Illus. 1961.—Plasmodesmata, called ectodesmata, in the outer walls of epidermal cells, have been investigated. Their occurrence and distribution in the epidermis of leaves of Plantago major and Helxine soleirolii have been examined in connection with foliar absorption. Leaf structures such as guard-cells, conical hairs, anticlinal walls and the epidermal cells adjacent to the leaf veins have been shown consistently to contain large numbers of ectodesmata, while in neighboring cells ectodesmata may be low in number or lacking. The same areas also are known to be especially pervious to water and dissolved dyes applied to the surface of the leaf. From special investigations, it appears that certain solutions that form visible crystals and precipitates in the outer wall enter the epidermal cell wall in localized pathways. The localization of these bodies coincides with that of ectodesmata. Therefore, it is concluded that the ectodesmata may be the pathways for transport of substances from the outside to the interior of tissues and vice versa. Nutrients applied to the surface of leaves are thought to enter by the same pathways, i.e., the ectodesmata, as those in which the penetration can be visibly detected. Some phenomena of foliar absorption which confirm this theory are explained in connection with the presence of ectodesmata.  相似文献

Summary The outer wall of adaxial epidermal cells of onion bulb scales shows punctate callose deposits. The abundance of these callose deposits is highly variable, and seems to correlate with the distribution of ectodesmata. Several peptides, including insulin, epidermal growth factor and vasoactive intestinal polypeptide bind to intestinal epithelial cells. However, it is unclear whether one binding site binds several peptides or whether separate sites exist for each peptide. These studies were designed to examine the specificity of peptide binding sites on intestinal epithelial cells. Peptide binding was measured directly with [125I]radiolabelled peptides to isolated enterocytes prepared from rabbit ileum. The characteristics of insulin and epidermal growth factor binding were similar. Both insulin and epidermal growth factor specific binding was saturable, directly correlated to cell concentration and temperature and pH dependent. The total number of insulin binding sites per cell was 4500, that for epidermal growth factor was 2280. Scatchard analysis for both peptides produced curvilinear plots. Dissociation of both peptides from the binding site was increased in the presence of their respective unlabelled peptide. However, insulin specific binding was not altered by epidermal growth factor, and epidermal growth factor specific binding was unaffected by insulin. Further, both insulin and epidermal growth factor failed to inhibit the specific binding of vasoactive intestinal polypeptide to ileal enterocytes, and vasoactive intestinal polypeptide did not inhibit insulin or epidermal growth factor specific binding. These studies demonstrate that insulin, epidermal growth factor and vasoactive intestinal polypeptide interact with three distinct membrane binding sites on the enterocyte.  相似文献

Summary When droplets of radioactive solutions are applied to the cuticle, micro-autoradiograms show that the guard cells and the anticlinal walls of the leaf epidermis ofSpinacia oleracea and ofViola tricolor are the favoured sites of absorption. Scattered black spots in the stripping film could also be observed above the periclinal walls of the epidermis.A comparison of such micro-autoradiograms with the distribution of the ectodesmata in the leaf epidermis of the same two species demonstrates that the areas of favoured foliar absorption correspond to the areas of the densest accumulation of the ectodesmata, while the ectodesmata in the less absorbing periclinal walls are more loosely dispersed. Thus, though ectodesmata cannot be localized exactly by the occurrence of silver grains in the stripping film, identity exists between the areas of foliar absorption and the distribution of ectodesmata in the leaves which proves that ectodesmata are functioning as pathways of transport in foliar absorption.The experiments reported cofirm earlier statements that guard cells participate essentially in foliar absorption. The micro-autoradiograms also clearly show that absorption does not take place through the pores of the guard cells.

Attenuation of UV radiation by plant cuticles from woody species   总被引:8,自引:0,他引:8  
Transmittance spectra of isolated plant cuticles were measured in the wavelength range from 270 to 600 nm. The cuticles were enzymatically isolated from the leaves of 27 species (26 evergreen or deciduous woody, one succulent herbaceous) and from four species of fruits. With the exception of subtropical and tropical species all plants were cultivated in the field. The cuticles of the species studied strongly attenuated ultraviolet (UV) radiation at wavelengths < 400 nm while they were practically translucent in the visible range. Relatively broad transmittance minima occurred at wavelengths from 280 to 320 nm (UV-B). Spectral transmittances at 300 nm ranged from 0.004 (Ilex aquifolium) to 0.50 (Prunus avium) for leaf cuticles and from 0.00023 (Cydonia oblonga) to 0.005 (Mains domestica) for fruit cuticles. The constitutive UV protection by cuticular pigments may be supplemented, to varying degrees, by pigments located in the epidermal cell wall and protoplast. Thus, it is concluded that only a small fraction of incident UV-B radiation may actually reach the sensitive tissues of the leaves of non-herbaceous species and of fruits.  相似文献

