Trisporic acid, its precursors and derivatives are used within zygomycete fungi as communication signals and sexual regulators, and also influence the production rate of the parent compound, β-carotene. Cultivation parameters during growth and the trisporoid production phase of Blakeslea trispora were studied in two-step shake flask cultures and up-scaled fermentations. Comparison of various fermentation protocols allowed the definition of parameters governing trisporoid production. Highest yields were obtained when the initial growth phase allowed for both rapid growth and fast exhaustion of nitrogen and phosporous sources. Onset of trisporoid production is accompanied by a pH drop in the medium and triggered by nutrient limitation, nitrogen depletion being the most important factor. Supplementation of cultures with carbon at low concentration after onset of trisporoid production led to prolonged growth and higher final product accumulation. B. trispora produces trisporoids in two major series, B and C. During a first peak in trisporic acid accumulation, production of trisporic acid B exceeds that of trisporic acid C, which later accumulates at the expense of the trisporic acid B, indicating a variable regulation of the ratio between these metabolites. These data are valuable for tailoring production systems for enrichment of specific intermediates of this complex signal family.  相似文献

The study of sexual interaction in seven (+) and four (−) Blakeslea trispora strains from the All-Russian Collection of Microorganisms showed that all the strains were capable of forming zygotes, albeit to a varying degree. Thus, pairs of strains with active zygote formation and not forming zygospores were identified. It was established that, irrespective of their capacity for zygote formation, all pairs of the (+) and (−) strains synthesized trisporoids in submerged culture. However, zygote-forming pairs accumulated significantly more trisporoids and carotenoids than the strain pairs incapable of forming zygospores. As a result, positive correlation between the capacity for zygote formation in surface culture and trisporoid and carotenoid synthesis in submerged culture was revealed in wild strains.  相似文献

In the course of their sexual interactions, zygomycete fungi communicate via an elaborate series of carotene-derived compounds, namely trisporic acid and its biosynthetic progenitors. A novel building-block strategy allowed the systematic generation of structurally modified trisporoids along with putative early biosynthetic precursors for physiological tests. The impact of discrete structural elements was documented by the ability of individual compounds to induce sexually committed hyphae in Mucor mucedo. The activity screening contributed to establish general structure-function relationships for trisporoid action. Most crucial for activity were the dimension of the longer side chain, the polarity of functional groups at C(4) and C(13), and the number of conjugated double bonds in the side chain. The presence of an oxygen substituent at the cyclohexene ring is not essential for function. The overall biological activity apparently results from the combination of the various structural elements.  相似文献

Stimulation of lycopene synthesis in the mycelium of the mucoraceous fungus Blakeslea trispora was accompanied by major changes in its lipid composition. The phospholipid content in the membrane lip-ids doubled, whereas the sterol and sphingolipid levels changed insignificantly. The amounts of phosphatidyl-choline, phosphatidylethanolamine, and phosphatidic acid in the phospholipids and the triacylglycerol content in the acylglycerols increased. The desaturation degree of fatty acids drastically increased against the background of a decrease in that of the neutral lipids. The data obtained are discussed in relation to the mechanism of action of the lycopene formation stimulator.  相似文献

The phenomenon of heterothallism in filamentous fungi is reviewed, with emphasis on the discussion of hormonal regulation of heterothallic strains of mucoraceous fungi. This process is viewed from the standpoint of current understanding that fungal cells communicate with each other using a special “language,” i.e., signaling chemicals (hormones or pheromones). Physiological and biochemical criteria of distinguishing between heterosexual strains, which make it possible to draw analogies with higher eukaryotes, are set forth for the first time, based on experimental data obtained with Blakeslea trispora. The synthetic pathway to trisporic acids (a zygogenic sex hormone of Mucorales), their relation to carotenoids, and biological functions are described. The similarity (both structural and functional) between fungal, plant, and animal hormones is another topic dealt with. Current understanding of the role of terpenoids in the evolution of sexual communication and transduction is presented with an excursion into microbial endocrinology, a novel field of research in biology. The concluding part of the review analyzes the data on biotechnological implications of the phenomenon of heterothallism. Specifically, it may be used to obtain a series of isoprenoid compounds, such as β-carotene and lycopene (which exhibit pronounced antioxidant activity), as well as sterols and trisporic acids.  相似文献

