昆虫病原线虫是一种有应用潜力的地下害虫生物防治因子。本研究利用形态学特征和ITS-rDNA分析方法对从河北省沧州分离的异小杆线虫一品系进行了鉴定, 并在室内测定比较了其对蛴螬的致病力。通过对该线虫侵染期幼虫和雄性成虫主要形态学特征的参数测量, 发现其与嗜菌异小杆线虫Heterorhabditis bacteriophora的形态特征最为相近;同时ITS1-rDNA序列比对和系统发育学分析结果显示其与嗜菌异小杆线虫亲缘关系最近。结合形态学和分子生物学特征, 确定该线虫为嗜菌异小杆线虫沧州品系。该线虫对蛴螬(华北大黑鳃金龟Holotrichia oblita、暗黑鳃金龟H. parallela和铜绿丽金龟Anomala corpulenta 3种金龟子的2龄幼虫)的致病力研究结果表明:处理72 h后, 暗黑鳃金龟幼虫死亡率显著高于另外两种金龟子幼虫 (P<0.05);处理120 h后, 暗黑鳃金龟和铜绿丽金龟幼虫的死亡率分别达到93.3%和80.0%, 二者无显著差异 (P>0.05), 可见该线虫对它们有较强的致病力。不同线虫对暗黑鳃金龟幼虫的致病力结果显示, 嗜菌异小杆线虫沧州品系侵染96 h后, 幼虫的死亡率显著高于Steinernema feltiaeS. longicaudum两种线虫的处理 (P<0.05) , 说明嗜菌异小杆线虫沧州品系的致病力显著高于另外两种线虫。  相似文献

Using isoelectric focusing the enzymes glucose phosphate isomerase, phosphoglucomutase, malate dehydrogenase and acid phosphatase were studied in four fellodistomine species Fellodistomum fellis, Steringotrema ovacutum, Steringophorus agnotus and Steringophorus furciger. Clear genetic separation has been demonstrated for these four species and, in the light of this result, some implications on the taxonomy of these species are discussed.  相似文献

DNA条形码技术在北京百花山地区夜蛾科物种鉴定中的应用   总被引:2,自引:0,他引:2  
为了探讨DNA条形码技术在夜蛾物种鉴定中的可行性, 本研究利用条形码通用引物扩增了北京百花山地区43种夜蛾75个样本的线粒体细胞色素C氧化酶亚基I (mitochondrial cytochrome c oxidase subunit I, COI)基因序列, 以Kimura双参数模型进行种内种间遗传距离分析、 使用邻接法(neighbor-joining, NJ)和最大简约法(maximum parsimony, MP)分别构建系统发育树, 并利用分子序列差异阈值对样本进行分子可操作分类单元(molecular defined operational taxonomic units, MOTU)划分。结果表明: 所有夜蛾种类通过系统发育树可以成功区分; 种内平均遗传距离(0.03%)远远小于种间平均遗传距离(11.29%); 采用较为保守的1%的序列差异阈值将75个夜蛾样本分为42个MOTU, 正确率为95%, 除了MOTU04包含2个物种外, 剩余41个MOTU与形态种呈现一一对应的关系。结果显示, 基于COI基因的DNA条形码对于本研究中所涉及的夜蛾具有较好的区分, 可以作为一种有效的工具在夜蛾科昆虫物种鉴定中进行应用。  相似文献