Dewaxed thin-sectioned and dewaxed isolated mature fruit cuticlesrevealed the unequivocal presence in situ of visibly discrete,ubiquitous, cuticular pores or orifices concomitant with anticlinally-orientedtranscuticular canals in 51 varieties of fruit among 20 plantfamilies. More than 66 per cent of the fruit cuticles have poresand/or canals. No correlation exists between either fruit sizeor pore size and cuticle thickness. Dewaxed cuticles rangedfrom 1.25–22.5 µm in thickness. Canal lengths aredirectly related to cuticle thickness. We report a formerly unknown mechanism in seeds which improvesgermination under relatively dry conditions. During seed developmentof several South American cacti, epidermal cells produce proteinaceousmaterial that appears to pass through ectodesmata in the outercell wall and which accumulates under the cuticle. Once moistened,this secretory layer readily absorbs water and distributes itover the seed surface. It thus improves water uptake and ensuresgermination with the minimum amount of available water, whichmay be advantageous in (semi-)arid regions. In experiments withseeds ofEchinopsis thionantha(Speg.) Werd. andGymnocalyciumgibbosum(Haw.) Pfeiff. under water stress, intact seeds tookup significantly more water and germinated better than seedsfrom which the hydrophilous layer had been artificially removed. Light and transmission electron microscopy revealed thatTyphula ishikariensis penetrated into bentgrass leaves either through cuticles or stomata either by single hyphae or infection cushions formed on host surfaces. Time course study on infected leaves showed that penetration through stomatal subsidiary cells and their adjacent cells seemed to occur earlier than that through epidermal cells located farther from stomata. More than 30% of epidermal cells were infected by 10 days after inoculation. When hyphae penetrated through an intact cuticle of epidermal cells, they seemed to dissolve host cell walls enzymatically at penetration sites. Plant cuticles have been traditionally classified on the basis of their ultrastructure, with certain chemical composition assumptions. However, the nature of the plant cuticle may be misinterpreted in the prevailing model, which was established more than 150 years ago. Using the adaxial leaf cuticle of Ficus elastica, a study was conducted with the aim of analyzing cuticular ultrastructure, chemical composition and the potential relationship between structure and chemistry. Gradual chemical extractions and diverse analytical and microscopic techniques were performed on isolated leaf cuticles of two different stages of development (i.e. young and mature leaves). Evidence for the presence of cutan in F. elastica leaf cuticles has been gained after chemical treatments and tissue analysis by infrared spectroscopy and electron microscopy. Significant calcium, boron and silicon concentrations were also measured in the cuticle of this species. Such mineral elements which are often found in plant cell walls may play a structural role and their presence in isolated cuticles further supports the interpretation of the cuticle as the most external region of the epidermal cell wall. The complex and heterogeneous nature of the cuticle, and constraints associated with current analytical procedures may limit the chance for establishing a relationship between cuticle chemical composition and structure also in relation to organ ontogeny.  相似文献

Summary Lambertz (1954) did not mention Gramineae as plants containing ectodesmata, although he stated that ectodesmata are not bound to special groups of angiosperms but should occur in all families of angiosperms. Schnepe (1959) expressly pointed out that he never succeeded in demonstrating such structures in leaves of Gramineae with his method. In our studies, however, ectodesmata could be shown in leaves of wheat and maize if the fixation mixture contained nitric acid. As in other objects, including species of gymnosperms and pteridophytes, the same distribution of ectodesmata could be observed which was characterized by the formation of rows along the anticlinal walls, crowding in guard-cells and seattering in periclincal walls.  相似文献

The pattern of cuticular protein synthesis by the epidermis of the tobacco hornworm larva changes during the final day of feeding, leading to an alteration in cuticular structure and a stiffening of the cuticle. We have isolated a small multigene family which codes for at least three of the new cuticular proteins made at this time. The five genes which were isolated from this family map to two different genomic regions. Sequencing shows that one of the genes is 1.9 kb and consists of three exons coding for a 12.2-kDa acidic (pI = 5.26) protein that is predominantly hydrophilic. The deduced amino acid sequence shows regions of similarity to proteins from flexible lepidopteran cuticles and from Drosophila larval and pupal cuticles, but not to proteins found in highly sclerotized cuticles. This gene family is first expressed late on the penultimate day (Day 2) of feeding in the final larval instar and ceases expression 2 days later when metamorphosis begins. In situ hybridization shows that this gene family is expressed in all the epidermal cells of Day 3 larvae except the bristle cells and those at the muscle attachment site. Expression can be induced in Day 1 epidermis by exposure to 50 ng/ml 20-hydroxyecdysone in vitro, but only if juvenile hormone is absent. Its developmental expression, tissue specificity, and hormonal regulation strongly suggest that this multigene family is involved in the structural changes that occur in the larval cuticle just prior to the onset of metamorphosis.  相似文献

The in vivo adminstration of l-triiodothyronine to normal adult rats produced a reduction in the number of binding sites in hepatic membranes for epidermal growth factor; hyperthyroidism had no effect on insulin binding. Leaf internal cuticle has not previously been studied in detail, and yet its existence has profound implications for the path of water movement. The internal cuticle forms a uniform layer on the inner periclinal epidermal walls that border substomatal cavities. This cuticle is continuous with the external cuticle through the stomatal pores. The thickness of the internal cuticle on nonstomatal epidermal cells is approximately one-third that of the external cuticle on the same cells. On both the abaxial and adaxial sides of the leaf the internal cuticle forms irregularly shaped islands bordered by mesophyll cells. The size of the islands coincides with the epidermal area of the substomatal cavity. The internal cuticle remains intact and connected to the external cuticle after incubation in cellulytic enzymes. After treatment with sulfuric acid or chloroform, both cuticles remain intact. The autofluorescence of both cuticles is increased by staining with auramine O. These results indicate that large portions of the leaf epidermis are covered by both an internal and an external cuticle.  相似文献