The addition of plant oils to the growth medium stimulated growth and lipid synthesis in the fungus Blakeslea trispora. However, only oils with high content of linoleic and especially linolenic acid enhanced lycopene formation. The increase in lycopene formation was accompanied by accumulation in the neutral lipid fraction of the fatty acids prevailing in plant oils. In contrast, the influence of exogenous lipids on the fatty acid composition of bulk fungal phospholipids was insignificant. Nonetheless, a marked increase in the content of membrane lipids and of their phosphatidylethanolamine content was revealed. Presumably, the main mechanism of stimulation of lycopene formation by the oils involves an increase in the solubility of lycopene in the triacylglycerols of the lipid bodies, which is due to an increase in the desaturation degree of their fatty acids. The predominance of linoleic and especially of linolenic fatty acid in plant oils is regarded as a criterion for selecting the oil species for the purpose of intensifying lycopene synthesis.  相似文献

It was demonstrated for the first time that the level of carotenogenesis by the heterothallic Blakeslea trispora strains intensively forming zygospores decreased under conditions of a surface cocultivation during their sexual interaction as compared with the strains grown separately. On the contrary, carotenogenesis was stimulated during a sexual interaction of the strains incapable of forming zygotes. In a submerged culture, the zygote-forming pairs of strains synthesized a considerably larger amount of trisporic acids but a smaller amount of carotenoids than the strains not forming zygospores. The discovered inverse dependence between zygote formation and carotenogenesis allowed us to suggest the inability to form zygotes as a criterion for selecting carotenogenic strain pairs.  相似文献

Spores of a minus strain of Mucor mucedo (Bref.) were treated with 1-methyl-[3-nitro]-1-nitro-soguanidine and mutants were isolated either by testing for zygophore induction with externally supplied trisporic acids (TA) or by mating with wild type plus colonies. Mutants were found defective (Tar?) or temperature-sensitive (Tar-Ts) in their reaction towards trisporic acids, blocked or temperature-sensitive in their mating with plus strain (Mat? or Mat-Ts) or temperature-sensitive in zygospore development (Zyg-Ts). The inability to react against externally supplied trisporic acids was not necessarily coupled with an inability to mate with plus strain (phenotype Tar? Mat+). This indicated that the diffusion and uptake of trisporic acids is not a necessary prerequisite to the sexual interaction of Mucor mating types.  相似文献

Separate plus and minus cultures of Blakeslea trispora synthesize small amounts of trisporic acids under specific conditions. These amounts are expressed as a percentage of the trisporic acids (50 mg/liter of medium) synthesized by mixed plus-minus cultures in 5 days. Plus cultures, without additives from minus cultures, synthesize 0.1% trisporic acids. Plus cultures synthesize 0.4% trisporic acids when stimulated by M-factor, a mating-type-specific component synthesized by minus cultures. Minus cultures, without additives from plus cultures, do not synthesize even 0.0001% trisporic acids. Minus cultures synthesize 1% trisporic acids when stimulated by P-factor, a mating-type-specific component synthesized by plus cultures. Minus cultures synthesize M-factor when stimulated by pi, a component synthesized by plus cultures. We speculate that (i) minus cultures synthesize a component, mu, which stimulates P-factor synthesis in plus cultures, and (ii) both M-factor and P-factor are precursors of trisporic acids.  相似文献

In Zygorhynchus moelleri, a homothallic Mucor, triglycendes are the main components of chloroform/methanol extractable lipids. The triglycerides accumulate in the aerial hyphae, particularly in the developing zygospores and in the lateral suspensors, but only after zygospore maturation. They are probably transported from the submerged mycelium to the aerial hyphae. Most of the fatty acid synthetase activity is found in the submerged mycelium. The fatty acid composition of the triglycerides does not change appreciably during sexual development. No influence of trisporic acids has been found on triglyceride synthesis or transport.  相似文献

Chromatographic and spectroscopic characterizations and comparative bioassay data are given for trisporic acid A, the separate 9-cis- and 9-trans-isomers of trisporic acids B and C, and trisporol C, all obtained from 'mated' (plus and minus) cultures of Blakeslea trispora. All five acids show comparable levels of hormone activity on both the mating types of Mucor mucedo, whereas natural trisporol C more specifically affects a plus strain and the laboratory-derived methyl esters are minus-specific. Similarly plus and minus strains of B. trispora convert trisporol C and the esters into trisporic acids at different rates, and they effect different transformations of administered methyl 14C-trisporate C.  相似文献