  • 1 The methods used to indicate the biological state of streams are often based on taxonomic composition, and the abundance of species or other taxa. This ‘taxonomic structure’ varies among ecoregions and cannot be applied to wider geographical areas. Therefore, we assessed the species traits of benthic macroinvertebrates from semi‐natural reference sites as a potential benchmark for large‐scale biomonitoring. Our purpose was to assess the stability of community structure, based on the representation of taxa and of traits, across large gradients of geology (sedimentary to granitic), altitude (65–1982 m), geographical coordinates (0° 48′ W to 7° 20′ E and 42° 52′ to 48° 44′ N), stream order (1–5) and slope (0.5–60‰).
  • 2 We used invertebrate abundance data from the 62 most natural French stream sites available. These abundance data served to weight the occurrence of ‘biological’ traits, such as reproductive characteristics, mobility, resistance forms, food, feeding habits, respiration, and ‘ecological’ traits, such as preferences for temperature, trophic level, saprobity, biogeographic distribution, longitudinal zonation, substratum and current velocity.
  • 3 Multivariate analyses of taxonomic composition demonstrated a clear site gradient from lowlands to uplands and from calcareous to granitic geology. In contrast, community structure based on both biological and ecological traits was stable across environmental gradients.
  • 4 The frequency distribution of biological traits indicated that the stream benthos of the ‘reference sites’ had a mixture of categories which confirmed theoretical predictions for temporally stable and spatially variable habitats. A mixture of ecological trait categories also occurred at our reference sites. Thus, semi‐natural benthic macroinvertebrate communities are functionally diverse. Moreover, we included an initial application of these traits to a case of slightly to moderately polluted sites to show that the impact of humans significantly changes this natural functional diversity.
  • 5 Future studies should focus on the potential for various biological and ecological traits to discriminate different human impacts on the benthic macroinvertebrates of running waters, and on the integration of this functional application into a general ‘reference‐condition’ approach.

The application of projection matrices in population biology to plant and animal populations has a parallel in infectious disease ecology when next-generation matrices (NGMs) are used to characterize growth in numbers of infected hosts ( R 0). The NGM is appropriate for multi-host pathogens, where each matrix element represents the number of cases of one type of host arising from a single infected individual of another type. For projection matrices, calculations of the sensitivity and elasticity of the population growth rate to changes in the matrix elements has generated insight into plant and animal populations. These same perturbation analyses can be used for infectious disease systems. To illustrate this in detail we parameterized an NGM for seven tick-borne zoonoses and compared them in terms of the contributions to R 0 from three different routes of transmission between ticks, and between ticks and vertebrate hosts. The definition of host type may be the species of the host or the route of infection, or, as was the case for the set of tick-borne pathogens, a combination of species and the life stage at infection. This freedom means that there is a broad range of disease systems and questions for which the methodology is appropriate.  相似文献

While there is strong evidence for hydraulic redistribution (HR) of soil water by trees, it is not known if common mycorrhizal networks (CMN) can facilitate HR from mature trees to seedlings under field conditions. Ponderosa pine (Pinus ponderosa) seedlings were planted into root-excluding 61-microm mesh barrier chambers buried in an old-growth pine forest. After 2 yr, several mature trees were cut and water enriched in D(2)O and acid fuchsin dye was applied to the stumps. Fine roots and mycorrhizal root tips of source trees became heavily dyed, indicating reverse sap flow in root xylem transported water from stems throughout root systems to the root hyphal mantle that interfaces with CMN. Within 3 d, D(2)O was found in mesh-chamber seedling foliage > 1 m from source trees; after 3 wk, eight of 10 mesh-chamber seedling stem samples were significantly enriched above background levels. Average mesh-chamber enrichment was 1.8 x greater than that for two seedlings for which the connections to CMN were broken by trenching before D(2)O application. Even small amounts of water provided to mycorrhizas by HR may maintain hyphal viability and facilitate nutrient uptake under drying conditions, which may provide an advantage to seedlings hydraulically linked by CMN to large trees.  相似文献

作为生态学理论应用于实践的有力工具与关键环节,生态系统服务一直是生态学、地理学等学科的研究热点;近些年,亦是自然资源管理、城乡规划设计等实践应用学科的研究前沿。因此,相关学科并驾齐驱,结合生态系统服务研究,不断丰富其涉猎领域,支撑决策者权衡利弊,促进社会与自然环境的可持续发展。然而,在从理论研究走向实践应用的研究链中,存在一些被忽视却值得回顾的问题,生态系统服务研究有没有与实践应用相结合,这种结合的程度如何。研究检索近十年间国内外发表的生态系统服务研究,建立“需求驱动—研究匹配—应用结合程度”的分析框架,通过文献计量和相关分析方法,从生态系统服务研究和实践应用之间结合的有效性这一视角分析国内外该领域进展、存在的不足及发展趋势。经筛选共获得文献122篇;分析发现,绝大部分的研究开篇自述受实践需求驱动,并表示他们的研究与应用相结合。这类结合多集中在自然资源管理、规划类应用,只有少数研究将生态系统服务应用于小尺度的工程,如生态恢复工程(10.7%)和设计(2.4%)。在研究匹配方面,尽管有些文献构建了特殊的指标和技术方法促使两者之间建立链接;但整体而言,生态系统服务研究对实践的影响有限,特别...  相似文献