利用光学显微镜和扫描电子显微镜技术,对臭椿(Ailanthus altissima(Mill.)Swingle)叶片从叶芽到成熟叶4个发育阶段(叶芽、幼叶、近成熟叶和成熟叶)的4种叶表面结构(表皮毛、气孔器、角质层、腺体)进行了详细地观察和比较。结果显示:叶片上、下表面具单细胞非腺毛和头状腺毛两种表皮毛,其密度在叶芽阶段最大,之后随着叶片的生长和展开表皮毛密度逐渐降低;气孔器主要分布于下表面,叶片近成熟时气孔器密度最大;角质层具有条纹状结构,其隆起程度在近成熟叶阶段达到最大;叶片基部两侧具锯齿状突起,每个突起上有一个腺体,腺体发育成熟后可分泌透明黏液。说明叶片表面各结构特征是臭椿对环境长期适应的结果。  相似文献

Binding sites for calcium and chloride ions, and urea on cuticular membrane surfaces were identified by microautoradiography. There was no localization of binding on either the smooth outer or irregular inner surfaces of the astomatous tomato fruit cuticle, and urea was bound to a lesser degree than inorganic ions. Calcium and chloride binding sites on surfaces of green onion leaf cuticles were closely associated with stomatal pores and above the periclinal cell walls. Binding of calcium was more pronounced than of chloride. These results are discussed in light of possible sites of entry for foliar applied nutrients.  相似文献

Bacteria were either isolated from leaf surfaces of Hedera helix or obtained from a culture collection in order to analyse their effect on barrier properties of isolated Hedera and Prunus laurocerasus cuticles. On the basis of the 16S rDNA sequences the genera of the six bacterial isolates from Hedera were identified as Pseudomonas sp., Stenotrophomonas sp. and Achromobacter. Water permeability of cuticles isolated from H. helix was measured before and after inoculation with the six bacterial strains. In addition water permeability of cuticles isolated from P. laurocerasus was measured before and after inoculation with the three bacterial strains Pseudomonas aeruginosa, Xanthomonas campestris and Corynebacterium fascians. Rates of water diffusing across isolated cuticles of both species significantly increased by up to 50% after inoculation with all bacterial strains. Obtained results show that epiphytic bacteria have the ability of increasing water permeability of Hedera and Prunus cuticles, which in turn should increase the availability of water and dissolved compounds in the phyllopshere. Consequently, living conditions in the habitat phyllosphere are improved. It can be concluded that the ability to change leaf surface properties will improve epiphytic fitness of leaf surface bacteria.  相似文献

An homogeneous cell population isolated from the inguinal tissue of 3-day-old rats is able to proliferate in primary culture. In the presence of a physiological concentration of insulin (1.5 nM) it converts into cells exhibiting the morphology and the biochemical characteristics of adipocytes. Insulin and epidermal growth factor (EGF) receptors were studied during both the exponential growth and the adipose conversion phases of these cells. Binding experiments with 125I-labelled peptides were performed directly in the culture dishes. The number of high affinity insulin binding sites increased, during the entire culture period studied, reaching 18 days after plating the value of 10,600 x 2360. Control cells (cultured in the presence of anti-insulin antibody) exhibited an increase of the concentration of insulin binding sites from no more than 500 sites/cell to 6880 +/- 1710 sites/cell between dat 0 and 9 (corresponding to the exponential growth phase); this increase was followed by a rapid reduction in insulin receptors during the stationary phase. The density of EGF binding sites increased between day 0 and 4 (one cell cycle), whether the cells were maintained or not with insulin, and plateaued thereafter. Mature adipocytes freshly isolated from the inguinal tissue of 3-day-old rats had no detectable EGF binding sites, but their content in high affinity binding sites for insulin was similar to that of cells after complete adipocyte conversion in primary culture.  相似文献

KOVACH, W. L. & DILCHER, D. L., 1984. Dispersed cuticles from the Eocene of North America. Macerations of organic-rich clay from the Claiborne Formation (Middle Eocene) of Tennessee have yielded a wide variety of well-preserved dispersed cuticles. Details of the epidermal cell patterns, arrangement of the stomata1 complex, trichomes and trichome attachment have made possible the association of some dispersed cuticles with leaf types known from this formation which have similar cuticle, and with modern families. All dispersed cuticles are classified in a morphologic system. Through both our own work and a review of previous investigations we have found that distinct dispersed cuticle types can be recognized and may be used biostratigraphically to characterize geologic strata, palaeo-ecologically to provide insights into environmental reconstruction, and systematically to follow the history of certain taxa.  相似文献