A tissue culture technique which permits the simultaneous measurement of collagen synthesis and bone resorption has been developed. Cultured neonatal mouse calvaria undergo resorption when stimulated by a number of agents including parathyroid hormone, vitamin D, and prostaglandin E2. Mouse calvaria are of sufficient size to measure the extent of proline incorporation into collagenase-digestible protein. Four chemically diverse stimulators of bone resorption were tested for their effect on collagen synthesis. For each stimulator tested, the dose-response relationships for the stimulation of resorption and the inhibition of collagen synthesis were found to coincide.  相似文献

Two different independent processes are operating in cultured thyroid cells to regulate adenylate cyclase/cyclic AMP responsiveness to thyroid stimulators (thyrotropin and prostaglandin E2): firstly, refractoriness or negative regulation [preceding paper], which is specific for each thyroid stimulator, is not mediated by cyclic AMP and is not accompanied by alteration of adenylate cyclase activity; secondly, positive regulation which is characterized by an augmentation of the cyclic AMP response stimulated by thyrotropin and prostaglandin E2. This process is not specific for each thyroid stimulator and is a state of increased susceptibility of cyclic AMP synthesis to stimulation, accompanied by increased activity of the catalytic subunit of adenylate cyclase. Positive regulation is apparently mediated by increased intracellular cyclic AMP levels. It is a time-dependent and dose-dependent process. Very low concentrations (5-50 micronU/ml) of thyrotropin augmented cyclic AMP synthesis stimulated by thyrotropin and prostaglandin E2 whereas higher concentrations (above 0.1 mU/ml) augmented prostaglandin E2 stimulation but induced refractoriness to thyrotropin. Prostaglandin E2 (0.1 to 10 micronM) augmented thyrotropin stimulation and dibutyryl adenosine 3':5'-monophosphate (0.3 to 2 mM) augmented thyrotropin and prostaglandin E2 stimulation. Positive regulation is a slow process which develops within days and increases up to day 5 in culture. Experiments using inhibitors suggested that protein synthesis is required for the full expression of the increase in adenylate cyclase activity induced by the studied thyroid stimulators.  相似文献

A study was made of some late reactions in the trisporic acid biosynthetic pathway in Mucor mucedo. Trisporic acids induce sexual reproduction in several Mucorales.Two enzymes involved in these reactions, a NADP-dependent dehydrogenase and an esterase, appeared to be highly specific for the minus mating type.The synthesis of these enzymes is stimulated by trisporic acids, indicating a positive control of these hormones upon their own synthesis.The dehydrogenase was histochemically shown to be concentrated in the zygophores of Mucor mucedominus. In the homothallic Zygorhynchus moelleri the copulating main branch (which is known to have a minus character) appeared to be the major site of dehydrogenase activity.  相似文献

When stimulated by equivalent amounts of progametangia-inducing hormones, cultures of the minus mating type of Blakeslea trispora produce about the same quantities of carotenoids as mated cultures of the fungus, which suggests that the stimulation of carotene synthesis during the sexual activity of mated cultures is the result of hormonal action. These hormones were isolated and purified. From spectroscopic analysis of purified samples, it appears that the hormones are identical with trisporic acids B and C. When both mating types of B. trispora were cultivated in one vessel but were kept apart by membrane filters, the formation of sex hormones was not inhibited. Physical contact between the mating types is obviously not required for the induction of sexual activity. The sex hormones also formed in combined cultures of B. trispora-plus and Zygorhynchus moelleri (a homothallic species), but not in combined cultures of B. trispora-minus and Z. moelleri. This is evidence for the hypothesis that the hormones are produced by B. trispora-plus only.  相似文献

The recent data on the pathways of carotenoid biosynthesis, which resulted in revision of the earlier concepts, are analyzed. Trisporoid diversity and their role in the sexual process of mucoraceous fungi, resulting in formation of sexual cells (zygospores) are discussed. Special attention is paid to the role of trisporic acids in carotenogenesis of combined culture of the (+) and (?) strains, in which zygospores are not formed.  相似文献