Marine micro-organisms have been playing highly diverse roles over evolutionary time: they have defined the chemistry of the oceans and atmosphere. During the last decades, the bioreactors with novel designs have become an important tool to study marine microbiology and ecology in terms of: marine microorganism cultivation and deep-sea bioprocess characterization; unique bio-chemical product formation and intensification; marine waste treatment and clean energy generation. In this review we briefly summarize the current status of the bioreactor technology applied in marine microbiology and the critical parameters to take into account during the reactor design. Furthermore, when we look at the growing population, as well as, the pollution in the coastal areas of the world, it is urgent to find sustainable practices that beneficially stimulate both the economy and the natural environment. Here we outlook a few possibilities where innovative bioreactor technology can be applied to enhance energy generation and food production without harming the local marine ecosystem.  相似文献

Nuclear matrices were isolated from maize leaves by the two conventional methods usually employed for the preparation of the corresponding structures of animal origin. It is demonstrated that functionally competent matrices, recognizing and specifically binding the MAR-containing DNA of the mousek-immunoglobulin gene may be prepared by both 2 M NaCl and LIS extractions of maize nuclei.A DNA region with a high affinity for the nuclear matrix was identified at the 5 end of the maizeAdh1-S gene, distal to the promoter region. The presence of sites of reported altered chromatin structure in this particular region is discussed. While the proximity and the cohabitation of MARs with different regulatory elements is a common feature of matrix association regions in animal systems, this is the first plant MAR identified in a region of known significance for gene regulation.  相似文献

Training in Population Ecology asks for scalable applications capable of embarking students on a trip from basic concepts to the projection of populations under the various effects of density dependence and stochasticity. Demography_Lab is an educational tool for teaching Population Ecology aspiring to cover such a wide range of objectives. The application uses stochastic models to evaluate the future of populations. Demography_Lab may accommodate a wide range of life cycles and can construct models for populations with and without an age or stage structure. Difference equations are used for unstructured populations and matrix models for structured populations. Both types of models operate in discrete time. Models can be very simple, constructed with very limited demographic information or parameter‐rich, with a complex density‐dependence structure and detailed effects of the different sources of stochasticity. Demography_Lab allows for deterministic projections, asymptotic analysis, the extraction of confidence intervals for demographic parameters, and stochastic projections. Stochastic population growth is evaluated using up to three sources of stochasticity: environmental and demographic stochasticity and sampling error in obtaining the projection matrix. The user has full control on the effect of stochasticity on vital rates. The effect of the three sources of stochasticity may be evaluated independently for each vital rate. The user has also full control on density dependence. It may be included as a ceiling population size controlling the number of individuals in the population or it may be evaluated independently for each vital rate. Sensitivity analysis can be done for the asymptotic population growth rate or for the probability of extinction. Elasticity of the probability of extinction may be evaluated in response to changes in vital rates, and in response to changes in the intensity of density dependence and environmental stochasticity.  相似文献

