effects, cross‐sectional evidence implicates a normalizing role for atypical antipsychotic medication; however, longitudinal studies remain inconclusive on this issue. Choline, creatine, and myo‐inositol levels remain largely unchanged and a time‐dependent role of glutamate finds confirmation in several spectroscopy studies. Other findings are less consistent and need further replication. Most studies lack power and methodological precision. Future studies should aim for standardization and for more distinguished study populations to gain more valid and reliable findings.  相似文献

Two new esterases (JEA and JEB) and a lipase (JL) were extracted from the seeds of Jatropha curas L. Lipase activity was only found during germination of the seeds and increased to a maximum after 4 days of germination. All enzymes were found to be most active in the alkaline range at around pH 8 and the purified (fractionated precipitation with ethanol and gel filtration) esterases were very stable at high temperatures. The molecular weight (SDS-PAGE) of both esterases was determined to be 21.6-23.5 kDa (JEA) and 30.2 kDa (JEB) and the isoelectric point was 5.7-6.1 for esterase JEA and 9.0 for esterase JEB. Most ions caused a negative influence on the activity of both esterases. Using p-nitrophenyl butyrate as a substrate JEA showed a K(m) of 0.02 mM and a v(max) of 0.26 micromol mg(-1) min(-1). Under the same conditions JEB showed a K(m) of 0.07 mM and a v(max) of 0.24 micromol mg(-1) min(-1). Both esterases hydrolyzed tributyrin, nitrophenyl esters up to a chain length of =C4 and naphtylesters up to a chain length =C6. In transesterification reactions, JL was found to be most active at very low water activities (0.2) and in high water activities, the lipase hydrolysed triglycerides into conversions above 80%. The lipase hydrolysed both short chain and long chain triglycerides at about the same rate but was inactive on alpha-methylbenzyl acetate. JL is a potentially useful biocatalyst in the hydrolysis of triglycerides in organic solvents.  相似文献

麻疯树幼苗对干旱胁迫的响应   总被引:18,自引:0,他引:18  
以不同浓度(5%~25%)的聚乙二醇(PEG-6000)模拟干旱胁迫处理麻疯树三叶期幼苗,研究了不同程度干旱胁迫下麻疯树叶片光合特性及其对干旱的耐受能力.结果表明:在较低浓度PEG(≤15%)处理下,随PEG浓度的增加,麻疯树叶片净光合速率(Pn)、气孔导度(Gs)、胞间CO2浓度(Ci)、PSⅡ实际光化学量子产量(ΦPSⅡ)、光化学猝灭(qP)和表观光合电子传递速率(ETR)下降,PSⅡ原初光能转化效率(Fv/Fm)轻微下降,水分利用效率(WUE)则逐渐升高,非光化学猝灭(NPQ)明显上升,初始荧光(Fo)无显著变化(P>0.05);在高浓度PEG(>15%)处理下,Ci随PEG浓度的增加而显著上升,Pn、Gs和WUE持续下降,Fv/Fm、ΦPSⅡ、qP和ETR下降幅度明显增大,Fo显著上升,而NPQ下降.低浓度PEG处理导致麻疯树叶片Pn下降主要是由气孔因素造成的;在高浓度PEG处理下,Pn的下降则是由非气孔和气孔因素的共同限制作用造成的.当PEG浓度<20%时,虽然出现Pn下降,但光合机构未受损伤.经15 d高浓度PEG处理的植株叶片,在胁迫解除后光合活性能够迅速恢复,且植株可以存活.说明麻疯树对干旱胁迫有较强的耐受能力.  相似文献

Variation is the phenomenon where individuals of a population differ from each other. The variation or diversity can have morphological manifestation or genetic basis. Individual identification based on morphological record is the most common practice in Jatropha. Therefore, in order to find a proper method for estimation of genetic diversity and genetic relationships among different germplasm of Jatropha curcas L., random amplified polymorphic DNA (RAPD) technique based on polymerase chain reaction (PCR) was used for described purpose. Out of 55 decamer primers tested, 26 primers produced good amplification products. A total of 6,011 amplification products were scored from which only 1,859 bands (30.92%) were found to be polymorphic and the size of bands ranged from 300 to 2,500 bp. Unweighted pair group method using arithmetic average cluster analysis revealed clear genetic difference among J. curcas germplasm. The scientific data presented in this study suggests that RAPD-PCR could be used as a valuable tool for estimation of genetic diversity and genetic relationship among germplasm of J. curcas L.  相似文献

Magnetic resonance imaging (MRI) is a well known diagnostic tool in radiology that produces unsurpassed images of the human body, in particular of soft tissue. However, the medical community is often not aware that MRI is an important yet limited segment of magnetic resonance (MR) or nuclear magnetic resonance (NMR) as this method is called in basic science. The tremendous morphological information of MR images sometimes conceal the fact that MR signals in general contain much more information, especially on processes on the molecular level. NMR is successfully used in physics, chemistry, and biology to explore and characterize chemical reactions, molecular conformations, biochemical pathways, solid state material, and many other applications that elucidate invisible characteristics of matter and tissue. In medical applications, knowledge of the molecular background of MRI and in particular MR spectroscopy (MRS) is an inevitable basis to understand molecular phenomenon leading to macroscopic effects visible in diagnostic images or spectra. This review shall provide the necessary background to comprehend molecular aspects of magnetic resonance applications in medicine. An introduction into the physical basics aims at an understanding of some of the molecular mechanisms without extended mathematical treatment. The MR typical terminology is explained such that reading of original MR publications could be facilitated for non-MR experts. Applications in MRI and MRS are intended to illustrate the consequences of molecular effects on images and spectra.  相似文献

能源短缺和环境污染是当前人类社会所面临的巨大挑战,生物柴油的推广和应用是现阶段解决替代燃油的重要途径之一。黑皂树种子油是很好的生物柴油原料,目前在许多热带地区大力种植黑皂树。我国华南、西南地区的一些干旱荒地可以种植黑皂树。我们对黑皂树油在柴油机上的应用研究证明:黑皂树油在气温35℃条件下,可以直接在各种柴油机上起动和应用;黑皂树油中掺入20%的轻柴油,其黏度大大降低,冷机起动性能和热效率等均与轻柴油相当。因此,黑皂树油的应用比目前国外使用的生物柴油更经济,是一种很有发展前途的能源植物。  相似文献

An efficient root induction system has been established for in vitro-regenerated Jatropha curcas L. shoots. Callus formation on shoots transferred to auxin containing medium was found to be a prominent and recurrent problem for rooting of in vitro-cultivated J. curcas. In particular, the type of auxins and cytokinins applied in the culture media were shown to strongly influence the severity of callus formation. Shoots cultivated on meta-methoxytopolin riboside (MemTR) were free of callus and produced elongated stems and well-developed leaves in comparison to the cytokinins benzyl adenine, zeatin, and thidiazuron. Subsequent root induction experiments were performed with shoots precultured on MemTR-containing medium. Shoots were excised and transferred to Murashige and Skoog (MS) medium supplemented with different concentrations of indole-3-butyric acid (IBA), indole-3-acetic acid (IAA), and α-naphtaleneacetic acid (NAA). The induction of excessive callus formation was avoided only on IBA-containing medium. The optimum rooting medium with good root induction (35%) and 1.2 roots per shoot contained half-strength MS salts supplemented with 2.5 μM IBA. The same medium supplemented with 0.25% (w/v) activated charcoal produced 46% rooted shoots. Further improvement of rooting was obtained by transferring in vitro grown shoots to woody plant medium containing phloroglucinol (PG). In the presence of 2.5 μM IBA and 238 μM PG, 83% of the shoots rooted with on average 3.1 roots per shoot. We also analyzed the impact of light quality on the rooting capacity of Jatropha in vitro grown shoots. In general, light-emitting diodes (LEDs) light sources were less efficient for root induction. Red LED light provided the most favorable growth conditions, inducing a rooting response in 65% of the shoots, which produced on average 5.5 roots per shoot. These results indicate that adventitious rooting in J. curcas is under control of photoreceptors and that optimal rooting requires fine-tuning of the salt concentration, auxin, and cytokinin balance and application of synergistic compounds.  相似文献

Jatropha curcas shows invasive characters and is a significant source of many phytochemicals with varying biological activities. Different plant parts of Jatropha curcas L exhibited variation in their phytochemical constituents. Leaves and ovary walls were found to contain higher contents of total phenols, tannins and phytic acid whereas free amino acids were greater in leaves. Young leaves of Jatropha show greater contents of all these metabolites. Further, plants exhibit seasonal differences as leaves collected during summer (May-June) have greater accumulation of total phenols, tannins and free amino acids however, phytic acid was more during rainy season. Leachates and extracts in their higher concentrations adversely affected the germination and growth of wheat seedlings however, lower concentrations were more or less stimulatory. These treatments not only decreased the length, fresh and dry weight of seedlings but also affected the chlorophyll contents and activity of enzymes such as nitrate reductase, aminotransferases in wheat seedlings however, the activity of superoxide dismutase and ascorbate peroxidases increased. Experiments indicate harmful allelopathic effects of Jatropha leachates /extracts on wheat seedlings, hence further experimentation and analysis is recommended before continued plantation of Jatropha particularly on fertile soils. However. Growth of Jatropha plants on saline soils and their potential for accumulating sodium, potassium and chloride are the attributes suggesting the possibility of use of Jatropha plants in improving saline soils.  相似文献

Jatropha curcas L. (jatropha) is a species identified for biofuel production because of the high quality of the oil produced by its seeds. However, jatropha is undomesticated and little information is available about its reproductive characteristics. Breeding and genetic improvement programs are much needed for the jatropha development as a bioenergy crop. Information about floral display and mode of reproduction are considered essential for breeding programs. In this study, the total number of female flowers, male to female flower ratio, fruit set, in vitro pollen germination, and the formation of fruits by apomixis, self‐pollination, and natural pollination were evaluated in 17 jatropha accessions planted in South Florida. The total number of female flowers per inflorescence and male to female flower ration ranged from 2.8 to 9.1 and 9.9:1 to 55.4 : 1, respectively. During summer 2011, high fruit setting average was observed (75.5%). In vitro pollen germination varied from 64.6% during spring 2011 to 51.6% during fall 2011. The fruit set observed was from 10.1% to 64.0% through natural pollination and apomixes, respectively. Characteristics such as fruit fresh weight, number of seeds per fruit, seed dry weight, and oil content were influenced by mode of reproduction.  相似文献