Studying the spread of a pathogen in a managed metapopulation such as cattle herds in a geographical region often requires to take into account both the within- and between-herd transmission dynamics. This can lead to high-dimensional metapopulation systems resulting from the coupling of several within-herd transmission models. To tackle this problem, we aim in this paper at reducing the dimension of a tractable but realistic dynamical system reproducing the within-herd spread. The context chosen to illustrate our purpose is bovine viral diarrhoea virus (BVDV) transmission in a cattle herd structured in two age classes and several epidemiological states, including two infectious states (transiently and persistently infected). Different time scales, corresponding to the epidemiological and demographic processes, are identified which allow to build a reduced model. Singular perturbation technique is used to prove that, under some non-restrictive conditions on parameter values, the behaviour of the original system is quite accurately approximated by that of the reduced system. Simulations are also performed to corroborate the approximation quality. Our study illustrates the methodological interest of using singular perturbations to reduce model complexity. It also rigorously proves the biologically intuitive assumption that transiently infected individuals can be neglected in a homogeneous population, when capturing the global dynamics of BVDV spread.  相似文献

Single nucleotide polymorphisms (SNPs) able to describe population differences can be used for important applications in livestock, including breed assignment of individual animals, authentication of mono-breed products and parentage verification among several other applications. To identify the most discriminating SNPs among thousands of markers in the available commercial SNP chip tools, several methods have been used. Random forest (RF) is a machine learning technique that has been proposed for this purpose. In this study, we used RF to analyse PorcineSNP60 BeadChip array genotyping data obtained from a total of 2737 pigs of 7 Italian pig breeds (3 cosmopolitan-derived breeds: Italian Large White, Italian Duroc and Italian Landrace, and 4 autochthonous breeds: Apulo-Calabrese, Casertana, Cinta Senese and Nero Siciliano) to identify breed informative and reduced SNP panels using the mean decrease in the Gini Index and the Mean Decrease in Accuracy parameters with stability evaluation. Other reduced informative SNP panels were obtained using Delta, Fixation index and principal component analysis statistics, and their performances were compared with those obtained using the RF-defined panels using the RF classification method and its derived Out Of Bag rates and correct prediction proportions. Therefore, the performances of a total of six reduced panels were evaluated. The correct assignment of the animals to its breed was close to 100% for all tested approaches. Porcine chromosome 8 harboured the largest number of selected SNPs across all panels. Many SNPs were included in genomic regions in which previous studies identified signatures of selection or genes (e.g. ESR1, KITL and LCORL) that could contribute to explain, at least in part, phenotypically or economically relevant traits that might differentiate cosmopolitan and autochthonous pig breeds. Random forest used as preselection statistics highlighted informative SNPs that were not the same as those identified by other methods. This might be due to specific features of this machine learning methodology. It will be interesting to explore if the adaptation of RF methods for the identification of selection signature regions could be able to describe population-specific features that are not captured by other approaches.  相似文献

The detection and identification of protein variants and abnormally increased modified proteins are important for clinical diagnosis. We applied soft ionization mass spectrometry (MS) to analyze proteins in blood and tissues from various patients. Over the past 8 years, we diagnosed 132 cases (55 kinds) of variant proteins including hemoglobin (Hb), transthyretin (TTR), and Cu/Zn-superoxide dismutase (SOD-1), using MS as the leading technology. Of these variants, eight were new, and nine were the first cases in Japan. Some abnormal Hb cause diseases, and most of them cause erroneous levels of glycated Hb, HbA1c, i.e., a popular index of diabetes. Most of the variant TTR causes amyloidotic polyneuropathy. Variant SOD-1 causes amyotrophic lateral sclerosis. We first showed that immunoprecipitation by a specific antiserum is a reliable and simple method to prepare protein from sera and tissues for analysis by matrix-assisted laser desorption time-of-flight MS, and liquid chromatography-electrospray ionization MS (LC-ESI-MS). The use of this technology has become widespread. Using an immunoprecipitated target protein and LC-ESI-MS, we showed that the ratios of tetra-, di- and a-sialo-transferrin from two cases of congenital glycoprotein deficient syndrome were clearly distinguishable from those of control samples. We first reported a unique modified form of TTR, that is, S-sulfonated TTR, which increased markedly and specifically in three cases with molibdenum cofactor deficiency. We proposed that S-sulfonated TTR is a useful marker for screening this disease. ESI-MS was successfully used for the accurate determination of HbA1c, and we clarified the extent of discrepancies between the HbA1c value measured by conventional methods and the accurate values for samples containing various Hb variants determined by the MS method.  相似文献
